Abstract:Metal-organic frameworks (MOFs), as drug delivery carriers, with high loading capacity and controllable release behavior can provide a more efficacious therapy in cancer treatments. In our work, a novel biocompatible zinc MOF Zn-cpon-1 with the (3,6)-connected rtl 3D topological network was designed and synthesized via employing ClO4- anion as template. The optically and chemically stable Zn-cpon-1 could be verified as a pH-responsive dual-emission platform and excellent drug delivery carrier with higher 5-fluorouracil (5-FU) (44.75 wt %) loading behavior than 6-mercaptopurine (6-MP) (4.79 wt %) ascribed to the influence of size and shape matching. The multiple interactions between Zn-cpon-1 and 5-FU drug molecules have been discussed and evidenced, which could be quantitatively estimated via the rate constant related to the topological structure. Specially, the gust release behavior of 5-FU@Zn-cpon-1 microcrystal was described and programmed via the Weibull distribution model and could be dual-triggered by the temperature and pH stimulus. This study illustrates that the Zn-cpon-1 without any postmodification performs a favorable potential of being used as biomedical drug delivery alternative carriers in effective drug payload, flexible release administration, and superior dual-stimuli responsiveness.
Immunoassay for Cardiac Troponin I with Fluorescent Signal Amplification by Hydrolyzed Coumarin Released from a Metal–Organic Framework

Abstract:Despite its simplicity and specificity, enzyme-linked immunosorbent assay (ELISA) requires conjugation of the enzyme to an antibody and preservation of enzymatic activity during storage and assay. In light of the high porosity and enormous surface area possessed by metal-organic frameworks (MOFs), we constructed an immunosensor by integrating antibody and fluorescent probe (coumarin or COU) with zeolitic imidazolate framework-8 (ZIF-8), in which about 34 COU molecules can be encapsulated per ZIF-8. After coating of ZIF-8 with the secondary antibody specific to cardiac troponin I (cTnI), the final MOF@COU/Ab(2) composite renders an enzyme-free assay that amplifies the detection signal via release of alkaline-hydrolyzed COU from the MOF interior. Owing to the isolation of the fluorescent probe before the readout step, our immunosensors are highly stable and reproducible, with a relative standard deviation of less than 1.5% for repetitive assays in a 30-day period. Our MOF-based immunoassay is also highly sensitive and possesses a wide dynamic range (11.1 fM to 35.6 pM). The detection limit was estimated to be 4.4 fM (0.099 pg.mL(-1)), which is similar to 180-fold lower than that of ELISA. We quantified cTnI in clinical serum samples, yielding results highly comparable to those measured by ELISA. Particularly remarkable is that the low detection limit enabled us to quantify cTnI in sera of healthy persons, including levels that are close to the high-risk threshold but not detectable by ELISA. The generality of this cost-effective and stable fluorescent immunosensor renders versatilities for sensitive detections of many other protein biomarkers.

Abstract:Kiwi‐type nanoarchitecture with good dispersity, fine uniformity, high magnetism and fluorescence are synthesized by spatially‐separated assembly strategy. The assembly structure serves as both a target‐enrichment substrate and an optical signal enhancement label for immunochromatography. The sensing platform shows perspective in determination of cardiac troponin I levels at point‐of‐care scenarios. Fluorescent lateral flow immunoassays (FLFIA) is a well‐established rapid detection technique for quantitative analysis. However, achieving accurate analysis of biomarkers at the pg mL−1 level using FLFIA still poses challenges. Herein, an ultrasensitive FLFIA platform is reported utilizing a kiwi‐type magneto‐fluorescent silica nanohybrid (designated as MFS) that serves as both a target‐enrichment substrate and an optical signal enhancement label. The spatially‐layered architecture comprises a Fe3O4 core, an endocarp‐fibers like dendritic mesoporous silica, seed‐like quantum dots, and a kiwi‐flesh like silica matrix. The MFS demonstrates heightened fluorescence brightness, swift magnetic response, excellent size uniformity, and dispersibility in water. Through liquid‐phase capturing and fluorescence‐enhanced signal amplification, as well as magnetic‐enrichment sample amplification and magnetic‐separation noise reduction, the MFS‐based FLFIA is successfully applied to the detection of cardiac troponin I that achieved a limit of detection at 8.4 pg mL−1, tens of times lower than those of previously published fluorescent and colorimetric lateral flow immunoassays. This work offers insights into the strategic design of magneto‐fluorescent synergetic signal amplification on LFIA platform and underscores their prospects in high‐sensitive rapid and on‐site diagnosis of biomarkers.

