Abstract:Fluoroquinolones (FQs) have raised significant concerns due to their ubiquitous occurrence and promoting of antimicrobial resistance (AMR). In this study, a molecularly imprinted polymer-diffusive gradient in thin-films (MIP-DGT) sampler is developed for selective measurement of FQs in waters by using a commercial available MIP material as the binding agent. The MIP-DGT shows selective adsorption of the FQs and linearly accumulates the FQs over the deployment time. MIP-DGT measurement is independent of pH (6–8) and ionic strength (IS) (0.01–0.5 M) but is affected by DOM at higher concentrations (~10 mg•L−1), which is due to the altered diffusion coefficients and reduced adsorption on the MIP binding gel. Significant interaction effects of DOM with pH or IS indicate that this is the predominant influence on the MIP-DGT performance, which results in a lower measurement by the MIP-DGT but this is curtailed to some extend with increasing IS or pH. The MIP-DGT measurements, however, correlate well with those by grab sampling in a wastewater treatment plant, suggesting it is reliable for measuring FQs in waters. For the first time, we demonstrate that key water chemistry parameters do have interaction effects on the DGT measurements, which should be considered for the data interpretation. The MIP-DGT is a promising tool to understand the interaction effects of the environmental parameters on the fate, behaviours and bioavailability/toxicity of organic contaminants and improve environmental risk assessments in the environment and modelling.

Abstract:Psychiatric pharmaceuticals are widely distributed in the aquatic environment and have attracted recent attention due to their potential for environmental effects. A robust and reliable in situ passive sampling approach, the diffusive gradients in thin-films (DGT) technique, is developed here to measure 14 psychiatric pharmaceuticals. A new binding material, mixed-mode cation exchange resin (Poly-Sery MCX, 40 mu m, CNW, Germany), was used for the first time in DGT and compared to XAD and HLB. Reliable elution efficiencies of the pharmaceuticals from the binding gels were obtained in methanol/ammonia, and diffusion coefficients for all the compounds were determined. The influence of diffusive layer thickness (0.515-2.015 mm), deployment time (3-168 h), and important environmental conditions-pH (3.02-9.45), ionic strength (0.0001-0.5 M), and dissolved organic matter (0-20 mg L-1)-were evaluated. The capacity of XAD, HLB, and MCX gels for binding all the test pharmaceuticals was similar to 335 mu g per disc, meaning that DGT could theoretically be deployed for over 30 months if there are no competitive effects or confounding factors. The uptake kinetics of psychiatric pharmaceuticals onto MCX gel were much faster than those onto XAD and HLB gels in the first hour. DGT measured concentrations of test pharmaceuticals at two sample points in a river (over 6 days) were comparable to those obtained by grab sampling. This study demonstrates the accuracy and reliability of DGT for measuring psychiatric pharmaceuticals across a wide range of freshwater conditions found in the natural environment.
Field evaluation of diffusive gradients in thin-film passive samplers for wastewater-based epidemiology

Abstract:There is a need for a simple water sampling technique to enable routine monitoring of community drug consumption through wastewater-based epidemiology (WBE). This study investigates the potential use of diffusive gradients in thin films to sample organic compounds (o-DGT) for WBE. Three types of resin gels (HLB, XAD 18, and XDA-1) within o-DGT samplers each were deployed in triplicate at the inlets of two sewage treatment plants of Southern Asian cities. The target compounds included 15 illicit drugs and 18 antibiotics. A comprehensive evaluation was undertaken regarding each resin's ability to accumulate the target compounds and accuracy by comparing active samples. The organic compounds accumulated on each resin gel were characterised at the molecular level using Fourier transform ion-cyclotron-resonance mass spectrometry (FT-ICR MS). The results showed that the HLB resin performed better than the XAD 18 and XDA-1 resins. Based on calculations using the HLB-DGT data, methamphetamine and heroin were the two most popular illicit drugs consumed among the studied populations, and were followed by ketamine and codeine, which agreed well with the authoritative reports and reference data. The total drug consumption in Hanoi was one order of magnitude higher than that in Guangzhou, thus implying a probably more serious drug situation in the former. Overall, the findings of this study demonstrate that o-DGT passive samplers are a promising tool for WBE studies, particularly at WWTPs or in urban streams where an automatic sampler for taking composite water samples is absent.

