Abstract:Abstract Aflatoxins are naturally occurring toxic substances produced mainly by species of the genus Aspergillus that can contaminate almost all foodstuffs. Apart from the harmful effects they have on human and animal health, they can be secreted unchanged in animal milk and cause contamination of milk and its products. Aflatoxin M 1 (AFM 1 ) is the major and most toxic type of aflatoxin after aflatoxin B 1 (AFB 1 ). The use of probiotic strains to reduce the amount of aflatoxin in milk and by-products has been observed in many studies. In this systematic review and meta-analysis, articles in PubMed, Scopus, Google Scholar and ISI Web of Science were searched to find eligible studies that reported reduction of AFM 1 using probiotics in dairy products. The results were pooled using a random-effects model. In most studies, the efficiency of probiotics in milk has been tested by Lactobacillus strains. The results indicated that probiotic microorganisms could significantly reduce AFM 1 by 55.76% (confidence interval (CI): 54.35%, 57.16%; I 2 = 100%). Besides, the rank order of AFM 1 reduction in dairy products based on probiotic strain subgroup was: Lactobacillus 51.99% (CI: 50.14%, 53.85%, I 2 = 100%), Saccharomyces 67.36% (CI: 65.05%, 69.67%, I 2 = 100%), Bifidobacterium 54.80% (CI: 54.18%, 55.43%, I 2 = 99.9%), and 61.90% (CI: 53.80%, 70.00%, I 2 = 100%) by a mix of strains. Considering the high binding potential of AFM 1 to probiotic strains, these microorganisms can be recommended as a safe system to reduce AFM 1 in dairy products.

Abstract:Milk is considered a perfect natural food for humans and animals. However, aflatoxin B1 (AFB1) contaminating the feeds fed to lactating dairy cows can introduce aflatoxin M1 (AFM1), the main toxic metabolite of aflatoxins into the milk, consequently posing a risk to human health. As a result of AFM1 monitoring in raw milk worldwide, it is evident that high AFM1 concentrations exist in raw milk in many countries. Thus, the incidence of AFM1 in milk from dairy cows should not be underestimated. To further optimize the intervention strategies, it is necessary to better understand the metabolism of AFB1 and its biotransformation into AFM1 and the specific secretion pathways in lactating dairy cows. The metabolism of AFB1 and its biotransformation into AFM1 in lactating dairy cows are drawn in this review. Furthermore, recent data provide evidence that in the mammary tissue of lactating dairy cows, aflatoxins significantly increase the activity of a protein, ATP-binding cassette super-family G member 2 (ABCG2), an efflux transporter known to facilitate the excretion of various xenobiotics and veterinary drugs into milk. Further research should focus on identifying and understanding the factors that affect the expression of ABCG2 in the mammary gland of cows.
84 Immuno-Safety of Recombinant Human C1 Inhibitor in Patients With Hereditary Angioedema: An Integrated Analysis

