Abstract:A unique multifunctional photoelectrochemical(PEC) platform for the simultaneous detection and inactivation of Salmonella enteritidis (SE) was proposed. The photoactive material in the platform was a Bi2MoO6/V2CTx composite, and gold nanoparticles were modified to link the aptamer via Au-S bonds. The aptamer was to link the complementary DNA (cDNA) to the electrode surface via base complementarity. In the presence of SE, the aptamer attached to Salmonella enteritidis selectively and then the cDNA was released from the aptamer electrode, the photocurrent signal was amplified, allowing for quantitative detection of Salmonella enteritidis. The linear range of the method was 1.82 × 102 CFU/mL to 1.82 × 108 CFU/mL based on the specific recognition of Salmonella enteritidis by aptamer. The detection limit was 26 CFU/mL. Furthermore, due to its photoelectrocatalytic action, the PEC platform demonstrated strong antibacterial activity (99.99 %). The new technique was created to detect and inactivate Salmonella enteritidis in a timely and sensitive manner.

Abstract:By means of the specific immuno-recognition and ultra-sensitive mass detection, a quartz crystal microbalance (QCM) biosensor for Escherichia coli O157: H7 detection was developed in this work. As a suitable surfactant, 16-mercaptohexadecanoic acid (MHDA) was introduced onto the Au surface of QCM, and then self-assembled with N-hydroxysuccinimide (NHS) raster as a reactive intermediate to provide an active interface for the specific antibody immobilization. The binding of target bacteria with the immobilized antibodies decreased the sensor's resonant frequency, and the frequency shift was correlated to the bacterial concentration. The stepwise assembly of the immunosensor was characterized by means of the electrochemical techniques. Using the immersion-dry-immersion procedure, this QCM biosensor could detect 2.0 x 10(2) colony forming units (CFU)/ml E. coli O157: H7. In order to reduce the fabrication time, a polyelectrolyte layer-by-layer self-assembly (LBL-SA) method was adopted for fast construction. Finally, the reproducibility of this biosensor was discussed.

Abstract:The sensitive determination of foodborne pathogens in foodstuffs or water system is essential for protecting food and environment safety. Herein, we have proposed a photoelectrochemical (PEC) aptasensor based on two-dimensional (2D)/3D organic–inorganic heterojunction for the efficient detection of Salmonella typhimurium ( S. typhimurium ). The heterojunction was established via the in situ growth of copper (II) selenide (CuSe) nanosheets around the photoactive triazine covalent-organic polymer (COP) nanospheres, which was synthesized using 2,4,6-Tris(4-aminophenyl)-1,3,5-Triazine (TAPT) and 2,4,6-trihydroxy-1,3,5-benzenetricarbaldehyde (Tp) as building blocks (denoted as ). Integrating the features of TAPT-Tp-COP (such as large specific surface and pore size, multi-functionality, and rich defects) and CuSe (such as high photoabsorption ability, fast electron transfer and effective separation of the photo-generated charge carrier) not only can accelerate the photoelectric transformation efficiency, but also can promote the aptamer immobilization. Consequently, the constructed PEC aptasensor based on illustrated a low detection limit of 3.4 CFU mL −1 with a linear range from 1 × 10 1 to 1 × 10 5 CFU mL −1 toward S. typhimurium . In addition, it demonstrated high selectivity in light of the specific recognition of aptamer toward S. typhimurium , good reproducibility and stability, and promising regeneration ability, as well as wide applications for the detection of S. typhimurium in diverse foodstuffs systems. The present work provides a new aptasensing strategy for the effective detection of foodborne pathogens from diverse foodstuffs by combining the merits of COPs and PEC technique.