In Situ Growing Triethanolamine-Functionalized Metal–Organic Frameworks on Two-Dimensional Carbon Nanosheets for Electrochemiluminescent Immunoassay

Abstract:A facile strategy for in situ growing triethanolamine (TEOA)-functionalized metal–organic framework () on the two-dimensional graphene oxide (GO) or g-C3N4 nanosheets via the self-assembly technique was introduced. In this method, Zn2+ was first attached on the carbon nanosheets by electrostatic interaction; then, trimesic acid (H3btc) acted as the complex agent and TEOA as a base for the deprotonation of H3btc and a template, which leads to in situ growing the MOF on the carbon nanosheets obtaining a sandwich-like structure. Different types of surface analysis techniques were employed to characterize the and g-C3N4 nanomaterials fabricated. The or g-C3N4 nanomaterial-modified electrode brings out obviously enhanced electrochemiluminescence (ECL) behaviors due to numerous TEOA in the framework structures. Specifically, both TEOA and GO can serve as the co-reactants for the ECL system of Ru(bpy)32+ and have the synergic effect of enhancing the signal. Based on the modified electrodes, we developed a sensitive and rapid label-free ECL immunoassay strategy for human copeptin, and the linear range was 5 pg mL–1 to 500 ng mL–1 as well as the limit of detection was 360 fg mL–1. This work exhibits excellent specificity and good stability of the prepared immunosensor in the practical sample determination, demonstrating it can serve as a very promising method for the clinical diagnostics of acute myocardial infarction disease.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) are commonly used as electrochemiluminescence (ECL) luminophores. However, PAHs are limited by the aggregation-caused quenching (ACQ) effect, which reduces ECL emission. Therefore, developing an ECL system from PAHs with efficient luminescence is a significant challenge. In this work, a two-ligand strategy was proposed to assemble 3, 4, 9, 10-perylenetetracarboxylic acid (PTCA) into zeolitic imidazole framework-8 (ZIF -8). The ACQ effect of PTCA was inhibited by participating in the formation of metal-organic frameworks (MOFs) and a luminescent ZIF-8-PTC (Z8P) composite was obtained. Satisfactorily, conceptual experiments demonstrated that the significantly higher ECL efficiency of Z8P was achieved compared with PTCA monomers and aggregates. On the sensing substrate, a novel co-reactant accelerator (Au@MFNC) was used to achieve continuous catalysis of the persulfate (K 2 S 2 O 8 ) with the Prussian blue analogues (PBAs) as a template under the efficient synergistic effect of MoS 2 . Through the reversible transformation of the transition metal valence states, a sufficient amount of sulfate radicals (SO 4 center dot- ), again amplifying the ECL signal of Pd@Z8P. Under ideal conditions, the constructed dual-MOFs ECL immunosensor had a wide response range (1 x 10 -4 -1 x 10 2 U/mL) and a low limit of detection (LOD) (6 x 10 -5 U/mL), which provided an alternative method for CA19-9 ultrasensitive detection, and broadened a new idea for the development of high -performance luminescent MOFs.
Dual amplification electrochemiluminescence of carbonized polymer dots embedded in the metal-organic framework with low trigger potential and nanoconfinement enhancement for sensitive CYFRA21-1 immunoassay

