Abstract:The objective of this study was to investigate the occurrence of aflatoxin M-1 (AFM(1)) in Chinese liquid milk products. A total of 190 liquid milk samples, including 168 ultrahigh-temperature-treated milk samples and 22 pasteurized milk (PM) samples, were collected in August 2019. A screening assay with the Charm rapid test kit and a confirmation method with high-performance liquid chromatography were used for AFM(1) analysis. Nine (4.74%) samples were screened positive, of which 5 (2.11%) samples were confirmed with concentration levels of 0.022 to 0.049 mu g/kg. The AFM(1) levels confirmed were all below the maximum residue levels set by China, the European Union, the United States, and the Codex Alimentarius Commission. The detection rate of AFM(1) in domestic milk samples was 3.39%, while no AFM(1) was detected in samples of imported milk. The prevalences of AFM(1) detected in three groups of brands were as follows: group I, the major brands of China. 2.70%; group II, the local city domestic brands, 4.55%; and group III, the brands imported into China, 0. The detection rate of AFM(1) was significantly higher in PM samples (9.09%) than in ultrahigh-temperature-treated samples (1.19%) (P < 0.05). Although the residue level of AFM(1) did not exceed the maximum residue levels in any of the samples, the higher detection rate in local Chinese brands, especially in PM samples, deserves the attention of the Chinese government and consumers.

Abstract:A comparison of experimental data and two-dimensional numerical simulations of polysilicon thin-film transistors (TFTs) is presented. It is shown that avalanche multiplication causes both the kink effect in the output characteristics and the reduction of threshold voltage in short-channel device. It is shown that exactly the same physical model for avalanche multiplication gives very good agreement between simulations and experimental data for both these effects. It is demonstrated that it is the presence of grain boundaries or traps in the polysilicon that causes avalanche effects to be much greater than in comparable single-crystal silicon devices.<<ETX>>

Abstract:In this study, the average level of aflatoxin M1 in various types of milk from 107 articles (297 studies with 16,274 milk samples) were meta-analyzed using random-effect model based on the milk varieties (animal species and heating processes), geographical regions, seasons, detection techniques and dairy farming subgroups. Studies on milk contamination with aflatoxin M1 in Iran were collected using universal and Persian databanks from January 1974 to the end of November 2021. The overall aflatoxin M1 mean concentration and prevalence in milk samples of Iran were 39.65 ng/l (95% CI: 36.00–43.30) and 80% (95% CI: 76–85%), respectively. The rank order of importance of various variables in mean levels of aflatoxin M1 in milk samples included milk type (animal species) > geographical regions > detection techniques > dairy farming types > milk types (heating processes) > seasons. Findings revealed that the overall content of aflatoxin M1 in milk samples of Iran was lower than that allowed by the European Union, Institute of Standards and Industrial Research of Iran, and the USA, possibly due to the milk monitoring by the Iranian regulatory systems.

Abstract:The current study aimed to evaluate aflatoxin M1 (AFM1) contamination in a total number of 240 samples collected during four consecutive seasons (from January to November 2022) from two western Iranian provinces (Ilam and Lorestan). AFM1 was determined by high-performance liquid chromatography with a fluorescence detector after the toxin extraction with immunoaffinity columns. The results showed 100% of AFM1 contamination in all the surveyed matrices. Considering the seasonal variability, AFM1 mean values in milk, cheese, and yoghurt samples collected during the summer were significantly higher than those detected in samples from other seasons (p < 0.01), and the lowest concentrations were observed in samples collected during the winter. AFM1 levels in cheese samples appeared safe, but an alarming rate of 46.6% of milk samples and 100% of the yoghurt samples exceeded the maximum permissible limit set by the European Union (50 ng/kg). Moreover, while the deterministic risk assessment demonstrated no risk of developing liver cancer in adults through the consumption of these dairy products, it showed significant concerns for children consuming milk and yoghurt (hazard index above 1). Overall, the results of this research indicate the necessity of a periodical monitoring program for AFM1 in milk and other dairy products.

