Abstract:Abstract 3D printing using conventional stereolithography is challenging because the polymerized layers adhere to the solid constraining interface. The mechanical separation forces lead to poor process reliability and limit the geometrical design space of the printed parts. Here, these challenges are overcome by utilizing a static inert immiscible liquid below the resin as the constraining interface. We elucidate the mechanisms that enable the static liquid to mitigate stiction in both discrete layer-by-layer and continuous layerless growth modes. The inert liquid functions as a dewetting interface during the discrete growth and as a carrier of oxygen to inhibit polymerization during the continuous growth. This method enables a wide range of process conditions, such as exposure and resin properties, which facilitates micrometer scale resolutions and dimensional accuracies above 95%. We demonstrate multi-scale microstructures with feature sizes ranging from 16 μm to thousands of micrometers and functional devices with aspect ratios greater than 50:1 without using sacrificial supports. This process can enable additive 3D microfabrication of functional devices for a variety of applications.
Beyond 100 nm Resolution in 3D Laser Lithography: Post Processing Solutions

Abstract:Laser polymerization has emerged as a direct writing technique allowing the fabrication of complex 3D structures with microscale resolution. The technique provides rapid prototyping capabilities for a broad range of applications, but to meet the growing interest in 3D nanoscale structures the resolution limits need to be pushed beyond the 100 nm benchmark, which is challenging in practical implementations. As a possible path towards this goal, a post processing of laser polymerized structures is presented. Precise control of the cross-sectional dimensions of structural elements as well as tuning of an overall size of the entire 3D structure was achieved by combining isotropic plasma etching and pyrolysis. The smallest obtainable feature sizes are mostly limited by the mechanical properties of the polymerized resist and the geometry of 3D structure. Thus the demonstrated post processing steps open new avenues to explore free form 3D structures at the nanoscale.

Abstract:Fabrication of microfluidic devices by soft lithography is by far the most popular approach due to simplicity and low cost. In this approach PDMS (polydimethylsiloxane) is cast on a photoresist master to generate replicas that are then sealed against glass slides using oxygen plasma. In this work, we demonstrated fabrication of soft photolithography masters using lamination of ADEX dry film as an alternative to the now classic SU-8 resist masters formed by spin coating. Advantages of using ADEX dry film include the easily-achievable uniform thickness without edge bead; simplicity of the process with significant time savings due to non-sticky nature of the film; and fewer health concerns due to less toxic developing solution and antimony-free composition. As we demonstrate, the process can be performed in a low-cost improvised fabrication room in ambient light, in place of a conventional yellow-light cleanroom environment. We believe this approach holds the promise of delivering state-of-the-art microfluidic techniques to the broad field of biomedical and pharmaceutical research.

Abstract:Abstract Conventional manufacturing techniques to fabricate microfluidic chips, such as soft lithography and hot embossing process, have limitations that include difficulty in preparing multiple-layered structures, cost- and labor-consuming fabrication process, and low productivity. Digital light processing (DLP) technology has recently emerged as a cost-efficient microfabrication approach for the 3D printing of microfluidic chips; however, the fabrication resolution for microchannels is still limited to sub-100 microns at best. Here, we developed an innovative DLP printing strategy for high resolution and scalable microchannel fabrication by dosing- and zoning-controlled vat photopolymerization (DZC-VPP). Specifically, we proposed a modified mathematical model to precisely predict the accumulated UV irradiance for resin photopolymerization, thereby providing guidance for the fabrication of microchannels with enhanced resolution. By fine-tuning the printing parameters, including optical irradiance, exposure time, projection region, and step distance, we can precisely tailor the penetration irradiance stemming from the photopolymerization of the neighboring resin layers, thereby preventing channel blockage due to UV overexposure or compromised bonding stability owing to insufficient resin curing. Remarkably, this strategy can allow the preparation of microchannels with cross-sectional dimensions of 20 μm × 20 μm using a commercial printer with a pixel size of 10 μm × 10 μm; this is significantly higher resolution than previous reports. In addition, this method can enable the scalable and biocompatible fabrication of microfluidic drop-maker units that can be used for cell encapsulation. In general, the current DZC-VPP method can enable major advances in precise and scalable microchannel fabrication and represents a significant step forward for widespread applications of microfluidics-based techniques in biomedical fields.

