Abstract:Abstract This study explores the enhancement of biocompatible titanium‐based implants through surface functionalization for improved bone healing. Specifically, a near‐beta type Ti‐13Nb‐13Zr alloy is 3D printed using laser powder bed fusion and subsequently textured using nanosecond (ns) and picosecond (ps) direct laser interference patterning (DLIP) to create single‐scale and multi‐scale surface textures. On these textures, the cell behavior, morphology, metabolic activity and osteogenic differentiation potential of human bone marrow stromal cells are assessed using fluorescence microscopy and MTS assays. Moreover, tissue non‐specific alkaline phosphatase activity served as an early osteoblast production marker. Compared to untextured specimens, both types of textures exhibited higher metabolic activity and cell proliferation. Single‐scale ns‐DLIP textures encouraged cell extensions anchored in groove regions, while ps‐DLIP textures with hierarchical structures promoted cell extensions attaching to nanostructures on sidewalls. The groove width and nanotopographies in groove areas facilitated cell spreading. Surface topography, roughness, and surface chemistry (surface energy, wettability) influenced cell adhesion, proliferation, and differentiation. A comprehensive evaluation of DLIP‐generated surface textures, including their topography and chemical states, complements the factors affecting in vitro cell behavior. Overall, this research demonstrates the potential of surface‐functionalized 3Dprinted titanium for a novel generation of biocompatible implants.

Abstract:It is well known that three-dimensional (3D) printing is an emerging technology used to produce customized implants and surface characteristics of implants, strongly deciding their osseointegration ability. In this study, Ti alloy microspheres were printed under selected rational printing parameters in order to tailor the surface micro-characteristics of the printed implants during additive manufacturing by an in situ, controlled way. The laser path and hatching space were responsible for the appearance of the stripy structure (S), while the bulbous structure (B) and bulbous–stripy composite surface (BS) were determined by contour scanning. A nano-sized structure could be superposed by hydrothermal treatment. The cytocompatibility was evaluated by culturing Mouse calvaria-derived preosteoblastic cells (MC3T3-E1). The results showed that three typical microstructured surfaces, S, B, and BS, could be achieved by varying the 3D printing parameters. Moreover, the osteogenic differentiation potential of the S, B, and BS surfaces could be significantly enhanced, and the addition of nano-sized structures could be further improved. The BS surface with nano-sized structure demonstrated the optimum osteogenic differentiation potential. The present research demonstrated an in situ, controlled way to tailor and optimize the surface structures in micro-size during the 3D printing process for an implant with higher osseointegration ability.
In vitro behaviour of human gingival fibroblasts cultured on 3D-printed titanium alloy with hydrogenated TiO2 nanotubes

