Abstract:The miniaturization of gas chromatography (GC) systems has made it possible to utilize the analytical technique in various on-site applications to rapidly analyze complex gas samples. Various types of miniaturized sensors have been developed for micro-gas chromatography (µGC). However, the integration of an appropriate detector in µGC systems still faces a significant challenge. We present a solution to the problem through integration of µGC with photonic crystal slab (PCS) sensors using transfer printing technology. This integration offers an opportunity to utilize the advantages of optical sensors, such as high sensitivity and rapid response time, and at the same time, compensate for the lack of detection specificity from which label-free optical sensors suffer. We transfer printed a 2D defect free PCS on a borofloat glass, bonded it to a silicon microfluidic gas cell or directly to a microfabricated GC column, and then coated it with a gas responsive polymer. Realtime spectral shift in Fano resonance of the PCS sensor was used to quantitatively detect analytes over a mass range of three orders. The integrated µGC-PCS system was used to demonstrate separation and detection of a complex mixture of 10 chemicals. Fast separation and detection (4 min) and a low detection limit (ng) was demonstrated.
On-chip Fabry–Pérot interferometric sensors for micro-gas chromatography detection

Abstract:We fabricated and characterized on-chip Fabry–Pérot (FP) vapor sensors for the development of on-column micro-gas chromatography (μGC) detectors. The FP sensors were made by coating a thin layer of polymer on a silicon wafer. The air–polymer and polymer–silicon interfaces form an FP cavity, whose resonance wavelengths change in response to the vapor absorption/desorption, thus allowing for rapid detection and quantification of vapors. For proof-of-concept, two polymers (PDMS and SU-8) were used independently and placed in an array in a microfluidic channel, and showed different sensitivities for different vapors. A sub-nano-gram detection limit and sub-second response time were achieved, representing orders of magnitude improvement over those previously reported. This on-chip design will enable the unprecedented integration of optical vapor sensors with μGC systems.
Microfabricated planar glass gas chromatography with photoionization detection

Abstract:We report the development of a microfabricated gas chromatography system suitable for the separation of volatile organic compounds (VOCs) and compatible with use as a portable measurement device. Hydrofluoric acid etching of 95x95mm Schott B270 wafers has been used to give symmetrical hemi-spherical channels within a glass substrate. Two matching glass plates were subsequently cold bonded with the channels aligned; the flatness of the glass surfaces resulted in strong bonding through van der Waals forces. The device comprised gas fluidic interconnections, injection zone and 7.5 and 1.4m long, 320microm internal diameter capillaries. Optical microscopy confirmed the capillaries to have fully circular channel profiles. Direct column heating and cooling could be achieved using a combination of resistive heaters and Peltier devices. The low thermal conductivity of glass allowed for multiple uniform temperature zones to be achieved within a single glass chip. Temperature control over the range 10-200 degrees C was achieved with peak power demand of approximately 25W. The 7.5m capillary column was static coated with a 2microm film of non-polar dimethylpolysiloxane stationary phase. A standard FID and a modified lightweight 100mW photoionization detector (PID) were coupled to the column and performance tested with gas mixtures of monoaromatic and monoterpene species at the parts per million concentration level. The low power GC-PID device showed good performance for a small set of VOCs and sub ng detection sensitivity to monoaromatics.

Abstract:We developed and characterized a rapid, sensitive and integrated optical vapor sensor array for micro-gas chromatography (mGC) applications. The sensor is based on the Fabry-Perot (FP) interferometer formed by a micrometre-thin vapor-sensitive polymer layer coated on a silicon wafer. The thickness and the refractive index of the polymer vary in response to the vapor analyte, resulting in a change in the reflected intensity of the laser impinged on the sensor. In our study, four different polymers were coated on four wells pre-etched on a silicon wafer to form a spatially separated sensor array. A CMOS imager was employed to simultaneously monitor the polymers' response, thus enabling multiplexed detection of a vapor analyte passing through the GC column. A sub-second detection time was demonstrated. In addition, a sub-picogram detection limit was achieved, representing orders of magnitude improvement over the on-chip vapor sensors previously reported.

