Abstract:This study aimed to diagnose the ability of two probiotic strains isolated from Siahmazgi cheese to reduce the concentration of aflatoxin M1 (AFM1), which was able to reduce it up to 50%. Two strains of Lactobacillus isolated from local Siahmazgi cheese, were inoculated into raw milk containing 10 ng/mL AFM1. AFM1 levels were assessed by HPLC at 0, 24, 48 and 72 h. Also, for the assessment of the possible reduction of AFM1 by non-viable bacterial cells, heat treatment was applied in milk. In general, significant differences were observed between the control group and AFM1 contaminated milk samples containing probiotics during 72 h of refrigeration ( p < 0.05). In the presence of Lactobacillus brevis , Lactobacillus plantarum and mixture probiotics, the concentrations of AFM1 ranged from 0.102 ± 0.004 ng/mL to 0.052 ± 0.002 ng/mL, 0.114 ± 0.002 ng/mL to 0.074 ± 0.003 ng/mL and 0.108 ± 0.003 ng/mL to 0.065 ± 0.002 ng/mL, respectively. Also, in the heated group, just a decrease was observed in the first 24 h. The results showed that native Iranian strains of Lactobacillus have good potential in reducing AFM1 in milk. The time factor is effective in reducing AFM1 by living bacterial cells.

Abstract:Abstract Aflatoxins are naturally occurring toxic substances produced mainly by species of the genus Aspergillus that can contaminate almost all foodstuffs. Apart from the harmful effects they have on human and animal health, they can be secreted unchanged in animal milk and cause contamination of milk and its products. Aflatoxin M 1 (AFM 1 ) is the major and most toxic type of aflatoxin after aflatoxin B 1 (AFB 1 ). The use of probiotic strains to reduce the amount of aflatoxin in milk and by-products has been observed in many studies. In this systematic review and meta-analysis, articles in PubMed, Scopus, Google Scholar and ISI Web of Science were searched to find eligible studies that reported reduction of AFM 1 using probiotics in dairy products. The results were pooled using a random-effects model. In most studies, the efficiency of probiotics in milk has been tested by Lactobacillus strains. The results indicated that probiotic microorganisms could significantly reduce AFM 1 by 55.76% (confidence interval (CI): 54.35%, 57.16%; I 2 = 100%). Besides, the rank order of AFM 1 reduction in dairy products based on probiotic strain subgroup was: Lactobacillus 51.99% (CI: 50.14%, 53.85%, I 2 = 100%), Saccharomyces 67.36% (CI: 65.05%, 69.67%, I 2 = 100%), Bifidobacterium 54.80% (CI: 54.18%, 55.43%, I 2 = 99.9%), and 61.90% (CI: 53.80%, 70.00%, I 2 = 100%) by a mix of strains. Considering the high binding potential of AFM 1 to probiotic strains, these microorganisms can be recommended as a safe system to reduce AFM 1 in dairy products.

Abstract:Aflatoxin M1 (AFM1) is a mycotoxin that often contaminates milk. Like other mycotoxins, it is thermostable and potentially carcinogenic. The present work was carried out to evaluate the ability of microorganisms isolated from Indonesian kefir grains to reduce AFM1 in contaminated phosphate buffer saline (PBS). Fourteen isolates of lactic acid bacteria, both aerobic (LAE) and anaerobic (LAN), and nine isolates of yeast (YEA) were used. The significantly highest AFM1 reduction percentage was shown by the isolate LAE7 (29.3 ± 0.6%) after 4 h incubation. DNA sequencing of LAE7 and YEA2 isolates showed that these isolates had homology (level of similarity) with species of Lactobacillus kefiri strain A/K and Saccharomyces cerevisiae NRRL Y-12632, respectively. The present work proved that isolates from Indonesian kefir grains could reduce AFM1 and have the potential for practical use.

