Abstract:Voltage imaging allows mapping of the membrane potential in living cells. Yet, current intensity-based imaging approaches are limited to relative membrane potential changes, missing important information conveyed by the absolute value of the membrane voltage. This challenge arises from various factors affecting the signal intensity, such as concentration, illumination intensity, and photobleaching. Here, we demonstrate electronic preresonance hyperspectral stimulated Raman scattering (EPR-hSRS) for spectroscopic detection of the membrane voltage using a near-infrared-absorbing microbial rhodopsin expressed in E. coli. This newly developed near-infrared active microbial rhodopsin enables electronic preresonance SRS imaging at high sensitivity. By spectral profiling, we identified voltage-sensitive SRS peaks in the fingerprint region in single E. coli cells. These spectral signatures offer a new approach for quantitation of the absolute membrane voltage in living cells.

Abstract:Introduction: Sinecatechins is a standardized extract of green tea leaves, containing a mixture of catechin compounds. The 15% ointment has been approved for the topical treatment of condylomata acuminata (CA), caused by particular human papillomaviruses (HPV), mainly HPV types 6 and 11. Objective: To update the use of sinecatechins for treatment of CA and to describe possible mechanisms involved in the mode of action. Areas covered: The clearance rate of wart lesions after treatment with sinecatechins is similar to other topically applied drugs, like imiquimod and podophyllotoxin, but recurrences are seen less frequently after treatment with sinecatechins. The molecular mode of action underlying the clinical efficacy has yet not been characterized, but the following properties are likely to be involved in the regression of genital warts: activation of cellular immune reactions, induction of cell cycle arrest and apoptosis, inhibition of HPV gene expression. Expert opinion: Sinecatechins is a safe and effective treatment option for CA. Due to the variety of molecular activities of catechins the product has great potential for application to other viral and tumor lesions in the future.

Abstract:By using the technique of optical second-harmonic generation, we have succeeded in obtaining the orientation, spectrum, and the change in dipole moment upon optical excitation for monolayers of retinal chromophores, which are of relevance for visual excitation. The change in dipole moment of the chromophore embedded in the purple membrane of Halobacterium halobium is found to be identical to that of the free chromophore. The spectrum of all-trans retinal mono layers exhibits a subsidiary one-photon forbidden peak in addition to the main absorption peak.

Abstract:Picosecond transient circular dichroism spectra are reported for the primary intermediates in the photocycle of reaction centers isolated from Rhodobacter sphaeroides. The time-resolved circular dichroism spectra of the two electron transfer intermediates (BChl2)+BPh(L)-Q(A) and (BChl2)+BPh(L)Q(A)- reveal a large, nonconservative, and fairly stationary CD band at 800 nm. These results suggest that mechanisms other than exciton interactions need to be included in order to explain the optical activity of this biological system.

Abstract:Abstract Photoisomerization of retinoids inside a confined protein pocket represents a critical chemical event in many important biological processes from animal vision, nonvisual light effects, to bacterial light sensing and harvesting. Light-driven proton pumping in bacteriorhodopsin entails exquisite electronic and conformational reconfigurations during its photocycle. However, it has been a major challenge to delineate transient molecular events preceding and following the photoisomerization of the retinal from noisy electron density maps when varying populations of intermediates coexist and evolve as a function of time. Here, I report several distinct early photoproducts deconvoluted from the recently observed mixtures in time-resolved serial crystallography. This deconvolution substantially improves the quality of the electron density maps, hence demonstrates that the all-trans retinal undergoes extensive isomerization sampling before it proceeds to the productive 13-cis configuration. Upon light absorption, the chromophore attempts to perform trans-to-cis isomerization at every double bond together with the stalled anti-to-syn rotations at multiple single bonds along its polyene chain. Such isomerization sampling pushes all seven transmembrane helices to bend outward, resulting in a transient expansion of the retinal binding pocket, and later, a contraction due to recoiling. These ultrafast responses observed at the atomic resolution support that the productive photoreaction in bacteriorhodopsin is initiated by light-induced charge separation in the prosthetic chromophore yet governed by stereoselectivity of its protein pocket. The method of a numerical resolution of concurrent events from mixed observations is also generally applicable.