Abstract:Rapid <i>in situ</i> detection and analysis of trace vapor concentrations at a sub-parts per billion to parts per trillion level remains a challenge for many applications such as indoor air-quality analysis and detection of explosives and narcotics. Micro-gas chromatography (μGC) together with a micro-photoionization detector (μPID) is a prominent method for portable analysis of complex vapor mixtures, but current μPID technology demonstrates poor detection performance compared to benchtop flame ionization detectors (FIDs). This work demonstrates the development of a significantly improved μPID with a sub-picogram detection limit (as low as ∼0.2 pg) comparable to or exceeding that of a benchtop FID, with a large linear dynamic range (>4 orders of magnitude) and robustness (high stability over 200 h of plasma activation). Based on this μPID, a complete μGC–PID system was built and tested on standard sample chromatograms in a laboratory setting to show the system's analytical capabilities and the detection limit down to sub-parts per trillion concentrations (as low as 0.14 ppt). Practical in-field chromatograms on breath and car exhaust were also generated to demonstrate applicability for <i>in situ</i> experimentation. Abstract:Abstract Ion mobility spectrometers can detect gaseous compounds at atmospheric pressure in the range of parts per trillion within a second. Due to their fast response times, high sensitivity, and limited instrumental effort, they are used in a variety of applications, especially as mobile or hand-held devices. However, most real-life samples are gas mixtures, which can pose a challenge for IMS with atmospheric pressure chemical ionization mainly due to competing gas-phase ionization processes. Therefore, we present a miniaturized drift tube IMS coupled to a compact gas chromatograph for pre-separation, built of seven bundled standard GC columns (Rtx-Volatiles, Restek GmbH) with 250 μm ID and 1.07 m in length. Such pre-separation significantly reduces chemical cross sensitivities caused by competing gas-phase ionization processes and adds orthogonality. Our miniaturized GC-IMS system is characterized with alcohols, halocarbons, and ketones as model substances, reaching detection limits down to 70 ppt v with IMS averaging times of just 125 ms. It separates test mixtures of ketones and halocarbons within 180 s and 50 s, respectively. The IMS has a short drift length of 40.6 mm and reaches a high resolving power of R P = 68.

Abstract:A polydimethylsiloxane micromolded separation column is presented with monolithically integrated gas injection, temperature control, and a nanomaterial chemiresistor array for multiparametric gas detection in complex mixtures on a unique single chip platform. Sensor arrays are made of carbon nanotubes, vanadium oxide, and reduced graphene oxide nanomaterials. The functionality of a micromolded gas separation device as opposed to the conventional etched silicon μGC column opens the door to disposable, easily manufactured, stand-alone gas chromatography for clinical diagnostics or remote fieldwork. Combining separation column with a diverse nanomaterial sensor array provides multiparametric detection of gases in a single chip device. The device has the dimensions of a glass slide while drawing <;2 W of power and capable of separation and detection of gases verified using a standard analytical mixture of aliphatic compounds.

Abstract:This study reports on dual on-column Fabry-Pérot (FP) cavity sensor-based gas chromatography (GC) of mixtures of volatile organic compounds (VOCs) utilizing an on-chip device, the so called "microfabricated passive preconcentrator/injector (μPPI)". Comprehensive analysis of the sampling, desorption/injection, and compound separation performance of the μPPI-based optofluidic GC system is described. Here, the combined use of the μPPI and on-column FP cavity sensors in a common GC platform enabled diffusion-based passive sampling, rapid (<7 min) chromatographic separation, and optical detection for the quaternary VOC mixtures of benzene, TCE, toluene, and m-xylene at sub-ppm concentrations with a simpler fluidic setup than conventional GC systems. The FP cavity sensor arrangement provided the means to study the dynamics of the thermal desorption/injection of VOCs by the μPPI and its effect on the GC separation resolution. Our analysis of obtained chromatograms revealed a presence of the competitive adsorptions of VOC mixtures onto the adsorption sites of trapping materials in the μPPI, which decreased the effective sampling rate by ~50% for compounds with high volatility. The validated performance of the optofluidic GC system promises future development of a field deployable GC microsystem incorporating the μPPI and the FP cavity sensors.

