Abstract:This study aimed to assess the effects of β-mannanase supplementation in metabolizable energy (ME)-reduced diets containing xylanase-phytase on performance, fecal score, blood biochemical and immunological profile, apparent total tract digestibility (ATTD), digesta passage rate, fecal microbiome, carcass traits and meat quality in finisher pigs ( n = 40 entire male hybrid, 26.0 ± 0.9 kg) randomly assigned to 1 of 4 dietary treatments: a control diet containing isolated phytase and xylanase valued at 40 kcal of ME/kg (CD0), CD0 + β-mannanase (0.3 g/kg valued at 30 kcal of ME/kg) (CD70), CD0 + β-mannanase (0.3 g/kg valued at 45 kcal of ME/kg) (CD85), and CD0 + β-mannanase (0.3 g/kg valued at 60 kcal of ME/kg) (CD100), with 10 pen replicates. Pigs fed CD0 diet showed ( P = 0.002) greater ADFI. However, pigs fed CD0 diet showed ( P = 0.009) lower G:F than those provided CD70 or CD85 diets. A greater ( P < 0.001) superoxide dismutase concentration was observed in pigs fed CD70 diet. Pigs fed CD85 diet showed ( P = 0.002) greater digestible protein than pigs fed CD0 or CD100 diets. Pigs fed CD70 diet showed an increase of 11.3% in digestible protein than those fed CD0 diet. In addition, greater ( P < 0.001) digestible energy was observed in pigs fed CD85 diet. Pigs fed CD0 or CD100 diets showed greater ( P < 0.05) Firmicutes:Bacteroidota ratio than those fed CD85 diet. The Muribaculaceae was more abundant ( P = 0.030) in pigs fed CD70 diet than in those fed CD0 diet. The Prevotella was more abundant ( P = 0.045) in pigs fed CD85 diet than in those fed CD100 diet. In conclusion, β-mannanase supplementation in diets containing xylanase-phytase allows reducing 85 kcal of ME/kg because it improves gain to feed ratio, energy and protein usage, and backfat thickness without metabolic and intestinal ecosystem disorders in finisher pigs.

Abstract:This study was conducted in two stages to investigate the potential of multi-enzyme supplementation on the nutrient digestibility, growth performance, and gut microbial composition of pigs. In stage 1, effects of multi-enzyme complex (xylanase, α-amylase, β-glucanase, and protease) supplementation on the ileal and total tract dry matter (DM) digestibility of feed-stuffs were investigated with in vitro two-stage and three-stage enzyme incubation methods. A wide range of feed ingredients, namely, corn meal, wheat meal, soybean meal, fish meal, Oriental herbal extract, Italian rye-grass (IRG) and peanut hull were used as substrates. Supplementation of the multi-enzyme complex increased (P < 0.05) the digestibility of the Oriental herbal extract and corn meal. In stage 2, in vivo animal studies were performed to further investigate the effects of the dietary multi-enzyme complex on the nutrient utilization, growth performance, and fecal microbial composition of pigs. A total of 36 weaned pigs were fed corn- and soybean meal-based diets without (control) and with the multi-enzyme complex (treatment) for 6 weeks. Fecal samples were collected from 12 pigs to analyze the microbial communities by using DNA sequencing and bioinformatics tools. Multi-enzyme supplementation had no effect on apparent digestibility of nutrients and growth performance of pigs compared to control. Taxonomic analysis of the fecal samples indicated that the bacteria in both control and treatment samples predominantly belonged to Firmicutes and Bacteroidetes. In addition, the proportion of the phylum Firmicutes was slightly higher in the treatment group. At the genus level, the abundance of Treponema and Barnesiella increased in the treatment group; whereas the numbers ofthe genera including Prevotella, Butyricicoccus, Ruminococcus and Succinivibrio decreased in the treatment group. These results suggest that multi-enzyme supplementation with basal diets have the potential to improve nutrient digestibility and modify microbial communities in the hind-gut of pigs.