Abstract:Mask Projection micro-Stereolithography (MPμSL) is an additive manufacturing technique capable of producing solid parts with micron-scale resolution from a vat of photocurable liquid polymer resin. Although the physical mechanism remains the same, the process differs from traditional laser-galvanometer based stereolithography (SL) in its use of a dynamic mask UV projector, or digital light processor (DLP), which cures each location within each 3D layer at the same time. One area where MPµSL has garnered considerable attention is in the field of microfluidics and Lab-on-a-Chip, where complex multistep microfabrication techniques adopted from the semiconductor industry are still widely used, and where MPµSL offers the ability to fabricate completely encapsulated fluidic channels in a single step and at low cost [1–3]. However, a significant obstacle exists in the prevention of channel blockage due to overcuring of the polymer resin [4, 5]. Overcuring can be attributed to the so-called ‘back side effect’ [2] which occurs during the build process as light from successive layers penetrates into the resin to a depth greater than the layer thickness. This effect is most prevalent in channels or features oriented horizontally (in a parallel plane to that of the build platform). Currently there are two main approaches in controlling the cure depth; 1. the chemical approach, which involves doping the resin material with a chemical light absorber [6–8]; and 2. by improving the system's hardware and optical elements to improve the homogeneity of the light dosage and control the cure depth [9]. Here we investigate a third approach through modification of the 3D CAD file prior to printing to mitigate for UV light leakage from successive build layers. Although used here in conjunction with the MPμSL technique, this approach can be applied to a range of SL techniques to improve printer resolution and enable production of internal features with higher dimensional accuracy.
High Resolution Dual Material Stereolithography for Monolithic Microdevices

Abstract:Functional 3D components such as perfusion channels and mechanical actuation elements at cellular length scales can support cell survival and tissue maturation in tissue modeling devices. These advanced requirements call for increasingly complex materials and 3D fabrication methods. Here, a high‐resolution dual‐material 3D printing concept is developed, where distinct materials are produced locally by orthogonal chemical reactions depending on the illumination wavelength. A tough, stiff epoxy network results from cationic polymerization in UV light, while a soft and diffusion‐open hydrogel forms by free‐radical polymerization initiated by blue light. Thus, dual‐exposure allows for selection of material properties in every voxel, while retaining the 3D design flexibility associated with stereolithography. This enables single‐process fabrication of devices integrating mechanically stable chip‐to‐world interconnects and compliant, diffusion‐open perfusable channel components of 150 µm in width and height, while also allowing structural and mechanical feature dimensions down to 60 µm. A perfusion chip capable of creating a stable uniaxial chemical gradient by passive dye diffusion through hydrogel sections, and a negative Poisson ratio structure based on the interplay between stiff rotators and compliant hinges, are manufactured as proof‐of‐concept microdevices. Lastly, week‐long culture of hydrogel‐encapsulated human liver cells demonstrates the cytocompatibility of both materials. A dual material stereolithographic 3D printing method based on orthogonal radical or cationic photopolymerization is presented. Two distinct materials can be locally polymerized in parallel, controlled by the illumination wavelength. The method is demonstrated by combining a diffusion‐open poly(ethylene glycol) diacrylate (PEGDA) hydrogel and a tough, diffusion‐closed interpenetrating PEGDA/epoxy material at high resolutions to create complex monolithic devices.

