Abstract:Bacterial cellulose is a bacterially derived polymer with great potential for application in wound healing due to its innate properties such as high biocompatibility and biodegradability. In addition to this, it is naturally biosynthesized by bacteria as a hydrogel, which makes it an optimal substrate for the treatment of dry wounds, where additional moisture is required to facilitate the healing process. However, this polymer lacks antibacterial properties. As bacterial infections are becoming increasingly common and difficult to treat due to antimicrobial resistance, it is of crucial importance to develop strategies for the modification of cellulose to ensure protection against bacterial contamination. In this study, a green-chemistry approach was proposed for the functionalization of cellulose to introduce antibacterial functional groups. Two different active agents, namely glycidyl trimethylammonium chloride and glycidyl hexadecyl ether, were used for the covalent derivatization of the hydroxyl groups of glucose through a heterogeneous reaction in basic aqueous conditions. The modified material was chemically and mechanically characterized by solid-state techniques and rheological measurements. A biological assessment was then carried out both using bacterial cells and human keratinocytes. It was observed that the functionalization performed induced a reduction of approximately half of the bacterial population within 24 h of direct contact withStaphylococcus aureussubsp. aureus Rosenbach 6538P(TM)andEscherichia coli(Migula) Castellani and Chalmers ATCC(R)8739(TM)(respectively, a reduction of 53% and 43% in the cell number was registered for the two strains). In parallel, cytotoxicity studies performed on keratinocytes (HaCaT cell line) showed cell viability in the range of 90 to 100% for up to 6 days of direct contact with both unmodified and modified samples. The morphology of the cells was also visually evaluated, and no significant difference was noted as compared to the control. Finally, thein vitroscratch assay evidenced good wound closure rates in the presence of the samples, with complete coverage of the scratched area after 5 days for both the modified cellulose and the positive control (i.e., keratinocytes growth medium). Overall, the modified hydrogel showed promising features, confirming its potential as an alternative substrate to develop a sustainable, antibacterial and biocompatible wound dressing.
Covalently grafting polycation to bacterial cellulose for antibacterial and anti-cell adhesive wound dressings

Abstract:Hydrogel-based wound dressings are becoming increasingly important for wound healing. Bacterial cellulose (BC) has been commonly used as wound dressings due to its good in vitro and in vivo biocompatibility. However, pure BC does not possess antibacterial properties. In this regard, polycation gel was grafted onto the BC using a surface-initiated activator regenerated by electron transfer atom transfer radical polymerization (SI-ARGET ATRP) with subsequent quaternization for antibacterial wound dressing. Dimethylethyl methacrylate (DMAEMA) was successfully polymerized on the BC surface which was confirmed by Fourier transform infrared spectroscopy and elemental analysis. The morphology structure, specific surface area, pore size, and mechanical properties were also characterized. The quaternized PDMAEMA grafted on the BC endowed it with excellent antibacterial activity against E. coli (Gram-negative) and S. aureus (Gram-positive) with a killing rate of 89.2 % and 93.4 %, respectively. The number of cells was significantly reduced on QPD/BC hydrogel, demonstrating its good anti-adhesion ability. In vitro cellular evaluation revealed that the antibacterial wound dressing exhibited good biocompatibility. Overall, this study provides a feasible method to develop antibacterial and anti-cell adhesive hydrogel, which has a promising potential for wound healing.
Barnacle-inspired and polyphenol-assisted modification of bacterial cellulose-based wound dressings for promoting infectious wound healing

Abstract:Bacterial cellulose (BC) is an ideal candidate for wound dressings due to its natural origin, exceptional water-holding capacity, pliability, biocompatibility, and high absorption capability. However, the lack of essential antimicrobial activity limits its biomedical applications. This study reported BC-based wound dressings containing silk fibroin protein (SF), offering the potential for biomimetic properties, and (-)-epigallocatechin-3-gallate (EGCG) for polyphenol-assisted surface modification to promote infectious wound healing. Glycerol was used as the carbon source to promote the formation of an adhesive layer by facilitating the β-sheet folding of SF, and different concentrations of EGCG were employed to interact with SF through strong hydrogen bonding facilitated by the polyphenolic groups. The functionalized membrane exhibited outstanding water-holding capacity, swelling ratio, and degradation properties, along with enhanced hydrophilicity, adhesiveness, and a remarkable free radical scavenging ability. Both in vitro and in vivo experiments confirmed its potent bacteriostatic activity. The composite membrane displayed excellent biocompatibility, including cellular and hemocompatibility. Importantly, it effectively promoted wound healing in murine back infections. These findings suggest the significant feasibility of the innovative modification approach, and that functionalized membranes have great potential as wound-dressing materials for infection management in future clinical applications.