Abstract:The experiment aimed to investigate the effects of dietary β-mannanase supplementation on growth performance, apparent total tract digestibility (ATTD) of nutrients, intestinal integrity, and the immunological and oxidative stress parameters in weaning pigs. A total of 64 newly weaning pigs (initial body weight: 6.96 ± 0.70 kg) were allotted to two dietary treatments in eight replicates per treatment with four pigs per pen based on body weight and sex. Dietary treatments were 1.) CON (control: corn-soybean meal based basal diet) and 2.) β-mannanase (basal diet +0.06% β-mannanase). The β-mannanase supplementation did not affect growth performance, concentrations of acute phase protein, superoxide dismutase and glutathione peroxidase. However, the pigs fed the β-mannanase-supplemented diet had greater ATTD of ether extract, jejunum villus height, and villus height-to-crypt depth ratio, and lower crypt depth compared with those fed the CON diet (p < 0.05). The pigs fed the β-mannanase-supplemented diet tended to have the lower count of E. coli in cecum than those fed the CON diet (p = 0.08). In conclusion, dietary β-mannanase supplementation did not affect growth performance, immune response and oxidative stress of weaning pigs, whereas it increased fat digestibility and had positive effects on intestinal integrity and cecum microflora by reducing the count of E.coli.

Abstract:This study aimed to investigate the nutritional and functional roles of β-mannanase on the intestinal health and growth of newly weaned pigs fed a typical or low-cost formulated feeds (LCF). Twenty-four newly weaned pigs at 6.2 kg ± 0.4 body weight (BW) were allotted to three dietary treatments based on a randomized complete block design with sex and initial BW as blocks. Three dietary treatments are as follows: Control, typical nursery feeds including animal protein supplements and enzyme-treated soybean meal; LCF with increased amounts of soybean meal, decreased amounts of animal protein supplements, and no enzyme-treated soybean meal; LCF+, low-cost formulated feed with β-mannanase at 100 g/t, providing 800 thermostable β-mannanase unit (TMU) per kg of feed. Pigs were fed based on a three-phase feeding program for a total of 37 d. On day 37 of feeding, all pigs were euthanized and the gastrointestinal tract was removed for sample collection to analyze intestinal health parameters, mucosa-associated microbiota, and gene expression of tight junction proteins. Pigs fed LCF increased (P < 0.05) the relative abundance of Proteobacteria and Helicobacter in the jejunal mucosa, tended to decrease (P = 0.097; P = 0.098) the concentration of malondialdehyde (MDA) and the expression of zona occluden 1 (ZO-1) gene in the jejunum, tended to decrease average daily gain (ADG; P = 0.084) and final BW (P = 0.090), and decreased (P < 0.05) average daily feed intake. Pigs fed LCF + tended to decrease (P = 0.088) digesta viscosity, decreased (P < 0.05) the relative abundance of Helicobacter, and increased (P < 0.05) Lactobacillus in the jejunal mucosa compared to LCF. Additionally, LCF + tended to increase final BW (P = 0.059) and ADG (P = 0.054), increased (P < 0.05) gain to feed ratio (G:F), and reduced (P < 0.05) fecal score compared to LCF. LCF with decreased amounts of animal protein supplements and increased amounts of soybean meal had negative effects on the composition of the mucosa-associated microbiota, intestinal integrity, and growth performance of nursery pigs. Beta-mannanase supplementation to LCF decreased digesta viscosity, increased the relative abundance of potentially health-benefitting microbiota such as Lactobacillus, and improved growth and fecal score, thus reflecting its efficacy in low-cost formulated feeds with increased amounts of soybean meal.

