Abstract:The heterogeneous composition of fungi plays an indispensable role in the foundation of the multifunctionalities of ecosystems within drylands. The precise mechanisms that govern fluctuations in soil fungal assemblages in dryland ecosystems remain incompletely elucidated. In this study, biological soil crusts (biocrusts) at different successional stages in the Gurbantunggut Desert were used as substrates to examine the characteristics and driving factors that influence fungal abundance and community dynamics during biocrust development using qPCR and high-throughput sequencing of the ITS2 region. The findings showed that the physicochemical properties changed significantly with the development of biocrusts. In particular, total nitrogen increased 4.8 times, along with notable increases in ammonium, total phosphorus (2.1 times) and soil organic carbon (6.5 times). Initially, there was a rise in fungal abundance, which was subsequently followed by a decline as the biocrust developed, with the highest abundance detected in lichen crust (2.66 × 10 7 copies/g soil) and the lowest in bare sand (7.98 × 10 6 copies/g soil). Ascomycetes and Basidiomycetes emerged as dominant phyla, collectively forming 85% of the fungal community. As the biocrust developed, noticeable alterations occurred in fungal community compositions, resulting from changes in the relative proportions of Dothideomycetes, Lecanoromycetes and unclassified ascomycetes. Nitrogen, phosphorus, organic carbon content, and pH of biocrusts were identified as direct or indirect regulators of fungal abundance and community structure. The complexity of fungal networks increased as biocrusts developed as revealed by network analysis, but reduced in the stability of fungal communities within algal and lichen crusts. Keystone species within the fungal community also underwent changes as biocrust developed. These results suggested that shifts in interspecies relationships among fungi could further contribute to the variation in fungal communities during the development of biocrusts.

Consistent community assembly but contingent species pool effects drive β-diversity patterns of multiple microbial groups in desert biocrust systems

Abstract:One of the key goals of ecology is to understand how communities are assembled. The species co-existence theory suggests that community β-diversity is influenced by species pool and community assembly processes, such as environmental filtering, dispersal events, ecological drift and biotic interactions. However, it remains unclear whether there are similar β-diversity patterns among different soil microbial groups and whether all these mechanisms play significant roles in mediating β-diversity patterns. By conducting a broad survey across Chinese deserts, we aimed to address these questions by investing biological soil crusts (biocrusts). Through amplicon-sequencing, we acquired β-diversity data for multiple microbial groups, that is, soil total bacteria, diazotrophs, phoD-harbouring taxa, and fungi. Our results have shown varying distance decay rates of β-diversity across microbial groups, with soil total bacteria showing a weaker distance-decay relationship than other groups. The impact of the species pool on community β-diversity varied across microbial groups, with soil total bacteria and diazotrophs being significantly influenced. While the contributions of specific assembly processes to community β-diversity patterns varied among different microbial groups, significant effects of local community assembly processes on β-diversity patterns were consistently observed across all groups. Homogenous selection and dispersal limitation emerged as crucial processes for all groups. Precipitation and soil C:P were the key factors mediating β-diversity for all groups. This study has substantially advanced our understanding of how the communities of multiple microbial groups are structured in desert biocrust systems.
Water and Heat-Triggered Changes of Bacterial Community Structure and Function in Biological Soil Crusts

Abstract:Abstract Background and Aims :The outstanding ability of biological soil crusts (BSCs) in soil microenvironments regulation is mainly attribute to microorganisms that colonizing in biocrusts. We aimed to investigate the changes of bacterial community structure and function with biocrust succession, as well as their responses to climatic changes across large geographical scales. Methods : Algal BSCs and lichen BSCs were sampled along an aridity gradient on alpine grasslands. Bacterial communities in biocrusts were measured using high-throughput sequencing, and soil underlying biocrusts (0-5 cm) was collected for nutrients determination. Results : Our results indicated that compared with algal BSCs, bacterial community in lichen BSCs was characterized by lower diversity, more complex co-occurrence network and mutually beneficial relationships. The bacterial community assembly was governed mainly by stochastic processes for lichen BSCs, which was different from the almost equally important roles of stochastic and deterministic processes for algal BSCs. Geographical location had a significant effect on bacterial communities in both algal and lichen BSCs, while had a greater effect on lichen BSCs. It is noteworthy that the bacterial diversity of algal BSCs was positively correlated with aridity index, while that of lichens was negatively correlated with aridity index. Moreover, we determined lower soil pH and higher soil phosphorus content underlying lichen BSCs, implying their advantages in soil improvement. Conclusions : Aridity index was one of important driving factors of bacterial community in biocrusts, and its effects were biocrust type dependent. Lichen BSCs had greater effects on soil improvement than that of algal BSCs.
Network analysis reveals the strengthening of microbial interaction in biological soil crust development in the Mu Us Sandy Land, northwestern China