Abstract:An integrated and miniaturized Micro-Gas Chromatography with real-time imaging capability for simultaneous chemical separation and detection was developed. Surface Plasmon Resonance imaging (SPRi) was used as a sensitive and real-time imaging based detector for various gaseous chemical mixtures and good gas chromatographs were obtained. The system integrated a home-made miniaturized molecular sieve packed spiral micro-channel column with the SPRi imaging chip and real-time chemical separation and detection were demonstrated using alkanes. The chemical separation processes were simulated using COMSOL and matched well with experimental results. The system enabled the study of chemical separation processes in real-time by miniaturizing and integrating the Micro-GC separation and detection units. This approach can be expanded to multidimensional GC development.
A microfabricated optofluidic ring resonator for sensitive, high-speed detection of volatile organic compounds

Abstract:Advances in microanalytical systems for multi-vapor determinations to date have been impeded by limitations associated with the microsensor technologies employed. Here we introduce a microfabricated optofluidic ring resonator (μOFRR) sensor that addresses many of these limitations. The μOFRR combines vapor sensing and fluidic transport functions in a monolithic microstructure comprising a hollow, vertical SiOx cylinder (250 μm i.d., 1.2 μm wall thickness; 85 μm height) with a central quasi-toroidal mode-confinement section, grown and partially released from a Si substrate. The device also integrates on-chip fluidic-interconnection and fiber-optic probe alignment features. High-Q whispering gallery modes generated with a tunable 1550 nm laser exhibit rapid, reversible shifts in resonant wavelength arising from polymer swelling and refractive index changes as vapors partition into the ~300 nm PDMS film lining the cylinder. Steady-state sensor responses varied in proportion to concentration over a 50-fold range for the five organic vapors tested, providing calculated detection limits as low as 0.5 ppm (v/v) (for m-xylene and ethylbenzene). In dynamic exposure tests, responses to 5 μL injected m-xylene vapor pulses were 710 ms wide and were only 18% broader than those from a reference flame-ionization detector and also varied linearly with injected mass; 180 pg was measured and the calculated detection limit was 49 pg without use of preconcentration or split injection, at a flow rate compatible with efficient chromatographic separations. Coupling of this μOFRR with a micromachined gas chromatographic separation column is demonstrated.

Abstract:Microfabricated gas chromatography (μGC) offers a powerful portable vapor analysis solution for various field applications. Here, we developed a microfluidically integrated architecture of a microfluidic photoionization detector (μPID) on a microfabricated column (μcolumn) via a silicon-on-insulator platform. By eliminating the off-chip interconnects, the microfluidically integrated column PID (iCPID) demonstrated a narrow peak width compared to the off-chip benchmark μPID. Furthermore, the iCPID sensitivity as a function of the bias voltage and temperature were characterized. Finally, an ultracompact (0.9 L, 0.9 kg), automated, and battery-operated μGC system without any benchtop components was constructed based on the iCPID. Rapid (2 min) isothermal separation of eight volatile compounds at room temperature was demonstrated.

Radioimmunoassay for the Measurement of Insulin-Like Growth Factor I in Patients with Pituitary Disease in Comparison with Commercially Available Somatomedin-C Radioimmunoassays

Abstract:A highly sensitive and specific radioimmunoassay for the measurement of insulin-like growth factor I (IGF I) has been developed. The IGF I concentrations were measured in sera of normal subjects, patients with acromegaly, hypophysectomized patients and patients with hyperprolactinaemia. The results were compared with the results obtained after measurement of IGF I in the sera of the same patients with two commercially available radioimmunoassays for somatomedin-C. IGF I was separated from its carrier protein using Sep-Pak C18 cartridges. The total recovery of IGF I by this method was about 100%. The in-house assay shows a high specificity for IGF I and a high sensitivity. As little as 0.1 microgram/l of IGF I can be detected. Only a small amount of serum (25 microliter) is necessary for the IGF I determination and a great number of serum samples (more than 100 per day) can be processed. The mean immunoreactive IGF I concentration was 379 +/- 159 micrograms/l in normal adult subjects, 3340 +/- 1094 micrograms/l in acromegalic patients, and 52 +/- 11 micrograms/l in growth-hormone deficient patients. Patients with hyperprolactinaemia had an IGF I concentration of 433 +/- 112 micrograms/l.
Modulated Ringdown Comb Interferometry for next-generation high complexity trace gas sensing