Abstract:The potential environmental risks of glucocorticoids (GCs) have attracted attention in the past few decades. In this study, a novel diffusive gradients in thin-films (DGT) device and analytical technique based on the second generation of polar enhanced phase (PEP-2), PEP-2-DGT, were developed for sampling and quantifying natural and synthetic GCs in aquatic systems. The capacity of PEP-2 gels for accumulating all target compounds was >600 μg per disc, sufficient for long-term passive sampling of selected GCs, even in wastewaters. Systematic tests were carried out to verify the application of DGT in natural waters and wastewaters. The performance of PEP-2-DGT devices was independent (CDGT/Csoln was in the acceptable range of 0.9-1.1) of a wide range of environmental conditions: ionic strength (0.001-0.5 mol L-1), dissolved organic matter (0-20 mg L-1), and pH (3.06-9.02). It was tested for various diffusive layer thicknesses (0.565-2.065 mm) and different deployment times (10-168 h). Diffusion coefficients (D) of selected GCs through an agarose-based diffusive gel were determined for the first time (3.80-4.85 × 10-6 cm-2 s -1 at 25 °C). Linear correlations between D and log Kow were established for three groups of target GCs (R2 = 0.96-0.99). This could enable prediction of D values for other GCs with similar structures in the future, which will help for rapid screening and emergency monitoring. Concentrations and distribution patterns of analytes obtained by PEP-2-DGT devices in five rivers after 7- and 14-day deployments were in accordance with those measured from grab samples, with total GC concentrations ranging from 7 to 27 ng L-1 at all sampling sites, confirming the reliability and robustness of the DGT devices for monitoring GCs in natural waters. The development of the new DGT technique will help improve understanding of the behavior and fate of these compounds in the aquatic environments.

Abstract:The widespread presence of pharmaceuticals in wastewater effluents after treatment stands as a significant challenge faced in the field of wastewater management and public health. Governments and the scientific community have worked to meet this urgent need for effective solutions. Nevertheless, the development of detection strategies for pharmaceutical monitorization capable of delivering rapid, on-site, and sensitive responses remains an ongoing necessity. In this work, the performance of a previously developed molecularly imprinted polymer (MIP) based electrochemical sensor for detecting atorvastatin (ATV) in wastewater effluents and surface waters is presented. A simple preconcentration method followed by electrochemical measurements by differential pulse voltammetry (DPV) in 0.1 M phosphate buffer (pH = 7), was implemented. The analytical results were validated with those obtained on a set of 16 water samples by ultra-high performance liquid chromatography coupled to tandem mass spectrometry (UHPLC-MS/MS). Additionally, a life cycle assessment (LCA) was conducted to compare the environmental impact of both methodologies. The results obtained demonstrated that ATV detection using MIP-sensor was reliable when compared to the results found by UHPLC-MS/MS presenting a robust linear correlation coefficient of 0.843. The LCA results show that the novel MIP-sensor technique has lower associated environmental impacts than UHPLC-MS/MS, when the current analytical protocol for pharmaceuticals detection is applied. These findings highlight the potential of the developed MIP-sensor as an eco-friendly analytical tool for routine analysis and point-of-care monitoring of ATV in WWTP wastewater and surface water samples.