Abstract:A comparison of experimental data and two-dimensional numerical simulations of polysilicon thin-film transistors (TFTs) is presented. It is shown that avalanche multiplication causes both the kink effect in the output characteristics and the reduction of threshold voltage in short-channel device. It is shown that exactly the same physical model for avalanche multiplication gives very good agreement between simulations and experimental data for both these effects. It is demonstrated that it is the presence of grain boundaries or traps in the polysilicon that causes avalanche effects to be much greater than in comparable single-crystal silicon devices.<<ETX>>

Abstract:The increased consumption of plant-based foods has intensified the concern related to mycotoxin intoxication. This study aimed to investigate the effect of selected lactic acid bacteria (LAB) strains on the growth of Aspergillus parasiticus NRRL 2999 and its production of aflatoxin (AF). The ability of the heat-killed (100°C for 1 h) LAB strains to bind aflatoxin M 1 (AFM 1 ) in milk and aflatoxin B 1 (AFB 1 ), ochratoxin A (OTA), and zearalenone (ZEN) in potassium phosphate buffer (PPB) was also evaluated in vitro . Ten LAB strains were tested individually, by inoculating them simultaneously with the fungus or after incubation of the fungus for 24 or 48 h at 25°C. Double layer yeast extract sucrose (YES) agar, de Man Rogosa and Sharpe (MRS) agar, and YES broth were incubated for 7 days at 25°C to follow the development of the fungus. Levilactobacillus spp. 3QB398 and Levilactobacillus brevis 2QB422 strains were able to delay the growth of A. parasiticus in YES broth, even when these strains were inoculated 24 h after the fungus. The inhibitory effect of these LAB strains was confirmed by the reduction of fungus colony size, suggesting dominance of LAB by competition (a Lotka-Voltera effect). The production of AFB 1 by A. parasiticus was inhibited when the fungus was inoculated simultaneously with Lactiplantibacillus plantarum 3QB361 or L. plantarum 3QB350. No AFB 1 was found when Levilactobacillus spp. 2QB383 was present, even when the LAB was inoculated 48 h after the fungus. In binding studies, seven inactivated LAB strains were able to promote a reduction of at least 50% the level of AFB 1 , OTA, and ZEN. This reduction varied depending on the pH of the PPB. In milk, however, only two inactivated LAB strains were able to reduce AFM 1 , with a reduction of 33 and 45% for Levilactobacillus spp. 3QB398 ( Levilactobacillus spp.) and L. brevis 2QB422, respectively. Nevertheless, these results clearly indicate the potential of using LAB for mycotoxin reduction.

Abstract:The health risks caused by aflatoxins, as one of the most important contaminants of human food and feed and the main cause of cancer, especially hepatocellular carcinoma (HCC) were investigated. The aim of the study was to assess the antimutagenic effects of Bifidobacterium lactis (B. lactis) probiotic against aflatoxin B1 (AFB1). The study was conducted with 27 treatments and three replications. The independent variables were aflatoxin concentrations at three levels of 5, 15, and 25 ng/g and probiotic content in three forms of cellular sedimentation (CS), cell-free supernatant (CFS), and cell suspension. The antimutagenic activity of B. lactis against AFB1 was measured. The lowest score of antimutagenic activity of B. lactis was observed in bacterial cellular sediment treatment at 10 7 CFU/g and 25 ng/g of AFB1 (20.8 ± 3.80%) and the highest score was achieved with cell suspension at 10 9 CFU/g and 5 ng/g of AFB1 (74.9 ± 7.11%). In addition, the lack of mutagenicity of probiotics was confirmed. Therefore, probiotics not only alleviate aflatoxin in food matrices and benefit the consumer, but also notably decrease mutagenicity of AFB1.

Virus enhancement following infection with antibody-coated infectious bursal disease virus (IBDV) in chickens.