Abstract:Self-powered photoelectrochemical (PEC) sensing is a novel sensing modality. The introduction of dual-mode sensing and photoelectrocatalysis in a self-powered system enables both detection and sterilization purposes. To this end, herein, a self-powered multifunctional platform for the photoelectrochemical-fluorescence (PEC-FL) detection and in-situ inactivation of Salmonella enteritidis (SE) was constructed. The platform utilized Bi4NbO8Cl/V2CTx/FTO as a photoanode and CuInS2/FTO as a photocathode and incubated quantum dot (QDs) signaling probes on the surface of the photocathode. During detection, the system drives the transfer of photogenerated electrons between the dual photoelectrodes through the Fermi energy level difference. The photoanode amplifies the photoelectric signal, while the photocathode is solely dedicated to the immune recognition process. QDs provide an additional fluorescence signal to the system. Under optimal experimental conditions, the multifunctional platform achieves detection limits of 3.2 and 5.3 CFU/mL in PEC and FL modes respectively, with a detection range of 2.91 × 102 to 2.91 × 108 CFU/mL. With the application of an external bias voltage, it further promotes electron transfer between the dual photoelectrodes, inhibits the recombination of photogenerated electrons and holes. It generates a significant amount of superoxide radicals (·O2-) in the cathodic region, resulting in strong sterilization efficiency (99%). The constructed self-powered multifunctional platform exhibits high sensitivity and sterilization efficiency, it provides a feasible and effective strategy to enhance the comprehensive capability of self-powered sensors.
Development of a Dual Mode UCNPs-MB Biosensor in Combination with PCR for Sensitive Detection of Salmonella

Abstract:In recent years, the high prevalence of Salmonella has emerged as a serious threat to public safety, prompting attempts to utilize accurate, rapid, and direct methods to ensure food safety. In this study, a multifunctional platform featuring dual-mode detection channels (colorimetric-fluorescence) combined with polymer chain reaction (PCR) was proposed for the sensitive and rapid detection of Salmonella. Additionally, the colorimetric measurements were achieved by color changes induced by methylene blue (MB) insertion into the double-stranded DNA, and the fluorescence measurements were performed by internal filter effect (IFE)-induced fluorescence quenching of upconversion nanoparticles (UCNPs) by MB. The results showed that the IFE and PCR amplification processes improved the sensitivity of the sensor towards Salmonella detection, with a limit of detection (LOD) of 21.8 CFU/mL. Moreover, this colorimetric-fluorescence dual-mode PCR biosensor was applied to determine Salmonella in food samples, such as chicken, egg, and fish, which produced satisfactory results. Overall, the present study results demonstrate the potential for combining PCR amplification with IFE to develop an efficient and reliable dual-mode analysis platform to safeguard food security.

Abstract:AIMS To determine the views of teachers, general practitioners (GPs) and paediatricians about whether there has been a recent change in the timing of the onset of puberty in children in the UK and in the timing of menarche in girls. DESIGN Questionnaires were sent to a stratified random sample of each professional group. RESULTS In relation to girls, almost 80% of teachers believe that the onset of puberty is occurring earlier and 73% feel that the menarche is occurring earlier. Those who had been working for longer than 10 years were more likely to say that the age of onset of puberty had decreased. GPs feel that both puberty and menarche are starting earlier, whereas paediatricians were evenly divided between those who thought puberty was starting earlier and those who thought that the timing had not changed. In boys, professionals generally believe that the timing of the onset of puberty has not altered significantly. CONCLUSION Further study is required to determine whether puberty is really occurring earlier in girls.

Abstract:A rapid and sensitive strategy for the detection of Salmonella was proposed by integrating simple DNA extraction, specific polymerase chain reaction (PCR) with an invA gene-based electrochemical DNA sensor. The amplified target sequence of invA gene could be specifically captured on the sensing interface, and further hybridized with biotinylated detection probe to form a sandwich-type hybridization structure. The electrochemical signal was amplified by streptavidin-alkaline phosphatase (ST-AP), producing sensitive enzyme-catalyzed electrochemical DNA sensing. The fabrication and hybridization processes were characterized with electrochemical impedance spectroscopy (EIS), square wave voltammetry (SWV) and surface plasmon resonance (SPR). The designed DNA sensor could discriminate satisfactorily the complementary and mismatched oligonucleotides, indicating good selectivity. The linear calibration range for target DNA detection was from 1 pM to 10 nM with a detection limit of 0.5 pM, showing high sensitivity. Under optimal conditions, the proposed strategy could quantitatively detect Salmonella from 10 to 10(5) CFU mL(-1) within 3.5 h. This strategy presented a simple, rapid and sensitive platform for Salmonella detection and would become a powerful tool for pathogenic microorganisms screening in clinical diagnostics, food safety, biothreat detection and environmental monitoring.