Abstract:This study aims to investigate the occurrence of aflatoxin B1 (AFB1) in total mixed rations (TMR) and aflatoxin M1 (AFM1) in raw and commercial milk in northern China during winter season. A total of 22 feed samples, as well as the corresponding 84 raw milk and 69 commercial milk samples were randomly collected from north-west, northeast and north areas of China from November 2019 to January 2020. High performance liquid chromatography and enzyme linked immunosorbent assay method were applied to detect AFB1 and AFM1. All samples presented AFB1 (30?370 ng/L) and AFM1 (10?430 ng/L) levels under the legal limit of China for raw milk (10 ?g/L) and commercial milk (500 ng/L). While 31.8% of TMRs and 34.5% of raw milk samples exceeded the European Union (EU) legal limit for AFM1 in raw milk (5 ?g/L) and commercial milk (50 ng/L). Compared with the samples from northeast and north areas of China with the similar AFB1 concentration, samples from northwest area of China exhibited higher AFM1 level (mean = 130 ng/L), thus presenting a higher transfer ratio from AFB1 to AFM1. A Linear relationship was observed between AFB1 in TMRs and AFM1 in raw milk. In conclusion, all samples from northern China presented AFB1 and AFM1 levels under Chinese legal limit and only about 30% of the tested samples exceeded the EU legal limit. However, the aflatoxin contamination in northwest area of China should be paid more attention to due to the high AFM1 levels and high variance in milk samples.

Abstract:In this study, a total amount of 868 samples of raw cows' milk, pasteurized and UHT cows' milk, and dairy products were analyzed for their AFM1 content. Milk and dairy consumption in Lebanon was evaluated through a food frequency questionnaire, and accordingly the exposure to AFM1 and its association with liver cancer risk in Lebanese population were evaluated. Results showed contamination in raw milk, pasteurized and UHT milk, and dairy products at a range of 0.011–0.440 μg/L, 0.013–0.219 μg/L, and 0.015–7.350 μg/L respectively; with 28%, 54.5%, and 45.5% respectively of samples with AFM1 above maximum tolerable limit (MTL) set by the European Commission. AFM1 consumption was shown to be associated with 0.0041 additional cancer cases per 100,000 persons per year. Based on these results, milk and dairy consumption in Lebanon can be considered hazardous and may present a significant risk on the health of the Lebanese population especially children.

Abstract:In this study, solid-phase extraction (SPE) combined with the dispersive liquid–liquid microextraction based on novel hydrophobic deep eutectic solvent (DLLME − DES) has been developed as an ultra-pre-concentration technique for the extraction of aflatoxin M 1 (AFM1) in milk-based baby food (MBBF) and infant formula milk (IFM) samples followed by HPLC combined with fluorescence detection (HPLC − FL). In addition, carcinogenic and non-carcinogenic risk assessment was performed by health-related risk factors including liver cancer risk (LCR), margin of exposure (MOE) and target hazard quotient (THQ) were calculated using the mean of AFM1 in different infant food samples. The results of the study showed that the mean of AFM1 was statistically significant different between various brands and types of IFM and MBBF. The results of the study showed that the percentage of positive samples higher than the allowable limit of AFM1 in 36 samples of domestic infant formula milk (DIFM), 24 samples of imported infant formula milk (IIFM), 36 samples of domestic milk-based baby food (DMBBF) and 18 samples of imported milk-based baby food (IMBBF) were 41.6, 12.5, 66.7 and 33.3%, respectively. In addition, estimated values for health risk-related factors including LCR, MOE and THQ indicated that for most infants less than one-year-old were higher than the acceptable levels. Based on the results, it can be concluded that the quality of IFM and MBBF consumed in Iran in terms of AFM1 is poor. Therefore, it is necessary to take appropriate measures to reduce the amount of AFM1 in DIFM and DMBBF, and in addition, the IIFM and IMBBF should be controlled qualitatively before supplying the market.