Abstract:Selective laser melting (SLM), as one of the most common 3D-printed technologies, can form personalized implants, which after further surface modification can obtain excellent osseointegration. To study the surface properties of SLM titanium alloy (Ti6Al4V) with hydrogenated titanium dioxide (TiO2)nanotubes (TNTs) and its influence on the biological behaviour of human gingival fibroblasts (HGFs), we used SLM to prepare 3D-printed titanium alloy samples (3D-Ti), which were electrochemically anodizing to fabricate 3D-TNTs and then further hydrogenated at high temperature to obtain 3D-H2-TNTs. Polished cast titanium alloy (MP-Ti) was used as the control group. The surface morphology, hydrophilicity and roughness of MP-Ti, 3D-Ti, 3D-TNTs and 3D-H2-TNTs were measured and analysed by scanning electron microscopy (SEM), contact angle metre, surface roughness measuring instrument and atomic force microscope, respectively. HGFs were cultured on the four groups of samples, and the cell morphology was observed by SEM. Fluorescence staining (DAPI) was used to observe the number of adhered cell nuclei, while a cell counting kit (CCK-8) was used to detect the early adhesion and proliferation of HGFs. Fluorescence quantitative real time polymerase chain reaction (RT-qPCR) and enzyme-linked immunosorbent assay (ELISA) were used to detect the expression of adhesion-related genes and fibronectin (FN), respectively. The results of this in vitro comparison study indicated that electrochemical anodic oxidation and high-temperature hydrogenation can form a superhydrophilic micro-nano composite morphology on the surface of SLM titanium alloy, which can promote both the early adhesion and proliferation of human gingival fibroblasts and improve the expression of cell adhesion-related genes and fibronectin. Abstract:3D printed medical titanium alloy has shown great advantages in manufacturing complex and personalized scaffolds for tissue engineering. To promote the repair of bone defects, fabrication of a scaffold with high accuracy and excellent cell behaviors is required. However, the partially melted or un-melted powders must be removed in the post-process. In this study, the effects of unfused metal powders on the biocompatibility of 3D printed Ti alloy scaffolds were analyzed by co-culture with MC3T3-E1 cells and RAW 264.7 cells, and the surface treatment strategy was proposed. Results showed that surface treatment can enhance the biocompatibility of the 3D printed Ti alloy scaffolds. The vitro tests with MC3T3-E1 cells demonstrated that the ETi5Cu (etched Ti5Cu) showed the better cell proliferation and osteogenic potential, and the lower cell apoptosis rates. Moreover, higher concentration of powders showed inhibitory effects on cytocompatibility and osteogenesis of MC3T3-E1 cells, and promoted the polarization of macrophages to M1 type. In , the removal of metal powders on the 3D printed scaffolds plays an important role in the biological performance, which provided a promising strategy for the application of Ti implants.

Fibroblasts and osteoblasts behavior after contact with different titanium surfaces used as implant abutment: An in vitro experimental study

Abstract:Background: The goal of this in vitro study was to compare three different surfaces: two types of implant surfaces commercially available ([a] smooth/machined and [b] acid-treated surface) versus (c) anodized surface. Discs were manufactured with commercially pure titanium (CP) grade IV, which were subsequently analyzed by scanning microscopy and fibroblastic and osteoblastic cell cultures. Methods: Ninety-nine discs (5 × 2 mm) were manufactured in titanium grade IV and received different surface treatments: (i) Mach group: machined; (ii) AA group: double acid etch; and (iii) AN group: anodizing treatment. Three discs from each group were analyzed by Scanning Electron Microscopy (SEM) to obtain surface topography images and qualitatively analyzed by EDS. Balb/c 3T3 fibroblasts and pre-osteoblastic cells (MC3T3-E1 lineage) were used to investigate each group's biological response (n = 10/cellular type). The data were compared statistically using the ANOVA one-way test, considered as a statistically significant difference p < 0.05. Results: The AA group had numerous micropores with diameters between 5 and 10 μm, while nanopores between 1 and 5 nm were measured in the AN group. The EDX spectrum showed a high titanium concentration in all the analyzed samples. The contact angle and wetting tension were higher in the AA, whereas similar results were observed for the other groups. A lower result was observed for base width in the AA, which was higher in the other two groups. The AN showed the best values in the fibroblast cells, followed by Mach and AA; whereas, in the culture of the MC3T3 cells, the result was precisely the opposite (AA > Mach > AN). There was similar behavior for cell adhesion for the test groups (Mach and AN), with greater adhesion of Balb/c 3T3 fibroblasts compared to MC3T3 cells; in the AA group, there was greater adherence for MC3T3 cells compared to Balb/c 3T3 fibroblasts. Conclusions: The findings suggest that different surface characteristics can produce different biological responses, possibly cell-line dependent. These findings have important implications for the design of implantable medical devices, where the surface characteristics can significantly impact its biocompatibility.
Comparison In Vitro Study on the Interface between Skin and Bone Cell Cultures and Microporous Titanium Samples Manufactured with 3D Printing Technology Versus Sintered Samples

