Abstract:The northern reef of Yongxing Island, the largest reef island of the Xisha Islands in the South China Sea, was in good condition with significant cover of scleractinian corals until 2002. Surveys in 2008 and 2010, however, found that coral coverage had declined rapidly and severely, implying that catastrophic coral mortality occurred during the past 8 years. A blackish mat was observed covering live and dead corals in both 2008 and 2010 that was identified as an encrusting sponge, Terpios hoshinota , by special surface morphology and spicule structure. In addition, spicule residues were found on the surface of long-dead corals, indicating a previous invasion of T. hosinota. T. hoshinota is referred to as the “black disease” because it rapidly overgrows and kills corals. Our evidence indicates that outbreaks of black disease are at least partially responsible for the massive coral mortality at the northern reef of Yongxing Island over the past 8 years, although human activities and heat-related coral bleaching cannot be discounted as minor causes for this coral decline.
Microbial and transcriptional response of Acropora valida and Turbinaria peltata to Vibrio coralliilyticus challenge: insights into corals disease resistance

Abstract:Background: Coral diseases are significant drivers of global coral reef degradation, with pathogens dominated by Vibrio coralliilyticus playing a prominent role in the development of coral diseases. Coral phenotype, symbiotic microbial communities, and host transcriptional regulation have been well-established as factors involved in determining coral disease resistance, but the underlying mechanisms remain incompletely understood. Methods: This study employs high-throughput sequencing to analyse the symbiotic microbial and transcriptional response of the hosts in order to evaluate the disease resistance of Acropora valida and Turbinaria peltata exposed to Vibrio coralliilyticus. Results: A. valida exhibited pronounced bleaching and tissue loss within 7 h of pathogen infection, whereas T. peltata showed no signs of disease throughout the experiment. Microbial diversity analyses revealed that T. peltata had a more flexible microbial community and a higher relative abundance of potential beneficial bacteria compared to A. valida. Although Vibrio inoculation resulted in a more significant decrease in the Symbiodiniaceae density of A. valida compared to that of T. peltata, it did not lead to recombination of the coral host and Symbiodiniaceae in either coral species. RNA-seq analysis revealed that the interspecific differences in the transcriptional regulation of hosts after Vibrio inoculation. Differentially expressed genes in A. valida were mainly enriched in the pathways associated with energy supply and immune response, such as G protein-coupled receptor signaling, toll-like receptor signaling, regulation of TOR signaling, while these genes in T. peltata were mainly involved in the pathway related to immune homeostasis and ion transport, such as JAK-STAT signaling pathway and regulation of ion transport. Conclusions: Pathogenic challenges elicit different microbial and transcriptional shifts across coral species. This study offers novel insights into molecular mechanisms of coral resistance to disease.
Broadscale coral disease interventions elicit efficiencies in endemic disease response

Abstract:The presence and abundance of reef-building corals are crucial to the long-term existence of Caribbean coral reef ecosystems, providing both direct and indirect, local and global, ecological, economic, and social benefits. In 2014, stony coral tissue loss disease (SCTLD) was first identified in southeast Florida and remains endemic to the region, while continuing to spread throughout the Caribbean. Effective in situ intervention treatments using antibiotic paste can halt lesion progression on Montastraea cavernosa up to 90% of the time. This study investigated intervention activities over a three-year period to identify efficiencies in disease response. Since May 2019, 1,037 corals, >85% of which were M. cavernosa , were treated during disease intervention dives in southeast Florida. Treated coral density, the number of treated corals per meter along a dive track, was significantly higher in the first year compared to subsequent years and displayed annual peaks in late summer each year. Season significantly influenced treatment density, leading to higher values in the wet season across all years, 2019 to 2022. Areas of highest treatment density were identified between Haulover Inlet and Government Cut near Miami and Hillsboro Inlet in northern Broward County. Areas with the highest treatment density were only identified in the first year, suggesting that broadscale interventions may have decreased disease prevalence in subsequent years. Results indicate that in endemic areas with sporadic and dynamic disease prevalence, intervention efforts should be weighted proportionally across space and time to maximize intervention efficiency. This study provides optimistic results for the potential of interventions reducing disease prevalence and supports that disease interventions are an effective coral restoration tool that can decrease the increasing burden on post hoc coral restoration.