Abstract:Aflatoxin M1 (AFM1) contamination in dairy products is a growing concern. In Iran, where dairy consumption is integral to daily life, understanding the extent of AFM1 exposure is crucial. This study presents a comprehensive human health risk assessment (HHRA) of AFM1 in six popular dairy products: yogurt, kashk, ice cream, doogh, butter, and tarkhineh through a systematic review. This study used Monte Carlo simulations to estimate the dietary exposure to AFM1 across different age groups. Other HHRA parameters, including liver cancer risk (LCR), hazard index (HI), and margin of exposure (MoE), were also used to explore safety concerns arising from AFM1 contamination. In addition, statistics, uncertainty, and sensitivity analysis for AFM1 in dairy products across various ages were performed. Our findings reveal varying levels of AFM1 contamination, with 67.54% of the products being contaminated with AFM1, of which 12.49% exceeded the regulatory limits set by the European Commission (EC) and Institute of Standards & Industrial Research of Iran (ISIRI) regulations. The results highlight the importance of strict monitoring and control measures to minimize AFM1 contamination and mitigate the associated health risks. This research is a valuable resource for food safety authorities and the dairy industry to enhance the safety and quality of dairy products in Iran.

Abstract:Abstract Aflatoxin M 1 (AFM 1 ) is a major metabolite of aflatoxin B 1 occurring in many feed materials commonly used in the ration of dairy cows. The level of contamination of feed materials is currently increasing in many parts of the world due to climate change. These changes emphasise the necessity to monitor AFM 1 levels in milk and dairy products as a precautionary measure to protect consumer health risk. In the current study, the AFM 1 levels were measured in raw, pasteurised, and ultra-high temperature processed (UHT) milk commercially available in Thailand. In total, 900 milk samples were analysed, making this investigation one of Thailand’s first large-scale monitoring of milk contamination. Over a period of three consecutive years (2019 to 2021), each year 100 individual samples of either AFM 1 , pasteurised and UHT milk were collected, extracted using an immunoaffinity column for AFM 1 and then quantified by a validated liquid chromatography analysis with fluorescence detection. The measured AFM 1 levels ranged between 65-1,810 ng/l, 12-87 ng/l, and 7-38 ng/l in the raw, pasteurised, and UHT milk, respectively, in 2019. In 2020, the corresponding AFM 1 concentration range was 58-1,232 ng/l, 15-65 ng/l, and 7-29 ng/l and 52-1,432 ng/l, 20-59 ng/l, and 7-33 ng/l in 2021, respectively. According to international guidance documents, a direct comparison and formal risk analysis revealed that the measured AFM 1 levels of the processed milk samples showed good compliance with the US regulatory limits and the Codex Alimentarius Commission recommendations. However, the latter applies particularly to milk and dairy products intended for the consumption of infants and young children, considering their relatively high consumption of milk and dairy products and the specific sensitivity of infants and toddlers under the age of 6 years to mutagenic and genotoxic contaminants in foods.

Abstract:This study aimed to diagnose the ability of two probiotic strains isolated from Siahmazgi cheese to reduce the concentration of aflatoxin M1 (AFM1), which was able to reduce it up to 50%. Two strains of Lactobacillus isolated from local Siahmazgi cheese, were inoculated into raw milk containing 10 ng/mL AFM1. AFM1 levels were assessed by HPLC at 0, 24, 48 and 72 h. Also, for the assessment of the possible reduction of AFM1 by non-viable bacterial cells, heat treatment was applied in milk. In general, significant differences were observed between the control group and AFM1 contaminated milk samples containing probiotics during 72 h of refrigeration ( p < 0.05). In the presence of Lactobacillus brevis , Lactobacillus plantarum and mixture probiotics, the concentrations of AFM1 ranged from 0.102 ± 0.004 ng/mL to 0.052 ± 0.002 ng/mL, 0.114 ± 0.002 ng/mL to 0.074 ± 0.003 ng/mL and 0.108 ± 0.003 ng/mL to 0.065 ± 0.002 ng/mL, respectively. Also, in the heated group, just a decrease was observed in the first 24 h. The results showed that native Iranian strains of Lactobacillus have good potential in reducing AFM1 in milk. The time factor is effective in reducing AFM1 by living bacterial cells.

