Abstract:Abstract Trimethylamine N-oxide (TMAO), a microbiota-derived metabolite has been implicated in human health and disease. Its early detection in body fluids has been presumed to be significant in understanding the pathogenesis and treatment of many diseases. Hence, the development of reliable and rapid technologies for TMAO detection may augment our understanding of pathogenesis and diagnosis of diseases that TMAO has implicated. The present work is the first report on the development of a molecularly imprinted polymer (MIP) based electrochemical sensor for sensitive and selective detection of TMAO in body fluids. The MIP developed was based on the polypyrrole (PPy), which was synthesized via chemical oxidation polymerization method, with and without the presence of TMAO. The MIP, NIP and the non-sonicated polymer (PPy-TMAO) were separately deposited electrophoretically onto the hydrolyzed indium tin oxide (ITO) coated glasses. The chemical, morphological, and electrochemical behavior of MIP, non-imprinted polymer (NIP), and PPy-TMAO were characterized using Fourier transform infrared spectroscopy (FT-IR), scanning electron microscopy (SEM), and electrochemical techniques. The detection response was recorded using differential pulse voltammetry (DPV), which revealed a decrease in the peak current with the increase in concentration of TMAO. The MIP sensor showed a dynamic detection range of 1–15 ppm with a sensitivity of 2.47 μA mL ppm −1 cm −2 . The developed sensor is easy to construct and operate and is also highly selective to detect TMAO in body fluids such as urine. The present research provides a basis for innovative strategies to develop sensors based on MIP to detect other metabolites derived from gut microbiota that are implicated in human health and diseases.

Abstract:A rapid, sensitive, low-cost device to detect trimethylamine was presented in this paper. The preparation of water soluble polyaniline was firstly studied. Then the polyaniline was characterized via Fourier transform infrared spectroscopy (FTIR), UV-visible spectroscopy and scanning electron microscopy (SEM). Based on the water soluble polyaniline film, a quartz crystal microbalance (QCM) sensor for trimethylamine detection was fabricated and its characteristics were examined. The sensor consisted of one quartz crystal oscillator coated with the polyaniline film for sensing and the other one for reference. Pretreated with trimethylamine, the QCM sensor had an excellent linear sensitivity to trimethylamine. Easily recovered by N2 purgation, the response of the sensor exhibited a good repeatability. Responses of the sensor to trimethylamine, ethanol and ethyl acetate were compared, and the results showed that the response was related to the polarity of the analyte vapor. Experimental result also showed that the sensitivity of the sensor was relatively stable within one month. The simple and feasible method to prepare and coat the polyaniline sensing film makes it promising for mass production.

Abstract:Trimethylamine is a good target for the assay of meat freshness,and also of clinical interest.Therefore,detection of trimethylamine is very meaningful.For this reason,we prepared polypyrrole by emulsion polymerization method,and coated it onto a quartz crystal which was used as the sensing QCM.The change of frequency difference between sensing QCM and reference QCM was used to monitor the atmosphere.When the sensor was exposed to trimethylamine vapor at room temperature,it responded rapidly and obviously.The sensor was characterized in its sensitivity,repeatability,selectivity and long-term stability.The results showed that the performance of sensor was excellent,though the method to prepare it was simple.