Abstract:Developing novel luminophores for electrochemiluminescence (ECL) that could operate smoothly under low-potential excitation while incorporating creative enhancement strategies is a challenging but very attractive initiative to achieve sensitive detection of disease markers. Herein, carbonized polymer dots (L-CPDs) with low cathode excitation potential were successfully synthesized and used for dual amplified sensitive electrochemiluminescence immunoassay of cytokeratin 19 fragment antigen 21–1 (CYFRA21–1). l -CPDs were first used as ECL luminophores and have an unprecedentedly low excitation potential of -0.95 V that has not been equipped for other cathodic carbon-base dots, avoiding side reactions, bioactive damage and electrode polarization due to high potential, thus obtaining excellent sensor performance. In addition, zeolitic imidazolate framework-8 (ZIF-8) synthesized at room temperature as a nanoconfinement carrier encapsulated l -CPDs and reduced the non-radiative energy loss of the luminophore, which led to substantially improved the ECL signal. The ECL efficiency of the encapsulated luminophores is 2.7 times higher than that of monomeric l -CPDs at the same mass concentration. Platinum composite iron oxide (Fe 3 O 4 /Pt) was used as a co-reaction acceleration platform to strengthen the ECL response. The sandwich ECL immunosensor fabricated achieved sensitive detection of CYFRA21–1 in the detection range of 150 fg/mL–110 ng/mL with a detection limit of 126 fg/mL. In conclusion, this work provides a consideration direction of developing ideal luminophores complemented by amplification strategies in ECL systems construction to achieve accurate and sensitive detection of targets.

Abstract:Polycyclic aromatic hydrocarbons (PAHs) are excellent alternative luminophores for electrochemiluminescence (ECL) immunoassays. However, they are inevitably limited by the aggregation-caused quenching effect. In this study, aimed at eliminating the aggregation quenching of PAHs, luminescent metal-organic frameworks (MOFs) with 1,3,6,8-tetra(4-carboxybenzene)pyrene (H4TBAPy) as the ligand were exploited as a novel nano-emitter for the construction of ECL immunoassays. The luminophore exhibits efficient aggregation-induced emission enhancement, good acid-base resistance property and unusual ECL reactivity. In addition, the simultaneous use of potassium persulfate and hydrogen peroxide as dual co-reactants resulted in a synergistic enhancement of the cathodic ECL efficiency. The use of magnetic iron-nickel alloys as the multifunctional sensing platform can further enhance the ECL activity, and its enriched zero-valent iron as a co-reactant accelerator effectively drives ECL analytical performance. Profiting from the excellent characteristics, signal-on ECL immunoassays have been constructed. With carcinoembryonic antigen as the model analysis target, a detection limit of 0.63 pg/mL was obtained within the linear range of 1 pg/mL to 50 ng/mL, accompanied by excellent analytical performance. This report opens a new window for the rational design of efficient ECL illuminators, and the proposed ECL immunoassays may find promising applications in the detection of disease markers.

Long-lasting chemiluminescence bioassays for glucose enabled by a MOFs-in-hydrogel hybrid platform

Abstract:Chemiluminescence (CL) bioassays with stable long-lasting signal outputs have rekindled interest due to their promising application potential in various fields. In this study, long-lasting chemiluminescence bioassays for glucose were achieved by incorporating metal–organic frameworks (MOFs) in an agarose hydrogel matrix. Zeolitic imidazolate framework-8 (ZIF-8) was used as a matrix to load glucose oxidase (GOx) and zeolitic imidazolate framework-67 (ZIF-67) was used as a nanomimic of peroxidase to promote the CL reactions of luminol. ZIF-8 protects the GOx from denaturation under high temperatures for gel formation and enables its slow release due to its pH-responsive capability. Agarose hydrogel was used as a scaffold to disperse the MOF nanocrystals and inhibit molecular diffusion. Based on the achieved strong and long-lasting chemiluminescence signals, cascade catalytic bioassays with a wide detection range were proposed for the sensitive detection of glucose. The detection range of glucose by CL analysis was 0.2–100 mmol L −1 under optimal conditions, and the detection limit was 0.011 mmol L −1 (S/N = 3).
Protein-driven interaction enhanced electrochemiluminescence biosensor of hydrogen-bonded biohybrid organic frameworks for sensitive immunoassay of disease markers