Abstract:Background Recombinant C1 inhibitor (rhC1INH) is a novel therapeutic option for the treatment of acute angioedema attacks in patients with hereditary angioedema (HAE). The amino acid sequence of rhC1INH is identical to that of endogenous C1INH. However, any recombinant protein may elicit antibodies against the protein and/or host related impurities (HRI). Clinical consequences of these antibodies can theoretically range from no clinical symptoms to allergic reactions and reduced C1INH activity due to neutralizing antibodies. Objective To analyze the immuno-safety of rhC1INH in symptomatic patients with HAE. Methods Plasma samples were collected pre-treatment and 22 and 90 days post-treatment of an acute angioedema attack. Plasma samples were tested for the presence of antibodies against plasma-derived C1INH and rhC1INH using 6 different, validated enzyme-linked immunosorbent assays (ELISAs), to detect IgM, IgG and IgA antibodies against plasma-derived C1INH or rhC1INH. Antibodies against HRI in plasma samples were measured in an ELISA testing for all antibody classes. Plasma samples from normal healthy controls and HAE patients, never exposed to rhC1INH, were used to estimate cut off levels of the assays. Plasma samples with antibody levels above the cut-off level in the screening assays were tested in confirmatory displacement assay in case of anti-HRI antibodies and in an assay for neutralizing antibodies in case of antibodies against C1INH. Results Data from 155 symptomatic HAE patients having received a total of 424 administrations of rhC1INH were analyzed. The frequency of anti-C1INH antibody levels above the assay cut-off was low and similar in pre- and post-exposure samples (1.7 and 1.8%, respectively). Results above the assay cut-off were sporadic and transient. Occurrence of anti-C1INH antibodies did not correlate with repeated treatment or time since last treatment. No neutralizing antibodies were detected. A total of 5/155 (3%) rhC1INH-treated patients had confirmed anti-HRI antibodies; these included 1 patient with presence of anti-HRI antibodies prior to exposure to rhC1INH. The presence of anti-C1INH and anti-HRI antibodies was not associated with clinical symptoms. The presence of anti-C1INH antibodies did not affect clinical efficacy. Conclusions rhC1INH used for the treatment of acute HAE attacks has a low potential to induce antibodies and has a reassuring immuno-safety profile.

Abstract:The aflatoxin producing fungi, <i>Aspergillus spp.</i>, are widely spread in nature and have severely contaminated food supplies of humans and animals, resulting in health hazards and even death. Therefore, there is great demand for aflatoxins research to develop suitable methods for their quantification, precise detection and control to ensure the safety of consumers' health. Here, the chemistry and biosynthesis process of the mycotoxins is discussed in brief along with their occurrence, and the health hazards to humans and livestock. This review focuses on resources, production, detection and control measures of aflatoxins to ensure food and feed safety. The review is informative for health-conscious consumers and research experts in the fields. Furthermore, providing knowledge on aflatoxins toxicity will help in ensure food safety and meet the future demands of the increasing population by decreasing the incidence of outbreaks due to aflatoxins.

Abstract:Aflatoxin M1 (AFM1) is the main secondary metabolites of aflatoxin B1 (AFB1) framed in the liver and discharged into milk when people and animals polish off AFB1-polluted food. The persist from feed to milk in dairy cows is affected by different dietary and physiological elements, including taking care of regimens, rate of ingestion, rate of absorption, animal health, hepatic biotransformation limit, and real milk production of the animals. Abstract:Aflatoxins (AF) are a group of toxic secondary metabolites mostly produced by strains of Aspergillus spp. fungi. There are several types of aflatoxins (AFB1, AFG1, AFB2, AFG2, AFM1, AFM2, etc.), but the most toxic and the most common is AFB1. Aflatoxins have been shown to possess powerful carcinogenic, mutagenic and teratogenic properties. AFB1 can contaminate feed and food during the growth and/or storage under inappropriate conditions. When animals consume AFB1-contaminated feed, it undergoes various metabolic pathways and finally, it is metabolized into hydroxylated metabolite AFM1, which is possibly excreted in milk if the animal is lactating. This issue presents a huge concern regarding the representation of milk and dairy products in the human diet. Furthermore, climate changes have a significant impact on aflatoxin production. Therefore, it is necessary to develop and improve strategies for controlling and mitigating the occurrence of AFM1 in milk and dairy products. The aim of this paper is to provide an overview of the latest scientific literature regarding the occurrence of AFM1 in milk and dairy products.
Metabolites and degradation pathways of microbial detoxification of aflatoxins: a review

