Abstract:Microplastics are ubiquitous in all environments and exert toxic effects in various organisms. However, the neurotoxicity and underlying mechanisms of long-term exposure to MPs aged under UV radiation remain largely unclear. In this study, Caenorhabditis elegans was treated with 0.1–100 μg/L virgin and aged polystyrene microplastics (PS-MPs) for 10 d, with locomotion behavior, neuronal development, neurotransmitter content, and neurotransmission-related to gene expression as endpoints. Using locomotion behavior as an endpoint, chronic exposure to aged PS-MPs at low concentrations (1 μg/L) caused more severe neurotoxicity than that to virgin PS-MPs. In transgenic nematodes, exposure to 10–100 μg/L aged PS-MPs significantly influenced the fluorescence intensity and percentage of worms with neurodegeneration of dopaminergic, glutamatergic, and serotonergic neurons compared with control. Further investigations showed that the content of glutamate, serotonin, and dopamine was significantly influenced in nematodes chronically exposed to 100 μg/L of aged PS-MPs. Similarly, neurotransmission-related gene (e.g., eat-4, dat-1, and tph-1) expression was also altered in nematodes. These results indicate that aged PS-MPs exert neurotoxicity owing to their effects on dopamine, glutamate, and serotonin neurotransmission. This study provides insights into the underlying mechanisms and potential risks of PS-MPs after UV radiation.

Abstract:Both of nanoplastics (NPs) and Tetrabromobisphenol A (TBBPA) are organic pollutants widely detected in the environment and organisms. The large specific surface area of NPs makes them ideal vectors for carrying various toxicants, such as organic pollutants, metals, or other nanomaterials, posing potential threats to human health. This study used Caenorhabditis elegans (C. elegans) to investigate the neurodevelopmental toxicity induced by combined exposure of TBBPA and polystyrene NPs. Our results showed that combined exposure caused synergistic inhibitory effects on the survival rate, body length/width, and locomotor ability. Furthermore, the overproduction of reactive oxygen species (ROS), lipofuscin accumulation, and dopaminergic neuronal loss suggested that oxidative stress was involved in induction of neurodevelopmental toxicity in C. elegans. The expressions of Parkinson's disease related gene (pink-1) and Alzheimer's disease related gene (hop-1) were significantly increased after combined exposure of TBBPA and polystyrene NPs. Knock out of pink-1 and hop-1 genes alleviated the adverse effects such as growth retardation, locomotion deficits, dopaminergic loss, and oxidative stress induction, indicating that pink-1 and hop-1 genes play an important role in neurodevelopmental toxicity induced by TBBPA and polystyrene NPs. In conclusion, TBBPA and polystyrene NPs had synergistic effect on oxidative stress induction and neurodevelopmental toxicity in C. elegans, which was mediated through increased expressions of pink-1 and hop-1.

Abstract:Microplastics (MPs) are nearly ubiquitous in aquatic ecosystems and may affect aquatic organisms. In this study, virgin and aged polystyrene MPs (PS-MPs) of size 1 μm were selected to analyze their adverse effects on larvae zebrafish. Exposure to PS-MPs significantly reduced the average swimming speed of zebrafish, and the behavioral effects caused by aged PS-MPs on zebrafish were more pronounced. Fluorescence microscopy revealed that 10–100 μg/L of PS-MPs accumulated in tissues of zebrafish. As an endpoint of neurotransmitter concentration, exposure to aged PS-MPs at doses ranging from 0.1 to 100 μg/L significantly increased the dopamine (DA), 5-hydroxytryptamine (5-HT), γ-aminobutyric acid (GABA), and acetylcholine (ACh) levels in zebrafish. Similarly, exposure to aged PS-MPs significantly altered the expression of genes related to these neurotransmitters (e.g., dat , 5ht1aa , and gabral genes). According to Pearson correlation analyses, neurotransmissions was significantly correlated with neurotoxic effects of aged PS-MPs. Thus, aged PS-MPs cause neurotoxicity in zebrafish through their effects on DA, 5-HT, GABA, and ACh neurotransmissions. The results highlight the importance of the neurotoxicity of aged PS-MPs in zebrafish, which has important implications for the risk assessment of aged MPs and the conservation of aquatic ecosystems.