Abstract:The oriented design of reticular materials as emitters can significantly enhance the sensitivity of electrochemiluminescence (ECL) sensing analysis for disease markers. However, due to the structural fragility of hydrogen bonds, relational research on hydrogen-bonded organic frameworks (HOFs) has not been thoroughly conducted. Additionally, the modulation of luminescence behavior through HOFs has been rarely reported. In view of this, hydrogen-bonded biohybrid organic frameworks (HBOFs) were synthesized and recruited for ECL immunoassay applications. HBOFs was easily prepared using 6,6',6″,6‴-(pyrene-1,3,6,8-tetrayl)tetrakis(2-naphthoic acid) as linkers via bovine serum albumin (BSA) activated hydrogen-bonded cross-linking. The material exhibited good fluorescence emission characteristics. And the highly ordered topological structure and molecular motion limitation mediated by BSA overcome aggregation-caused quenching and generate strong aggregation induced emission, expressing hydrogen-bond interaction enhanced ECL (HIE-ECL) activity with the participation of tri-n-propylamine. Furthermore, a sandwich immunosensor was constructed employing cobalt-based metal-phenolic network (CMPN) coated ferrocene nanoparticles (FNPs) as quenchers (CMPN@FNPs). Signal closure can be achieved by annihilating the excited state through electron transfer from both CMPN and FNPs. Using a universal disease marker, carcinoembryonic antigen, as the analysis model, the signal-off sensor obtained a detection limit of 0.47 pg/mL within the detection range of 1 pg/mL - 50 ng/mL. The synthesis and application of highly stable HBOFs triggered by proteins provide a reference for the development of new reticular ECL signal labels, and electron transfer model provides flexible solutions for more sensitive sensing analysis.
Framework-Induced Electrochemiluminescence Enhancement of an AIEgen-Based MOF Coupled with Heterostructured TiO2@Ag NPs as an Efficient Coreaction Accelerator for Sensitive Biosensing

Abstract:In conventional metal-organic framework (MOF) luminophore-involved electrochemiluminescence (ECL) systems, the aggregation-caused quenching commonly exists for the organic luminescent ligands, limiting the ECL efficiency and detection sensitivity. Herein, by employing the aggregation-induced emission luminogen (AIEgen) 1,1,2,2-tetra(4-carboxylbiphenyl)ethylene (H4TCBPE) as a ligand, one high-efficiency ECL emitter (Zr-MOF) was synthesized through a simple hydrothermal reaction. Compared with H4TCBPE monomers and their aggregates, the resultant Zr-MOF possesses the strongest ECL emission, which is mainly attributed to the framework-induced ECL enhancement. Specifically, the heterostructure was prepared by the deposition of silver nanoparticles on TiO2 microflowers and utilized as an efficient coreaction accelerator. Remarkably, the formative heterojunction can increase the interfacial charge transfer efficiency and promote the carrier separation, facilitating the oxidation of coreactant tripropylamine. In this way, a novel aptamer-mediated ECL sensing platform is constructed, achieving the sensitive analysis of adenosine triphosphate with a low detection limit of 0.17 nM. As a proof-of-concept study, this work may enlighten the rational design of new-type MOF-based ECL materials and expand the application scope of the ECL technology.

