Abstract:In order to improve the stability and bioavailability of camellia oil, as well as expanding its application in aqueous, a complex (SPIQU) was prepared using soybean protein isolate (SPI) and quercetin (Qu) as material. The structural composition, emulsification properties, and antioxidant properties of SPIQU were evaluated using infrared, particle size, contact angle, bromophenol blue binding rate, and free radical clear rate tests. On this basis, Pickering emulsions were prepared using SPIQU as wall material and curcumin dissolved in camellia oil as core material, and the emulsion properties, antioxidant activity, microstructure, rheology and stability tests were performed on Pickering emulsions. The results showed that when quercetin was added 1.0%, the particle size of SPIQU was 514.61 ± 16.35 nm, the complex binding rate was 89.06%, and the ABTS radical scavenging rate was 78.72%, showing good surface hydrophobicity and emulsification properties. The prepared Pickering emulsion had uniform droplets and good emulsification characteristics, with an average particle size of 8.3 ± 2.7μm. It was a pseudoplastic fluid by rheological test, and Pickering emulsion showed good stability in storage, centrifugation and heat within four weeks. Therefore, SPIQU can form a stable Pickering emulsion system from oil and oil-soluble substances, which can be used as a good emulsion preparation method in the field of food, pharmaceuticals, and cosmetics.

Development of high-internal-phase emulsions stabilized by soy protein isolate-dextran complex for the delivery of quercetin

Abstract:Background: Protein-polysaccharide complexes have been widely used to stabilize high-internal-phase emulsion (HIPEs). However, it is still unknown whether soy protein isolate-dextran (SPI-Dex) complexes can stabilize HIPEs or what is the effect of Dex concentration on the HIPEs. Furthermore, the non-covalent interaction mechanism between SPI and Dex is also unclear. Therefore, we fabricated SPI-Dex complexes and used them to stabilize HIPEs-loaded quercetin and explore the interaction mechanism between SPI and Dex, as well as the effect of Dex concentration on the particle size, ζ-potential, microstructure, rheology, quercetin encapsulation efficiency, and gastrointestinal fate of the HIPEs. Results: Spectral analysis (fourier transform infrared spectroscopy, ultraviolet spectroscopy, and fluorescence spectroscopy) results identified the formation of SPI-Dex complexes, and indicated that the addition of Dex changed the spatial structure of SPI, whereas thermodynamic analysis (ΔH > 0, ΔS > 0) showed that hydrophobic interactions were the main driving forces in the formation of SPI-Dex complexes. Compared with HIPEs stabilized by SPI, the SPI-Dex complex-stabilized HIPEs had smaller particles (3000.33 ± 201.22 nm), as well as higher ζ-potential (-21.73 ± 1.10 mV), apparent viscosities, modulus, and quercetin encapsulation efficiency (98.19 ± 0.14%). In addition, in vitro digestion revealed that SPI-Dex complex-stabilized HIPEs significantly reduced the release of free fatty acid and improved quercetin bioaccessibility. Conclusion: HIPEs stabilized by SPI-Dex complexes delayed the release of free fat acid and improved the bioaccessibility of quercetin, and may be help in designing delivery systems for bioactive substances with specific properties. © 2022 Society of Chemical Industry.
Pickering emulsions stabilized by soybean protein isolate/chitosan hydrochloride complex and their applications in essential oil delivery