Abstract:Microplastics (MPs) are ubiquitous environmental contaminants and cause neurotoxicity in various organisms. However, previous studies that analyzed the effects of MPs mainly focused on virgin polystyrene (V-PS) as representative models of MPs, and the mechanism underlying the neurotoxicity of photoaged polystyrene (P-PS) remains largely unknown. In this study, zebrafish ( Danio rerio ) were exposed to environmentally relevant concentrations (0.1–100 μg/L) of V-PS and P-PS(10 μm). The results indicated that UV radiation accelerated the aging process and changed physical and chemical properties of PS. Whereas exposure to both V-PS and P-PS at low concentrations (100 μg/L) significantly reduced the locomotor behavior of zebrafish larvae, P-PS caused more severe neurotoxicity compared to V-PS. The activity of antioxidant enzymes (SOD, CAT, and GST) and MDA content were significantly altered in zebrafish exposed to 10–100 μg/L of P-PS. Similarly, exposure to P-PS significantly increased neurotransmitter (5-HT, GABA, DA, and ACh) levels and activity of AChE, ChAT, and ChE. Star plots based on integrated biomarker response (IBR) values showed more incline toward neurotransmitter biomarkers in response to increasing P-PS concentration, and the behavioral parameters negatively correlated with the neurotransmitter biomarkers. Further investigations revealed that the expression of neurotransmission- (e.g., ache , drd3 , 5th2c , and gat1 ) and oxidative stress- (e.g., cat1 , sod1 , gpx1a , and gstrl ) related genes was significantly affected by PS in larval zebrafish. Thus, this study provides new insights on the potential risks of MPs into the environment.

Abstract:Microplastics are widely detected in the environment and induce toxic effects in various organisms. However, the properties and toxicity associated with environmentally persistent free radicals (EPFRs) in photoaged nanopolystyrene (NPS) remain largely unknown. We investigated the generation of EPFRs on photoaged NPS and their neurotoxicity and underlying mechanism in Caenorhabditis elegans . The results suggested that photoaging induces the generation of EPFRs and reactive oxygen species (O 2 •− , •OH, and 1 O 2 ), which altered the physicochemical properties (morphology, crystallinity, and functional groups) of NPS. Acute exposure to 1 μg/L of NPS-60 (NPS with light irradiation time of 60 d) significantly decreased locomotion behaviors and neurotransmitter contents (e.g., glutamate, serotonin, dopamine, and γ-aminobutyric acid). Treatment with N- acetyl-L-cysteine (NAC) by radical quenching test significantly reduced EPFRs levels on the aged NPS, and the toxicity of NAC-quenching NPS was decreased in nematodes compared to those in photoaged NPS. EPFRs also caused dysfunction of neurotransmission-related gene expression in C. elegans. Thus, EPFRs generated on photoaged NPS contributed to neurotoxicity by affecting dopamine, glutamate, serotonin, and γ-aminobutyric acid neurotransmission. The study highlights the potential risks of photoaged NPS and the contributions of EPFRs to toxicity.

Abstract:The increased emission and accumulation of micro- or nanoplastics (M-NPs) have posed a severely threaten to organisms in the environment. Though the toxicity of M-NPs has been observed in many species, the fundamental factors determining the biotoxicity are rarely expounded on. In this work, typical polystyrene (PS) M-NPs were set up with a multiparameter variation in size gradient, surface charge contrast and concentration variant, and evaluated by the Caenorhabditis elegans (C. elegans) model. From the endpoints of body length, brood size, survival rate and lifespan, an adverse effect was found on the growth and development of C. elegans caused by PSs. In general, the toxicity of PS was found to be concentrated- and size-dependent, with 100 nm positively charged nano-PS having the highest physio-toxicity. Monitoring by fluorescent imaging, it showed that positively charged nano-PS was mainly ingested and accumulated in the intestinal tract of C. elegans. In addition, the penetrated PS induced severe biological stress reactions with the increase of reactive oxygen species (ROS) and lipofuscin. Furthermore, the following expression of antioxidation-related enzymes was activated in vivo as indicated by the GFP-labelled C. elegans. All the results supplied visually toxic parameters of M-NPs to organisms, which sheds light on the biosecurity and ecological risks of M-NPs in the future.