Abstract:Catecholamines are neurotransmitters in the peripheral and central nervous systems, which maintain the normal physiological state of the human body. Studies have shown that the occurrence of many diseases are often accompanied by changes in catecholamine levels. Clinically, the detection of catecholamine levels in human plasma or urine can assist in the diagnosis of endocrine-related diseases and neurological diseases. However, catecholamines are easily oxidized, and the content in human body is very low. So the rapid detection of catecholamines is very important for the prevention and diagnosis of related diseases. In this paper, the electrochemical response characteristics of Ti(3)C(2)Tx MXene and noble metal nanoparticles modified Ti(3)C(2)Tx to dopamine (one type of catecholamine) were explored. The results shown that Ti(3)C(2)Tx would undergo irreversible oxidation at the anode potential and the single Ti(3)C(2)Tx modified electrode was not suitable for the detection of dopamine. So we further explored the modification of Pt and Au nanoparticles on the Ti(3)C(2)Tx/GCE electrode, and the experimental results shown that Au/Ti(3)C(2)Tx/GCE had higher sensitivity for measuring dopamine, while Pt/Ti(3)C(2)Tx/GCE had a larger linear range. Both electrodes had good stability and repeatability and could be used for the detection of catecholamines.
MAGNETIC RESONANCE IMAGING OF POROSITY HETEROGENEITY IN BIOTURBATED SANDSTONE FROM THE WHITE ROSE RESERVOIR, ATLANTIC CANADA

Abstract:The determination of porosity heterogeneity within hydrocarbon reservoirs is important because cost-effective, efficient production of hydrocarbons depends on accurate knowledge of reserve estimates, and hydrocarbon distribution. A new magnetic resonance imaging (MRI) technique, based on the conical SPRITE (Single-Point Ramped Imaging with T1 Enhancement) MRI method is capable of revealing the three-dimensional geometry of porosity distribution in core samples taken from reservoir rocks. Porosity heterogeneity at the core scale is difficult to assess by conventional petrophysical methods. Conventional MRI methods also fail because of complications associated with the multi-exponential nature of spin-spin relaxation time decay and restrictions on the minimal echo time. The SPRITE MRI technique allows direct quantitative measurements of porosity and imaging of samples with short MR signal lifetimes. We illustrate the application of this method to two core samples from the White Rose reservoir: one containing shell fragments and the other fine grained and massive in texture. The data presented in this paper show the potential of the conical SPRITE MRI technique as a tool for porosity heterogeneity evaluation by application of the method to core plugs taken from sandstone reservoir rocks of the White Rose oilfield, located in offshore Newfoundland. The results show that the MRI technique can significantly enhance petrophysical studies on reservoir rocks by providing: 1) direct and non-destructive visualization of three-dimensional data required to model the porous network; 2) spatially resolved porosity throughout the core sample, which allow reveals the porosity heterogeneity and will result in better understanding of reservoir quality.
Polyaniline–TiO2 Nano-Composite-based Trimethylamine QCM Sensor and Its Thermal Behavior Studies

Abstract:Inorganic/organic composites are very attractive due to synergetic behavior and a wide range of potential use. A polyaniline–TiO2 nano-composite, obtained by combination of chemical polymerization and a sol–gel method, was deposited on the electrode of quartz crystal to implement a quartz crystal microbalance (QCM) chemical sensor. The morphology of the composite film was studied by scanning electron microscopy (SEM) measurements. The coated quartz crystal and a non-coated quartz crystal were mounted in a sealed chamber, and their frequency difference was monitored. When analyte vapor was injected into the chamber, gas absorption decreased the frequency of the coated quartz crystal and thereby caused an increase of the frequency difference between the two crystals. The frequency difference change response towards trimethylamine was evident and could be recovered by N2 purgation easily. The calibration curve towards trimethylamine, its long-term stability and selectivity were investigated. The thermal behavior of the sensing characteristics was compared with that of a polyaniline QCM sensor. Fourier transform infrared (FTIR) spectra of polyaniline and polyaniline–TiO2 nano-composite and QCM data under various conditions were used to study the effect of thermal treatment.

