Abstract:This paper proposed a multiple headspace single-drop microextraction (MHS-SDME) method coupled to gas chromatography with flame-ionization detection (GC-FID) for direct determination of residual solvents in solid drug product. The MHS-SDME technique is based on extrapolation to an exhaustive extraction of consecutive extractions from the same sample which eliminates the matrix effect on the quantitative analysis of solid samples. The total peak area of analyte is calculated with a beta constant which can be obtained from the slope of the linear regression that related to the peak area of each extraction and the number of extraction times. In this work, a model drug powder was chosen and the amounts of residues of two solvents, methanol and ethanol, were investigated. The factors influencing the extraction process including extraction solvent, microdrop volume, extraction time, sample amount, thermostatting temperature and incubation time were studied. 10mg of drug powder was incubated for 3h at 140°C prior to the first extraction and thermostatted for 15min at 140°C between each extraction. Extraction was carried out with 2μL of dimethyl sulfoxide (DMSO) as the microdrop for 5min. The features of the method were established using standard solutions. Validation of the proposed method showed good agreement with the traditional dissolution method for analysis of residual solvents in drug product. The results indicated that MHS-SDME has a great potential for the quantitative determination of residual solvents directly from the solid drug products due to its low cost, ease of operation, sensitivity, reliability and environmental protection.

Abstract:The expansion of industrial demand for lignocellulosic feedstocks increases the urgency for sustainable and rapid characterization of these raw materials, whose response could infer the suitability of all kinds of plants and agro-industrial residues for biorefinery processes. Thus, we developed an alternative composition and crystallinity analysis method via deconvolution of differential thermogravimetry (DTG) curves, which quantifies water and volatiles, extractives, hemicellulose, amorphous cellulose, crystalline cellulose allomorphs (I and II) and lignin fractions, both in natural lignocellulose and treated holocelluloses. The statistical assessment proved its high accuracy (R 2 > 0.9993, F > 514,345 and SE < 0.1406) via analysis of variance (ANOVA), and the results for figures of merit showed its high precision (SD < 1.58, MSE < 2.13, MAE < 1.37 and ASRE < 5.45 × 10 –3 ) in a 95% confidence level. Data acquisition is significantly faster than other standard methodologies, such as the analytical procedure NREL/TP-510-42618 created by the National Renewable Energy Laboratory. The alternative method can be performed in 1 day, requires less expensive apparatus and low energy consumption, in addition to producing almost no residue. In addition, it can be employed for an all-in-one evaluation (for composition, crystallinity, polymorphism, treatment efficiency and reactivity) of all kinds of lignocellulosic biomass, either raw or treated, with focus on enhancing biomass research and biorefinery processes.

Abstract:Two-dimensional gas chromatography (GC × GC) and two-dimensional liquid chromatography (LC × LC) are nowadays widely used in academia and industry due to their high separation power. However, as far as we know, the complementarity of these two techniques has not yet been thoroughly studied based on the analysis of the same sample. Therefore, this was undertaken here by analysing the liquid fraction obtained after depolymerising a natural waste - lignin - with GC × GC and off-line comprehensive LC × SFC (SFC: supercritical fluid chromatography). Using complementary techniques is also important for lignin valorisation, as thorough structural characterisation of the depolymerised product can aid with developing and improving valorisation processes. For the tentative identification, NIST library was used for GC × GC-MS results and MS-DIAL together with SIRIUS for LC × SFC-MS/MS data. This allowed to study which compounds are detectable with the different 2D methods but also to discuss the limitations of the data analysis processes. The previous knowledge that LC techniques are more suitable than GC × GC for the analysis of larger oligomers and other low volatility compounds was confirmed; however, it was seen that GC × GC enabled the analysis of smaller compounds, such as aliphatic alcohols and saturated compounds. Overall, the study demonstrates the complementarity of the two techniques but also draws attention to the different detectable compound groups and classifications that the two techniques can provide.

Abstract:In this work, thermal desorption-gas chromatography-flame ionization detection (TD-GC-FID) was adapted to enable the determination of residual solvents (RS) in mesoporous silica (MPSi). MPSi is often utilized in various pharmaceutical formulations or drug delivery systems and the accurate determination of RS is an important part of pharmaceutical quality control. Seven commonly used solvents (methanol, ethanol, acetone, isopropanol, dichloromethane, tetrahydrofuran and hexafluoroisopropanol) were evaluated in combination with 3 types of MPSi having pore sizes of 2-3, 15 and 25nm. Validation results showed general recovery values >98% and good linearity over the concentration ranges studied. The limits of detection (LOD) and limits of quantification (LOQ) for the different solvents ranged from 0.03 to 0.08μg and from 0.1 to 0.2μg per tube, respectively. Verification of the accuracy of the TD method was investigated by using an alternative method based on complete dissolution of MPSi in hydrofluoric acid (HF) followed by full evaporation headspace-GC (HS-GC). The results obtained from both procedures were not statistically different (p>0.05) when applied to actual experimental drug samples consisting of itraconazole loaded on MPSi.

