Abstract:Early infection and peri-implantitis after implant restoration are major reasons for dental implant failure. Implant-associated infections are majorly attributed to biofilm formation. In this study, co-incorporated zinc- (Zn-) and strontium- (Sr-) nanorod coating on sandblasted and acid-etched (SLA) titanium (SLA-Zn/Sr) was fabricated by hydrothermal synthesis. It was aimed at promoting osteogenesis while inhibiting biofilm formation. The nanorod-like particles (phi 30-50 nm) were found to be evenly formed on SLA-Zn/Sr (Zn: 1.49 +/- 0.16 wt%; Sr: 21.69 +/- 2.74 wt%) that was composed of well-crystallized ZnTiO3 and SrTiO3 phases. With a sufficient interface bonding strength (42.00 +/- 3.00 MPa), SLA-Zn/Sr enhanced the corrosion resistance property of titanium. Besides, SLA-Zn/Sr promoted the cellular initial adhesion, proliferation and osteogenic differentiation of rBMSCs in vitro while inhibiting the adhesion of Staphylococcus aureus and Porphyromonas gingivalis . In addition, through down-regulating icaA gene expression, this novel surface reduced the secretion of polysaccharide intercellular adhesion (reduced by 87.9% compared to SLActive) to suppress the S. aureus biofilm formation. We, therefore, propose a new chemical modification on titanium for multifunctional implant material development. Due to the Zn/Sr co-doping in coating, material properties, early osteogenic effect and antibacterial ability of titanium can be simultaneously enhanced, which has the potential to be applied in dental implantation in the future.

Abstract:Background For dental implants, early infection and peri-implantitis after implant restoration are major reasons for implant failure. Plaque biofilm formation is the initiator for implant-related infection. Therefore, it is urgent to develop a new antibacterial implant materials or treatment methods. Aim/Hypothesis In this study, Sr- and Zn- incorporated micro nano-structured titanium surface with nanorods (MNT-Sr Zn) was fabricated by a two-step hydrothermal process, aiming to improve osseointegration and reduce implant failure caused by implant-related infection and broaden new ideas for the future research Material and Methods The Mod Ti surfaces were used as a control. The characterization of different surfaces was detected by a field emission scanning electron microscope (FE-SEM) equipped with energy dispersive spectra (EDS) detector, x-ray diffractometer (XRD) and inductively coupled plasma mass spectroscopy (ICP-MS). Meanwhile, Staphylococcus aureus (S. aureus) was selected to evaluate antibacterial properties through Live Dead Staining and watching the adhesive bacterial morphology under FE-SEM. What's more, rat bone marrow-derived mesenchymal stem cells (rBMSCs) were cultured on the surfaces to detect their early osteogenic properties, like the alkaline phosphatase(ALP) activity and the expressions of osteogenesis related genes. Results The results confirmed that nanorod-like particles with a diameter of about 30-50 nm of MNT-Sr Zn composed of well-crystallized ZnO and SrTiO3 phases. In the EDS spectra, the elements of Zn and Sr were evenly distributed, and approximately 1.49 ± 0.16 wt% and 21.69 ± 2.74 wt% respectively. Besides, the release of Sr2+ and Zn2+ was a controlled and long-term process. More importantly, Zn-containing nanorod-like particles and the morphology could work together, slightly decreasing adhesion of S. aureus but increasing the proportion of dead cells, thus inhibiting subsequent biofilm formation, compared with the controlled group. In addition to this, MNT-Sr Zn could enhance the osteogenic differentiation via the upregulation of the transcription of osteogenic related genes and the expression of ALP in vitro. Conclusion and Clinical Implications This research provides a new surface modification method on titanium substrate for multi-functional implant material development. Due to the co-doping of Sr and Zn in the coating, biomechanical stability, antibacterial capability and early osteogenic inductive effect are enhanced, which has the potential to be applied in dental implantation in the future.