Abstract:Although studies examining changes in the structure and diversity of microbial communities in biological soil crusts (BSCs) have increased, microbial interactions in BSCs are not currently fully understood. In this study, we applied a random matrix theory (RMT)-based network approach to construct bacterial, fungal and bacteria-fungi interaction networks for the four developmental stages of BSCs (bare sand, physical crusts, algae crusts and moss crusts) in the Mu Us Sandy Land, northwestern China. Our results showed that Firmicutes, Proteobacteria, Cyanobacteria and Actinobacteria were the keystone phylum in bacterial networks in the four developmental stages, respectively. Ascomycota was the keystone phylum in the fungal networks in all stages, and Basidiomycota was a further keystone phylum in the algae and moss crust networks. With the development of BSCs, complexity in bacterial and fungal networks increased, becoming more clustered and connected, indicating a strengthened pattern in microbial interactions with BSC development. Positive links dominated in all networks, showing that microbial synergism was important in the development of BSCs. Interestingly, with the development of BSCs, the proportion of negative links in bacterial networks increased and they decreased in fungal networks, suggesting more inter-community competition was present in the bacterial communities and less inter-community competition was present in the fungal communities. Environmental factors, especially total carbon and total organic carbon, constrained most bacterial and fungal communities, respectively. Our findings provided a new insight into understanding BSC developmental mechanisms and ecological functions in a desert ecosystem composed of a sandy land landscape in arid and semi-arid regions.

Abstract:Biological soil crusts (biocrusts) are considered "desert ecosystem engineers" because they play a vital role in the restoration and stability maintenance of deserts, including those cold sandy land ecosystems at high latitudes, which are especially understudied. Microorganisms participate in the formation and succession of biocrusts, contributing to soil properties' improvement and the stability of soil aggregates, and thus vegetation development. Accordingly, understanding the composition and successional characteristics of microorganisms is a prerequisite for analyzing the ecological functions of biocrusts and related applications. Here, the Hulun Buir Sandy Land region in northeastern China—lying at the highest latitude of any sandy land in the country—was selected for study. Through a field investigation and next-generation sequencing (Illumina MiSeq PE300 Platform), our goal was to assess the shifts in diversity and community composition of soil bacteria and fungi across different stages during the succession of biocrusts in this region, and to uncover the main factors involved in shaping their soil microbial community. The results revealed that the nutrient enrichment capacity of biocrusts for available nitrogen, total nitrogen, total phosphorus, total content of water-soluble salt, available potassium, soil organic matter, and available phosphorus was progressively enhanced by the succession of cyanobacterial crusts to lichen crusts and then to moss crusts. In tandem, soil bacterial diversity increased as biocrust succession proceeded but fungal diversity decreased. A total of 32 bacterial phyla and 11 fungal phyla were identified, these also known to occur in other desert ecosystems. Among those taxa, the relative abundance of Proteobacteria and Cyanobacteria significantly increased and decreased, respectively, along the cyanobacterial crust–lichen–moss crust successional gradient. However, for Actinobacteria , Chloroflexi , and Acidobacteria their changed relative abundance was significantly hump-shaped, increasing in the shift from cyanobacterial crust to lichen crust, and then decreasing as lichen crust shifted to moss crust. In this process, the improved soil properties effectively enhanced soil bacterial and fungal community composition. Altogether, these findings broaden our understanding about how soil microbial properties can change during the succession of biocrusts in high-latitude, cold sandy land ecosystems.