Abstract:Considered one of the most devastating coral disease outbreaks in history, stony coral tissue loss disease (SCTLD) is currently spreading throughout Florida's coral reefs and the greater Caribbean. SCTLD affects at least two dozen different coral species and has been implicated in extensive losses of coral cover. Here we show Pseudoalteromonas sp. strain McH1-7 has broad-spectrum antibacterial activity against SCTLD-associated bacterial isolates. Chemical analyses indicated McH1-7 produces at least two potential antibacterials, korormicin and tetrabromopyrrole, while genomic analysis identified the genes potentially encoding an L-amino acid oxidase and multiple antibacterial metalloproteases (pseudoalterins). During laboratory trials, McH1-7 arrested or slowed disease progression on 68.2% of diseased Montastraea cavernosa fragments treated ( n = 22), and it prevented disease transmission by 100% ( n = 12). McH1-7 is the most chemically characterized coral probiotic that is an effective prophylactic and direct treatment for the destructive SCTLD as well as a potential alternative to antibiotic use.

Abstract:Caribbean coral reefs are currently facing a rapid decline caused by a plethora of threats including disease outbreaks. Octocorals appear to be unaffected by the majority of diseases impacting scleractinian corals, including stony coral tissue loss disease (SCTLD) that emerged in 2014 and resulted in a mass mortality of scleractinian coral populations inhabiting Florida, the USA, and Caribbean reefs. Although the Caribbean Sea is considered a disease hot spot, few investigations into the mechanism(s) responsible for the resistance of octocorals have been conducted. In response, the capacity for octocoral-derived extracts and natural products to inhibit strains of Vibrio coralliilyticus , pathogenic bacteria that can cause bleaching and disease in stony corals and can co-occur in SCTLD infections, was explored. Extracts obtained from each of the four octocoral species studied demonstrated antimicrobial activity against V. coralliilyticus . Bioassay-guided fractionations of crude extracts from Antillogorgia americana were employed to identify the antimicrobial compounds, revealing the presence of secosterols in the most bioactive fractions. These results suggest that octocoral species may utilize chemical defenses to protect themselves against infection by strains of a known coral pathogen and contribute to the body of knowledge regarding the success of octocorals on Caribbean reefs.

Abstract:Abstract Coral reefs are in global decline with coral diseases playing a significant role. This is especially true for Acroporid corals that represent ~25% of all Pacific coral species and generate much of the topographic complexity supporting reef biodiversity. Coral diseases are commonly sediment-associated and could be exacerbated by overharvest of sea cucumber detritivores that clean reef sediments and may suppress microbial pathogens as they feed. Here we show, via field manipulations in both French Polynesia and Palmyra Atoll, that historically overharvested sea cucumbers strongly suppress disease among corals in contact with benthic sediments. Sea cucumber removal increased tissue mortality of Acropora pulchra by ~370% and colony mortality by ~1500%. Additionally, farmerfish that kill Acropora pulchra bases to culture their algal gardens further suppress disease by separating corals from contact with the disease-causing sediment—functioning as mutualists rather than parasites despite killing coral bases. Historic overharvesting of sea cucumbers increases coral disease and threatens the persistence of tropical reefs. Enhancing sea cucumbers may enhance reef resilience by suppressing disease.

Abstract:Ciliates are considered the most harmful parasites of fish, causing mass mortalities and thus substantial economic losses to the aquaculture and ornamental fish industries. To better elucidate their pathogenic mechanisms and facilitate the development of effective prevention and control strategies, it is essential to apply novel technologies to ensure the continuous and sufficient supply of parasite specimens. This review discusses the latest research progress in the artificial culture of six important parasitic ciliates affecting both freshwater and marine fish globally: Ichthyophthirius multifiliis, Cryptocaryon irritans, Chilodonella (C. hexasticha, C. piscicolaand C. uncinata), Balantidium ctenopharyngodoni, Scuticociliatida gen. spp., and Tetrahymena spp. Methods for in vitro and in vivo culture as well as cryopreservation/preservation (depending on the specific species) of these organisms are described herein. Challenges and future applications of parasite culture are also discussed.

