Abstract:Changes in the starch digestibility and the properties of the starches isolated from waxy, low-, and high-amylose rice kernels subjected to heat-moisture treatment (HMT) at 100 degrees C for 16 h at a moisture content of 25% were investigated. The results showed that amylose content is important in determining pasting properties for both HMTrice starches and in-kernel HMTrice starches. Amylose leaching, swelling power, and solubility index decreased in hydrothermal treatment of both isolated rice starches and rice kernels. The gelatinization temperatures of both hydrothermal-treated samples were affected by different treatments to different extents, and the changes were more pronounced for starches isolated from the treated rice kernels. The relative crystallinity increased in the starch from treated waxy and low-amylose rice kernels but decreased in the starch from treated high-amylose rice kernels. Starch digestibility study suggested that thermal treatment of rice kernels was more effective in producing low-glycemic-index starch-based foods than the same treatment of isolated starch. The study showed that HMT on rice kernels had significant impacts of physicochemical properties and starch digestibility of waxy, low-, and high-amylose rice starches to various extents.
Physicochemical and Crystalline Properties of Heat-Moisture-Treated Rice Starch: Combined Effects of Moisture and Duration of Heating

Abstract:BACKGROUND Currently there is much interest in the application of physical modification techniques such as heat-moisture treatment (HMT). The effects of HMT on normal and waxy rice starches, subject to different levels of moisture content and duration of heating, were investigated. RESULTS Water solubility index (determined at 90 °C) decreased after HMT for normal and waxy rice starches, while swelling power (determined at 90 °C) showed inconsistent results (decrease for normal type, increase for waxy type) after HMT. Values in pasting parameters of normal and waxy rice starch increased initially, but the extent of increase slowed down with moisture content and length of treatment increasing. HMT decreased gelatinization temperatures with 4 h and 8 h treatment, but when length of treatment was prolonged to 16 h gelatinization temperature increased. Degree of crystallinity decreased for all treatments, and decreased much more at higher levels of moisture content. CONCLUSION Variations in levels of moisture content and duration of heating had significant effects on physicochemical and crystalline properties to different extents.

Abstract:High hydrostatic pressure (HHP) is a novel technology used in the food-processing industry. Starch is an important renewable natural resource. The applications of starch are determined by its properties, which in turn are determined by its structure. In this study, the effects of HHP treatment on starch structure (granular structure, crystalline structure, molecular structure, and molecular conformation) and properties (pasting, retrogradation, thermal, digestive, rheological, swelling, solubility, water absorption, and oil absorption properties) are summarised. Additionally, the mechanism of HHP-induced gelatinisation is discussed. First, the strong hydration ability of starch molecules under high pressure facilitates the binding of water molecules to starch molecules via hydrogen bonding. These bound water molecules may block the channels inside the starch granules, leading to the formation of a sealed space. Finally, the granules disintegrate because of the intra/extra pressure difference. This study provides a reference for the application of HHP to starch processing and modification.

Abstract:Ornithine transcarbamoylase (carbamoyl phosphate:l-ornithine carbamoyltransferase, EC 2.1.3.3) has been partially purified from the blue-green alga Nostoc muscorum Kützing, an organism in which the enzyme seems to be involved in a bicarbonate-fixing pathway leading to citrulline. Pertinent to possible regulation of this pathway, the enzyme shows hyperbolic substrate kinetics, has a molecular weight estimated at 75,000 daltons, and its catalytic capability is little influenced by a selection of metabolites that might conceivably act as regulators in vivo. Thus it seems unlikely that this enzyme is the control point for bicarbonate fixation. In terms of energy of activation (12.3 kcal/mole), size and Km for carbamoylphosphate, the Nostoc enzyme resembled preparations from liver and higher plants more than preparations from Streptococcus and Mycoplasma. The enzymes from Streptococcus and Mycoplasma are probably specialized for citrulline breakdown rather than citrulline synthesis. The Km for ornithine was 2.5 mm at a saturating concentration of carbamoylphosphate and the Km for carbamoylphosphate was 0.7 mm at an ornithine concentration of 2 mm. Ornithine was inhibitory at concentrations greater than 2 mm. Phosphate was a competitive inhibitor with respect to carbamoylphosphate. The pH optimum for citrulline synthesis was 9.5.