Abstract:Breast milk (BM) is considered as the best source of nutrition which could have prevention effects on various diseases in the first years of a child. Along with nutritive compounds, presence of contaminants such as mycotoxins in BM could be transmitted into neonate. The aim of this study was to determine the occurrence, levels, and factors associated with the presence of aflatoxin M 1 (AFM 1 ) and ocratoxin a (OTA) in BM samples of nursing mothers in rural centers of Yazd, Iran. The presence and average AFM 1 and OTA concentration in 72 BM samples was measured by competitive ELISA. The demographic and diet parameters of nursing mothers were collected by a questionnaire and were analyzed using SPSS 18 software. AFM 1 and OTA were detected in 63 (87.5%) and 47 (65.2%) samples with the mean concentration levels of 19.46 ± 13.26 ng/L (ranges from 5.1 to 53.9) and 200 ± 160 ng/L (ranges from 100 to 2460), respectively. Of these, 32 samples (50.7%) for AFM 1 and 23 samples (48.9%) for OTA had values exceeding the limit set by the European Union regulation for infant foods (25 ng/L for AFM 1 and 500 ng/L for OTA). It was also found that the risk of AFM 1 and OTA occurrence in BM increased significantly with the consumption of beans, bread, cereals, fruit juice and crackers, and cream, respectively. This study showed that the estimated daily intake for AFM 1 and OTA by 1 month of age infants was 2.7 and 28.5 ng/kg bw/day, respectively, while, as the age of the infant increased, the values were lower and close to 0.9 and 9.9 ng/kg bw/day for AFM1 and OTA in 12 months of age infants, respectively. The high occurrence and noticeable levels of AFM 1 and OTA detected in this study indicated that some infants receive undesirable exposures to AFM 1 and OTA with breast milk. Therefore, it is recommended that mothers are advised to avoid certain foods during pregnancy and breastfeeding that are likely sources of mycotoxins.

Abstract:The highly toxic Aflatoxin M1 (AFM1) is most often detected in milk using an Enzyme-Linked-Immunosorbent Assay (ELISA) for screening purposes, while High-Performance Liquid Chromatography with Fluorescence Detector (HPLC-FL) is the reference method used for confirmation. The aim of the present study was the comparison between three commercially available ELISA kits and a newly developed HPLC-FL method for the determination of the AFM1 in milk samples. The developed HPLC-FL method was validated for the AFM1 and Aflatoxin M2 (AFM2), determining the accuracy, precision, linearity, decision limit, and detection capability with fairly good results. All three ELISA kits were also validated and showed equally good performance with high recovery rates. Moreover, the Limit Of Detection (LOD) and Limit Of Quantification (LOQ) values were found to be significantly lower than the Maximum Residue Limit (MRL) (50 ng kg−1). After the evaluation of all three commercial kits, the ELISA kit with the optimum performance along with the HPLC method was used for the determination of AFM1 in raw cow’s, goat’s, and sheep’s milk samples (396) obtained from producers in different regions of Greece. The evaluation of both methods showed that this ELISA kit could be considered as a faster and equally reliable alternative method to HPLC in routine analysis for the determination of AFM1 in milk.

