Abstract:The individual toxicity and bioaccumulation of cadmium, copper and zinc for common carp juveniles was evaluated in a direct comparison in two experimental setups. First, fish were exposed for 10 days to different metal concentrations in order to link metal bioaccumulation to LC50 values (concentration lethal to 50% of the animals) and incipient lethal levels (ILL, concentration where 50% survives indefinitely). Accumulated metals showed a positive dose dependent uptake for cadmium and copper, but not for zinc. Toxicity was in the order cadmium>copper>zinc with 96h LC50 values for cadmium at 0.20±0.16 μM, for copper at 0.77±0.03 μM, and for zinc at 29.89±9.03 μM respectively. For copper, the 96h exposure was sufficient to calculate the incipient lethal level and therefore 96h LC50 and ILL levels were the same, while for cadmium and zinc 5 to 6 days were needed to reach ILL resulting in slightly lower values at 0.16 μM and 28.33 μM respectively. Subsequently, a subacute exposure experiment was conducted, where carp juveniles were exposed to 2 equitoxic concentrations (10% and 50% of LC50 96 h) of the three metals for 1, 3 and 7 days. Again a significant dose-dependent increase in gill cadmium and copper, but not in zinc, was observed during the 7-day exposure. Copper clearly affected sodium levels in gill tissue, while zinc and cadmium did not significantly alter any of the gill electrolytes. The overall histopathological effects (e.g. hyperemia and hypertrophy) of the metal exposures were mild for most of the alterations. Our study showed that copper an cadmium (but not zinc) showed dose dependent metal accumulation, however this bioaccumulation was only correlated with mortality for cadmium. Metal specific alterations were reduced gill sodium levels in copper exposed fish and oedema of the primary epithelium which typically occurred in both levels of zinc exposure.

Abstract:Copper (Cu) is a common contaminant in aquatic environments, which could cause physiological dysfunction in aquatic organisms. However, few studies have comprehensively examined the impact of copper toxicity in freshwater fish over the past decade. In this research, the oxidative stress, liver transcriptome, intestinal microbiota, and histopathology of common carp (C. carpio) in response to Cu exposure were studied, by exposing juvenile carp to 0.2 mg/ml Cu2+ for 30 days. The results revealed that Cu2+ could induce significant changes in malondialdehyde (MDA) content and antioxidant enzyme (superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx)) activity. The changes in antioxidant enzyme activities indicate that Cu can induce oxidative stress by generating reactive oxygen species (ROS) content. RNA-seq analysis of the liver identified 1069 differentially expressed genes (DEGs) after treatment with 2.0 mg/L Cu2+. Among the DEGs, 490 genes were upregulated and 579 genes were downregulated. GO functional enrichment analysis revealed that Cu could affect the fatty acid biosynthetic process, carnitine biosynthetic process, and activity of carboxylic acid transmembrane transporter. Meanwhile, the most significantly enriched KEGG pathway also included the lipid metabolism pathway. In addition, Cu2+ exposure increased bacterial richness and changed bacterial composition. At the phylum level, we found that the ratio of Bacteroidetes to Firmicutes was increased in the treatment carps, which can regulate intestinal epithelium function and reduce inflammation and immune responses. At the genus level, the abundances of 11 genera were significantly altered after exposure to Cu2+. The altered composition of the microbial community caused by Cu exposure may play a useful role in compensation of the intestinal lesions by Cu exposure. Furthermore, we found that Cu2+ exposure could cause histological alterations such as structural damage to the liver and intestines. The results of this research contribute to a better understanding of mechanisms related to Cu toxicity in fish.