Abstract:Diffusive gradients in thin films (DGTs) have been well-documented for the measurement of a broad range of organic pollutants in surface water. However, the performance has been challenged by the inherent periodic concentration fluctuations for most organic pollutants. Therefore, there is an urgent need to assess the true time-weighted average (TWA) concentration based on fluctuating concentration profiles. The study aimed to evaluate the responsiveness of DGT and accuracy of TWA concentrations, considering various concentration fluctuating scenarios of 20 pharmaceuticals in surface water. The reliability and accuracy of the TWA concentrations measured by the DGT were assessed by comparison with the sum of cumulative mass of DGT exposed at different stages over the deployment period. The results showed that peak concentration duration (1-5 days), peak concentration fluctuation intensity (6-20 times), and occurrence time of peak concentration fluctuation (early, middle, and late stages) have minimal effect on DGT's response to most target pharmaceutical concentration fluctuations (0.8 < CDGT/CTWA < 1.2). While the downward-bent accumulations of a few pharmaceuticals on DGT occur as the sampling time increases, which could be accounted for by capacity effects during a long-time sampling period. Additionally, the DGT device had good sampling performance in recording short fluctuating concentrations from a pulse event returning to background concentrations with variable intensity and duration. This study revealed a satisfactory capacity for the evaluation of the TWA concentration of pharmaceuticals integrated over the period of different pulse deployment for DGT, suggesting that this passive sampler is ideally suited as a monitoring tool for field application. This study represents the first trial for evaluating DGT sampling performance for pharmaceuticals with multiple concentration fluctuating scenarios over time, which would be valuable for assessing the pollution status in future monitoring campaign.

Abstract:A device of graphene nanoplatelet-based diffusion gradients in thin-films (G-DGT) was developed for in situ sampling of tetracycline (TC), oxytetracycline (OTC) and chlortetracycline (CTC) in aquatic environment. The accumulation of antibiotics in a synthetic solution by the proposed G-DGT was consistent with the theoretical curves predicted by the DGT equation. The values of the detection and quantification limits of G-DGT using high-performance liquid chromatography over the deployment time of 7 days were at the level of μg L−1 for the three antibiotics. The performance of the proposed G-DGT was unaffected by pH (3–9) and ionic strength (0.001–0.7 mol L−1 NaNO3). Fulvic acid did not significantly interfere with the performance of the proposed G-DGT device when the mass ratios between the three antibiotics and fulvic acid were within the range of 1:10-1:100. Humic acid had a significant effect on the performance of the proposed G-DGT for the sampling of the three antibiotics due to strong complexation and coprecipitation between the antibiotics and humic acid. The proposed G-DGT was used for the in situ sampling in spiked freshwaters and livestock culture wastewater and exhibited good precision and accuracy without notable interference from the matrices.

Abstract:Due to their wide applications, occurrence and "PFAS-like" environmental behaviors, ionic liquids (ILs) represent a new challenge for the environmental monitoring community, who require robust analytical methods that can determine accurately and efficiently their environmentally relevant concentrations. A new passive sampling method based on the diffusive gradients in thin films (DGT) technique was developed for the measurement of imidazole-based ILs in waters using a mixed-mode cation exchange (MCX) resin as the adsorbent. The selected binding gel had a high binding capacity (>170 μg per disc) for ILs. Diffusion coefficients measured using a diffusion cell correlated well with alkyl chain lengths ( r 2 = 0.95) and retention times ( r 2 = 0.88), providing a simple and rapid prediction approach for other ILs. The assembled MCX-DGT sampler exhibited a linear accumulation for at least 120 h. MCX-DGT also showed good performance under typical freshwater conditions (pH 5–8, ionic strength 0.001–0.01 M, and humic acid 0–5 mg L −1 ), while still being problematic for aquatic conditions with higher ionic strength (>0.1 M) or DOM (>10 mg L −1 ). Laboratory deployment (for up to 3 days) in spiked natural freshwater (SNW) resulted in linear mass uptakes for the short-chain ILs (C2–C8), and their DGT-measured concentrations agreed well with solution concentrations. However, MCX-DGT significantly overestimated the concentrations of the long-chain ILs (C10–C12) when deployed in SNW for one day or more, which is attributed to the strong competitive adsorption of the long-chain ILs by natural organic matter. In situ field evaluation along with grab sampling found no target ILs in a wastewater treatment plant and its receiving river, implying that these new chemicals might not be widely used in South China now. This is the first report on the DGT technique for ILs and might provide an effective tool for monitoring short chain length ILs in the aquatic environment in the near future.