Abstract:In this work, fabrication of soybean protein isolate (SPI)/chitosan hydrochloride (CHC) composite particles stabilized O/W Pickering emulsions using soybean oil as an oil phase was optimized by examining the effects of pH, SPI/CHC mass ratio, SPI/CHC composite particle concentration and oil phase fraction on the stability of the emulsions. The results showed that under the conditions of SPI/CHC mass ratio 1:1, pH 4 and particle concentration 2 %, the SPI/CHC composite particles could stabilize the emulsions with oil phase fraction up to 80 %. At an oil phase fraction of 60 %, the emulsions had a minimum particle size. The microstructure, storage and oxidation stabilities and rheological properties of the emulsions were determined. Using this SPI/CHC composite particle-stabilized Pickering emulsion template, citrus essential oil (CEO) Pickering emulsion (CEOP) was prepared. CEOP was found to markedly inhibit two food-related microorganisms, Staphylococcus aureus and Escherichia coli . In addition, the CEOP emulsion dilution (containing 4500 μL CEO/L) not only improved the water solubility of CEO, but also effectively retarded the browning and bacterial growth of fresh-cut apple. The SPI/CHC-stabilized Pickering emulsion template constructed in this work provides a promising alternative for the delivery of antimicrobial essential oils in the food industry.

Abstract:Summary The use of two emulsifiers to fabricate a composite emulsifier is a useful strategy used to strengthen the emulsifying capacity. To investigate the feasibility of using soy protein isolate (SPI) to stabilize Pickering emulsions stabilized by soy peptide nanoparticles (SPNs), the SPI was introduced into the SPN system to fabricate SPI‐SPN complex emulsifiers, and the effects of the SPI on the properties of the SPNs and SPN‐stabilized emulsions were investigated with various oil fractions. The addition of SPI significantly reduced the particle sizes and surface hydrophobicities. Furthermore, the SPI increased the structural rearrangements of the SPNs at the oil‐water interface. Compared with emulsions stabilized with the SPN alone, the droplet sizes of the SPI‐SPN emulsions were smaller, and the interface protein adsorption percentages were lower for all oil fractions. The viscosity of the SPI‐SPN emulsion was higher than that of the SPNs emulsion at Φ≤0.60. Decreases in G′ and G′′ were observed when the SPI was added at all Φ values tested. More importantly, the added SPI markedly enhanced the creaming stabilities of the SPN emulsions at low Φ (≤0.40). These results confirmed that SPI improved the emulsifying activity of the SPNs and enhanced the stabilities of Pickering emulsions stabilized with SPNs at low Φ.

Abstract:Astaxanthin (AXT) is a lipid-soluble carotenoid with good anti-oxidation, hepatic steatosis reduction, anti-inflammation and intestinal microbiota regulation ability, whose poor stability and pH vulnerability limit its bioavailability. Spirulina protein (SP) derived from Spirulina has good emulsifying ability with potential application in nutraceuticals, medicines and cosmetics. In this study, Pickering emulsions were fabricated using SP-chitosan (CS) complex as an emulsifier. The particle size, zeta potential and three-phase contact angle of SP-CS complex with different SP to CS ratios were investigated. A mass ratio of 1:2.5 SP-CS complex showed good emulsifying ability in preparing Pickering emulsion. Higher storage modulus and viscoelasticity were observed with higher SP-CS complex concentrations and oil fractions. The SP-CS Pickering emulsion significantly improved the stability and bioavailability of AXT. The results indicated that the SP-CS complex could act as a Pickering emulsion stabilizer and had the potential to deliver protective hydrophobic AXT.

Abstract:The potential of ultrasonication-driven molecular self-assembly of whey protein isolate (WPI) with chitosan (CS)/chitooligosaccharide (COS) to stabilize Pickering emulsions was examined, based on CS/COS ligands-induced partial unfolding in remodeling the Pickering particles features. Multi-spectral analysis suggested obvious changes in conformational structures of WPI due to interaction with CS/COS, with significantly higher unfolding degrees of WPI induced by COS. Non-covalent interactions were identified as the major forces for WPI-CS/COS conjugates. Ultrasonication enhanced electrostatic interaction between CS's -NH 3 groups and WPI's -COO − groups which improved emulsification activity and storability of WPI-COS stabilized Pickering emulsion. This was attributed to increased surface hydrophobicity and decreased particle size compared to WPI-CS associated with differential unfolding degrees induced by different saccharide ligands. CLSM and SEM consistently observed smaller emulsion droplets in WPI-COS complexes than WPI-CS/COS particles tightly adsorbed at the oil-water interface. The electrostatic self-assembly of WPI with CS/COS greatly enhanced the encapsulation efficiency of quercetin than those stabilized by WPI alone and ultrasound further improved encapsulation efficiency. This corresponded well with the quantitative affinity parameters between quercetin and WPI-CS/COS complexes. This investigation revealed the great potential of glycan ligands-induced conformational transitions of extrinsic physical disruption in tuning Pickering particle features.