Abstract:Abstract One-hundred-and-ninety-two weanling pigs (6.7 kg body weight) were used to evaluate the impact of a carbohydrases-protease enzyme complex (CPEC) on growth performance, nutrient digestibility, and gut microbiome. Pigs were assigned to one of the four dietary treatments for 42 d according to a 2 × 2 factorial arrangement of diet type (low fiber [LF] or high fiber [HF]) and CPEC supplementation (0 or 170 mg/kg diet). The LF diet was prepared as corn–wheat-based diet while the HF diet was wheat–barley-based and contained wheat middlings and canola meal. Each dietary treatment consisted of 12 replicate pens (six replicates per gender) and four pigs per replicate pen. Over the 42-d period, there was no interaction between diet type and CPEC supplementation on growth performance indices of pigs. Dietary addition of CPEC improved (P < 0.05) the body weight of pigs at days 28 and 42 and the gain-to-feed ratio of pigs from days 0 to 14. During the entire experimental period, dietary CPEC supplementation improved (P < 0.05) the average daily gain and gain-to-feed ratio of pigs. There were interactions between diet type and CPEC supplementation on apparent total tract digestibility (ATTD) of dry matter (DM; P < 0.01), gross energy (GE; P < 0.01), and neutral detergent fiber (NDF; P < 0.05) at d 42. Dietary CPEC addition improved (P < 0.05) ATTD of DM, GE, and NDF in the HF diets. At day 43, dietary CPEC addition resulted in improved (P < 0.05) apparent ileal digestibility (AID) of NDF and interactions (P < 0.05) between diet type and CPEC supplementation on AID of DM and crude fiber. Alpha diversity indices including phylogenetic diversity and observed amplicon sequence variants of fecal microbiome increased (P < 0.05) by the addition of CPEC to the HF diets on day 42. An interaction (P < 0.05) between diet type and CPEC addition on Bray-Curtis dissimilarity index and Unweighted UniFrac distances was observed on day 42. In conclusion, CPEC improved weanling pig performance and feed efficiency, especially in wheat–barley diets, while dietary fiber composition had a more significant impact on fecal microbial communities than CPEC administration. The results of this study underscores carbohydrase’s potential to boost pig performance without major microbiome changes.

Abstract:Abstract This study aimed to investigate the effects of reductions in energy and amino acids on the efficacy of supplementing corn-wheat-soybean meal diets with a multi-carbohydrase and phytase complex (MCPC) on growth performance and ATTD of energy and nutrients. A total of 700 pigs (BW = 29.5; SD = 4.2 kg) were housed in 70 pens and fed 5 diets (14 pens/diet). Diets were positive control (PC); and negative control 1 (NC1) or negative control 2 (NC2) without or with MCPC. The PC diet was adequate in nutrients according to NRC (2012). Relative to this PC diet, both NC diets were similarly reduced in digestible P (-0.134% unit) and calcium (-0.12% unit) and were differently reduced in NE and dig. AA (NC1 = -173 kcal/kg and -5% dig. AA and NC2 = -124 kcal/kg and -7% dig. AA). Fecal samples were collected from each pen at around 50 kg of BW. Final BW, ADG and feed conversion ratio were impaired (P < 0.05) with the lower nutrient specifications (NC1 and NC2) in comparison with the PC diet. ADFI did not differ among treatments. MCPC supplementation improved the final BW (+2.2%, P = 0.05), ADG (+2.8%, P = 0.05) and feed conversion ratio (-3.3%, P = 0.001). Compared with PC diet, ATTD of DM, OM, CP, EE, energy, Ca and P were reduced in pigs fed NC diets (P < 0.05). The addition of MCPC improved the ATTD of DM (P = 0.001), OM (P = 0.02), CP (P < 0.0001), energy (P = 0.01), Ca (P < 0.0001), and P (P < 0.0001). The present study indicates that the MCPC supplementation was efficient to compensate the dietary protein and amino acid reductions (up to 7%). However, the reduction in dietary energy should be limited below 5% to guarantee feed efficiency and optimal performance.

Abstract:The effects of supplementing a barley-based diet for weaned piglets with exogenous beta-glucanase and xylanase on gastrointestinal digestive enzyme activities were investigated. Thirty-six cross-bred weaned piglets were randomly assigned to two groups with three pens based on sex and mass. Each group was fed on the diet based on barley with or without added beta-glucanase and xylanase (0.15%) for a 4-week period. The results showed that enzyme supplementation improved growth performance of piglets significantly (p < 0.05), but had no effect (p = 0.091) on average daily feed intake. The results also showed that supplementation of beta-glucanase and xylanase had no effect on pepsin activity in gastric contents but slightly decreased (p = 0.092) the pepsin activity in gastric mucosa. Meanwhile, no effect of enzyme supplementation on trypsin activity in duodenal contents was observed. However, the activities of amylase and lipase in duodenal contents were significantly (p < 0.05) decreased, whereas the activities of maltase, sucrase and gamma-glutamyl transpeptidase (gamma-GT) in jejunal and ileal mucosa were enhanced significantly (p < 0.05). The improvement of disaccharidase and gamma-GT activity may be attributed to the positive impacts of exogenous enzymes on digestion and absorption of the nutrients. In conclusion, the current results indicated that supplementation with enzymes in barley-based diets could improve the growth performance of piglets, decrease the activities of amylase and lipase in duodenal contents and increase the activities of disaccharidase and gamma-GT in jejunal and ileal mucosa.
Effects of Supplemental Beta-mannanase on Digestible Energy and Metabolizable Energy Contents of Copra Expellers and Palm Kernel Expellers Fed to Pigs