Comparison of Transgenerational Neurotoxicity between Pristine and Amino-Modified Nanoplastics in C. elegans

Abstract:Increasing evidence has suggested that nanoplastic pollution has become a global concern. More importantly, transgenerational toxicity can be induced by nanoplastics at predicted environmentally relevant doses (ERDs). Considering that amino modification could increase nanoplastic toxicity, we compared transgenerational neurotoxicity between pristine polystyrene nanoparticle (PS-NP) and amino-modified PS-NP (NH2-PS-NP) in Caenorhabditis elegans. At 0.1-10 μg/L, NH2-PS-NP caused more severe transgenerational toxicity on locomotion and neuronal development. Accompanied with a difference in transgenerational neuronal damage, compared to PS-NP (10 μg/L), NH2-PS-NP (10 μg/L) induced more severe transgenerational activation of mec-4, crt-1, itr-1, and tra-3, which are required for the induction of neurodegeneration. Moreover, NH2-PS-NP (10 μg/L) caused more severe transgenerational inhibition in expressions of mpk-1, jnk-1, dbl-1, and daf-7 than PS-NP (10 μg/L), and RNA interference (RNAi) of these genes conferred susceptibility to the toxicity of PS-NP and NH2-PS-NP on locomotion and neuronal development. NH2-PS-NP (10 μg/L) further caused more severe transgenerational activation of germline ligand genes (ins-3, ins-39, daf-28, lin-44, egl-17, efn-3, and lag-2) than PS-NP (10 μg/L), and RNAi of these ligand genes caused resistance to the toxicity of PS-NP and NH2-PS-NP on locomotion and neuronal development. Our results highlighted more severe exposure risk of amino-modified nanoplastics at ERDs in causing transgenerational neurotoxicity in organisms.

Abstract:Caenorhabditis elegans was employed as model to compare reproductive toxicity between pristine and aged polylactic acid microplastics (PLA-MPs). Aged PLA-MPs induced by UV irradiation showed degradation reflected by decrease in size and alteration in morphological surface. Aged PLA-MPs also exhibited some certain changes of chemical properties compared to pristine PLA-MP. Compared with pristine PLA-MPs, more severe toxicity on reproductive capacity and gonad development was detected in 1–100 μg/L aged PLA-MPs. Meanwhile, aged PLA-MPs caused more severe enhancement in germline apoptosis and alterations in expressions of ced-9 , ced-4 , ced-3 , and egl-1 governing cell apoptosis. In addition, aged PLA-MPs resulted in more severe increase in expression of DNA damage related genes ( cep-1 , mrt-2 , hus-1 , and clk-2 ) compared to pristine PLA-MPs, and the alterations in expression of ced-9 , ced-4 , ced-3 , and egl-1 in pristine and aged PLA-MPs could be reversed by RNAi of cep-1 , mrt-2 , hus-1 , and clk-2 . Besides this, enhanced germline apoptosis in pristine and aged PLA-MPs exposed animals was also suppressed by RNAi of cep-1 , mrt-2 , hus-1 , and clk-2 . Therefore, our results suggested the more severe exposure risk of aged PLA-MPs than pristine PLA-MPs in causing reproductive toxicity, which was associated with the changed physicochemical properties and DNA damage induced germline apoptosis.

