Abstract:Designing and developing antibacterial hydrogel dressing with multiple functions including adhesiveness, good mechanical performance, self-healing, and hemostatic to improve full-thickness skin wound regeneration is greatly desirable in healthcare. In this work, a versatile hydrogel was fabricated based on bacterial cellulose decorated with polydopamine (PDA@BC), polyvinyl alcohol (PVA) via dynamic borate (PB) ester bond crosslinking. The tissue adhesiveness, self-healing mechanical property, and hemostatic ability of the hydrogels were improved. Doxycycline hydrochloride (Doxy)-load PB-PDA@BC hydrogels exhibit excellent antibacterial activity by continuous drug release of Doxy. The hydrogels also exhibited high swelling ratio, good biocompatibility, and suitable mechanical property. Compared with commercial Tegaderm film dressing, PB-PDA@BC/Doxy hydrogels dressing could better promote wound closure in a mouse full-thickness wound model. In generally, our results demonstrated that the PB-PDA@BC/Doxy hydrogels dressing can be considered an excellent wound dressing for wound healing.

Mussel-Inspired Hydrogel with Potent in Vivo Contact-Active Antimicrobial and Wound Healing Promoting Activities

Abstract:Open wounds (e.g., burns and trauma) are always challenged by various opportunistic bacteria. There is an urgent need for developing novel wound dressing that is able to prevent bacterial infection and promote the healing simultaneously. Herein, we developed a new type of antimicrobial hydrogels for the open wound healing through imitating a facile mussel-inspired catechol/polyamine chemistry. This hydrogel was prepared using catechol (CT) and ε-poly-l-lysine (EPL) by oxidative cross-linking directly in the open air at room temperature. This nonleaching CT/EPL hydrogel not only exhibited excellent contact-active antimicrobial activities against Gram-negative bacteria Escherichia coli (E. coli) and Gram-positive "superbug" methicillin-resistant Staphylococcus aureus (MRSA) but also inhibited the biofilm formation in vitro. Moreover, the animal burn wound model study clearly validated the in vivo anti-infective property of CT/EPL hydrogel against MRSA infection. More importantly, the CT/EPL hydrogel possessed low cytotoxicity and enhanced cell migration in vitro. A full-thickness cutaneous wound model study revealed that CT/EPL hydrogel upregulated the expression of vascular endothelial growth factor (VEGF) and reduced the production of the pro-inflammatory cytokines, thus promoted the wound healing. These findings suggested that the CT/EPL hydrogel have great potential as a wound dressing for preventing bacterial infection and accelerating healing of open wounds.

Abstract:Hydrogel dressings have been widely used for wound management due to their ability to maintain a hydrated wound environment, restore the skin's physical barrier and facilitate regular dressing replacement. However, the therapeutic functions of standard hydrogel dressings are restricted. In this study, an injectable hybrid hydrogel dressing system was prepared from a polyethylene glycol (PEG)-based thermoresponsive hyperbranched multiacrylate functional copolymer and thiol-modified hyaluronic acid in combination with adipose-derived stem cells (ADSCs). The cell viability, proliferation and metabolic activity of the encapsulated ADSCs were studied in vitro, and a rat dorsal full-thickness wound model was used to evaluate this bioactive hydrogel dressing in vivo. It was found that long-term cell viability could be achieved for both in vitro (21days) and in vivo (14days) studies. With ADSCs, this hydrogel system prevented wound contraction and enhanced angiogenesis, showing the potential of this system as a bioactive hydrogel dressing for wound healing.
Rapid Preparation of Highly Stretchable and Fast Self-Repairing Antibacterial Hydrogels for Promoting Hemostasis and Wound Healing.

