Abstract:Different sources of nitrogen pose diverse effects to algal community, but the mechanism of inhibitory effects of nitrogen sources on freshwater diatoms is not fully understood. The purpose of this study was to compare biomass, photosynthetic activity, and morphological structure of three common freshwater diatoms ( Cyclotella meneghiniana , Nitzschia sp., and Gomphonema parvulum ) under different nitrogen sources (NO 3 − or NH 4 + ). The sorption characteristic of each diatom was investigated, and chlorophyll a (Chl-a) content and oxygen evolution rate were analyzed to investigate stress of different nitrogen sources on each diatom in the batch experiments. Ammonium lowered the growth rate of C. meneghiniana and Nitzschia sp. when it was supplied in addition to growth-saturating nitrate concentrations, suggesting a combined effect of inhibition of nitrate uptake and direct ammonium stress. Oxygen evolution rate of Nitzschia sp. showed that the direct ammonium stress on the photosynthetic activity can be alleviated by coexistence of nitrate in the nitrogen enriched treatment, but not for C. meneghiniana and G. parvulum , which may be caused by a different nitrate transporter system within algal cells. Transmission electron microscopy was used to assess the toxicity of ammonium on ultrastructural chloroplast of each diatom. Ultrastructural changes in chloroplasts showed undefined electron-dense granules and lipid droplets, but the membrane integrity of cell was maintained, suggesting an adaptation to adjustment to ammonia stress. Results showed that Cyclotella meneghiniana and Nitzschia sp. were more sensitive to ammonium stress than Gomphonema parvulum on growth, but the mechanism remains unclear.

Abstract:From April 2006 to April 2007, the geographical and seasonal variation in nitrogen content in terricolous lichen (Cladonia portentosa) and atmospheric ammonia concentrations were measured at five heathland sites. The seasonal variation in the nitrogen content of the lichen was small, even though there was a large seasonal variation in the air concentration of ammonia. A sizable local variation in the nitrogen content of the lichen was found even at the scale of a few kilometres. The nitrogen content in the lichen showed a high correlation to the yearly mean value of the measured ammonia concentration in air at the different locations. This investigation is part of a larger attempt to incorporate effects of nitrogen in the conservation status of terrestrial habitat types.

Abstract:Anthropogenic nitrogen (N) and sulfur (S) emissions triggered acid deposition and affected the environmental quality and ecosystem functions. Lichen N and S concentrations have been found increasing with N and S deposition at relatively low levels, respectively. However, it remains unclear whether lichen N and S concentrations can respond to corresponding deposition fluxes under high pollution environments and how to use lichen N isotopes (δ15N) to evaluate source contributions of N deposition quantitatively.Along an urban-to-rural transect surrounding a polluted area in northern China, we investigated lichen N and S to examine their sensitivity and applicability in estimating N and S deposition. Moreover, we established a new method to reconstruct site-based δ15N values of ammonium N (NHx) and oxidized N (NOy) deposition using lichen δ15N and then to differentiate relatively contributions of major N emission sources.Lichen N (1.0–3.9%, 2.5 ± 0.6%) and S (0.09–0.33%, 0.21 ± 0.06%) decreased linearly with distances from the polluted center area. Wet inorganic N and sulfate deposition (29.0 ± 6.1 kg-N/ha/yr and 25.8 ± 7.9 kg-S/ha/yr, respectively) estimated by integrating relationships from previous literature data were comparable with levels based on direct deposition observations in the study area. Lichen δ15N varied between −12.1 to −4.1‰ and averaged −7.1 ± 2.0‰ among our study sites, and reconstructed δ15N values of NHx and NOy deposition averaged −12.8 ± 1.0‰ and −3.2 ± 0.5‰, respectively. Source contribution analyses revealed significant contributions of volatilization ammonia (66 ± 3%) from wastes and fertilizers to NHx deposition and non-fossil fuel N oxides (62 ± 7%) from biomass burnings and microbial N cycles to NOy deposition.These results indicate that lichen N and S are sensitive to deposition fluxes under high pollution and applicable to estimating deposition levels. This work improves the methodology of atmospheric deposition biomonitoring based on lichen element and isotope records.

