Abstract:Microplastics (MPs) can enter aquatic food webs through direct ingestion from the environment or indirectly via trophic transfer, but their fate and biological effects within local freshwater food chains remain largely unexplored. In this study, we conducted the first investigation on the trophic transfer and impacts of fluorescently labeled polystyrene microplastics (PS-MPs) (100-nm and 10-μm) in a model freshwater food chain consisting of the snail Bellamya aeruginosa and the commercially important fish Mylopharyngodon piceus, both prevalent in Chinese freshwater ecosystems. Quantitative analysis revealed substantial accumulation of MPs in B. aeruginosa, reaching an equilibrium state within 12 h of exposure. While steady-state was not observed, a pronounced time-dependent bioaccumulation of MPs was evident in M. piceus over a five-week period following dietary exposure through the consumption of contaminated B. aeruginosa. Notably, MPs of both sizes underwent translocation from the gastrointestinal tract to the muscle tissue in M. piceus. High-throughput sequencing of the gut microbiota revealed that exposure to 100-nm MPs significantly altered the microbial community composition in M. piceus, and both particle sizes led to increased relative abundance of potentially pathogenic bacterial genera. Our findings provide novel insights into the trophic transfer, tissue accumulation, and biological impacts of MPs in a model freshwater food chain, highlighting the need for further research to assess the ecological and food safety risks associated with microplastic pollution in freshwater environments.

Abstract:Microplastics (MPs) have received increasing attention in the last decade and are now considered among the most concerning emerging pollutants in natural environments. Here, the current knowledge on microplastic ingestion by wild freshwater fish is reviewed with a focus on the identification of possible factors leading to the ingestion of MPs and the consequences on fish health. Within the literature, 257 species of freshwater fishes from 32 countries have been documented to ingest MPs. MPs ingestion was found to increase with rising level of urbanization, although a direct correlation with MPs concentration in the surrounding water has not been identified. MPs ingestion was detected in all the published articles, with MPs presence in more than 50% of the specimens analyzed in one study out of two. Together with the digestive tract, MPs were also found in the gills, and there is evidence that MPs can translocate to different tissues of the organism. Strong evidence, therefore, exists that MPs may represent a serious risk for ecosystems, and are a direct danger for human health. Moreover, toxicological effects have also been highlighted in wild catches, demonstrating the importance of this problem and suggesting the need for laboratory experiments more representative of the environmental situation.

Abstract:Pollution of microplastics (MPs) has become a global environmental issue due to the difficulty in its degradation and may cause unexpected ecological effects. Nevertheless, little is known about the potential effects of MPs on reproduction toxicity in aquatic species. In this study, adult loach (Paramisgurnus dabryanus, F0 generation) were exposed to two concentrations (1 and 10 mg/L) of polyethylene MPs (PE-MPs) for 15 or 30 days, and the toxic effects in parental loach and the offspring (F1 generation) were examined. Our results showed that PE-MPs exposure could change the indicators content of antioxidant system in the brain, liver, and gonad. PE-MPs can accumulate in the gonads, disrupt the transcription of HPG-axis related genes, alter sex hormone levels, increase cell apoptosis and gonadal pathological lesions, lead to the damage of biological characteristics of semen, and affect the reproduction in F0 generation. PE-MPs remaining in the parental gonads can be transferred to the F1 generation embryos and accumulated on the embryonic chorionic membrane, increasing mortality and malformation rates, accelerating hatching time, and decreasing hatching rate and body length. These results suggest that PE-MPs leads to a potential adverse influence on reproduction and serious impacts on population sustainability. This work provides a new perspective into the effects of MPs on reproductive damage and cross-generational effects in teleost fish, which have implications in fields of freshwater ecology and environmental toxicology.