Abstract:In this study, we rationally combined target-induced nanoprobe deactivation with a static headspace microextraction strategy for highly selective and sensitive colorimetric/SERS dual-mode sensing of trimethylamine N-oxide (TMAO). Prussian blue–polyacrylic acid–gold (PB-PAA-Au) nanohybrids were elaborately constructed as excellent peroxidase-like and SERS-active nanoprobes owing to the synergistic effect between PB and Au nanoparticles, which, however, could be deactivated by trimethylamine (TMA), the reduction product of TMAO. Accordingly, the nonvolatile TMAO in sample solution was catalytically reduced to volatile TMA, which could be headspace extracted and in turn cause the deactivation of PB-PAA-Au in the extracting droplet. The deactivation degree was totally dependent on TMA concentration and could be indicated by the catalytic efficiency of PB-PAA-Au toward chromogenic reaction of H2O2–3,3′,5,5′-tetramethylbenzidine, as well as the SERS intensity of PB-PAA-Au. Thus, the quantitative determination of TMAO could be realized by tracking the changes in colorimetric and SERS signals. Under the optimal conditions, the proposed sensing strategy exhibited a limit of detection down to 1.57 μM in colorimetric mode and 5.33 μM in SERS mode. Furthermore, the dual-mode system was successfully applied for TMAO detection in real samples with satisfactory outcomes. This work provides an efficient strategy for clinical monitoring of TMAO and holds great promise for further evaluation of the impact of dietary and environmental factors on TMAO levels.

Abstract:One of the gut-derived uremic toxins 4-ethylphenyl sulfate (4-EPS) exhibits significantly elevated plasma levels in chronic kidney diseases and autism, and its early quantification in bodily fluids is important. Therefore, the development of rapid and sensitive technologies for 4-EPS detection is of significant importance for clinical diagnosis. In the current work, the synthesis of a molecularly imprinted biopolymer (MIBP) carrying 4-EPS specific cavities only using the biopolymer polydopamine (PDA) and molybdenum disulfide (MoS2) nanosheets has been reported. The fabricated electrode was prepared using screen-printed carbon electrodes on a polyvinyl chloride substrate. The synthesized material was characterized using several techniques, and electrochemical studies were performed using cyclic voltammetry (CV) and differential pulse voltammetry (DPV) techniques. The DPV technique for the electrochemical sensing of 4-EPS using the fabricated sensor (PDA@MoS2-MIBP) determined a sensitivity of 0.012 μA/ng mL/cm2 and a limit of detection of 30 ng/mL in a broad linear range of 1-2200 ng/mL. Also, the interferent study was performed to evaluate the selectivity of the fabricated sensor along with the control and stability study. Moreover, the performance of the sensor was evaluated in the spiked urine sample, and a comparison was made with the data obtained by ultraperformance liquid chromatography-tandem mass spectroscopy.

Abstract:The phenomenon of angiogenesis-the growth of capillary blood vessels and the development of new blood vesselshas been familiar to morphologists and physiologists since the late 19th century. Most of the early research revolved around tumour neovascularization and was initiated by Goldman who published a key paper in 19071. The pioneering work of Folkman began in the early 1960s and was directed mainly at elucidating the mechanisms of early tumour angiogenesis2. Under normal conditions, angiogenesis is highly regulated and occurs when an imbalance exists between the supply and demand of oxygen and nutrients to a vascular bed, or if inflammation is present. Initially the concept of angiogenesis was resisted, because tumour hypervascularity was believed to result from dilatation of pre-existing host vessels rather than growth and development of vessels de novo. Opinions changed, however, when avascular rabbit corneas were seen to acquire new vessels when implanted with tumours; and these studies eventually led to the isolation and purification of an angiogenic factor from tumour cells3. For a time it was thought that angiogenic initiating factors must be endothelial mitogens. Basic fibroblast growth factor (b-FGF), first purified by Shing and Folkman, is an endothelial mitogen4; but the next angiogenic factor to be discovered, angiogenin, was not5. Whereas Folkman6 and his associates were instrumental in establishing the basic mechanisms of tumour vessel growth and development, it was Schaper who greatly increased our understanding of angiogenesis in the heart and the development of the coronary collateral circulation7. Since the work of these early investigators, the advent of recombinant technology and the development of novel drug delivery and biocompatible systems to deliver peptides have increased the potential for both scientific investigation and therapeutic angiogenesis. The fact that collateral vessels develop and grow during gradual coronary occlusion in animal models and in patients, along with the observation that certain mitogenic growth factors and cytokines can induce angiogenesis, suggests that angiogenic substances could be of value in the treatment of occlusive coronary artery disease. Currently, myocardial ischaemia and its consequences can be treated with coronary artery bypass grafting (CABG), percutaneous transluminal coronary angioplasty (PTCA), and more recently with transmyocardial laser revascularization (TMLR). However, a subset of patients exists who are not candidates for CABG, PTCA or other conventional methods of revascularization. The lack of efficacy and hazards of TMLR leave a void that may be filled by other approaches such as therapeutic angiogenesis. Here we discuss the potential mechanisms and the therapeutic implications.
Investigation of Electrocatalysis for Tiered-Tower Micro-Electro-Mechanical-Systems Biosensors: Applied to Early Prevention of Thrombosis Factor Trimethylamine N-Oxide