Abstract:Owing to a supposed quantitative transformation, oximation of dialdehyde cellulose (DAC) with hydroxylamine hydrochloride is commonly employed in chemical DAC analysis, e.g., for the determination of the degree of oxidation (DO) by titration or elemental analysis. In this study, this modification was utilized for the indirect determination of molecular weight distributions (MWD) by gel permeation chromatography (GPC). The presumably quantitative conversion of aldehyde groups in DAC to the corresponding oxime also breaks up the intermolecular and intramolecular hemiacetal crosslinks, which were associated with solubility issues in the DMAc/LiCl solvent system in previous studies. The limits of the procedure and the material's stability during oximation were investigated. For samples with a DO up to approximately 9% a good applicability was observed, before at higher DO values residual crosslinks led to solubility problems. The oximation/GPC protocol was used to examine the development of the MWD in the early stages of DAC formation under different reaction conditions. The time-dependent partial depolymerization of the polymer backbone was observed. Furthermore, the stability of DAC towards different pH conditions ranging from strongly acidic to strongly alkaline was tested. The depolymerization of DAC in alkaline media occurred with concomitant degradation of aldehyde moieties. In turn, DAC proved to be remarkably stable in acidic and neutral solutions up to a pH of 7.

Abstract:In this research, for the first time, we introduce a vapor-monitoring headspace strategy to establish a new methodology for determining water absorbability of cellulose-derived materials. The method involves detecting the water in the gas phase from various cellulose-derived materials after achieving the equilibrium state. By utilizing the headspace technique to monitor the change in water vapor pressure from bound water to free water, a change point indicating the water absorbability can be identified through fitting procedures. The study showed that the newly-established method possesses high precision (relative standard deviation ≤ 2.92%) and accuracy (relative differences ≤ 5.74%) for water absorbability analysis. The present method emerges as a facile and reliable tool for measuring water absorbability, and the introduction of the vapor-monitoring headspace strategy is anticipated to inspire the development of a new type of analytical method.

Abstract:Cellulose, mostly in the form of toilet paper, forms a major component of the particulates in raw municipal wastewater, which could lead to significant consequences due to the potential accumulation of cellulosic fibers and slow biodegradability. Despite the sparse reports on cellulose content and degradation in wastewater and sludge, an accurate and validated method for its quantification in such matrices does not exist. In this paper, four different methods were compared including dilute acid hydrolysis, concentrated acid hydrolysis, enzymatic hydrolysis, and the Schweitzer reagent method. The Schweitzer reagent method, applied to municipal wastewater and sludge, was found to be a very robust and reliable quantification method in light of its reproducibility, accuracy, and ideal (100%) recovery. The determination of cellulose content is critical to understand its fate in wastewater treatment plants as well as improve sludge management and enhance resource recovery.

Abstract:Heavy alcohol drinking may trigger the onset of acute myocardial infarction. Although the underlying mechanisms are poorly understood, alcohol-induced changes in blood homeostasis, endothelial dysfunction, decreased fibrinolysis, and coronary spasm are the possible explanations. There are rare reports of acute myocardial infarction in young individuals triggered by acute heavy alcohol intake. We present a case of acute ST segment elevation myocardial infarction triggered by heavy alcoholic binge drinking in a man and discuss the available explanations of this rare association.

Abstract:This work presents a novel application of gas-diffusion microextraction (GDME) for the extraction and identification of volatile carbonyl compounds from a dry-process fibreboard (MDF) by combining high-performance liquid chromatography with diode array detection (HPLC–DAD) and mass spectrometry (MS). GDME is a simple, inexpensive, and environmentally friendly technique that allows the simultaneous extraction and derivatization with 2,4-dinitrophenylhydrazine (DNPH) of selected volatiles. The extraction conditions were optimized using design of experiments through a 24 full fractional design followed by a Box–Behnken design, resulting in 35 min of extraction at 45 °C using 500 μL of DNPH 0.15%. The analysis of commercial MDF samples with different characteristics, such as thickness or colour, showed a distinct emission profile of volatile carbonyls. The principal emitting compounds found were formaldehyde, acetaldehyde, acetone, butanal, pentanal, hexanal, heptanal, octanal, and nonanal. A total of 25 compounds were identified using MS, including saturated and unsaturated aldehydes, ketones, dicarbonyls, and benzaldehyde-derivatives. This method can be a valuable tool for the qualitative evaluation of VOCs released from wood-based panels and for the assessment of indoor-air quality.