In particular, the most relevant changes in protein expression were observed in the fungal partner, with NO3−mostly affecting the energetic metabolism and NH4+affecting transport and regulation of proteins and the energetic metabolism much more than NO3−did.Excess deposition of reactive nitrogen is a well-known agent of stress for lichens, but which symbiont is most affected and how, remains a mystery. Using proteomics can expand our understanding of stress effects on lichens. We investigated the effects of different doses and forms of reactive nitrogen, with and without supplementary phosphorus and potassium, on the proteome of the lichen Cladonia portentosa growing in a ‘real-world’ simulation of nitrogen deposition. Protein expression changed with the nitrogen treatments but mostly in the fungal partner, with NO3− mainly affecting the energetic metabolism and NH4+ also affecting the protein synthesis machinery. The photobiont mainly responded overexpressing proteins involved in energy production. This suggests that in response to nitrogen stress, the photobiont mainly supports the defensive mechanisms initiated by the mycobiont with an increased energy production. Such surplus energy is then used by the cell to maintain functionality in the presence of NO3−, while a futile cycle of protein production can be hypothesized to be induced by NH4+ excess. External supply of potassium and phosphorus influenced differently the responses of particular enzymes, likely reflecting the many processes in which potassium exerts a regulatory function.

Enhanced foliar 15 N enrichment with increasing nitrogen addition rates: Role of plant species and nitrogen compounds

Abstract:Determining the abundance of N isotope (δ15 N) in natural environments is a simple but powerful method for providing integrated information on the N cycling dynamics and status in an ecosystem under exogenous N inputs. However, whether the input of different N compounds could differently impact plant growth and their 15 N signatures remains unclear. Here, the response of 15 N signatures and growth of three dominant plants (Leymus chinensis, Carex duriuscula, and Thermopsis lanceolata) to the addition of three N compounds (NH4 HCO3 , urea, and NH4 NO3 ) at multiple N addition rates were assessed in a meadow steppe in Inner Mongolia. The three plants showed different initial foliar δ15 N values because of differences in their N acquisition strategies. Particularly, T. lanceolata (N2 -fixing species) showed significantly lower 15 N signatures than L. chinensis (associated with arbuscular mycorrhizal fungi [AMF]) and C. duriuscula (associated with AMF). Moreover, the foliar δ15 N of all three species increased with increasing N addition rates, with a sharp increase above an N addition rate of ~10 g N m-2 year-1 . Foliar δ15 N values were significantly higher when NH4 HCO3 and urea were added than when NH4 NO3 was added, suggesting that adding weakly acidifying N compounds could result in a more open N cycle. Overall, our results imply that assessing the N transformation processes in the context of increasing global N deposition necessitates the consideration of N deposition rates, forms of the deposited N compounds, and N utilization strategies of the co-existing plant species in the ecosystem.
Acute responses of bio-denitrification to short-term clopyralid exposure: Kinetic analysis and biological mechanisms

Abstract:The mass production and extensive application of clopyralid (CLP) have attracted much attention regarding its environmental effect in recent decades. However, studies on the acute responses and the underlying inhibition mechanisms of denitrifiers to CLP exposure are scarce. Herein, the crucial factors in denitrification were quantified through kinetics, metabolic activity analysis, and structural equation modeling (SEM). The acute response mechanisms of denitrifiers under short-term CLP exposure were further proposed according to the quantitative correlation of denitrification performance with C and N metabolism, ETS activity, and oxidative stress. The modified non-competitive inhibition model fitted well to describe the inhibitory characteristics of CLP on the nitrate reduction (R-2 = 0.99), which was significantly inhibited with increasing CLP exposure concentrations. Short-term exposure to CLP could inhibit C and N metabolism and electron transport by inducing oxidative stress during denitrification. Quantitative PCR (qPCR) analysis demonstrated that CLP restrained the abundance of narG and nirS, suggesting that down-regulation of denitrification enzyme expression was highly related to the reduction in nitrogen conversion. Moreover, SEM indicated that ETS activity and NAR were the major factors in predicting NO3- reduction rate (DNR) and were mainly influenced by NADH and reactive oxygen species (ROS). Consistently, NADH and NIR were closely correlated with total nitrite accumulation (NIA). Shortterm CLP exposure could affect denitrification performance by directly affecting microbial physicochemical properties or indirectly interfering with the abundance of denitrifying functional genes and metabolic activity through oxidative stress. This study provided new insights into acute responses of denitrifiers to short-term CLP exposure.