Abstract:Aquatic acidification has been becoming a major issue in recent years. Concomitant with aquatic acidification, Aluminum (Al) toxicity also pose threat to the fish. The purpose of the present study was to assess the combined effect of aquatic acidification and Al on ion concentrations in plasma, ATPase activities in liver and kidney, and health status of fish. Juvenile common carp were exposed to solutions with different combined concentrations of pH (7, 5) and Al (0, 800 mu g L-1) for 1, 3, 9 days. We found that 9 days exposure to Al increased the levels of Na+ and Cl-. The K+ level significantly increased in low pH and low pH/Al treatments after exposure for 1 and 9 days. Furthermore, the activities of Na+/K+-ATPase and Ca2+-ATPase in liver were significantly decreased after exposure for 3 and 9 days, which indicated that the liver played an important role in coping with the stress. Besides, the increase of RBC and decrease of WBC also reconfirmed the toxic effect of low pH and Al on common carp. These results provide some evidence on understanding the impact of low pH and Al on the health status of fish.

Abstract:Background: Heavy metal pollution, particularly lead (Pb), poses a significant threat to aquatic ecosystems and their inhabitants, threatening their delicate balance and long-term viability. This study highlights the urgent need to mitigate heavy metal pollution in aquatic ecosystems. Objective: This study investigates Pb(NO3)2 exposure effects on tissue accumulation, behavioral abnormalities, and hemato-biochemical parameters in common carp (Cyprinus carpio), a widely distributed freshwater fish species. Methodology: Fish (115 ± 5.23 g) were exposed to various Pb(NO3)2 concentrations for 10 and 20 days, representing control (0 %), 25 %, 50 %, and 75 % of the LC50 equivalent to 19.33, 38.66, and 58.0 mg/l, respectively. The standard manual procedure was used for blood sampling. The lead concentration in fish tissue was determined using an atomic absorption spectrophotometer. Results: Results revealed that fish gills showed significant (P < 0.05) increase in Pb(NO3)2 after 10 days, further rising after 20 days. Liver concentrations also rose significantly (P < 0.05) with prolonged exposure and increasing Pb levels. Muscle had lower concentrations. Hematological parameters (RBC, WBC, HB, HCT) decreased with higher Pb(NO3)2 levels. Behavioral and morphological changes were significantly more pronounced in the exposure groups when compared to the control group. Hepatic enzyme activities (AST, ALT), glucose, and lipid levels increased, while total protein decreased. Conclusions: The study highlights Pb(NO3)2 harmful effects on common carp, impacting tissue accumulation, hematological parameters, and biochemical disruptions. It emphasizes the need to monitor and mitigate heavy metal pollution in aquatic environments to safeguard freshwater organisms and ecosystems, and to further increase our understanding of Pb toxicity in freshwater ecosystems.
Impaired intestinal immunity and microbial diversity in common carp exposed to cadmium

Abstract:Waterborne cadmium (Cd) accumulates in the fish intestine and causes irreversible toxicity by disrupting intestinal immunity and microbial diversity. To explore the toxicity of environmentally available high Cd concentration on intestinal immunity and microbial diversity of fish, we selected the widely used bioindicator model species, Common carp (Cyprinus carpio). Literature review and Cd pollution data supported sequential doses of 0.2, 0.4, 0.8, 1.6, 3.2, and 6.4 mg/L Cd for 30 days. Based on intestinal tissue Cd accumulation, previous studies, and environmentally available Cd data, 0.4 and 1.6 mg/L Cd were selected for further studies. Intestinal Cd bioaccumulation increased significantly to ~100 times in fish exposed to 1.6 mg/L Cd. We observed villous atrophy, increased goblet cells with mucus production, muscularis erosion, and thickened lamina propria due to intense inflammatory cell infiltration in the intestine at this Cd concentration. Cd-induced immunosuppression occurred with increased lysozyme, alkaline phosphate (AKP), and acid phosphate (ACP). High levels of catalase (CAT), total antioxidant capacity (T-AOC), malondialdehyde (MDA), and hydrogen peroxide (H2O2) suggested induced oxidative stress and poor metabolism by α-amylase and lipase suppression for Cd toxicity. Proteobacteria (41.2 %), Firmicutes (21.8 %), and Bacteroidetes (17.5 %) were the dominant bacterial phyla in the common carp intestine. Additionally, potential pathogenic Cyanobacteria increased in Cd-treated fish. The decrease of beneficiary bacteria like Aeromonas, and Cetobacterium indicated Cd toxicity. Overall, these findings indicate harmful consequences of high Cd concentration in the intestinal homeostasis and health status of fish.