Abstract:Biocrusts (biological soil crusts) are a living consortium of microbes that form a protective cover on many dryland soil and rock surfaces and provide important ecological functions in ecosystems. Characterising biocrust microbial communities is an important step in understanding their functional traits. Our aims were to explore the composition, diversity, and ecological function of the microbial community in two types of biocrusts in the Mu Us Sandland, China. High-throughput sequencing (Illumina MiSeq) and Functional Annotation of Prokaryotic Taxa (FAPROTAX) were used to analyze the soil bacterial and fungal communities in two types of biocrusts and their potential functions. In our study we showed that the richness and diversity of bacterial and fungal communities was significantly different between the lichen and moss biocrusts based on high-throughput sequencing. Dominant bacteria in lichen biocrusts (68.0%) were Actinomycetes, Cyanobacteria, and Proteobacteria, with 59.9% in moss biocrusts. In lichen crusts the dominant phylum of fungi was mainly Ascomycota. Microcoleus (11.1%) and Exophiala (7.4%) lichen crusts, while the dominant genus in moss crusts was RB41 (5.2%). The top three factors that were correlated with both bacterial and fungal community structures were pH, soil dissolved organic carbon, and soil organic carbon. Further analysis by FAPROTAX corroborated that the function of the bacterial community in two types of biocrusts was quite different. The relative abundances of metabolic functions in moss crusts, such as chemoheterotrophy, ureolysis, aromatic compound degradation, and nitrate reduction, were significantly higher than those in lichen crusts, however, the relative abundances of cyanobacteria, oxygenic photoautotrophy, photoautotrophy, and phototrophy were significantly lower (P < 0.05). Together, our results showed that the taxonomic and metabolic diversity, including specific species, have significant effects on the biocrusts types and their functional traits. This study provides a theoretical basis for elements of the soil microbiome restoration that may influence the establishment of plant communities.

Abstract:Abstract Soil microorganisms including many rare taxa and few abundant taxa, have different contributions to the ecosystem functions and services. High throughput sequencing technology was used to analyze the species composition of soil samples by DNA sequencing. Soil microorganisms were divided into abundant taxa and rare taxa to reveal their composition. Correlation analysis and random forest method were used to further analyze the influence of environmental factors on the community. Finally, the beta nearest taxon index (βNTI) based on the null model was used to reveal the mechanism of soil microbial community assembly. The findings indicate that in desert soils, the community assembly of rare bacteria is almost entirely dominated by a homogeneous selection of deterministic processes. For comparison, stochastic processes had more pronounced effects on the abundant bacteria. However, both abundant and rare fungi exhibited similar patterns of community assembly, that is, deterministic and stochastic processes almost jointly determined the assembly processes of fungal communities. We observed that community assembly shifted from stochastic to deterministic processes with increasing mean annual precipitation (MAP) and mean annual temperature (MAT) for abundant bacteria. Conversely, for rare fungi, there was an inclination towards a shift from deterministic to stochastic processes with rising MAT. In conclusion, our findings provide compelling evidence that MAT and MAP regulate the community assembly process of abundant and rare microbial communities in desert soil. These findings establish a theoretical foundation for future investigations into the community structure and ecological functions of soil microorganisms.

Abstract:Biological soil crusts (BSCs) are common and play critical roles in semi-arid and arid ecosystems. Bacteria, as an important community in BSCs, play critical roles in biochemical processes. However, how bacterial diversity and community change in different successional stages of BSCs is still unknown. We used 454 pyrosequencing of 16S rRNA to investigate the bacterial composition and community, and the relationships between bacterial composition and environmental factors were also explored. In different successional stages of BSCs, the number of bacteria operational taxonomic units (OTUs) detected in each sample ranged from 2572 to 3157. Proteobacteria, Cyanobacteria, Bacteroidetes were dominant in BSCs, followed by Firmicutes, Acidobacteria, and Actinobacteria. At the successional stages of BSCs, bacterial communities, OTU composition and their relative abundance notably differentiated, and Cyanobacteria, especially Microcoleus vaginatus, dominated algal crust and lichen crust, and were the main C-fixing bacteria in BSCs. Proteobacteria and Bacteroidetes increased with the development of BSCs. OTUs related to Planomicrobium Chinese, Desulfobulbus sp., Desulfomicrobium sp., Arthrobacter sp., and Ahhaerbacter sp. showed higher relative abundance in bare sand than other successional stages of BSCs, while relative abundance of Sphingomonas sp. Niastella sp., Pedobacter, Candidatus solobacter, and Streptophyta increased with the development of BSCs. In successional stages of BSCs, bacterial OTUs composition demonstrated strong correlations with soil nutrients, soil salts, and soil enzymes. Additionally, variation of bacterial composition led to different ecological function. In bare sand, some species were related with mineral metabolism or promoting plant growth, and in algal crust and lichen crust, C-fixing bacteria increased and accumulated C to the desert soil. In later developed stage of BSCs, bacteria related with decomposition of organic matter, such as Sphingomonas sp. Niastella sp., Pedobacter, and Candidatus solobacter increased. Therefore, bacterial community composition and their key ecological roles shifted to the development of BSCs.