Abstract:Microplastics (MPs) and methylmercury (MeHg) have attracted increasing attention due to ubiquitous occurrence and toxicity. This study aimed to investigate whether MPs could absorb MeHg and thus modify its bio-concentration and neurotoxicity in the zebrafish larvae (Danio rerio). The pseudo-second-order model (R-2 = 0.989) was found to be suitable for describing the adsorption kinetics of MeHg onto MPs. Compared with Freundlich and Temkin models, the Langmuir isotherm model provided a better fit with the experimental data exhibiting a maximum monolayer adsorption capacity of 54.945 mg/g. These results suggested that adsorption occurs mainly by a chemical process dominated by monolayer adsorption. MPs adsorbed MeHg to form MPs/ MeHg complex, which was ingested by zebrafish larvae, and promoted accumulation of MeHg. Thus, the presence of MPs aggravated the reduction of locomotor activity induced by MeHg, and downregulation of neuro-transmitters related genes, such as ache, gfap and scl1A3b. Metabolome analysis also revealed disrupted glutathione (GSH) metabolism upon exposure of MeHg alone and in combination with MPs, as reflected by the increased in the ratio of GSH and oxidized glutathione. These effects were also confirmed by upregulation of oxidative stress-related genes, such as sod, sod mt and gpx4a. Collectively, these results indicated that MPs could act as a carrier of MeHg and enhance its accumulation in zebrafish, thereby disrupting locomotor activity by excessive oxidative stress. This study provides a scientific basis for improving health risk assessment of environmental pollutants, particularly those potentially able to adsorb to MPs by virtue of their chemical nature.

Abstract:Micro- and nano-plastic (MP/NP) pollution represents a threat not only to marine organisms and ecosystems, but also a danger for humans. The effects of these small particles resulting from the fragmentation of waste of various types have been well documented in mammals, although the consequences of acute and chronic exposure are not fully known yet. In this review, we summarize the recent results related to effects of MPs/NPs in different species of fish, both saltwater and freshwater, including zebrafish, used as model organisms for the evaluation of human health risk posed by MNPs. The expectation is that discoveries made in the model will provide insight regarding the risks of plastic particle toxicity to human health, with a focus on the effect of long-term exposure at different levels of biological complexity in various tissues and organs, including the brain. The current scientific evidence shows that plastic particle toxicity depends not only on factors such as particle size, concentration, exposure time, shape, and polymer type, but also on co-factors, which make the issue extremely complex. We describe and discuss the possible entry pathways of these particles into the fish body, as well as their uptake mechanisms and bioaccumulation in different organs and the role of blood response (hematochemical and hematological parameters) as biomarkers of micro- and nano-plastic water pollution.

Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.

Abstract:Microplastics (MPs) are regarded as a global issue due to their toxicity effects on fish and humans. Fish is a vital origin of human protein, which is necessary for body growth. Contamination of fish by MPs is a major hazard that requires special focus. After exposure to MPs alone or in combination with other pollutants, fish may experience a variety of health issues. MPs can cause tissue damage, oxidative stress, and changes in immune-related gene expression as well as antioxidant status in fish. After being exposed to MPs, fish suffer from neurotoxicity, growth retardation, and behavioral abnormalities. The consequences of MPs on human health are poorly understood. Due to the abundance of MPs in environment, exposure may occur via consumption, inhalation, and skin contact. Humans may experience oxidative stress, cytotoxicity, neurotoxicity, immune system disruption, and transfer of MPs to other tissues after being exposed to them. The toxic effects of MPs in both fish and human are still unknown. This detailed review has the potential to add to existing knowledge about the ecotoxicity effects of MPs in both fish and humans, which will be useful for the forthcoming study.

