Abstract:Quality changes in grass carp during superchilled storage at -2C were evaluated in terms of the microstructure, drip loss, pH value, total volatile basic nitrogen (TVB-N), trimethylamine nitrogen (TMA-N), water holding capacity (WHC), salt soluble protein (SSP) and Ca2+-ATPase activity by comparison with frozen storage at -18C. The microstructure of grass carp muscle was seriously damaged with the exhibition of myofibril shrinkage and breakage under frozen storage. Both TVB-N and TMA-N contents in superchilled grass carp were controlled under the acceptable values within 21 days of storage. Significant lower drip loss and higher WHC under superchilled storage was observed compared with frozen storage. Superchilling attenuated the denaturation of myofibrillar protein and the damage of muscle structure associated with freezing, where extractability of SSP and Ca2+-ATPase activity were reduced. A shelf life of 21 days for superchilled grass carp could be estimated by the overall quality evaluation.PRACTICAL APPLICATIONSTraditional frozen fish can be extended for a long time. However, large amounts of water frozen out lead to serious texture change and drip loss after thawing. This study evaluated the potential of superchilling technology to keep the quality of grass carp with little damage caused by freezing storage. The results confirmed that superchilled grass carp could maintain the fresh quality up to 3 weeks. The present study provided a possible application of superchilling for medium period storage of freshwater fish.

Abstract:The crisp grass carp (CGC; Ctenopharyngodon idellus C. et V.), known for its unique texture and flavour, is a culinary delicacy whose quality is significantly influenced by thermal processing. This study employed 4D label-free proteomics and data mining techniques to investigate the proteomic changes in CGC muscle tissue induced by various heating temperatures. CGC samples were subjected to a series of heat treatments at increasing temperatures from 20 °C to 90 °C. Proteins were extracted, digested, and analysed using high-resolution mass spectrometry. The proteomic data were then subjected to extensive bioinformatics analysis, including GO and KEGG pathway enrichment. We identified a total of 1085 proteins, 516 of which were shared across all the temperature treatments, indicating a core proteome responsible for CGC textural properties. Differential expression analysis revealed temperature-dependent changes, with significant alterations observed at 90 °C, suggesting denaturation or aggregation of proteins at higher temperatures. Functional enrichment analysis indicated that proteins involved in amino acid metabolism, glutathione metabolism, and nucleotide metabolism were particularly affected by heat. Textural analysis correlated these proteomic changes with alterations in CGC quality attributes, pinpointing 70 °C as the optimum temperature for maintaining the desired texture. A strong positive correlation between specific upregulated proteins was identified, such as the tubulin alpha chain and collagen alpha-1(IV) chain, and the improved textural properties of CGC during thermal processing, suggesting their potential as the potential biomarkers. This study offers a comprehensive proteomic view of the thermal stability and functionality of CGC proteins, delivering invaluable insights for both the culinary processing and scientific management of CGC. Our findings not only deepen the understanding of the molecular mechanisms underpinning the textural alterations in CGC during thermal processing but also furnish practical insights for the aquaculture industry. These insights could be leveraged to optimize cooking techniques, thereby enhancing the quality and consumer appeal of CGC products.

Abstract:This study aimed to reveal amino acid deamination and decarboxylation activities of spoilage microbiota in chill-stored grass carp fillets. Results showed that microbial deamination activities of umami/sweet-taste amino acids were higher than that of bitter-taste amino acids. The total deamination activity of tested amino acids decreased during the late period of storage, which inhibited the increase of ammonia in fish flesh. Microbial decarboxylation activity of ornithine was much higher than lysine and histidine, which was consistent with the rapid increase of putrescine in fish fillets. Meanwhile, putrescine could be produced in large quantities through arginine deiminase pathway of spoilage bacteria. Glucose utilization by spoilage microbiota was active during the late period of storage, which was consistent with the rapid consumption of lactate and total sugar in fish flesh. Overall, results of this study could be beneficial for revealing fish spoilage mechanisms and providing theoretical guidance for developing fish preservation technologies.
The apoptosis of grass carp (Ctenopharyngodon idella) muscle during postmortem condition regulated by the cytokines via TOR and NF-κB signaling pathways