Abstract:Percutaneous implants osseointegrated into the residuum of a person with limb amputation need to provide mechanical stability and protection against infections. Although significant progress has been made in the biointegration of percutaneous implants, the problem of forming a reliable natural barrier at the level of the surface of the implant and the skin and bone tissues remains unresolved. The use of a microporous implant structure incorporated into the Skin and Bone Integrated Pylon (SBIP) should address the issue by allowing soft and bone tissues to grow directly into the implant structure itself, which, in turn, should form a reliable barrier to infections and support strong osseointegration. To evaluate biological interactions between dermal fibroblasts and MC3T3-E1 osteoblasts in vitro, small titanium discs (with varying pore sizes and volume fractions to achieve deep porosity) were fabricated via 3D printing and sintering. The cell viability MTT assay demonstrated low cytotoxicity for cells co-cultured in the pores of the 3D-printed and sintered Ti samples during the 14-day follow-up period. A subsequent Quantitative Real-Time Polymerase Chain Reaction (RT-PCR) analysis of the relative gene expression of biomarkers that are associated with cell adhesion (α2, α5, αV, and β1 integrins) and extracellular matrix components (fibronectin, vitronectin, type I collagen) demonstrated that micropore sizes ranging from 200 to 500 µm of the 3D printed and sintered Ti discs were favorable for dermal fibroblast adhesion. For example, for representative 3D-printed Ti sample S6 at 72 h the values were 4.71 ± 0.08 (α2 integrin), 4.96 ± 0.08 (α5 integrin), 4.71 ± 0.08 (αV integrin), and 1.87 ± 0.12 (β1 integrin). In contrast, Ti discs with pore sizes ranging from 400 to 800 µm demonstrated the best results (in terms of marker expression related to osteogenic differentiation, including osteopontin, osteonectin, osteocalcin, TGF-β1, and SMAD4) for MC3T3-E1 cells. For example, for the representative 3D sample S4 on day 14, the marker levels were 11.19 ± 0.77 (osteopontin), 7.15 ± 0.29 (osteonectin), and 6.08 ± 0.12 (osteocalcin), while for sintered samples the levels of markers constituted 5.85 ± 0.4 (osteopontin), 4.45 ± 0.36 (osteonectin), and 4.46 ± 0.3 (osteocalcin). In conclusion, the data obtained show the high biointegrative properties of porous titanium structures, while the ability to implement several pore options in one structure using 3D printing makes it possible to create personalized implants for the best one-time integration with both skin and bone tissues.
Surface Modification And Its Effect On Attachment, Spreading, And Proliferation Of Human Gingival Fibroblasts