Abstract:Atmospheric nitrogen (N) deposition is coupled with organic nitrogen (ON) and inorganic nitrogen (IN); however, little is known about plant growth and the balance of elements in Torreya grandis growing under different ON/IN ratios. Here, we investigated the effects of ON/IN ratios (1/9, 3/7, 7/3, and 9/1) on leaf stoichiometry (LF), chlorophyll content, and chlorophyll fluorescence of T. grandis. We used ammonium nitrate as the IN source and an equal proportion of urea and glycine as the ON source. The different ON/IN ratios altered the stoichiometry and photochemical efficiency in T. grandis. Although the leaf P content increased significantly after treatment, leaf N and N:P maintained a certain homeostasis. Torreya grandis plants performed best at an ON/IN ratio of 3/7, with the highest values of chlorophyll-a, total chlorophyll, maximum photochemical efficiency, and photosynthetic performance index. Thus, both ON and IN types should be considered when assessing the responses of plant growth to increasing N deposition in the future. Our results also indicated that the leaf P concentration was positively correlated with Chl, Fv/Fm, and PIabs. This result further indicates the importance of the P element for plant growth against the background of nitrogen deposition. Overall, these results indicate that T. grandis might cope with changes in the environment by maintaining the homeostasis of element stoichiometry and the plasticity of PSII activity.

Abstract:The kinetic characteristics of ammonium and nitrate uptake by duckweed(Spirodela oligorrhiza) plants could be illustrated with the Michaelis-Menten equation.The results indicated that the Km value of ammonium uptake was less than that of nitrate uptake by duckweed.The finding reinforced the insight about " preferential uptake and clean-up ammonium in solution by duckweed plants".It is also found that the maximum rate of nitrate uptake by duckweed plants was larger than that of ammonium uptake.Based on the kinetic characteristics of ammonium and nitrate uptake by duckweed(Spirodela oligorrhiza) plants,a new complex system as physical oxygen enrichment—microbial nitrification—plant (duckweed) absorption for waste water clean-up was suggested.

Abstract:Erythropalum scandens Bl. is a woody vegetable with high nitrogen demand that inhabits southern China. Ammonium and nitrate are the two main forms of inorganic nitrogen that plants directly absorb. A pot experiment was performed to determine the growth, physiological responses, and preferences of 12-month-old E. scandens seedlings for ammonium and nitrate. Aboveground and underground growth indexes, biomass, physiological and biochemical indexes (chlorophyll [Chl], soluble sugar, soluble protein and free proline contents), and substrate pH and nitrogen contents were determined under different nitrate and ammonium ratios (0 NO3-: 100 NH4+, 25 NO3-: 75 NH4+, 50 NO3-: 50 NH4+, 75 NO3-: 25 NH4+, and 100 NO3-: 0 NH4+), and the control (0 NO3-: 0 NH4+). The results showed that ammonium and nitrate improved the growth and physiological status of E. scandens seedlings in most of the treatments compared to the control. The aboveground growth status and biomass accumulation of E. scandens seedlings were significantly better under the 0 NO3-: 100 NH4+ treatment during fertilization compared with all other treatments. However, the growth status of the underground parts was not significantly different among treatments. Significant differences in osmoregulator content, except for soluble sugars, and Chl content were observed. Soluble sugars and soluble proteins were highest under the 0 NO3-: 100 NH4+ treatment at the end of fertilization (day 175). However, free proline accumulated during fertilization and the increase in NO3- indicated that excessive use of NO3- had a negative effect on the E. scandens seedlings. The order of accumulating nitrogen content was leaves > roots > stems. The highest N accumulation occurred in the aboveground parts under the 0 NO3-: 100 NH4+ treatment, whereas the highest N accumulation occurred in the underground parts under the 50 NO3-: 50 NH4+ treatment. Substrate pH increased at the end of fertilization (day 175) compared with the middle stage (day 75), while total nitrogen, ammonium, and nitrate were highly significantly different among the treatments. Total nitrogen and NH4+ content were the highest under the 0 NO3-: 100 NH4+ treatment, while NO3- content was the highest under the 100 NO3-: 0 NH4+ treatment. In conclusion, 12-month-old E. scandens seedlings grew best, and had better physiological conditions in NH4+ than NO3-. The 0 NO3-:100 NH4+ treatment (ammonium chloride 3.82 g/plant) resulted in the best growth and physiological conditions. Most of the growth and physiological indexes were inhibited with the increase in nitrate.
Improved Utilization of Nitrate Nitrogen Through Within-Leaf Nitrogen Allocation Trade-Offs in Leymus chinensis

