Abstract:In recent years, quartz crystal microbalance (QCM) plays an important role in many biological detections, and the enhancement of a single strain DNA probe linked to the sensor surface is of crucial importance in DNA molecule recognition. By means of nanogold modification and mass amplification, a QCM biosensor for Escherichia coli O157:H7 DNA detection was explored and developed in this work. As a suitable surfactant, 1,6-Hexanedithiol was introduced onto the Au surface of QCM, then self-assembled to provide a thioled interface for the inner goldnano immobilization. Based on the Au-SH binding mechanism, thiolated single-stranded DNA (ssDNA) probes which was specific to E. coli O157:H7 eaeA gene would link to the nanoparticle-modified electrode surface. The hybridization was induced by exposing the ssDNA probe to the complementary target DNA, but the frequency change caused by the captured target DNA was negligible. In order to amplify the signal, outer nanogold as the mass enhancer were applied to combine with the target DNA by the biotin-avidin system. The electrochemical techniques, cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS), are adopted to manifest and character each step, which supplied some reasonable and powerful supports for the frication and detection of this QCM biosensor. The target DNA corresponding to 2.0 times 103 colony forming unit (CFU)/ml E. coli O157:H7 can be detected by this biosensor, so it is practical to develop a sensitive and effective QCM biosensor for pathogenic bacteria detection based on specific DNA analysis. In order to attract more attention, this DNA biosensor should be combined with some micro- and nano-fabrication techniques to realize more promising and pratical applications, and more attention should be focused on the further improvement of the sensitivity and the shorten of analysis time.
Dual DNA recycling amplifications coupled with Au NPs@ZIF-MOF accelerator for enhanced electrochemical ratiometric sensing of pathogenic bacteria

Abstract:Sensitive and accurate quantification of pathogenic bacteria is vastly significant to the related food safety. Herein, a sensitive ratiometric electrochemical biosensor was developed for the detection of Staphylococcus aureus ( S. aureus ) based on dual DNA recycling amplifications and Au accelerator. Gold nanoparticles-loaded Zeolitic imidazolate metal-organic framework (Au ) as electrode substrate possessed a large specific surface area for nucleic acid adsorption, and as an accelerator promoted the transfer of electrons. The strong recognition of aptamer to target S. aureus could initiate the padlock probe-based exponential rolling circle amplification (P-ERCA, as the first DNA recycling amplification), generating large numbers of trigger DNA strands. The released trigger DNA further activated the catalytic hairpin assembly (CHA, as the second DNA recycling amplification) on electrode surface. Consequently, P-ERCA and CHA continuously brought about one target to many signal transduction, leading to an exponential amplification. To achieve the accuracy of detection, the signal ratio of methylene blue (MB) and ferrocene (Fc) (I MB /I Fc ) was applied for intrinsic self-calibrating. Taking advantages of dual DNA recycling amplifications and Au , the proposed sensing system displayed high sensitivity for S. aureus quantification with a linear range of 5–10 8 CFU/mL, and the limit of detection was 1 CFU/mL. Moreover, this system represented excellent reproducibility, selectivity, and practicability for S. aureus analysis in foods.

Abstract:In this work, a novel surface-enhanced Raman scattering (SERS) sandwich strategy biosensing platform has been established for simultaneously detecting Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ). Fe 3 O 4 @SiO 2 -Au nanocomposites (NCs) with varying amounts of Au nanocrystals were prepared, and the effect of interparticle gaps on SERS activity was studied by finite-difference time-domain (FDTD) method. The optimal magnetic SERS-active substrates (FS-A5) were functionalized with the specific aptamers to act as capture probes. Meanwhile, graphene oxide-Au nanostars (GO-Au NSs) decorated with Raman reporters and aptamers were used as SERS tags. The loading density of Au NSs on GO was tuned to change the number of SERS active sites. In this proposal, E. coli and S. aureus were first captured by capture probes and then bound with SERS tags to form a sandwich-like structure, which caused enhanced electromagnetic field because of the dual enhancement strategy. Under optimal conditions, SERS platform could detect E. coli and S. aureus simultaneously, and the detection limit was as low as 10 cfu/mL. Our sandwich assay-based dual-enhanced SERS platform provides a new idea for simultaneously detecting multiple pathogens with high selectivity and sensitivity, and thus will have more hopeful prospects in the field of food safety.