Abstract:This study prepared SPI-Pol-HPMC (SPH) nanoparticles from soybean protein isolate (SPI), hydroxypropyl methyl cellulose (HPMC), and broccoli leaf polyphenol (Pol) and used them as a stabilizer for the Pickering emulsion. The SPH (2:1) nanoparticles have the best ability to encapsulate broccoli leaf polyphenols, with uniform particle size distribution, and a more dense and stable structure. The chemical and hydrogen bonding forces between the SPH nanoparticle components were enhanced. Additionally, the 1.5 % SPH nanoparticle-stabilized emulsions exhibited good physical stability, manifesting as small particle droplets with good rheological properties and uniform dispersion. The volume fraction of the emulsified phase of the 1.5 % SPH nanoparticle-stabilized emulsions was the greatest after 21 days of storage. Interestingly, SPH nanoparticles also improved the oxidative stability of the emulsions, as evidenced through their lower peroxide values and thiobarbituric acid active substances. The aforementioned results suggest that SPH nanoparticles may be used as food-grade emulsifiers that stabilize emulsions and inhibit their lipid oxidation.

Abstract:<p>Herein, we investigated the influence of covalent complexation between (−)-epigallocatechin gallate (EGCG) and soy protein isolate (SPI) on the structural and emulsifying properties of the protein as well as the effect of chitosan (CS) on the stability of emulsions formed by the covalent conjugates. The formation of the covalent complex of SPI and EGCG was confirmed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Three-dimensional fluorescence spectroscopy and circular dichroism spectroscopy were conducted to analyze the changes in the protein structure at different EGCG concentrations (0.005, 0.015, and 0.025 wt%). The addition of EGCG changed the tertiary conformation and secondary structure of the protein—unfolding of the protein polypeptide chain, decrease in the β-strand content, and increase in α-helix and random coil contents were observed. After covalent bonding of SPI with EGCG (0.015 wt %), the complex exhibited optimal solubility and emulsifying properties. SPI–EGCG–CS emulsions were prepared by layer-by-layer electrostatic assembly at pH 5.0. The effect of environmental stress on the stability of the CS–loaded SPI–EGCG (0.015 wt%) emulsion oil droplets was studied by freezing (−20 ℃, pH 5) and thermal treatment (90 ℃, 30 min, pH 5). The stability of the emulsion was improved when the CS concentration was >0.25 wt%; this improvement was attributed to the increase in electrostatic force and steric hindrance among droplets and the formation of a relatively thick biopolymer interface on the surface of oil droplets. In addition, the rheological properties showed that thicker interface layers and network structures caused deviations from the flow behavior of emulsions. These results indicate that SPI − EGCG conjugates can be used as food additives and for the construction of the lipophilic active ingredient delivery system.</p>

