Abstract:This study investigated the effects of low-temperature and high-pressure collaborative and conventional air freezing (CAF) on myofibrillar protein (MP) characteristics from grass carp. Turbidity, solubility, and particle size analysis showed that the pressure shift freezing (PSF) treatment could inhibit the formation of large MP aggregates to a certain extent during freezing. Chemical interaction studies unveiled that PSF treatment not only mitigates damage to the inherent structure of MP during freezing but also enhances its crosslinking ability. Microstructure analysis revealed a densely woven fibrous protein network structure within the MP gel in the PSF group. MP gel with fiber shape network structure in the PSF group exhibited stronger water-holding capacity and mechanical properties compared to the CAF group having an incomplete protein network structure. This suggests that the combined application of low-temperature and high-pressure treatment holds promising potential for utilization in the surimi gel processing industry and aquatic products processing.Industrial relevance: The quality and safety of food are closely related to the commodity value of food. Combined low-temperature and high-pressure treatment can enhance the commodity value of low-value freshwater fish products and has potential in surimi gel new product development. This study provides theoretical guidance for high-pressure and low-temperature collaborative processing and industrial production of related aquatic products.
High-Release, Residue-Free Polysaccharide Hydrogel for Refrigerated Food Preservation.

Abstract:Hundreds of millions of tons of food resources are wasted annually due to microbial contamination. Effective food packaging can prevent food contamination and wastage. However, traditional food packaging has the problem of low release of bioactive substances. This study aimed to prepare a pH-responsive polysaccharide hydrogel (GDPP) by double cross-linking of ester and hydrogen bonds that could result in a high release of bioactive substances and no residual peeling. The infrared results showed the existence of ester bonds in the hydrogel, and the scanning electron microscopy results showed the porous network structure of the hydrogel. The results of texture profile analysis and self-healing tests showed that GDPP-1 has good mechanical and self-healing properties. Moreover, the ester bond of the hydrogel broke in response to the pH in the environment, improving the swelling and release properties of the hydrogel. The equilibrium swelling ratio of GDPP-1 was greater than 1000%, and the release rate of bioactive substances was more than 80%. Notably, the results of peeling experiments showed that only 0.1 N external force was needed to separate the hydrogel from the salmon, and no residue was observed on the salmon surface. The final freshness test results showed that the hydrogel effectively prolonged the shelf life of refrigerated salmon for 3-6 days. These findings indicated that hydrogels could be used in food packaging to extend the shelf life of refrigerated food. Furthermore, their advantages of low cost and simple preparation can better meet the needs of food industry applications.

Abstract:In this work, konjac glucomannan (KGM)-based film reinforced with pullulan (PL) and acai berry extract (ABE) was developed by solvent casting method. The as-prepared films performed pH-sensitive properties, which can be potentially applied for fish freshness detection. Rheology, Fourier transform infrared spectroscopy (FT-IR), X-ray diffraction (XRD), scanning electron microscopy (SEM) were used to characterize chemical structure and morphology of ABE-loaded KGM/PL (KP) films (KP-ABE). FT-IR spectrum indicated that hydrogen bond dominated the formation of KP-ABE films. Adding PL contributed to enhanced mechanical properties of KGM film with increased tensile strength (TS) from 21.25 to 50.27 MPa and elongation at break (EAB) from 10.64 to 19.19 %. Incorporating ABE upgraded flexibility, UV-shielding, thermostability, water barrier (decreased Water vapor permeability (WVP) from 2.07 to 1.67 g·mm/m2·day kPa), antioxidant, and antibacterial ability of KP films, but weakened TS. In addition, KP-ABE films can reflect fish freshness in real time through color variability. Therefore, KP-ABE films exhibited potential applications in intelligent food packaging materials.

Abstract:Coating preservation has a remarkable effect on the preservation of aquatic products. This work prepared a composite coating using konjac glucomannan (KGM) as the film-forming matrix and ε-polylysine hydrochloride (ε-PL) and ferulic acid (FA) as the preservative. Three types of treated sea bass (KGM, KGM-ε-PL, and KGM-ε-PL-FA) and untreated sea bass were stored at 4 °C for 20 days to compare freshness changes under different treatment conditions. The results showed that the surface color and texture of sea bass in refrigerated storage changed dramatically and deteriorated as storage time increased. The composite coating treatment was significantly different from the control group. Using Gas-phase ion migration spectrometry (GC-IMS) technology, 32 volatile compounds, such as aldehydes, alcohols, and ketones, were found in fillets during flavor quality analysis. The composite coating can successfully inhibit the formation of odor compounds such as 2-nonenone, isoamyl alcohol monomer, ammonia, and trimethylamine, delaying the deterioration of fish and improving freshness. Among them, KGM-ε-PL-FA composite coating has the most remarkable preservation performance, which significantly inhibits the occurrence of rotten odor, and has a potential application prospect in the field of food preservation.