Abstract:Food-borne pathogens are one of the leading causes of food poisoning, which vigorously affect food safety and human health. Therefore, the development of early and rapid detection methods for food pollution evaluation is the key to food safety and quality control. Herein, a simple and inexpensive photoelectrochemical (PEC) sensor is developed for highly selective and ultrasensitive detection of Staphylococcus aureus (S. aureus). The technique is based on "signal-off" that employs Cu-C3N4-TiO2 heterostructures as photoactive materials and monolayer Cu-C3N4 nanozyme as a signal amplifier. In the presence of S. aureus, the aptamer-modified Cu-C3N4 (Cu-C3N4@Apt, a signal amplifier) and S. aureus were specifically anchored on the surface of the ligand-modified photoelectrode. The Cu-C3N4@Apt nanozyme acted as a peroxidase to catalyze the oxidation of 4-chloro-1-naphthol (4-CN) to produce insoluble precipitate on the electrode surface and resulted in a significant decrease in photocurrent. Based on the signal-amplification by the Cu-C3N4@Apt nanozyme, the constructed PEC sensor demonstrated a wide linear range between 10-108 CFU/mL for the S. aureus detection with the detection limit (LOD) as low as 3.40 CFU/mL. Furthermore, the PEC sensor was capable of determining S. aureus in spiked orange juice and milk, with the recovery of 91%-113%, indicating the reliability of the sensor for S. aureus detection in real samples. This investigation provides a feasible strategy for the design of highly selective and ultrasensitive PEC sensors to determine analytes in complex systems.

Post‐extraction implant placement into infected versus non‐infected sites: A multicenter retrospective clinical study

Abstract:PURPOSE The aim of this study was to assess whether immediate implant placement into post-extraction sites presenting a chronic infection poses a greater risk of implant failure than immediate placement in non-infected sites. MATERIALS AND METHODS Records of patients who underwent extraction and immediate implant placement into both infected and non-infected sites from January 1998 to September 2014 at 5 different dental centers were considered for inclusion. Included records were subjected to statistical analysis of survival rates, along with a number of other patient-, implant-, surgery-, and prosthesis-related variables. RESULTS The inclusion criteria were met by 369 patients who received a total of 527 implants. The follow-up averaged 53.2 months (range 0.9-158.3) for implants placed into non-infected sockets (N = 334) and 50.1 months (range 1.6-146.1) for those placed into infected sites (N = 193). Seven implants failed in non-infected sites and 3 in infected ones. All failures occurred within 1 year of placement. Cumulative implant survival rate for non-infected and infected sites was, respectively, 97.9% ± 0.8% and 98.4% ± 0.9%, being not significantly different (P = .66). None of the investigated variables affected the outcome. CONCLUSIONS Placement of implants into periodontally or endodontically infected sites immediately after tooth extraction is a safe option, even when the implants are loaded immediately or early.

Abstract:Healthcare-associated infections (HAIs) have increasingly become a threat to global health. Rapid, accurate, and reliable detection of HAI-related bacteria is urgently needed. Here, we presented a novel antimicrobial peptide-based surface-enhanced Raman scattering lateral flow assay (SERS-LFA) for the detection of multiple HAI-related bacteria. Antimicrobial peptide cecropin 1(CP1) was immobilized on the surface of Fe 3 O 4 @Au by the streptavidin-biotin system to capture Pseudomonas aeruginosa ( P. aeruginosa ) and Escherichia coli O157:H7 ( E. coli O157:H7) simultaneously. The synthesized Fe 3 O 4 @Au-CP1 exhibits excellent broad-spectrum capturing ability, rapid magnetic enrichment function, and superior antibacterial activity. LFA strips enable the specific detection of two target bacteria in a quick and simple manner, with bacterial type differentiation via corresponding monoclonal antibodies sprayed. The highly sensitive SERS signals and the amplification effect of streptavidin-biotin significantly improved the sensitivity of detection. The LODs of P. aeruginosa and E. coli O157:H7 were 12 and 16 cells/mL, respectively. This assay also exhibited good specificity and reproducibility when applied to urine samples. Therefore, Fe 3 O 4 @Au-CP1-based SERS-LFA has great potential for detecting multiple bacteria after infection as well as regularly monitoring therapeutic efficacy.