Abstract:The aim of this study was to conduct an up-to-date investigation on the occurrence levels of aflatoxin M1 (AFM1) in samples of raw milk (n = 40), pasteurized milk (n = 44), ultra-high temperature (UHT) milk (n = 27), Minas cheese (n = 57), and yogurt (n = 44) traded in São Paulo state, Brazil. AFM1 was extracted from fluid milks and dairy products using immunoaffinity columns and determined by high performance liquid chromatography. AFM1 was detected at the mean level of 0.080 ± 0.071 μg/L or kg in 72 samples (34.0%) evaluated in the study (n = 212). Detectable levels of AFM1 were observed in five samples of raw milk (12.5%), 16 samples of pasteurized milk (36.4%), 13 samples of UHT milk (48.1%), 27 samples of cheese (47.4%), and 11 samples of yogurt (25.0%), although none of them had concentrations above the maximum permitted levels (MPL) for AFM1 adopted in Brazil. However, 11.7% (n = 13) of samples of raw, pasteurized, and UHT milks would have AFM1 concentrations above the MPL of 0.05 μg/L adopted in the EU. The maximum level was detected in one cheese sample containing 0.695 μg/kg. Although none of the samples exceeded the Brazilian MPL, the high frequencies of AFM1 in Brazilian milk products warrant concern about their contribution to the human exposure to aflatoxins. Because aflatoxins are among the most potent carcinogens known, the results of this trial stress the need for stringent measures in the milk production system to avoid AFM1 in milk and derived products.

Abstract:Aflatoxin B1 (AFB1) is widespread and seriously threatens public health worldwide. This study aimed to investigate AFB1 in imported hazelnut samples in northwest of Iran (Eastern Azerbaijan Province) using High-Performance Liquid Chromatography with a Fluorescent Detector (HPLC-FLD). In all tested samples AFB1 was detected. The mean concentration of AFB1 was 4.20 μg/kg and ranged from 3.145 to 8.13 μg/kg. All samples contained AFB1 levels within the maximum acceptable limit except for one sample. Furthermore, the human health risk assessment of AFB1 from consuming imported hazelnuts by Iranian children and adults was evaluated based on the margin of exposure (MoE) and quantitative liver cancer risk approaches. The MoE mean for children was 2529.76, while for adults, it was 8854.16, indicating a public health concern. The present study found that the risk of developing liver cancer among Iranian children was 0.11100736 per 100,000 people, and in the Iranian adult population was 0.0314496 cancers per 100,000 people. Since environmental conditions potentially affect aflatoxin levels in nuts, countries are advised to monitor aflatoxin contents in imported nuts, especially from countries with a conducive climate for mold growth.

Abstract:Aflatoxin M1 (AFM1) is a major carcinogenic compound found in milk and dairy products, posing a constant risk to consumers in the Mediterranean region. This study systematically reviewed AFM1 presence in these products in Mediterranean countries over a period of 20 years and estimated the relevant nutritional exposure for consumers. Using data from three databases, 596 articles were retrieved with 123 meeting the inclusion criteria. The frequency of AFM1 occurrence was 40% regardless of milk type, with non-cow milk showing an increasing trend. Moreover, the Estimated Daily Intake of AFM1 from milk and dairy product consumption and the associated Hazard Index were determined. Notably, Southeastern Mediterranean countries presented higher contamination levels, leading to elevated Estimated Daily Intake and Hhazard Index values. Thermally processed milk showed high contamination levels, and among dairy products, cheeses were the most contaminated. Reduced Graphene Oxide-Zinc Iron Oxide Nanomaterial As Selective Dispersive Solid-Phase Extraction Sorbent for Extraction and Enrichment of Aflatoxins from Milk Combined with UHPLC-MS/MS Analysis