Abstract:Pickering emulsions are increasingly being explored for their potential applications within the food industry. However, the stability of these emulsions is often limited when stabilized by single colloidal particles. Herein, we examined the possibility of improving the performance of Pickering emulsions by using a combination of colloidal particles (zein nanoparticles, Z) and small molecular emulsifiers (quillaja saponin, Q). The stability mechanism of Pickering emulsions was explored from microscopic to macroscopic scales using QCM-D, microrheology and Cyro-SEM, et. Corn oil-in-water Pickering emulsions were prepared by adding the Z and Q in different sequences: (i) mix Z and Q and then homogenize ("ZQ-P′′); (ii) add Z, homogenize, and then add Q ("Z-QP"); (iii) add Q, homogenize, and then add Z ("Q-ZP"). The sequence significantly influenced the interfacial structure and properties of Pickering emulsions. Macroscopic rheology and microrheology showed that the Pickering emulsions formed an elastic-like structure (G'>G′′) and their viscoelastic properties increased over time. QCM-D analysis showed that the properties of the interfacial layers depended on the emulsifier addition sequence used to form the Pickering emulsions. The interfacial membranes formed using the Q-ZP sequence were the thickest (5.8 nm). Cyro-SEM imaging showed that nanoparticle bridges formed between neighboring droplet surfaces, which led to the formation of a 3D network that provided mechanical strength. Overall, this study showed that the Q-ZP addition sequence was the most effective at forming stable Pickering emulsions. It provides new insights that may facilitate the formation of plant-based Pickering emulsions with improved functional performance in food applications.

Abstract:In this work, we prepared concentrated Pickering emulsions stabilized by using soybean protein isolate (SPI) and TEMPO-oxidized bacterial cellulose (TOBC) complex. The effects of solid particle content, pH and oil phase volume fraction on the morphology, stability and rheological behaviors of the emulsions had been investigated. It indicated that the emulsion droplets had uniform particle size distribution when the ratio of SPI/TOBC was controlled to 12.5:1 (wt/wt). The emulsification performance could be improved with the increase of solid content from 1.0% to 3.0%. Moreover, the pH played an important role in regulating the structure and properties of the emulsions. With microscopic observation, the emulsions at pH 7.0 had smaller droplet size (10–15 μm) than that (15–40 μm) of the emulsions at pH 3.0. While the emulsions at pH 3.0 maintained a low creaming index (CI) value than those at pH 7.0, and uniform cellular network structure could be seen by SEM observation. It suggested that the electrostatic complexation between soy proteins and anionic bacterial cellulose nanofibrils at pH 3.0 was more favorable for stabilizing emulsions than those at pH 7.0. This work would provide some guidance for regulating the properties of emulsions that stabilized by soy protein and cellulose nanofibrils.

Abstract:In contrast to their well-known physiological properties, phytochemicals, such as flavonoids, have been less frequently examined for their physiochemical properties (e.g., surface activity). A natural quercetin self-stabilizing Pickering emulsion was fabricated and characterized in the present study. The antisolvent precipitation method was used to modify quercetin (in dihydrate form), and the obtained particles were characterized by light microscope, atom force microscope, XRD, and contact angle. The antisolvent treatment was found to reduce the particle size, crystallinity, and surface hydrophobicity of quercetin. We then examined the effects of the antisolvent ratio, particle concentration, and oil fraction on the properties of the quercetin particle-stabilized emulsions. In addition, increasing the antisolvent ratio (1:1~1:10) effectively improved the emulsification performance of the quercetin particles. The emulsion showed good storage stability, and the particle size of the emulsion decreased with the rising particle concentration and increased with the rising oil phase ratio. The findings indicate that natural quercetin treated with antisolvent method has a good ability to stabilize Pickering emulsion, and this emulsion may have good prospective application potential for the development of novel and functional emulsion foods.

Abstract:Abstract This study investigated the emulsifying properties of a protein–polysaccharide hybrid nanoconjugate system comprising cellulose nanocrystals (CNC, 1% w/v) and soy protein isolate at various concentrations (SPI, 1–3% w/v). The average particle size of the nanoconjugate increased, and the zeta potential decreased when 3% (w/v) of SPI was used. The contact angle and thermal stability of CNC improved with the conjugation of SPI. Upon Pickering emulsification, 0.5% (w/v) of CNC–SPI nanoconjugate as particle stabilizer was sufficient to obtain stable emulsions. The CNC–SPI1 formulation (CNC to SPI, 1:1) provided the emulsion with the smallest droplet size and higher emulsifying activity. Intriguingly, ultrasound (US) pre-treatment on nanoconjugates before emulsification significantly reduced the size of the emulsion. The rheological assessment demonstrated that the CNC–SPI-stabilized emulsions exhibit shear thinning behavior at a lower shear rate and shear thickening behavior at a higher shear rate, indicating the interruption of existing attractive interactions between the CNC particles. All emulsions exhibited higher elastic modulus ( G ′) than viscous modulus ( G ′′), suggesting high viscoelastic properties of the emulsions. This study demonstrates that CNC–SPI nanoconjugate with optimum protein to polysaccharide ratio has great potential as a natural particle stabilizer in food and nutraceutical emulsion applications.