Abstract:Trimethylamine N-Oxide (TMAO) metabolism has been recognized as a biomarker for early thrombosis detection. However, testing for TMAO typically requires expensive laboratory equipment and skilled technicians, making it unsuitable as a pre-screening device for home care. To enable widespread use in home applications, it is crucial to develop a scalable and sensitive device capable of catalyzing TMAO with a specific enzyme, tailored for point-of-care use. This study presents an investigation of a MEMS based two tiered-layer tower biosensor array with a detection limit of 0.1 μM for TMAO, aimed at diagnosing chronic metabolic diseases in urine or serum. Based on augmented Cole-Cole model, the proposed parameters R_catalyzed, C_catalyzed, and Rp_catalyzed in the model can predict the catalytic impedance of enzyme activities, such as the redox effects of analytes, and characterize the small signal current caused by catalysis. The proposed MEMS biosensor, integrated with readout circuitry, demonstrates high sensitivity of 41 ADC counts/μM TMAO (or 4.5 mV/μM TMAO), a response time of 1 second, a repetition rate of 98.9%, and a drift over time of 0.5 mV. The sensor effectively discriminates TMAO based on minute capacitance changes induced by the TorA enzyme, resulting in a discernible 10.6% distinction. These measurements have been successfully compared to the conventional cyclic-voltammetry (CV) method, showing a mere 0.024% variance. The proposed biosensor is well-suited for pre-screening thrombosis factors in early prevention for point-of-care applications. The device is cost-effective, lightweight, and demonstrates excellent performance, with an 88% conversion rate of TMAO and a 97% selectivity rate for the by-product TMA, allowing for the prediction of cardiovascular risk in advance.

Abstract:Methamphetamine (MA), a psychoactive substance with many medicinal applications in different countries, has destructive impacts on the nervous system and brain and can lead to addiction. The optimal system for MA determination must be able to measure the tiny amount of MA in complex matrixes accurately. In the current work, a simple and biocompatible sensitive optical probe was developed based on molecularly imprinted polymers (MIPs) technique and by using green CQDs and mesoporous structured imprinting microspheres (SiO2@CQDs@ms-MIPs). CQDs (ФF = 33%) were synthesized via the hydrothermal method using natural chewing gum as carbon source. SiO2 nanoparticles were used as the backup substrate for the placement of CQDs. In spite of biocompatibility, porosity and having high specific area are the unique features of SiO2 nanoparticles. When MA is present, the fluorescence response of MIPs enhances. This is caused by the passivation and adjustment of active clusters that are present on the surface of CQDs. By this optical sensor, the favorable linear dynamic range (5.0-250 μM) and the detection limit (1.6 μM) were obtained. The applicability of the advanced sensor was studied in real samples such as human urine and human blood plasma. Acceptable results were obtained and recovery amounts were in the 92-110% interval.
Atovaquone: A Valuable Therapeutic Option in Toxoplasma Retinochoroiditis