Abstract:Scuticociliates are recognized as the causative agents of mass mortalities in certain cultured marine fishes, resulting in enormous economic losses. This study aimed to investigate a fatal infection caused by scuticociliates in farmed large yellow croaker (Larimichthys crocea) in Fujian province, China. Microscopic examinations of focal organs, including the brain, eyes, gills, and skin, revealed the presence of parasites. Active masses of scuticociliates were observed in these organs, and the ciliates were subsequently isolated and maintained in vitro. An immersion challenge experiment revealed that L. crocea experienced cumulative mortalities reaching 73% within 7 d upon exposure to 1.0 × 104 ciliates mL-1. Based on the microscopic and PCR testing of infected fishes, the brain was comprehensively inferred as the main infection organ for the isolated strain. Microscopic and submicroscopic observations of the isolated scuticociliate, coupled with cortical ciliature patterns revealed through α-tubulin indirect immunofluorescence techniques, identified these scuticociliates as Miamiensis avidus. The sequencing of two genetic markers (small subunit ribosomal RNA, SSU rRNA and cytochrome c oxidase subunit I, COI) further confirmed that the isolated strains exhibited the highest sequence similarity to most M. avidus sequences in GenBank. However, significant differences in SSU sequences compared to the M. avidus strain Ma/2, and the lack of published COI and ITS (internal transcribed spacer) sequences for Ma/2, indicate the need for further molecular data to resolve whether there are potential cryptic species within the M. avidus complex.

Abstract:Stony coral tissue loss disease (SCTLD) is destructive and poses a significant threat to Caribbean coral reef ecosystems. Characterized by the acute loss of coral tissue, SCTLD has impacted over 22 stony coral species across the Caribbean region, leading to visible declines in reef health. Based on the duration, lethality, host range, and spread of this disease, SCTLD is considered the most devastating coral disease outbreak ever recorded. Researchers are actively investigating the cause and transmission of SCTLD, but the exact mechanisms, triggers, and etiological agent(s) remain elusive. If left unchecked, SCTLD could have profound implications for the health and resilience of coral reefs worldwide. To summarize what is known about this disease and identify potential knowledge gaps, this review provides a holistic overview of SCTLD research, including species susceptibility, disease transmission, ecological impacts, etiology, diagnostic tools, host defense mechanisms, and treatments. Additionally, future research avenues are highlighted, which are also relevant for other coral diseases. As SCTLD continues to spread, collaborative efforts are necessary to develop effective strategies for mitigating its impacts on critical coral reef ecosystems. These collaborative efforts need to include researchers from diverse backgrounds and underrepresented groups to provide additional perspectives for a disease that requires creative and urgent solutions.

Microbiome changes in Sinularia spp. soft corals relative to health status

Abstract:Coral reefs are globally important ecosystems with high species diversity at both the macro and micro scales. In recent years, coral reefs have been heavily impacted by anthropogenic and natural stressors, including emerging diseases. Many of these diseases have been identified in reef-building corals, but other invertebrate taxa, such as soft corals, are increasingly at risk. This study focuses on a hybrid species complex within soft corals of the genus Sinularia , which dominate the shallow reefs of Guam, and the broader Indo-Pacific. These soft corals exhibit varying levels of disease susceptibility to Sinularia tissue loss disease (STLD), a chronic wasting disease. In the current study, we used next-generation amplicon sequencing of prokaryotic and eukaryotic communities within these soft corals to characterize their microbiomes, and develop a better understanding of the etiology of STLD. There were differences in specific ASVs across the microbiomes of healthy colonies of Sinularia maxima, Sinularia polydactyla and their hybrid ( S. maxima x S. polydactyla) . There was also a decline in the relative abundance of putatively beneficial symbionts (Symbiodinaceae and Endozoicomonas ) in STLD-affected soft corals, but no consistent shifts towards a specific microbial community associated with STLD. The soft coral microbiomes also contained a high relative abundance of ASVs typically associated with terrestrial runoff. Our results suggest that the STLD phenotype may be due to a combination of factors, including infection by a yet unknown etiologic agent, shifts in putatively beneficial symbionts, and anthropogenic impacts on this shallow nearshore reef.

