Abstract:Background: Coral diseases are significant drivers of global coral reef degradation, with pathogens dominated by Vibrio coralliilyticus playing a prominent role in the development of coral diseases. Coral phenotype, symbiotic microbial communities, and host transcriptional regulation have been well-established as factors involved in determining coral disease resistance, but the underlying mechanisms remain incompletely understood. Methods: This study employs high-throughput sequencing to analyse the symbiotic microbial and transcriptional response of the hosts in order to evaluate the disease resistance of Acropora valida and Turbinaria peltata exposed to Vibrio coralliilyticus. Results: A. valida exhibited pronounced bleaching and tissue loss within 7 h of pathogen infection, whereas T. peltata showed no signs of disease throughout the experiment. Microbial diversity analyses revealed that T. peltata had a more flexible microbial community and a higher relative abundance of potential beneficial bacteria compared to A. valida. Although Vibrio inoculation resulted in a more significant decrease in the Symbiodiniaceae density of A. valida compared to that of T. peltata, it did not lead to recombination of the coral host and Symbiodiniaceae in either coral species. RNA-seq analysis revealed that the interspecific differences in the transcriptional regulation of hosts after Vibrio inoculation. Differentially expressed genes in A. valida were mainly enriched in the pathways associated with energy supply and immune response, such as G protein-coupled receptor signaling, toll-like receptor signaling, regulation of TOR signaling, while these genes in T. peltata were mainly involved in the pathway related to immune homeostasis and ion transport, such as JAK-STAT signaling pathway and regulation of ion transport. Conclusions: Pathogenic challenges elicit different microbial and transcriptional shifts across coral species. This study offers novel insights into molecular mechanisms of coral resistance to disease.

Abstract:Variability in coral hosts susceptibility to Vibrio coralliilyticus is well-documented; however, the comprehensive understanding of tolerance of response to pathogen among coral species is lacked. Herein, we investigated the microbial communities and transcriptome dynamics of two corals in response to Vibrio coralliilyticus . Favites halicora displayed greater resistance to Vibrio coralliilyticus challenge than Pocillopora damicornis . Furthermore, the relative abundances of Flavobacteriaceae , Vibrionacea, Rhodobacteraceae, and Roseobacteraceae increased significantly in Favites halicora following pathogen stress, whereas that of Akkermansiaceae increased significantly in Pocillopora damicornis, leading to bacterial community imbalance. In contrast to the previous results, pathogen infection did not have much effect on the community structures of Symbiodiniaceae and fungi, but led to a decrease in the density of Symbiodiniaceae. Transcriptome analysis indicated that Vibrio infection triggered a coral immune response, resulting in higher expression of immune-related genes, which appeared to have higher transcriptional plasticity in Favites halicora than in Pocillopora damicornis . Specifically, the upregulated genes of Favites halicora were predominantly involved in the apoptosis pathway, whereas Pocillopora damicornis were significantly enriched in the nucleotide excision repair and base excision repair pathways. These findings suggest that coral holobionts activate different mechanisms across species in response to pathogens through shifts in microbial communities and transcriptomes, which provides novel insight into assessing the future coral assemblages suffering from disease outbreaks.

Abstract:Algal symbionts of corals can influence host stress resistance; for example, in the Pacific Ocean, whereas Cladocopium (C-type) is generally dominant in corals, Durusdinium (D-type) is found in more heat-resistant corals. Thus, the presence of D-type symbiont likely increases coral heat tolerance, and this symbiotic relationship potentially provides a hint to increase the stress tolerance of coral-algal symbioses. In this study, transcriptome profiles of Cladocopium- and Durusdinium-harboring Acropora solitaryensis (C-coral and D-coral, respectively) and algal photosystem functioning (F v /F m ) under bleaching conditions (high temperature and light stress) were compared. Stress treatment caused algal photoinhibition that the F v /F m value of Symbiodiniaceae was immediately reduced. The transcriptome analysis of corals revealed that genes involved in the following processes were detected: endoplasmic reticulum (ER) stress, mitophagy, apoptosis, endocytosis, metabolic processes (acetyl-CoA, chitin metabolic processes, etc.), and the PI3K-AKT pathway were upregulated, while DNA replication and the calcium signaling pathway were downregulated in both C- and D-corals. These results suggest that unrepaired DNA and protein damages were accumulated in corals under high temperature and light stress. Additionally, some differentially expressed genes (DEGs) were specific to C- or D-corals, which includes genes involved in transient receptor potential (TRP) channels and vitamin B metabolic processes. Algal transcriptome analysis showed the increased expression of gene encoding photosystem and molecular chaperone especially in D-type symbiont. The transcriptome data imply a possible difference in the stress reactions on C-type and D-type symbionts. The results reveal the basic process of coral heat/light stress response and symbiont-type-specific coral transcriptional responses, which provides a perspective on the mechanisms that cause differences in coral stress tolerance.