Abstract:In this study, gelatin/wheat gliadin electrospun films containing different amounts of chlorogenic acid (CA; 0–150 mg) were fabricated and characterized, and the protective effect of the films with CA on food preservation was investigated, specifically for grass carp fillets. The results of the morphology and structure of the films indicated that CA was successfully encapsulated in the film and interacted with proteins. Thermal stability, mechanical properties, and water contact angle (WCA) analysis suggested that the film had good stability and hydrophobicity. Furthermore, films with CA exhibited good antioxidant and antibacterial activities and these properties were enhanced with increasing CA content. The antioxidant activity was greater than 90% when the amount of CA added was greater than 100 mg. The CA release mechanism of the film supplemented with 125 mg of CA (CA125) was in line with the first-order kinetic model, and CA release reached 84.38% ± 0.26% at 16 h. Furthermore, CA125 was used to perform 10-day preservation assays. During the storage period, the quality of grass carp fillets deteriorated and the film with CA exhibited a better protective effect on muscle quality, thus, possibly delaying the deterioration of the fish fillets and prolonging their acceptance period, demonstrating its applicability in the protection of the freshness of aquatic products.

Abstract:Hydrogels have become promising materials for food packaging due to their unique microstructure. However, hydrogel materials suitable for seafood preservation have rarely been reported. In this study, a tamarind polysaccharide-polyvinyl alcohol hydrogel with the ability to maintain seafood freshness was prepared and characterized. The hydrogel possesses quick self-healing, good tissue fitting, and freezing tolerance capability. Moreover, a peeling force of only 0.1 N between the hydrogel and the fillet tissue confirmed the non-stick properties. The FTIR characteristic peak at 1600 cm-1 and 1450 cm-1 proved the ester bond-based chemical cross-linking of the hydrogel. Release profiles at pH 6.0 to 8.0 verified the pH-responsive release of quorum-quenching (QQ) enzymes over 120 h, which enabled the hydrogel to achieve biofilm and protease inhibitory activities. In vivo spoilage tests showed that the shelf life of hydrogel-coated red snapper fillets was extended by >3 days. These results illustrate the potential of the prepared hydrogel as functional packaging for seafood preservation.

Abstract:This work is aimed to investigate the effects of molecular weights on physicochemical properties of fish gelatins and their potential application to food capsule film fabrication. Firstly, high/medium/low component fish gelatins (HCG/MCG/LCG) were obtained by ultrafiltration. Physicochemical property assay results showed that high molecular weight content of gelatin contributed to the increase in gelling strength, viscosity, melting point, and denaturation temperature due to extensive inter–intra molecular interactions. Meanwhile, parameters (temperature, weight ratio, and drying time) had been comprehensively investigated to determine the effects of three molecular weights of gelatins on capsule film manufacture. MCG-derived capsule film was found to achieve good balance among tensile strength, elongation at break, and dissolution rate constant. Furthermore, 90-day acceleration storage stability experiment suggested that MCG showed an appropriate ɛ -amino acid residue content and crosslink density. This work provides a comprehensive insight into the effect of molecular weights on physicochemical properties of fish gelatins and their application in food capsule film manufacture.
Effect of konjac glucomannan-based preservation pads on quality changes in refrigerated large yellow croaker (Pseudosciaena crocea)

Abstract:The purpose of this study was to evaluate the preservation effects of konjac glucomannan (KGM)/oregano essential oil (OEO) Pickering emulsion-based pads (K/OPE pads) on large yellow croaker (Pseudosciaena crocea) fillets stored at 4 °C. The K/OPE pads were fabricated using a freeze-drying technique. The homogeneous distribution of the OEO Pickering emulsions in the KGM matrix was observed using scanning electron microscopy. Fourier transform infrared spectroscopy confirmed that the OEO emulsions were encapsulated in the KGM and there was hydrogen bonding interaction between them. Compared with the KGM pads, the K/OPE pad groups demonstrated enhanced antioxidant and antimicrobial properties. When the content of OPE was increased from 20 % to 40 %, the antioxidant performance of the K/OPE pads increased from 48.09 % ± 0.03 % to 86.65 % ± 0.02 % and the inhibition range of Escherichia coli and Staphylococcus aureus increased to 13.84 ± 0.81 and 16.87 ± 1.53 mm, respectively. At the same time, K/OPE pads were more effective in inhibiting the formation of total volatile alkaline nitrogen and the production of thiobarbituric acid-reactive substances, thereby helping in reducing water loss and maintaining the muscle tissue structure of fish fillets for a longer storage time. Consequently, these K/OPE40 pads extended the shelf life of the fish fillets by an additional 4 days and delayed spoilage during refrigerated storage. The findings suggest that the K/OPE pads can effectively safeguard the quality of refrigerated large yellow croaker fillets, presenting their potential as an active packaging material in the fish preservation industry.
Effects of Enzymatic Konjac Glucomannan Hydrolysates on Textural Properties, Microstructure, and Water Distribution of Grass Carp Surimi Gels