Effects of microplastics on key reproductive and biochemical endpoints of the freshwater microcrustacean Daphnia magna

Abstract:Human activities have directly impacted the environment, causing significant ecological imbalances. From the different contaminants resulting from human activities, plastics are of major environmental concern. Due to their high use and consequent discharge, plastics tend to accumulate in aquatic environments. There, plastics can form smaller particles (microplastics, MPs), due to fragmentation and weathering, which are more prone to interact with aquatic organisms and cause deleterious effects, including at the basis of different food webs. This study assessed the effects of two microplastics (polyethylene terephthalate, PET; and polypropylene, PP; both of common domestic use) in the freshwater cladoceran species Daphnia magna. Toxic effects were assessed by measuring reproductive traits (first brood and total number of offspring), and activities of biomarkers involved in xenobiotic metabolism (phase I: cytochrome P-450 isoenzymes CYP1A1, 1A2 and 3A4; phase II/conjugation: glutathione S-transferases; and antioxidant defense (catalase)). Both MPs showed a potential to significantly reduce reproductive parameters in D. magna. Furthermore, PET caused a significant increase in some isoenzymes of CYP450 in acutely exposed organisms, but this effect was not observed in chronically exposed animals. Similarly, the activity of the antioxidant defense (CAT) was significantly increased in acutely exposed animals, but not in chronically exposed organisms. This pattern of effects suggests a possible mechanism of long-term adaptation to the presence of the tested MPs. In conclusion, the herein tested MPs have shown the potential to induce deleterious effects on D. magna mainly observed in terms of the reproductive outcomes. Changes at the biochemical level seems transient and are not likely to occur in long term, environmentally exposed crustaceans.

Abstract:Aquatic ecosystems are among the main destination for microplastics (MPs) in the environment. MPs that enter aquatic ecosystems can contribute to pollution together with other co-existing pollutants. However, whether such pollution results in higher or lower toxicity to fish than that caused by co-existing pollutants alone remains controversial. This study aimed at closing this research gap based on 1380 biological endpoints under the background of environmental MP concentrations collected from 55 laboratory studies. Overall, MPs in co-existing pollutant solutions significantly increased the toxicity to fish. Specifically, MPs elevated negative effects on the immune system, metabolism, and oxidative damage. Subgroup analysis indicated that changes in toxicity were related to fish life stage and MP size, but not to co-existing pollutant or MP type. Meta-regression analysis indicated that changes in toxicity were not related to the logarithm of the octanol-water partition coefficient (logKow) or exposure time. Finally, the differences between laboratory research and the actual aquatic environment were discussed from four aspects: MPs, co-existing pollutants, environmental factors, and experimental objects. Our study provides a basis for further understanding the potential impact of MPs on aquatic organisms from a combined pollution perspective. Moreover, our results can provide a reference for the conservation and management of aquatic ecosystems.

Abstract:The widespread occurrence of microplastics (MPs) has resulted in their interaction with biological processes. Thus, there is a great concern about the potential toxicity of MPs on animal and plant cells and on the possibility that MPs reach humans through the food web. In order to shed light on both issues, laboratory assays were performed for evaluating the effects of polyethylene (PE) microparticles on the aquatic plant Spirodela polyrhiza (L.) Schleid. and the gammarid Echinogammarus veneris (Heller, 1865). Moreover, a stock of MP-treated Spirodela plants was used to feed gammarid individuals, and the presence of MP particles in their digestive tracts was analyzed. Results evidenced the lack of toxic effects of MPs on plants, evaluated at growth and physiological level by biometric parameters, pigment content, and photosynthetic performance estimated by chlorophyll fluorescence imaging through the ETPT (EcoTox Photosystem Tool). Only a slight reduction in pigment-related indices in MP-treated plants was observed. A remarkable genotoxic effect was instead highlighted by Comet assay in the hemocytes of gammarid individuals exposed to MPs, with three times more DNA damage (expressed as Tail Moment) in MP-treated individuals compared to control ones. Finally, the gut content of the gammarids fed with MP-treated plants revealed the presence of 7.6 MP particles/individual, highlighting the occurrence of trophic transfer of MPs among freshwater ecosystem organisms. Novel indications about the potential impact of the PE microparticles in the aquatic compartment are provided. Notably, the transfer of MP particles between primary producer and primary consumer organisms of the freshwater trophic chain and the genotoxic effects associated with the ingestion of such particles by gammarids are issues of concern for the aquatic ecosystem and the food web leading to the human diet.

