Abstract:Memory formation is usually associated with Hebbian learning, using synaptic plasticity to change the synaptic strengths but omitting structural changes. Recent work suggests that structural plasticity can also lead to silent memory engrams, reproducing a conditioned learning paradigm with neuron ensembles. However, this work is limited by its way of synapse formation, enabling the formation of only one memory engram. Overcoming this, our model allows the formation of many engrams simultaneously while retaining high neurophysiological accuracy, e.g., as found in cortical columns. We achieve this by substituting the random synapse formation with the Model of Structural Plasticity ( ). As a homeostatic model, neurons regulate their activity by growing and pruning synaptic elements based on their current activity. Utilizing synapse formation based on the Euclidean distance between the neurons with a scalable algorithm allows us to easily simulate 4 million neurons with 343 memory engrams. These engrams do not interfere with one another by default, yet we can change the simulation parameters to form long-reaching associations. Our model paves the way for simulations addressing further inquiries, ranging from memory chains and hierarchies to complex memory systems comprising areas with different learning mechanisms.

Abstract:The hippocampus plays a critical role in the rapid learning of new episodic memories. Many computational models propose that the hippocampus is an autoassociator that relies on Hebbian learning (i.e., "cells that fire together, wire together"). However, Hebbian learning is computationally suboptimal as it does not learn in a way that is driven toward, and limited by, the objective of achieving effective retrieval. Thus, Hebbian learning results in more interference and a lower overall capacity. Our previous computational models have utilized a powerful, biologically plausible form of error-driven learning in hippocampal CA1 and entorhinal cortex (EC) (functioning as a sparse autoencoder) by contrasting local activity states at different phases in the theta cycle. Based on specific neural data and a recent abstract computational model, we propose a new model called Theremin (Total Hippocampal ERror MINimization) that extends error-driven learning to area CA3-the mnemonic heart of the hippocampal system. In the model, CA3 responds to the EC monosynaptic input prior to the EC disynaptic input through dentate gyrus (DG), giving rise to a temporal difference between these two activation states, which drives error-driven learning in the EC→CA3 and CA3↔CA3 projections. In effect, DG serves as a teacher to CA3, correcting its patterns into more pattern-separated ones, thereby reducing interference. Results showed that Theremin, compared with our original Hebbian-based model, has significantly increased capacity and learning speed. The model makes several novel predictions that can be tested in future studies.

Abstract:Memory is a complex process across different brain regions and a fundamental function for many cognitive behaviors. Emerging experimental results suggest that memories are represented by populations of neurons and organized in a categorical and hierarchical manner. However, it is still not clear how the neural mechanisms are emulated in computational models. In this paper, we present a spatio-temporal memory (STM) model using spiking neurons to explore the memory formulation and organization in the brain. Unlike previous approaches, this model employs temporal population codes as the neural representation of information and spike-timing-based learning methods to formulate the memory structure. It explicitly demonstrates that the complex spatio-temporal patterns are the internal neural representations of memory items. Two types of memory processes are analyzed and emulated: associative memory, i.e., spatio-temporal patterns driven by intra-assembly connections, and episodic memory, i.e., temporally separated spatio-temporal patterns linked by inter-assembly connections. Our model will provide a computational substrate based on low-level neural circuits for developing neuromorphic cognitive systems with wide applications.

Abstract:Memory consolidation involves a process of engram reorganization and stabilization that is thought to occur primarily during sleep through a combination of neural replay, homeostatic plasticity, synaptic maturation, and pruning. From a computational perspective, however, this process remains puzzling, as it is unclear how the underlying mechanisms can be incorporated into a common mathematical model of learning and memory. Here, we propose a solution by deriving a consolidation model that uses replay and two-factor synapses to store memories in recurrent neural networks with sparse connectivity and maximal noise robustness. The model offers a unified account of experimental observations of consolidation, such as multiplicative homeostatic scaling, task-driven synaptic pruning, increased neural stimulus selectivity, and preferential strengthening of weak memories. The model further predicts that intrinsic synaptic noise scales sublinearly with synaptic strength; this is supported by a meta-analysis of published synaptic imaging datasets.

Abstract:Memory engrams are formed through experience-dependent remodeling of neural circuits, but their detailed architectures have remained unresolved. Using 3D electron microscopy, we performed nanoscale reconstructions of the hippocampal CA3-CA1 pathway following chemogenetic labeling of cellular ensembles with a remote history of correlated excitation during associative learning. Projection neurons involved in memory acquisition expanded their connectomes via multi-synaptic boutons without altering the numbers and spatial arrangements of individual axonal terminals and dendritic spines. This expansion was driven by presynaptic activity elicited by specific negative valence stimuli, regardless of the co-activation state of postsynaptic partners. The rewiring of initial ensembles representing an engram coincided with local, input-specific changes in the shapes and organelle composition of glutamatergic synapses, reflecting their weights and potential for further modifications. Our findings challenge the view that the connectivity among neuronal substrates of memory traces is governed by Hebbian mechanisms, and offer a structural basis for representational drifts.

Abstract:Empirical evidence shows that memories that are frequently revisited are easy to recall, and that familiar items involve larger hippocampal representations than less familiar ones. In line with these observations, here we develop a modelling approach to provide a mechanistic understanding of how hippocampal neural assemblies evolve differently, depending on the frequency of presentation of the stimuli. For this, we added an online Hebbian learning rule, background firing activity, neural adaptation and heterosynaptic plasticity to a rate attractor network model, thus creating dynamic memory representations that can persist, increase or fade according to the frequency of presentation of the corresponding memory patterns. Specifically, we show that a dynamic interplay between Hebbian learning and background firing activity can explain the relationship between the memory assembly sizes and their frequency of stimulation. Frequently stimulated assemblies increase their size independently from each other (i.e. creating orthogonal representations that do not share neurons, thus avoiding interference). Importantly, connections between neurons of assemblies that are not further stimulated become labile so that these neurons can be recruited by other assemblies, providing a neuronal mechanism of forgetting.