Abstract:Microplastics (MPs), emerging environmental contaminants, exhibit multiple toxicities in organisms. However, the transgenerational neurotoxicity of MPs has received little attention. Caenorhabditis elegans has been used as a model organism for studying transgenerational toxicity. In this study, the transgenerational neurotoxicity and oxidative stress of MPs were investigated over five generations (F0–F4) of C. elegans. The parental generation (F0) was exposed to polystyrene microplastics (PS-MPs) at concentrations of 0.1–100 μg/L, and subsequent generations (F1–F4) were cultured under toxicant-free conditions. The results indicated that exposure to PS-MPs at concentrations of 10–100 μg/L significantly decreased head thrash and body bends in nematodes, and this reduction was also observed in subsequent generations (F1–F2). This suggested that neurotoxicity induced by PS-MPs can be transferred from the parent to subsequent generations. Maternal exposure to 100 μg/L PS-MPs significantly enhanced ROS production and lipofuscin accumulation in subsequent generations (F1–F2), indicating that the induction of oxidative stress plays an important role in the transgenerational neurotoxicity in C. elegans. Moreover, maternal exposure to PS-MPs resulted in the transgenerational upregulation of genes related to oxidative stress (clk-1, ctl-1, sod-3, sod-4, and sod-5) in the F1–F3 generations, which indicated that these genes may be involved in regulating transgenerational neurotoxicity in C. elegans.

Abstract:Silver nanoparticles (AgNPs) have become widespread in the environment with increasing industrial applications. But the studies about their potential health risks are far from enough, especially in neurotoxic effects. This study aimed to investigate the neurotoxic effects of longer‐term exposure (prolonged exposure for 48 h and chronic exposure for 6 days) of 20nm AgNPs with/without polyvinylpyrrolidone (PVP) coating at low concentrations (0.01–10 mg·L−1) to Caenorhabditis elegans. The results suggested that exposure to AgNPs induced damage to nematode survival, with the longest and relative average life span reduced. Exposure to AgNPs caused neurotoxicity on locomotion behaviors (head thrashes, body bends, pharyngeal pumping frequency, and defecation interval) and sensory perception behaviors (chemotaxis assay and thermotaxis assay), as well as impaired dopaminergic, GABAergic, and cholinergic neurons, except for glutamatergic, based on the alters fluorescence intensity, in a dose‐ and time‐dependent manner. Further investigations suggested that the low‐dose AgNPs (0.01–0.1 mg·L−1) exposure raises receptors of GABAergic and dopamine in C. elegans at the genetic level, whereas opposite results were observed at higher doses (1–10 mg·L−1), which implied that AgNPs could cause neurotoxicity by impairing neurotransmitter delivery. The PVP‐AgNPs could cause a higher fatality rate and neurotoxicity at the same dose. Notably, AgNPs did not cause any deleterious effect on nematodes at the lowest dose of 0.01 mg·L−1. In general, these results suggested that AgNPs possess the neurotoxic potential in C. elegans and provided useful information to understand the neurotoxicity of AgNPs, which would offer an inspiring perspective on the safe application.

Reproductive toxicity of UV-photodegraded polystyrene microplastics induced by DNA damage-dependent cell apoptosis in Caenorhabditis elegans

Abstract:Although many studies have investigated the toxic effects of polystyrene microplastics (PS-MPs), toxicity of natural aged of PS-MPs to soil organisms remains unclear. The photodegradation of virgin PS-MPs under UV irradiation was investigated, and reproductive toxicity of pristine and UV-photodegraded PS-MPs at environmental concentrations (0.1-100 μg/L) was examined to Caenorhabditis elegans. Using brood size and egg ejection rate as endpoints, acute exposure to aged PS-MPs resulted in more severe reproductive toxicity than pristine PS-MPs. Exposure to 100 μg/L aged PS-MPs significantly increased the number of HUS-1::GFP foci and the expression of genes required for DNA damage, such as clk-2, cep-1, and egl-1, suggesting induction of DNA damage. Additionally, the number of cell corpses and apoptosis-related gene expression (e.g., ced-3, ced-4, and ced-9) were significantly altered, indicating induction of apoptosis. Germline apoptosis induced by aged PS-MPs was altered in egl-1, hus-1, cep-1, ced-3, ced-4, and ced-9 mutants. Thus, the reproductive toxicity of aged PS-MPs may be due to DNA damage-induced cell apoptosis, and the HUS-1-CEP-1-EGL-1-CED-9-CED-4-CED-3 signalling pathway is involved in regulating cell apoptosis in nematodes.