Abstract:Eating food contaminated by foodborne pathogens can lead to illness. The development of electrochemical sensors for pathogen detection has received widespread attention. However, the analytical performance of electrochemical sensors is inevitably affected by the non-specific adsorption of molecules in the sample. Moreover, the external signal probes might be affected by the complex components in the sample accompanied with signal suppression. This work presents an electrochemical aptasensor for Salmonella typhimurium detection based on the self-signal of poly-xanthurenic acid and the antifouling ability of chondroitin sulfate. The detection time was 60 min. The linear range was from 101 to 107 CFU/mL, and the detection limit was 3 CFU/mL. The biosensors presented good repeatability and storage stability. And the biosensors has been successfully applied in milk and orange juice. This strategy is expected to be applied in the design of other antifouling biosensors, to achieve rapid detection of pathogens and ensure food safety.

Abstract:Efficient detection of pathogenic bacteria is paramount for ensuring food safety and safeguarding public health. Herein, we developed a label-free and signal-on dual-target recognition electrochemical DNA sensing platform based on the conformational formation of split G-quadruplex. This platform focused on achieving sensitive and low-cost detection of Salmonella and its most human-infecting S. typhimurium serotype. In simple terms, the dual-target recognition probe (DTR-6P) was ingeniously designed for the loop sequence on the loop-mediated isothermal amplification (LAMP) amplicons. It could recognize two different genes and release their corresponding G-rich sequences. The exfoliated G-rich sequences could be captured by the capture probes on the electrode, and then the bimolecular G-quadruplex or the tetramolecular G-quadruplex would be formed to capture hemin, thereby enabling dual-signal reporting. The minimum detection amount of target genes can be as low as 2 copies/μL. Encouragingly, the real food samples contaminated by Salmonella and the S. typhimurium serotype can be readily identified. The sensing platform with ingenious design paves a new way for label-free, multi-target simultaneous detection, whose advantage of rapidity, sensitivity, cost-effectiveness, and specificity also lay a solid foundation for practical applications.

Abstract:Salmonella is one of the most common causes of food-associated disease. An electrochemical biosensor was developed for Salmonella detection using a Salmonella-specific recognition aptamer. The biosensor was based on a glassy carbon electrode modified with graphene oxide and gold nanoparticles. Then, the aptamer ssDNA sequence could be linked to the electrode. Each assembly step was accompanied by changes to the electrochemical parameters. After incubation of the modified electrode with Salmonella, the electrochemical properties between the electrode and the electrolyte changed accordingly. The electrochemical impedance spectrum was measured to quantify the Salmonella. The results revealed that, when more Salmonella were added to the reaction system, the current between the electrode and electrolyte decreased; in other words, the impendence gradually increased. A detection limit as low as 3 cfu/mL was obtained. This novel method is specific and fast, and it has the potential for real sample detection.

Abstract:Salmonella is a principal causal agent of pathogenic outbreaks via food. A universal, highly sensitive and visual Salmonella detection method was proposed in this paper, based on a universal linker PCR (UL-PCR)-triggered Strand Displacement Amplification (SDA). In this research, the UL-PCR achieved the primary amplification. The universal linker primer was ingeniously designed and composed of two parts, one of which was the binding sequence of the target, and the other was the universal linker. Complementary sequences of the G-quadruplex and the nicking endonuclease recognition sequence were included in the universal linker. Therefore, the G-quadruplexes and nicking sites were successfully introduced into the UL-PCR products, providing a basis for further SDA triggering. SDA achieved the secondary signal amplification and generated a large amount of label-free DNAzymes. Following SDA, DNAzymes catalyzed 3,3',5,5'-tetramethylaniline (TMB) into colored compounds visible to the naked eye. We obtained the best experimental conditions by univariate analysis. Under optimal conditions, this proposed universal label-free method could detect Salmonella genome at level as low as 22 copies mL-1, with an excellent linear range between 102 copies mL-1 and 107 copies mL-1. And the limit of quantification (LOQ) was 102 copies mL-1. This strategy shows promise for broad applications.