Abstract:Few studies have holistically examined successive changes in coral holobionts in response to increased temperatures. Here, responses of the coral host Pocillopora damicornis , its Symbiodiniaceae symbionts, and associated bacteria to increased water temperatures were investigated. High temperatures induced bleaching, but no coral mortality was observed. Transcriptome analyses showed that P. damicornis responded more quickly to elevated temperatures than its algal symbionts. Numerous genes putatively associated with apoptosis, exocytosis, and autophagy were up‐regulated in P. damicornis , suggesting that Symbiodiniaceae can be eliminated or expelled through these mechanisms when P. damicornis experiences heat stress. Furthermore, apoptosis in P. damicornis is presumably induced through tumor necrosis factor and p53 signaling and caspase pathways. The relative abundances of several coral disease‐associated bacteria increased at 32°C which may affect immune responses in heat‐stressed corals and potentially accelerates loss of algal symbionts. Additionally, consistency of Symbiodiniaceae community structures under heat stress suggests non‐selective loss of Symbiodiniaceae. Abstract:Abstract A worldwide increase in the prevalence of coral diseases and mortality has been linked to ocean warming due to changes in coral-associated bacterial communities, pathogen virulence, and immune system function. In the Mediterranean basin, the worrying upward temperature trend has already caused recurrent mass mortality events in recent decades. To evaluate how elevated seawater temperatures affect the immune response of a thermophilic coral species, colonies of Astroides calycularis were exposed to environmental (23 °C) or elevated (28 °C) temperatures, and subsequently challenged with bacterial lipopolysaccharides (LPS). Using immunolabeling with specific antibodies, we detected the production of Toll-like receptor 4 (TLR4) and nuclear factor kappa B (NF-kB), molecules involved in coral immune responses, and heat shock protein 70 (HSP70) activity, involved in general responses to thermal stress. A histological approach allowed us to characterize the tissue sites of activation (epithelium and/or gastroderm) under different experimental conditions. The activity patterns of the examined markers after 6 h of LPS stimulation revealed an up-modulation at environmental temperature. Under warmer conditions plus LPS-challenge, TLR4-NF-kB activation was almost completely suppressed, while constituent elevated values were recorded under thermal stress only. An HSP70 up-regulation appeared in both treatments at elevated temperature, with a significantly higher activation in LPS-challenge colonies. Such an approach is useful for further understanding the molecular pathogen-defense mechanisms in corals in order to disentangle the complex interactive effects on the health of these ecologically relevant organisms related to global climate change.

Abstract:SUMMARY Disease epizootics are increasing with climatic shifts, yet within each system only a subset of species are identified as the most vulnerable. Understanding ecological immunology patterns as well as environmental influences on immune defenses will provide insight into the persistence of a functional system through adverse conditions. Amongst the most threatened ecosystems are coral reefs, with coral disease epizootics and thermal stress jeopardizing their survival. Immune defenses were investigated within three Caribbean corals, Montastraea faveolata, Stephanocoenia intersepta and Porites astreoides, which represent a range of disease and bleaching susceptibilities. Levels of several immune parameters were measured in response to elevated water temperature and the presence of a commercial pathogen-associated molecular pattern (PAMP) – lipopolysaccharide (LPS) – as an elicitor of the innate immune response. Immune parameters included prophenoloxidase (PPO) activity, melanin concentration, bactericidal activity, the antioxidants peroxidase and catalase, and fluorescent protein (FP) concentration. LPS induced an immune response in all three corals, although each species responded differently to the experimental treatments. For example, M. faveolata, a disease-susceptible species, experienced significant decreases in bactericidal activity and melanin concentration after exposure to LPS and elevated temperature alone. Porites astreoides, a disease-resistant species, showed increased levels of enzymatic antioxidants upon exposure to LPS independently and increased PPO activity in response to the combination of LPS and elevated water temperature. This study demonstrates the ability of reef-building corals to induce immune responses in the presence of PAMPs, indicating activation of PAMP receptors and the transduction of appropriate signals leading to immune effector responses. Furthermore, these data address the emerging field of ecological immunology by highlighting interspecific differences in immunity and immunocompetences among Caribbean corals, which are reflected in their life-history characteristics, disease susceptibilities and bleaching-induced mortality.