Abstract:Hydrogel dressings have emerged as a vital resource in wound management, offering several advantages over conventional wound dressing materials. Their inherent biocompatibility, ability to replicate the native extracellular matrix, and capacity to provide an ideal environment for cell survival make them particularly valuable. Nevertheless, the mechanical properties of many hydrogel dressings are an area that warrants improvement, as it currently constrains their application range. This limitation is especially evident when skin wounds are addressed in highly active or easily scratched areas. In this study, we present the development of a highly stretchable self-repairing hydrogel by cross-linking poly(vinyl alcohol) (PVA) through dynamic boron ester bonds, coupled with the hydrogen bonding of carboxymethyl cellulose sodium (CMC) via an efficient one-pot method without adding any catalyst. This innovative PVA/CMC hydrogel exhibited remarkable antibacterial properties achieved through the incorporation of bergamot oil, which was dispersed in a β-cyclodextrin solution. The hydrogel's elongation at the point of rupture reached an impressive 1910%, and it was capable of rapid self-healing in just 3 min upon bonding. Additionally, the hydrogel demonstrated excellent hemostatic properties, effectively mitigating blood loss and exudation. In vivo wound models have shown that PVA/CMC significantly expedites wound healing by reducing bacterial infections, inflammatory responses, and blood loss and by promoting collagen deposition. In summary, this research provides crucial insights into its potential as an advanced wound dressing material, particularly well-suited for addressing wounds in places with frequent activities or easy scratches.

Abstract:One of the main reasons impeding wound healing is wound infection caused by bacterial colonization with a continuous stage of inflammation. Traditional wound treatments like gauze are being replaced by tissue adhesives with strong wet tissue adhesion and biocompatibility. Herein, a fast-crosslinking hydrogel is developed to achieve both strong antimicrobial properties and excellent biocompatibility. In this study, a simple and non-toxic composite hydrogel was prepared by the Schiff base reaction between the aldehyde group of 2,3,4-trihydroxybenzaldehyde (TBA) and the amino group of ε -Poly- L -lysine (EPL). Subsequently, a succession of experiments toward this new hydrogel including structure characterization, antimicrobial properties, cell experiment and wound healing were applied. The results of the experiments show that the EPL-TBA hydrogel not only exhibited excellent contact-active antimicrobial activities against Gram-negative bacteria Escherichia coli ( E. coil ) and Gram-positive Bacteria Staphylococcus aureus ( S. aureus ), but also inhibited the biofilm formation. More importantly, the EPL-TBA hydrogel promoted the wound healing with low cytotoxicity in vivo . These findings indicate that the EPL-TBA hydrogel has a promising use as a wound dressing in the bacterial infection prevention and wounds healing acceleration.

An antibacterial and anti-oxidant hydrogel containing hyperbranched poly-l-lysine and tea polyphenols accelerates healing of infected wound

Abstract:Both bacteria-infection and excessive inflammation delay the wound healing process and even create non-healing wound, thus it is highly desirable to endow the wound dressing with bactericidal and anti-oxidation properties. Herein an antibacterial and antioxidation hydrogel based on Carbomer 940 (CBM) and hydroxypropyl methyl cellulose (HPMC) loaded with tea polyphenols (TP) and hyperbranched poly-l-lysine (HBPL) was designed and fabricated. The hydrogel killed 99.9 % of methicillin-resistant Staphylococcus aureus (MRSA) and Escherichia coli (E. coli) at 107 CFU mL-1, and showed strong antioxidation against H2O2 and 2,2-di(4-tert-octylphenyl)-1-picryl-hydrazyl (DPPH) radicals without noticeable cytotoxicity in vitro. The CBM/HPMC/HBPL/TP hydrogel significantly shortened the inflammatory period of the MRSA-infected full-thickness skin wound of rats in vivo, with 2 orders of lower MRSA colonies compared with the blank control, and promoted the wound closure especially at the earlier stage. The inflammation was suppressed and the vascularization was promoted significantly as well, resulting in reduced pro-inflammatory factors including interleukin-6 (IL-6), interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α), and increased anti-inflammatory factors such as interleukin-4 (IL-4) and interleukin-10 (IL-10).
Double-network cellulose-based hybrid hydrogels with favourable biocompatibility and antibacterial activity for wound healing