Abstract:A novel concept of vaccination, employing virus-antibody complex has been reported for the control of infectious bursal disease in chickens. A comparison of virus replication, serum neutralizing antibody response and pathogenicity in chickens inoculated with the antibody coated virus, prepared by mixing virus and antibody in different ratios (1:1, 1:0.1, 1: 0.01) and virus alone without antibody, has been made. Antibody coated virus (when mixed in certain crucial ratios) replicated to a higher magnitude in the target organ, caused more severe pathogenesis but induced a primary serum neutralizing antibody response almost comparable. The results may have important implications in understanding of pathogenesis and development of control strategies against infectious bursal disease virus, specially employing immune complex vaccine.

Abstract:The current study aimed to evaluate aflatoxin M1 (AFM1) contamination in a total number of 240 samples collected during four consecutive seasons (from January to November 2022) from two western Iranian provinces (Ilam and Lorestan). AFM1 was determined by high-performance liquid chromatography with a fluorescence detector after the toxin extraction with immunoaffinity columns. The results showed 100% of AFM1 contamination in all the surveyed matrices. Considering the seasonal variability, AFM1 mean values in milk, cheese, and yoghurt samples collected during the summer were significantly higher than those detected in samples from other seasons (p < 0.01), and the lowest concentrations were observed in samples collected during the winter. AFM1 levels in cheese samples appeared safe, but an alarming rate of 46.6% of milk samples and 100% of the yoghurt samples exceeded the maximum permissible limit set by the European Union (50 ng/kg). Moreover, while the deterministic risk assessment demonstrated no risk of developing liver cancer in adults through the consumption of these dairy products, it showed significant concerns for children consuming milk and yoghurt (hazard index above 1). Overall, the results of this research indicate the necessity of a periodical monitoring program for AFM1 in milk and other dairy products.

Abstract:Transcription factor IID (TFIID) recognizes the TATA element of promoters transcribed by RNA polymerase II (RNAPII) and serves as the base for subsequent association by other general transcription factors and RNAPII. The carboxyl-terminal domain of TFIID is highly conserved and contains an imperfect repetition of a 60-amino acid sequence. These repeats are separated by a region rich in basic amino acids. Mutagenesis of the lysines in this region resulted in a conditioned phenotype in vivo, and the mutant proteins were defective for interactions with transcription factor IIA in vitro. Binding of TFIID to DNA was unaffected. These results suggest that the basic domain of TFIID is important for protein-protein interactions.
Effects of Lactic Acid Bacteria Reducing the Content of Harmful Fungi and Mycotoxins on the Quality of Mixed Fermented Feed

Abstract:The contamination of fermented feeds and foods with fungi and mycotoxins is a major food safety issue worldwide. Certain lactic acid bacteria (LAB), generally recognized as safe (GRAS) fermentation probiotics, are able to reduce microbial and mycotoxins contamination. In this study, Lactiplantibacillus (L.) plantarum Q1-2 and L. salivarius Q27-2 with antifungal properties were screened as inoculants for mixed fermenting feed, and the fermentation and nutritional qualities, microbial community, and mycotoxins of mixed fermented feed were analyzed at different fermentation periods (1, 3, 7, 15, and 30 days, respectively). The findings indicated that the utilization of Q1-2 and Q27-2 strains in fermenting feed led to a decrease in pH and an increase in lactic acid concentration and the proportion of Lactiplantibacillus, while effectively restraining the proliferation of undesirable microorganisms. In particular, Q1-2 reduced the relative abundance of fungi including Fusarium and Aspergillus. Compared to the control group, the Q1-2 and Q27-2 groups reduced aflatoxin B1 by 34.17% and 16.57%, and deoxynivalenol by up to 90.61% and 51.03%. In short, these two LAB inoculants could reduce the contents of aflatoxin B1 and deoxynivalenol to the limited content levels stipulated by the Chinese National Standard GB 13078-2017. These findings suggest that the LAB strains of Q1-2 and Q27-2 have potential applications in the feed industry for the mitigation of mycotoxin pollution, thereby enhancing the quality of animal feed.