Abstract:Background: Toxoplasma gondii is a leading cause of severe foodborne illness in first world countries and the most common cause of posterior uveitis. Neither a standard treatment protocol has been established so far, nor has the efficacy of the mostly applied medications and their combinations be proven in well-designed clinical trials. Atovaquone, a hydroxy-1,4-naphtoquinone, is a safe and effective treatment against tachyzoites and cyst forms of Toxoplasma, but it is only FDA and EMEA approved for the treatment of pneumocystis infections. We have started using Atovaquone in ocular toxoplasmosis on a regular, off-label basis in 1996. Methods: We report about the use of Atovaquone 750 mg twice daily in punctate outer retinal toxoplamosis as well as in Toxoplasma retinochoroiditis resistant to four different antibiotics. Results: Toxoplasma activity stopped within three weeks of Atovaquone therapy without any side-effects. Conclusions: In our hands the three-week Atovaquone treatment is a safe and efficient therapeutic option in ocular toxoplasmosis. Placebo-controlled randomized trials of anti-Toxoplasma treatment in patients presenting with Toxoplasma retinochoroiditis are required. These trials should include Atovaquone.

Abstract:The combination of two-dimensional material MXene and one-dimensional metal oxide semiconductor can improve the carrier transmission rate, which can effectively improve sensing performance. We prepared a trimethylamine gas sensor based on MoO3 nanofibers and layered Ti3C2Tx MXene. Using electrospinning and chemical etching methods, one-dimensional MoO3 nanofibers and two-dimensional Ti3C2Tx MXene nanosheets were prepared, respectively, and the composites were characterized via XPS, SEM, and TEM. The Ti3C2Tx MXene–MoO3 composite material exhibits excellent room-temperature response characteristics to trimethylamine gas, showing high response (up to four for 2 ppm trimethylamine gas) and rapid response–recovery time (10 s/7 s). Further, we have studied the possible sensitivity mechanism of the sensor. The Ti3C2Tx MXene–MoO3 composite material has a larger specific surface area and more abundant active sites, combined with p–n heterojunction, which effectively improves the sensitivity of the sensor. Because of its low detection limit and high stability, it has the potential to be applied in the detection system of trimethylamine as a biomarker in exhaled air.

Abstract:The gut microbiota plays a critical role in chronic kidney disease (CKD) and hypertension. Trimethylamine-N-oxide (TMAO) and trimethylamine (TMA) are gut microbiota-derived metabolites, and both are known uraemic toxins that are implicated in CKD, atherosclerosis, colorectal cancer and cardiovascular risk. Therefore, the detection and quantification of TMAO, which is a metabolite from gut microbes, are important for the diagnosis of diseases such as atherosclerosis, thrombosis and colorectal cancer. In this study, a new “colour-switch” method that is based on the combination of a plasma separation pad/absorption pad and polyallylamine hydrochloride-capped manganese dioxide (PAH@MnO2) nanozyme was developed for the direct quantitative detection of TMAO in whole blood without blood sample pretreatment. As a proof of concept, a limit of quantitation (LOQ) of less than 6.7 μM for TMAO was obtained with a wide linear quantification range from 15.6 to 500 μM through quantitative analysis, thereby suggesting potential clinical applications in blood TMAO monitoring for CKD patients.
An Off-Chip Capacitance Biosensor Based on Improved Cole-Cole Model for the Detection of Trimethylamine N-Oxide in Early Cardiovascular Disease