Abstract:Abstract Solvent extracts of mammalian tissues and blood contain a large amount of co-extracted matrix components, in particular lipids, which can adversely affect instrumental analysis. Clean-up typically degrades non-persistent chemicals. Alternatively, passive sampling with the polymer polydimethylsiloxane (PDMS) has been used for a comprehensive extraction from tissue without altering the mixture composition. Despite a smaller fraction of matrix being co-extracted by PDMS than by solvent extraction, direct analysis of PDMS extracts was only possible with direct sample introduction (DSI) GC-MS/MS, which prevented co-extracted matrix components entering the system. Limits of quantitation (LOQ) ranged from 4 to 20 pg μL −1 ethyl acetate (PDMS extract) for pesticides and persistent organic pollutants (POPs). The group of organophosphorus flame retardants showed higher LOQs up to 107 pg μL −1 due to sorption to active sites at the injection system. Intraday precision ranged between 1 and 10%, while the range of interday precision was between 1 and 18% depending on the analyte. The method was developed using pork liver, brain, and fat as well as blood and was then applied to analyze human post-mortem tissues where polychlorinated biphenyls (PCBs) as well as dichlorodiphenyltrichloroethane (DDT) and DDT metabolites were detected.

Abstract:In the search for a replacement for fossil fuel and the valuable chemicals currently obtained from crude oil, lignocellulosic biomass has become a promising candidate as an alternative biorenewable source for crude oil. Hence, many research efforts focus on the extraction, degradation, and catalytic transformation of lignin, hemicellulose, and cellulose. Unfortunately, these processes result in the production of very complex mixtures. Further, while methods have been developed for the analysis of mixtures of oligosaccharides, this is not true for the complex mixtures generated upon degradation of lignin. For example, high-performance liquid chromatography/multiple stage tandem mass spectrometry (HPLC/MS(n)), a tool proven to be invaluable in the analysis of complex mixtures derived from many other biopolymers, such as proteins and DNA, has not been implemented for lignin degradation products. In this study, we have developed an HPLC separation method for lignin degradation products that is amenable to negative-ion-mode electrospray ionization (ESI doped with NaOH), the best method identified thus far for ionization of lignin-related model compounds without fragmentation. The separated and ionized compounds are then analyzed by MS(3) experiments to obtain detailed structural information while simultaneously performing high-resolution measurements to determine their elemental compositions in the two parts of a commercial linear quadrupole ion trap/Fourier-transform ion cyclotron resonance mass spectrometer. A lignin degradation product mixture was analyzed using this method, and molecular structures were proposed for some components. This methodology significantly improves the ability to analyze complex product mixtures that result from degraded lignin.

Abstract:A sensitive and rapid analytical methodology based on derivatization followed by gas chromatography‐mass spectrometry ( GC ‐ MS ) was developed for the quantitative determination of the toxic contaminant ethyl carbamate ( EC , urethane, C 2 H 5 OCONH 2 ) in alcoholic samples. EC was extracted using liquid–liquid extraction technique, and then silylated with bis ‐(trimethylsilyl)trifluoroacetamide, analysed finally by GC ‐ MS . The isopropyl carbamate was used as the internal standard for quantitative analysis of EC in alcoholic samples. In this work, the sample extraction and derivatization reaction conditions were investigated, and the optimal extraction conditions obtained were: pH 9 and solvent of ethyl acetate, and the derivatization conditions were: derivatization reaction temperature of 80°C and time duration of 30 min. With the optimal conditions, the method validations were also studied. In the validation studies, EC exhibited good linearity with a regression coefficient of 0.9999. The limit of detection and limit of quantification were 0.30 and 5.0 μg/kg, respectively. The precision was less than 8.4%. Finally, the proposed technique was successfully applied to the analysis of EC in 35 kinds of alcoholic samples. The experimental results have demonstrated that the proposed technique is a fast, reliable and low‐cost method for determination of EC in alcoholic samples.
Trace Analysis of Phenols and Chlorophenols in Water by in Situ Derivatization Headspace Solid-Phase Microextraction Coupled with Gas Chromatography/Mass Spectrometry

Abstract:An in situ derivatization headspace solid phase microextraction method has been developed for the determination of phenols and related chlorophenols in water. Acetylated derivatives were selectively determined using gas chromatography/mass spectrometry (GC/MS). Parameters affecting both derivatization and SPME procedures, such as fiber coating, extraction time, temperature, volume of derivatizating reagent and ionic strength, were studied. Then the polydimethylsiloxane/divinylbenzene fiber (PDMS/DVB, 65 mu m) was selected. The performance of the method was evaluated in terms of accuracy, linearity, precision and limits of detection. Under the selected conditions, i.e. 4.0 g NaCl, 0.10 g Na2HPO4 in every 10 mL sample solution, headspace extraction at 60 degrees C and 600 r/min for 30 min, quantitative recoveries and satisfactory precision (RSD <= 13.7%) were obtained. Limits of detection for all target compounds ranged from 0.014 to 0.044 mu g/L. Linearity was found in a wide range of concentrations. Finally, the applicability of the proposed method is demonstrated by determining phenols in real wastewater samples.