Abstract:Abstract Microplastics have become pervasive environmental pollutants in both freshwater and marine ecosystems. The presence of microplastics have been recorded in the tissues of many wild fish species, and laboratory studies have demonstrated that microplastics can exert adverse health effects. To further investigate the biological mechanisms underlying microplastics toxicity we applied an integrated approach, analyzing the effects of microplastics at transcriptomic, histological and behavioral level. Adult zebrafish have been exposed to two concentrations of high-density polyethylene and polystyrene microplastics for twenty days. Transcriptomic results indicate alterations in the expression of immune system genes and the down-regulation of genes correlated with epithelium integrity and lipid metabolism. The transcriptomic findings are supported by tissue alterations and higher occurrence of neutrophils observed in gills and intestinal epithelium. Even the daily rhythm of activity of zebrafish appears to be affected, although the regular pattern of activity is recovered over time. Considering the transcriptomic and histological findings reported, we hypothesize that the effects on mucosal epithelium integrity and immune response could potentially reduce the organism defense against pathogens, and lead to a different utilization of energy stores.

Abstract:The presence of microplastics in aquatic environments has raised concerns about their abundance and potential hazards to aquatic organisms. This review provides insight into the problem that may be of alarm for freshwater fish. Plastic pollution is not confined to marine ecosystems; freshwater also comprises plastic bits, as the most of plastic fragments enter oceans via rivers. Microplastics (MPs) can be consumed by fish and accumulated due to their size and poor biodegradability. Furthermore, it has the potential to enter the food chain and cause health problems. Evidence of MPs s ingestion has been reported in >150 fish species from both freshwater and marine systems. However, microplastic quantification and toxicity in freshwater ecosystems have been underestimated, ignored, and not reported as much as compared to the marine ecosystem. However, their abundance, influence, and toxicity in freshwater biota are not less than in marine ecosystems. The interaction of MPs with freshwater fish, as well as the risk of human consumption, remains a mystery. Nevertheless, our knowledge of the impacts of MPs on freshwater fish is still very limited. This study detailed the status of the toxicity of MPs in freshwater fish. This review will add to our understanding of the ecotoxicology of microplastics on freshwater fish and give subsequent research directions.

Abstract:To identify surface proteins of eimerian sporozoite, Eimeria tenella sporozoites were labeled with biotin-LC-hydrazide, and analyzed by two-dimensional polyacrylamide gel electrophoresis (2D-PAGE) and Western blotting. More than 200 sporozoite proteins were recognized with silver staining after 2D-PAGE. Among them, 7 biotin-labeled proteins were detected as surface molecules after Western blotting and reaction with horseradish peroxidase conjugated streptavidin. Their molecular masses ranged from 23 to 117 kilo daltons and their isoelectric points from 4.3 to 6.3. It appears that the biotin-labeling technique can be used to analyze of surface proteins of eimerian parasites.

Abstract:ABSTRACT— Resective surgical treatment of medically intractable epilepsy requires accurate identification of the site and extent of the epileptogenic zone responsible for habitual seizures. Epileptogenicity per se is demonstrated electrophysiologically, but interictal and ictal EEG transients, whether recorded extracranially or intracranially, propagate widely and can give rise to false lateralizing and false localizing information. Neuroimaging techniques provide additional important information which greatly enhances confidence in localization derived electrophysiologically. Structural imaging with X‐ray computed tomography and magnetic resonance imaging, as well as functional imaging with positron emission tomography, single photon emission computed tomography, and computerized mapping of electromagnetic activity, used together with other tests of focal functional deficit, 1) increase the confidence with which surgical resection can be performed on the basis of noninvasive tests alone, 2) aid in developing appropriate strategy for intracranial electrode recording when this is necessary, and 3) supplement results of invasive studies sufficiently to justify surgical resection in some patients who otherwise might be rejected for surgery. Addition of these new techniques, therefore, has increased the number of patients considered candidates for surgery, decreased the number of invasive procedures necessary before surgery can be performed, and increased the accuracy of surgical resection.