Abstract:Food contamination with aflatoxin is one of the most critical concerns of health professionals. One of the best ways to reduce aflatoxin content in food is probiotics. Therefore, this study was performed to isolate Bifidobacterium from the chick's intestine; evaluate its probiotic activities and its application with Polyvinylpyrrolidone (PVP) to reduce aflatoxin B1 (AFB1) in the medium were investigated. Samples were isolated from the chick's intestine, and Bifidobacterium was isolated and identified by biochemical and molecular methods. Next, the potential probiotic characterization was assessed. Afterward, the effect of selected isolate and PVP on reducing AFB1 in the medium was studied using ELISA and HPLC. Biochemical and molecular evaluations indicated isolation of Bifidobacterium bifidum strain from chick's intestine. One of the B. bifidum strains was selected for the next steps, which showed potential probiotic characterization and the ability to reduce the concentration of AFB1 in the medium (50% reduction). When used in combination with PVP showed synergistic effects in reducing the concentration of AFB1 from the medium (up to 90%). In conclusion, it was found that selected B. bifidum strains and PVP could have synergistic effects in reducing AFB1 toxin in a medium up to 90%.

Safety, probiotic properties, antimicrobial activity, and technological performance of Lactobacillus strains isolated from Iranian raw milk cheeses

Abstract:The objective of this study was to investigate probiotic, antimicrobial, technological and safety properties of lactobacillus strains isolated from local Iranian cheese made from raw milk. Six different samples were prepared, after serial dilution, culture was performed on MRS culture medium. The gram-positive and catalase-negative lactobacillus strains were subjected to grouping and identifying using biochemical tests, carbohydrates fermentation profiles, and 16S rDNA analysis. The results of sequence analysis showed the Lactobacillus spp. belonged to Lactobacillus brevis, Lactobacillus acidophilus, Lactobacillus plantarum, and Lactobacillus casei. After 3 hr incubation at pH=2, 3–6 log units of strains decreased which Lactobacillus acidophilus (B14) and Lactobacillus brevis (B2) showed highest resistance to low pH as well as simulated GIT juices. The highest and lowest hydrophobicity degree was belonged to L. acidophilus (B14) (65.9%) and L. casei (B22) (25.6%), respectively. Also, the highest auto-aggregation and coaggregation were observed in L. acidophilus (B14) (51.3%) and L. plantarum (B20) (43.6%). The adhered percentage of strains varied from 2.5% to 14.6%. L. plantarum (B20) showed highest proteolytic activity followed by L. acidophilus (B14). Also, the highest autolytic activity belonged to L. acidophilus (B14). All of the strains showed low acidifying potential, except for L. acidophilus (B17) which decreased 2.05 unit of pH after 24 hr. The isolates did not show lipolytic activity as well as biogenic amines production (except L. brevis B3). All of the strains were sensitive to chloramphenicol and erythromycin except L. acidophilus (B15) and L. casei (B22). All strains showed no hemolysis activity which make them safe for consumption. Based on the obtained results, L. acidophilus (B14) presented the best probiotic and technological characteristics and is proposed for using as coculture in the dairy industrial.

Abstract:The objective of this study was to investigate the occurrence of aflatoxin M-1 (AFM(1)) in Chinese liquid milk products. A total of 190 liquid milk samples, including 168 ultrahigh-temperature-treated milk samples and 22 pasteurized milk (PM) samples, were collected in August 2019. A screening assay with the Charm rapid test kit and a confirmation method with high-performance liquid chromatography were used for AFM(1) analysis. Nine (4.74%) samples were screened positive, of which 5 (2.11%) samples were confirmed with concentration levels of 0.022 to 0.049 mu g/kg. The AFM(1) levels confirmed were all below the maximum residue levels set by China, the European Union, the United States, and the Codex Alimentarius Commission. The detection rate of AFM(1) in domestic milk samples was 3.39%, while no AFM(1) was detected in samples of imported milk. The prevalences of AFM(1) detected in three groups of brands were as follows: group I, the major brands of China. 2.70%; group II, the local city domestic brands, 4.55%; and group III, the brands imported into China, 0. The detection rate of AFM(1) was significantly higher in PM samples (9.09%) than in ultrahigh-temperature-treated samples (1.19%) (P < 0.05). Although the residue level of AFM(1) did not exceed the maximum residue levels in any of the samples, the higher detection rate in local Chinese brands, especially in PM samples, deserves the attention of the Chinese government and consumers.
Quantitative analysis and carcinogenic/non-carcinogenic risk assessment of aflatoxin M1 in milk-based baby food and infant formula milk – a case study in Iran