Abstract:An aggregation-induced emission (AIE)-based strategy was proposed for fluorescence immunoassays of protein biomarkers using Cu-based metal-organic frameworks (Cu-MOFs) to load recombinant targets and enzymes for dual signal amplification. The immunosensing platform was built based on the sequestration and consumption of the substrates of pyrophosphate (PPi) ions by Cu-MOFs and enzymatic catalysis. The negatively charged PPi could trigger the aggregation of positively charged tetraphenylethene (TPE)-substituted pyridinium salt nanoparticles (TPE-Py NPs) by electrostatic interactions, lighting up the fluorescence due to the AIE phenomenon. The consumption of PPi by the captured Cu-MOFs through the Cu2+-PPi chelation interaction and ALP-enzymatic hydrolysis depressed the aggregation of TPE-Py NPs. Capture of the tested targets in samples by the antibodies on the plate surface could prevent the attachment of target/ALP-loaded Cu-MOFs due to the competitive immunoreactions. The "signal-on" competitive immunoassay was applied for the detection of procalcitonin (PCT) as the model analyte with a linear range of 0.01-10 pg/mL and a detection limit down to 8 pg/mL. The conceptual integration of AIE with enzymatic and MOFs-based dual signal amplification endowed fluorescence immunoassays with high sensitivity and selectivity. The surface modification of Cu-MOFs with hexahistine (His6)-tagged recombinant proteins through metal coordination interactions should be evaluable for the design of novel biosensors.

Abstract:Enzyme-like nanomaterials (nanozyme) has been used to generate amplified response signals in biosensors instead of the bio-enzyme, on account of their higher stability and comparable catalytical activity. Especially the metal-organic framework (MOF) materials with ultra-high porosity and high specific surface area, which is more suitable for deriving superior nanozyme. In order to deal with the increasing demand of healthcare diagnostics and biomedical research, here we propose a novel approach for ultrasensitive photoelectrochemical (PEC) immunoassay. The bio-inspired MOF-derived nanozyme Hemin/BSA@ZIF-8 was introduced into the target biorecognition and sensing processes to accelerate the oxidization of substrate to produce insoluble precipitate, which will impede the electron transfer and lead to a significant reduction in photocurrent. The proposed PEC sensor exhibits a wide linear range of 0.1 pg mL −1 ~ 100 ng mL −1 , and a low detection limit of 11.9 fg mL −1 for cardiac troponin I (cTnI) detection. This work brings a new insight of the bio-inspired MOF nanozyme-mediated PEC immunoassay designing, and offers a novel sensing strategy for accurate human biomarker analysis.

Abstract:Metal-Organic Frameworks (MOFs) can deliver many advantages when acting as enzyme mimics to assist with signal amplification in molecular detection: they have abundant active catalytic sites per unit volume of the material; their structures and elemental compositions are highly tunable, and their high specific surface area and porous property can assist with target separation and enrichment. In the present work, we have demonstrated that, by adding the pore partition agent, 2,4,6-tris(4-pyridyl)pyridine (TPY) during synthesis of the bimetallic Fe/Co-MIL-88(NH2) MOF to block the open metal sites, a highly porous MOF of Fe/Co-TPY-MIL-88(NH2) can be produced. This material also exhibits high stability in basic solutions and biofluids and possesses high peroxidase-mimicking activity, which can be utilized to produce long-lasting chemiluminescence (CL) from luminol and H2O2. Moreover, acting as the peroxidase-mimic, the Fe/Co-TPY-MIL-88(NH2) MOF can form the enzymatic cascade with glucose oxidase (GOx) for biomarker detection. When applied to detect extracellular vesicles (EVs), the MOF material and GOx are brought to the proximity on the EVs through two surface proteins, which triggers the enzyme cascade to produce high CL from glucose and luminol. EVs within the concentration range of 5 × 105 to 4 × 107 particles/mL can be detected with an LOD of 1 × 105 particles/mL, and the method can be used to analyze EV contents in human serum without sample preparation and EV purification. Overall, our work demonstrates that the high versatility and tunability of the MOF structures could bring in significant benefits to biosensing and enable ultrasensitive detection of biomarkers with judicious material designs.