Abstract:The degradation of aflatoxins using nonpathogenic microbes and their enzymes is emerging as a safe and economical alternative to chemical and physical methods for the detoxification of aflatoxins in food and feeds. Many bacteria and fungi have been identified as aflatoxin degraders. This review is focused on the chemical identification of microbial degradation products and their degradation pathways. The microbial degradations of aflatoxins are initiated by oxidation, hydroxylation, reduction, or elimination reactions mostly catalyzed by various enzymes belonging to the classes of laccase, reductases, and peroxidases. The resulting products with lesser chemical stability further undergo various reactions to form low molecular weight products. Studies on the chemical and biological nature of degraded products of aflatoxins are necessary to ensure the safety of the decontamination process. This review indicated the need for an integrated approach including decontamination studies using culture media and food matrices, proper identification and toxicity profiling of degraded products of aflatoxins, and interactions of microbes and the degradation products with food matrices for developing practical and effective microbial detoxification process.
Reduced Graphene Oxide-Zinc Iron Oxide Nanomaterial As Selective Dispersive Solid-Phase Extraction Sorbent for Extraction and Enrichment of Aflatoxins from Milk Combined with UHPLC-MS/MS Analysis

Abstract:Fungal species capable of producing aflatoxins frequently contaminate the feed provided to dairy animals, leading to the subsequent occurrence of aflatoxin contamination in milk. Milk is renowned for its exceptional nutritional composition, and the consumption of milk contaminated with aflatoxins by humans can engender a multitude of health complications. In this study, reduced graphene oxide zinc iron oxide (rGO-Zn/Fe2O4) nanomaterial was utilized as a dispersive solid phase extraction adsorbent to separate and enrich aflatoxins (aflatoxin B1, aflatoxin B2, aflatoxin G1, aflatoxin G2, and aflatoxin M1) from milk. The rGO-Zn/Fe2O4 nanomaterial was morphologically characterized by scanning electron microscopy, transmission electron microscopy, and X-ray diffraction, indicating that Zn/Fe2O4 nanoparticles were embedded on the surface of rGO. The acetonitrile/formic acid (99/1, v/v) combined with ultrasonic treatment for a duration of 40 min expressed an efficient extraction solvent. Employing the matrix-spiked technique, different parameters of the dispersive solidphase extraction (d-SPE) method were meticulously optimized, yielding the following conditions: 2% acetonitrile as loading solvent, 15 mg of rGO-Zn/Fe2O4 as the adsorbent quantity, 15 min of ultrasonication as adsorption time, and n-hexane as the washing solvent. The desorption reagent was 5 mL acetonitrile, with a desorption time of 5 min. The combination of the dispersive solid-phase extraction (d-SPE) technique with validated UHPLC-MS/ MS quantitative analysis demonstrated excellent selectivity, a robust linear relationship (R-2 >= 0.994), remarkable sensitivity (limit of quantification ranging from 0.006 to 0.013 mu g/L), and satisfactory recovery rates of 78%-104% at three different matrix-spiked levels. Moreover, the method exhibited precision with relative standard deviations (RSD) ranging from 2.4% to 10.8%. Following appropriate pretreatment procedures, the milk samples underwent rigorous aflatoxin analysis, leading to the identification of AFM1 presence in all of the examined milk samples (n = 9). However, the contamination levels detected were found to be within the permissible limits as defined by regulatory standards.

Abstract:Aflatoxin M1 (AFM1) is a mycotoxin that often contaminates milk. Like other mycotoxins, it is thermostable and potentially carcinogenic. The present work was carried out to evaluate the ability of microorganisms isolated from Indonesian kefir grains to reduce AFM1 in contaminated phosphate buffer saline (PBS). Fourteen isolates of lactic acid bacteria, both aerobic (LAE) and anaerobic (LAN), and nine isolates of yeast (YEA) were used. The significantly highest AFM1 reduction percentage was shown by the isolate LAE7 (29.3 ± 0.6%) after 4 h incubation. DNA sequencing of LAE7 and YEA2 isolates showed that these isolates had homology (level of similarity) with species of Lactobacillus kefiri strain A/K and Saccharomyces cerevisiae NRRL Y-12632, respectively. The present work proved that isolates from Indonesian kefir grains could reduce AFM1 and have the potential for practical use.