Abstract:Postmortem alteration by apoptosis has significant effects on flesh quality. Currently, the information necessary to understand the apoptotic behavior and the molecular mechanisms during postmortem alteration in fish muscle is still lacking. Activation of apoptosis and the cytokines involved in regulating apoptosis in fish muscle were evaluated during postmortem condition at 4 °C for 5 days in terms of apoptotic morphology changes, nucleus DNA fragmentation, caspases activation and related gene expressions. The triggering apoptotic mechanisms associated with multiple cytokines transcriptional levels showed that the up-regulated pro-apoptotic mediators [IFN-γ2, TNF-α, IL-6, IL-1β, IL-17D, IL-12p35 and IL-10 (except IL-15)] and the down-regulated anti-apoptotic mediators of [IL-8 and IL-11 (except TGF-β and IL-4)] both regulated apoptosis at early stage, which were regulated by NF-κB and TOR, respectively. Results suggested that transcriptional regulation of multiple cytokines produce a positive outcome on triggering apoptosis.

Abstract:Crisp grass carp products from China are becoming more prevalent in the worldwide fish market because muscle hardness is the primary desirable characteristic for consumer satisfaction of fish fillet products. Unfortunately, current instrumental methods to evaluate muscle hardness are expensive, time-consuming, and wasteful. This study sought to develop classification models for differentiating the muscle hardness of crisp grass carp on the basis of blood analysis. Out of the total 264 grass carp samples, 12 outliers from crisp grass carp group were removed based on muscle hardness (<9 N), and the remaining 252 samples were used for the analysis of seven blood indexes including hydrogen peroxide (H2O2), glucose 6-phosphate dehydrogenase (G6PD), malondialdehyde (MDA), glutathione (GSH/GSSH), red blood cells (RBC), platelet count (PLT), and lymphocytes (LY). Furthermore, six machine learning models were applied to predict the muscle hardness of grass carp based on the training (152) and testing (100) datasets obtained from the blood analysis: random forest (RF), naïve Bayes (NB), gradient boosting decision tree (GBDT), support vector machine (SVM), partial least squares regression (PLSR), and artificial neural network (ANN). The RF model exhibited the best prediction performance with a classification accuracy of 100%, specificity of 93.08%, and sensitivity of 100% for discriminating crisp grass carp muscle hardness, followed by the NB model (93.75% accuracy, 91.83% specificity, and 94% sensitivity), whereas the ANN model had the lowest prediction performance (85.42% accuracy, 81.05% specificity, and 85% sensitivity). These machine learning methods provided objective, cheap, fast, and reliable classification for in vivo crisp grass carp and also prove useful for muscle quality evaluation of other freshwater fish.

Abstract:The yield of freshwater fish has reached more than 20 million tons in China,accounting for about 50% total output of the world fisheries production.The proportion of aquatic product processing is about 80% in developed countries,while the proportion of deep processing of aquatic products in China is only 30% out of total production.Low percentage of freshwater fish deep processing,which has only about 10%,is mainly because of its unpleasant earthy flavor and lack of attraction to many consumers.Thus,the study of earthy taste and odor of freshwater fish not only enriches the theoretical knowledge of flavor chemistry,but also provide an effective solution to the current situation of low proportion of fish processing.Grass carp is one of the most major freshwater fish species in China.In this paper,the different parts of cultured grass carp were used as research object which were killed by exsanguinations or knock-head.The volatile compounds in different parts of grass carp meat were compared by electronic nose.The volatile compounds of grass carp meat were extracted and concentrated by solid phase micro-extraction(SPME).Then the volatiles were identified by gas chromatography-mass spectrometry(GC-MS).The results of electronic nose showed that exsanguination had effect on the volatile compounds of dorsal meat and belly meat,and little effect on red meat of grass carp.There are 27,29 and 47 kinds of volatile compounds identified in dorsal meat,belly meat and red meat of grass carp killed by exsanguination,respectively.Meanwhile,there are 42,41 and 43 kinds of volatile compounds identified in dorsal meat,belly meat and red meat of grass carp killed by knock-head,respectively.Among these components,most of them were volatile carbonyl components and alcohols and their content is above 90%.The results also show slaughter measure had effect on the volatile compounds of dorsal meat and belly meat,and little effect on red meat of grass carp.There are 46 kinds of volatile compounds identified in blood of grass carp,among them 34 kinds are aromatic hydrocarbon and the relative content could reach 75.59%.Therefore,the exsanguination was suggested to apply ahead of processing and consumption of grass carp which will improve food safety and product flavor.