Abstract:Ciliate parasites can lead to great economic losses to aquaculture. Among them facultative parasites have not emerged yet as important pathogens. In this study, the facultative ciliate parasite Chilodonella uncinata was isolated from the gills of goldfish. Its morphological characters were investigated by observations of both living cells and specimens stained with silver nitrate and protargol methods. Sequence alignments revealed that the present ITS gene sequences were consistent with C. uncinata strain ATCC 50194 and the SSU rDNA gene sequences were similar to the sequences of existing C. uncinata in NCBI website with 99.28% of similarity. Using phylogenetic analyses based on SSU rDNA gene sequences, ciliates collected in this study were confirmed to be C. uncinata. To further validate the infectivity of C. uncinata, an infection model of goldfish was established by adding the C. uncinata isolates from cultures to the goldfish's gills. Microscopic examination and pathological analysis were also used to identify the success establishment of the model, which have shown the presence of C. uncinata, infiltration of inflammatory cells and epithelial hyperplasia on gill tissues. This model has the potential to be used in subsequent diagnostic test and drug screening, which may be beneficial to control chilodonellosis infections in aquaculture. Moreover, this is the first record that C. uncinata demonstrated to be parasitic to fish.
Caryospora-Like Coccidia Infecting Green Turtles (Chelonia mydas): An Emerging Disease With Evidence of Interoceanic Dissemination

Abstract:Protozoa morphologically consistent with Caryospora sp. are one of the few pathogens associated with episodic mass mortality events involving free-ranging sea turtles. Parasitism of green turtles (Chelonia mydas) by these coccidia and associated mortality was first reported in maricultured turtles in the Caribbean during the 1970s. Years later, epizootics affecting wild green turtles in Australia occurred in 1991 and 2014. The first clinical cases of Caryospora-like infections reported elsewhere in free-ranging turtles were from the southeastern US in 2012. Following these initial individual cases in this region, we documented an epizootic and mass mortality of green turtles along the Atlantic coast of southern Florida from November 2014 through April 2015 and continued to detect additional, sporadic cases in the southeastern US in subsequent years. No cases of coccidial disease were recorded in the southeastern US prior to 2012 despite clinical evaluation and necropsy of stranded sea turtles in this region since the 1980s, suggesting that the frequency of clinical coccidiosis has increased here. Moreover, we also recorded the first stranding associated with infection by a Caryospora-like organism in Hawai'i in 2018. To further characterize the coccidia, we sequenced part of the 18S ribosomal and mitochondrial cytochrome oxidase I genes of coccidia collected from 62 green turtles found in the southeastern US and from one green turtle found in Hawai'i. We also sequenced the ribosomal internal transcribed spacer regions from selected cases and compared all results with those obtained from Caryospora-like coccidia collected from green turtles found in Australia. Eight distinct genotypes were represented in green turtles from the southeastern US. One genotype predominated and was identical to that of coccidia collected from the green turtle found in Hawai'i. We also found a coccidian genotype in green turtles from Florida and Australia with identical 18S and mitochondrial sequences, and only slight inter-regional differences in the internal transcribed spacer 2. We found no evidence of geographical structuring based on phylogenetic analysis. Low genetic variability among the coccidia found in green turtle populations with minimal natural connectivity suggests recent interoceanic dissemination of these parasites, which could pose a risk to sea turtle populations.