Abstract:A highly structured SERS biosensor was proposed for Staphylococcus aureus ( S. aureus , ATCC 29213) detection. Polydimethylsiloxane (PDMS) film was chemically modified and gold and silver core–shell nanoparticles (Au@Ag NPs) were self-assembled on it. Sulfhydryl-modified aptamer of S. aureus was immobilized on the SERS substrate to form a capture substrate and the aptamer modified with 6-carboxy-x-rhodamine (ROX) was used as a signal molecular probe. The aptamer and target constructed a sandwich structure "capturing substrate-target-signal probe". Concurrently, the bacterial surface was covered with Au@Ag NPs which resulted in more "hot spots" and improved the detection sensitivity. The SERS intensity was plotted against the logarithmic concentration of S. aureus (3.6 × 10 to 3.6 × 10 8 cfu/mL) and achieved an ultra-low detection response of 16 cfu/mL. S. aureus was detected in tap water, milk and fish samples with satisfactory recoveries (93.25%∼106.42%) and validation results ( p > 0.05). Significantly, the method offered a speedy and accurate S. aureus examining in food/environmental settings.

Abstract:The accurate and sensitive detection of foodborne pathogens, which is critical for timely food quality supervision and human health. To address this issue, herein, we developed a simple and novel surface enhanced Raman scattering (SERS) assay, using p-mercaptobenzoic acid (MBN) modified gold nanoparticles (Au NPs) and magnetic beads, for interference-free detection of escherichia coli (E.coli). The assay technique cleverly reduced silver ions (Ag+) on the surface of E.coli (bacteria@Ag NPs), and the functionalized magnetic beads (capture probes) captured and enriched bacteria@Ag NPs, forming the structure of capture probes-bacteria@Ag NPs, then capture probes-bacteria@Ag NPs was dissolved in the acidic medium, the Ag NPs on the surface of E.coli was converted to Ag+ again. Due to the special coordination between Ag+ and MBN modified Au NPs (functionalized Au NPs), the SERS intensity of MBN exhibited positive correlation with the E.coli concentration, the SERS detection assay of E.coli was eatablished. The signal of functionalized Au NPs located at 2228 cm-1, can perfectly avoided the spectral overlap with coexisting materials in Raman fingerprint region, which ensured the accuracy of the technique; The controlled aggregation of functionalized Au NPs ensured the reproducibility and reliability of the detection system; The emergence of MBs, greatly reduced the reaction time, make sure the operation be rapid, simple and portable. The limit of detection (LOD) for E.coli was as low as 10 cfu/mL, the detection assay was also successfully applied to the detection of E.coli in bottled water and milk. As a sensitive and accurate analytical technique for detection of pathogens, this SERS based method had great potential to be applied in the field of food safety.

Abstract:Surface-enhanced Raman spectroscopy (SERS) has been successfully applied to detect various biomolecules, but it is still in challenge to assay living cells or bacteria sensitively, selectively and quantitatively in complex environments. In this paper, 4-ATP and DTNB are assembled on Ag nanoparticle (NP) -decorated poly (styrene-co-acrylic acid) (PSA) nanospheres and then sealed by silica shells to form sensitive SERS labels based on the localized surface plasmon resonance of Ag NPs and large light scattering cross-sections of PSA nanospheres. They are further developed as encoding tags for dual detection of S. aureus and E. coli after assembling corresponding aptamers, which demonstrate ultralow detection limits of 8 cell L-1 for S. aureus and 2 cell L-1 for E. coli. Such a bioassay indicates a point-of-care strategy of ultrasensitively biomedical detections by encoding specific SERS tags.