Abstract:Microplastics (MPs) can be ingested by a variety of species and mainly accumulate in the gut. However, the consequences of MPs exposure in the gut are largely unknown. Here we evaluated the impacts of MPs exposure in zebrafish gut. Animals were experimentally exposed to polystyrene MPs (5-μm beads; 50 μg/L and 500 μg/L) for 21 days and monitored for alterations in tissue histology, enzymatic biomarkers, gut microbiome and metabolomic responses. Inflammation and oxidative stress were observed in the zebrafish gut after exposed to MPs. Furthermore, significant alterations in the gut microbiome and tissue metabolic profiles were observed, with most of these were associated with oxidative stress, inflammation and lipid metabolism. This study provides evidence that MPs exposure causes gut damage as well as alterations in gut metabolome and microbiome, yielding novel insights into the consequences of MPs exposure.

Abstract:<p>Microplastics (MPs) attract global concern due to their ubiquitous existence in aquatic environments. However, the genotoxic effect of MPs on aquatic organisms in the natural environment remains controversial. Therefore, this meta-analysis was conducted by recompiling 44 individual studies from 12 publications to determine whether MPs could induce genotoxicity in aquatic organisms at environmentally relevant concentrations (≤1 mg/L, median = 0.5 mg/L). Multiple genotoxic endpoints were involved, including the percentage of DNA in tail (TDNA%), tail length (TL), olive tail moment (OTM), and the number of micronuclei (NM), and their increases represented the biologically adverse effects (i.e. genotoxicity). The results showed that all included endpoints tended to increase after exposure to MPs, among which TDNA%, TL and NM were significantly increased by 20%, 32% and 81% compared with the control group, respectively. The overall estimate of all endpoints in the MPs-treated groups was remarkably increased by 24%, with high statistical power and no obvious publication bias, suggesting the evident genotoxicity caused by MPs. In addition, the magnitudes of MPs-induced genotoxicity were independent of selected endpoint, MP composition, morphology, exposure concentration and duration, but closely correlated with particle size, living habitat and tested species. Overall, this work provided a reference for the health risk assessment of MPs in the natural environment, contributing to our understanding the action mode of MPs at environmentally relevant concentrations.</p>

Abstract:Microplastics (MPs) are a big and growing environmental concern, with studies showing sublethal to acute biological impacts on typical aquatic organisms. However, little is known about the biological effects of naturally weathered MPs, particularly focusing on mitochondria dysfunction as the key trigger of the biological effects. Therefore, in this study, naturally weathered MPs were produced from day-to-day life products, characterized, and chronically exposed (21 days) to adult zebrafish at the concentration of 0.1 and 1 mg/L. Locomotion and unconditioned anxiety-like behaviour was assessed. Mitochondrial respiration, membrane potential, mitochondrial complex activity and oxidative-related parameters were evaluated in the brain and liver. The results revealed the weathered MPs as a copolymer of propylene and ethylene that induced anxiety-like behaviour. There was an increase in brain catalase activity while the brain lactate dehydrogenase activity was inhibited after exposure to 1 mg/L. Brain glutathione levels were increased while their ratio was not affected. Mitochondrial respiratory chain complex Ⅱ and IV were also significantly decreased in the brain, although not compromising mitochondrial function. On the other hand, exposure to 1 mg/L caused a deficiency in liver mitochondrial respiration and decreased mitochondrial membrane potential, which were associated with the mitochondrial respiratory chain inhibition. An increase in hepatic superoxide dismutase and catalase activity was noticed, supporting the occurrence of ROS-induced ROS release as the potential trigger for the mitochondrial dysfunction. Overall, these findings highlight the potential indirect and cumulative environmental effects these particles may pose to aquatic ecosystems.