Abstract:The Sharply increasing atmospheric nitrogen (N) deposition may substantially impact the N availability and photosynthetic capacity of terrestrial plants. Determining the trade-off relationship between within-leaf N sources and allocation is therefore critical for understanding the photosynthetic response to nitrogen deposition in grassland ecosystems. We conducted field experiments to examine the effects of inorganic nitrogen addition (sole NH 4 + , sole NO 3 – and mixed NH 4 + /NO 3 – : 50%/50%) on N assimilation and allocation by Leymus chinensis . The leaf N allocated to the photosynthetic apparatus (N PSN ) and chlorophyll content per unit area (Chl area ) were significantly positively correlated with the photosynthetic N-use efficiency (PNUE). The sole NO 3 – treatment significantly increased the plant leaf PNUE and biomass by increasing the photosynthetic N allocation and Chl area . Under the NO 3 treatment, L. chinensis plants devoted more N to their bioenergetics and light-harvesting systems to increase electron transfer. Plants reduced the cell wall N allocation or increased their soluble protein concentrations to balance growth and defense under the NO 3 treatment. In the sole NH 4 + treatment, however, plants decreased their N allocation to photosynthetic components, but increased their N allocation to the cell wall and elsewhere. Our findings demonstrated that within-leaf N allocation optimization is a key adaptive mechanism by which plants maximize their PNUE and biomass under predicted future global changes.

Differential effects of nitrate and ammonium on the growth of algae and microcystin production by nitrogen-fixing Nostoc sp. and non-nitrogen-fixing Microcystis aeruginosa

Abstract:Cyanotoxins produced by cyanobacteria are a significant threat to human health. However, their responses to nitrogen (N) supplies could differ between N-fixing and non-N-fixing species, which has been poorly understood. This study aimed to compare the responses of the non-N-fixing Microcystis aeruginosa and N-fixing Nostoc sp. to varying concentrations of nitrate and ammonium. This comparison had been conducted by analyzing chlorophyll-a contents, maximum quantum efficiencies of photosystem II, microcystin production, and related gene expressions. Our findings revealed that nitrate substantially stimulated the growth of both M. aeruginosa and Nostoc sp. with biomass increase by 366.2 ± 56.5 and 93.0 ± 14.0%, respectively, at 16 mg-N/L. In contrast, high ammonium concentrations suppressed their growth. Furthermore, the intracellular concentration of microcystins produced by M. aeruginosa was higher under high nitrate. Extracellular microcystins showed an opposite trend to increases in nitrate and ammonium. Ammonium increases the production and releases microcystin from Nostoc sp. N metabolism genes showed a similar trend with toxin formation genes, which were up-regulated under the high N treatments. This study provides valuable insights into the impacts of N supplies on growths of N- and non-N-fixing cyanobacteria, as well as microcystin production, which helps to develop effective strategies for managing cyanobacterial blooms.
Nitrate and Ammonium Nitrogen Addition Did Not Alter Nutrient Resorption of Dominant Plant in an Alpine Grassland