Abstract:Bacterial infections are among the leading causes of delayed wound healing. At present, a series of antibacterial materials, such as antibiotics, antimicrobial peptides (AMPs), metals and metal oxides (MMOs), have been used to fabricate antibacterial wound dressings. However, their translational potential is limited owing to their poor biocompatibility. ε-Polylysine (ε-PL) is a natural macromolecule with excellent biocompatibility and broad-spectrum antibacterial activity. Herein, ε-PL was incorporated into a cellulose/γ-polyglutamic acid (γ-PGA) composite hydrogel to form a novel double-network hydrogel termed as CGLH. The elastic modulus of CGLH increased from 0.097 ± 0.015 MPa to 0.441 ± 0.096 MPa, and the equilibrium swelling ratio increased from 382.7 ± 24.3 % to 611.2 ± 8.6 %. Several preclinical models were used to investigate the translational potential of this hydrogel. CGLH exhibited good biocompatibility and antibacterial activity, which promoted the healing of infected and critical-size wounds within 12 days. CGLH had positive effects on collagen synthesis, vascularization and cell proliferation. As a result, this study not only provided an effective alternative for wound healing but also proposed a double-network strategy for creating biocompatible and antibacterial biomaterials.
Barnacle-inspired and polyphenol-assisted modification of bacterial cellulose-based wound dressings for promoting infectious wound healing

Abstract:Bacterial cellulose (BC) is an ideal candidate for wound dressings due to its natural origin, exceptional water-holding capacity, pliability, biocompatibility, and high absorption capability. However, the lack of essential antimicrobial activity limits its biomedical applications. This study reported BC-based wound dressings containing silk fibroin protein (SF), offering the potential for biomimetic properties, and (-)-epigallocatechin-3-gallate (EGCG) for polyphenol-assisted surface modification to promote infectious wound healing. Glycerol was used as the carbon source to promote the formation of an adhesive layer by facilitating the β-sheet folding of SF, and different concentrations of EGCG were employed to interact with SF through strong hydrogen bonding facilitated by the polyphenolic groups. The functionalized membrane exhibited outstanding water-holding capacity, swelling ratio, and degradation properties, along with enhanced hydrophilicity, adhesiveness, and a remarkable free radical scavenging ability. Both in vitro and in vivo experiments confirmed its potent bacteriostatic activity. The composite membrane displayed excellent biocompatibility, including cellular and hemocompatibility. Importantly, it effectively promoted wound healing in murine back infections. These findings suggest the significant feasibility of the innovative modification approach, and that functionalized membranes have great potential as wound-dressing materials for infection management in future clinical applications.

Abstract:<p>Treatment of wounds in special areas is challenging due to inevitable movements and difficult fixation. Common cotton gauze suffers from incomplete joint surface coverage, confinement of joint movement, lack of antibacterial function, and frequent replacements. Hydrogels have been considered as good candidates for wound dressing because of their good flexibility and biocompatibility. Nevertheless, the adhesive, mechanical, and antibacterial properties of conventional hydrogels are not satisfactory. Herein, cationic polyelectrolyte brushes grafted from bacterial cellulose (BC) nanofibers are introduced into polydopamine/polyacrylamide hydrogels. The 1D polymer brushes have rigid BC backbones to enhance mechanical property of hydrogels, realizing high tensile strength (21–51 kPa), large tensile strain (899–1047%), and ideal compressive property. Positively charged quaternary ammonium groups of tethered polymer brushes provide long‐lasting antibacterial property to hydrogels and promote crawling and proliferation of negatively charged epidermis cells. Moreover, the hydrogels are rich in catechol groups and capable of adhering to various surfaces, meeting adhesive demand of large movement for special areas. With the above merits, the hydrogels demonstrate less inflammatory response and faster healing speed for in vivo wound healing on rats. Therefore, the multifunctional hydrogels show stable covering, little displacement, long‐lasting antibacteria, and fast wound healing, demonstrating promise in wound dressing.</p>

Fabrication of Bacterial Cellulose-Based Dressings for Promoting Infected Wound Healing