Abstract:In this study, solid-phase extraction (SPE) combined with the dispersive liquid–liquid microextraction based on novel hydrophobic deep eutectic solvent (DLLME − DES) has been developed as an ultra-pre-concentration technique for the extraction of aflatoxin M 1 (AFM1) in milk-based baby food (MBBF) and infant formula milk (IFM) samples followed by HPLC combined with fluorescence detection (HPLC − FL). In addition, carcinogenic and non-carcinogenic risk assessment was performed by health-related risk factors including liver cancer risk (LCR), margin of exposure (MOE) and target hazard quotient (THQ) were calculated using the mean of AFM1 in different infant food samples. The results of the study showed that the mean of AFM1 was statistically significant different between various brands and types of IFM and MBBF. The results of the study showed that the percentage of positive samples higher than the allowable limit of AFM1 in 36 samples of domestic infant formula milk (DIFM), 24 samples of imported infant formula milk (IIFM), 36 samples of domestic milk-based baby food (DMBBF) and 18 samples of imported milk-based baby food (IMBBF) were 41.6, 12.5, 66.7 and 33.3%, respectively. In addition, estimated values for health risk-related factors including LCR, MOE and THQ indicated that for most infants less than one-year-old were higher than the acceptable levels. Based on the results, it can be concluded that the quality of IFM and MBBF consumed in Iran in terms of AFM1 is poor. Therefore, it is necessary to take appropriate measures to reduce the amount of AFM1 in DIFM and DMBBF, and in addition, the IIFM and IMBBF should be controlled qualitatively before supplying the market.

Abstract:Aflatoxin M1 (AFM1) contamination in dairy products is a growing concern. In Iran, where dairy consumption is integral to daily life, understanding the extent of AFM1 exposure is crucial. This study presents a comprehensive human health risk assessment (HHRA) of AFM1 in six popular dairy products: yogurt, kashk, ice cream, doogh, butter, and tarkhineh through a systematic review. This study used Monte Carlo simulations to estimate the dietary exposure to AFM1 across different age groups. Other HHRA parameters, including liver cancer risk (LCR), hazard index (HI), and margin of exposure (MoE), were also used to explore safety concerns arising from AFM1 contamination. In addition, statistics, uncertainty, and sensitivity analysis for AFM1 in dairy products across various ages were performed. Our findings reveal varying levels of AFM1 contamination, with 67.54% of the products being contaminated with AFM1, of which 12.49% exceeded the regulatory limits set by the European Commission (EC) and Institute of Standards & Industrial Research of Iran (ISIRI) regulations. The results highlight the importance of strict monitoring and control measures to minimize AFM1 contamination and mitigate the associated health risks. This research is a valuable resource for food safety authorities and the dairy industry to enhance the safety and quality of dairy products in Iran.