Abstract:Microfluidics is a crucial technology in biological and medical fields, and its traditional fabrication methods include soft photolithography poly(dimethyl siloxane) (PDMS) technology and polymethyl methacrylate (PMMA) injection moulding technology. However, these techniques face challenges in manual alignment and bonding issues when they are used to manufacture microfluidic chips with complex flow channels. To overcome these limitations, digital light processor (DLP) 3D printing technology has been proposed as a promising alternative. However, this method is prone to problems such as channel blockage or shape distortion caused by the optical proximity effect or curing light transmission during the manufacture of microfluidic chips with small-diameter channels. This paper introduces a method to enhance the manufacturing technology by controlling the local greyscale of the projection image during DLP 3D printing. This allows for modulation of the curing light intensity, thus reducing the optical proximity effect and the impact of transmitted light on extra areas. The methodology can be employed in the production of microfluidic devices with circular and square apertures in the microchannels, utilizing commercially accessible general-purpose resins that are readily available in the market. The printed microfluidic devices show improved quality, with a channel size that closely matches the preset size, in contrast to significant channel blockage in devices printed without greyscale optimization. This method provides a new approach for enhancing the quality of low-cost microfluidic chip production and improves the print quality based on the resolution of the printer and resin used.

Abstract:With the recent advances of additive manufacturing, three-dimensional (3D) bioprinting has enabled the printing of biocompatible materials, cells and supporting components into complex 3D functional living tissues to address the need for tissues and organs suitable for transplantation. As vascularization is generally identified as one of the main technological barriers for building 3D organs, the freeform printing of 3D biological vascular constructs is of great importance to prove the overall feasibility of 3D bioprinting techniques. In this study, vascular-like constructs are fabricated by both laser- and inkjet-based printing techniques using a liquid support-based approach, which utilizes a calcium chloride-based solution as a cross-linking agent and buoyancy-based support material. 3D constructs such as Y-shaped tubular constructs have been successfully fabricated from alginate-based and cell-laden bioinks with a satisfactory post-printing cell viability.

Abstract:Negative features in microdevices find a wide range of applications. The process of 3D printing has revolutionized their fabrication due to the combination of good resolution and integration capability. Herein, we report on a systematic study of the effects of materials and print directions on the 3D printing of microfluidic channels as negative features under PolyJet technology. Specifically, the Statasys Objet500 printer was used for this study. We printed two sets of chips (n=10 each), each of which contains channel pairs of a high-contrast reference material and a sacrificial material, respectively. Both materials were embedded in a clear photopolymer resin. The channel pairs ranged in planned width from 64 to 992 μm. To explore the effect on print orientation, channels were printed either parallel or perpendicular with respect to the jetting head's movement. The width of each channel of a pair was compared for each planned width and each combination of materials. The effect of print orientation on channel morphology was also investigated. We found that reproducibility and accuracy were highest at a planned channel width of approximately ≥600 μm and that channel morphology was most suitable when the jetting head of the printer moved parallel to the channel's longitudinal axis. The results should be of interest to any users who wish to create negative features using PolyJet 3D technology.

Microfluidic devices manufacturing with a stereolithographic printer for biological applications

Abstract:Stereolithographic printers have revolutionized many manufacturing processes with their capacity to easily produce highly detailed structures. In the field of microfluidics, this technique avoids the use of complex steps and equipment of the conventional technologies. The potential of low force stereolithography technology is analysed for the first time using a Form 3B printer and seven printing resins through the fabrication of microchannels and pillars. Manufacturing performance of internal and superficial channels and pillars is studied for the seven printing resins in different configurations. A complete characterization of printed structures is carried out by optical, confocal and SEM microscopy, and EDX analysis. Internal channels with unobstructed lumen are obtained for diameters and angles greater than 500 μm and 60°, respectively. Outward and inward superficial channels in the range of hundreds of microns can be fabricated with an accurate profile, printing them with a perpendicular orientation respect to the base, allowing a proper uncured resin evacuation. Outward channels are replicated by soft lithography using polydimethylsiloxane. Clear, Model and Tough resins show a good behaviour to be used as master, but Amber and Dental resins present a poor topology transference from the master to the replica. According to the needs of devices used for biological and biomedical research, transparency as well as superficial biocompatibility of some resins is evaluated. Human umbilical vein endothelial cells (HUVEC) adhesion is confirmed on Amber, Dental and Clear resins, but these cells were only able to grow and progress as a cell culture over the Amber resin. Therefore, Amber showed an adequate biocompatibility, in terms of cell adhesion and growth for HUVEC.
Rapid High-Resolution Visible Light 3D Printing