Abstract:In recent years, as the abuse of antibiotics, hormones, and herbicides has worsened in aquaculture industry, it is important to monitor the concentrations of those trace contaminants in aquaculture water more effortlessly. The aim of this study was to develop a reliable sampling method for chemical monitoring in aquaculture ponds based on the technique of diffusive gradients in thin films (DGT). A binding material (XDA-1 resin) with high adsorption capacity for antibiotics, hormones, and herbicides was selected. In laboratory, the diffusion coefficients of 11 antibiotics, 3 hormones, and 3 herbicides of the XDA-1-DGT devices were tested and ranged from 1.0×10−6 to 8.7×10−5 cm2/s. During the in situ application of XDA-1-DGT devices in different aquaculture ponds, concentrations of 11 antibiotics, 3 hormones, and 3 herbicides in aquaculture water (CSOLN) via grab sampling were also analyzed and found ranging from 0.03 to 6.3 ng/L, lower than the results based on DGT (CDGT) (1.2–1.3×102 ng/L). The values of CDGT/CSOLN were larger than one unit, suggesting that the rates of resupply of target chemicals (antibiotics, hormones, and herbicides) by the desorption and diffusion of pond water were higher than the rates of uptake by XDA-1-DGT, and pond water was proved a sufficient reservoir of antibiotics for DGT application. The value of Log (CDGT/CSOLN) of 1.1 derived from the application in the aquaculture ponds of 14 regions in Eastern China can be a reference value for future chemical monitoring based on DGT technique. And the accuracy of this value was found hardly affected by the physical and chemical properties of chemicals with pKa ranging from −1 to 12, Log Kow ranging from 0 to 4.5, and Log solubility ranging from 0.0 to 4.0.

Abstract:Important biogeochemical processes occur in sediments at fine scales. Sampling techniques capable of yielding information with high resolution are therefore needed to investigate chemical distributions and fluxes and to elucidate key processes affecting chemical fates. In this study, a high-resolution diffusive gradients in thin-films (DGT) technique was systematically developed and tested in a controlled sediment system to measure organic contaminants , antibiotics, for the first time. The DGT probe was used to resolve compound distributions at the mm scale. It also reflected the fluxes from the sediment pore-water and remobilization from the solid phase, providing more dynamic information. Through the fine scale detection, a reduction of re-supply was observed over time, which was concentration and location dependent. Compared to the Rhizon sampling method, antibiotic concentrations obtained by DGT probes were less than the pore-water concentrations, as DGT measures the labile fraction of the compounds. The DGT probe was also tested on an intact sediment core sampled from a lake in China and used to measure the distribution of labile antibiotics with depth in the core at the mm scale. Environmental Implication The abuse of antibiotics and widespread of their residues influences the ecosystem, induces the generation of super-bacteria, and finally poses threat to human health. Sediments adsorbs pollutants from the aquatic environment, while may also release them back to the environment. We systematically developed DGT probe approach for measuring antibiotics in sediment in situ in high resolving power, it provides information at fine scale to help us investigate biogeochemical processes take place in sediment and sediment-water interface.

Abstract:Extensive and long-term use of per- and polyfluoroalkyl substances (PFASs) has caused their widespread distribution in aquatic systems. A new diffusive gradients in thin-films (DGT) passive sampling method based on weak anion exchanger (WAX) binding layer is developed here for monitoring five perfluoroalkyl carboxylic acids (PFCAs), five perfluoroalkanesulfonic acids (PFSAs) and two PFASs (6:2 FTSA and GenX) in waters. Performance of WAX-DGTs was independent of environmental conditions, namely pH (3.03-8.96), ionic strength (1-500 mM), and DOM content (4-30 mg L-1). Diffusion coefficients (D) of the 12 PFASs in the diffusive gels were measured, 9 for the first time. Linear correlations between D and perfluoroalkyl chain lengths (CF2) were established to obtain D for congener chemicals with the similar functional group and structure. The binding capacity of the WAX-DGT sampler was at least 440 μg PFASs per sampler, sufficient for applications in waters across a wide range of conditions and PFASs concentrations. Successful applications of WAX based DGT samplers in a wastewater treatment plant (WWTP) and three rivers has demonstrated that DGT is a powerful tool for monitoring, surveillance and research of these 12 PFASs in aquatic systems, and can be extended to wider suites of PFs in future.
In Situ Measurement of Perfluoroalkyl Substances in Aquatic Systems Using Diffusive Gradients in Thin-Films Technique.