Abstract:Anthropogenic climate change is warming sea water worldwide, pushing the limits of tolerance for marine organisms and driving a decline in biodiversity. The risk of thermal anomalies has increased particularly in the Mediterranean region over the last 30 yrs, where intense warming has been identified as one of the main stressors in coastal regions. To determine the influence of warmer conditions on the immunity of an endemic Mediterranean coral species, different immune activity parameters were compared in response to elevated temperature (~ 28 °C) and the presence of a pathogen-associated molecular pattern— Escherichia coli lipopolysaccharide (LPS)—as an elicitor of the innate immune response of Astroides calycularis . Immune parameters, which included phenoloxidase-like, glutathione peroxidase, lysozyme-like, alkaline phosphatase, and esterase enzyme activity, were measured over time after LPS balneation (0-, 12-, 48-, and 120 h time point). All five enzymes demonstrated constant values under environmental conditions (~ 23 °C), indicating a constituent activity. LPS at environmental temperature induced significant upregulation immediately after exposure (0 h-time point), demonstrating an immune response to the pathogen elicitor. Under warmer conditions (~ 28 °C), constituent values increased over time, indicating a shift in the immune strategy to maintain homeostasis. However, warmer sea water, within the summer range experienced by this coral species, impaired the immune response to LPS, delaying it over time. These changes in immune strategy indicate that temperature affects coral immunity and, in thermophilic A. calycularis , results in an energy trade-off that could maintain its health-state through suboptimal conditions during multiple perturbations, such as summertime diseases.

Interspecies differences in the transcriptome response of corals to acute heat stress

Abstract:Rising sea surface temperatures threaten the survival of corals worldwide, with coral bleaching events becoming more commonplace. However, different coral species are known to exhibit variable levels of susceptibility to thermal stress events. To elucidate genetic mechanisms that may underlie these differences, we compared the gene complement of four coral species, Favites colemani, Montipora digitata, Acropora digitifera, and Seriatopora caliendrum, that were previously demonstrated to have differing responses to acute thermal stress. We found that more tolerant species, like F. colemani and M. digitata, possess a greater abundance of antioxidant protein families and chaperones. Under acute thermal stress conditions, only S. caliendrum showed a significant bleaching response, which was accompanied by activation of DNA damage response network and drastic upregulation of stress response genes (SRGs). This suggests that differences in SRG complement, as well as the mechanisms that control SRG expression response, contribute to the ability of corals to maintain stable physiological functions that is required to survive shifts in seawater temperature.

Warmer Water Affects Immunity of a Tolerant Reef Coral

Abstract:Corals are multipartite sedentary organisms, which have evolved complex, physiological networks in order to survive perturbations and environmental fluctuations. However, climate change is warming tropical waters, pushing the limits of coral tolerance and driving global declines. Coral susceptibility to thermal anomalies is variable among species and through time, and directly relates to constituent immunity. Constituent immunity refers to immune activities required to ensure homeostasis, whereas an immune response is acutely heightened immune activity to a perturbation. Understanding the mechanisms behind coral health, and sustained health through adverse conditions, is increasingly important for establishing effective reef conservation and restoration projects. However, most experimental studies of coral health use species that are highly susceptible to thermal events, potentially skewing our understanding. To determine the influence of warmer water on immunity, activities of key coral immune pathways and an antioxidant were compared under ambient (27°C) and warmer water (32°C), and between injured and uninjured (control) branches of the tolerant reef coral Porites cylindrica. Three types of phenoloxidase, mono-phenoloxidase, ortho-diphenoloxidase and para-diphenloxidase, indicative of two melanin synthesis pathways (the tyrosinase and laccase-type), and peroxidase were measured at 0 (control), 1, 6, 24, 48, and 168 h post-injury. All four enzymes demonstrated consistent levels of activity under ambient conditions (27°C), indicating constituent immunity. Upon injury at ambient temperature, all enzyme activities were significantly higher 1 h post-injury as compared to uninjured controls, demonstrating a comprehensive immune response to tissue disruption. Under warmer water, constituent immunity increased through time indicative of immune modulation to maintain homeostasis. However, warmer water, within the non-bleaching summer range, suppressed the immune response to injury, delaying it by 24 h. Therefore, upon the environmental cue of warmer water, the tolerant coral P. cylindrica may divert resources away from immune responses (immunosuppression) while enhancing constituent immunity (immune modulation) so as to maintain health through sub-optimal conditions. These changes in immunity with warmer water demonstrate that temperature affects coral immunity and, for this tolerant coral, triggers immune-modulation that may provide cross-tolerance to perturbations more frequent in summer months, such as bleaching and disease.