Abstract:Bacterial cellulose (BC) is a promising biomaterial that has been widely used for wound healing. However, the lack of antimicrobial properties limits its biological applications. Herein, a BC-based wound dressing with a biological antimicrobial agent was developed. The process involved in situ cultivation of BC by adding carboxymethyl cellulose (CMC), subsequently it was combined with cationic antibacterial agent ε -poly- L -lysine (PLL) through covalent bonding with CMC. The prepared hydrogel showed a nano-porous structure, proper mechanical properties (120 kPa) and desired swelling capacity (over 1000%). It also exhibited superior antibacterial activity against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ) bacteria. From in vitro biological experiments, BC/CMC/PLL presented good biocompatibility containing cell proliferation and cell spreading. The resulting BC/CMC/PLL will be one of the competitive wound dressings for preventing infection.

Abstract:Abstract Bacterial cellulose is an extracellular polysaccharide produced by microorganisms, offering advantages such as high water‐holding capacity, flexibility, and biocompatibility. However, its lack of bactericidal activity hampers its wide application. Usnic acid, a secondary metabolite derived from lichens of the Usnea genus, is recognized for its antibacterial and anti‐biofilm efficiency, coupled with anti‐inflammatory properties. Its water insolubility presents challenges for wide utilization and stable release. Sanxan gel, a novel polysaccharide, exhibits exceptional freeze‐thaw stability, suspension properties, and high elasticity, rendering it effective as a suspending agent to improve the bioavailability of water‐insoluble drugs. In this study, a hydrogel membrane was designed by combining bacterial cellulose and usnic acid suspended in sanxan gel through a simple in situ microorganism fermentation. The obtained membranes demonstrated excellent ability for sustained drug release, strong eradication capability against tested bacteria in both in vitro and in vivo experiments, effective inhibition of biofilm formation, and excellent hemocompatibility and cytocompatibility. Additionally, the composite membranes promoted wound healing with reduced inflammation and bacterial infection in a full‐thickness wound infection model in mice. This study provides innovative insights and strategies for the development of functional dressings for infected wounds in future clinical applications. This article is protected by copyright. Abstract:Bacterial cellulose (BC) is a promising material for use as an artificial skin in wound healing application, however, its applications are limited due to its poor malleability. Incorporating non-cellulosic polysaccharides such as dextran and xyloglucan (XG) may enhance its respective wound healing and malleability. This study presents a novel in situ biopreparation method to produce BC-based hybrid hydrogels containing dextran (BC-D) and xyloglucan-dextran (BC-XG-D) with unique mechanical and rheological properties. Structural analysis revealed that dextran of different sizes (10 k, 70 k and 2 M of Mw) form micron-sized particles by loosely binding to cellulosic fibres. The addition of xyloglucan resulted acts as a lubricant in mechanical testing. The BC-XG-D hybrid hydrogels showed a reduced Young's modulus of 4 MPa and a higher maximum tensile strain of 53 % compared to native BC. Moreover, they displayed less plastic but more viscous behaviour under large shear strain deformation. The wound healing animal model experiments demonstrated that the BC-XG-D hybrid hydrogels promoted wound healing process and skin maturation. Overall, these findings contribute to the development of functional BC-based medical materials with desired mechanical and rheological properties that have the potential to accelerate wound healing.

Abstract:Bacterial cellulose (BC) holds several unique properties such as high water retention capability, flexibility, biocompatibility, and high absorption capacity. All these features make it a potential material for wound healing applications. However, it lacks antibacterial properties, which hampers its applications for infectious wound healings. This study reported BC-based dressings containing ε-polylysine (ε-PL), cross-linked by a biocompatible and mussel-inspired polydopamine (PDA) for promoting infectious wound healing. BC membranes were coated with PDA by a simple self-polymerization process, followed by treating with different contents of ε-PL. The resulted membranes showed strong antibacterial properties against tested bacteria by both in vitro and in vivo evaluations. The membranes also exhibited hemocompatibility and cytocompatibility by in vitro investigations. Moreover, the functionalized membranes promoted infected wound healing using Sprague–Dawley rats as a model animal. A complete wound healing was observed in the group treated with functionalized membranes, while wounds were still open for control and pure BC groups in the same duration. Histological investigations indicated that the thickness of newborn skin was greater and smoother in the groups treated with modified membranes in comparison to neat BC or control groups. Abstract:Ideal wound dressing materials should be active components in the healing process. Bacterial cellulose (BC) has attracted great attention as a novel wound dressing material, however, BC itself has no antimicrobial activity. To explore the practical application values of BC and develop novel wound dressing materials, a series of composite membranes (BC/PILs) based on BC and polymeric ionic liquids (PILs, composed of choline and different amino acids) with antimicrobial activity were synthesized by ex-situ method. The physicochemical, antimicrobial properties and biocompatibility of these membranes were systematically investigated. The results indicated that BC/PILs membranes with excellent properties could be obtained by adjusting the concentration and type of PILs. Several kinds of BC/PILs membranes exhibited good biocompatibility and high antimicrobial activity against Gram-positive, Gram-negative bacteria and fungus. The anionic backbone of PILs played important roles in the antimicrobial activity of BC/PILs membranes. The obtained membranes provided a very promising candidate for wound dressing materials.