Abstract:A new technique, headspace single-drop microextraction (HS-SDME) with in-drop derivatization, was developed. Its feasibility was demonstrated by analysis of the model compounds, aldehydes in water. A hanging microliter drop of solvent containing the derivatization agent of O-2,3,4,5,6-(pentaflurobenzyl)hydroxylamine hydrochloride (PFBHA) was shown to be an excellent extraction, concentration, and derivatization medium for headspace analysis of aldehydes by GC-MS. Using the microdrop solvent with PFBHA, acetaldehyde, propanal, butanal, hexanal, and heptanal in water were headspace extracted and simultaneously derivatized. The formed oximes in the microdrop were analyzed by GC-MS. HS-SDME and in-drop derivatization parameters (extraction solvent, extraction temperature, extraction time, stirring rate microdrop volume, and the headspace volume) and the method validations (linearity, precision, detection limit, and recovery) were studied. Compared to liquid-liquid extraction and solid-phase microextraction, HS-SDME with in-drop derivatization is a simple, rapid, convenient, and inexpensive sample technique.
Monitoring residual solvents in pharmaceutical products using a portable pre-concentration GC-PID

Abstract:Pharmaceutical manufacturing utilizes solvents at different stages of production. Some of the harmful solvent residuals may be retained in the final product; therefore, they need to be monitored for quality control and to meet the regulation requirement. Here, a novel method capable of rapidly analyzing residual solvents in pharmaceutical products was developed using a compact-portable gas chromatography with a photoionization detector (GC-PID). The method consists of modified Tedlar® bag sampling, online pre-concentration, separation of volatiles by miniaturized GC, and micro-PID detection. The method detection limits of selected residual solvents were in the range of 26.00 - 52.03 pg/mL which is much lower than the pharmaceutical compliance concentration limits. Limits of detection > 520 pg of analyte per grams of sample was also determined for the over-the-counter drugs. The method performance showed rapid speed (5 min), linear calibration (r2 < 0.99), and repeatable retention time (RSD < 0.4 %). Direct analysis of residual solvents in solid samples was conducted without the need for complex sample preparation. The method validation using over-the-counter pharmaceutical products yielded excellent accuracy (recovery > 91.2 %) and precision (RSD < 6.5 %) for the selected residual solvents, including 1,4-dioxane, benzene, chlorobenzene, cyclohexane, xylenes, and toluene. This portable and rapid method could be deployed during the pharmaceutical drug manufacturing processes for quality control.

Abstract:A sensitive quantification of decabrominated diphenyl ether (deca-BDE) was achieved by optimizing the injection temperature, pressure and column system with gas chromatography mass spectrometry in selected ion monitoring mode. A simple one-step cleanup method with acid silica gel column chromatography showed the optimum efficiency and minimum matrix interference. Ultrasonic-assisted extraction with toluene provided the best performance (111%, 1 h) as compared to Soxhlet extraction (110%, 5 h) and microwave-assisted extraction (63%, 15 min) taking consideration of extraction efficiency and consumed time. The proposed method could meet the requirement for determination of deca-BDE in polymers and has the potential to monitor the content of deca-BDE in electrical and electronic equipment for routine analysis.
Investigation of Deep Eutectic Solvent-Based Microwave-Assisted Extraction and Efficient Recovery of Natural Products

Abstract:A systematic study of the principles of deep eutectic solvent-based microwave-assisted extraction (DES-MAE) was performed. It was found that the heating rates of most DESs decreased (heat capacity increased) under microwave irradiation with increasing water content, allowing high-efficiency extraction for thermally sensitive compounds. In addition, DESs containing carboxylic acids reacted with hydroxyl groups of sugar and choline chloride, resulting in cell wall destruction and inhibition of cellulose, hemicellulose, and lignin reconnection in cell walls through hydrogen bonds, thus leading to better extraction performance. This was verified by extracting anthraquinones from <i>Rheum palmatum</i> using DES-MAE and optimizing extraction conditions. DES with citric acid as the hydrogen bonding donor gave the highest extraction efficiency under the optimized conditions. In addition, anthraquinones in the DES extract were recovered using three kinds of silica modified by different functional groups. The results showed that material containing a phenyl group is beneficial to the recovery of anthraquinones in acid-based DESs because it can facilitate strong hydrophobic and π–π interactions. This study showcases the green chemistry applications of DES-MAE in a laboratory and industry alike, and demonstrates the recovery of natural products from DES extracts.