Abstract:The increasing threat of ocean warming has led to the more frequent endangerment of coral reefs, including the heat-tolerant Pavona decussata . To shed light on the molecular mechanisms involved in the response of coral to ocean warming, we investigated the gene expression profiles of P. decussata after natural thermal stress. Using PacBio Sequel II sequencing technology, we obtained relatively complete transcriptome data for P. decussata and then analyzed its gene expression quantitatively with Illumina RNA-seq technology. We acquired information on gene function, structure, and expression profile from coral host and zooxanthellae. Analysis of Illumina sequencing data revealed that unbleached coral host might rely on the active utilization of amino acids to maintain a stable living condition based on the tricarboxylic acid cycle under high temperature stress, and that zooxanthellae might benefit from ammonium produced by coral host. Moreover, the downregulation of unbleached coral host gene expression in innate immune pathways centered on the transcription factors that heat shock factor and nuclear factor (NF)-κB, as well as the tyrosine kinase pathway, might be crucial for maintaining the equilibrium of the zooxanthellae under thermal stress. Thus, the differences in these molecular mechanisms could determine, to some extent, whether coral host can maintain a symbiotic relationship with algae under heat stress. This study elucidated the molecular mechanisms underlying differences in thermal tolerance within P. decussata species and supported further theoretical basis in coral molecular biology and ecological conservation, which enhance our comprehension of coral responses to future climate change.

Abstract:Under homoeostatic conditions, the relationship between the coral Pocillopora damicornis and Vibrio coralliilyticus is commensal. An increase in temperature, or in the abundance of V. coralliilyticus, can turn this association pathogenic, causing tissue lysis, expulsion of the corals’ symbiotic algae (genus Symbiodinium), and eventually coral death. Using a combination of microfluidics, fluorescence microscopy, stable isotopes, electron microscopy and NanoSIMS isotopic imaging, we provide insights into the onset and progression of V. coralliilyticus infection in the daytime and at night, at the tissue and (sub-)cellular level. The objective of our study was to connect the macro-scale behavioural response of the coral to the micro-scale nutritional interactions that occur between the host and its symbiont. In the daytime, polyps enhanced their mucus production, and actively spewed pathogens. Vibrio infection primarily resulted in the formation of tissue lesions in the coenosarc. NanoSIMS analysis revealed infection reduced 13C-assimilation in Symbiodinium, but increased 13C-assimilation in the host. In the night incubations, no mucus spewing was observed, and a mucus film was formed on the coral surface. Vibrio inoculation and infection at night showed reduced 13C-turnover in Symbiodinium, but did not impact host 13C-turnover. Our results show that both the nutritional interactions that occur between the two symbiotic partners and the behavioural response of the host organism play key roles in determining the progression and severity of host-pathogen interactions. More generally, our approach provides a new means of studying interactions (ranging from behavioural to metabolic scales) between partners involved in complex holobiont systems, under both homoeostatic and pathogenic conditions.

Abstract:Coral reefs are essential for marine ecology and biodiversity. Global climate change has resulted in severe coral reef degradation, partly via coral bleaching, which is caused by rising sea temperatures and solar light intensity. In this study, we examined the impact of strong light (300 µmol.m-2.s-1) and high temperature (33°C) on the growth, immunity, and gene expression of Galaxea fascicularis. Strong light caused coral bleaching in the absence of high sea temperatures, while no obvious bleaching was observed under high temperature alone. The effect of strong light on calcification rate of G. fascicularis is significantly weaker than that of high temperature. Both strong light and high temperatures significantly affected the immune enzyme activity of G. fascicularis symbionts, with the former having a strong effect on their photosystem. Temperature affected the digestive system, replication and repair, and cell growth and death of coral hosts, as indicated by transcriptomics analysis. These results provide a valuable for strategies to mitigate coral bleaching. TEASER: We explored the effects of strong light exposure and high temperature on coral reefs and their symbiont algae.