Picosecond Absorption and Circular-Dichroism Studies of Proteins

Abstract:Picosecond absorption and circular dichroism (CD) studies of photosynthetic reaction centers (RC) from Rhodabacter Sphaeroides are reported. The transient absorption spectra obtained are consistent with previous studies and provide definitive evidence for the formation of the two charge transfer intermediates: (BChl2)+BPhL-QA and (BChl2)+BPhLQA+ with rate constants whose half lives are <10 picosecond and $OM200 picoseconds, respectively. The transient circular dichroism spectra of these two charge separated intermediates are characterized by a nonconservative CD band centered at 800 nm with similar shape and intensity. This result clearly demonstrates that the negative CD band at 814 nm arises from the exciton coupling of the bacteriochlorophyll dimer. This transient CD cannot be due to excitonic interactions between the pigments in the reaction center and suggest that the monomer bacteriochlorophyll is either intrinsically chiral through distortions induced by the surrounding protein structure or obtains rotational strength from coupled oscillator interactions with surrounding aromatic residues in the protein.
Ab initio simulation of the ultrafast circular dichroism spectrum of provitamin D ring-opening

Abstract:We present a method to simulate ultrafast pump-probe time-resolved circular dichroism (TRCD) spectra based on time-dependent density functional theory trajectory surface hopping. The method is applied to simulate the TRCD spectrum along the photoinduced ring-opening of provitamin D. Simulations reveal that the initial decay of the signal is due to excited state relaxation, forming the rotationally flexible previtamin D. We further show that oscillations in the experimental TRCD spectrum arise from isomerizations between previtamin D rotamers with different chirality, which are associated with the helical conformation of the triene unit. We give a detailed description of the formation dynamics of different rotamers, playing a key role in the natural regulation vitamin D photosynthesis. Going beyond the sole extraction of decay rates, simulations greatly increase the amount of information that can be retrieved from ultrafast TRCD, making it a sensitive tool to unravel details in the sub-picosecond dynamics of photoinduced chirality changes.

Abstract:In this study, the electronic transport properties of 11- Cis and Trans retinal, components of rhodopsin, were investigated as optical molecular switches using the nonequilibrium Green's function (NEGF) formalism combined with first-principles density functional theory (DFT). These isomers, which can be reversibly converted into each other, were examined in detail. The structural and spectroscopic properties, including infrared (IR), Raman, nuclear magnetic resonance (NMR), and ultraviolet (UV) spectra, were analyzed using the hybrid B3LYP/6–311 + + G** level of theory. Complete vibrational assignments were performed for both forms utilizing the scaled quantum mechanical force field (SQMFF) methodology. To evaluate the conductivity of these molecules, we utilized current–voltage (I-V) characteristics, transmission spectra, molecular projected self-consistent Hamiltonian (MPSH), HOMO–LUMO gap, and second-order interaction energies (E 2 ). The trendline extrapolation of the current–voltage plots confirmed our findings. We investigated the effect of different electrodes (Ag, Au, Pt) and various connection sites (hollow, top, bridge) on conductivity. The Ag electrode with the hollow site exhibited the highest efficiency. Our results indicate that the Cis form has higher conductivity than the Trans form.

Quantum Yields from Stationary States: Cis-Trans Isomerization of Model Retinal

Abstract:Cis-trans isomerization in retinal, the first step in vision, is often computationally studied from a time dependent viewpoint. Motivation for such studies lies in coherent pulsed laser experiments that explore the isomerization dynamics. However, such biological processes take place naturally in the presence of incoherent light, which excites a non-evolving mixture of stationary states. Here the isomerization problem is considered from the latter viewpoint and applied to a standard two-state, two-mode linear vibronic coupling model of retinal that explicitly includes a conical intersection between the ground and first excited electronic states. The calculated quantum yield at 500 nm agrees well with both the previous time-dependent calculations of Hahn and Stock (0.63) and with experiment ($0.65\pm0.01$), as does its wavelength dependence. Significantly, the effects of environmental relaxation on the quantum yield in this well-established model are found to be negligible. The results make clear the connection of the photoisomerization quantum yield to properties of stationary eigenstates, providing alternate insights into conditions for yield optimization.

Abstract:Isomerization is a key process in many (bio)chemical systems. In microbial rhodopsins, the photoinduced isomerization of the all-trans retinal to the 13-cis isomer initiates a cascade of structural changes of the protein. The interplay between these changes and the thermal relaxation of the isomerized retinal is one of the crucial determinants for rhodopsin functionality. It is therefore important to probe this dynamic interplay with chromophore specific markers that combine gapless temporal observation with spectral sensitivity. Here we utilize the near-UV and mid-IR fingerprint region in the framework of a systematic (time-resolved) spectroscopic study on H+- (HsBR, (G)PR), Na+- (KR2, ErNaR) and Cl-- (NmClR) pumps. We demonstrate that the near-UV region is an excellent probe for retinal configuration and - being sensitive to the electrostatic environment of retinal - even transient ion binding, which allows us to pinpoint protein specific mechanistic nuances and chromophore-charge interactions. The combination of the near-UV and mid-IR fingerprint region hence provides a spectroscopic analysis tool that allows a detailed, precise and temporally fully resolved description of retinal configurations during all stages of the photocycle.