Abstract:Studies have shown that exposure to either microplastics (MPs) or di-(2-ethylhexyl) phthalic acid (DEHP) alone can cause neurotoxicity in animals, but it remains uncertain whether and to what extent co-exposure to these two substances, which often occur together in reality, can also induce neurotoxicity. This study aimed to investigate the neurotoxicity and molecular mechanisms of combined exposure to DEHP and polypropylene microplastics (synthetic PP-MPs were used), the microplastics most commonly encountered by young children, in immature mice. The results showed that exposure to PP-MPs and/or DEHP did cause neurotoxic effects in immature mice, including induction of neurocognitive and memory deficits, damage to the CA3 region of the hippocampus, increased oxidative stress, and decreased AChE activity in the brain. The severity of the neurotoxicity increased with increasing concentrations of PP-MPs, combined exposure to PP-MPs and DEHP exhibited additive or synergistic effects. Transcriptomic analyses revealed that the PP-MPs and/or DEHP exposure altered the expression profiles of gene clusters involved in the stress response, and in protein processing in endoplasmic reticulum. Quantitative analyses further indicated that PP-MPs and/or DEHP exposure inhibited the activity of the heat shock response mediated by heat shock transcription factor 1, while chronically activated the unfolded protein response, consequently inducing neurotoxicity through neuronal apoptosis and neuroinflammation in the immature mice. As a pioneer study to highlight the neurotoxicity induced by combined exposure to PP-MPs and DEHP in immature mice, this research provides new insights into mitigating the health risks of PP-MPs and DEHP exposure in young children.

Abstract:Microplastics (MPs) are emerging pollutants widely distributed in the environment, inducing toxic effects in various organisms. However, the neurotoxicity and underlying mechanisms of simulated sunlight-aged MPs have rarely been investigated. In this study, zebrafish (Danio rerio) were exposed to environmentally relevant concentrations (0, 0.1, 1, 10, and 100 μg/L) of virgin polystyrene (V-PS) and aged polystyrene (A-PS) for 120 hpf to evaluate the neurotoxicity. The results demonstrated that simulated sunlight irradiation altered the physicochemical properties (morphology, functional groups, and chemical composition) of V-PS. Exposure to A-PS causes greater toxicity on locomotor ability in larval zebrafish than V-PS. Motor neuron development was disrupted by transgenic (hb9-GFP) zebrafish larvae exposed to A-PS, with significant alterations in neurotransmitter levels (ACh, DA, 5-HT, and GABA) and enzyme activity (AChE, ChAT, and ChE). Further investigation found that exposure to A-PS had a significantly impact on the expression of neurotransmission and neurodevelopment-related genes in zebrafish. These findings suggest that A-PS induces neurotoxicity by its effects on neurotransmission and neurodevelopment. This study highlights the neurotoxic effects and mechanisms of simulated sunlight irradiation of MPs, providing new insights for assessing the ecological risks of photoaged MPs in the environment.

Abstract:Tetrabromobisphenol-A-bis(2,3-dibromopropyl ether) (TBBPA-BDBPE), a novel additive brominated flame retardant, is being developed for use in polyolefin and copolymers. Despite its emerging application, the neurotoxicity and mechanisms of action of TBBPA-BDBPE remain unexplored. Caenorhabditis elegans was utilized as the model organism to study the neurotoxic effects of TBBPA-BDBPE across environmental concentrations ranging from 0 to 100 μg/L. This investigation focused on various toxicological endpoints such as locomotive behavior, neuronal injury, neurotransmitter transmission, and the regulation of nervous system-related gene expression. Acute exposure to TBBPA-BDBPE at concentrations of 10–100 μg/L significantly impaired nematode movement, indicating potential neurotoxicity. In transgenic nematodes, this exposure also caused damage to γ-aminobutyric acid (GABAergic) and serotonergic neurons, along with notable changes in the levels of GABAergic and serotonergic neurotransmitters. Further molecular studies indicated alterations in neurotransmission-related genes ( cat-4 , mod-1 , unc-25 , and unc-47 ). Molecular docking analysis confirmed the binding affinity of TBBPA-BDBPE to key neurotransmission proteins—CAT-4, MOD-1, UNC-25, and UNC-47. These findings demonstrate that TBBPA-BDBPE exerts neurotoxic effects by impacting GABAergic and serotonergic neurotransmission in nematodes. This study provides new insights into the potential environmental risks of TBBPA-BDBPE.