Abstract:The glutathione metabolism contains crucial antioxidant molecules to defend the organisms against oxidants. Thus, the aim of this study was to investigate the response of the glutathione metabolism in the liver of freshwater fish Oreochromis niloticus exposed to metals (Cu, Cd, Cr, Pb, Zn) in different periods. Fish were exposed to metals (as 1 μg/mL) individually for 1, 7, and 14 days and subsequently antioxidant enzymes (glutathione peroxidase, GPX; glutathione reductase, GR and glutathione S-transferase, GST) and glutathione levels (total glutathione, tGSH; reduced glutathione, rGSH; oxidized glutathione, GSSG and GSH/GSSG ratios) in the liver were measured. There was no fish mortality during the experiments, except Cu exposure. The antioxidant enzymes responded differently to metal exposures depending on metal types and exposure durations. GPX activity increased only after Cd exposure, while GST activity increased following 7 days of all metal exposures. However, GR activity did not alter in most cases. Total GSH and GSH/GSSG levels generally decreased, especially after 7 days. Data showed that metal exposures significantly altered the response of antioxidant system parameters, particularly at day 7 and some recovery occurred after 14 days. This study suggests that the response of antioxidant system could help to predict metal toxicity in the aquatic environments and be useful as an “early warning tool” in natural monitoring studies.

Abstract:Nowadays, pollution of aquatic ecosystems with heavy metals is one of the most important environmental challenges due to toxicity, stability, and bioaccumulation in the environment. Heavy metals accumulate in aquatics and plant tissues and can eventually threaten human health by transmission into the food chain. For this reason, in the present study, the effect of copper sulfate (CuSO4) and its transmission in the food chain of Artemia franciscana, Danio rerio (Zebra), and Astronotus ocellatus (Oscar) were investigated. Initially, lethal concentration of Cu on Artemia was obtained in 24 h. In the first step, Zebra fishes fed with Artemia (under copper sulfate exposure (LC50)) and water (clean and 10% of CuSO4) treatments. In the second step, Oscar fishes fed with Zebra (under copper sulfate exposure (LC50)) and water (clean and 10% of CuSO4) treatments. In the last step, the concentration of Cu was measured in liver tissues of Zebra and Oscar fishes. Also, the variation of glucose and enzymes including Alanine aminotransferase (ALT), Aspartate aminotransferase (AST), and Autologous Conditioned Plasma (ACP) was measured. The results of this study showed that Cu causes histopathological changes liver tissues including hyperemia, necrosis, hepatic portal vein, hepatocyte destruction, nucleolysis, bile stasis, and biliary edema. Also, a strong positive and significant correlation was observed between Cu and increasing glucose and ALT, AST, and ACP.

Abstract:The current study was performed to examine the acute toxicity of mercuric chloride (HgCl2) on the silver carp (Hypophthalmichthys molitrix) larvae. Probit analysis was used to determine the median lethal concentration (LC50). The LC50 values of Hg2+ for the fish larvae at 24, 48, 72, and 96 h were 267.72, 252.97, 225.57, and 97.80 μg/L, respectively. The safe concentration of Hg was 9.78 μg/L for fish larvae. Based on the 96 h LC50, fish were exposed to four different groups including 0, 6.11, 12.23, and 24.45 μg/L for 96 h to assess the effects of different concentrations of Hg2+ on antioxidant capacity, energy metabolism parameters, and related gene expression. The findings revealed that there were no significant differences in the activities of superoxide dismutase (SOD) and total antioxidant capacity (T-AOC) in fish larvae among all the groups (P < 0.05). In the 12.23 μg/L group, fish larvae had a maximum in catalase (CAT) activity. The creatine kinase (CK) activities of fish larvae in control and 6.11 μg/L groups were significantly lower than those groups (P < 0.05). A high concentration of Hg2+ significantly upregulated the mRNA levels of heat shock protein 70 (HSP70) and metallothionein (MT) genes in fish larvae. Furthermore, the IBR index value showed the highest value in the 24.45 μg/L group. Overall, this study provides an increased understanding of the effects of Hg-acute toxicity on silver carp larvae.