Abstract:Worldwide increases in nitrogen deposition rates are influenced by human activities. Although the total amount of N deposition tends to be stable in our country, atmospheric N deposition of the reactive N forms (NO4+-N, NH4+; NO3−-N, NO3−) is remarkably different, and the ratios of NH4+ to NO3− change continuously. Nutrient resorption is a crucial driver of plant nutrient conservation strategies and litter quality. Therefore, the plant nutrient resorption pattern has remarkable ecological significance for nutrient cycling and the community structure of the ecosystem. However, previous studies have found that plants have different preferences in NH4+ and NO3−, and the response of nutrient resorption to different N forms remains unclear. In 2017 and 2018, we conducted field experiments simulating five NO3− and NH4+ addition ratios to examine the responses of the nutrient resorption of the dominant species, Stipa capillata, in the alpine grassland of northwest China. Results showed that N addition treatments did not affect the nutrient resorption efficiencies. The results showed that N application had little effect on soil inorganic N composition, so there was no significant change in the nutrient resorption efficiencies. In addition, the nutrient resorption efficiencies were unaffected across the five different ratios of nitrate and NH4+ addition, suggesting that nutrient resorption was not only controlled by the nutrient supply and nutrient form. Our results have important implications in understanding the significance of the frequency of N addition due to N loss through leaching and denitrification. The results also highlight that low intensity and high frequency N addition are required to investigate the response of plant nutrient resorption to the N supply in the future study.

Nitrogen and Water Addition Alter Nitrogen Uptake Preferences of Two Dominant Plant Species in a Typical Inner Mongolian Steppe

Abstract:The preferences for different nitrogen (N) forms by plants play an important role in determining the fate and dynamics of N in ecosystems. Nevertheless, it remains unclear whether and how resource addition influences N uptake preferences of plants. Here we investigated the effects of long-term (10 a) N and water addition on plant N uptake in a temperate semi-arid grassland. We used the N-15 natural abundance technique to measure N content and isotope in soils and leaves of two dominant plant species, Stipa krylovii and Agropyron cristatum, and quantified the proportion of plant N derived from different N sources with an isotopic mixing model. We found that N addition, but not water addition, significantly increased soil nitrate and ammonium content, and also enhanced plant leaf N concentration. Soil nitrate accounted for 80% and ammonium for 20% contributing to plant N in non-treated controls. Compared to the controls, N addition reduced nitrate uptake of the two species, whereas water addition decreased nitrate uptake of S. krylovii but promoted the nitrate uptake of A. cristatum. Water interacted with N to significantly affect plant N uptake preference, indicating that the effect of water addition on plant N uptake varies with soil N availability. At the ambient N level, water addition enhanced ammonium utilization of S. krylovii but decreased that of A. cristatum. At the increased N level, water addition reduced ammonium uptake and enhanced nitrate uptake in both species. Our results demonstrate that the dominant plant species of the typical Inner Mongolian steppe mainly take up the relatively abundant nitrate rather than the scarce ammonium in this semiarid grassland. Plant preferences for different N forms changed with resource addition, suggesting that future N deposition and increases in nitrate availability may promote the growth of the dominant plant species in temperate grasslands, which may contribute to alleviating the global climate change.
Thalli Growth, Propagule Survival, and Integrated Physiological Response to Nitrogen Stress of Ramalina calicaris var. japonica in Shennongjia Mountain (China)

Abstract:In this study, effects of nitrogen (N) availability on growth, survival of Ramalina calicaris var. japonica, and whether it respond nitrogen stress in an integrated physiological way was evaluated. Thalli growth and propagule survival, thalli N and phosphorus (P) content, and activity of phosphomonoesterase (PME) of R. calicaris var. japonica were determined in a field experiment. Its differentiate adsorption in ammonia and nitrate, the activity of glutamine synthetase (GSA) and nitrate reductase (NRA) also were investigated in a series of indoor experiments. The results showed that N deposition significantly decreased the growth and survival of this lichen, and the N sensitivity threshold was suggested at 6.0 kg N⋅ha-1⋅y-1. When the N deposition increased from 8.59 kg N⋅ha-1⋅y-1 to 14.24, 20.49, 32.99 and 57.99 kg N⋅ha-1⋅y-1, the growth rates of lichen thalli decreased by 26.47, 39.01, 52.18 and 60.3%, respectively; Whereas the survival rate of the lichen propagules decreased from 92.8% of control (0.0 kg N⋅ha-1⋅y-1) to 10.7% of 50.0 kg N⋅ha-1⋅y-1, when they were treated with 0.00, 6.25, 12.5, 25.0, and 50.0 kg N⋅ha-1⋅y-1 deposition. Compared with an adequate adsorption of ammonium N, no nitrate adsorption occurred when thalli was submerged in solution lower than 0.4 mM. Our results also suggested that thalli total nitrogen, N:P ratio increased with N availability, and the activity of PME was significantly correlated with thalli total nitrogen. These all indicated that phosphorus limitation occurred when R. calicaris var. japonica treated with higher nitrogen deposition. Compared with slightly effects of NRA, GSA of R. calicaris var. japonica responded nitrogen availability significantly; In addition, GSA and NRA negatively correlated with thalli growth rate and propagule survival significantly. These results indicated that nitrogen stress do decrease growth and survival of R. calicaris var. japonica, and lichen would be impacted by excess nitrogen in a integrated, not a fragmentary way, including nitrogen uptake, assimilation, even nutrient balance of nitrogen and phosphorous.
Responses of Plant 15N Natural Abundance and Isotopic Fractionation to N Addition Reflect the N Status of a Temperate Steppe in China