Abstract:Fungal species capable of producing aflatoxins frequently contaminate the feed provided to dairy animals, leading to the subsequent occurrence of aflatoxin contamination in milk. Milk is renowned for its exceptional nutritional composition, and the consumption of milk contaminated with aflatoxins by humans can engender a multitude of health complications. In this study, reduced graphene oxide zinc iron oxide (rGO-Zn/Fe2O4) nanomaterial was utilized as a dispersive solid phase extraction adsorbent to separate and enrich aflatoxins (aflatoxin B1, aflatoxin B2, aflatoxin G1, aflatoxin G2, and aflatoxin M1) from milk. The rGO-Zn/Fe2O4 nanomaterial was morphologically characterized by scanning electron microscopy, transmission electron microscopy, and X-ray diffraction, indicating that Zn/Fe2O4 nanoparticles were embedded on the surface of rGO. The acetonitrile/formic acid (99/1, v/v) combined with ultrasonic treatment for a duration of 40 min expressed an efficient extraction solvent. Employing the matrix-spiked technique, different parameters of the dispersive solidphase extraction (d-SPE) method were meticulously optimized, yielding the following conditions: 2% acetonitrile as loading solvent, 15 mg of rGO-Zn/Fe2O4 as the adsorbent quantity, 15 min of ultrasonication as adsorption time, and n-hexane as the washing solvent. The desorption reagent was 5 mL acetonitrile, with a desorption time of 5 min. The combination of the dispersive solid-phase extraction (d-SPE) technique with validated UHPLC-MS/ MS quantitative analysis demonstrated excellent selectivity, a robust linear relationship (R-2 >= 0.994), remarkable sensitivity (limit of quantification ranging from 0.006 to 0.013 mu g/L), and satisfactory recovery rates of 78%-104% at three different matrix-spiked levels. Moreover, the method exhibited precision with relative standard deviations (RSD) ranging from 2.4% to 10.8%. Following appropriate pretreatment procedures, the milk samples underwent rigorous aflatoxin analysis, leading to the identification of AFM1 presence in all of the examined milk samples (n = 9). However, the contamination levels detected were found to be within the permissible limits as defined by regulatory standards.
Occurrence of Aflatoxin M1 in Raw Milk Marketed in Italy: Exposure Assessment and Risk Characterization

Abstract:The current study is based on the AFM1 contamination of milk determined from April 2013 to December 2018 in the framework of a self-control plan of six milk processing plants in Italy. These data – together with the consumption data of milk consumers – were evaluated and used for the calculation of the Estimated Daily Intake (EDI), the Hazard Index (HI), and the fraction of hepatocarcinoma cases (HCC) due to AFM1 exposure in different population groups. Altogether a total of 31,702 milk samples were analyzed, representing 556,413 tons of milk, which is an outstanding amount compared to published studies. The results indicate the monthly fluctuation of AFM1 levels through a period of nearly 6 years. The EDI of AFM1 in different population groups was in the range of 0.025–0.328 ng kg−1 body weight (bw) per day, based on the average consumption levels and weighted mean contamination of the milk in the study period. Considering average consumptions, in the groups of infants and toddlers, the HI calculation resulted in 1.64 and 1.4, respectively, while for older age groups, it was <1. The estimated fractions of HCC incidences attributable to the AFM1 intakes were 0.005 and 0.004 cases per 100,000 individuals in the 0–0.9 and 1–2.9-year age groups, respectively, and below 0.004 cases in the other age categories. The monthly average AFM1 contamination of tested milk consignments ranged between 7.19 and 22.53 ng kg−1. Although the results of this extensive investigation showed a low risk of HCC, the variability of climatic conditions throughout years that influence AFB1 contamination of feed and consequently AFM1 contamination of milk justifies their continuous monitoring and update of the risk assessment.

Abstract:Anthranylic acid derivatives are direct structural analogs of salicylic acid derivatives. They possess analgesic, anti-inflammatory, and antipyretic activity. They are similar to pyrazolones in terms of analgesic and antipyretic activity, yet they exceed the anti-inflammatory activity of salicylates. The mechanism of action of this series of nonsteroid, anti-inflammatory analgesics is not conclusively known. One of the early advances in the search for nonnarcotic analgesics was centered in the N-arylanthranilic acids. The outstanding characteristic of mefenamic acid is primarily anti-inflammatory , and secondarily, some possess analgesic properties.