Abstract:Purpose: The purpose of this study was to exploit potential methods of surface modification for improving the seal between the neck portion of a dental implant and the surrounding soft tissue. Materials and Methods: Titanium surfaces were modified by machining (SM-Ti group); machining and acid etching (AE-Ti group); or machining, acid etching, and depositing 4.5 collagen/hyaluronic acid (col/HA) polyelectrolyte bilayers (CHC-Ti group). These were analyzed using scanning electron microscopy, scanning force microscopy, x-ray photoelectron spectroscopy, contact angle measurement, and quartz crystal microbalance measurement. The degradation behavior of the col/HA multilayer coating was measured. Next, human gingival fibroblasts (HGFs) were cultured on the different surfaces, and cell morphology and spreading were observed using fluorescence microscopy and a shape factor measurement. Cell proliferation was examined by fluorometric quantification of the amount of cellular DNA. Matrix formation of HGFs was determined via enzyme-linked immunosorbent assay. Gene expression was analyzed via reverse transcriptase polymerase chain reaction. Results: Similar surface topology for these three groups was observable on a microscopic scale, and morphologic differences were apparent on the nanoscale. Both acid etching and col/HA deposition improved the hydrophilicity of the titanium surface, in contrast to machining alone. Each col/HA bilayer was about 5 nm thick. The col/HA coating degraded in about a week. Attachment and spreading of HGFs was better on the CHC-Ti surface than on the SM-Ti or AE-Ti surfaces. Moreover, the proliferation and differentiation of HGFs were greatly stimulated when cultured on CHC-Ti. Conclusion: In contrast to two control surfaces (one machined, one machined and acid-etched), col/HA treatment of Ti improved the attachment, spreading, proliferation, and differentiation of HGFs. Abstract:Three-dimensional (3D) printing technology has been widely used to fabricate of various titanium and its alloy implants. However, engineering the 3D printing nanoscaled feature to realize a hierarchical micronano structured surface topography still remains a challenge. On one hand, enhanced bioactivity is always expected on micronano-hybrid biomimetic topography; on the other hand, a typical functional protein in extracellular matrix (ECM) is nanoscaled; therefore, nanoscaled features might affect its binding to specific receptor and subsequent cell response. Here, we engineered a novel hierarchical structure with microparticles and rutile TiO2 nanorods topography that fabricated by 3D printing of pure titanium followed by a hydrothermal process. Although there was no difference on the microscaled feature before/after nanonization, cellular behaviors including adhesion, proliferation, and osteogenic differentiation of mesenchymal stem cells (MSCs) were significantly upregulated on the hierarchical micronano structured topography. Moreover, we demonstrated that the distinct conformation of the initially fibronectin proteins adsorbed on nanorods was more beneficial to cellular adhesion. In vivo test in a rabbit femur model also demonstrated the favorable for new bone formation on the novel hierarchical micronano structured implant-bone interface. These results therefore suggest that the hierarchical micronano structured topography might be a promising surface feature for the new generation of bone implants.

Abstract:The topography at the micro/nanoscale level for biomaterial surfaces has been thought to play vital roles in their interactions with cells. However, discovering the interdisciplinary mechanisms underlying how cells respond to micro-nanostructured topography features still remains a challenge. In this work, ∼37 μm 3D printing used titanium microspheres and their further hierarchical micro-nanostructured spheres through hydrothermal treatment were adopted to construct typical model surface topographies to study the preosteoblastic cell responses (adhesion, proliferation, and differentiation). We here demonstrated that not only the hierarchical micro-nanostructured surface topography but also their distribution density played critical role on cell cytocompatibility. The microstructured topography feature surface with middle-density distributed titanium microspheres showed significantly enhanced cell responses, which might be attributed to the better cellular interaction due to the cell aggregates. However, the hierarchical micro-nanostructured topography surface, regardless of the distribution density of titanium microspheres, improved the cell-surface interactions because of the enhanced initial protein adsorption, thereby reducing the cell aggregates and consequently their responses. This work, therefore, provides new insights into the fundamental understanding of cell-material interactions and will have a profound impact on further designing micro-nanostructured topography surfaces to control cell responses.

Abstract:This work aims to provide a topographical and biological characterization of Additively Manufactured (AMed) Ti6Al4V ELI surfaces subjected to different surface finishing techniques. Unlike traditional biomedical sandblasting methods, femtosecond laser texturing generates new surface roughness and morphologies, which show promise for enhancing the differentiation and proliferation of human bone cells. This work lays the groundwork for optimizing the surfaces of AMed medical devices. Among Additive Manufacturing (AM) technologies, Laser Powder Bed Fusion (LPBF) has made a great contribution to optimizing the production of customized implant materials. However, the design of the ideal surface topography, capable of exerting the best biological effect without drawbacks, is still a subject of study. The aim of the present study is to topographically and biologically characterize AM‐produced Ti6Al4V ELI (Extra Low Interstitial) samples by comparing different surface finishing. Vertically and horizontally samples are realized by LPBF with four surface finishing conditions (as‐built, corundum‐sandblasted, zirconia‐sandblasted, femtosecond laser textured). Bioactivity in vitro tests are performed with human osteoblasts evaluating morphology, metabolic activity, and differentiation capabilities in direct contact with surfaces. Scanning electron microscope and profilometry analysis are used to evaluate surface morphology and samples' roughness with and without cells. All tested surfaces show good biocompatibility. The influence of material surface features is evident in the early evaluation, with the most promising results of morphological study for laser texturing. Deposition orientations seem to influence metabolic activities, with XZ orientation more effective than XY. Current data provide the first positive feedback on the biocompatibility of laser texturing finishing, still poorly described in the literature, and support the future clinical development of devices produced with a combination of LPBF and different finishing treatments.