Abstract:In this work, we demonstrated an off-chip capacitance array biosensor with limit of detection 1 μM of TMAO to diagnose chronic metabolism disease in urine. Based on the improved cole-cole model, the parameters of R catalyzed and C catalyzed and Rp catalyzed can predict the catalytic resistance of enzyme and redox effects of the analyte, and identify the AC properties of ion strength due to enzymatic catalysis. With the investigation of the off-chip capacitance biosensor electrical field change with respect to pixel geometry and associated driving and sensing electrode width in a pixel, the proposed off-chip biosensor with readout SoC exhibits high sensitivity of 21 ADC counts/μM (or 25 mV/μM), response time of 0.1 s, repetition of 98.9%, and drift over time of 0.5 mV. The proposed off-chip biosensor can distinguish TMAO in phosphate buffered saline (PBS) solution from the tiny capacitance change of TMAO induced by enzyme TorA with a difference of 2.15%, and the measurements has been successful verified with traditional cyclic-voltammetry (CV) method, which shows a difference of only 0.024%. The off-chip biosensor is designed to specifically detect TMAO by excluding the redox reactions of TMAO-specific enzyme TorA with creatine and creatinine in urine. This exclusion ensures that the biosensor focuses solely on TMAO, enhancing its specificity for accurate and reliable TMAO detection.

Abstract:Pharmaceutical and personal care products are emerging as a new category of environmental pollution. Analytical drug detection from a biological sample for detection is still crucial today. Mefenamic acid (MA) is an anti-inflammatory drug utilized for its antipyretic and analgesic properties, which is harmful to patients at higher dosages and is also recognized as a chemical pollutant that harms the environment. In this view, Dysprosium manganite/carbon nanofiber (DMO/CNF) was prepared by hydrothermal method for the electrochemical detection of MA. DMO/CNF/GCE exhibits high selectivity, excellent anti-interference, good stability, and reproducibility toward the detection of MA. The enhanced electrochemical performance of DMO/CNF/GCE was attributed to their synergetic interaction. Under optimized conditions, DMO/CNF/GCE shows a wide linear range of 0.01–741 μM and a low LOD of 0.009 μM. Satisfactory recoveries were obtained for human blood and tablet samples. Thus, the proposed DMO/CNF nanocomposite emerges as a promising material for the detection of MA.

Abstract:A novel electrochemical sensor using the molecularly imprinted (MIP) oxygen-containing polypyrrole (PPy) decorated carbon nanotubes (CNTs) composite was proposed for in vivo detection of dopamine (DA). The prepared sensor exhibits a remarkable sensitivity of (16.18μA/μM) with a linear range of 5.0×10−11–5.0×10−6M and limit of detection as low as 1.0×10−11M in the detection of DA, which might be due to the plenty cavities for binding DA through π–π stacking between aromatic rings and hydrogen bonds between amino groups of DA and oxygen-containing groups of the novel PPy.

Abstract:Polypyrrole composite film was formed on the interdigital carbon electrodes by in-situ polymerization to make a gas sensor. The electrical response to trimethylamine was investigated. The results indicate that the organic film has high sensitivity and rapid response to trimethylamine. The gas-sensitivity of polypyrrole composite film to trimethylamine of 5.14×10-7mol/mL, which is up to 3-6 orders of magnitude within 1 min, is much higher than those of phthalocyanine, porphyin, perylene and their fluorination derivatives. The sensor exhibits a good reproducibility as well as a good selectivity to analogy vapors while the film can be completely recovered with high-purity N2 at room temperature.

Abstract:In this study, a new dual-signal electrochemical sensor using a glassy carbon electrode (GCE) modified with molecularly imprinted polyaniline/polythionine/gold imidazolate framework-67 composite (MIP/pTH/) is fabricated for the quantitative analysis of tyrosine (Tyr). is used as a matrix to improve the surface area and electron-transfer ability of the sensor. The pTH as a built-in probe provides a signal-off response, meanwhile the oxide current of Tyr acts as a signal-on response. The MIP is prepared in the presence of aniline monomer and Tyr template by electro-polymerization for Tyr specific recognition. Under the optimized conditions, the sensor demonstrates a linear range from 1 × 10−8 M to 4 × 10−6 M for Tyr determination and a limit of detection (LOD) of 7.9 × 10−10 M (S/N = 3). Furthermore, the sensor exhibits excellent selectivity, superior stability and good reproducibility toward Tyr. More importantly, the sensor is successfully employed to detect Tyr in human serum with satisfactory recoveries.