Abstract:Staphylococcus aureus (S. aureus) is one of the most common foodborne pathogens worldwide, which poses a great threat to public health. It is of utmost importance to develop rapid, simple, and sensitive methods for the determination of S. aureus. A signal-on photoelectrochemical (PEC) aptasensor is constructed herein based on titanium carbide (Ti3C2Tx)-Au nanobipyramids (NBPs)/ZnO nanoarrays (NRs). The reliability and capability of the PEC aptasensor make it suitable for the sensitive and selective determination of S. aureus. First, the electrostatically self-assembled Ti3C2Tx-Au NBP nanomaterial was coated on the ZnO NR surface by a spin-coating method. On the one hand, Ti3C2Tx-Au NBPs can broaden the spectral absorption of ZnO NRs, resulting in Ti3C2Tx-Au NBPs/ZnO NR composites that exhibit a wide range of absorption from the ultraviolet to the infrared region. On the other hand, Ti3C2Tx can reduce the agglomeration of nanoparticles, while Au NBPs can effectively fix the aptamer through the Au-S bond. Specifically, the experimental results show that when S. aureus is present, the Au NBPs-aptamer-S. aureus complex is shed from the electrode surface, altering the interfacial electron transfer model and reducing the steric hindrance. Consequently, an amplified photocurrent signal for the quantitative determination of S. aureus is obtained. Under optimal experimental conditions, a linear correlation is observed between the current response of the aptasensor and the logarithm of the S. aureus concentration (ranging from 1.0 to 1.0 × 106 CFU/mL), with an impressive detection limit as low as 0.5 CFU/mL. Furthermore, the aptasensor has been successfully employed for the detection of S. aureus in milk, with the recovery of 93.0%-99.0%. Hence, this research offers a novel approach for the detection of foodborne pathogens and other noxious substances.

Abstract:Staphylococcus aureus ( S. aureus ) is a widespread pathogenic bacterium, causing foodborne disease and posing threat to public safety. Thus, it is of great importance for rapid and effective determination of S. aureus . In this protocol, we synthesized a smart DNA-modified Au/CuMOF bioconjugate material (ssDNA-Au/CuMOF) with multiple functions of biorecognition, electrochemical and catalytical activity, and explored its application in dual-mode determination of S. aureus . Magnetic bead (MB) modified with aptamers (MB-Apt) was employed for specific S. aureus capture. The incubation of ssDNA-Au/CuMOF and MB-Apt with different concentration of S. aureus led to the difference in quantity of ssDNA-Au/CuMOF confined on MB or released in supernatant. After magnetic separation, dual-mode detection was realized by the electrochemical measurement and the material-catalyzed colorimetry. This method demonstrated a satisfied analytical performance with a broad linear range from 10 to 10 8 CFU mL −1 , a colorimetric minimum resolution as low as 48 CFU mL −1 , an electrochemical detection limit of 5 CFU mL −1 , and good selectivity in distinguishing target from non-target bacteria. Its application in detecting S. aureus in complex samples also showed good response, indicating the great potential for S. aureus detection in real samples and providing a methodological and technical support for food monitoring.