Abstract:The ecological function of biological crusts in arid and semi-arid areas is of great importance. Bacteria, as a crucial microbial group in biological crusts, play a key role in the formation, nutrient cycling, and regulation of these crusts. However, the succession of biological crusts and the diversity of bacterial communities, along with key environmental factors in the Loess Plateau's hilly and gully areas, remain unclear. This study investigated soil bacterial abundance and diversity in bare soil (BS), alga-lichen mixed crust (MC), and alga-lichen mixed crust subsoil (MCS) using high-throughput sequencing methods. It explored the relationship between the bacterial community in biological crusts and key environmental factors. The results indicated that the Chao1, Shannon index, and phylogenetic diversity of bacteria significantly increased with the succession of biological crusts. There were notable differences in the community composition and structure of bacteria at different stages of crust development, with Rubrobacteria and Cyanobacteriia dominating in MCS. Effective phosphorus, available potassium, nitrogen, pH, and total organic carbon were identified as key environmental factors affecting soil bacterial communities. In summary, the succession of biological crusts alters soil physicochemical characteristics and creates different ecological niches for bacterial communities. Soil nutrients and pH play a crucial role in the selection of bacterial species and the shaping of bacterial communities in the Loess Plateau's hilly and gully areas.

Eukaryotic algae, as the primary group of photosynthetic autotrophs, exert a significant influence on the development and functions of biological crusts in dryland ecosystems. Despite their importance, there are substantial knowledge gaps on the composition of eukaryotic algal communities and their effects on the distributions of bacteria and fungi in dryland soils. This study examined the eukaryotic algal community along a successional sequence of biocrusts in the Gurbantunggut desert, while also investigating their patterns of co‐occurrence with bacteria and fungi through high‐throughput sequencing and bioinformatic analyses. The results showed that nitrogen and phosphorus levels played a crucial role in the regulation of changes in the abundance and composition of the algal community. In particular, changes in the structure of the algal community arise primarily from fluctuations in the main species, rather than from loss and appearance of species during the biocrust succession. The accumulation of nitrogen and phosphorus in the biocrust was associated with the decrease in the relative abundance of Chlorophyta. The results also indicated that eukaryotic algae played an important role in the affecting bacterial and fungal communities and significantly improved the stability of the microbial community, reflected by the robustness of co‐occurrence networks. The network analysis further indicated that eukaryotic algae affected the stability of microbial co‐occurrence networks either by acting as keystone taxa or associating with the keystone bacterial and fungal taxa. These findings reveal a clear mechanism by which soil nitrogen and phosphorus levels affected the composition of eukaryotic algae communities and further regulated bacterial and fungal communities during biocrust development, providing valuable information on the development and functional execution of biocrusts in dryland ecosystems. Abstract:Introduction: Biodiversity maintenance and its underlying mechanisms are central issues of ecology. However, predicting the composition turnovers of microbial communities at multiple spatial scales remains greatly challenging because they are obscured by the inconsistent impacts of climatic and local edaphic conditions on the assembly process. Methods: Based on the Illumina MeSeq 16S/18S rRNA sequencing technology, we investigated soil bacterial and eukaryotic communities in biocrusts with different successional levels at a subcontinental scale of Northern China. Results: Results showed that irrespective of spatial scale, bacterial α diversity increased but eukaryotic diversity decreased with the primary succession, whereas both β diversities decreased at the subcontinental scale compared with smaller scales, indicating that the biogeographic pattern of soil microorganisms was balanced by successional convergence and distance decay effect. We found that the convergence of bacterial and eukaryotic communities was attributed to the turnovers of generalist and specialist species, respectively. In this process, edaphic and climatic factors showed unique roles in the changes of diversity at local/subcontinental scales. Moreover, the taxonomic diversity tended to be more susceptible to climatic and edaphic conditions, while biotic factors (photosynthesis and pigments) were more important to phylogenetic diversity. Conclusion: Taken together, our study provided comprehensive insights into understanding the pattern of microbial diversity at multiple spatial scales of drylands.