Mitigation of Vibrio coralliilyticus -induced coral bleaching through bacterial dysbiosis prevention by Ruegeria profundi

Abstract:ABSTRACT Vibrio species are prevalent in ocean ecosystems, particularly Vibrio coralliilyticus , and pose a threat to corals and other marine organisms under global warming conditions. While microbiota manipulation is considered for coral disease management, understanding the role of commensal bacteria in stress resilience remains limited. Here, a single bacterial species ( Ruegeria profundi ) rather than a consortium of native was used to combat pathogenic V. coralliilyticus and protect corals from bleaching. R. profundi showed therapeutic activity in vivo , preventing a significant reduction in bacterial diversity in bleached corals. Notably, the structure of the bacterial community differed significantly among all the groups. In addition, compared with the bleached corals caused by V. coralliilyticus , the network analysis revealed that complex interactions and positive correlations in the bacterial community of the R. profundi protected non-bleached corals, indicating R. profundi ’s role in fostering synergistic associations. Many genera of bacteria significantly increased in abundance during V. coralliilyticus infection, including Vibrio , Alteromonas , Amphritea , and Nautella , contributing to the pathogenicity of the bacterial community. However, R. profundi effectively countered the proliferation of these genera, promoting potential probiotic Endozoicomonas and other taxa, while reducing the abundance of betaine lipids and the type VI section system of the bacterial community. These changes ultimately influenced the interactive relationships among symbionts and demonstrated that probiotic R. profundi intervention can modulate coral-associated bacterial community, alleviate pathogenic-induced dysbiosis, and preserve coral health. These findings elucidated the relationship between the behavior of the coral-associated bacterial community and the occurrence of pathological coral bleaching. IMPORTANCE Changes in the global climate and marine environment can influence coral host and pathogen repartition which refers to an increased likelihood of pathogen infection in hosts. The risk of Vibrio coralliilyticus -induced coral disease is significantly heightened, primarily due to its thermos-dependent expression of virulent and populations. This study investigates how coral-associated bacterial communities respond to bleaching induced by V. coralliilyticus . Our findings demonstrate that Ruegeria profundi exhibits clear evidence of defense against pathogenic bacterial infection, contributing to the maintenance of host health and symbiont homeostasis. This observation suggests that bacterial pathogens could cause dysbiosis in coral holobionts. Probiotic bacteria display an essential capability in restructuring and manipulating coral-associated bacterial communities. This restructuring effectively reduces bacterial community virulence and enhances the pathogenic resistance of holobionts. The study provides valuable insights into the correlation between the health status of corals and how coral-associated bacterial communities may respond to both pathogens and probiotics.

Abstract:Studies of drug-membrane interactions witness an ever-growing interest, as penetration, accumulation, and positioning of drugs play a crucial role in drug delivery and metabolism in human body. Molecular dynamics simulations complement nicely experimental measurements and provide us with new insight into drug-membrane interactions; however, the quality of the theoretical data dramatically depends on the quality of the force field used. We calculated the free energy profiles of 11 molecules through a model dimyristoylphosphatidylcholine (DMPC) membrane bilayer using five force fields, namely Berger, Slipids, CHARMM36, GAFFlipids, and GROMOS 43A1-S3. For the sake of comparison, we also employed the semicontinuous tool COSMOmic. High correlation was observed between theoretical and experimental partition coefficients (log K). Partition coefficients calculated by all-atomic force fields (Slipids, CHARMM36, and GAFFlipids) and COSMOmic differed by less than 0.75 log units from the experiment and Slipids emerged as the best performing force field. This work provides the following recommendations (i) for a global, systematic and high throughput thermodynamic evaluations (e.g., log K) of drugs COSMOmic is a tool of choice due to low computational costs; (ii) for studies of the hydrophilic molecules CHARMM36 should be considered; and (iii) for studies of more complex systems, taking into account all pros and cons, Slipids is the force field of choice.
Microbiomes of three coral species in the Mexican Caribbean and their shifts associated with the Stony Coral Tissue Loss Disease