Abstract:Elevated anthropogenic nitrogen (N) deposition could alter N status in temperate steppe. However, threshold observations of N status change from N limit to N saturation by far are not conclusive in these ecosystems. Research on the natural abundance of N-15 (N-15) could greatly help provide integrated information about ecosystem N status. The goal of this study was to investigate the suitability of measurements of N-15 of major ecosystem N pools and several key species, plant N-15 fractionation, together with key vegetation and soil indicators in response to N fertilization as a tool to identify the N status in a temperate steppe in Inner Mongolia. We carried out a N addition experiment during 201114 on a Stipa krylovii steppe in Inner Mongolia, Northern China. We investigated the response of several key N transformation processes, vegetation and soil properties to N addition. Aboveground biomass and belowground biomass (BGB) N-15, root and foliar N-15 of three dominant species (Artemisia frigida, S. krylovii and Leymus chinensis), N-15 of soil total N and soil KCl-extractable NO3-N were determined. The responses of isotope fractionation during plant N uptake and reallocation to N addition were also determined. Our results suggest that the N addition rate of 5g N m(2) yr(1) could be regarded as threshold of early N saturation in this S. krylovii steppe as indicated by an increase in plant fractionation and a decrease in plant N-15. When N input rate is > 10 g N m(2) yr(1), increased N deposition can lead to an apparent reduction in species richness and BGB as well as an increase in NO3 in extractable soil pools < 30-cm soil profile. With N addition, S. krylovii and A. frigida undergo earlier N status shift from N limitation toward N excess compared with L. chinensis, contributing to L. chinensis out-competing other species. Overall, this study provides a better understanding of N status change in temperate steppe based on isotope evidence and several other functional variables and contributes to predicting the responses of temperate steppe to future global N deposition scenario.

Abstract:Soil nitrogen (N) transformations between labile N forms (extractable organic N [EON], ammonium [NH4+], and nitrate [NO3−]) regulate soil N availability. However, it has long been difficult to quantify the transformations of total soil organic and labile N forms in soils, which has left large uncertainties in evaluating atmospheric N deposition effects on soil N dynamics. Based on concentrations and natural abundances of N isotopes of soil organic N, EON, NH4+, and NO3− across 11 forests with variant N deposition levels, we established a quantitative isotopic framework to estimate the fractions of soil N depolymerization (fD), mineralization (fM), nitrification (fN), and of NO3− losses (fL) via denitrification and leaching. Based on the fractions, the gross production and storage of corresponding soil labile N were estimated for forests of China and Japan. We found that fD, fM, and fN increased, while fL decreased with the increase of N deposition among the study forests. And the contribution of denitrification (relative to the NO3− leaching) to total NO3− losses also increased with increasing N deposition. Our method provides new and straightforward insights into the present soil N transformations and allows to evaluate the soil N status. These findings are useful for modeling forest N cycles under different N deposition regimes. Effect of ammonium fixation on determination of n mineralized from soil microbial biomass