Abstract:This present work investigated the influence of konjac glucomannan (KGM) enzymatic hydrolysates on the textural properties, microstructure, and water distribution of surimi gel from grass carp (Ctenopharyngodon idellus). The molecular weight (Mw) of KGM enzymatic hydrolyzed by β-dextranase degraded from 149.03 kDa to 36.84 kDa with increasing enzymatic time. In the microstructure of surimi gels, KGM enzymatic hydrolysates with higher Mw showed entangled rigid-chains, while KGM enzymatic hydrolysates with lower Mw (36.84 kDa) exhibited swelled fragments. The hardness of surimi gel with a decline in KGM Mw exhibited first increasing then decreasing trends, while the whiteness of surimi gel increased. When KGM Mw decreased, the immobile water percentage of total signals decreased from 96.7% to 93.6%, and mobile water increased from 3.03% to 6.37%. In particular, the surimi gel with the addition of K2 showed better gel strength and water distributions. KGM enzymatic hydrolysates are expected to be used as a low-calorie healthy gel enhancer in surimi processing.
Insight into konjac glucomannan-retarding deterioration of steamed bread during frozen storage: Quality characteristics, water status, multi-scale structure, and flavor compounds

Abstract:Konjac glucomannan (KGM), a water-soluble hydrocolloid, holds considerable potential in the food industry, especially for improving the quality and nutritional properties of frozen products. This study explored the alleviative effect of KGM on the quality characteristics, water status, multi-scale structure, and flavor compounds of steamed bread throughout frozen storage. KGM significantly improved the quality of steamed bread by slowing down the decrease in water content and the increase in water migration while maintaining softness and taste during frozen storage. Notably, KGM also delayed amylopectin retrogradation and starch recrystallization, thus preserving the texture and structure of the steamed bread. At week 3, the microstructure of the steamed bread with 1.0 % KGM remained intact, with the lowest free sulfhydryl content. Additionally, heat map analysis revealed that KGM contributed to flavor retention in steamed bread frozen for 3 weeks. These results indicate that KGM holds promise as an effective cryoprotectant for improving the quality of frozen steamed bread.
The application of ice glazing containing D-sodium erythorbate combined with vacuum packaging to maintain the physicochemical quality and sweet/umami non-volatile flavor compounds of frozen stored large yellow croaker (Pseudosciaena crocea)

Abstract:Ice glazing containing 0.3 % D-sodium erythorbate (DSE), combined with vacuum packaging, was used as a method to maintain the quality of large yellow croaker during frozen storage. This study aimed to assess various aspects, including water properties (water holding capacity and moisture distribution), protein-related characteristics (secondary and tertiary structure of myofibrillar protein), freshness indicators (K value and total volatile base nitrogen (TVB-N)), and non-volatile flavor compounds (free amino acids (FAAs) and nucleotides) in samples stored for 300 days at -23 °C. The results showed that vacuum packaging had a significant inhibitory effect on the growth of ice crystal. Notably, it successfully maintained the cross-sectional area of nearly all ice crystals below 20,000 μm2, effectively curtailing water loss. Simultaneously, the combination of vacuum packaging with the complex ice glaze effectively mitigated the degradation of IMP and free amino acids, maintaining low levels of K value (12.85 %) and TVB-N (11.28 mg N/100 g) throughout the 300-day frozen storage, retaining first-class freshness. Among the various treatment modalities assessed, the combined application of vacuum packaging and 0.3 % DSE-infused ice glazing emerged as the most effective in terms of preservation outcomes. This efficacious combination shows promising potential for the frozen storage of aquatic products and is therefore recommended for practical implementation.