Abstract:The rheological, structural properties and in vitro digestibility of starch with high methoxyl pectin (HMP) and further modified by dynamic high-pressure microfluidization (DHPM) were investigated. The viscosity and elasticity increased on addition of HMP and were more pronouncedly affected by 10% HMP. However, after DHPM treatment, the viscosity and elasticity decreased with increasing DHPM pressure. After 100 MPa DHPM treatment, the ordered and crystalline structures were further increased compared with starch-HMP mixtures. A compact and dense surface of starch paste was formed under 100 MPa DHPM and 10% HMP treatment, thus significantly slowing down the digestibility. In contrast, the crystalline and semicrystalline structure of starch were disrupted by intense shear force under 200 MPa DHPM. This study provides theoretical information regarding starch-HMP interaction and improves their functional and physicochemical properties through a promising strategy for better applications in food formulation.
A Study on the Structural and Digestive Properties of Rice Starch–Hydrocolloid Complexes Treated with Heat–Moisture Treatment

Abstract:Rice starch-hydrophilic colloid complexes (SHCs) were prepared by incorporating xanthan gum and locust bean gum into natural rice starch. Subsequently, they underwent hygrothermal treatment (H-SHC) to investigate their structural and digestive properties with varying colloid types and added amounts of H-SHC. The results demonstrated that heat–moisture treatment (HMT) led to an increase in resistant starch (RS) content in rice starch. This effect was more pronounced after the addition of hydrophilic colloid, causing RS content to surge from 8.42 ± 0.39% to 38.36 ± 3.69%. Notably, the addition of locust bean gum had a more significant impact on enhancing RS content, and the RS content increased with the addition of hydrophilic colloids. Enzyme digestion curves indicated that H-SHC displayed a lower equilibrium concentration (C∞), hydrolysis index (HI), and gluconeogenesis index (eGI). Simultaneously, HMT reduced the solubility and swelling power of starch. However, the addition of hydrophilic colloid led to an increase in the solubility and swelling power of the samples. Scanning electron microscopy revealed that hydrophilic colloid encapsulated the starch granules, affording them protection. X-ray diffraction (XRD) showed that HMT resulted in the decreased crystallinity of the starch granules, a trend mitigated by the addition of hydrophilic colloid. Infrared (IR) results demonstrated no formation of new covalent bonds but indicated increased short-range ordering in H-SHC. Rapid viscosity analysis and differential scanning calorimetry indicated that HMT substantially decreased peak viscosity and starch breakdown, while it significantly delayed the onset, peak, and conclusion temperatures. This effect was further amplified by the addition of colloids. Rheological results indicated that H-SHC displayed lower values for G′, G′′, and static rheological parameters compared to natural starch. In summary, this study offers valuable insights into the development of healthy, low-GI functional foods.

Abstract:Thermal treatment has been used to modify starch properties, but few studies pay attention to the change in highland barley starch (HBS) under the whole kernel treatment. The different effect of superheated steam (SS) on the HBS properties from other traditional thermal treatment is still unclear, not mention to the exactly structural change of HBS. This study explored the different effects of SS, steam and roasting treatments of HB kernel on the multi-scale structure, rheological and digestive properties of HBS. Thermal treatments led to the aggregation of HBS granules, the rough surface, and the absence of Maltese cross, as well as the increase in short-chain length ratio and amylose content. Compared with steam and roasting treatments, SS treatment increased the average molecular weight of HBS at 140 °C while reduced it at 180 °C due to starch interactions and depolymerization effect. Multi-scale structure analysis revealed that SS-180 showed the most significant disruption of HBS crystalline structure, the disorder of short-range structure, the decrease in double helix content and the disruption of lamellar structure. These structural changes of HBS resulted in the enhanced thermal stability, and decreased values of G′ and G″ representing the weaker gel structures. Meanwhile, much more resistant starch was converted from rapid digestible starch and slowly digestible starch in SS-treated HB than other thermal-treated HB. In particularly, the most change in multi-scale structure of HBS caused by SS-180 was responsible for its best thermal stability, weakest gel structures and slowest starch digestive rate.