Effects of the micro-nano surface topography of titanium alloy on the biological responses of osteoblast

Abstract:In clinical applications, osseointegration is essential for the long-term stability of dental implants. Inspired by the hierarchical structure of natural bone, we applied the electrochemical etching (EC) technique to form a micro-nano structure on a titanium alloy (Ti6Al4V) substrate, called EC surface. Sand blasting and acid etching (SLA) and machined (M) methods were employed to generate micro and smooth textures, respectively, as the control groups. The surface topographies of the three substrates were characterized using scanning electron microscopy (SEM). Then, human osteoblast-like cells (MG63) were cultured on substrates, and adhesion, proliferation, morphology, alkaline phosphatase activity (ALP), and gene expression levels of Runt-related transcription factor 2 (RUNX2), osteocalcin (OCN), osteopontin (OPN), and type I collagen (COLIA 1) were analyzed. MG63 cells cultured on the EC Ti alloy substrates displayed better cell adhesion, significant proliferation, and a higher production level of ALP, gene expressions of RUNX2, OCN, OPN and COLIA 1 (p < 0.01 or p < 0.05) compared with those of SLA and M substrates. These results indicate that the micro-nano structure fabricated by electrochemical etching method is beneficial for the biological functions of MG63 cells and may be a promising candidate in dental implants. © 2016 Wiley Periodicals, Inc. Nucleolar organizer regions (NORs) in normal and pathological liver: a quantitative analysis.

Abstract:By means of a silver staining method, Nucleolar Organizer Region-associated proteins (NORs) have been evaluated on paraffin sections of a series of 58 ultrasound-guided liver biopsy specimens. These included 12 normal livers, 12 cirrhotic livers, 12 cases of chronic hepatitis and 22 cases of hepatocellular carcinoma. A significant difference (P less than 0.001) was found between the mean AgNOR scores of the normal and pathological biopsies, and between the non-neoplastic and the carcinomatous lesions. The authors suggest that AgNOR counts, in combination with conventional histocytological criteria, may be a useful method in the diagnosis of liver diseases.
Efficacy of bone defect therapy involving various surface treatments of titanium alloy implants: an in vivo and in vitro study

Abstract:Multiple surface treatment methods for titanium alloy prostheses, widely used in orthopedics, are available; however, these can affect bone integration and regeneration efficiency. In this study, through cell and animal experiments, we devised seven bone implant categories of Ti6Al4V based on surface preparation and post-processing technology (polishing, grit-blasting, fine titanium spraying, coarse titanium spraying, electron beam melting [EBM] printing, selective laser melting [SLM] printing, and post-processed SLM printing) and imaged each microscopic surface structure with a scanning electron microscope (SEM). Mechanical testing revealed excessive post-processing damaged the mechanical properties of the implants. In vitro, human bone marrow mesenchymal stem cells (hBMSCs) were cultured with implants, and the morphology of the cells adhering to the implant surface was observed using SEM and confocal laser scanning microscopy. Cell Counting Kit-8 (CCK-8) semi-quantitatively determined cell activity, indirectly reflecting the proliferation of hBMSCs. Alizarin red and alkaline phosphatase experiments assessed osteogenic differentiation. In vivo, experiments utilized the New Zealand rabbit femoral condyle bone defect model to assess bone regeneration and integration using micro-computed tomography, Van Giesen staining, and Masson staining. We found that 3D-printed implants with regular pore structures were more conducive to hBMSC osteogenic differentiation, while the presence of metal powder on NPT-SLM-printed implants hindered such differentiation. The post-treatment SLM scaffold surface may have some residual semi-melted powder; however, these powder residues have no significant effect on cell activity and differentiation. Surface treatment (grit-blasting and titanium spraying) of planar structures can enhance hBMSC adhesion but does not necessarily promote their differentiation. The framework structure of 3D printing may affect the osteogenic differentiation of hBMSCs, and for SLM-printed implants, excessive pursuit of a "powderless" state will damage the mechanical properties of the implant.