Abstract:Light-driven 3D printing to convert liquid resins into solid objects (i.e., photocuring) has traditionally been dominated by engineering disciplines, yielding the fastest build speeds and highest resolution of any additive manufacturing process. However, the reliance on high-energy UV/violet light limits the materials scope due to degradation and attenuation (e.g., absorption and/or scattering). Chemical innovation to shift the spectrum into more mild and tunable visible wavelengths promises to improve compatibility and expand the repertoire of accessible objects, including those containing biological compounds, nanocomposites, and multimaterial structures. Photochemistry at these longer wavelengths currently suffers from slow reaction times precluding its utility. Herein, novel panchromatic photopolymer resins were developed and applied for the first time to realize rapid high-resolution visible light 3D printing. The combination of electron-deficient and electron-rich coinitiators was critical to overcoming the speed-limited photocuring with visible light. Furthermore, azo-dyes were identified as vital resin components to confine curing to irradiation zones, improving spatial resolution. A unique screening method was used to streamline optimization (e.g., exposure time and azo-dye loading) and correlate resin composition to resolution, cure rate, and mechanical performance. Ultimately, a versatile and general visible-light-based printing method was shown to afford (1) stiff and soft objects with feature sizes <100 μm, (2) build speeds up to 45 mm/h, and (3) mechanical isotropy, rivaling modern UV-based 3D printing technology and providing a foundation from which bio- and composite-printing can emerge.Liquid resins that rapidly harden upon exposure to low-energy visible light are developed and applied to high-resolution projection-based 3D printing across the color spectrum.This article has not yet been cited by other publications.

Upconversion 3D printing enables single-immersion multi-material stereolithography

Abstract:Photopolymer 3D printing techniques have a common limitation: the layers have to be deposited in a sequential order, which constrains multi-material fabrication to techniques where the vats have to be exchanged many times, in addition to repeating the cleaning step for each vat change. In this work, we present a technique that allows multi-material stereolithography by overcoming the layer-by-layer limitation, enabling one single immersion per resin. This is enabled by selective crosslinking of voxels at any depth and position inside the stereolithography (SLA) resin vat. The concept uses invisibility windows and upconversion to achieve selective volumetric crosslinking. This enables printing inside and through previously 3D printed parts. A photopolymer resin is developed, in which a visible light photoinitiator (fluorinated diphenyl titanocene) with a broad absorption and an optical absorber (naphthalimide class dye, maximum absorption at 445 nm) are carefully matched with the upconversion emissions of lanthanide-doped phosphors (both micron-sized phosphors and customized core/shell/shell nanoparticles). The lateral printing resolution of the formulation was found to be 134 ± 13 μm, which was improved to 103 ± 11 μm through the addition of light-absorbing dye. The vertical resolution was defined by the layer thickness achieved by lowering the build platform, which was set to 100 μm. The NIR light used for the excitation of upconversion phosphors enabled enhanced penetration depths of up to 5.8 cm, which allowed SLA printing of interwoven multi-color samples, multi-material rigid/flexible (acrylate/elastomer) samples and dielectric/metallic plated samples. This opens up a myriad of new possibilities, such as 3D printing objects inside cavities in a different material, restoration of broken objects and artefacts, 3D circuitry, insitu bioprinting, etc.