Abstract:AimsCyanobacterial and algal communities have a large effect on biocrust formation and development. Biocrust species and abundance vary spatially and temporally due to different environmental factors. The relationships among cyanobacterial and algal communities, and biocrust function have been studied extensively. Lacking, however, are studies of temporal changes in a similar landscape where environmental conditions are similar, but where biocrust formation is different.MethodsBiocrusts of different ages were located in the Loess Plateau in an area that had experienced a landslide. We examined changes in cyanobacterial and algal communities, carbon, nutrients, and the composition of dissolved organic matter in the topsoil, and the relationships among the community and soil characteristics using redundancy analysis.ResultsPhormidium tenue (Cyanophyta) dominated in all biocrusts, and co-dominated in a newly formed crust with Euglena sp. (Euglenophyta). Oscillatoria sp. (Cyanophyta) increased with biocrust age. Oscillatoria was positively correlated with carbon fixation and nutrient (nitrogen and phosphorus) accumulation in topsoils.ConclusionsWhile incubation of Phormidium tenue and Euglena sp. is suggested for rapid biocrust formation at an early stage, increasing the abundance of Phormidium tenue and Oscillatoria sp. can promote carbon fixation and nutrient (nitrogen and phosphorus) accumulation, thereby accelerating biocrusts into a later stage.

Abstract:Introduction: Biological soil crusts (BSCs) constitute a substantial portion of primary production in dryland ecosystems. They successionally mature to deliver a series of ecosystem services. Bacteria, as an important community in BSCs, play critical roles in maintaining the structure and functions of BSCs. However, the process by which bacterial diversity and community are altered with BSC development is not fully understood. Methods: In this study, amplicons sequencing was used to investigate bacterial diversity and community compositions across five developmental stages of BSCs (bare sand, microbial crusts, algae crusts, lichen crusts, and moss crusts) and their relationship with environmental variables in the Gonghe basin sandy land in Qinghai-Tibet Plateau, northwestern China. Results: The results showed that Proteobacteria, Actinobacteria, Cyanobacteria, Acidobacteria, Bacteroidetes, and Firmicutes were predominant in different developmental stages of BSCs, accounting for more than 77% of the total relative abundance. The phyla of Acidobacteria and Bacteroidetes were abundant in this region. With BSC development, bacterial diversity significantly increased, and the taxonomic community composition significantly altered. The relative abundance of copiotrophic bacteria, such as Actinobacteria, Acidobacteria, Bacteroidetes, Verrucomicrobia, Planctomycetes, and Gemmatimonadetes significantly increased, whereas the relative abundance of oligotrophic bacteria, such as Proteobacteria and Firmicutes significantly decreased. The relative abundance of Cyanobacteria in the algae crusts was significantly higher than that in the other developmental stages ( p < 0.05). Conclusion: Variations in bacterial composition suggested that the potential ecological functions of the bacterial community were altered with BSC development. The functions varied from enhancing soil surface stability by promoting soil particle cementation in the early stages to promoting material circulation of the ecosystem by fixing carbon and nitrogen and decomposing litter in the later stages of BSC development. Bacterial community is a sensitive index of water and nutrient alterations during BSC development. Abstract:Recent studies have shown distinct soil microbial assembly patterns across taxonomic types, habitat types and regions, but little is known about which factors play a dominant role in soil microbial communities. To bridge this gap, we compared the differences in microbial diversity and community composition across two taxonomic types (prokaryotes and fungi), two habitat types (Artemisia and Poaceae) and three geographic regions in the arid ecosystem of northwest China. To determine the main driving factors shaping the prokaryotic and fungal community assembly, we carried out diverse analyses including null model, partial mantel test and variance partitioning analysis etc. The findings suggested that the processes of community assembly were more diverse among taxonomic categories in comparison to habitats or geographical regions. The predominant driving factor of soil microbial community assembly in arid ecosystem was biotic interactions between microorganisms, followed by environmental filtering and dispersal limitation. Network vertex, positive cohesion and negative cohesion showed the most significant correlations with prokaryotic and fungal diversity and community dissimilarity. Salinity was the major environmental variable structuring the prokaryotic community. Although prokaryotic and fungal communities were jointly regulated by the three factors, the effects of biotic interactions and environmental variables (both are deterministic processes) on the community structure of prokaryotes were stronger than that of fungi. The null model revealed that prokaryotic community assembly was more deterministic, whereas fungal community assembly was structured by stochastic processes. Taken together, these findings unravel the predominant drivers governing microbial community assembly across taxonomic types, habitat types and geographic regions and highlight the impacts of biotic interactions on disentangling soil microbial assembly mechanisms.