Abstract:Copper (Cu) contamination is serious in China, with ≤2.76 mg/L in some waters. Exposure to Cu causes a high toxicity to the aquatic organisms and subsequent ecological risk. To understand fish responses to Cu exposure, we analyzed the metabonomic changes in multiple tissues (gill, liver, and muscle) of Cyprinus flammans using an nuclear magnetic resonance-based metabonomic technique. Our results showed that metabolic alterations are dose-dependent. No significant metabolic alterations in three tissues of fish are caused by 0.25 mg/L Cu. However, 1.53 mg/L Cu caused changes of energy-related metabolites and amino acids, which we suggest are due to enhanced metabolic acidosis in gill and muscle, decreased tricarboxylic acid cycle activity in muscle, increased gluconeogenesis from amino acids in liver, and improved glycogenesis in liver and muscle. The Cori cycle between liver and muscle is concurrently triggered. Furthermore, high concentration of Cu resulted in the alteration of choline metabolism such that we hypothesize that Cu induces membrane damage and detoxification of CuSO4 in gill as well as altered osmoregulation in all three tissues. Choline-O-sulfate in gill may be used as a biomarker to provide an early warning of Cu exposure in C. flammans. Moreover, Cu exposure caused alterations of nucleoside and nucleotide metabolism in both gill and muscle. These findings provide a new insight into the metabolic effects of Cu exposure on C. flammans and highlight the value of metabonomics in the study of metabolic metal disturbance in fish.

Interactions of free copper (II) ions alone or in complex with iron (III) ions with erythrocytes of marine fish Dicentrarchus labrax

Abstract:As a consequence of human activity, various toxicants - especially metal ions - enter aquatic ecosystems and many fish are exposed to considerable levels. As the free ion and in some complexes, there is no doubt that copper promotes damage to cellular molecules and structures through radical formation. Therefore, we have investigated the influence of copper uptake by the red blood of the sea bass (Dicentrarchus labrax), and its oxidative action and effects on cells in the presence of complexed and uncomplexed Fe3+ ions. Erythrocytes were exposed to various concentrations of CuSO4, Fe(NO3)3, and K3Fe(CN)6 for up to 5h, and the effects of copper ions alone and in the combination with iron determined. The results show that inside the cells cupric ion interacts with hemoglobin, causing methemoglobin formation by direct electron transfer from heme Fe2+ to Cu2+. Potassium ferricyanide as a source of complexed iron decreases Met-Hb formation induced by copper ions unlike Fe(NO3)3. We also found that incubation of fish erythrocytes with copper increased hemolysis of cells. But complexed and uncomplexed iron protected the effect of copper. CuSO4 increased the level of lipid peroxidation and a protective effect on complexed iron was observed. Incubation of erythrocytes with copper ions resulted in the loss of a considerable part of thiol content at 10 and 20 microM. This effect was decreased by potassium ferricyanide and Fe(NO3)3 only after 1 and 3h of incubation. The level of nuclear DNA damage assayed by comet assay showed that 20 microM CuSO4 as well as 20 microM Fe(NO3)3 and 10 mM K3Fe(CN)6 induce single- and double-strand breaks. The lower changes were observed after the exposure of cells to K3Fe(CN)6. The data suggest that complexed iron can act protectively against copper ions in contrast to Fe(NO3)3.