Abstract:The sequential derivation of an expression that describes an X-ray natural circular dichroism (XNCD) signal in the most general case is performed. The obtained expression is used as a basis to consider XNCD signals and their structural origins in copper metaborate CuB2O4 crystals at an incident radiation energy near the K absorption edge of copper and lanthanum–gallium silicate La3Ga5SiO14 crystals at an incident radiation energy near the K absorption edge of gallium. It has been demonstrated that the measurements of circularly polarized X-ray absorption spectra in combination with corresponding first-principle calculations provide an efficient method for studying the contributions of crystallographically non-equivalent atoms to the X-ray optical activity of crystals.

Abstract:The synthesis and characterization of a subphthalocyanine-bisazobenzene-subphthalocyanine triad are reported. Evidence for E,E <-> E,Z isomerization of the linking bisazobenzene moiety is observed in the optical and NMR spectra when the subphthalocyanine rings are used as light-harvesting units. Electronic coupling and communication between the two para-azo units decrease the photoisomerization efficiency. An obvious decrease in fluorescence intensity is observed on moving from E -> Z with a recovery in intensity observed on moving back from Z -> E that can be attributed to a change in the rate of non-radiative decay.

Abstract:In this paper, planar chiral helicenes based on [2.2]paracyclophane PCPH5 acted as both chiral inducers and energy donors to construct the circularly polarized luminescence (CPL)-active ternary cholesteric liquid crystals (T-N*-LCs) in a commercial nematic liquid crystal (SLC1717, N-LCs) medium. Induced red CPL emission based on the energy acceptor of the achiral polymer DTBTF8 could be successfully promoted by the intermolecular Förster resonance energy transfer mechanism. The resulting T-N*-LCs can generate intensive CPL signals with glum up to +0.70/-0.67. Interestingly, this kind of "on-off" CPL switching of T-N*-LCs can be manipulated by the applied direct current electric field.

Abstract:Using a dual-beam flow technique, we have obtained resonance Raman spectra of the M412 photointermediates of both native purple membrane (15H M412) and purple membrane regenerated with 15-deuterioretinal (15D M412). For comparison, we have also obtained Raman spectra of the n-butylamine Schiff bases of the 13-cis and all-trans isomers of 15H and 15D retinal. The 15D model compound spectra, when compared to the 15H spectra, show isotopically induced spectral changes that are markedly different for the two isomers. There is a very close agreement between the frequency and intensity changes which occur upon deuteration of M412 and those which occur upon deuteration of the 13-cis model compound, but not even a qualitative correspondence exists when M412 and the all-trans model compound are similarly compared. These data demonstrate that the chromophore of M412 is an unprotonated Schiff base of 13-cis-retinal rather than all-trans-retinal. An analogous spectral comparison of 15H and 15D light-adapted bacteriorhodopsin (bRLA) with the 15H and 15D protonated Schiff bases of 13-cis- and all-trans-retinal demonstrates that bRLA contains an all-trans chromophore, in agreement with previous extraction experiments. Thus, a trans leads to cis isomerization occurs in the proton-pumping photocycle of Halobacterium halobium.

Abstract:We observed an orientational photo-refractive effect in semiconductor CdSe/CdS nanorods-doped nematic liquid crystal cells homeotropically aligned. In order to study the photo-refractive and the orientational responses of this cell, we employed both asymmetric two-beam coupling (TBC) to measure two beam coupling gain under the influence of an applied electric field and forced light scattering technique. An optical index grating was written in the device in a holographic tilted configuration by overlapping two laser beams at 514 nm. Self diffraction of the writing beams and diffraction of a probe beam at 633 nm were measured. A high TBC gain coefficient of 270 cm -1 was achieved at a DC field applied of 3.4 V/μm. First-order diffraction efficiencies were evaluated with a maximum efficiency of 7% in the dynamic grating with a nonlinear refractive index, n 2 , 4 × 10 -2 cm 2 /W for E 0 = 3 V /μ m .