Abstract:In order to promote wound healing, self-healing hydrogels with moisturizing property are employed as wound dressing. In this study, bacterial cellulose nanofibers (BCN) with high mechanical strength are used as reinforcement to improve the mechanical properties of self-healing hydrogels. A multifunctional self-healing hydrogel has been constructed by incorporating natural biomass, including Ag hybrid bacterial cellulose nanofiber (Ag-BCN), resveratrol (Res), and carbon nanodots (CNDs). The results of in vitro experiments demonstrate that the mechanical strength of the hybrid hydrogel was increased by 6 times with the addition of Ag-BCN, which also offers excellent antibacterial efficiency ( S. aureus 99.99 % and E. coli 99.68 %). The hydrogel with CNDs can observe the healing process of the crack in real time and realize the controlled release of Res through photothermal effect. Moreover, the results of animal model experiments indicate that the prepared hydrogel could reduce the infection of the wound, effectively shorten the progress of wound healing (from 21d to 14 d). All the results imply that the prepared hydrogel has great promise in the application of skin wound healing.

Abstract:Bacterial cellulose (BC) with excellent flexibility and biocompatibility has aroused great attention for wound healing. However, the surface inertness of BC nanofibers tremendously hindered its biofunctionalized to improve the antibacterial ability and cytocompatibility. Herein, the biofunctionalized BC was fabricated by in situ growing a uniformly polydopamine (PDA)/polyethyleneimine (PEI) coating and grafting bacitracin (BA) on the coating (BA-PPBC). The results confirmed that the BA-grafted PDA/PEI coating was distributed uniformly on the surface of BC nanofibers, and the natural three-dimensional pore structure and other physical properties of pristine BC were retained. Compared to BC, the obtained BA-PPBC materials exhibited excellent antibacterial activities against the adhesion and colonization of S. aureus and MRSA . Moreover, the BA-PPBC not only had no obvious cytotoxicity to NIH-3T3 cells, but also improved the adhesion, proliferation, and spread out of cells. This work provides a biofunctionalized BC dressing to simultaneously meet the requirements of wound healing.

Abstract:This study aims to modify bacterial cellulose (BC) produced by Komagataeibacter nataicola TISTR 975 with collagen hydrolysate (CH) and Quercus infectoria gall (QI) via an ex-situ method to develop a bioactive wound dressing. Initially, QI gall was extracted in ethanol, and the crude extract showed a high total phenolic content (TPC) of 778.7 ± 76.2 mg GAE/g. After loading CH and QI onto the BC film, the morphology, FTIR spectra, mechanical properties, antibacterial properties, and biocompatibility of the prepared films were systematically evaluated. SEM images revealed that all prepared films were porous with multiple layers, and FTIR results confirmed the successful incorporation of CH and QI into the BC film. The TPC on the films ranged from 149.7 to 563.1 mg GAE/g, depending on the QI loading concentration. The tensile strength of the BC/CH/QI films was higher, while Young's modulus and % elongation at break were comparable to the BC. The swelling ratio of the composite films was increased to nearly doubled, which can be attributed to the high-water absorption capacity of CH. Moreover, disk agar tests revealed that adding QI in the BC film significantly inhibited the growth of Escherichia coli and Staphylococcus aureus . Hemolysis assay results supported that the BC/CH/QI films were biocompatible. Overall, the results of this study demonstrate that the BC/CH/QI films are considered as a bioactive material and has the great potential for biomedical applications.