A robust and facile colorimetric aptasensor for the detection of Salmonella Typhimurium based on the regulation of Fe3O4@Cu@PCPy yolk-shell nanozyme activity

Abstract:Due to their superparamagnetism and enzyme-like activity, iron oxide (Fe3O4) nanozymes can be readily used for sample pretreatment and the generation of detection signals, and have, thus, attracted much attention in the field of bioanalysis and diagnosis. However, the low catalytic activity of Fe3O4 nanozymes does reduce the sensitivity of Fe3O4-based methods, limiting their application. In this study, Fe3O4@Cu@poly(pyrrole-2-carboxylic acid) yolk-shell nanozymes (Fe3O4@Cu@PCPy YSNs) were synthesized using a facile approach and selective chemical etching technology. Compared with Fe3O4 nanozymes, the Fe3O4@Cu@PCPy YSNs demonstrated a three-fold increase in the peroxidase-like activity, good dispersity and strong superparamagnetism. In addition, the flower-shaped structure of aptamer-complementary strand (Apt-CS) conjugates was designed on the surface of the Fe3O4@Cu@PCPy YSNs, which effectively inhibited their peroxidase-like activity by creating a physical barrier that hindered the access of substrates to the center of the Fe3O4@Cu@PCPy YSNs. Based on this principle, a robust and facile colorimetric aptasensor was developed for detecting Salmonella Typhimurium. The flower-shaped Apt-CS were dissociated in the presence of S. Typhimurium, promoting the recovery of Fe3O4@Cu@PCPy YSN catalytic activity. Under optimized conditions, this proposed aptasensor successfully detected S. Typhimurium in a linear range of 3 to 3 × 106 CFU/mL, achieving a detection limit of 1 CFU/mL. Finally, the feasibility of this novel aptasensor was further validated by three actual samples, with recoveries of between 84.3% and 102%, thereby demonstrating the huge potential of the proposed aptasensor for detecting S. Typhimurium in foods.

Abstract:A method based on surface plasmon resonance (SPR) DNA biosensor has been developed for label-free and high-sensitive detection of Salmonella. A biotinylated single-stranded oligonucleotide probe was designed to target a specific sequence in the invA gene of Salmonella and then immobilized onto a streptavidin coated dextran sensor surface. The invA gene was isolated from bacterial cultures and amplified using a modified semi-nested asymmetric polymerase chain reaction (PCR) technique. In order to investigate the hybridization detection, experiments with different concentration of synthetic target DNA sequences have been performed. The calibration curve of synthetic target DNA had good linearity from 5 nM to 1000 nM with a detection limit of 0.5 nM. The proposed method was applied successfully to the detection of single-stranded invA amplicons from three serovars of Salmonella, i.e., Typhimurium, Enterica and Derby, and the responses to PCR products were related to different S. typhimurium concentrations in the range from 10(2) to 10(10) CFU mL(-1). While with this system to detect E. coli and S. aureus, no significant signal was observed, demonstrating good selectivity of the method. In addition, the hybridization can be completed within 15 min, and the excellent sensor surface regeneration allows at least 300 assay cycles without obvious loss of performance.
A multimetallic nanozyme enhanced colorimetric biosensor for Salmonella detection on finger-actuated microfluidic chip

Abstract:Salmonella screening is essential to avoid food poisoning. A simple, fast and sensitive colorimetric biosensor was elaborately developed for Salmonella detection on a microfluidic chip through limiting air chambers for precise air control, switching rotary valves for accurate fluid selection, a convergence-and-divergence passive micromixer and an extrusion-and-suction active micromixer for efficient fluid mixing, and immune gold@platinum palladium nanocatalysts for effective signal amplification. The mixture of bacteria, immune magnetic nanobeads and nanocatalysts was first rapidly mixed to form nanobead-bacteria-nanocatalyst conjugates and magnetically separated for enrichment. After washing with water, the conjugates were used to catalyze colorless substrate and blue product was finally analyzed using ImageJ for quantifying bacterial concentration. The finger-actuated microfluidic chip enabled designated control of designated fluids in designated places towards designated directions by simple press-release operations on designated air chambers without any external power. Under optimal conditions, this sensor could detect Salmonella at 45 CFU/mL in 25 min.
A dual-mode composite nanozyme-based cascade colorimetric-fluorescence aptasensor for Salmonella Typhimurium detection