Abstract:Objectives/Hypothesis: The objective of this study was to determine whether glycopyrrolate is useful as a vestibular suppressant in patients with Meniere's disease. The tested hypotheses were that glycopyrrolate would decrease the perception of dizziness measured by the Dizziness Handicap Inventory in patients with Meniere's disease and that placebo would cause no such decrease. Study Design: Randomized, prospective. Methods: Thirty‐seven subjects with a diagnosis of Meniere's disease were administered either 2 mg of glycopyrrolate or placebo twice daily as needed for vertigo. All were also administered the regimen of 1500 mg sodium/day diet and diuretic. The following indices were examined: Dizziness Handicap Inventory, Tinnitus Handicap Inventory, Modified Somatic Perception Questionnaire, Beck Depression Inventory, hearing examination, and electronystagmography. After 4 to 6 weeks of the drug regimen, Dizziness Handicap Inventory, Tinnitus Handicap Inventory, Modified Somatic Perception Questionnaire, and Beck Depression Scale were reexamined. Paired t tests were performed to verify the significance of improvement before and after treatment. Results: Subjects who received glycopyrrolate had statistically significant reduction in Dizziness Handicap Inventory, Beck Depression Score, and Modified Somatic Perception Score. In the placebo group, no improvement in any index was found. Abstract:To investigate the optimal delivery system of quercetin, in this paper, cellulose nanocrystals (CNCs) extracted from rice bran were used to stabilize the Pickering emulsion and Pickering emulsion gels (PEGs) with quercetin. To compare the emulsion properties, stability, antioxidation activity, encapsulation rate, and bioaccessibility of the quercetin, four emulsions of CNC Pickering emulsion (C), CNC Pickering emulsion with quercetin (CQ), CNC Pickering gel emulsion (CG), and CNC Pickering gel emulsions with quercetin (CQG) were prepared. All four emulsions exhibited elastic gel network structure and good stability. The quercetin significantly reduced the particle size, increased the stability, and improved the antioxidant capacity of CQ and CQG. Compared to C and CG, the ABTS+ radical scavenging capacities of CQ and CQG were respectively enhanced by 46.92% and 3.59%. In addition, CQG had a higher encapsulation rate at 94.57% and higher bioaccessibility (16.17) compared to CQ. This study not only indicated that CNC from rice bran could be exploited as an excellent stabilization particle for Pickering emulsions, but also provided a highly stable and bioaccessible delivery system for water-insoluble functional active factors.