Abstract:The purpose of this study was to determine the effect of β-mannanase supplementation on digestible energy (DE) and metabolizable energy (ME) contents of copra expellers (CE) and palm kernel expellers (PKE) fed to pigs. Six barrows with an initial body weight of 38.0 kg (standard deviation = 1.5) were randomly allotted to a 6×6 Latin square design with 6 dietary treatments and 6 periods. Six experimental diets were prepared in a 3×2 factorial treatment arrangement with 3 diets of a corn-soybean meal-based diet, a CE 30% diet, and a PKE 30% diet and with 2 concentrations of supplemental β-mannanase at 0 or 2,400 U/kg. All diets had the same proportion of corn:soybean meal ratio at 2.88:1. The marker-to-marker procedure was used for fecal and urine collection with 4-d adaptation and 5-d collection periods. No interactive effects were observed between diet and β-mannanase on energy digestibility and DE and ME contents of experimental diets. However, diets containing CE or PKE had less (p<0.05) DE and ME contents compared with the corn-soybean meal-based diet. The DE and ME contents in CE and PKE were not affected by supplemental β-mannanase. Taken together, we failed to find the effect of β-mannanase supplementation on energy utilization in CE and PKE fed to pigs.

Effects of fibre-degrading enzymes in combination with different fibre sources on ileal and total tract nutrient digestibility and fermentation products in pigs

Abstract:The study was conducted to determine effects of a complex of fibre-degrading enzymes (xylanase, cellulase and β-glucanase) on nutrient digestibility, fibre fermentation and concentrations of short chain fatty acids (SCFA) at different parts of digestive tract in pigs fed different fibre-rich ingredients. A total of 36 barrows fitted with T-cannulas in the distal ileum (initial body weight of 41.1 ± 2.7 kg) were randomly allotted to six dietary treatments with three different high-fibre diets including maize bran (MB), sugar beet pulp (SBP) and soybean hulls (SH) with or without supplementation of fibre-degrading enzymes. Enzyme supplementation improved (p < 0.05) apparent ileal digestibility (AID) of dietary gross energy (GE), crude protein, dry matter (DM), organic matter (OM), total dietary fibre (TDF), neutral detergent fibre (NDF) and apparent total tract digestibility (ATTD) of dietary GE, DM, OM, TDF, insoluble dietary fibre (IDF) when pigs were fed MB, SBP or SH diets. When compared to the SBP and SH diets, the AID of GE, DM, ash, OM and NDF in diet MB was higher (p < 0.05), but the hindgut disappearance and ATTD of nutrients, except for ether extract and crude ash, were lower (p < 0.05). Enzyme supplementation increased acetate and total SCFA concentrations in ileal digesta and faeces of pigs. In conclusion, enzyme addition improved IDF fermentation and SCFA concentration in the whole intestine of pigs, and there was a large variation of digestibility of fibre components among MB, SH and SBP owing to their different fibre composition. Therefore, fibre-degrading enzymes should be applied to fibrous diets to improve efficient production of swine, especially considering low fibre digestibility of fibre-rich ingredients.

Abstract:This study investigated the effects of supplementing feed with various levels of brewer grain meal (BGM) and enzymes (amylase, xylanase, β-glucanase, lipase, cellulase, β-mannanase, phytase, and pectinase) on growth performance, nutrient digestibility, intestinal morphology, immunity, and oxidative status in growing pigs. Eighty growing pigs were subjected to four feed treatments (five replicates per treatment), based on a corn-soybean basal diet: feeds with 0.1% enzyme combination supplementation (PC), no enzyme supplementation (NC), 20% BGM with 0.1% enzyme combination (BGM20), and 40% BGM with 0.1% enzyme combination (BGM40). Supplementing the feed with both BGM-supplemented diets significantly increased final body weight, average daily gain, the digestibility of crude protein and ash, serum concentration of total proteins, superoxide dismutase activity, villus height in the duodenum and jejunum, and duodenal villus height to crypt depth ratio; however, it did not significantly increase blood urea nitrogen, tumor necrosis factor-alpha, malondialdehyde levels, and duodenal crypt depth compared to the NC diet (p 0.05). The addition of up to 40% BGM combined with 0.1% enzyme supplementation positively promotes the growth performance, nutrient utilization, and intestinal health of growing pigs.