Abstract:Tetrachlorobisphenol A (TCBPA) has been used as an alternative flame retardant in various fields. However, the long-term effects of TCBPA on the nervous system remain unclear. Thus, Caenorhabditis elegans (L4 larvae) were selected as a model animal to investigate the neurotoxic effects and underlying mechanisms after 10 d of TCBPA exposure. Exposure to TCBPA (0.01–100 μg/L) decreased locomotive behavior in a concentration-dependent manner. In addition, reactive oxygen species (ROS) formation and lipofuscin accumulation were significantly increased, and the expression of sod-3 was upregulated in the exposed nematodes, indicating that TCBPA exposure induced oxidative damage. Furthermore, 100 μg/L TCBPA exposure caused a reduction in dopamine and serotonin levels, and damage in dopaminergic and serotoninergic neurons, which was further confirmed by the downregulated expression of related genes (e.g., dop-1 , dop-3 , cat-1 , and mod-1 ). Molecular docking analysis demonstrated the potential of TCBPA to bind to the neurotransmitter receptor proteins DOP-1, DOP-3, and MOD-1. These results indicate that chronic exposure to TCBPA induces neurotoxic effects on locomotive behavior, which is associated with oxidative stress and damage to dopaminergic and serotoninergic neurons.

Abstract:The increase of plastic production together with the incipient reuse/recycling system has resulted in massive discards into the environment. This has facilitated the formation of micro- and nanoplastics (MNPs) which poses major risk for environmental health. Although some studies have investigated the effects of pristine MNPs on reproductive health, the effects of weathered MNPs have been poorly investigated. Here we show in Caenorhabditis elegans that exposure to photoaged polystyrene nanoplastics (PSNP-UV) results in worse reproductive performance than pristine PSNP (i.e., embryonic/larval lethality plus a decrease in the brood size, accompanied by a high number of unfertilized eggs), besides it affects size and locomotion behavior. Those effects were potentially generated by reactive products formed during UV-irradiation, since we found higher levels of reactive oxygen species and increased expression of GST-4 in worms exposed to PSNP-UV. Those results are supported by physical-chemical characterization analyses which indicate significant formation of oxidative degradation products from PSNP under UV-C irradiation. Our study also demonstrates that PSNP accumulate predominantly in the gastrointestinal tract of C. elegans (with no accumulation in the gonads), being completely eliminated at 96 h post-exposure. We complemented the toxicological analysis of PSNP/PSNP-UV by showing that the activation of the stress response via DAF-16 is dependent of the nanoplastics accumulation. Our data suggest that exposure to the wild PSNP, i.e., polystyrene nanoplastics more similar to those actually found in the environment, results in more important reprotoxic effects. This is associated with the presence of degradation products formed during UV-C irradiation and their interaction with biological targets.