Abstract:Biological soil crusts (biocrusts) cover about 12% of the Earth’s land masses, thereby providing ecosystem services and affecting biogeochemical fluxes on a global scale. They comprise photoautotrophic cyanobacteria, algae, lichens and mosses, which grow together with heterotrophic microorganisms, forming a model system to study facilitative interactions and assembly principles in natural communities. Biocrusts can be classified into cyanobacteria-, lichen-, and bryophyte-dominated types, which reflect stages of ecological succession. In this study, we examined whether these categories include a shift in heterotrophic communities and whether this may be linked to altered physiological properties. We analyzed the microbial community composition by means of qPCR and high-throughput amplicon sequencing and utilized flux measurements to investigate their physiological properties. Our results revealed that once 16S and 18S rRNA gene copy numbers increase, fungi become more predominant and alpha diversity increases with progressing succession. Bacterial communities differed significantly between biocrust types with a shift from more generalized to specialized organisms along succession. CO2 gas exchange measurements revealed large respiration rates of late successional crusts being significantly higher than those of initial biocrusts, and different successional stages showed distinct NO and HONO emission patterns. Thus, our study suggests that the photoautotrophic organisms facilitate specific microbial communities, which themselves strongly influence the overall physiological properties of biocrusts and hence local to global nutrient cycles.
Bacterial contributions of bio-crusts and litter crusts to nutrient cycling in the Mu Us Sandy Land

Abstract:Desertification has become an important issue for the sustainable development of human society at global scale and has led to the changes in soil properties and vegetation cover. Biocrusts and litter crusts play roles in improving the soil microhabitat of sandy ecosystems. Soil microbial communities mediate ecosystem functions in various ecosystems, e.g., soil biogeochemical processes. However, limited information is available about how the underlying processes of bio-crusts/litter crusts restoration are driven by soil bacterial communities in sandy land. Here, we investigated the changes in soil bacteria from three groups (sandy land, bio-crusts, and litter crusts) and three soil layers (0–2 cm, 2–5 cm, 5–10 cm) with nine replicates each collected in July 2019 utilized high-throughput pyrosequencing of the V4-V5 rRNA gene region. Most soil nutrients (SOM, AP, AK, and TN) and enzyme activities (BG and DHA) had differences among the three groups and three soil layers. OTU richness and diversity of bacteria were positively correlated with most soil variables. The constructed co-occurrence networks between soil variables and bacterial communities, and within bacterial communities showed that bacterial taxa had closer relationships with all soil variables in crusts than sandy land and varied among the three sand groups (sandy land, bio-crusts, and litter crusts). The result showed that the composition of bacterial community was regulated mainly by soil variables and crust types. Compared with sandy land, more predictors in nutrient cycling were found in crust types. They played major roles in nutrient cycling in desert ecosystem restoration on the basis of random forest modeling. Our findings indicate some bacterial taxa may played the predominant roles in connecting with soil variables and other bacterial taxa across crusts types, and litter crusts and bio-crusts drive the nutrient cycling by mediating the restoration of bacterial taxa in sandy ecosystems.

Abstract:Biocrust communities are often heterogeneous and affected by small-scale environmental features, including both physical and biotic factors. The presence of moss in biocrusts, for example, is likely to create a set of physical conditions distinct from those found in microbial biocrusts that lack mosses. Similarly, hypolithic organisms living under and on the belowground surface of translucent and opaque stones (typically quartz) experience a distinct environment relative to surrounding soil surface organisms. To understand the impact of these factors on biocrust bacterial community composition, we performed 16S rRNA sequencing from surface and hypolithic biocrusts with and without the common biocrust moss, Syntrichia caninervis. While alpha diversity indices did not differ significantly between any of the biocrust habitats sampled, we did observe differences in overall community composition. Cyanobacterial abundance and diversity decreased in the presence of moss and in surface samples compared to hypolithic biocrusts, while Proteobacteria showed the opposite pattern. Acidobacteria were significantly more abundant in hypolithic niches, and Patescibacteria were found to be restricted to moss-dominated surface biocrusts. Notably, bacterial community composition was found to shift significantly between surface and hypolithic microbial biocrusts (Adonis, R2 = 0.122, p = 0.002) and between surface moss and microbial biocrusts (R2 = 0.107, p = 0.002). These findings support the idea that even at small spatial scales (e.g., within <10 cm), desert biocrust bacterial community composition varies based on the habitat structure and cohabitants.