Abstract:During the degradation of Microcystis spp. blooms, ammonia and microcystin are released into water column at the same time, which may impact aquatic organisms. This study assessed the responses of oxidative stress to a combination of ammonia and microcystin in larval fish. Newly-hatched larvae and 21-day-old larvae of silver carp Hypophthalmichthys molitrix were exposed to solutions with different concentrations of ammonia (NH3–N) and microcystin-LR (MC-LR). Results indicated significant interaction between NH3–N and MC-LR on glutathione (GSH) and malondialdehyde (MDA) levels in newly-hatched larvae. In the 21-day-old larvae, MDA fluctuated significantly with time and the two toxins, and significant interactions were detected among any two or three factors (time, NH3–N, MC-LR); the superoxide dismutase (SOD) and GSH changed significantly with time and concentrations of toxins and were time or dose-dependent on NH3–N and MC-LR exposure; results also indicated significant interaction on SOD or GSH among any two or three factors.
Algal blooms exacerbate cadmium/zinc bioaccumulation in filter-feeding fish in contaminated water: Mechanisms and environmental implications

Abstract:Eutrophic waters experiencing cyanobacterial blooms often contain high levels of heavy metal pollutants, including cadmium (Cd) and zinc (Zn). The transfer mechanisms of Cd and Zn within the water-algae-fish system, however, remain poorly understood. This study investigates the accumulation and transfer of Cd and Zn in a simulated water-algae‐silver carp ( Hypophthalmichthys molitrix ) ecosystem during Microcystis aeruginosa ( M. aeruginosa ) blooms. Algae demonstrated significant bioaccumulation of Cd and Zn, with Zn proving more toxic to the algae than Cd. In silver carp, the accumulation of Cd and Zn was strengthened in the intestine, liver, and skin following ingestion of M. aeruginosa enriched with these metals. This resulted in a 31% increase in Cd enrichment in the intestine, liver, and a 72% increase in the skin, compared to systems without M. aeruginosa . Zn enrichment in the liver and skin increased by 53% and 61%, respectively. However, muscle accumulation of Cd and Zn showed only minor increases of 0.59 and 3.93 mg/kg. This study elucidated that algae exacerbated the bioaccumulation of Cd and Zn in silver carp through mechanisms such as digestion, absorption, and skin mucosal contact in ecosystems experiencing M. aeruginosa blooms. The accumulation and food chain amplification of heavy metals in eutrophic water bodies increase the ecological toxicity risk to aquatic organisms and local residents. Measures should be considered to simultaneously control both eutrophication and heavy metal pollution in water bodies, with overall results contributing to the scientific management of eutrophic waters containing heavy metals.

Abstract:Bardawil Lake is a unique aquatic ecosystem that provides a habitat for various fish and other marine organisms. This study aimed to analyze the quality of fish species to prove that this lake is free of pollution, not other Egyptian lakes, due to the accumulation of some heavy metals (Cd, Pb, Cu, and Zn) in various tissues of fish species that were caught from this lake. Thirty-five fish samples were caught during the Spring of 2018 from seven different species: Mugil cephalus, Liza auratus, Sparus aurata, Dicentrarchus labrax, Siganus rivulatus, Anguilla angilla, and Solae solea . The Association of Official Analytical Chemists methods using a spectrophotometer determined the biochemical composition. In contrast, atomic absorption spectrometry (AAS) was employed to determine the heavy metals expressed by μg/g wet weight. Results exposed that the accumulation of essential micronutrient (Cu, Zn) content was higher than toxic elements (Cd & Pb) in muscles in order to Zn > Cu > Pb > Cd. Muscles < gills < liver in order of all metals except Pb with order muscles < liver < gills. The metals studied in the muscles were lower than those set by the WHO and the EU standards. The carcinogenic risk with lower allowable limits of 1 × 10 –6 to 1 × 10 –4 in both normal and high consumption groups; target and total target hazard quotients (THQ & HI) in muscles were < 1. The biochemical composition level was highest in the liver, except for protein, which was highest in muscle for all fish species. There is no evidence of harmful contaminants in the muscular tissue of the fish sampled from Bardawil Lake, although fishing activity. Detection of Arsenic, Chromium, Cadmium, Lead, and Mercury in Fish: Effects on the Sustainable and Healthy Development of Aquatic Life and Human Consumers