Abstract:Titanium (Ti) has been commonly used as an implant material in dentistry and bone surgery for several decades. Meanwhile, surface modification of titanium can enhance the osseointegration of implants. In this study, a graphene oxide/chitosan/hydroxyapatite (GO/CS/HA) composite coating was fabricated by electrophoretic deposition on Ti substrates. Subsequently, the surface morphology, phase composition, wettability, and bonding strength of this composite coating were researched. Additionally, in vitro cytological examination was performed, including evaluations of cell adhesion, cell viability, cell differentiation, cell mineralization, and osteogenetic factor expression. Finally, the in vivo osteogenetic properties were evaluated through an animal study, including a histological analysis, a microcomputed tomography, and biomechanical tests. The results showed that a homogeneous and crack-free GO/CS/HA composite coating was coated on Ti, and the wettability and bonding strength of the GO/CS/HA composite coating were enhanced compared with HA, GO/HA, and CS/HA coatings. Furthermore, the GO/CS/HA coating greatly heightened the cell-material interactions in vitro. Additionally, this GO/CS/HA-Ti implant could enhance osseointegration in vivo. Consequently, GO/CS/HA-Ti may have potential applications in the field of dental implants. © 2018 Wiley Periodicals, Inc. In Vitro Biological Characterization of Silver-Doped Anodic Oxide Coating on Titanium

Abstract:Despite the high biocompatibility and clinical effectiveness of Ti-based implants, surface functionalization (with complex osteointegrative/antibacterial strategies) is still required. To enhance the dental implant surface and to provide additional osteoinductive and antibacterial properties, plasma electrolytic oxidation of a pure Ti was performed using a nitrilotriacetic acid (NTA)-based Ag nanoparticles (AgNP)-loaded calcium–phosphate solution. Chemical and structural properties of the surface-modified titanium were assessed using scanning electron microscopy (SEM) with energy dispersive X-ray (EDX) and contact angle measurement. A bacterial adhesion test and cell culture biocompatibility with collagen production were performed to evaluate biological effectiveness of the Ti after the plasma electrolytic process. The NTA-based calcium–phosphate solution with Ag nanoparticles (AgNPs) can provide formation of a thick, porous plasma electrolytic oxidation (PEO) layer enriched in silver oxide. Voltage elevation leads to increased porosity and a hydrophilic nature of the newly formed ceramic coating. The silver-enriched PEO layer exhibits an effective antibacterial effect with high biocompatibility and increased collagen production that could be an effective complex strategy for dental and orthopedic implant development.

Abstract:Titanium implants are surface-modified to achieve bioactivity and often antibacterial properties. Such surface coatings may increase corrosion degradation and be weakly attached to the substrate. In the present research, biodegradable composite coatings, investigated so far as smart environment-sensitive, slowly releasing silver to the implant neighborhood, were produced as a combination of chitosan (CS) with Eudragit E 100 (EE100) or poly(4-vinylpyridine) (P4VP) polymers incorporating mesoporous bioactive glass nanoparticles (MBGNs). Such coatings were designed to positively affect corrosion resistance, bioactivity in vitro , and the bonding to the surface. The coating was obtained in a single electrophoretic deposition (EPD) process on the surface of chemically etched grade 2 titanium. The deposition was carried out using an ethanol-based suspension for 5 min at 60 V. Suspension stability was assessed through the quantification of zeta potential. The fabricated coatings underwent microstructural analysis, chemical composition profiling, surface roughness measurement, wettability assessment, thickness determination, evaluation of corrosion resistance, and investigation of their adherence to metallic substrates. Additionally, an immersion test extending up to 14 days was employed to assess the coatings' capacity for calcium phosphate formation. The resulting coatings tightly covered the metallic substrate; the CS/EE100/MBGNs coating exhibited superior uniformity, which can be attributed to the increased stability of this suspension, indicated by a higher zeta potential value. The deposited MBGNs showed a diameter of <100 nm. The prepared coatings have contributed to improved corrosion resistance of the system and facilitated the deposition of calcium phosphates. All investigated surfaces showed hydrophilic properties. Substrate-coating adhesion was notably enhanced for the CS/EE100/MBGNs coating. The proposed coatings, after in vitro and in vivo cytotoxicity studies, can successfully be applied on long-term load-bearing titanium bone implants.