Abstract:Experimental manipulation of the intestinal microbiota influences health of the host and is a common application for synbiotics. Here Clostridium butyricum (C. butyricum, C.B) combined with corn bran (C.B + Bran) was taken as the synbiotics application in a waned pig model to investigate its regulation of intestinal health over 28 days postweaning. Growth performance, fecal short chain fatty acids (SCFAs) and bacterial community were evaluated at day 14 and day 28 of the trial. Although the C.B + Bran treatment has no significant effects on growth performance (P > 0.05), it optimized the composition of intestinal bacteria, mainly represented by increased acetate-producing bacteria and decreased pathogens. Microbial fermentation in the intestine showed a shift from low acetate and isovalerate production on day 14 to enhanced acetate production on day 28 in the C.B + Bran treatment. Thus, C.B and corn bran promoted intestinal microbial fermentation and optimized the microbial community for pigs at an early age. These findings provide perspectives on the advantages of synbiotics as a new approach for effective utilization of corn barn.

Abstract:Background: Dietary fiber can be fermented in gut of pigs and the end products of fermentation were short-chain fatty acids (SCFA). The SCFA had positive effects on gut bacteria and host immune system. In addition, SCFA can provide a part of available energy for pigs. However, there were limited reports on the relationship between dietary fiber, gut bacteria, and energy metabolism. Therefore, this study investigated how dietary fiber and enzyme addition impacted energy metabolism by acting on the microbial community and SCFA. Methods: Wheat bran (WB) was added to the corn-soybean meal-based diet at the levels of 12% and 27%, and oat bran (OB) at 15% and 36%. One of each diet was supplemented with or without 5000 U/kg feed of xylanase, so a total of 10 diets were allotted to 60 growing pigs (initial body weight: 27.2 ± 1.2 kg) using a randomized complete block design. The experiment was conducted in 10 consecutive periods using 6 similar open-circuit respiration chambers. Each pig was used for one 20-day period. During each period, six pigs were allowed 14 d to adapt to the diets in metabolic cages followed by 6 d (from d 15 to d 20) in respiration chambers to measure heat production (HP). Results: Pigs fed 36% OB diets had greater (P < 0.05) nutrient digestibility and net energy (NE) values compared to those fed 27% WB diets. Apparent digestibility coefficients of dry matter (DM) and crude protein (CP) were lower (P < 0.05) in pigs fed 27% WB diets compared with those fed 12% WB diets. Enzyme addition improved (P < 0.05) the NE values (11.37 vs. 12.43 MJ/kg DM) in diets with 27% WB. Supplementation of xylanase did not affect NE values for basal diets, OB diets and 12%WB diets. Compared with diets with 36% OB, pigs fed 27% WB-based diets excreted more total SCFA, acetate and propionate (expressed as g/kg feed DM) in fecal samples of pigs (P < 0.05). Pigs in the WB diets had greater proportion of phylum Bacteroidetes while phylum Firmicutes were greater in pigs fed OB diets (P < 0.05). Pigs fed WB diets had greater (P < 0.05) abundance of Succinivibrio and Prevotella, which were associated with fiber degradation and SCFA production. Conclusion: Our results indicated diets supplied by high level of OB or WB promote the growth of fiber-degrading bacteria. The differences in fiber composition between WB and OB led to differences in nutrient digestibility and bacterial communities, which were ultimately reflected in energy metabolism. Enzyme supplementation improved nutrient digestibility as well as NE values for 27% WB diets but not for other diets, which indicated that effects of enzyme were related to type and level of dietary fiber in diets.