Abstract:An infant who suffered acute subdural hematoma due to minor head trauma twice in a short period is presented. Each subdural hematoma, showing high density on computed tomographic scanning, resolved with unusual rapidity, resulting in full recovery after nonsurgical management. The mechanism of this rapid resolution of each hematoma was thought to be participation of cerebrospinal fluid secondary to a tearing of the arachnoid membrane.

Probiotics and Antimicrobial Proteins

Abstract:Aflatoxins (AFs) are secondary metabolites produced by fungi, and they are deemed the most perilous mycotoxin and food safety predicament. The exposure of humans to mycotoxins transpires either directly through the consumption of contaminated agricultural commodities or indirectly through the ingestion of items derived from animals that have been nourished with tainted substances of animal origin. To ensure the detoxification of AFs in animal and plant food products and to mitigate the risks they pose to public health and the economy, diverse techniques (physical, chemical, and biological) have been subject to scrutiny. By altering and eradicating the molecular structure of the toxin, all of these approaches impede its transmission to the digestive system and potentially diminish the accessibility of toxins to the target tissue, ultimately eliminating them. Given the pervasive predicaments attributed to the contamination of foods and feeds by AFs, it is of utmost importance to urgently devise cost-effective and appropriate strategies to combat this hazard. This review highlights the concept of AFs, definitions, and benefits of postbiotics and their biological role in the detoxification of AFs, as well as their benefits in the food-pharmaceutical industry.

Biodegradation Characteristics and Mechanism of Aflatoxin B1 by Bacillus amyloliquefaciens from Enzymatic and Multiomics Perspectives

Abstract:Aflatoxin B1 (AFB1), a mycotoxin and natural carcinogen, commonly contaminates cereals and animal feeds, posing serious health risks to human and animal. In this study, Bacillus amyloliquefaciens ZG08 isolated from kimchi could effectively remove 80.93% of AFB1 within 72 h at 37 °C and pH 7.0. Metabolome and transcriptome analysis showed that metabolic processes including glycerophospholipid metabolism and amino acid metabolism were most affected in B. amyloliquefaciens ZG08 exposed to AFB1. The adaptation mechanism likely involved activation of the thioredoxin system to restore intracellular redox equilibrium. The key genes, tpx and gldA, overexpressed in Escherichia coli BL21, achieved degradation rates of 60.15% and 47.16% for 100 μg/kg AFB1 under optimal conditions of 37 °C and pH 8.0 and 45 °C and pH 7.0, respectively. The degradation products, identified as AFD1, were less cytotoxic than AFB1 in HepG2 cells. These findings suggest potential strategies for utilizing probiotics and engineered enzymes in AFB1 detoxification.
Current major degradation methods for aflatoxins: A review

Abstract:Background Aflatoxins are strong cancerogenic compounds predominantly produced by certain strains of the Aspergillus genus. Due to their extreme stability in different conditions, it is very difficult to remove them completely in human diet and animal feeds. In this way aflatoxins are triggering numerous healthy problems (such as liver cancer) and thus becoming a huge burden to the hygiene system and food industry worldwide. Scope and approach Therefore, seeking for an effective technique to degrade aflatoxins to a threshold level has been a “hot-topic” among researchers. Traditional methods to detoxify aflatoxins include physical and chemical treatments, such as an extrusion cooking process and ammoniation, respectively. Meanwhile a bio-degradation by microorganisms gains its popularity due to its friendliness to both environment and body health. Natural phytochemicals (plant extracts) which have great capability to remove aflatoxins without causing any damage on human and animals come out as an improvement. Key findings and conclusion However, a fully and systematically discussion of the methods of detoxification for aflatoxins is still not available. Therefore, in the present review we briefly enumerate several traditional strategies, update newly methods, particularly the potential use of natural phytochemicals, and discuss some mechanisms during the detoxification period, summarizing merits and demerits of these methods. We suggest that this important information and our humble opinions could help researchers to understand the degradation of methods for aflatoxins.