Abstract:The construction of an antibacterial biological coating on titanium surface plays an important role in the long-term stability of oral implant restoration. Graphene oxide (GO) has been widely studied because of its excellent antibacterial properties and osteogenic activity. However, striking a balance between its biological toxicity and antibacterial properties remains a significant challenge with GO. ε-poly-L-lysine (PLL) has broad-spectrum antibacterial activity and ultra-high safety performance. Using Layer-by-layer self-assembly technology (LBL), different layers of PLL/GO coatings and GO self-assembly coatings were assembled on the surface of titanium sheet. The materials were characterized using scanning electron microscope (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS) and contact angle test. The antibacterial properties of Porphyromonas gingivalis ( P.g .) were analyzed through SEM, coated plate experiment, and inhibition zone experiment. CCK-8 was used to determine the cytotoxicity of the material to MC3T3 cells, and zebrafish larvae and embryos were used to determine the developmental toxicity and inflammatory effects of the material. The results show that the combined assembly of 20 layers of GO and PLL exhibits good antibacterial properties and no biological toxicity, suggesting a potential application for a titanium-based implant modification scheme.

Silver-deposited titanium as a prophylactic 'nano coat' for peri-implantitis

Abstract:Dental implant failures caused by bacterial infections are a significant concern for dental implantologists. We modified the titanium surface by depositing silver (Ti-Ag) using direct current (DC) sputtering and confirmed the formation of a 'nano coat' by X-ray photoelectron spectroscopy (XPS), surface profilometry and energy dispersive spectroscopy (EDS). Scanning electron microscopy (SEM) and Atomic force microscopy (AFM) revealed the deposition of a uniform nano Ag thin film. A gradual increase in thickness was observed and the film thickness (530 nm) at 5 min deposition time (Ti-Ag5) resulted in the reduction of water contact angle (WCA, 15%) and an increase in surface energy (SFE, 22%) in comparison to the uncoated Ti surface. Using inductively coupled plasma-atomic emission spectroscopy (ICP-AES), the slow, steady release of Ag from the coating was observed over 21 days. The Ti-Ag5 surface exhibited excellent antibacterial activity against Streptococcus oralis, Streptococcus sanguinis, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis which belong to the yellow, purple, and red complex representing specific periodontal pathogens. Furthermore, we observed excellent cytocompatibility of Ag-deposited Ti towards MG-63 osteoblasts with no inhibitory effect on their proliferative potential. Quantitation of alkaline phosphatase (ALP) activity, mineralization efficiency, and osteogenesis-related gene expression of the MG-63 cells over a 21-day period was suggestive of rapid osseointegration. Overall, the 'nano coat' of Ag on Ti, is indeed a prophylactic against peri-implantitis ensuring increased implant success.

Abstract:Variety of ion-doped apatite coatings, especially strontium substituted hydroxyapatite (SrHA), have attracted eventful attention for medical metal surface modifications due to their excellent cytocompatibility and organizational affinity. However, they are still not commercially used in clinics because of their inappropriate biomechanical properties, and lack of osteoinductivity and antibacterial capabilities. Here, silver (Ag) doped SrHA (SrAgHA)/graphene oxide (GO) composite coatings were fabricated on titanium (Ti) substrate via electrodeposition (ED) and this combination has been reported for the first time. Morphological observations revealed that GO facilitated HA pricker-like crystals to grow in longitudinal directions, changing into a bone-like microneedles structure. SrAgHA/GO showed good mechanical properties to ensure mechanical strength required for the bone repair coating. The roughness of samples decreased from 120 ± 19 μm for the SrAgHA/GO coated Ti to 527 ± 66 μm for HA coated Ti. Although the introduction of GO makes the composite coating have a significant decreased roughness, it has no effect on the hydrophilicity of the composite coating, which is almost superpotropic. Ion release evaluation demonstrated that the presence of GO slowed the release rate of Sr and Ag, which was beneficial to reduce the cytotoxicity accompanying high concentration of Ag release. Simultaneously, GO increased the surface hydrophilicity and mechanical strength of the SrAgHA/GO. In addition, the corrosion rates of investigated coatings increased in the following order: SrAgHA/GO<SrHA/GO<SrAgHA<HA<Ti. SrAgHA/GO coatings demonstrated an effective antibacterial effect against common bacteria in clinical. The co-doping of Sr and GO in SrAgHA/GO coatings can effectively mitigate the cytotoxicity of Ag and promote the cytocompatibility of HA. The SrAgHA/GO coating did not exhibit any cytotoxicity, which can greatly promote adhesion, extension, proliferation and early differentiation of osteoblasts. Therefore, high cytocompatibility, excellent antimicrobial activity, and good mechanical properties of the SrAgHA/GO-coated Ti samples combined with its exceptional corrosion resistance performance displayed the beautiful prospect of the SrAgHA/GO-coated Ti for applications in hard tissue implant.