Abstract:The aim of this study was to evaluate the effects of mannan oligosaccharides (MOS) on gut health and performance in post-weaning piglets. In total, 40 piglets were divided into two experimental groups and fed a basal diet with (TRT) or without (CON) 0.2% mannan oligosaccharides for 35 days. Growth performance was determined weekly and faecal microbial composition on days 0, 14 and 35. On day 36, histometrical evaluations were performed on duodenal, jejunal, ileal, and colon samples. mRNA gene expression of inflammation-related genes was evaluated in samples of ileal Peyer’s patches (IPP). MOS administration improved feed efficiency in the last two weeks of the trial (p < 0.05), and a decreased clostridia content was found in faeces at day 14 (p = 0.05). TRT piglets showed increased duodenal villi height (p < 0.05), and reduced mRNA levels of Tumour Necrosis Factor α (p < 0.05) and Toll-Like Receptor 4 (p < 0.01) in IPP. Our results suggest beneficial effects of MOS supplementation on gut morphology and the expression of inflammation-related genes in post-weaning piglets, accompanied by increased feed efficiency.

Abstract:This study was designed to determine the effect of dietary supplementation of xylanase on growth performance, nutrient digestibility, organ weight, digesta pH, and concentration of short-chain fatty acids (SCFA) of weaned pigs fed wheat-canola meal (CM) diets over a 35-d period. A total of 144 piglets (72 barrows and 72 gilts) weaned at 18 ± 2 d of age, with initial body weight (BW) of 6.2 ± 0.7 kg, received one of eight dietary treatments based on randomized complete block design. BW and feed intake were recorded weekly to calculate average daily gain (ADG), average daily feed intake (ADFI), and feed efficiency (G:F). Treatments consisted of a control wheat-soybean meal-based diet and wheat-regular (RCM), dehulled (DCM), or coarse CM (CCM) without and with 16,000 BXU/kg xylanase (Econase XT). All diets contained 500 FTU/kg of phytase (Quantum Blue 5G) and titanium dioxide (0.3%). Apparent total tract digestibility (ATTD) of neutral detergent fiber (NDF), crude protein (CP), phosphorous (P), calcium (Ca), dry matter, and diet were determined. On day 35, one pig per pen was euthanized to evaluate the main factors of protein, xylanase supplementation, and sex on organ weight, ileal and colon digesta pH, and colon digesta concentrations of SCFA. The main factors did not affect growth performance. Xylanase supplementation improved nutrient digestibilities in all diets and increased ileal and colonic digesta pH without affecting the growth performance of weaned pigs fed wheat and CM-based diets. A protein-xylanase effect (P < 0.05) resulted in increasing the ATTD of NDF from 28% to 32% and from 29% to 37% for RCM and DCM, respectively. The ATTD of CP was greater (P < 0.05) with xylanase supplementation (75% vs. 70%). Xylanase supplementation increased ATTD of P and Ca. A three-way interaction (P < 0.05) for protein-xylanase-gender for colon pH, acetic, and propionic acid in the colon digesta of pigs indicated that, in addition to the protein source, piglet sex could have influenced how xylanase works. Xylanase supplementation increased (P < 0.05) the weight of the liver and spleen and tended (P < 0.10) to increase the size of the kidney. In conclusion, dietary supplementation of xylanase increased nutrient digestibility and digesta pH but did not influence the growth performance of weaned pigs fed wheat and CM-based diets over a 35-d period.

Abstract:Abstract An experiment was conducted to test the hypothesis that probiotic Bacillus toyonensis M15750 improve the apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of energy and nutrients when included in diets fed by weanling pigs. A control diet was formulated based on corn, soybean-meal, and distillers dried grains with solubles and a second diet was formulated by supplementing the probiotic Bacillus toyonensis M15750 (1 × 109 cfu/kg feed) to the control diet. Titanium dioxide (0.50 %) was added to the diets as an indigestible marker. Sixteen weanling barrows (10.7 ± 0.7 kg) that had a T-cannula in the distal ileum were allotted to the two diets with 8 replicate pigs per diet in a randomized complete block design with body weight being the blocking factor. Pigs were adapted to the diets and to the metabolism crates for 7 days, urine and fecal materials were collected during the following 4 days according to standard procedures using the marker-to-marker approach, whereas ileal digesta were collected for 8 h during the following 2 days. The AID and ATTD of energy and nutrients were calculated for each diet. The AID of dry matter and gross energy was greater (P < 0.05) and the AID of starch tended to be greater (P = 0.05) in the diet supplemented with the probiotic Bacillus toyonensis M15750 compared with the control diet (Table 1). However, no differences were observed for the ATTD of energy or nutrients between the 2 diets, which indicates that the increased digestibility in the small intestine of pigs fed the diet supplemented with Bacillus toyonensis M15750 was offset by a reduction in hindgut fermentation. These results indicate that inclusion of probiotic Bacillus toyonensis M15750 in diets for weanling pigs have the potential to improve the AID of dry matter, gross energy and starch.