Abstract:Gas samples relevant to health and environment typically contain a plethora of molecular species that span a huge concentration dynamic range. High-concentration molecules impose a strong absorption background that hinders robust identification of low-concentration species. While mid-infrared frequency comb spectroscopy with high-finesse cavity enhancement has realized many of the most sensitive multi-species trace gas detection to date, its robust performance requires gas samples to contain only weak absorption features to avoid dispersing cavity resonances from the comb line frequencies. Here we introduce a new technique that is free from this restriction, thus enabling the development of next-generation multi-species trace gas sensing with broad applicability to complex and dynamic molecular compositions. The principle of Modulated Ringdown Comb Interferometry is to resolve ringdown dynamics carried by massively parallel comb lines transmitted through a length-modulated cavity. This method leverages both periodicity of the field dynamics and Doppler frequency shifts introduced from a Michelson interferometer. Scalable enhancement of both spectral coverage and cavity finesse is enabled with dispersion immune and high-efficiency data collection. Built upon this platform, we realize in the mid-infrared a product of finesse and spectral coverage that is orders of magnitude better than all prior experiments. We demonstrate the power of this technique by measuring highly dispersive exhaled human breath samples over a vastly expanded spectral coverage of 1,010 cm-1 and with cavity finesse of 23,000. This allows for the first time simultaneous quantification of 20 distinct molecular species at > 1 part-per-trillion sensitivity with their concentrations varying by 7 orders of magnitude.

Abstract:Growing concerns about environmental conditions, public health, and disease diagnostics have led to the rapid development of portable sampling techniques to characterize trace-level volatile organic compounds (VOCs) from various sources. A MEMS-based micropreconcentrator (μPC) is one such approach that drastically reduces the size, weight, and power constraints offering greater sampling flexibility in many applications. However, the adoption of μPCs on a commercial scale is hindered by a lack of thermal desorption units (TDUs) that easily integrate μPCs with gas chromatography (GC) systems equipped with a flame ionization detector (FID) or a mass spectrometer (MS). Here, we report a highly versatile μPC-based, single-stage autosampler-injection unit for traditional, portable, and micro-GCs. The system uses μPCs packaged in 3D-printed swappable cartridges and is based on a highly modular interfacing architecture that allows easy-to-remove, gas-tight fluidic, and detachable electrical connections (FEMI). This study describes the FEMI architecture and demonstrates the FEMI-Autosampler (FEMI-AS) prototype (9.5 cm × 10 cm x 20 cm, ≈500 gms). The system was integrated with GC-FID, and the performance was investigated using synthetic gas samples and ambient air. The results were contrasted with the sorbent tube sampling technique using TD-GC-MS. FEMI-AS could generate sharp injection plugs (≈240 ms) and detect analytes with concentrations <15 ppb within 20 s and <100 ppt within 20 min of sampling time. With more than 30 detected trace-level compounds from ambient air, the demonstrated FEMI-AS, and the FEMI architecture significantly accelerate the adoption of μPCs on a broader scale.

Abstract:Portable and cost-effective gas sensors are gaining demand for a number of environmental, biomedical, and industrial applications, yet current devices are confined into specialized labs and cannot be extended to general use. Here, we demonstrate a part-per-billion-sensitive refractive index gas sensor on a photonic chip based on silicon nitride waveguides functionalized with a mesoporous silica top-cladding layer. Low-concentration chemical vapors are detected by monitoring the output spectral pattern of an integrated unbalanced Mach-Zehnder interferometer having one coated arm exposed to the gas vapors. We retrieved a limit of detection of 65 ppb, 247 ppb, and 1.6 ppb for acetone, isopropyl alcohol, and ethanol, respectively. Our on-chip refractive index sensor provides, to the best of our knowledge, an unprecedented limit of detection for low gas concentrations based on photonic integrated circuits. As such, our results herald the implementation of compact, portable, and inexpensive devices for on-site and real-time environmental monitoring and medical diagnostics.

Abstract:Isoprene is a typical physiological marker that can be used to screen for chronic liver disease (CLD). This work developed a portable micro-integrated chromatography analysis system based on micro-electromechanical system (MEMS) technology, nanomaterials technology and embedded microcontroller technology. The system integrated components such as graphene oxide quantum dots (GOQDs) modified semi-packed microcolumn, In2O3 nanoflower (NF) gas-sensitive detector and 3D printed miniature solenoid valve group. The effectiveness of the separation effect of the micro-integrated system was verified by gas mixture test; the laws of the influence of carrier gas pressure and column temperature on the chromatographic separation performance, respectively, were investigated, and the working conditions (column temperature 90 °C and carrier gas pressure 7.5 kPa) for system testing were determined. The percentages of relative standard deviation (RSD) of the peak areas and retention times obtained for the separated gases were in the range of 0.95% to 6.06%, indicating the good reproducibility of the system. Meanwhile, the microintegrated system could detect isoprene down to 50 ppb at small injection volume (1 mL). The system response increased with increasing isoprene concentration and was linearly correlated with isoprene concentration (R2 = 0.986), indicating that the system was expected to be used for trace detection of isoprene, a marker gas for liver disease, in the future.