Abstract:The flavor alterations in bighead carp subjected to varying storage temperatures and the underlying metabolic mechanism were elucidated. Analysis of volatile flavor compounds, electronic nose, free amino acids, ATP-related compounds, and sensory evaluations uncovered a progressive flavor deterioration during storage, especially at 25 °C. Metabolomics-based flavor relating component profiling analysis showed that free fatty acids formed various fatty aldehydes including (E, E)-2,4-heptadienal and nonanal under lipoxygenase catalysis. Alcohol dehydrogenase and alcohol acyltransferases were intimately involved in alcohol and ester generation, while alkaline phosphatase, 5'-nucleotidase, and acid phosphatase were closely associated with IMP, Hx, and HxR conversion, respectively. Aeromonas, Serratia, Lactococcus, Pseudomonas, and Peptostreptococcus notably influenced flavor metabolism and enzyme activities. The metabolism disparities of valine, leucine, isoleucine, lysine, and α-linolenic acid could be the primary factors contributing to flavor metabolism distinctions. This study offers novel insights into the flavor change mechanisms and potential regulation strategies of bighead carp during storage.
Diet Affects Muscle Quality and Growth Traits of Grass Carp (Ctenopharyngodon idellus): A Comparison Between Grass and Artificial Feed

Abstract:Fish muscle, the main edible parts with high protein level and low fat level, is consumed worldwide. Diet contributes greatly to fish growth performance and muscle quality. In order to elucidate the correlation between diet and muscle quality, the same batch of juvenile grass carp (Ctenopharyngodon idellus) were divided into two groups and fed with either grass (Lolium perenne, Euphrasia pectinata and Sorghum sudanense) or artificial feed, respectively. However, the different two diets didn't result in significant differences in all the detected water quality parameters (e.g., Tm, pH, DO, NH3/[Formula: see text]-N, [Formula: see text]-N, [Formula: see text], TN, TP, and TOC) between the two experimental groups. After a 4-month culture period, various indexes and expression of myogenic regulatory factor (MRFs) and their related genes were tested. The weight gain of the fish fed with artificial feed (AFG) was nearly 40% higher than the fish fed with grass (GFG). Significantly higher alkaline phosphatase, total cholestrol, high density cholestrol and total protein were detected in GFG as compared to AFG. GFG also showed increased hardness, resilience and shear force in texture profile analysis, with significantly bigger and compact muscle fibers in histologic slices. The fat accumulation was most serious in the abdomen muscle of AFG. Additionally, the expression levels of MyoG, MyoD, IGF-1, and MSTNs were higher, whereas Myf-5, MRF4, and IGF-2 were lower in most positional muscles of GFG as compared to AFG. Overall, these results suggested that feeding grass could promote muscle growth and development by stimulating muscle fiber hypertrophy, as well as significantly enhance the expression of CoL1As. Feeding C. idellus with grass could also improve flesh quality by improving muscle characteristics, enhancing the production of collagen, meanthile, reducing fat accumulation and moisture in muscle, but at the cost of a slower growth.

Effects of Salinity Treatment on Muscle Quality and Off‐flavour Compounds of Grass Carp ( Ctenopharyngodon Idella ) and Black Carp ( Mylopharyngodon Piceus )