Abstract:A reliable lateral flow immunoassay (LFIA) based on a facile one-step synthesis of single microspheres in combining with immunochromatography technique was developed to establish a new point-of-care test (POCT) for the rapid and early detection of cardiac troponin I (cTnI), a kind of cardiac specific biomarker for acute myocardial infarction (AMI). The double layered microspheres with clear core-shell structures were produced using soap-free emulsion polymerization method with inexpensive compounds (styrene and acrylic acid). The synthetic process was simple, rapid and easy to control due to one-step synthesis without any complicated procedures. The microspheres are nanostructure with high surface area, which have numerous carboxyl groups on the out layer, resulting in high-efficiency coupling between the carrier and antibody via amide bond. Meanwhile, the red fluorescent dye, Nile-red (NR), was wrapped inside the microspheres to improve its stability, as well to reduce the background noise, because of its higher emission wavelength than interference from real plasma samples. The core-shell structures provided different functional areas to separate antibody and dyes, so the immunoassay has highly sensitive, wide working curves in the range of 0⁻40 ng/mL, low limits of detection (LOD) at 0.016 ng/mL, and limits of quantification (LOQ) at 0.087 ng/mL with coefficient of variations (CV) of 10%. This strategy suggested an outstanding platform for LFIA, with good reproducibility and stability to straightforwardly analyze the plasma samples without washing steps, thereby reducing the operating procedures for non-professionals and promoting detection efficiency. The whole detection process can be completed in less than 15 min. This novel immunoassay offers a reliable and favorable analytical result by detecting the real samples, indicating that it holds great potential as a new alternative for biomolecule detection in complex samples, for the early detection of cardiac specific biomarkers.

Abstract:A simple strategy for one-step fabrication of tris(bipyridine)ruthenium(II) (Ru(bpy)32+)-functionalized metal-organic framework (Ru-MOF) thin films using a self-assembly approach assisted by an electrochemical way was introduced. In this protocol, the electrochemically driven cooperative reaction of Ru(bpy)32+ as an electrochemiluminescent (ECL) probe and a structure-directing agent, trimesic acid (H3btc) as a ligand, and Zn(NO3)2 as the Zn2+ source leads to an one-step and simultaneous synthesis and deposition of the MOF onto the electrode surface. Characterization of the Ru-MOF thin films was performed with scanning electron microscopy, Fourier transform infrared, and X-ray photoelectron spectroscopy. Scanning ion conductance microscopy was specially applied in situ to image the topography and thickness of the Ru-MOF thin films. The Ru-MOF thin films as a sensing platform show excellent ECL behavior because of plenty of Ru(bpy)32+ molecules encapsulated in the frameworks. On the basis of the Ru-MOF modified electrodes, an ultrasensitive label-free ECL immunosensing method for the human heart-type fatty-acid-binding protein has been developed with a wide linear response range (150 fg mL-1-150 ng mL-1) and a very low limit of detection (2.6 fg mL-1). The prepared immunosensor also displayed excellent stability and good specificity in the test of practical samples.

Abstract:The precise and real-time monitoring of localized pH changes is of great importance in many engineering and environmental fields, especially for monitoring small pH changes in biological environments and living cells. Metal-organic frameworks (MOFs) with their nanoscale processability show very promising applications in bioimaging and biomonitoring, but the fabrication of nanoscale MOFs is still a challenge. In this study, we synthesized a nanoscale mixed-lanthanide metal-organic framework by a microemulsion method. The morphology and size of the NMOF can be simply adjusted by the addition of different amounts of the CTAB surfactant. This NMOF exhibits significant pH-dependent luminescence emission, which can act as a self-referenced pH sensor based on two emissions of Tb3+ at 545 nm and Eu3+ at 618 nm in the pH range from 3.00 to 7.00. The MTT assay and optical microscopy assay demonstrate the low cytotoxicity and good biocompatibility of the nanosensor.