Abstract:Aims This study was conducted to investigate the effects of dietary supplementation with a mixture ofBacillus,which serves as an alternative of antibiotics on the intestinal ecosystem of weaned piglets. Methods and Results We randomly assigned 120 piglets to three groups: a control group (a basal diet), a probiotics group (a basal diet supplemented with 4 x 10(9) CFU per gramBacillus licheniformis-Bacillus subtilismixture; BLS mix), and an antibiotics group (a basal diet supplemented with 0 center dot 04 kg t(-1)virginiamycin, 0 center dot 2 kg t(-1)colistin and 3000 mg kg(-1)zinc oxide). All groups had five replicates with eight piglets per replicate. On days 7, 21 and 42 of the trial, intestine tissue and digesta samples were collected to determine intestinal morphology, gut microbiota and bacterial metabolite composition, and the expression of genes related to the gut barrier function and inflammatory status. The results showed that the BLS mix decreased the jejunum crypt depth, while increased the ileum villus height and the jejunum and ileum villus height to crypt depth ratio. The BLS mix increased Simpson's diversity index in the gut microbiota and the relative abundances of o_Bacteroidetesand f_Ruminococcaceae, but decreased the relative abundances ofBlautiaandClostridium. Dietary BLS mix supplementation also modified the concentration of several bacterial metabolites compared to the control group. In addition, BLS mix upregulated the expression level ofE-cadherinin the colon and pro-inflammatory cytokines and TLR-4 in ileum and colon. Lastly, Spearman's rank-order correlation revealed a potential link between alterations in gut microbiota and health parameters of the weaned piglets. Conclusion These findings suggest that dietary BLS mix supplementation modifies the gut ecosystem in weaned piglets. The potential advantages of such modifications in terms of intestinal health are discussed. Significance and Impact of the Study Weaning is the most important transition period of piglet growth and development. This study showed that dietary supplementation of a probiotic mixture ofBacillus, an effective alternative of antibiotics, was beneficial in improving the intestinal ecosystem of weaned piglets.

Abstract:The field of mobile ad hoc networking has enjoyed dramatic increase in popularity over the last few years. However, owing to the fact that such networks have dynamic, sometimes rapidly changing, random, multihop topologies, the development of efficient and applicable multicast routing protocols presents many issues and challenges. In this paper we propose a Mobility-based Hybrid Multicast Routing (MHMR) protocol suitable for mobile ad hoc networks. The main features that our proposed protocol introduces are the following: (i) mobility-based clustering and group-based hierarchical structure, in order to effectively support stability and scalability; (ii) group-based (limited) mesh structure and forwarding tree concepts, in order to support the robustness of the mesh topologies, which provides ‘limited’ redundancy and the efficiency of tree forwarding simultaneously; and (iii) combination of proactive and reactive concepts that provide low route acquisition delay and low overhead. The use of dynamic mobility-based clustering as the underlying structure is motivated by the observation that in mobile ad hoc networks communications are often among teams that tend to coordinate their movements, and as a result we can dynamically and adaptively partition the network into several groups, each with its own mobility characteristics and behaviors. In our protocol we support the creation of a limited mesh structure based on the clusterheads of the various created clusters, and not among all the members that participate in the multicast session and route, thereby reducing significantly the complexity of the created mesh topology. The performance evaluation of the proposed protocol is achieved via modeling and simulation. The corresponding results demonstrate the proposed multicast protocol's efficiency in terms of packet delivery ratio, scalability, control overhead, end-to-end delay as a function of mobility, packet transmission rate, and multicast group size.