Abstract:Microbial colonization and biofilm formation on implanted devices represent an important complication in orthopaedic and dental surgery and may result in implant failure. Controlled release of antibacterial agents directly at the implant site may represent an effective approach to treat these chronic complications. Resistance to conventional antibiotics by pathogenic bacteria has emerged in recent years as a major problem of public health. In order to overcome this problem, non-conventional antimicrobial agents have been under investigation. In this study, polyacrylate-based hydrogel thin coatings have been electrosynthesised on titanium substrates starting from poly(ethylene glycol diacrylate)–co–acrylic acid. Silver nanoparticles (AgNPs) with a narrow size distribution have been synthesized using a “green” procedure and immobilized on Ti implant surfaces exploiting hydrogel coatings’ swelling capabilities. The coatings have been characterized by XPS and SEM/EDX, while their silver release performances have been monitored by ICP–MS. The antibacterial activity of these AgNP-modified hydrogel coatings was tested evaluating in vitro inhibition growth of Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli, among the most common pathogens in orthopaedic infections. Moreover, a preliminary investigation of the biocompatibility of silver-loaded coatings versus MG63 human osteoblast-like cells has been performed. An important point of strength of this paper, in fact, is the concern about the effect of silver species on the surrounding cell system in implanted medical devices. Abstract:Titanium (Ti) implants are widely used in orthopedic and dental applications due to their excellent biocompatibility and mechanical properties. However, bacterial adhesion and subsequent biofilm formation on implant surfaces pose a significant risk of postoperative infections and complications. Conventional surface modifications often lack long-lasting antibacterial efficacy, necessitating the development of novel coatings with enhanced antimicrobial properties. This study aims to develop a novel Ag-TiO 2 -OTS (Silver-Titanium dioxide-Octadecyltrichlorosilane, ATO) nanocomposite coating, through a chemical plating method. By employing a 'resist-killing-disintegrate' approach, the coating is designed to inhibit bacterial adhesion effectively, and facilitate pollutant removal with lasting effects. Characterization of the coatings was performed using spectroscopy, electron microscopy, and contact angle analysis. Antibacterial efficacy, quantitatively evaluated against E. coli and S. aureus over 168 h, showed a significant reduction in bacterial adhesion by 76.6% and 66.5% respectively, and bacterial removal rates were up to 83.8% and 73.3% in comparison to uncoated Ti-base material. Additionally, antibacterial assays indicated that the ratio of the Lifshitz-van der Waals apolar component to electron donor surface energy components significantly influences bacterial adhesion and removal, underscoring a tunable parameter for optimizing antibacterial surfaces. Biocompatibility assessments with the L929 cell line revealed that the ATO coatings exhibited excellent biocompatibility, with minimal cytotoxicity and no significant impact on cell proliferation or apoptosis. The ATO coatings provided a multi-functionality surface that not only resists bacterial colonization but also possesses self-cleaning capabilities, thereby marking a substantial advancement in the development of antibacterial coatings for medical implants.