Abstract:The advent of three-dimensional (3D) printing technology has revolutionized the production of customized titanium (Ti) alloy implants. The success rate of implantation and the long-term functionality of these implants depend not only on design and material selection but also on their surface properties. Surface modification techniques play a pivotal role in improving the biocompatibility, osseointegration, and overall performance of 3D-printed Ti alloy implants. Hence, the primary objective of this review is to comprehensively elucidate various strategies employed for surface modification to enhance the performance of 3D-printed Ti alloy implants. This review encompasses both conventional and advanced surface modification techniques, which include physical–mechanical methods, chemical modification methods, bioconvergence modification technology, and the functional composite method. Furthermore, it explores the distinct advantages and limitations associated with each of these methods. In the future, efforts in surface modification will be geared towards achieving precise control over implant surface morphology, enhancing osteogenic capabilities, and augmenting antimicrobial functionality. This will enable the development of surfaces with multifunctional properties and personalized designs. By continuously exploring and developing innovative surface modification techniques, we anticipate that implant performance can be further elevated, paving the way for groundbreaking advancements in the field of biomedical engineering.

Screening the Optimal Patterned Surfaces Consisting of Cell Morphology Mimicking Micro-pillars and Nanotube Arrays for the Design of Titanium Implants

Abstract:Micron/nano scale topographic modification has been a significant focus of interest in current titanium (Ti) surface design. However, the influence of micron/nano structured surface on cell or bacterium behavior on the Ti implant has rarely been systematically evaluated. Moreover, except for popular microgrooves, little work has been carried out on the reaction of cells to the bionic structure. In this study, several micro-pillars mimicking cell morphology were prepared on Ti surfaces by lithography and contact printing (ICP) method, and they were further decorated with nanotube arrays by anodization technology. These surface modifications remarkablly increased the surface roughness of pristine Ti surface from 91.17 nm ± 5.57 nm to be more than 1000 nm, and reduced their water contact angles from 68.3° ± 0.7° to be 16.9° ± 2.4°. Then, the effects of these hierarchical micron/nano scale patterns on the behaviors of MG63 osteoblasts, L929 fibroblasts, SCC epithelial cells and P. gingivalis were studied, aiming to evaluate their performance in osseointegration, gingival epithelial sealing and antibacterial ability. Through an innovative scoring strategy, our findings showed that square micro-pillars with 6 µm width and 2 µm height combined with 85 nm diameter nanotubes was suitable for implant neck design, while square micro-pillars with 3 µm width and 3.6 µm height combined with 55 nm diameter nanotubes was the best for implant body design. Our study reveals the synergistic effect of the hierarchical micron/nano scale patterns on MG63 osteoblasts, L929 fibroblasts, SCC epithelial cells and P. gingivalis functions. It provides insight into the design of biomedical implant surfaces.

Abstract:One of the critical issues in the development of novel metallic biomaterials is the design and fabrication of metallic scaffolds and implants with hierarchical structures mimicking human bones. In this work, selective laser melting (SLM) and electrochemical anodization were applied to fabricate dense Ti-6Al-4V components with macro-micron-nanoscale hierarchical surfaces. Scanning electron microscopy (SEM), 3D laser scanning microscopy (3D LSM), contact angle video system, fluorescence microscopy and spectrophotometer were used to investigate the properties of the samples. The results reveal that the SLMed post-anodization (SLM-TNT) exhibits enhanced or at least comparable wettability, protein adsorption and biological response of mesenchymal stem cells (MSCs) in comparison with the three reference configurations, i.e., the polished Ti-6Al-4V (PO-Ti64), the SLMed Ti-6Al-4V (SLM-Ti64) and the polished Ti-6Al-4V post-anodization (PO-TNT). The improved cytocompatibility of the samples after SLM and anodization should be mainly attributed to the nanoscale tubular features, while the macro-micron-scale structures only lead to slight preference for cell attachment.