Abstract:Human health is greatly threatened by bacterial infection, which raises the risk of serious illness and death in humans. For early screening and accurate treatment of bacterial infection, there is a strong desire to undertake ultrasensitive detection and effective killing of pathogenic bacteria. Herein, a novel surface-enhanced Raman scattering (SERS) biosensor based on sandwich structure consisting of capture probes/bacteria/SERS tags was established for specific identification, capture and photothermal killing of Escherichia coli (E. coli). Finite-difference time-domain (FDTD) technique was used to simulate the electromagnetic field distribution of capture probes, SERS tags and sandwich-type SERS substrate, and a possible SERS enhancement mechanism based on sandwich structure was presented and discussed. Sandwich-type SERS biosensor successfully achieved distinctive identification and magnetic beneficiation of E. coli. In addition, a single SERS substrate, including capture probes and SERS tags, could also achieve outstanding photothermal effects as a consequence of localized surface plasmon resonance (LSPR) effect. Intriguingly, sandwich-type SERS biosensor demonstrated a higher photothermal conversion efficiency (50.03 %) than the single substrate, which might be attributed to the formation of target bacterial clusters. The superior biocompatibility and the low toxicity of the sandwich-type biosensor were confirmed. Our approach offers a fresh method for constructing sandwich-type biosensor with multiple SERS hotspots based on extremely effective hybrid plasmonic nanoparticles, and has a wide range of potential applications in the recognition and treatment of bacteria.

Abstract:Food safety represents a critical global public health issue, with safety challenges posed by foodborne pathogens garnering extensive attention. Therefore, we introduce a co-recognition, enrichment and sensing (CES) all-in-one strategy for analysis of bacteria with low background and high specificity. This method employs antimicrobial peptide (AMP) functionalized magnetic nanoparticles (MNPs) to enrich bacteria and uses aptamer@Au@PBA (K x MFe(CN) 6 (M = Pb and Ni)) NPs as silent SERS tags. When both S. aureus and E. coli O157:H7 are present, the silent SERS probes could specifically label the target bacteria, forming a sandwich-like structure. This binding induces silent Raman shifts (2139 cm −1 and 2197 cm −1 ), enabling quantification of two bacteria. Coupling with the modular flexible microfluidics and magnetic control slider device, this platform facilitates rapid switching between magnetic loading and elution. The CES SERS method demonstrated linear relationships for both S. aureus and E. coli O157:H7 at 50–1600 cfu mL −1 , with detection limits of 14 and 18 cfu mL −1 , respectively. The method achieved recovery rates of 85.6–112% and relative standard deviations of 1.5–8.6%. Validation using the ELISA method revealed relative errors between −7.5 and 4.3%. The CES approach has potential applications in food safety, environmental monitoring, and biomedical diagnosis.

Abstract:In this study, we introduced a Raman detection technique based on a combination of functionalized magnetic beads and surface-enhanced Raman scattering (SERS) tags to develop a rapid and sensitive strategy for the detection of Staphylococcus aureus (S. aureus), a typical foodborne pathogen. Polyethylene glycol (PEG) and bovine serum albumin (BSA) dual-mediated teicoplanin functionalized magnetic beads (TEI-BPBs) were prepared for separation of target bacteria. SERS tags were used to immobilize antibodies on gold surfaces with bifunctional linker proteins to ensure specific recognition of S. aureus. Under optimal conditions, the combination of TEI-BPBs and SERS tags showed reliable performance, exhibiting good capture efficiency even in the presence of 106 CFU mL-1 of non-target bacteria. The SERS tag provided an effective hot spot for subsequent Raman detection, presenting good linearity in the range of 102-107 CFU mL-1. Good performance has also been shown in detecting target bacteria in milk samples, where it has a recovery of 95.5-101.3%. Thus, the highly sensitive Raman detection technique combined with TEI-BPBs capture probes and SERS tags is a promising method for the detection of foodborne pathogens in food or clinical samples.