Abstract:In recent years, miniaturized analytical instruments have been developing to meet the needs of portable and rapid analysis. The key of miniaturized analytical equipment is the miniaturization and integration of functional modules. This paper aims to develop a miniaturized photometric detector and separation microfluidic chip for a liquid chromatography (LC) system. The detector uses a light-emitting diode to emit ultraviolet light, which is collimated by an internal double lens. A Z-shaped flow cell with a long optical path is designed and fabricated in the separation microfluidic chip with a three-layer structure, which provides a tubing-free connection between the separation and detection unit. Detector performance is evaluated using hemoglobin (Hb) samples, with an upper limit of detection linearity (95 %) of 0.345 AU and stray light level as low as 0.08 %. Additionally, the microchip channel can be filled with cation exchange resin and C18 particles. Finally, an ion LC system and a reversed-phase LC system were constructed based on the miniaturized photometric detector and two microchips with different packed columns, respectively, and were successfully used in the separation and detection of two metabolic markers (glycated hemoglobin or bilirubin). The results of this study are expected to facilitate the development of a portable LC system and their application in community health services and family health management of chronic diseases.

Abstract:Abstract This paper presents the design, fabrication, and characterization of a middle-infrared (MIR) linear variable optical filter (LVOF) and thermopile detectors that will be used in a miniaturized mixed gas detector for CH 4 /CO 2 /CO measurement. The LVOF was designed as a tapered-cavity Fabry-Pérot optical filter, which can transform the MIR continuous spectrum into multiple narrow band-pass spectra with peak wavelength in linear variation. Multi-layer dielectric structures were used to fabricate the Bragg reflectors on the both sides of tapered cavity as well as the antireflective film combined with the function of out-of-band rejection. The uncooled thermopile detectors were designed and fabricated as a multiple-thermocouple suspension structure using micro-electro-mechanical system technology. Experimentally, the LVOF exhibits a mean full-width-at-half-maximum of 400 nm and mean peak transmittance of 70% at the wavelength range of 2.3~5 μm. The thermopile detectors exhibit a responsivity of 146 μV/°C at the condition of room temperature. It is demonstrated that the detectors can achieve the quantification and identification of CH 4 /CO 2 /CO mixed gas.

Gas chromatographic sensing on an optical fiber by mode-filtered light detection

Abstract:A chemical sensor for gas phase measurements is reported which combines the principles of chemical separation and fiber optic detection. The analyzer incorporates an annular column Chromatographic sensor, constructed by inserting a polymer-clad optical fiber into a silica capillary. Light from a helium-neon laser is launched down the fiber, producing a steady intensity distribution within the fiber, but a low background of scattered light. When sample vapor is introduced to the sensor, and an analyte-rich volume interacts with the polymer cladding, Chromatographic retention is observed simultaneously with a change in the local refractive index of the cladding. An increase in cladding refractive index (RI) causes light to be coupled out of the fiber, with detection at a right-angle to the annular column length to provide optimum S/N ratio. This detection mechanism is called mode-filtered light detection. We report a gas Chromatographic separation on a 3.1 m annular column (320 microm i.d. silica tube, 228 microm o.d. fiber with a 12 microm fluorinated silicone clad) of methane, benzene, butanone and chlorobenzene in 6 min. The annular column length was reduced to 22 cm to function as a sensor, with selected organic vapors exhibiting unique retention times and detection selectivity. The detection selectivity is determined by the analyte RI and the partition coefficient into the cladding. The calculated limit of detection (LOD) for benzene vapor is 0.03% by volume in nitrogen, and several chlorinated species had LOD values less than 1%. For binary mixtures of organic vapors, the detected response appears to be the linear combination of the two organic standards, suggesting that the annular column may be useful as a general approach for designing chemical sensors that incorporate separation and optical detection principles simultaneously.