Abstract:Nanoplastics are emerging contaminants that may co-exist with organochlorine pesticides and adversely affect invertebrates in the environment. However, the impact of environmental aging on the combined toxicity of nanoplastics and organochlorine pesticides remains unclear. This study investigated the effects of aging on the combined toxicity of polystyrene nanoplastics (PS NPs) and chlordane against Caenorhabditis elegans. The results showed that photo-aging altered the physicochemical properties of PS NPs and promoted the combined toxicity of PS NPs and chlordane to nematodes by reducing survival rate, body length and enhancing germline apoptosis. Additionally, combined exposure of nematodes to aged PS NPs and chlordane significantly increased reactive oxygen species production and intestinal permeability, suggesting that aging enhances combined toxicity through oxidative stress and intestinal damage. Moreover, aging increased chlordane contents in nematodes without promoting PS NPs accumulation, potentially leading to increased combined toxicity of PS NPs and chlordane. Notably, aging significantly increased the accumulation of PS NPs in the posterior intestine of the nematode during co-exposure, which may be responsible for the most sensitive and highest degree of change in germline apoptosis. These observations emphasize the significance of accounting for environmental aging as well as the accumulation and distribution of nanoplastics in organisms when assessing the combined effects of nanoplastics and coexisting pollutants.
Toxic effect of chronic exposure to polyethylene nano/microplastics on oxidative stress, neurotoxicity and gut microbiota of adult zebrafish (Danio rerio)

Abstract:The rapid development of aquaculture industry has provided a large amount of high-quality animal protein, while the food safety caused by microplastics and nanoplastics (MP/NPs) has become a major concern. In addition, recent evidence has shown the potential toxic effect of PE-MP/NPs, highlighting the need for further research into their environmental and health impacts. Chronic exposure of polyethylene microplastics (PE-MPs) and nanoplastics (PE-NPs) on adult zebrafish were conducted in the present study for 21 d. Organ-dependent oxidative damage induced by MP/NPs was observed. Insignificant differences in neurotoxicity and dysbiosis of gut microbiota were found between MPs and NPs. Changes in glutathione S-transferase (GST), glutathione (GSH), catalase (CAT), lipid peroxidation (LPO), and superoxide dismutase (SOD) showed that MP/NPs induced oxidative damage in gill and intestinal cells of zebrafish. The inhibited AChE activity suggested the potential neurotoxicity of microplastics and nanoplastics (MP/NPs). In addition, chronic exposure increased the alpha-diversity of intestinal microbiota. At the phylum level, the average relative abundance of Proteobacteria increased from 29.73% (control group) to 66.10% (microplastics), 54.84% (nanoplastics) and 60.03% (combined exposure), respectively. Tenericutes decreased from 55.43% (control group) to 20.02% (microplastics), 22.44% (nanoplastics) and 31.77% (combined exposure), respectively. Overall, this study provides new insights and objective evidence for the toxicity assessment of PE-MPs.

Abstract:Polystyrene nanoplastics (PS-NPs) are emerging environmental contaminants that are ubiquitously detected in various environments and have toxic effects on various organisms. Nevertheless, the transgenerational reproductive toxicity and underlying mechanisms of PS-NPs remain largely unknown, especially for photoaged PS-NPs under ultraviolet irradiation. In this study, only the parental generation (P0) was exposed to virgin and aged PS-NPs at environmentally relevant concentrations (0.1-100 μg/L), and subsequent generations (F1-F4) were cultured under normal conditions. Ultraviolet irradiation induced the generation of environmentally persistent free radicals and reactive oxygen species, which altered the physical and chemical characteristics of PS-NPs. The results of toxicity testing suggested that exposure to aged PS-NPs caused a more severe decrease in brood size, egg ejection rate, number of fertilized eggs, and hatchability than did the virgin PS-NPs in the P0, F1, and F2 generations. Additionally, a single maternal exposure to aged PS-NPs resulted in transgenerational effects on fertility in the F1 and F2 generations. Increased levels of H3K4 and H3K9 methylation were observed in the F1 and F2 generations, which were concomitant with the transgenerational downregulation of the expression of associated genes, such as spr-5, set-17, and met-2. On the basis of correlation analyses, the levels of histone methylation and the expression of these genes were significantly correlated to transgenerational reproductive effects. Further research showed that transgenerational effects on fertility were not observed in spr-5(by134), met-2(n4256), and set-17(n5017) mutants. Overall, maternal exposure to aged PS-NPs induced transgenerational reproductive effects via H3K4 and H3K9 methylation, and the spr-5, met-2, and set-17 genes were involved in the regulation of transgenerational toxicity. This study provides new insights into the potential risks of photoaging PS-NPs in the environment.