Abstract:Bacterial cellulose (BC) holds several unique properties such as high water retention capability, flexibility, biocompatibility, and high absorption capacity. All these features make it a potential material for wound healing applications. However, it lacks antibacterial properties, which hampers its applications for infectious wound healings. This study reported BC-based dressings containing ε-polylysine (ε-PL), cross-linked by a biocompatible and mussel-inspired polydopamine (PDA) for promoting infectious wound healing. BC membranes were coated with PDA by a simple self-polymerization process, followed by treating with different contents of ε-PL. The resulted membranes showed strong antibacterial properties against tested bacteria by both in vitro and in vivo evaluations. The membranes also exhibited hemocompatibility and cytocompatibility by in vitro investigations. Moreover, the functionalized membranes promoted infected wound healing using Sprague–Dawley rats as a model animal. A complete wound healing was observed in the group treated with functionalized membranes, while wounds were still open for control and pure BC groups in the same duration. Histological investigations indicated that the thickness of newborn skin was greater and smoother in the groups treated with modified membranes in comparison to neat BC or control groups. Covalently grafting polycation to bacterial cellulose for antibacterial and anti-cell adhesive wound dressings

Abstract:Hydrogel-based wound dressings are becoming increasingly important for wound healing. Bacterial cellulose (BC) has been commonly used as wound dressings due to its good in vitro and in vivo biocompatibility. However, pure BC does not possess antibacterial properties. In this regard, polycation gel was grafted onto the BC using a surface-initiated activator regenerated by electron transfer atom transfer radical polymerization (SI-ARGET ATRP) with subsequent quaternization for antibacterial wound dressing. Dimethylethyl methacrylate (DMAEMA) was successfully polymerized on the BC surface which was confirmed by Fourier transform infrared spectroscopy and elemental analysis. The morphology structure, specific surface area, pore size, and mechanical properties were also characterized. The quaternized PDMAEMA grafted on the BC endowed it with excellent antibacterial activity against E. coli (Gram-negative) and S. aureus (Gram-positive) with a killing rate of 89.2 % and 93.4 %, respectively. The number of cells was significantly reduced on QPD/BC hydrogel, demonstrating its good anti-adhesion ability. In vitro cellular evaluation revealed that the antibacterial wound dressing exhibited good biocompatibility. Overall, this study provides a feasible method to develop antibacterial and anti-cell adhesive hydrogel, which has a promising potential for wound healing.
A lignocellulose-based nanocomposite hydrogel with pH-sensitive and potent antibacterial activity for wound healing

Abstract:Hydrogel dressings with similar structural characteristics to the extracellular matrix and tunable physicochemical properties have become promising candidates for wound healing. However, the fabrication of an ideal hydrogel dressing with low-cost, good biocompatibility, excellent hemostatic capacity, potent and broad-spectrum antibacterial activity remains a huge challenge. Herein, a lignocellulose-based nanocomposite hydrogel (ATC/SA/PVA) is fabricated by simply mixing Ag nanoparticles loaded, tannic acid-decorated lignocellulose nanofibrils with sodium alginate and polyvinyl alcohol. Based on the dynamic borate ester bonds and multiple weak hydrogen bonds, the fabricated hydrogel exhibits excellent flexibility and self-healing performance. Its highly porous structure endows the gel excellent blood and tissue exudates absorption ability. Interestingly, the release behavior of Ag nanoparticles from hydrogel displays pH dependence, which can facilitate the accumulation of Ag nanoparticles at the wound site, thereby accelerating the process of wound healing. In vitro antibacterial assay demonstrates the potent antibacterial ability of hydrogel against both Gram-positive (S. aureus) and negative bacteria (E. coli). More importantly, in vivo investigations reveal that such hydrogel can effectively accelerate tissue regeneration and wound healing with no obvious adverse effects. All these results suggest that this nanocomposite hydrogel would be a promising candidate to accelerate wound healing.
Biocompatible gellan gum/sericin hydrogels containing halloysite@polydopamine nanotubes with hemostasis and photothermal antibacterial properties for promoting infectious wound repair