Abstract:Bacterial cellulose (BC) is produced by several microorganisms as extracellular structures and can be modified by various physicochemical and biological strategies to produce different cellulosic formats. The main advantages of BC for biomedical applications can be summarized thus: easy moldability, purification, and scalability; high biocompatibility; and straightforward tailoring. The presence of a high amount of free hydroxyl residues, linked with water and nanoporous morphology, makes BC polymer an ideal candidate for wound healing. In this frame, acute and chronic wounds, associated with prevalent pathologies, were addressed to find adequate therapeutic strategies. Hence, the main characteristics of different BC structures-such as membranes and films, fibrous and spheroidal, nanocrystals and nanofibers, and different BC blends, as well as recent advances in BC composites with alginate, collagen, chitosan, silk sericin, and some miscellaneous blends-are reported in detail. Moreover, the development of novel antimicrobial BC and drug delivery systems are discussed.

Abstract:Due to their biodegradability and biocompatibility, chitosan-based hydrogels have great potential in regenerative medicine, with applications such as bacteriostasis, hemostasis, and wound healing. However, toxicity and high cost are problems that must be solved for chitosan-based hydrogel crosslinking agents such as formaldehyde, glutaraldehyde, and genipin. Therefore, we developed a biocompatible yet cost-effective chitosan-based hydrogel system as a candidate biomaterial to prevent infection during wound healing. The hydrogel was fabricated by crosslinking chitosan with dialdehyde chitosan (CTS–CHO) via dynamic Schiff-base reactions, resulting in a self-healable and injectable system. The rheological properties, degradation profile, and self-healable properties of the chitosan-based hydrogel were evaluated. The excellent antibacterial activity of the hydrogel was validated by a spread plate experiment. The use of Live/Dead assay on HEK 293 cells showed that the hydrogel exhibited excellent biocompatibility. The results demonstrate that the newly designed chitosan-based hydrogel is an excellent antibacterial wound dressing candidate with good biocompatibility.
Fungal Carboxymethyl Chitosan-Impregnated Bacterial Cellulose Hydrogel as Wound-Dressing Agent

Abstract:Bacterial cellulose (BC) produced by Gluconoacetobacter hansenii is a suitable polymeric fiber network for wound-dressing purposes, but its lack of antibacterial properties limits it from healing bacterial wounds. We developed hydrogels by impregnating fungal-derived carboxymethyl chitosan to BC fiber networks using a simple solution immersion method. The CMCS–BC hydrogels were characterized using various characterization techniques such as XRD, FTIR, water contact angle measurements, TGA, and SEM to know the physiochemical properties. The results show that the impregnation of CMCS into BC fiber networks greatly influences BC's improving hydrophilic nature, which is crucial for wound healing applications. Furthermore, the CMCS–BC hydrogels were studied for biocompatibility analysis with skin fibroblast cells. The results revealed that by increasing the CMCS content in the BC, biocompatibility, cell attachment, and spreading capacity also increase. The antibacterial activity of CMCS–BC hydrogels is shown using the CFU method against Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus). As a result, the CMCS–BC hydrogels exhibit more suitable antibacterial properties than those without BC due to the CMCS having amino groups that enhance antibacterial properties. Therefore, CMCS–BC hydrogels can be considered suitable for antibacterial wound dressing applications.

Abstract:Cellulose/chitosan composite, as a mature commercial antibacterial dressing, is an important type of wound repair material. However, how to achieve the perfect compound of two components and improve antibacterial activity is a major, lingering issue. In this study, a bifunctional group modified bacterial cellulose (DCBC) was prepared by carboxymethylation and selective oxidation. Further, the chitosan (CS) was compounded in the network of DCBC by self-crosslinking to form dialdehyde carboxymethyl bacterial cellulose/chitosan composites (S-DCBC/CS). The aldehyde group can react with amino of CS by Schiff base reaction. The carboxyl group of DCBC and the amorphous distribution of CS molecular chains increase the antimicrobial properties of composites. The bacteriostatic rate of composites could be higher than 95%. Bacteria can be attracted onto the surface of composites, what we call it "directional adhesion antibacterial effects". In particular, a kind of large animal wound model, deep Ⅱ degree infected scald of Bama miniature pig, was used to research the antimicrobial and healing properties of materials. The S-DCBC/CS can effectively inhibit bacterial proliferation of wound and kill the bacteria. The wound healing rate of S-DCBC/CS was up to 80% after three weeks. The composites show better antibacterial and promoting concrescence effects than traditional chitosan dressings.