Abstract:Stony Coral Tissue Loss Disease (SCTLD) has caused widespread coral mortality in the Caribbean Region. However, how the disease presence alter the microbiome community, their structure, composition, and metabolic functionality is still poorly understood. In this study, we characterized the microbial communities of the tissues of apparently healthy and diseased SCTLD colonies of the species Siderastrea siderea, Orbicella faveolata, and Montastraea cavernosa to explore putative changes related to the presence of SCTLD. Gammaproteobacteria, Alphaproteobacteria, and Bacteroidia were the best represented classes in the healthy tissues of all coral species, and alpha diversity did not show significant differences among the species. The microbial community structure between coral species was significantly different (PERMANOVA: F=3.46, p=0.001), and enriched genera were detected for each species: Vibrio and Photobacterium in S. siderea, Spirochaeta2 and Marivivens in O. faveolata and SAR202_clade and Nitrospira in M. cavernosa. Evidence of SCTLD in the microbial communities was more substantial in S. siderea, where differences in alpha diversity, beta diversity, and functional profiles were observed. In O. faveolata, differences were detected only in the community structure, while M. cavernosa samples showed no significant difference. Several microbial groups were found to have enriched abundances in tissue from SCTLD lesions from S. siderea and O. faveolata, but no dominant bacterial group was detected. Our results contribute to understanding microbial diversity associated with three scleractinian coral species and the shifts in their microbiomes associated with SCTLD in the Mexican Caribbean.

Current Knowledge of Coral Diseases Present Within the Red Sea

Abstract:Disease is defined as a disorder of structure or function in an organism, which produces specific signs or symptoms and is not simply a direct result of physical injury. In scleractinian corals, as in all other organisms, diseased individuals show morphologically distinct lesions, which manifest themselves in alterations of colour, shape, size, or texture. Coral diseases are routinely described by macroscopic signs, with notes on the extent of tissue loss, tissue colour, and exposure of coral skeleton aiding in the nomenclature and diagnosis. However, despite their reported significance in reefs around the world, the aetiologies of the majority of diseases are still unknown and coral epidemiology remains poorly understood. The majority of published literature suggests numerous microorganisms are capable of causing coral disease, including bacteria, viruses, fungi, and protozoa. That said, the initial trigger for disease is often recorded as being abiotic factors such as temperature, UV exposure, pollution (such as heavy metals, and agricultural chemicals) or predation scars, for example. Coral diseases have been shown to be prevalent on both well-managed (for example marine protected areas) and unmanaged reefs alike. Despite the serious negative implications associated with coral disease, not all geographic areas are as well documented as others. For example, diseases affecting corals throughout the Red Sea are one of the least well explored and documented. Here we sum up the current knowledge of the diseases described on the reefs in this area and give a brief background into what is currently known about the cause of such diseases.
Bacterial Metabolic Potential and Micro-Eukaryotes Enriched in Stony Coral Tissue Loss Disease Lesions

Abstract:The epizootic disease outbreak known as stony coral tissue loss disease (SCTLD) is arguably the most devastating coral disease in recorded history. SCTLD emerged off the coast of South Florida in 2014 and has since moved into the Caribbean, resulting in coral mortality rates that have changed reef structure and function. Currently, the cause of SCTLD is unknown, but there is evidence from 16S rRNA gene sequencing and bacterial culture studies that the microbial community plays a role in the progression of SCTLD lesions. In this study, we applied shotgun metagenomics to characterize the potential function of bacteria, as well as the composition of the micro-eukaryotic community, associated with SCTLD lesions. We re-examined samples that were previously analyzed using 16S rRNA gene high-throughput sequencing from four coral species: Stephanocoenia intersepta, Diploria labyrinthiformis, Dichocoenia stokesii , and Meandrina meandrites . For each species, tissue from apparently healthy (AH) corals, and unaffected tissue (DU) and lesion tissue (DL) on diseased corals, were collected from sites within the epidemic zone of SCTLD in the Florida Keys. Within the micro-eukaryotic community, the taxa most prominently enriched in DL compared to AH and DU tissue were members of Ciliophora. We also found that DL samples were relatively more abundant in less energy-efficient pathways like the pentose phosphate pathways. While less energy-efficient processes were identified, there were also relatively higher abundances of nucleotide biosynthesis and peptidoglycan maturation pathways in diseased corals compared to AH, which suggests there was more bacteria growth in diseased colonies. In addition, we generated 16 metagenome-assembled genomes (MAGs) belonging to the orders Pseudomonadales, Beggiatoales, Rhodobacterales, Rhizobiales, Rs-D84, Flavobacteriales, and Campylobacterales, and all MAGs were enriched in DL samples compared to AH samples. Across all MAGs there were antibiotic resistance genes that may have implications for the treatment of SCTLD with antibiotics. We also identified genes and pathways linked to virulence, such as nucleotide biosynthesis, succinate dehydrogenase, ureases, nickel/iron transporters, Type-1 secretion system, and metalloproteases. Some of these enzymes/pathways have been previously targeted in the treatment of other bacterial diseases and they may be of interest to mitigate SCTLD lesion progression.