The Effects of Graphene on the Biocompatibility of a 3D-Printed Porous Titanium Alloy

Abstract:3D-printed titanium (Ti) materials have attracted much attention in the field of bone tissue repair. However, the combination strength of traditional alloy materials with bone tissue is lower, and the elastic modulus is higher than that of natural bone tissue, which makes the titanium alloy susceptible to stress shielding phenomena after implantation. Therefore, it is urgent to find better surface modification technology. In this study, the physical and chemical properties, toxicity, and proliferation of adipose stem cells of composite graphene-coated titanium alloy (Gr–Ti) were investigated using 3D-printed titanium alloy as a material model. Physical and chemical property tests confirmed that 3D printing could produce porous titanium alloy materials; the compressive strength and elastic modulus of the titanium alloy scaffolds were 91 ± 3 MPa and 3.1 ± 0.4 GPa, matching the elastic modulus of normal bone tissue. The surface characterization shows that graphene can be coated on titanium alloy by a micro-arc oxidation process, which significantly improves the surface roughness of titanium alloy. The roughness factor (Ra) of the Ti stent was 4.95 ± 1.12 μm, while the Ra of the Gr–Ti stent was 6.37 ± 0.72 μm. After the adipose stem cells were co-cultured with the scaffold for 4 h and 24 h, it was found that the Gr–Ti scaffold could better promote the early cell adhesion. CCK-8 tests showed that the number of ADSCs on the G–Ti scaffold was significantly higher than that on the Ti scaffold (p < 0.01). The relative growth rate (RGR) of ADSCs in Gr–Ti was grade 0–1 (non-toxic). In the in vivo experiment of repairing a critical bone defect of a rabbit mandible, the bone volume fraction in the Gr–Ti group increased to 49.42 ± 3.28%, which was much higher than that in the Ti group (39.76 ± 3.62%) (p < 0.05). In conclusion, the porous graphene–titanium alloy promotes the proliferation and adhesion of adipose stem cells with multidirectional differentiation potential, which has great potential for the application of bone tissue engineering in repairing bone defects in the future.

Abstract:There is an evident advantage in personalized customization of orthopedic implants by 3D-printed titanium (Ti) and its alloys. However, 3D-printed Ti alloys have a rough surface structure caused by adhesion powders and a relatively bioinert surface. Therefore, surface modification techniques are needed to improve the biocompatibility of 3D-printed Ti alloy implants. In the present study, porous Ti6Al4V scaffolds were manufactured by a selective laser melting 3D printer, followed by sandblasting and acid-etching treatment and atomic layer deposition (ALD) of tantalum oxide films. SEM morphology and surface roughness tests confirmed that the unmelted powders adhered on the scaffolds were removed by sandblasting and acid-etching. Accordingly, the porosity of the scaffold increased by about 7%. Benefiting from the self-limitation and three-dimensional conformance of ALD, uniform tantalum oxide films were formed on the inner and outer surfaces of the scaffolds. Zeta potential decreased by 19.5 mV after depositing tantalum oxide films. The in vitro results showed that the adhesion, proliferation, and osteogenic differentiation of rat bone marrow mesenchymal stem cells on modified Ti6Al4V scaffolds were significantly enhanced, which may be ascribed to surface structure optimization and the compatibility of tantalum oxide. This study provides a strategy to improve the cytocompatibility and osteogenic differentiation of porous Ti6Al4V scaffolds for orthopedic implants.