Abstract:The present study reports development of multi-functional coatings on titanium for orthopaedic implant application. The multi-functional coating approach involves development of TiO2 nanotubes by anodization followed by dip coating of polycaprolactone (PCL) in which the polymer matrix is incorporated with bioglass (BG), Ag doped bioglass (AgBG), Ag doped bioglass-graphene oxide (AgBG-GO), Ag doped bioglass-reduced graphene oxide (AgBG-rGO) and bioglass-silver functionalized reduced graphene oxide (BG-AgrGO). The structural and morphological characteristics, bioactivity and antibacterial activity of the multi-functional coatings were evaluated. AgBG-GO/PCL and BG-AgrGO/PCL composite coatings exhibit better antibacterial activity than AgBG/PCL and AgBG-rGO/PCL composite coatings. Among them, the BG-AgrGO/PCL composite coating with higher loading and slow release of silver ions is suitable for long term antibiotic delivery. The BG-AgrGO/PCL composite coating shows a Ca/P ratio of 1.53. The formation of Ca-deficient apatite of this composite coating suggests that the AgrGO did not hinder the bioactivity. Further, rGO nanosheets enables a higher loading of Ag and the persistence of 0.98 at.% of Ag even after 10 days of immersion reveal sustained release of antibiotic in the physiological medium. The retention of coarse BG particles and lower number of oxygen functional groups of rGO and AgrGO could have limited the dissolution of BG particles in the AgBG-rGO/PCL and BG-AgrGO/PCL composite coatings even after 14 days of immersion in HBSS. With a higher loading, slow release of antibiotics and having adequate bioactivity, BG-AgrGO/PCL composite coating can be considered as a potential candidate for biomedical implant materials.

Near-infrared Excited Graphene Oxide/silver Nitrate/chitosan Coating for Improving Antibacterial Properties of Titanium Implants

Abstract:Objective:To design and construct a graphene oxide (GO)/silver nitrate (Ag3PO4)/chitosan (CS) composite coating for rapidly killing bacteria and preventing postoperative infection in implant surgery. Methods:GO/Ag3PO4 composites were prepared by ion exchange method, and CS and GO/Ag3PO4 composites were deposited on medical titanium (Ti) sheets successively. The morphology, physical image, photothermal and photocatalytic ability, antibacterial ability, and adhesion to the matrix of the materials were characterized. Results:The GO/Ag3PO4 composites were successfully prepared by ion exchange method and the heterogeneous structure of GO/Ag3PO4 was proved by morphology phase test. The heterogeneous structure formed by Ag3PO4 and GO reduced the band gap from 1.79 eV to 1.39 eV which could be excited by 808 nm near-infrared light. The photothermal and photocatalytic experiments proved that the GO/Ag3PO4/CS coating had excellent photothermal and photodynamic properties. In vitro antibacterial experiments showed that the antibacterial rate of the GO/Ag3PO4/CS composite coating against Staphylococcus aureus reached 99.81% after 20 minutes irradiation with 808 nm near-infrared light. At the same time, the composite coating had excellent light stability, which could provide stable and sustained antibacterial effect. Conclusion:GO/Ag3PO4/CS coating can be excited by 808 nm near infrared light to produce reactive oxygen species, which has excellent antibacterial activity under light.

Abstract:Abstract Titanium (Ti) is the widely used implant material in clinic, however, failures still frequently occur due to its bioinertness and poor antibacterial property. To improve the biological and antibacterial properties of Ti implants, micro-nanostructured hydroxyapatite (HA) coating was prepared on Ti surface by micro-arc oxidation (MAO), and then the antibacterial agent of chitosan (CS) was loaded on the HA surface through dip-coating method. The results showed that the obtained HA/CS composite coating accelerated the formation of apatite layer in SBF solution, enhanced cell adhesion, spreading and proliferation, and it also inhibited the bacterial growth, showing improved biological and antibacterial properties. Although, with the increased CS amount, the coverage of HA coating would be enlarged, resulting in depressed biological property, however, the antibacterial property of the composite coating was enhanced, and the cytotoxicity about CS was not detected in this work. In conclusion, the HA/CS coating has promising application in orthopedics, dentistry and other biomedical devices.