Therefore Zn2+-loaded adhesive bacterial cellulose hydrogel was designed to observe its angiogenic and antibacterial abilities in the wound healing process. Methods The characterization, tensile strength, swelling behaviors and antibacterial activity of bacterial cellulose/polydopamine/zeolitic imidazolate framework-8 (BC/PDA/ZIF8) hydrogels were tested. Cell-Counting-Kit-8 (CCK8), transwell, tube formation and real time qunantitative PCR (qRT-PCR) assays were performed to evaluate the cell compatibility of BC/PDA/ZIF8 hydrogels in vitro. A full-thickness defect wound model and histological assays were used to evaluate the BC/PDA/ZIF8 hydrogels in vivo. Results The prepared BC/PDA/ZIF8 hydrogels exhibited suitable mechanical strength, excellent swelling properties, good tissue adhesion, efficient angiogenic and antibacterial effects and good performance as a physical barrier. In vivo experiments showed that the BC/PDA/ZIF8 hydrogels accelerated wound healing in a full-thickness defect wound model by stimulating angiogenesis. Conclusions This study proved that BC/PDA/ZIF8 hydrogels possess great potential for promoting satisfactory wound healing in full-thickness wound defects through antibacterial effects and improved cell proliferation, tissue formation, remodeling and re-epithelialization.

Abstract:Composite bacterial cellulose (BC) based hydrogel with alginate (A) or pectin (P) or alginate and pectin was fabricated via a physical crosslinking technique using calcium chloride (CaCl2) solution and incorporated with polyhexamethylene biguanide (PHMB) as an effective antimicrobial drug by immersion method. After that, the physicochemical properties of all hydrogel formulations were characterized. The result showed that the formulations with PHMB performed better physicochemical properties than the hydrogel without PHMB. Fourier transform infrared spectroscopy (FT-IR) showed the interaction between PHMB and the carboxylic group of alginate and pectin. BC/A-PHMB hydrogel performed suitable mechanical strength, fluid uptake ability, water retention property, drug content, high integrity value, and maximum swelling degree. Moreover, in vitro cell viability of BC/A-PHMB hydrogel revealed high biocompatibility with human keratinocyte cell line (HaCaT) and demonstrated prolong released of PHMB in Tris-HCl buffer pH 7.4, while rapid release in phosphate buffer saline pH 7.4. BC/A-PHMB hydrogel demonstrated good anti-bacterial activity against S. aureus and P. aeruginosa. In conclusion, BC/A-PHMB hydrogel could be a potential dual crosslinked ion-based hydrogel for wound dressing with anti-bacterial activity.

Abstract:Bacterial infections are a common problem associated with wound treatment that imposes a significant burden on healthcare systems and patients. As a result, healthcare providers urgently need new treatment strategies to protect people. Hydrogel biomaterials with inherent antimicrobial properties offer an attractive and viable solution to this issue. Here, for the first time, we have developed a new efficient synthetic strategy to prepare cationic hydrogels (PHCI) with intrinsically efficient antimicrobial properties by chemically cross-linking trans-1,4-cyclohexanediamine with 1,3-dibromo-2-propanol using a condensation reaction without the use of toxic cross-linking agents. As expected, the prepared PHCI hydrogel possessed an inherent antibacterial ability that can adsorb and kill Staphylococcus aureus and Escherichia coli electrostatically. Notably, in vivo experiments on normal and diabetic rat models confirmed that the PHCI hydrogel can quickly stop bleeding, efficiently kill bacteria, promote the conversion of macrophages from the proinflammatory M1 phenotype to the repaired M2 phenotype, and accelerate collagen deposition and blood vessel formation, thereby achieving rapid wound healing. Overall, this work presents an effective antibacterial dressing that might provide a facile but effective approach for clinical wound management.

Abstract:Bacterial cellulose (BC) produced by Acetobacter xylinum has great advantages in wound dressing. However, the structural limitation under static culture, and lack of antibacterial properties restrict its application, especially for infectious wound healing. The present study reported an original wound dressing, which was composed of a Janus BC membrane with antibacterial nano-sized copper oxide (CuO) through polydopamine (PDA) conjugation to promote wound healing under infectious condition. The finished product (CuO/PDA/BC membrane) exhibited favorable air permeability, high hydrophilicity and good mechanical properties, as well as strong antibacterial effects by the sustained release of CuO and photothermal effect of CuO/PDA. Furthermore, CuO/PDA/BC membrane inhibited inflammatory response and promoted wound healing in an infectious wound model in vivo. These results suggested that our CuO/PDA/BC membrane had great potential as wound dressing for infectious wound healing.