Potential of Animal Excreta as a Source of Probiotic Lactic Acid Bacteria for Aflatoxin B1 Detoxification by the Surface Binding Mechanism

Abstract:Aflatoxins (AFs) are the most potent and ubiquitously found mycotoxins, capable of causing contamination in agricultural products. Aflatoxin B1 (AFB1) is the most toxic and primarily produced Aflatoxin and will be a real threat to the safety of food and feeds. The current study searched for the potential of Lactic acid bacteria (LAB) isolated from animal excreta for AFB1 mitigation. Three LAB out of 56 isolates were found to exhibit more than 50% sorbent action with AFB1 in phosphate-buffered saline (PBS) and were identified as Lactococcuslactis strain CF_6 (OP183481) (65.38%), Lacticaseibacillus casei strain CW_3 (OP183482)(52.63%) and Lactobacillus acidophilus strain CE_4 (OP183483)(63.13%). More than 60% of the total AFB1 removal was observed in 2 hr of incubation, and maximum sorbent action was found at a pH 6-7 range at 37oC for 24 hours. In the Scanning Electron Microscope (SEM) analysis, heat-killed cells showed a significant increase in cell surface binding area, which improved the surface binding for all isolates except L. casei strain CW_3; however, it proves that LAB surface binding is strain-specific rather than heat treatment. Moreover, the rise in AFB1 concentration improved the rate of the sorbent action but did not observe any substantial changes in total AFB1 detoxification. So, it is concluded that the animal excreta may be a versatile source of probiotic LAB for AFB1 detoxification by surface binding.

Abstract:Aflatoxins are toxic secondary metabolites that often contaminate food and animal feed, causing huge economic losses and serious health hazards. Aflatoxin contamination has become a major concern worldwide. Biological methods have been used to reduce aflatoxins in food and feed by inhibiting toxin production and detoxification. Among biological methods, lactic acid bacteria are of significant interest because of their safety, efficiency, and environmental friendliness. This study aimed to review the mechanisms by which lactic acid bacteria degrade aflatoxins and the factors that influence their degradation efficiency, including the action of the lactic acid bacteria themselves (cell wall adsorption) and the antifungal metabolites produced by the lactic acid bacteria. The current applications of lactic acid bacteria to food and feed were also reviewed. This comprehensive analysis provided insight into the binding mechanisms between lactic acid bacteria and aflatoxins, facilitating the practical applications of lactic acid bacteria to food and agriculture.

Abstract:Aflatoxin B1 (AFB1) is one of the most harmful mycotoxins, raising serious global health and economic problems. Searching for biological approaches for effective and safe AFB1 degradation is imminent. In our study, Microbacterium proteolyticum B204 isolated from bovine faeces degraded 77% of AFB1 after 24 hour, becoming the first reported bacteria from the Microbacterium family to possess AFB1 degradation characteristics. Temperature variation showed little effect on its degradation ratio, demonstrating high thermostability of 75% and 79% after boiling and sterilization, respectively. We suppose that the components playing a key role during this process were proteins, considering the decreased degradation rate caused by Proteinase K. Cell viability detection on HepG2 cells indicated that the degradation products were much less toxic than pure AFB1. Furthermore, B204 cell-free culture supernatant also degraded AFB1-contaminated food, such as peanuts, corn and cheese. These results suggested that this strain with AFB1 degradation properties could be a prospective candidate for application in the food and feed industries.