Abstract:Most edible films/coatings including those made of gelatin cannot be used independently as packaging films. In this study, active fish gelatin-based bilayer packaging films were developed and applied to preserve Japanese sea bass. The coating method was applied to produce polylactic acid/tilapia fish gelatin-sodium alginate bilayer film (PLA/FGSA). β-cyclodextrin inclusion complexes of cinnamaldehyde (β-CD-CI) and thymol (β-CD-Ty) were added into the gelatin-based layer of the PLA/FGSA to obtain active bilayer films (PLA/FGSA-CI and PLA/FGSA-Ty). This combination improved the oxygen and water vapor barrier abilities of the films, compared with PLA and edible gelatin-based monolayer films. Both active bilayer films possessed strong antibacterial and antioxidant activities. PLA/FGSA-Ty film showed the most effective inhibition against the growth of Pseudomonas.spp and H2S-producing Shewanella, generation of total volatile basic nitrogen (TVB-N) and thiobarbituric acid reactive substances (TBARS) of fish meat. These results indicate the active bilayer films are promising in the preservation of seafood.

Abstract:Strawberry is a seasonal and regional fruit. Thus, strawberry waste caused by spoilage and decay is an urgent problem that must be solved. Developing hydrogel films (HGF) for multifunctional food packaging can effectively slow down strawberry. Based on the carboxymethyl chitosan/sodium alginate/citric acid with excellent biocompatibility, preservation effect, and ultrafast (10 s) coating on the strawberry surface, HGF specimens were designed and prepared through the electrostatic interaction of opposite charges between polysaccharides. The prepared HGF specimen exhibited excellent low moisture permeability and antibacterial properties. Its lethality rates against both Escherichia coli and Staphylococcus aureus were more than >99 %. The HGF could keep strawberries fresh for up to 8, 19, and 48 days at 25.0, 5.0, and 0 °C, respectively, by delaying the fruits' ripening, dehydration, microbial invasion, and respiration rate. The HGF dissolved and regenerated five times still exhibited good performance. The water vapor transmission rate of the regenerative HGF could reach 98 % of that of the original HGF. The regenerative HGF could maintain the freshness of strawberries for up to 8 days at 25.0 °C. This study provides new insight into an alternative film design for convenient, green, and renewable alternative films to delay perishable fruit spoilage.

Abstract:Konjac glucomannan (KGM) gel has attracted widespread interest in the field of food. However, the low texture properties of gel limit its further application. In this study, the effect of KGM powder under different processing times (8 h, 12 h, 24 h and 48 h) and temperatures (4 °C, −18 °C, −80 °C) on the texture properties of gels were investigated. According to result of texture profile analysis (TPA) and puncture test, the gel prepared by pretreatment showed good texture characteristics. The gel prepared by freezing KGM powder at −18 °C for 8 h has the best gel strength in the sample, which is 1.6 times that of native KGM. The molecular weight of KGM decreased significantly after pretreatment (from 12.30 × 10 5 Da to 0.29 × 10 5 Da). In addition, low temperature and freezing treatment can influence the rheological behavior and regulate water composite in gel. According to experimental result, it can be attributed to that low temperature and the growth of ice crystal can make structures denser, promote KGM molecular chain entangled and increase the number of hydrogen bond. This study provides a convenient and effective method for improving the gel properties.

Abstract:Fish gelatin (FG) is a natural biodegradable biopolymer. However, solution casting, which is commonly used to prepare gelatin films, limits large-scale film production. In this study, melt extrusion (ME) commonly used in traditional plastics was successfully applied to the preparation of gelatin composite films. The non-covalent crosslinking between hydrophobic beeswax (BW) and hydrophilic FG not only reduced the film stickiness during rolling but also decreased the water solubility of the film by 45.17% (p < 0.05). Transglutaminase (TGase) promoted covalent crosslinking between the FG networks, thereby improving the properties of the film. The results of Fourier transform infrared (FTIR) spectroscopy confirmed the crosslinking effect of BW and TGase on the gelatin films, and appropriate crosslinking maintained the forward movement of gelatin in the rotating screw according to a linear mode. The FG–BW–1%TGase film exhibited the lowest water solubility of 36.63% and the highest tensile strength of 6.26 MPa. The co-crosslinking of TGase and BW contributed to the preparation of melt-extruded composite films exhibiting desirable properties and were potential candidates for commercial food packaging.