Abstract:Fast, continuous, 3D printing Although three-dimensional (3D) printing is now possible using relatively small and low-cost machines, it is still a fairly slow process. This is because 3D printers require a series of steps to cure, replenish, and reposition themselves for each additive cycle. Tumbleston et al. devised a process to effectively grow solid structures out of a liquid bath. The key to the process is the creation of an oxygen-containing “dead zone” between the solid part and the liquid precursor where solidification cannot occur. The precursor liquid is then renewed by the upward movement of the growing solid part. Rapid Micromolding of Sub-100 µm Microfluidic Channels Using an 8K Stereolithographic Resin 3D Printer

Abstract:Engineering microfluidic devices relies on the ability to manufacture sub-100 micrometer fluidic channels. Conventional lithographic methods provide high resolution but require costly exposure tools and outsourcing of masks, which extends the turnaround time to several days. The desire to accelerate design/test cycles has motivated the rapid prototyping of microfluidic channels; however, many of these methods (e.g., laser cutters, craft cutters, fused deposition modeling) have feature sizes of several hundred microns or more. In this paper, we describe a 1-day process for fabricating sub-100 µm channels, leveraging a low-cost (USD 600) 8K digital light projection (DLP) 3D resin printer. The soft lithography process includes mold printing, post-treatment, and casting polydimethylsiloxane (PDMS) elastomer. The process can produce microchannels with 44 µm lateral resolution and 25 µm height, posts as small as 400 µm, aspect ratio up to 7, structures with varying z-height, integrated reservoirs for fluidic connections, and a built-in tray for casting. We discuss strategies to obtain reliable structures, prevent mold warpage, facilitate curing and removal of PDMS during molding, and recycle the solvents used in the process. To our knowledge, this is the first low-cost 3D printer that prints extruded structures that can mold sub-100 µm channels, providing a balance between resolution, turnaround time, and cost (~USD 5 for a 2 × 5 × 0.5 cm3 chip) that will be attractive for many microfluidics labs.

Abstract:Mycobacterium tuberculosis (Mtb) infections are causing serious health concerns worldwide. Antituberculosis drug resistance threatens the current therapies and causes further need to develop effective antituberculosis therapy. GlmU represents an interesting target for developing novel Mtb drug candidates. It is a bifunctional acetyltransferase/uridyltransferase enzyme that catalyzes the biosynthesis of UDP-N-acetyl-glucosamine (UDP-GlcNAc) from glucosamine-1-phosphate (GlcN-1-P). UDP-GlcNAc is a substrate for the biosynthesis of lipopolysaccharide and peptidoglycan that are constituents of the bacterial cell wall. In the current study, structure and ligand based computational models were developed and rationally applied to screen a drug-like compound repository of 20,000 compounds procured from ChemBridge DIVERSet database for the identification of probable inhibitors of Mtb GlmU. The in vitro evaluation of the in silico identified inhibitor candidates resulted in the identification of 15 inhibitory leads of this target. Literature search of these leads through SciFinder and their similarity analysis with the PubChem training data set (AID 1376) revealed the structural novelty of these hits with respect to Mtb GlmU. IC50 of the most potent identified inhibitory lead (5810599) was found to be 9.018 ± 0.04 μM. Molecular dynamics (MD) simulation of this inhibitory lead (5810599) in complex with protein affirms the stability of the lead within the binding pocket and also emphasizes on the key interactive residues for further designing. Binding site analysis of the acetyltransferase pocket with respect to the identified structural moieties provides a thorough analysis for carrying out the lead optimization studies.
Development of a Custom-Made 3D Printing Protocol with Commercial Resins for Manufacturing Microfluidic Devices