Abstract:Aflatoxins (AFTs) are toxic products of fungal metabolism, associated with serious health consequences and substantial economic losses to agriculture, livestock and poultry sectors, particularly in the developing countries. This review outlines the current information on AFTs in terms of historical background, classification, relative occurrence and co-existence with other mycotoxins in various food commodities. The phenomenon of aflatoxin (AFT) biosynthesis has been elucidated with reference to molecular basis, genetic regulation and factors affecting the AFT production. Moreover, the in vivo disposition kinetics, toxicological action and toxico-pathological consequences of AFTs have also been highlighted. Currently employed strategies for the detection and detoxification of AFTs, biomarkers of exposure assessment, potential economic impact and regulatory considerations regarding the AFTs have been emphasized.
Effect of Lactic Acid Bacteria Strains on the Growth and Aflatoxin Production Potential of Aspergillus parasiticus, and Their Ability to Bind Aflatoxin B1, Ochratoxin A, and Zearalenone in vitro

Abstract:The increased consumption of plant-based foods has intensified the concern related to mycotoxin intoxication. This study aimed to investigate the effect of selected lactic acid bacteria (LAB) strains on the growth of Aspergillus parasiticus NRRL 2999 and its production of aflatoxin (AF). The ability of the heat-killed (100°C for 1 h) LAB strains to bind aflatoxin M 1 (AFM 1 ) in milk and aflatoxin B 1 (AFB 1 ), ochratoxin A (OTA), and zearalenone (ZEN) in potassium phosphate buffer (PPB) was also evaluated in vitro . Ten LAB strains were tested individually, by inoculating them simultaneously with the fungus or after incubation of the fungus for 24 or 48 h at 25°C. Double layer yeast extract sucrose (YES) agar, de Man Rogosa and Sharpe (MRS) agar, and YES broth were incubated for 7 days at 25°C to follow the development of the fungus. Levilactobacillus spp. 3QB398 and Levilactobacillus brevis 2QB422 strains were able to delay the growth of A. parasiticus in YES broth, even when these strains were inoculated 24 h after the fungus. The inhibitory effect of these LAB strains was confirmed by the reduction of fungus colony size, suggesting dominance of LAB by competition (a Lotka-Voltera effect). The production of AFB 1 by A. parasiticus was inhibited when the fungus was inoculated simultaneously with Lactiplantibacillus plantarum 3QB361 or L. plantarum 3QB350. No AFB 1 was found when Levilactobacillus spp. 2QB383 was present, even when the LAB was inoculated 48 h after the fungus. In binding studies, seven inactivated LAB strains were able to promote a reduction of at least 50% the level of AFB 1 , OTA, and ZEN. This reduction varied depending on the pH of the PPB. In milk, however, only two inactivated LAB strains were able to reduce AFM 1 , with a reduction of 33 and 45% for Levilactobacillus spp. 3QB398 ( Levilactobacillus spp.) and L. brevis 2QB422, respectively. Nevertheless, these results clearly indicate the potential of using LAB for mycotoxin reduction.

Biological Degradation of Aflatoxin B1 by Cell-Free Extracts of Bacillus velezensis DY3108 with Broad PH Stability and Excellent Thermostability

Abstract:(1) Background: Aflatoxin contamination in food and grain poses serious problems both for economic development and public health protection, thus leading to a focus on an effective approach to control it; (2) Methods: Aflatoxin B1 (AFB1) degrading bacteria were isolated using a medium containing coumarin as the sole carbon source, and the biodegradation of AFB1 by the isolate was examined by high performance liquid chromatography, and liquid chromatography mass spectrometry; (3) Results: a bacterial strain exhibiting strong AFB1 degradation activity (91.5%) was isolated and identified as Bacillus velezensis DY3108. The AFB1 degrading activity was predominantly attributed to the cell-free supernatant of strain DY3108. Besides, it was heat-stable and resistant to proteinase K treatment but sensitive to sodium dodecyl sulfate treatment. The optimal temperature for the maximal degradation of AFB1 was 80 °C. Even more notable, the supernatant showed a high level of activity over a broad pH (4.0 to 11.0) and exhibited the highest degradation (94.70%) at pH 8.0. Cytotoxicity assays indicated that the degradation products displayed significantly (p < 0.05) lower cytotoxic effects than the parent AFB1; (4) Conclusions: B. velezensis DY3108 might be a promising candidate for exploitation in AFB1 detoxification and bioremediation in food and feed matrices.