Abstract:Two soils with relatively high (Soil 1) and low (Soil 2) ammonium fixation capacities were used in this study to examine the effect of ammonium fixation on the determination of N mineralized from soil microbial biomass. Organism suspension was quantitatively introduced to Soil 1 at various rates. Both fumigationincubation (FI) and fumigation-extraction (FE) methods were used to treat the soil. The amount of fixed NH-N increased with increasing rate of organism-N addition. A close correlation was found between the amount of fixed ammonium and the rate of organism-N addition. The net increases of fixed NH4 +-N were equivalent to 38% and 12% of the added organism-N for FI and FE treatments, respectively, in this specific soil. To provide isotopic evidence, 15N-labelled organism-N was added to Soils 1 and 2 at 121.4 mg N kg-1. In FI treatment, 22 and 3 mg N kg-1 of labelled N were found in the fraction of fixed NH4 +-N in Soils 1 and 2 respectively; while in FE treatment, 9 mg N kg-1 of labelled N was found in the fraction of fixed NH4 +-N in Soil 1 only. There was no labelled N in the fraction of fixed NH4 +-N in Soil 2. In all of the unfumigated (check) soils, there was little or no labelled N in the fixed fractions, probably because the organism-N added was easily mineralized and nitrified. A mean of 0.64 for AN value, the fraction of N mineralized in the killed microbial biomass, was obtained with inclusion of the net increase of fixed NH4 +-N. The corresponding value calculated with exclusion of the net increase of fixed NH4 +-N was 0.46. It was concluded that ammonium fixation was a problem in determination of KN, particularly for soils with a high ammonium fixation capacity. Abstract:In freshwaters, algal species are exposed to different inorganic nitrogen (Ni) sources whose incorporation varies in biochemical energy demand. We hypothesized that due to the lesser energy requirement of ammonium ( <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> )-use, in contrast to nitrate ( <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> )-use, more energy remains for other metabolic processes, especially under CO2- and phosphorus (Pi) limiting conditions. Therefore, we tested differences in cell characteristics of the green alga Chlamydomonas acidophila grown on <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> or <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> under covariation of CO2 and Pi-supply in order to determine limitations, in a full-factorial design. As expected, results revealed higher carbon fixation rates for <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> -grown cells compared to growth with <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> under low CO2 conditions. <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> -grown cells accumulated more of the nine analyzed amino acids, especially under Pi-limited conditions, compared to cells provided with <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> . This is probably due to a slower protein synthesis in cells provided with <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> . In contrast to our expectations, compared to <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> -grown cells <math><msubsup><mtext>NO</mtext> <mrow><mn>3</mn></mrow> <mo>-</mo></msubsup> </math> -grown cells had higher photosynthetic efficiency under Pi-limitation. In conclusion, growth on the Ni-source <math><msubsup><mtext>NH</mtext> <mrow><mn>4</mn></mrow> <mo>+</mo></msubsup> </math> did not result in a clearly enhanced Ci-assimilation, as it was highly dependent on Pi and CO2 conditions (replete or limited). Results are potentially connected to the fact that C. acidophila is able to use only CO2 as its inorganic carbon (Ci) source.

Abstract:Organic nitrogen and inorganic nitrogen were the main nitrogen sources in the north and south of Dianchi Lake, respectively. Thus, the effect of the different concentration (0.5, 1.0, 2.0, 5.0, 10, 20, 50 mg/L) of ammonium nitrogen (NH4-N) and alanine on growth of Microcystis aeruginosa and Scenedesmus quadricauda was investigated. In NH4Cl vitro co-culture, M. aeruginosa grew better than S. quadricauda, and its chlorophyll a increased to 2893.94 ug/L in 2 mg/L NH4Cl group. In vitro alanine treatments, S. quadricauda’s chlorophyll a was measured at 5034.34 ug/L in 50 mg/L alanine, which was higher than M. aeruginosa. M. aeruginosa in NH4Cl vitro monocultures showed better cell structure in 20 mg/L NH4Cl, and its chlorophyll a was higher than that of the corresponding concentration of alanine. The photosynthetic activity (Fv/Fm), the maximum electron transfers rate (ETRmax), and the saturated light intensity point (Ik) of M. aeruginosa increased with the ammonium concentration, and decrease with the alanine concentration, indicating that M. aeruginosa can tolerate higher concentrations of ammonium chloride. In high concentrations of NH4Cl, S. quadricauda’s cell was seriously damaged, but of alanine alone, it was intact. Fv/Fm, ETRmax, and Ik of S. quadricauda increased with the alanine concentrations, showing that S. quadricauda makes better use of organic nitrogen.