Abstract:The Agrobacterium single-stranded DNA (ssDNA) intermediate T-strand is likely transferred to the plant cell nucleus as a complex with a single VirD2 molecule at its 5' end and multiple VirE2 molecules along its length. VirD2 contains a nuclear localization signal (NLS); however, because the T-strand is principally coated with VirE2 molecules, VirE2 also might assist in nuclear uptake. Indeed, VirE2 fused to a reporter protein localizes to plant cell nuclei, a process mediated by two amino acid sequences with homology to the bipartite NLS of Xenopus nucleoplasmin. Moreover, tumorigenicity of an avirulent virE2 mutant is restored when inoculated on transgenic plants expressing VirE2, supporting in planta function of VirE2.
Preparation and Properties of Hydroxyapatite/Chitosan Coatings on Ti Surface by Different Electrochemical Methods

Abstract:Titanium (Ti) is the widely used implant material in clinic; however, failures still frequently occur due to its poor bioactivity and antibacterial property. To improve the bioactivity and antibacterial property, in this work, petaling micro-nanostructured hydroxyapatite (HA) coatings were firstly prepared on Ti surface by micro-arc oxidation (MAO), and then, the antibacterial agent—chitosan (CS) was loaded on the HA surfaces. The CS was loaded by two methods: One is anodic oxidation, and the other is the in situ MAO. The degradability, bioactivity, biocompatibility and antibacterial activity of the two kinds of HA/CS coatings were examined and compared by in vitro tests. The results showed that, the two-step method induced HA/CS coating had more CS on Ti surface, showing better antibacterial property; however, the in situ MAO-induced HA/CS coating had longer degradation time and better biological property. The better antibacterial property was due to the increased CS amount on HA surface. The prolonged degradation time was due to the mixed structure of HA and CS. And the good biological property was attributed to the improved surface roughness, hydrophilicity and exposed HA area. Considering the long-term needs of both antibacterial and biological properties for implant materials, the in situ MAO-induced HA/CS coating has a greater potential to be used as an implant material in clinics.

Abstract:Dental implants have great potential in the global market, around $3.7 billion in 2015, which will increase to $7 billion in 2023 with an annual increase rate of 8.2%. Incorporating antibacterial and osteogenic agents into implants is helpful to make the dental implants successful, which can be endowed by coatings. In recent years, graphene oxide (GO) and its composite materials have shown advances in the biomedical field. Lysozyme (Lys) and tannic acid (TA) are naturally derived, with promising antibacterial and osteogenic properties as well. In the present study, the strong antibacterial and enhanced osteogenic multilayer coating is fabricated using the facile and controllable layer by layer (LBL) technique to integrate GO, Lys, and TA. The thickness of coating exhibited a continuous growth with the deposited process as proved from UV-vis and ellipsometry data, and the physical properties of the coating, such as wettability, roughness, and stiffness are well characterized. The coatings exhibited the synergic effect on the killing bacteria, both Gram-negative bacteria and Gram-positive bacteria represented by E. coli and S. aureus, respectively, and enhancing osteogenesis of dental pulp stem cells (hDPSCs), showing the potential application on coatings of dental implants. Thus, the strategy applied here will inspire the design and development of dual functional surfaces for the success of implanted dental surface in future.

Abstract:Infection-related titanium implant failure rates remain exceedingly high in the clinic. Functional surface coating represents a very promising strategy to improve the antibacterial and bioactive properties of titanium alloy implants. Here, we describe a novel bioactive surface coating that consists of a mussel-inspired carboxymethyl chitosan hydrogel loaded with silver nanoparticles (AgNPs) to enhance the bioactive properties of the titanium alloy. The preparation of hydrogel is based on gallic acid grafted carboxymethyl chitosan (CMCS-GA) catalyzed by DMTMM (4-(4,6-dimethoxy-1,3,5-triazin-2-yl)-4-methylmorpholinium chloride). To build a firm bonding between the hydrogel and titanium alloy plate, a polydopamine layer was introduced onto the surface of the titanium alloy. With HRP/H2O2 catalysis, CMCS-GA can simply form a firm gel layer on the titanium alloy plate through the catechol groups. The surface properties of titanium alloy were characterized by scanning electron microscope (SEM), X-ray photoelectron spectroscopy (XPS), and water contact angle. Silver nanoparticles were loaded into the gel layer by in situ reduction to enhance the antibacterial properties. In vitro antibacterial and cell viability experiments showed that the AgNPs-loaded Ti-gel possesses excellent antibacterial properties and did not affect the proliferation of rabbit mesenchymal stem cells (MSCs).