On the mechanism behind enhanced antibacterial activity of alkyl gallate esters against foodborne pathogens and its application in Chinese icefish preservation

Abstract:The objective of this study was to investigate antibacterial activities and action mode of alkyl gallates against three food-related bacteria. Results show that the length of the alkyl chain plays a critical role in eliciting their antibacterial activities and octyl gallate (GAC8) exhibited an outstanding bactericidal effect against these strains. A possible bactericidal mechanism of GAC8 against E. coli was fully elucidated by analyzing associated changes in cellular functions of E. coli, including assessments of membrane modification and intracellular oxidation state. Our data strongly suggested that GAC8 functions outside and inside the bacterial membrane and causes increased intracellular reactive oxygen species (hydroxyl radicals) and subsequent oxidative damage. We demonstrated that the hydroxyl radical formation induced by GAC8 is the end product of an oxidative damage cellular death pathway involving a transient depletion of NADH, the tricarboxylic acid cycle, intrinsic redox cycling activities, and stimulation of the Fenton reaction. Also, chitosan-based edible films containing GAC8 have unique superiorities for icefish preservation at 4 °C. This research highlights the effectiveness of GAC8 as an attractive antibacterial, which possesses both antioxidant and antibacterial activities and can be used as a multifunctional food additive combined with the benefit of active packaging for food preservations.
Formation of high amylose corn starch/konjac glucomannan composite film with improved mechanical and barrier properties

Abstract:Recently, starch-based packaging materials have become one of research hot points. In the present study, glycerol-plasticized composite films based on high amylose corn starch (HCS) and konjac glucomannan (KGM) were developed. The influence of KGM on the film-forming properties of HCS and the physicochemical properties of the resulting films were characterized by X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), thermogravimetric (TG) analysis and water vapor permeability (WVP). The crystallinity and the proportion of short-range order structure of the films increased first and then declined with the addition of KGM. The micromorphology of the films exhibited the more even texture after KGM was incorporated in. The tensile strength, elongation at break and water resistance of HCS film were also improved significantly. The synergistic effect between HCS and KGM improved the film-forming ability of HCS. The optimal addition amount of KGM was 0.3 %.

Abstract:<p>The present study aims to develop the new composite films by blending Konjac glucomannan (KGM) and pullulan with different ratios and concentrations. The structural, physical, barrier properties and morphology of the films were investigated and the practical use on strawberry preservation at 4 ± 1 °C, 85%±5% relative humidity (RH) and 25 ± 1 °C, 55%±5% RH was evaluated. Fourier transform infrared and scanning electron microscopy indicated the well-dispersion of film matrix was due to the good compatibility of the components. The mechanical and barrier properties of blend films were markedly enhanced although the light transmittance of which were decreased slightly. It was a further proof that 1% (w/v) KGM/pullulan (with the mass ratio of 2:1) blend film could decrease the weight loss significantly and maintain the titratable acidity, soluble solids and skin color on the strawberry preservation, thus improving the qualities of strawberries during storage time and offering a potential alternative to synthetic materials.</p>

Abstract:The designated composite zein edible film containing dill extract and encapsulated dill essential oil can be introduced as an active edible packaging in the shelf life improvement of common carp fillets during cold storage. The aim of this study was to examine the impacts of corn zein edible film (Z) fortified with dill leaves extract (DE) and encapsulated dill essential oil with β‐cyclodextrin (nDEO) on the quality of refrigerated common carp fillet. Gas chromatography/mass spectrometry (GC/MS) analysis showed that the most frequent substances of DEO were apiol (35.1%) and carvone (31.4%), respectively. Designated treatments were as follows: (1) Control (C), (2) Z, (3) Z‐DE, (4) Z‐DEO, (5) Z‐nDEO, (6) Z‐DE‐DEO, and (7) Z‐DE‐nDEO. The physicochemical properties (thickness, moisture percent, tensile strength, elongation at break, Young's modulus, color, morphology, functional groups, and thermal resistance) of the activated films significantly improved (p ≤ .05). The total viable counts, lactic acid bacteria, Enterobacteriaceae, and psychrotrophic bacteria significantly decreased in all wrapped fillets compared to the unwrapped ones (p ≤ .05). Throughout storage period, the wrapped fillets exhibited lower changes in pH, thiobarbituric acid reactive substances, and total volatile‐based nitrogen values than the unwrapped fillets. According to the sensory findings, incorporating DE and nDEO in the zein films created significantly desirable aroma and flavor in the wrapped samples during storage time (p ≤ .05). Encapsulation of DEO with β‐cyclodextrin significantly fortified preservative effects of the films in fish fillets during storage period (p ≤ .05). In conclusion, the designated composite zein edible film containing DE and nDEO can be introduced as an active edible packaging in the shelf life improvement of common carp fillets during cold storage.