Abstract:High-pressure homogenization (HPH) is a common physical method used for starch modification. In this study, starch molecular structure in terms of chain-length distribution (CLD) and molecular size is characterized to explore the structural variations during HPH and its internal relations. It is found that: 1) the molecular size is significantly reduced by HPH treatments and further gradually decreases with HPH pressure increasing; 2) HPH degrades the long amylose chains with degree of polymerization (DP) ~ 2000–20,000 into short- and intermediate-amylose chains with DP ~ 100–1000 and DP ~ 1000–2000; 3) by HPH treatment, the proportion of amylopectin chains with DP ~ 6–12 and DP ~ 12–24 decreases while that with DP ~ 24–36 and DP ~ 36–100 increases, whereas, the amylopectin CLDs between HPH treated starch samples are not significantly varied; and 4) by a subtraction analysis, the molecular size of HPH treated starches shows a strong correlation with the proportion of degraded long amylose chains, indicating these long amylose chains might play a critical role in maintaining the large molecular size of starch. This study provides a further understanding of molecular features from the individual chains assembling into a whole branched molecule.

Abstract:To investigate the effect of heat-moisture treatment (HMT) on the physiochemical properties and in vitro di-gestibility of corn starch, the pasting behavior, viscoelasticity, thermal properties, long/short range structure, morphology and in vitro digestion of corn starch treated with different HMT conditions (HMT-20, 25, 30, 35 and 40 %) were characterized. Results indicated that after HMT, the pasting and disintegration behaviors of corn starch were affected and correlated with the moisture content. The dynamic viscoelasticity of corn starch was changed, and when glassy conditions were reached, the elastic properties decreased with increasing moisture while the viscous properties increased, especially for the HMT-40 %. The thermal stability of starch was improved by HMT, although the enthalpy of pasting (& UDelta;H) was reduced. Additionally, the HMT processing also promoted the conversion of RDS to SDS and/or RS (SDS and RS increased to 39.80 % and 31.68 % for HMT-40 %, respectively), which might attribute to the rearrangement of free starch molecules. The present work provides a potential approach to make functional starches with healthy properties.

Abstract:This paper evaluated the relationship between multi-scale structure and high-pressure gelation properties of nine types of starches with different amylose (AM) content and crystalline polymorphic structure by RVA 4800. Higher average chain lengths of long amylopectin (AP) chains (DP > 36) and AM content, and lower relative content of short AP chains (DP <= 36) contributed to the higher peak temperature and peak time at 95-140 degrees C, and lower peak viscosity, through viscosity, and final viscosity at 95-110 degrees C. Rheological and texture parameters including storage modulus, loss modulus, hardness, and gumminess, had no significant correlation with starch structural parameters at 95-120 degrees C, but were instead controlled by AM molecular structure at 130-140 degrees C. AM content was mainly responsible for the rheological behaviors of starch gels at 130-140 degrees C, and short and intermediate AM chains were mainly associated with the texture of starch gels at the temperature ranges.

Abstract:The starch suspension was treated by ultra‐high pressure homogenization under various pressures, the starch granules were broken. Subsequent pullulanase debranching produced large amounts of short linear amylose. The final temperature‐cycled crystallization promoted the formation of the double helix structure by hydrogen bonds between starch chains. This study examined the structural characterization and in vitro digestibility of rice starch processed with the combination of ultra‐high pressure homogenization (UHPH), debranching, and temperature‐cycled crystallization under various pressures (0–160 MPa). After UHPH treatment, the degree of branching (DB) of processed starches (0.65%–2.03%) was lower than that of the native rice starch (NS, 2.96%). Assessment of chain length distribution suggested that the considerable amylose was produced after treatments. Processed starches exhibited an irregular fragmented structure, and the crystalline structure was transformed from A‐type (native rice starch) into B + V‐type (processed starches). Moreover, the relative crystallinity (RC) and the absorbance ratio of 1047/1022 cm−1 (R1047/1022) of processed starches decreased from NS (RC, 22.99%; R1047/1022, 0.79) to 13.47%–18.21% (RC) and 0.51–0.78 (R1047/1022), respectively. Iodine binding curves revealed that processed starches possessed the higher degree of polymerization (DP). Furthermore, the contents of slowly digestible starch (SDS) and resistant starch (RS) increased in all processed starches, and the RS content of starch treated under 160 MPa (35.33%) was notably greater than that of native starch (9.37%). These results suggested that the UHPH can be used as a pretreatment strategy to reduce the digestibility of starch products. Practical applications UHPH pretreatment disrupts starch molecular chains to produce the large amounts of short linear amylose. Debranching followed by temperature‐cycled crystallization can increase the content of resistant starch (RS), yielding a raw material suitable for boosting the health of the digestive system and overall health. Therefore, UHPH pretreatment has a high application value in the field of starch modification.