Abstract:Previously, functional coatings on 3D-printed titanium implants were developed to improve their biointegration by separately incorporating Ga and Ag on the biomaterial surface. Now, a thermochemical treatment modification is proposed to study the effect of their simultaneous incorporation. Different concentrations of AgNO3 and Ga(NO3)3 are evaluated, and the obtained surfaces are completely characterized. Ion release, cytotoxicity, and bioactivity studies complement the characterization. The provided antibacterial effect of the surfaces is analyzed, and cell response is assessed by the study of SaOS-2 cell adhesion, proliferation, and differentiation. The Ti surface doping is confirmed by the formation of Ga-containing Ca titanates and nanoparticles of metallic Ag within the titanate coating. The surfaces generated with all combinations of AgNO3 and Ga(NO3)3 concentrations show bioactivity. The bacterial assay confirms a strong bactericidal impact achieved by the effect of both Ga and Ag present on the surface, especially for Pseudomonas aeruginosa, one of the main pathogens involved in orthopedic implant failures. SaOS-2 cells adhere and proliferate on the Ga/Ag-doped Ti surfaces, and the presence of gallium favors cell differentiation. The dual effect of both metallic agents doping the titanium surface provides bioactivity while protecting the biomaterial from the most frequent pathogens in implantology.

Titanium Dioxide/Graphene Oxide Composite Coatings for 316 Stainless Steel Dental Implants

Abstract:Stainless steel has been used in orthopedics and orthodontic fields. However, it cannot be used for fabrication of dental implants due to its inertness, low biocompatibility and weak resistance to corrosion. A composite coating of titanium oxide /graphene oxide has been prepared for stainless steel to improve its biological properties. Stainless steel discs were polished, cleaned and pre-treated with a mixture of HNO3 and HF acid for 15 min. The composite coating composed of TiO2 produced by sol-gel technique and doped with 0.75 wt% graphene oxide. XRD, SEM-EDX and AFM were employed to characterize the composite coating. The anti-bacterial action of the composite coating was investigated against S. aureus and E. coli. The corrosion resistance of coated and noncoated samples was assessed in SBF using electrochemical technique. Cytotoxicity was assessed using osteoblast-like cells. The wettability was determined by contact angle, and bioactivity assessed by immersion in SBF. The results revealed that the composite coating was dense with few micro-cracks, and was not cytotoxic to osteoblast-like cells. The composite coating reduced bacterial colonies and the corrosion rate of the steel was improved. The wettability of the sample was increased with the composite coating and apatite formation appeared after 21 days.

Abstract:One of the most common causes of implant failure is peri-implantitis, which is caused by bacterial biofilm formation on the surfaces of dental implants. Modification of the surface nanotopography has been suggested to affect bacterial adherence to implants. Silver nanoparticles are also known for their antibacterial properties. In this study, titanium alloy implants were surface modified following silver plating, anodisation and sintering techniques to create a combination of silver, titanium dioxide and hydroxyapatite (HA) nanocoatings. Their antibacterial performance was quantitatively assessed by measuring the growth of Streptococcus sanguinis, proportion of live/dead cells and lactate production by the microbes over 24 h. Application of a dual layered silver-HA nanocoating to the surface of implants successfully inhibited bacterial growth in the surrounding media (100% mortality), whereas the formation of bacterial biofilm on the implant surfaces was reduced by 97.5%. Uncoated controls and titanium dioxide nanocoatings showed no antibacterial effect. Both silver and HA nanocoatings were found to be very stable in biological fluids with material loss, as a result of dissolution, to be less than 0.07% for the silver nanocoatings after 24 h in a modified Krebs-Ringer bicarbonate buffer. No dissolution was detected for the HA nanocoatings. Thus, application of a dual layered silver-HA nanocoating to titanium alloy implants creates a surface with antibiofilm properties without compromising the HA biocompatibility required for successful osseointegration and accelerated bone healing.