Abstract:To investigate the effects of short-term salinity treatment on muscle quality and earthy and musty off-flavour compounds of freshwater fish, adult grass carp (1.25 +/- 0.25 kg) and black carp (1.50 +/- 0.30 kg) were reared at water salinities of 0 parts per thousand, 5 parts per thousand and 7.5 parts per thousand for 48 h. Muscle nutrients, amino acid, and fatty acid contents, texture properties, and earthy and musty off-flavour compounds were then determined. The content of total amino acid (TAA), Lys, Arg and essential amino acid index (EAAI) in the muscle of C7.5 group were elevated significantly compared with that in the control group (p < 0.05). The content of delicious amino acids (DAA) and EAAI in the muscle of M7.5 group was significantly higher than M0 and M5 groups (p < 0.05). The fatty acid content of the muscle was affected by salinity. In grass carp, the saturated fatty acid (SFA) content level was decreased significantly in the C5 and C7.5 groups (p < 0.05), while the monounsaturated fatty acid (MUFA) and polyunsaturated fatty acid (PUFA) content levels were increased significantly in the C5 and C7.5 groups (p < 0.05). The ratio of n3/n6 PUFA increased significantly in the C7.5 group (p < 0.05). The PUFA content and the n3/n6 PUFA ratio were increased significantly in the M7.5 group (p < 0.05). With increasing salinity, the relative content levels of volatile compounds causing earthy and musty off-flavour in muscle were decreased significantly in 5 parts per thousand and 7.5 parts per thousand salinity groups both grass carp and black carp (p < 0.05). The textural properties of muscle, such as hardness, springiness, gumminess and chewiness, were improved in the C7.5 and M5 groups. These results suggest that a short-term salinity treatment in freshwater fish prior to selling can effectively improve muscle quality and decrease the content of earthy and musty off-flavour compounds, with ideal salinities of 5 parts per thousand or 7.5 parts per thousand for grass carp and 7.5 parts per thousand for black carp.

Abstract:Rigor mortis occurs in a relatively early postmortem period and is a complex biochemical process in the conversion of muscle to meat. Understanding the quality changes and biomarkers during rigor mortis can provide a theoretical basis for maintaining and improving meat quality. Herein, a tandem mass tag proteomic method is used to investigate the effects of differentially expressed proteins on the meat quality of cattle Longissimus lumborum muscle postmortem (0, 6, and 24 h). The pH, total sulfhydryl content and sarcomere length decrease significantly during storage. In contrast, meat color values (L*, a*, and b*) and the myofibril fragmentation index increase significantly. Altogether, 147 differentially expressed proteins are identified, most being categorized as metabolic enzymes, mitochondrial proteins, necroptosis and ferroptosis proteins and structural proteins. The results also reveal additional proteins that are potentially involved in rigor mortis, such as cardiac phospholamban, acetyl-coenzyme A acyltransferase, and ankyrin repeat domain 2. The current results provide proteomic insights into the changes in meat quality during rigor mortis.

Abstract:To characterize the involvement of microorganisms in amino acid degradation and fish quality deterioration, three major grass carp spoilage bacteria were artificially inoculated in amino acid solutions (in-vitro) and grass carp flesh (in-situ). Results showed that Pseudomonas putida largely degraded free amino acids and produced 3.78 mM/100 g ammonia in grass carp flesh, relying on its high amino acid deamination1 activity. Aeromonas rivipollensis produced 3-Methyl-butanol and 2-Methyl-butanol through leucine and isoleucine degradation. Shewanella putrefaciens had potent ornithine-decarboxylation activity (423.91 × 10-9 µg/CFU) and released 22.98 mg/kg putrescine in situ. S. putrefaciens could produce more putrescine when cooperating with P. putida through the arginine deiminase pathway. To conclude, the biochemical activities identified through in-vitro tests correlated well with quality changes in inoculated grass carp flesh. The outcomes of this study provided fundamental information on the spoilage mechanisms of freshwater fish and important guidance for the development of quality control strategies.
Effects of Frozen Storage Condition Abuse on the Textural and Chemical Properties of Grass Carp (Ctenopharyngodon idella) Fillets

Abstract:Fish is highly perishable and frozen storage is normally used to maintain its quality. This study employed freezing-thawing (F-T) cycles to simulate the abuse in frozen storage conditions, and their effects on the quality changes of grass carp fillet were investigated. Results indicated that with the increase in F-T cycles, the hardness exhibited an increase during cycle 0 and cycle 1 and then decreased in the following two cycles. Measurement points had little influence on textural properties. Total volatile basic nitrogen (TVB-N) value showed an increase from 8.72 to 22.55mg N/100g during four F-T cycles, while thiobarbituric acid reactive substances (TBARS) exhibited gradual increase with the cycles and K value was also increased from 20.57 to 76.29% with the cycles. The results demonstrated that frozen storage condition abuse showed an important effect on textural properties and TVB-N, TBARS and K value of grass carp fillet. Practical ApplicationsInvestigations of fish and fish fillet freshness are much more significant in research and development for providing premium quality for the acceptance of consumers as well as international fishery trade. However, little work has been reported about the frozen storage condition abuse on textural and chemical properties of grass carp fillet. Therefore, in the current study, freezing-thawing cycles were used to simulate the temperature abuse during frozen storage, and their effects on the quality changes of grass carp fillet muscle based upon fish textural parameters were investigated. The results should provide practical guidance for the fish industry.
Mechanisms of chicken processing quality changes during the early postmortem time: the role of the changes in myofibrillar protein