Abstract:New strategy of a rapid, sensitive bacteria detection is highly desirable. Constructing a dual-mode colorimetric/surface-enhanced Raman scattering (SERS) biosensor is key to obtain highly sensitive discriminating and detecting of different pathogen bacteria with high accuracy. Herein, we developed a multifunctional colorimetric catalytic nanozyme ( MOF) composed of AuAg-doping Prussian Blue analogue-based metal-organic framework (MOF) that fabricated by self-assembled sheet growth of Ag-etched silver-coated gold nanoparticles ( NPs) mediated by addition of ferricyanide (K 3 [Fe(CN) 6 ]). MOF displays ultrahigh peroxidase-mimicking activity, enhanced colorimetric signal brightness, and notably SERS enhancement. Through optimizing its Fe-doping level, peroxidase-like activity of MOF is approximately 1900-fold higher than that of horseradish peroxidase (HRP), while its 3-fold enhancement in SERS signal compared with that of gold nanoparticles (Au NPs). Integrated with nanozyme-based sandwich system that made of bacteria/4-MPBA (4-mercaptophenylboronic acid)/AuA MOF, this dual-modal biosensor achieves a highly sensitive colorimetric assay of bacteria with a lower limit of detection (6 CFU/mL), and simultaneously discriminate E. coli and S. aureus by bacterial "fingerprints" spectra. Furthermore, this proposed colorimetric assay was combined with a smartphone readout-sensing platform for determining bacteria with ultra-sensitivity (2 CFU/mL). This work exhibits a novel colorimetric nanozyme used in dual-mode colorimetric/SERS sensing of bacteria.

Abstract:Foodborne pathogens, especially bacteria, are explicitly threatening public health worldwide. Biosensors represent advances in rapid diagnosis with high sensitivity and selectivity. However, multiplexed analysis and minimal pretreatment are still challenging. We fabricate a gold nanoparticle (Au NP)-amplified microcantilever array biosensor that is capable of determining ultralow concentrations of foodborne bacteria including Escherichia coli O157:H7, Vibrio parahaemolyticus, Salmonella, Staphylococcus aureus, Listeria monocytogenes, Shigella, etc. The method is much faster than using conventional tools without germiculturing and PCR amplification. The six pairs of ssDNA probes (ssDNA1 + ssDNA2 partially complementary to the target gene) that originated from the sequence analysis of the specific gene of the bacteria were developed and validated. The ssDNA1 probes were modified with -S-(CH2)6 at the 5′-end and ready to immobilize on the self-assembled monolayers (SAMs) of the sensing cantilevers in the array and couple with Au NPs, while 6-mercapto-1-hexanol SAM modification was carried out on the reference cantilevers to eliminate the interferences by detecting the deflection from the environment induced by non-specific interactions. For multianalyte sensing, the target gene sequence was captured by the ssDNA2-Au NPs in the solution, and then the Au NPs-ssDNA2-target complex was hybridized with ssNDA1 fixed on the beam of the cantilever sensor, which results in a secondary cascade amplification effect. Integrated with the enrichment of the Au NP platform and the microcantilever array sensor detection, multiple bacteria could be rapidly and accurately determined as low as 1–9 cells/mL, and the working ranges were three to four orders of magnitude. There was virtually no cross-reaction among the various probes with different species. As described herein, it holds great potential for rapid, multiplexed, and ultrasensitive detection in food, environment, clinical, and communal samples.

A multiplex nanoparticles-based DNA electrochemical biosensor for the simultaneous detection of Escherichia coli O157:H7 and Staphylococcus aureus

Abstract:A B S T R A C T In this study, we evaluated the sensitivity of a multiplex nano particle-based DNA electrochemical biosensor for the simultaneous multiple detection of gene of E. coli O157:H7 and the nuc gene of S. aureus. The biosensor system was mainly composed of a sandwich structure: fixing probe (fDNA), target DNA (tDNA) and detective probe (dDNA). The fixing probe carries the sulfhydryl group strongly binded with the gold electrode and it was the complementary sequence of one end of the tDNA. The dDNA was complementary sequence of another end of the tDNA, which binds with the nanoparticle tracers (NTs), such as lead sulfite (PbS) and cadmium sulfite (CdS), that act as a signal reporter and amplifier. The sandwich structures were immersed in 1 M nitric acid, thereby dissolving the PbS and CdS. Another gold electrode was used to detect NTs ions by Square wave voltammetry (SWV). The results show that the detection limits of this multiplex DNA biosensor are 1×10 -12 mol/L of the gene of E. coli O157:H7 using PbS, and 1×10 -12 mol/L of the nuc gene of S. aureus, using CdS NTs respectively. The multiplex nanoparticles-based DNA electrochemical biosensor has potential application in rapid detection of multiple food -borne pathogens.