Identification of quorum sensing-regulated Vibrio fortis as potential pathogenic bacteria for coral bleaching and the effects on the microbial shift

Abstract:Coastal pollution, global warming, ocean acidification, and other reasons lead to the imbalance of the coral reef ecosystem, resulting in the increasingly serious problem of coral degradation. Coral bleaching is often accompanied by structural abnormalities of coral symbiotic microbiota, among which Vibrio is highly concerned. In this study, Vibrio fortis S10-1 (MCCC 1H00104), isolated from sea cucumber, was used for the bacterial infection on coral Seriatopora guttatus and Pocillopora damicornis . The infection of S10-1 led to coral bleaching and a significant reduction of photosynthetic function in coral holobiont, and the pathogenicity of V. fortis was regulated by quorum sensing. Meanwhile, Vibrio infection also caused a shift of coral symbiotic microbial community, with significantly increased abundant Proteobacteria and Actinobacteria and significantly reduced abundant Firmicutes; on genus level, the abundance of Bacillus decreased significantly and the abundance of Rhodococcus , Ralstonia , and Burkholderia–Caballeronia–Paraburkholderia increased significantly; S10-1 infection also significantly impacted the water quality in the micro-ecosystem. In contrast, S10-1 infection showed less effect on the microbial community of the live stone, which reflected that the microbes in the epiphytic environment of the live stone might have a stronger ability of self-regulation; the algal symbionts mainly consisted of Cladocopium sp. and showed no significant effect by the Vibrio infection. This study verified that V. fortis is the primary pathogenic bacterium causing coral bleaching, revealed changes in the microbial community caused by its infection, provided strong evidence for the "bacterial bleaching" hypothesis, and provided an experimental experience for the exploration of the interaction mechanism among microbial communities, especially coral-associated Vibrio in the coral ecosystem, and potential probiotic strategy or QS regulation on further coral disease control.

Abstract:Endemic to the Brazilian fauna, the brain coral Mussismilia hispida is the second most widespread zooxanthellate coral of the South-west Atlantic and, most importantly, is within the main reef-building species of the region. Counterintuitively, M. hispida has one of its most abundant populations near its southernmost distributional limit, the Alcatrazes Archipelago off the coast of São Paulo State. On this archipelago, colonies thrive from 2 to over 20 m deep, and in some localities, M. hispida covers more than 50% of the rocky shore. Although more resistant to bleaching than other coral species, a capacity enhanced by colder water resurgence around the archipelago, signs of a coral disease affecting some colonies were recorded in 2019, simultaneously with a severe bleaching event. Diseased corals had tissue loss and a distinct white lesion on the corals' tissue, suggesting that it may be a White Plague disease. Using 16S rRNA metabarcoding, we compared the microbial community associated with the part of the colonies presenting signs of disease to those apparently healthy. Results indicate that the microbiota genera from healthy and diseased colony portions are highly variable, suggesting community dysbiosis and alterations in the metabolic pathways of the microbiome. While it was not possible to identify a pathogen or a pathogenic consortium associated with the disease, the overall microbial signature, characterized by the presence of Roseimarinus , Carboxylicivirga , Tepidibacter , Vallitalea , and Halodesulfovibrio , is similar to that found in diseased Caribbean massive corals.