Abstract:The properties of white meat, such as chicken and fish, changed quickly in the early postmortem period. This study focused on the relationship between myofibrillar protein and the eating quality of chicken in this period. The results showed that the eating quality of chicken, such as texture, water holding capacity and shear force value, decreased in the early postmortem period. The western blot analysis indicated that the desmin and troponin T were degraded significantly with postmortem time. Meanwhile, the myofibril protein degeneration in this period was evaluated by Fourier transform infrared and Differential Scanning Calorimetry. The findings demonstrated that the ordered structure of the myofibrillar protein was destroyed, and its thermal stability was reduced with the prolongation of postmortem time. Subsequently, the transmission electron microscope results suggested that the heating processing could destroy the connection between the weakened Z-line and the myofilament. Overall, the decline in thermal stability and degradation of myofibrillar may worsen the structural instability of chicken during heating, which may be a significant contributor to the variations of chicken quality in the early postmortem period.

Abstract:The micro-flowing water system can improve the flesh quality of freshwater fish using the traditional pond farming method. However, the mechanism of this phenomenon has not yet been explored. This study intends to examine the changes of metabolites in freshwater fish after treatment with the micro-flowing purification system (MFPS). The UPLC-QTOF/MS based metabolomics method was utilized to screen the metabolites and predict the major possible metabolic pathways after MFPS treatment. There were 377 types of metabolites identified in the fish muscle, of which 54-71 represented significant different metabolites identified during different stages of MFPS treatments. The main mechanism of MFPS treatment in improving the quality of grass carp fish muscle was investigated, and the MFPS treatment was shown to improve the flesh quality and the flavor of grass carp fish muscle. This study could provide the theoretical basis for improving the quality of aquatic products.

Abstract:Background: Among various cooking methods, steaming is favored by many because it can cause less damage to nutrient components in muscle, retain the inherent food flavor, and reduce the generation of harmful substances. Steaming conditions are closely related to fish flavor, fat and protein oxidation, and digestibility. Results: The black carp steamed for 4 to 14 min was studied in this article based on sensory assessment, electronic tongue, free amino acids, adenosine triphosphate (ATP)-related compounds, total nitrogen and non-protein nitrogen to explore the effect of steaming time on the taste substances and protein degradation of pickled black carp. The experimental result showed that the meat steamed within 8 min tasted better, showing high tastiness. The sensory assessment score increased significantly to the maximum value of 82.33 at 6 min. The content of umami and sweet amino acids increased significantly to the maximum value of 1.6801 g kg-1 at 6 min. In the meantime, the IMP (inosine monophosphate) content was 1.9128 g kg-1 , with its taste activity value (TAV) reaching 7.65, which proved that IMP affected the taste most. Furthermore, the total nitrogen content was 30.77 g kg-1 , which meant protein degraded a great deal. Based on equivalent umami concentration (EUC) and its TAV, the meat tasted best at 6-8 min. The longer the steaming time, the faster the protein degradation and the more the flavor precursors. Conclusion: The black carp pickled products (with a weight of 20 g, with the size of 3 cm × 3 cm × 2 cm) is suggested to be steamed for 6 to 8 min. This conclusion provides a theoretical basis for its better taste quality. © 2020 Society of Chemical Industry.