Abstract:The study presents an innovative approach using heat-moisture treatment (HMT) to prepare rice starch-oleic acid (OA)/linoleic acid (LOA) complex. Relationship of the multi-scale structure-in vitro digestibility of rice starch-OA/LOA complex induced by HMT was established in this work. Herein, HMT induced the formation of single helices complex between OA/LOA and amylose or long side chains of amylopectin, which formed more V-type crystallite. HMT assisted complexation with OA/LOA resulted in higher ordered molecular chain aggregations in amorphous amylose background area, reduced the amorphous lamellae and made the double helices and A-type crystallite perfectly arranged. These structural alternations inhibited the molecular interactions (e.g., binding ability, catalyzed location) between rice starch and α-amylase, which eventually increased the resistant starch (RS) content and decreased the slow digestible starch (SDS) content. Moreover, HMT treated rice starch formed type II complex with OA and type I complex with LOA, and all the referred changes went more obvious with the increasing amount of OA than LOA. The results suggested that the OA/LOA was inserted inside the amylose spiral cavity and the amorphous lamellae of rice starch. The current study exhibited an in-depth research of the multi-scale structure within rice starch-OA/LOA complex and proposes a promising approach to ensure the starch products have designated digestibility.
Revisiting the Evolution of Multi-Scale Structures of Starches with Different Crystalline Structures During Enzymatic Digestion

Abstract:Porous starch has been created through hydrolysis by amyloglucosidase and α-amylase. However, little information is known about the precise evolution of multi-scale structures of starch during digestion. In this study, rice starch and potato starch, containing different crystalline structures, were hydrolyzed by amyloglucosidase and α-amylase for 20 and 60 min, respectively, and their resulting structural changes were examined. The digestion process caused significant degradation of the molecular structures of rice and potato starches. In addition, the alterations in the ordered structures varied between the two starches. Rice starch exhibited porous structures, thicker crystalline lamellae as determined by small-angle X-ray scattering, and enhanced thermostability after digestion using differential scanning calorimetry. For rice starch, the extent of crystalline structures was analyzed with an X-ray diffractometer; it was found to first increase after 20 min of digestion and then decrease after 60 min of digestion. In contrast, potato starch did not display porous structures but exhibited thicker crystalline lamellae and a reduction in ordered structures after digestion. These findings suggest that it is possible to intentionally modulate the multi-scale structures of starch by controlling the digestion time, thereby providing valuable insights for the manipulation of starch functionalities.
Hierarchical structure and physicochemical properties of highland barley starch following heat moisture treatment

Abstract:Herein, we concern the multi-scale structure and properties (digestibility and pasting) of highland barley starch following heat moisture treatment (HMT). With the moisture content (MC) rose, HMT reduced the molecular weight (molar mass) but increased the amylose content and the V-type polymorph. When the MC was lower than 25%, a higher MC resulted in increases in the long- (crystallites) and short-range orders (double helices, etc.); nonetheless, a further elevated MC (i.e., 30%) tended to reduce the amounts of these ordered structures. Also, the SAXS results reveal that the lamellar structure could be gradually vanished by the increased MC. These structural changes on multiple scales transformed part of rapidly digestible starch into the slowly digestible and/or resistant forms, accompanied by higher pasting temperature and lower paste viscosity. Hence, highland barley starch following HMT can serve as a food ingredient with reduced digestion rate and paste viscosity.
Effect of Annealing Treatment on the Physicochemical Properties and Enzymatic Hydrolysis of Different Types of Starch.