Abstract:Inorganic lanthanide nanomaterials as photoluminescent biolabels have attracted increasing attention due to their superior physicochemical properties. However, unstable conjugation of inorganic lanthanide nanomaterials with biological function units (such as antibodies) induces instability of conjugated complexes in aqueous solution, limiting their clinical application. In this study, we developed a rapid point-of-care testing (POCT) platform strategy based on coordination-enhanced time-resolved luminescence of specially nanostructural lanthanide particles for lateral flow immunoassay (CE-TRFIA). This strategy integrates a nanoprobe via a dendritic mesoporous silica nanosphere (DMSN) loading a large amount of ultrasmall amorphous europium oxide (Eu2O3) nanoparticles, which rapidly dissolve to release Eu3+ cations under neutral pH value and form luminescent complexes with photosensitizers (such as β-NTA and TOPO) in an LFIA system. This innovative strategy achieves high-sensitivity detection and long-term stability primarily through high-loading probes, excellent dissolution enhancement, stable covalent coupling, and time-resolved detection. With Procalcitonin (PCT) antigen selected as the detection sample, this approach achieves high-sensitivity detection of PCT with a limit of detection (LoD) as low as 1.9 pg/mL, significantly lower than that of commercial LFIA (0.1 ng/mL), and excellent clinical correlation (r = 0.989). The method offers chemiluminescence-level sensitivity without the need for large instruments while retaining the real-time detection characteristics of LFIA. Our results highlight CE-TRFIA as a highly sensitive, specific, and rapid POCT solution for detecting low-abundance biomarkers such as PCT, enhancing the diagnostic capabilities of traditional LFIA and offering significant potential for ultrasensitive and rapid clinical diagnostics.

Abstract:Performance of nanoprobes can often determine the detection level of Lateral immunochromatography. Traditional probes were limited by the quantity and orientation of antibodies, immune activity of the Fab region or binding strength between protein and substrate. This study developed a new efficient and robust technology to construct fluorescent nanoprobes with oriented modified antibodies, based on specific binding of the Fc region of antibody with streptococcal protein G (SPG) on the surface of polystyrene microspheres (MS) and subsequent covalent cross-linking at binding sites to firm them. Lateral flow immunoassay using these probes was applied for the detection of cardiac troponin I (cTnI). The significantly improved detection sensitivity demonstrated that antibody orientation on MS surfaces effectively enhanced immunological activities of probes compared with random immobilizing methods. Furthermore, performance evaluation results of lateral flow test strips met clinical requirements perfectly, including limit of detection (0.032 ng/mL), linearity ( R > 0.99), repeatability (CV < 10%), correlation ( R > 0.99), and heat aging stability. This research also employed heterophilic blocking reagent (HBR) to actively block redundant binding sites of SPG for the first time in order to eliminate false positive interferences, improving the sensitivity and precision of test results further.

Abstract:Lanthanide metal organic frameworks (L-MOFs) are emerging as promising electrochemiluminescence (ECL) emitters for bioanalysis. This work proposed a copper doped terbium MOF as a luminescent tag for construction of a "signal-on" ECL immunosensing method. The Tb-Cu-PA MOF was prepared using Tb3+ and Cu2+ ions as metal linkers and m-phthalic acid as bridge ligand, and exhibited strong ECL emission with K2S2O8 as a coreactant. The immunosensor was prepared by immobilizing capture antibody on Pd nanoparticles modified Ni-Co layered double hydroxide (Pd-ZIF-67@LDH) nanoboxes, which showed strong electrocatalytic activity toward the reduction of S2O82- for amplifying the ECL signal. Upon the sandwich-typed immunoreactions, Tb-Cu-PA MOF labeled antibody was introduced onto the immunosensor for sensitive ECL detection of target protein. Using cytokeratin 19 fragment 21-1 (CYFRA21-1), a representative lung cancer biomarker, as target model, the ECL immunosensing method showed a linear range of 0.01-100 ng/mL and a detection limit of 2.6 pg/mL (S/N = 3). This immunosensing strategy highlighted the advances of using luminescent and electroactive MOFs in the developments of highly efficient immunosensors for bioanalysis.