Abstract:There is fast increasing interest in the development of food grade high internal phase emulsions (HIPEs; with oil fractions > 0.74) stabilized by protein-based particles, due to their potential applications in food, cosmetics and biomedical fields. This work reported that soy glycinin (SG), an important soy storage globulin, exhibited a potential to act as Pickering stabilizers for HIPEs; and the glycation with soy soluble polysaccharide (SSPS) greatly improved the emulsification performance of SG, the gel network formation and stability (against heating or freeze-thawing) of the resultant HIPEs. In the work, two selected glycation incubation periods (24 and 72 h) were applied to obtain the glycated SG. The results indicated that the glycation progressively decreased the minimal concentration required for the formation of gel-like HIPEs, and droplet size, but increased the gel network strength, in an incubation period dependent manner. The improvement of emulsification performance by the glycation was mainly associated with the enhanced adsorption, as well as facilitated subunit dissociation at the interface, during the HIPE formation. All the HIPEs stabilized by untreated or glycated SG exhibited an excellent coalescence stability against a long-term storage. The structural analyses of non-adsorbed and adsorbed SG indicated that although untreated SG showed a high whole structural integrity with no distinct changes in structures after adsorption at interface, it tended to aggregate (due to its high surface hydrophobicity); in contrast, glycated SG easily dissociated into subunits at the interface, but the dissociated subunits exhibited a high structural stability at the tertiary and secondary levels. The findings would be of importance not only for the development of soy protein-based Pickering stabilizers for emulsions or HIPEs, but also for extending the knowledge about the modification of emulsifying properties of proteins by glycation with carbohydrates.

Abstract:Curcumin, as a lipid-soluble and environment-sensitive (light, heat and oxidants) polyphenol, need to be embedded to develop health care products with stable storage-stability and of higher bioaccessibility. Enzymatically modified quinoa starch (EMQS) based emulsion gel was fabricated and used as the carrier for curcumin. The emulsifying property and mechanism of EMQS, and the micromorphology, stability, rheological properties and in vitro digestion properties of the emulsion gel were studied. The decreased particle size and increased contact angle of QS after enzymatic hydrolysis contributed to the improved emulsifying properties of EMQS. The curcumin-encapsulated EMQS Pickering emulsion gel showed better emulsifying stability, much clearer solid-like behavior, higher gel strength and viscosity than that of Tween 80 emulsion. The retention rates of curcumin in the EMQS Pickering emulsion gel were higher than 85% after subjected to the different treatments. And under 7 h UV and 8-day storage conditions, the protective effect of EMQS-based Pickering emulsion gel on curcumin was even higher than that of tween 80 emulsion system. The lipid digestion extent in EMQS Pickering emulsion gel was higher than that in bulk oil system and lower than that in tween 80 system. Compared with that of bulk oil system (5.37%), the bioaccessibility of curcumin in EMQS Pickering emulsion gel (38.57%) was greatly improved, proving that EMQS Pickering emulsion gel could be used as delivery vehicle for curcumin.

Abstract:Summary Tea processing often generates significant waste, resulting in resource inefficiency. However, extracting green tea polysaccharide conjugate (gTPC) from this waste has shown promising bioactivity. Previous studies have confirmed the emulsifying properties of gTPC. Yet, its use as an emulsifier is limited due to its requirement for high surface activity. The findings revealed that a 0.40 wt% soy protein isolate (SPI) and 0.50 wt% gTPC conjugate required 300°C for complete decomposition, demonstrating the highest stability among other ratios. The Pickering emulsion, using a 0.30 wt% emulsifier ratio, exhibited exceptional stability. Moreover, it showed remarkable zeta potential and particle size stability under stressful conditions (temperature, pH, and metal ions). Additionally, it effectively protected curcumin from adverse pH, light, and temperature effects, with a retention rate of over 74.00%. These results highlight the gTPC/SPI combination as an effective stabilizer, offering new prospects for gTPC development and utilization.

Abstract:Forty hospitalized schizophrenics and 40 volunteers from the community listened to sentences in which a click was embedded. Half of the schizophrenic subjects and half of the normal subjects were asked to reproduce the sentences and indicate precisely where the click occurred. The remaining subjects were given the previously heard sentences and asked to recognize the click locations. Results showed that schizophrenic subjects tend to be less accurate than normal subjects in locating the clicks. However, this inferior performance was not due to an inability to use syntactic rules. Rather, the pattern of errors demonstrated that schizophrenic subjects distinguish between sentences which are acoustically identical but syntactically distinct, and that this distinction is maintained under response conditions with varying emphases on sentence retrieval. It is concluded that schizophrenic subjects appear to use syntax as a basis for sentence processing at least to the same extent as normal subjects do.