Abstract:Skin wounds accompanied by massive bleeding and bacterial infection can be life-threatening and pose a significant challenge to clinical wound management. Herein, we prepared gellan gum/sericin hydrogels containing PDA-encapsulated halloysite nanotubes () to achieve fast hemostasis and photothermally antibacterial effects. The morphology and compressive properties of the composite hydrogels were characterized, and the biocompatibility was investigated by hemocompatibility and cytocompatibility tests. The hemostatic properties were further evaluated in the rat liver hemorrhage and tail amputation model. The mechanical performance (compressive strength 353.2 KPa) and the hemostatic effect (30.33±3.68 s in the liver injury model and 28.33±2.35 s in the caudal amputation model) of the composite hydrogel were improved due to the introduction of In addition, the composite hydrogel rapidly heated under 808 nm near-infrared (NIR) light irradiation, making the hydrogel extremely effective against E. coli and S. aureus . Furthermore, the wound closure rate, granulation tissue thickness, collagen deposition, and expression of inflammation-associated cytokines (CD68) and angiogenesis-associated cytokines (CD31) were investigated in a rat full-thickness skin defect model infected with S. aureus. All results showed that this multifunctional hydrogel provided better healing effects, suggesting that the composite hydrogel has become a promising alternative for infected wound repair.

Antimicrobial and Biocompatible Ε-Polylysine–γ-poly(glutamic Acid)–based Hydrogel System for Wound Healing

Abstract:Biocompatible and antimicrobial wound dressings are important biomaterials for tissue adhesion and healing. Hydrogels derived from natural polyamino acids are ideal wound dressing because of their good water solubility, biocompatibility, and biodegradability. In this study, we introduce poly(amino acid)–based hydrogels as a new type of wound dressing for wound healing. The composite hydrogels were derived from water-soluble ε-polylysine and γ-poly(glutamic acid) cross-linked by 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide and N-hydroxy-succinimide. The gelation is attributed to the reaction between the amino groups of ε-polylysine and carboxyl groups of γ-poly(glutamic acid). Fourier transform infrared spectroscopy was used to characterize the composite hydrogels. The effect of the ratio of ε-polylysine and γ-poly(glutamic acid) on the gelation time, surface morphology, equilibrium swelling, elastic moduli, adhesive strength, and in vitro degradation of composite hydrogels was examined. The hydrogels showed excellent antimicrobial efficacy against Escherichia coli and Staphylococcus aureus. Indirect cytotoxicity assessment indicated that the composite hydrogels were nontoxic to the L929 cell. The potential of the composite hydrogel as wound dressing was demonstrated by applying it in the skin incision. Results demonstrated that the composite hydrogel showed superior healing effects compared with suture, fibrin glue, and compont skin adhesive, while eliciting less inflammatory response. In conclusion, antimicrobial and biocompatible ε-polylysine–γ-poly(glutamic acid) composite may have great application for wound healing.

Abstract:Hydrogels have attracted widespread attention in controlling bleeding due to their superiority in matching irregular defects. However, inadequate mechanical performance, insufficient adhesion and long gelation times still hinder their application for hemostasis. In this study, a novel hydrogel with self-healing and adhesive properties was designed and synthesized by introducing reversible borate ester bonds of polyvinyl alcohol-borax into tannic acid-modified bacterial cellulose (TA@BC). As demonstrated by tensile stress‒strain tests and rheological measurements, the hydrogels have excellent mechanical performance owing to the synergistic multiple crosslinked bonds among tannic acid-modified bacterial cellulose (TA@BC), polyvinyl alcohol polymer chains and borax in a covalent polymer network. The hydrogels show good self-healing properties and adhesion to different substrates. Moreover, blood loss from a wound treated with the designed hydrogel was approximately 1/5th that from an untreated wound, demonstrating the excellent hemostatic capacity of the hydrogels. Therefore, the designed hydrogels have potential applications in hemostasis and wound dressing.Graphical abstract

Abstract:Wound dressings play a critical role in the wound healing process; however, conventional dressings often address singular functions, lacking versatility in meeting diverse wound healing requirements. Herein, a dual-network, multifunctional hydrogel (PSA/CS-GA) has been designed, synthesized through a one-pot approach. The in vitro and in vivo experiments demonstrate that the optimized hydrogel has exceptional antifouling properties, potent antibacterial effects and rapid hemostatic capabilities. Notably, in a full-thickness rat wound model, the hydrogel group displays a remarkable wound healing rate exceeding 95% at day 10, surpassing both the control group and the commercially 3M group. Furthermore, the hydrogel exerts an anti-inflammatory effect by reducing inflammatory factors IL-6 and tumor necrosis factor-α (TNF-α), enhances the release of vascular endothelial growth factor (VEGF) to promote blood vessel proliferation, and augments collagen deposition in the wound, thus effectively accelerating wound healing in vivo. This innovative hydrogel presents a novel and highly effective approach to wound healing.