Abstract:Heavy metals are among the most important pollutants that threaten the aquatic environment when their concentrations exceed certain limits. Some of these metals and metalloids are beneficial and necessary for fish, but others, such as arsenic (As), chromium (Cr), cadmium (Cd), lead (Pb) and mercury (Hg), are non-essential and toxic. In reviewing the recent relevant literature, 4 different continents, 13 different countries, and more than 50 different fish species were analyzed in terms of As, Cr, Cd, Pb and Hg concentrations. According to the comparative results, it was found that in Tercan Dam Lake, Turkey, the highest concentration of Cr was detected in Capoeta umbla (2.455 mg/kg), and of As in Ctenopharyngodon idella (0.774 mg/kg) species. Greater values than the permissible limits of FAO/WHO in terms of As were also found in Andalusia, Southern Spain, in Mullus surmuletus (0.427 mg/kg), and Sardina pilchardus (0.561 mg/kg) and in Sprattus sprattus (0.636 mg/kg) in the Baltic Sea, but a remarkably high content of As (8.48 mg/kg) was determined in Penaeus notialis, found in Guinea, Africa. Moreover, Cd concentration was low to nil in almost all cases, with the exception of Amblyceps mangois species collected from the Dhaleshwari River in Bangladesh, which showed the highest value (0.063 mg/kg). Finally, extremely high levels of Pb were found in Plectropomus pessuliferus (5.05 mg/kg) and Epinephelus summana (2.80 mg/kg) in Jeddah, Saudi Arabia. The Hg content in fish was under the permissible limit in almost all cases, with megrim and red mullet from the Andalusian Sea exhibiting a relatively higher content (0.091 and 0.067 mg/kg). In general, the sequence of accumulation of toxic elements in fish was As > Cr > Pb > Cd > Hg.

Abstract:Increasing toxic metal content in aquatic products has become a universal burden due to the risks to aquatic organisms and human health associated with the consumption of these products. In this study, toxic metal distribution and accumulation in the organs of fish and bivalve species of economic and culinary importance from the lower reaches of the Yangtze River are examined, and the corresponding health risks are also investigated. In general, the viscera and gill show higher concentration of metals than other tissues. The order of the accumulation sequence of metals in muscle tissue of fish and bivalve is Zn > Cu > Mn > Cr > As > Hg > Pb > Cd and Mn > Zn > Cu > As > Cr > Pb > Cd > Hg respectively. Maximum accumulation of Mn (507.50 μg g−1) and Pb (0.51 μg g−1) in the gill tissues indicates the major uptake of these metals from the water column. According to the Hazard Index (HI) calculations (based on USEPA), the analyzed metals will not cause any harmful health effects to individuals for both normal and habitual fish consumers, except for Hg and As in habitual consumers, if these species are consumed at a larger amount. Compared to the Chinese Food Health Criterion and other international standards (WHO/FAO), metal concentrations in the edible muscle tissues of the studied species are lesser than the acceptable levels and found to be fit for human consumption.

Abstract:In sustainable aquaculture systems, copper sulfate (CuSO4) is widely applied as a disinfectant to control parasitic infections and algal growth. However, aquatic organisms may suffer from exposure to excessive concentrations of Cu. Elevated Cu concentrations could activate damage to the respiratory functions of aquatic animals. Thus, this study explored the effects exerted by ferulic acid (FA) on respiratory metabolism, oxidation-related lesions, and the apoptosis parameters of the gills and red blood cells in copper sulfate pentahydrate (CuSO4·5H2O)-treated carp (Cyprinus carpio var. Jian). When the 30-day feeding experiment was completed, the carp were exposed to 12.5 μM of Cu for 4 days. The results indicated that the Cu decreased the oxygen consumption and ammonia excretion rates in the carp, reduced the metabolic- and antioxidant-related enzyme activities and glutathione levels in the carp, and enhanced the caspase activities and reactive oxygen species and malondialdehyde levels in the gills of the carp. Moreover, in addition to the changes in the above parameters, the Cu decreased the cell numbers and hemoglobin concentrations and increased the phosphatidylserine exposure and cytochrome c levels in the red blood cells of the carp. These results demonstrate that Cu is capable of decreasing respiratory metabolism and increasing oxidation-related lesions and apoptosis inside the gills and red blood cells of the fish. However, dietary FA quenched the Cu-induced apoptosis and oxidative lesions by reversing the same biomarker indicators, thereby suppressing the Cu-induced decrease in respiratory metabolism. Thus, FA can be used as a suppressor of Cu stress in fish.