Abstract:Ultra-high pressure (UHP) and magnetic field (MF) are emerging food preservation technologies, but the research on the effects of combined treatment (UHP-MF) on aquatic products quality is insufficient. Hence, we explored the effects of UHP-MF treatment on the protein properties, microbial action and quality characteristics of stored grass carp fillets. Results showed that 300 MPa UHP treatment combined with MF storage (UHP300-MF) delayed the protein oxidation of stored fillets by inhibiting the accumulation of protein radical and the formation of lipid pro-oxidation products, thus the quality (texture, color) deterioration caused by oxidation was deferred. Additionally, 16S rDNA bacteria flora analysis revealed that UHP300-MF treatment inhibited the flora activity and diversity of fillets, delayed the protein degradation and quality deterioration caused by microbial action. Overall, UHP300-MF treatment suppressed the lipid and protein oxidation as well as microbial action, retarded the quality deterioration, and prolonged the shelf life of stored fillets.

Abstract:Mechanism on texture change of Wuchang bream (Megalobrama amblycephala) muscle during live transportation was explored by UPLC-QTOF-MS. The shear force (hardness) of fish muscle decreased gradually as the trans-portation time was extended from 3 to 24 h, with a maximum decrease of 43.49 %. With the extension of the time, the cellular and ultrastructure of fish muscle fiber were gradually disrupted. There was a significant dif-ference between metabolites of the samples after transporting for 6, 12, 24 h versus 3 h, with organic acids and their derivatives upregulated while nucleotide metabolites downregulated. The texture deterioration might be related to metabolism of amino acids, purine, histidine, and choline. The results showed that the deterioration of Wuchang bream texture during live transportation might be mainly related to the changes of the above metabolic pathways caused by oxidative stress, which resulting in the destruction of the muscle cells and fibers.

Abstract:The aim of this study was to investigate the effect of dietary protein level (soybean meal) on growth performance, flesh quality of grass carp, and the related molecular mechanisms. The results showed that appropriate dietary protein levels improved the growth performance, hardness, and pH of muscle while decreasing muscle crude lipid content and cooking loss and altering the antioxidant capacity and metabolic enzymes activities. In addition, appropriate dietary protein promoted the gene expression of myhc-1, myhc - 4, myf5, myod, myog , and fgf6a , whereas inhibited that of myhc-7, myhc - 2, mrf4 , and mstn . Transcriptome profiling of muscle revealed that the flesh quality-specific differences were related to tight junctions and intramuscular fat (IMF) accumulation. GSEA showed that fatty acid metabolism and oxidative phosphorylation were downregulated in SM5 compared with SM1. To conclude, appropriate protein levels improved the growth and flesh quality by regulating muscle antioxidant capacity and gene expression of myhc s and fat metabolism-related signaling molecules.

Influence of short-term starvation on the muscle quality of commercial-sized grass carp (Ctenopharyngodon idella)

Abstract:The effects of short-term starvation on muscle common nutrition component and texture index of grass carp (Ctenopharyngodon idella) were studied,in order to provide theoretical basis and reference data for improvement of the quality of grass carp in production practice.Ten common grass carp that reached the listing specifications and ten grass carp that starvation for 15 days were selected respectively in this research.The normal nutrition component,physic-chemical parameters,composition of fatty acid,muscle fibers diameter and the main texture iudex of grass carp muscle were determined.And according to the effect of short-term starvation on muscle common nutritional values,physic-chemical parameters and the main texture index,whether need to extend the time for starvation to 50 d was decided eventually.The results showed that there were no significant changes in water content,crude protein and crude fat content in grass carp after short-term(15 days)starvation,but the hardness,adhesiveness and chewiness decreased significantly,ash content,pH and conductivity increased significantly.In addition,short-term starvation significantly increased the content of PUFA(carbon twenty alkyl acid)in grass carp muscle.After short-term starvation the characteristic tasty amino acids (glutamic acid and asparagines) increased,and the characteristic sweet amino acids(alanine and glycin)decreased.The data starvation for 50 days showed that the water content and crude fat content of the grass carp decreased significantly in comparison with the control group and the hardness,chewiness and adhesiveness of the grass carp were significantly higher than those of the group starvation for 15 days,but the hardness and adhesiveness were still significantly lower than the control group,there was no significant difference of chewing,the ash content was significantly higher than the control group.Therefore,the effects of starvation for 50 days are better than shot-term starvation(15 d),when reduction of muscle fat content was one of the goals of starvation and the muscle fat content and the normal texture index of hardness,gumminess and chewiness were both considered.