Abstract:Bacterial infections significantly deteriorate the process of wound healing. The wound dressings loaded with antimicrobials are widely used to reduce bacterial infections. However, release-based sterilization may increase the risk of drug resistance of bacteria and complicate translation. Thus, the development of long-term intrinsic antibacterial wound dressings is highly desirable. In this study, an intrinsic antibacterial hydrogel (PVA/PPG-HBPL) consisting of poly(vinyl alcohol) (PVA), poly(polyethylene glycol methyl ether methacrylate-co-glycidyl methacrylate) (PPG), and hyperbranched poly-l-lysine (HBPL) was designed and fabricated. The mechanical properties of the PVA/PPG-HBPL hydrogel were enhanced by hydrogen bonding and semi-interpenetrating networks. It also possessed a favorable ability to absorb the wound exudates. The release of antibacterial HBPL was significantly decreased by the methods of cyclic freeze-thawing and covalent cross-linking during hydrogel fabrication, enabling the PVA/PPG-HBPL hydrogel with intrinsic and long-term antibacterial performance. The PVA/PPG-HBPL hydrogel dressing killed 99.9% of methicillin-resistant Staphylococcus aureus (MRSA) cultured on its surface without observable cytotoxicity in vitro. It observably shortened the healing process by 2 orders of magnitude of MRSA colonies compared with the control in the MRSA-infected full-thickness skin wound of rats in vivo even after being soaked in phosphate-buffered saline (PBS) for 21 days (PBS was changed every 3 days). The antibacterial hydrogels could kill wound bacteria in a timely manner, significantly reduce inflammatory cell infiltration, and promote neovascularization and collagen deposition.

Abstract:Bacterial infections in wounds often delay the healing process which threaten human health and life. The development of biocompatible hydrogels with antibacterial activity and self-healing to achieve excellent wound closure effectiveness is highly desirable in clinical applications. Herein, a new class of highly stretchable, biocompatible, and antibacterial hydrogel dressings called BPP hydrogel is designed and prepared by chemically cross-linking β-CD with PEI and then physically cross-linking with PVA. The BPP hydrogel exhibits superior properties including excellent antibacterial properties against Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli, good biocompatibility for mouse fibroblasts, mouse embryonic fibroblasts and human umbilical vein endothelial cells, outstanding deformability and self-healing performance to protect the wound completely even in special scenes such as pulling and bending. Besides, after damage, the broken BPP hydrogel could heal together, and the healed BPP hydrogel could be stretched to 150% breaking deformation. Furthermore, in vivo experiments further verify that BPP hydrogel has excellent sterilization and healing effect on infectious wounds. This work proves that BPP hydrogel would be a promising material for wound healing and has good prospect for clinical translation.

Abstract:Hydrogels can relieve wound pain and provide a moist environment for wound healing. Although hydrogels based on natural materials have proven to be biocompatible and biodegradable, their antibacterial and anti-inflammatory properties are not desirable. Natural extracts with antimicrobial and antioxidant properties are becoming increasingly attractive because of their environmentally friendly nature and good biocompatibility. Herein, composite cellulose/silk fibroin/polyacrylic acid hydrogels based on natural materials were fabricated by crosslinking between oxidized cellulose and silk fibroin together with radical polymerization of acrylic acid. The fabricated hydrogels displayed a uniform hierarchical topography with high porosity, as well as good mechanical properties, swelling properties, air permeability and biocompatibility. With the addition of tea polyphenols to natural extracts, the composite hydrogels exhibited significant antimicrobial and antioxidant properties such that they could prevent bacterial infection and relieve wound inflammation in the inflammation stage and could promote the synthesis of extracellular matrix (ECM), angiogenesis, collagenization and re-epithelization during wound healing. The tea polyphenols/cellulose/silk fibroin/polyacrylic acid hydrogels fabricated in this study can accelerate wound healing, demonstrating their great potential for application in wound dressings.