In this study, viable and heat-inactivated L. acidophilus CIP 76.13T and L. delbrueckii subsp. bulgaricus CIP 101027T cells were investigated for their ability to remove aflatoxin B1 (AFB1), ochratoxin A (OTA), zearalenone (ZEA), and deoxynivalenol (DON) from MRS medium and PBS buffer over a 24 h period at 37 °C. LAB decontamination activity was also assessed in a ZEA-contaminated liquid feed (LF). Residual mycotoxin concentrations were determined by UHPLC-FLD/DAD analysis. In PBS, viable L. acidophilus CIP 76.13T and L. delbrueckii subsp. bulgaricus CIP 101027T cells removed up to 57% and 30% of ZEA and DON, respectively, while AFB1 and OTA reductions were lower than 15%. In MRS, 28% and 33% of ZEA and AFB1 were removed, respectively; OTA and DON reductions were small (≤15%). Regardless of the medium, heat-inactivated cells produced significantly lower mycotoxin reductions than those obtained with viable cells. An adsorption mechanism was suggested to explain the reductions in AFB1 and OTA, while biodegradation could be responsible for the removal of ZEA and DON. Both viable LAB strains reduced ZEA by 23% in contaminated LF after 48 h of incubation. These findings suggest that LAB strains of L. acidophilus CIP 76.13T and L. delbrueckii subsp. bulgaricus CIP 101027T may be applied in the feed industry to reduce mycotoxin contamination.

Abstract:Background: Milk and dairy products are essential for global nutrition, and ensuring their safety is crucial. Aflatoxin M1 (AFM1), a carcinogen from contaminated animal feed, poses a significant threat to consumers. Objectives: This “health policy brief" highlights the critical need for meticulous and constant monitoring of Iran's animal feed quality and storage conditions to minimize AFM1 contamination in dairy products. Methods: Comprehensive review papers on AFM1 were reviewed in Iranian dairy products, drawing evidence from various databases. Results: The prevalence of AFM1 contamination in Iranian dairy products, particularly risky for children due to extensive consumption, exceeds safety limits, with livestock feed identified as the primary source. Public health demands rigorous monitoring practices, such as stricter feed quality and storage regulations, regular AFM1 testing, stakeholder collaboration, and public awareness. There are many benefits associated with these measures, including lowering AFM1 levels in dairy products, improving food safety standards, minimizing AFM1 exposure risks, especially for children, and increasing consumer confidence in the integrity of the Iranian dairy industry. Conclusions: Continuous monitoring of animal feed is crucial to safeguard public health and ensure the safety of Iranian dairy products, particularly for children. This comprehensive approach, collaboration, and public awareness are vital to mitigating AFM1 contamination and promoting Iran's safe and robust dairy industry.

Abstract:In the present study, the antimicrobial activity of kefir and probiotic yogurt produced from cow, camel, ewe, and goat milk on pathogenic bacteria (Staphylococcus aureus, Listeria monocytogenes, Escherichia coli, and Salmonella enterica) and fungi (Aspergillus niger, Penicillium sp., and Fusarium sp.) during 20-day storage period at 4 ºC was investigated. The carbohydrate content and pH of milk samples decreased during the fermentation process to produce probiotic yogurt and kefir, also the acidity increased significantly. The results revealed that kefir samples had stronger antifungal and antibacterial effect than probiotic yogurt samples. Among kefir samples, the ewe and cow milk kefir expressed the highest and the lowest antimicrobial activity, respectively. A. niger, S. aureus, and L. monocytogenes were the most sensitive and Penicillium sp. and E. coli were the most resistant microorganisms against treatment by kefir and probiotic yogurt.Graphic abstract

Abstract:This study aimed to investigate the potential ability of simultaneously used L. acidophilus(LA‐5), L.rhamnosus(LGG), and L.casei(LC‐01) in encapsulated (E) and nonencapsulated (NE) forms in mycelial growth of Aspergillus spp and aflatoxin production by A. flavus. In order to assess the zone of fungal growth inhibition by E and NE lactic acid bacteria, the agar well diffusion method was applied. Quantification of aflatoxin was performed using a high‐performance liquid chromatography technique. Lactic acid bacteria exhibited high antifungal activity and significantly reduced AFB1, AFB2, AFG1, and AFG2 production in both E and NE forms compared to the control group. The percentage of reduction in total AFs production in treated samples with E and NE lactic acid bacteria was 94.1% and 95.5%, respectively. These results suggested that simultaneously used lactic acid bacteria in E and NE forms can prevent growth and decrease aflatoxin production of toxigenic aspergilla.