Abstract:Background Salmonella Typhimurium poses a serious threat to human health worldwide, necessitating the development of a rapid, sensitive, and convenient method for S . Typhimurium detection. Nanozymes are considered ideal signal report elements, which are extensively used for developing colorimetric methods. However, single-component nanozymes display low catalytic activity, and colorimetric methods are susceptible to environmental interference, reducing the sensitivity and accuracy of detection results. To address these drawbacks, this study constructed a dual-mode composite nanozyme-based cascade colorimetric-fluorescence aptasensor for S . Typhimurium detection in food. Results In this study, the composite Fe 3 O 4 @MIL-100(Fe) nanozymes were successful synthesized and demonstrated substantial peroxide-like activity, with 4.76-fold higher specificity activity (SA) than that of Fe 3 O 4 nanozymes. Then, a glucose oxidase (GOx)-Fe 3 O 4 @MIL-100(Fe) cascade reaction was developed for colorimetric detection via an aptamer to facilitate the formation of Fe 3 O 4 @MIL-100(Fe)/ S . Typhimurium/carboxylated g-C 3 N 4 (CCN)-GOx sandwich complexes. Meanwhile, the fluorescence mode was achieved by measuring the fluorescence intensity of the sandwich complexes. In optimum conditions, the dual-mode detection limits (LOD) were 1.8 CFU/mL (colorimetric mode) and 1.2 CFU/mL (fluorescence mode), respectively, with the S . Typhimurium concentration ranging from 10 CFU/mL to 10 7 CFU/mL. Finally, the feasibility of the dual-mode colorimetric-fluorescence method was validated via three actual samples, yielding recovery rates of 77.32 % to91.17 % and 82.17 % to 103.7 %, respectively. Significance and novelty This study successfully develops a composite nanozyme-based cascade colorimetric and fluorescence dual-mode aptasensor for S . Typhimurium detection. It presents several distinct benefits, such as a broader linear range (10–10 7 CFU/mL), a lower LOD value (1.2 CFU/mL), and more accurate results. More importantly, the proposed dual-mode method displays a low LOD in colorimetric mode, demonstrating considerable potential for S . Typhimurium on-site detection in food.

Abstract:Early screening of Salmonella is of utmost importance to minimize the incidence of related foodborne diseases. Here, we reported an immunosensor to fast detect Salmonella using Fe-MIL-88NH2 metal–organic framework (Fe-MOF) nanocubes to load platinum nanoparticles (PtNPs) for signal amplification and smartphone thermal imaging to monitor the temperature change. The Fe-MOF nanocubes decorated with PtNPs (Fe-MOF/PtNPs) were prepared as peroxidase mimics to catalyze hydrogen peroxide (H2O2) to produce O2. The immune magnetic nanoparticles (MNPs), the target bacteria, and the immune Fe-MOF/PtNPs were first mixed by the three-dimensional (3D) chaotic micromixer and conjugated in the microfluidic chip to form MNP–bacteria–Fe-MOF/PtNPs sandwich complexes. After magnetic separation and washing, H2O2 was injected and decomposed by Fe-MOF/PtNPs on the sandwich complexes to generate O2 in a sealed chamber. The increase in the pressure pushed the preloaded water to react with calcium oxide powder, resulting in the generation of heat, which was measured using the smartphone-based thermal sensor to determine the bacteria. An Architecture for Optimized Inter-Technology Handovers: Experimental Study

Abstract:With the large number of access technologies and the emergence of user mobility, the need to "access the world", anywhere and anytime has gained a significant importance. It is essential to ensure the quality of the services when moving between access networks and technologies in a transparent manner. This paper presents a new architecture that enables seamless handovers in wireless heterogeneous environments. Towards this aim, the proposed architecture combines the MIPv6 mobility management protocol, the IEEE 802.21 Media Independent Handover (MIH) framework, and a mobility control entity responsible for controlling the handover decisions in a make-before-break approach. In order to validate and evaluate this new solution, a real testbed was developed, combining Wi-Fi, WiMAX and 3G technologies. The results obtained with this experimental platform cover the analysis of the handover processes, and important aspects as the handover performance, QoS and QoE of the services while handover is taking place. The results show the significant benefits of the proposed approach.