Abstract:Lead (Pb) is one of the most common trace metals in water, and its high concentration in the environment can cause harm to aquatic animals and humans. In the present study, the effects of Pb exposure (3.84 mg/kg) on the morphology, digestive enzyme activity, immune function and microbiota structure of silver carp (Hypophthalmichthys molitrix) intestines within 96 h were detected. Moreover, the correlation between them was analyzed. The results showed that Pb exposure on the one hand severely impaired the intestinal morphology, including significantly shortening the intestinal villi's length, increasing the goblet cells' number, causing the intestinal leukocyte infiltration, and thickening the intestinal wall abnormally, on the other hand, increasing the activity of intestinal digestive enzyme (trypsin and lipase). In addition, the mRNA expressions of structure-related genes (Claudin-7 and villin-1) were down-regulated, and the immune factors genes (IL-8, IL-10 and TNF-α) were up-regulated after Pb exposure. Furthermore, data of the MiSeq sequencing showed that the abundance of membrane transport, immune system function and digestive system of silver carp intestinal microbiota all decreased, while cellular antigens increased. Finally, the canonical correlation analysis (CCA) showed that there were correlations between silver carp's intestinal microbiota and intestinal morphology and immune factors. In conclusion, it is speculated that the entry of Pb into the intestine leads the microbiota dysbiosis, affects the intestinal immunity and digestive function, and further damages the intestinal barrier of silver carp.

Abstract:Due to the non-degradable and persistent nature of metal ions in the environment, they are released into water bodies, where they accumulate in fish. In order to assess pollution in fish, the enzyme, glucose 6-phosphate dehydrogenase (G6PD), has been employed as a biomarker due to sensitivity to various ions. This study investigates the kinetic properties of the G6PD enzyme in yellow catfish (Pelteobagrus fulvidraco), and analyzes the effects of these metal ions on the G6PD enzyme activity in the ovarian cell line (CCO) of channel catfish (Ictalurus punctatus). IC 50 values and inhibition types of G6PD were determined in the metal ions Cu 2+ , Al 3+ , Zn 2+ , and Cd 2+ . While, the inhibition types of Cu 2+ and Al 3+ were the competitive inhibition, Zn 2+ and Cd 2+ were the linear mixed noncompetitive and linear mixed competitive, respectively. In vitro experiments revealed an inverse correlation between G6PD activity and metal ion concentration, mRNA levels and enzyme activity of G6PD increased at the lower metal ion concentration and decreased at the higher concentration. Our findings suggest that metal ions pose a significant threat to G6PD activity even at low concentrations, potentially playing a crucial role in the toxicity mechanism of metal ion pollution. This information contributes to the development of a biomonitoring tool for assessing metal ion contamination in aquatic species.

Abstract:Hazardous materials, such as ammonia and microcystin, are released into lakes during cyanobacterial bloom degradation and may severely impact aquatic organisms. To assess the combined effects of ammonia and microcystin on survival, growth, and oxidative stress of larval fish, 14-day-old larvae of bighead carp Hypophthalmythys nobilis were exposed to solutions with different combined concentrations of ammonia (0, 0.06, 0.264mgL(-1)) and microcystin (0, 2, 10, 30μgL(-1)) for 10 days. Microcystin significantly decreased body length, while ammonia significantly increased body weight, specific growth rate, and condition factor, but there was no significant interaction between ammonia and microcystin on them. Superoxide dismutase, catalase, and malondialdehyde significantly changed with microcystin concentration, whereas glutathione was not affected by microcystin. Ammonia significantly affected the antioxidant system. There were significant interactions between ammonia and microcystin on superoxide dismutase and malondialdehyde. Our data clearly demonstrate that ammonia and microcystin adversely affect bighead carp larvae.