Abstract:To better understand the environmental impact of ubiquitous perfluoroalkyl substances (PFASs) in waters, reliable and robust measurement techniques are needed. As one of the most widely used passive sampling approaches, diffusive gradients in thin-films (DGT) is not only easy to handle but also provides time-weighted analyte concentrations. Based on DGT with XAD18 as a binding agent, we developed a new methodology to measure two frequently detected PFASs in surface waters and wastewaters, i.e. perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS). Their diffusion coefficients in the diffusive gel, measured using an independent diffusion cell, were 4.37 × 10-6 and 5.08 × 10-6 cm2 s-1 at 25 °C, respectively. DGT had a high capacity for PFOA and PFOS at 196 and 246 μg per gel disk, suggesting the DGT sampler was suitable for deployment of several weeks. Time-integrated concentrations of PFOA and PFOS in a natural lake and river, and a municipal wastewater treatment plant effluent using DGT samplers deployed in situ for 12-33 d were comparable to those measured by a solid-phase extraction method coupled with high-frequency grab sampling. This study demonstrates that DGT is an effective tool for in situ monitoring of PFASs in natural waters and wastewaters.
Rapid Determination of Pharmaceuticals from Multiple Therapeutic Classes in Wastewater by Solid-Phase Extraction and Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry

Abstract:A new analytical method utilizing ultra-performance liquid chromatography (UPLC) tandem mass spectrometry (MS/MS) has been developed to determine 16 pharmaceuticals from 8 therapeutic classes in wastewater: bezafibrate, clofibric acid, carbamazepine, caffeine, chloramphenicol, diclofenac, gemfibrozil, indomethacin, ketoprofen, mefenamic acid, metoprolol, nalidixic acid, N,N-diethyl-meta- toluamide, propranolol, sulpiride and trimethoprim. Key parameters of MS/MS, UPLC and solid phase extraction (SPE) were optimized. In general, recovery of target pharmaceuticals was over 70% for the wastewater effluent samples and 50% for the influent samples. The effects of matrix suppression, loss during the pretreatment as well as instrument variability were successfully corrected by two internal standards, and acceptable relative recovery was obtained. Target compounds were quantitatively analyzed using multiple reaction monitoring (MRM) mode, and the detection limits ranged from 0.3 to 20 ng/L. A detailed study, matrix effect in effluent wastewater was also present. The method was applied to detecting pharmaceuticals in the wastewater from three wastewater treatment plants (WWTPs) in Beijing, China and the results demonstrated that most target compounds were detectable in both the influent and effluent, with the mean concentrations ranging from 20.5 to 5775.6 ng/L and 4.6 to 418.6 ng/L, respectively.

This innovative design tackled issues of filter membrane sorption in traditional o-DGT devices and potential gel damage in membrane-less o-DGT devices, showing promising application prospects. The mass accumulation of 15 target BPs was linear over 10 days in both freshwater (r2 ≥ 0.92) and seawater (r2 ≥ 0.94), with no saturation observed. The diffusion coefficients (D) through polyacrylamide diffusive gels ranged from 4.04 × 10-6 to 5.77 × 10-6 cm2 s-1 in freshwater and from 1.74 × 10-6 to 4.69 × 10-6 cm2 s-1 in seawater for the target BPs (except for bisphenol PH) at 22 °C. The D values of the target BPs in seawater were lower than those in freshwater due to the high salinity in seawater (35 ‰). The o-DGT samplers demonstrated good integrity in field applications. The total concentrations of the eight detected BPs ranged from 9.2 to 323 ng L-1, which was consistent with the measurements obtained by grab sampling. Among all BPs, bisphenol S, bisphenol F, and bisphenol A were consistently detected at all sites using both sampling methods. The concentrations of some novel BPs in coastal water measured by grab sampling were comparable to those measured in rivers, suggesting the need to strengthen pollution control of BPs in coastal areas. These results indicate that the o-DGT passive sampling method developed in the present study can be effectively used for monitoring BPs in freshwater and coastal environments.
Wastewater Surveillance for 168 Pharmaceuticals and Metabolites in a WWTP: Occurrence, Temporal Variations and Feasibility of Metabolic Biomarkers for Intake Estimation.