Abstract:The combination of microfluidics and photo-polymerization techniques such as stereolithography (SLA) has emerged as a new field which has a lot of potential to influence in such important areas as biological analysis, and chemical detection among others. However, the integration between them is still at an early stage of development. In this article, after analyzing the resolution of a custom SLA 3D printer with commercial resins, microfluidic devices were manufactured using three different approaches. First, printing a mold with the objective of creating a Polydimethylsiloxane (PDMS) replica with the microfluidic channels; secondly, open channels have been printed and then assembled with a flat cover of the same resin material. Finally, a closed microfluidic device has also been produced in a single process of printing. Important results for 3D printing with commercial resins have been achieved by only printing one layer on top of the channel. Abstract:Soft lithography microfluidics offer many benefits over conventional biological assays; however, the impact this field is inhibited by the lack of widespread adoption of this technology in part due to prohibitive cost and fabrication time. Recent improvements in three-dimensional (3D) printing technologies such as digital light processing (DLP) printing offer a cost-effective and rapid prototyping solution to microfluidic fabrication. Limited information is available about how 3D printing parameters and resin cytocompatibility impact the performance of 3D printed molds for fabrication of polydimethylsiloxane (PDMS)-based microfluidics for cellular studies. Using a low-cost, commercially available DLP 3D printer, we assess the cytocompatibility of several resins, optimize printer settings and characterize minimum feature size of our system. We demonstrate the applications of DLP printing for soft lithography microfluidics by developing four assays to characterize cell viability, drug response, establish concentration gradients, and monitor live-cell 3D invasion into a hydrogel.

Light-oriented 3D printing of liquid crystal/photocurable resins and in-situ enhancement of mechanical performance

Abstract:Additive manufacturing technology has significantly impacted contemporary industries due to its ability to generate intricate computer-designed geometries. However, 3D-printed polymer parts often possess limited application potential, primarily because of their weak mechanical attributes. To overcome this drawback, this study formulates liquid crystal/photocurable resins suitable for the stereolithography technique by integrating 4'-pentyl-4-cyanobiphenyl with a photosensitive acrylic resin. This study demonstrates that stereolithography facilitates the precise modulation of the existing liquid crystal morphology within the resin. Furthermore, the orientation of the liquid crystal governs the oriented polymerization of monomers or prepolymers bearing acrylate groups. The products of this 3D printing approach manifest anisotropic behavior. Remarkably, when utilizing liquid crystal/photocurable resins, the resulting 3D-printed objects are approximately twice as robust as those created using commercial resins in terms of their tensile, flexural, and impact properties. This pioneering approach holds promise for realizing autonomously designed structures that remain elusive with present additive manufacturing techniques.

Abstract:Three-dimensional printing technology has been implemented in microfluidic mold fabrication due to its freedom of design, speed, and low-cost fabrication. To facilitate mold fabrication processes and avoid the complexities of the soft lithography technique, we offer a non-sacrificial approach to fabricate microscale features along with mesoscale features using Stereolithography (SLA) printers to assemble a modular microfluidic mold. This helps with addressing an existing limitation with fabricating complex and time-consuming micro/mesoscale devices. The process flow, optimization of print time and feature resolution, alignments of modular devices, and the advantages and limitations with the offered technique are discussed in this paper.

Abstract:Organ-on-chip and Lab-on-chip are microfluidic devices widely applied in the biomedical field. They are traditionally produced by soft lithography: starting from a mold fabricated by optical photolithography, a Polydimethylsiloxane (PDMS) device is obtained by casting and baking. While this technique offers the possibility to produce features with high resolution, it is not flexible enough to respond to the necessity of customization and prototyping. In this study, we propose as alternative the production of devices by digital light processing (DLP), a vat photopolymerization technology, in combination with a commercially available, biocompatible resin. Studying the process factors by a statistical methodology called Design of Experiment (DoE), we were able to achieve small features with high aspect ratio (60). DoE method allowed us to have a deep understanding of the process without the need of any physical inspection of the involved phenomena, and to generate empirical models, correlating the process factors to the dimensions of the final printed object. We proved that this optimization was beneficial also in terms of transparency (evaluated by UV-Vis spectrophotometry), and mechanical strength (evaluated by a compression test) of the printed resin. Finally, a proof-of-concept microfluidic device was fabricated, sealed to a PDMS membrane through an oxygen plasma treatment, and tested against leakage on a microfluidic circuit for one week. As result, we proved that DLP printing is not only a suitable method to develop microfluidic devices, but if correctly optimized it can also reproduce small features in the order of tens of micrometers rapidly.