Abstract:Introduction: Mossy biocrust represents a stable stage in the succession of biological soil crust in arid and semi-arid areas, providing a microhabitat that maintains microbial diversity. However, the impact of mossy biocrust rhizoid soil and different particle sizes within the mossy biocrust layer and sublayer on microbial diversity and soil enzyme activities remains unclear. Methods: This study utilized Illumina MiSeq sequencing and high-throughput fluorometric technique to assess the differences in microbial diversity and soil extracellular enzymes between mossy biocrust rhizoid soil and different particle sizes within the mossy biocrust sifting and sublayer soil. Results: The results revealed that the total organic carbon (TOC), total nitrogen (TN), ammonium (NH 4 + ) and nitrate (NO 3 − ) in mossy biocrust rhizoid soil were the highest, with significantly higher TOC, TN, and total phosphorus (TP) in mossy biocrust sifting soil than those in mossy biocrust sublayer soil. Extracellular enzyme activities (EAAs) exhibited different responses to various soil particle sizes in mossy biocrust. Biocrust rhizoid soil (BRS) showed higher C-degrading enzyme activity and lower P-degrading enzyme activity, leading to a significant increase in enzyme C: P and N: P ratios. Mossy biocrust soils were all limited by microbial relative nitrogen while pronounced relative nitrogen limitation and microbial maximum relative carbon limitation in BRS. The diversity and richness of the bacterial community in the 0.2 mm mossy biocrust soil (BSS 0.2 ) were notably lower than those in mossy biocrust sublayer, whereas the diversity and richness of the fungal community in the rhizoid soil were significantly higher than those in mossy biocrust sublayer. The predominant bacterial phyla in mossy biocrust were Actinobacteriota, Protebacteria, Chloroflexi, and Acidobacteriota, whereas in BSS 0.2 , the predominant bacterial phyla were Actinobacteriota, Protebacteria, and Cyanobacteria. Ascomycota and Basidiomycota were dominant phyla in mossy biocrust. The bacterial and fungal community species composition exhibited significant differences. The mean proportions of Actinobacteriota, Protebacteria, Chloroflexi, Acidobacteriota, Acidobacteria, Cyanobacteria, and Bacteroidota varied significantly between mossy biocrust rhizoid and different particle sizes of mossy biocrust sifting and sublayer soil ( p < 0.05). Similarly, significant differences ( p < 0.05) were observed in the mean proportions of Ascomycota, Basidiomycota, and Glomeromycota between mossy biocrust rhizoid and different particle sizes within the mossy biocrust sifting and sublayer soil. The complexity and connectivity of bacterial and fungal networks were higher in mossy biocrust rhizoid soil compared with different particle sizes within the mossy biocrust sifting and sublayer soil. Discussion: These results offer valuable insights to enhance our understanding of the involvement of mossy biocrust in the biogeochemical cycle of desert ecosystems.

Abstract:Biological soil crust can effectively prevent wind erosion of sand and soil, which is of great significance to ecological restoration in desert areas. There have been few studies that have screened different planting patterns by analyzing biological crust properties. In this study, high-throughput sequencing was used to analyze the bacterial 16S rRNA community structure of biologically crusted soil in sandy bare land (CK), planting corn soil crust (CB), planting potato soil crust (TB) and planting alfalfa soil crust (AB), and nutrient profiles of each soil type were also analyzed. The results showed that soil nutrient content was significantly increased by the biological layer, and the organic matter, available phosphorus, and available potassium contents were significantly increased. In terms of bacterial communities, the TB treatment had more endemic genera (100). The top 10 phyla in terms of relative abundance accounted for around 95% of the total community, with Actinobacteria being the highest. The community structure of CB and AB was similar, and the diversity and richness indices of CB were significantly increased compared to those of CK. The available phosphorus and total nitrogen were found to be the main factors affecting bacterial community structure. The dominant role of Actinobacteria was mainly determined by pH. Overall, the results show that corn planting could be most helpful in the early restoration period of sandy land.