Abstract:Wastewater-based epidemiology (WBE) is amply used for mining information about public health such as the estimation of consumption/intake of certain substances. Yet, proper biomarker selection is critical to obtain reliable data. This study measured a broad range of pharmaceuticals and metabolites in a wastewater treatment plant in Beijing, China, and evaluated their suitability as consumption estimation biomarkers. Wastewater sampling was conducted during a normal week and two holiday weeks to assess the impact of the holiday on population normalized daily mass loads (PNDLs). One hundred and forty-nine out of 168 pharmaceuticals were detected, with 94 analytes being quantified in all sampling events. Moreover, digestive drug cimetidine (< MDL~672 ng L-1) and anabolic steroid trenbolone (< MDL~53 ng L-1) were only detected during holiday weeks. PNDLs of some substances showed disparities between weekdays and weekends during the normal week. This study proposed a framework to diagnose whether a parent compound or its metabolite is suitable for intake/ prevalence rate estimation. Our results support that not all the metabolites can be employed as biomarkers for back-calculation when the in-sewer stability of these compounds is unclear, such as metoprolol acid and O-desmethyl venlafaxine. Public healthcare data for drug utilization were applied to validate the prevalence of average substance use in this study. As a popular anti-epileptic ranging from hundreds to thousands of ng L-1 in this study, the parent compound levetiracetam is more appropriate to be used in WBE under our framework, referring to public healthcare data. This WBE study illustrates the changes in pharmaceutical use and population lifestyle that stem from holidays and commutes. In addition, it can provide data support for the selection of more suitable biomarkers in WBE studies.
TNF‐α Has Not Been Shown to Be Elevated in Headache Patients: A Response

Abstract:This is a comment to the Letter to the Editor by Saxon made on our manuscript published in Headache 8 years ago. The question at hand pertains to the lab values of cerebrospinal fluid (CSF) tumor necrosis factor-α (TNF-α) levels that were utilized as normal in our study of new daily persistent headache (NDPH) patients. ARUP Laboratories (Salt Lake City) was used to run the CSF samples, and they were the ones to set the normal values for serum and CSF. The comment of the writer contends that ARUP had no norms for CSF TNF-α levels, and thus our results are not accurate. It is correct that ARUP created their serum normal values from control subjects, and after re-speaking with ARUP they did not run control specimens for CSF (this information was not provided correctly to the authors 8 years ago when we spoke to the lab), but they continue to run lab values for CSF TNF-α levels currently as they did 8 years ago, suggesting they feel the test is adequate enough to provide results and they provide the same values as a normal for CSF and serum.Also, after speaking with the head of the immunology lab at ARUP (Buck Lozier – senior technologist), the levels of TNF-α in the CSF in normal individuals (without inflammatory conditions) should be about the same as in serum, thus their stated lab value numbers. This is verified in other studies, such as in normal male controls where CSF TNF-α values are actually the same or lower than serum TNF-α values. In addition, other studies that were stated in our original paper looking at neurological populations with non-headache sufferers have shown undetectable CSF TNF-α production with levels of serum TNF-α being higher than CSF. Thus, the elevated CSF levels we presented in our NDPH patients with normal serum levels would seem to be significant values. So in conclusion we do agree with Dr. Saxon that further CSF studies for TNF-α levels in headache patients are needed, especially for NDPH, to verify past results. However, the results of our study we feel should not be questioned as the lab values we utilized to analyze our data were set forth by a national laboratory that continues to evaluate CSF samples for TNF-α and have provided normal values. If anything, serum TNF-α and CSF TNF-α levels should be about the same, and the published data even suggest that normal serum levels are higher than CSF levels; thus, our results showing elevated CSF TNF-α in NDPH patients with normal serum levels would suggest significance.