Abstract:Annealing treatment on the physicochemical properties and hydrolysis of maize, potato and pea starch were investigated in present study. Results indicated that annealing treatment did not change the morphology of the starch. However, the relative crystallinity of maize and potato starch showed a peak trend as the annealing time extended, while pea starch showed a lower relative crystallinity. Besides, all the annealed starch showed a decrease in peak viscosity and an increase in pasting time. Little difference in the rapidly digestive starch (RDS) and resistant starch (RS) contents of annealed maize starch and pea starch were observed after annealing, while annealed potato starch (72 h) showed an increased RS content (23.37 +/- 5.36 %) and a decreased RDS content (52.60 +/- 6.14 %), respectively. The obtained results may provide a better understanding of the physicochemical properties and enzymatic hydrolysis of annealed starch with different semi crystalline type.
Impacts of Hydrothermal Treatments on the Morphology, Structural Characteristics, and In Vitro Digestibility of Water Caltrop Starch

Abstract:The influence of hydrothermal treatments on the structural properties and digestibility of water caltrop starch was investigated. Scanning electron microscopy (SEM) showed some small dents on the surface of starch granules for samples treated with heat moisture treatment (HMT), but not for samples treated with annealing (ANN) which generally showed smoother surfaces. The gelatinization temperature of starch was generally increased by hydrothermal treatments, accompanied by a trend of decreasing breakdown viscosity. These results implied the improvement of thermal and shearing stability, particularly for HMT in comparison to ANN. After being cooked, the native and ANN-modified water caltrop starch granules were essentially burst or destroyed. On the other hand, the margin of starch granules modified by HMT and dual hydrothermal treatments remained clear with some channels inside the starch granules. X-ray diffraction revealed that the crystalline pattern of water caltrop starch changed from the CA-type to the A-type and the relative crystallinity reduced with increasing moisture levels of HMT. Results of ANN-modified water caltrop starch were mostly similar to those of the native one. Moreover, water caltrop starch modified with HMT20 and dual modification contained a pronouncedly higher resistant starch content. These results suggested that HMT, ANN, and dual modification effectively modified the functional properties of water caltrop starch.

Abstract:From a supramolecular structure viewpoint, we concern how concurrent microwave and heat moisture treatment (M/HMT) tailors the pasting and digestion features of indica rice starch (IRS) and waxy rice starch (WRS), using combined analytical methods such as scanning electron microscopy and small-angle X-ray scattering. The results confirm that, for the starches (especially IRS), M/HMT led to a concave granule surface, weakened alignment of helices in lamellae, loosened packing of chains in amorphous regions, and somewhat disrupted crystallites. Consequently, the susceptibility of starch structures to hydrothermal effects was insignificantly altered, as reflected by unchanged pasting temperature; the enzyme absorption/hydrolysis could be properly accelerated, as affirmed by a modestly increased digestion rate and slightly reduced enzymatic resistance. Also, compared to WRS, IRS had more amylose with relatively long branches, which probably contributed to starch chain interactions during M/HMT. This tended to reduce the paste viscosity related to the swelling degree of starch granules, and increase the paste stabilities during heating and cooling related to the robustness of swelled granules and the reorganization of glucan chains.
Effects of Low-pressure Treatment on Physical and Chemical Properties of Chestnut Starch

Abstract:With Dahongpao chestnut starch from Anhui as raw material,the changes of apparent form and crystal feature of the said chestnut were analyzed by a field scanning electron microscope and a light microscopy at different pressures and material-water ratio.The results showed that chestnut starch granules easily swelled and broke along the increase of the pressure under the same time.Some starch granules ruptured when the pressure reached 0.05 MPa.Chestnut starch granules were much easier for rupture with the increases of water content,and some starch granules ruptured when the material-water ratio reached 1∶1 and the pressure was only 0.05 MPa.In the solubility test,after being treated under different pressure and water content,the solubility of chestnut starch increased when the pressure increased;the solubility became higher when the pressure was constant and the water content was higher.And after being treated under different pressures and water contents,the chestnut starch presented the following crystal features:the pressure did not change the crystal type of the chestnut starch;when the pressure increased,the water content in the starch increased and the treatment time was prolonged,the crystal area of the chestnut starch reduced and the crystal content decreased.