Abstract:Widespread use of bisphenols (BPs) in our daily life results in their elevated concentrations in waters and the need to study their environmental impact, which demands reliable and robust measurement techniques. Diffusive gradients in thin films (DGT) is an in situ passive sampling approach which provides time-integrated data. In this study we developed a new methodology, based on DGT with activated charcoal (AC) as a binding agent, for measuring three BPs (BPA, BPB, and BPF) which incorporated and tested its performance characteristics. Consistent elution efficiencies were obtained using methanol when concentrations of BPs were low and a methanol-NaOH mixture at high concentrations. The diffusion coefficients of BPA, BPB, and BPF in the diffusive gel, measured using an independent diffusion cell, were 5.03 x 10(-6), 5.64 x 10(-6), and 4.44 x 10(-6) cm(2) s(-1) at 25 degrees C, respectively. DGT with an AC binding gel had a high capacity for BPA, BPB, and BPF at 192, 140, and 194 mu g/binding gel disk, respectively, and the binding performance did not deteriorate with time, up to 254 d after production. Time-integrated concentrations of BPs measured in natural waters using DGT devices with AC gels deployed in situ for 7 d were comparable to concentrations measured by an active sampling method. This study demonstrates that AC-based DGT is an effective tool for in situ monitoring of BPs in waters.

Abstract:On-site therapeutic drug monitoring (TDM) is important for providing a quick and accurate dosing to patients in order to improve efficacy and minimize toxicity. Aminoglycosides such as amikacin, gentamicin, and tobramycin are important antibiotics that have been commonly used to treat infections of chronic bacterial infections in the urinary tract, lung, and heart. However, these aminoglycosides can lead to vestibular and auditory dysfunction. Therefore, TDM of aminoglycosides is important due to their ototoxicity and nephrotoxicity. Here, we have developed a hot embossed poly (methyl methacrylate) (PMMA) microfluidic device featuring an electrokinetic size and mobility trap (SMT) to purify, concentrate, and separate the aminoglycoside antibiotic drugs amikacin, gentamicin, and tobramycin. These drugs were separated successfully from whole blood within 3 min, with 30-fold lower detection limits compared to a standard pinched injection. The limit of detections (LOD) were 3.75 μg/mL for gentamicin, 8.53 μg/mL for amikacin, and 6.00 μg/mL for tobramycin. These are sufficient to cover the therapeutic range for treating sepsis of 6–10 μg/mL gentamicin and tobramycin and 12–20 μg/mL of amikacin. The device is simple and could be mass produced via embossing or injection molding approaches.

Abstract:Intense rainfall and snowmelt events may affect the safety of drinking water, as large quantities of fecal material can be discharged from storm or sewage overflows or washed from the catchment into drinking water sources. This study used β-D-glucuronidase activity (GLUC) with microbial source tracking (MST) markers: human, bovine, porcine mitochondrial DNA markers (mtDNA) and human-associated Bacteroidales HF183 and chemical source tracking (CST) markers including caffeine, carbamazepine, theophylline and acetaminophen, pathogens ( Giardia, Cryptosporidium , adenovirus, rotavirus and enterovirus), water quality indicators ( Escherichia coli , turbidity) and hydrometeorological data (flowrate, precipitation) to assess the vulnerability of 3 drinking water intakes (DWIs) and identify sources of fecal contamination. Water samples were collected under baseline, snow and rain events conditions in urban and agricultural catchments (Québec, Canada). Dynamics of E. coli , HF183 and WWMPs were similar during contamination events, and concentrations generally varied over 1 order of magnitude during each event. Elevated human-associated marker levels during events demonstrated that urban DWIs were impacted by recent contamination from an upstream municipal water resource recovery facility (WRRF). In the agricultural catchment, mixed fecal pollution was observed with the occurrences and increases of enteric viruses, human bovine and porcine mtDNA during peak contaminating events. Bovine mtDNA qPCR concentrations were indicative of runoff of cattle-derived fecal pollutants to the DWI from diffuse sources following rain events. This study demonstrated that the suitability of a given MST or CST indicator depend on river and catchment characteristics. The sampling strategy using continuous online GLUC activity coupled with MST and CST markers analysis was a more reliable source indicator than turbidity to identify peak events at drinking water intakes.