Abstract:BackgroundCorals are capable of launching diverse immune defenses at the site of direct contact with pathogens, but the molecular mechanisms of this activity and the colony-wide effects of such stressors remain poorly understood. Here we compared gene expression profiles in eight healthy Acropora hyacinthus colonies against eight colonies exhibiting tissue loss commonly associated with white syndromes, all collected from a natural reef environment near Palau. Two types of tissues were sampled from diseased corals: visibly affected and apparently healthy.ResultsTag-based RNA-Seq followed by weighted gene co-expression network analysis identified groups of co-regulated differentially expressed genes between all health states (disease lesion, apparently healthy tissues of diseased colonies, and fully healthy). Differences between healthy and diseased tissues indicate activation of several innate immunity and tissue repair pathways accompanied by reduced calcification and the switch towards metabolic reliance on stored lipids. Unaffected parts of diseased colonies, although displaying a trend towards these changes, were not significantly different from fully healthy samples. Still, network analysis identified a group of genes, suggestive of altered immunity state, that were specifically up-regulated in unaffected parts of diseased colonies.ConclusionsSimilarity of fully healthy samples to apparently healthy parts of diseased colonies indicates that systemic effects of white syndromes on A. hyacinthus are weak, which implies that the coral colony is largely able to sustain its physiological performance despite disease. The genes specifically up-regulated in unaffected parts of diseased colonies, instead of being the consequence of disease, might be related to the originally higher susceptibility of these colonies to naturally occurring white syndromes.

The role of gene expression and symbiosis in reef-building coral acquired heat tolerance

Abstract:Abstract Predicting how reef-building corals will respond to accelerating ocean warming caused by climate change requires knowledge of how acclimation and symbiosis modulate heat tolerance in coral early life-history stages. We assayed transcriptional responses to heat in larvae and juveniles of 11 reproductive crosses of Acropora tenuis colonies along the Great Barrier Reef. Larvae produced from the warmest reef had the highest heat tolerance, although gene expression responses to heat were largely conserved by cross identity. Juvenile transcriptional responses were driven strongly by symbiosis – when in symbiosis with heat-evolved Symbiodiniaceae, hosts displayed intermediate expression between its progenitor Cladocopium and the more stress tolerant Durusdinium , indicating the acquisition of tolerance is a conserved evolutionary process in symbionts. Heat-evolved Symbiodiniaceae facilitated juvenile survival under heat stress, although host transcriptional responses to heat were positively correlated among those hosting different genera of Symbiodiniaceae. These findings reveal the relative contribution of parental environmental history as well as symbiosis establishment in coral molecular responses to heat in early life-history stages.

Changes in DNA methylation in response to heat stress and symbiotic conditions in coral primary polyps

Abstract:The symbiotic relationship between scleractinian corals and dinoflagellates is a cornerstone of coral reef ecosystems. Various mechanisms underlying the adaptability of corals to environmental changes have been studied, but the effects of epigenetic mechanisms (i.e. DNA methylation, DNAm) on heat stress and the symbiotic conditions of corals remain unclear. Moreover, the DNAm patterns relevant to the primary polyp life stage of corals are not known. Hence, we conducted an aquarium factorial experiment to evaluate the effects of symbiotic conditions and temperatures on global DNAm rates in primary Acropora digitifera polyps. The tested conditions included symbiotic (infected with Symbiodinium ITS2 type A3) and aposymbiotic (without Symbiodinium ) groups, at either heated (32°C) or ambient (27°C) temperatures. The results indicated that the presence of Symbiodinium within primary A. digitifera polyps significantly decreased basal DNAm rates of the coral holobiont. In addition, the presence of Symbiodinium in the early primary polyp stage increased DNAm in the coral holobiont when exposed to heat stress. These results suggest that the symbiotic relationship between host corals and Symbiodinium is one of the major factors related to global DNAm rate changes in primary A. digitifera polyps.

Abstract:Coral reefs are extremely vulnerable to ocean warming, which triggers coral bleaching-the loss of endosymbiotic microalgae (Symbiodiniaceae) from coral tissues, often leading to death. To enhance coral climate resilience, the symbiont, Cladocopium proliferum was experimentally evolved for >10 years under elevated temperatures resulting in increased heat tolerance. Bacterial 16S rRNA gene metabarcoding showed the composition of intra- and extracellular bacterial communities of heat-evolved strains was significantly different from that of wild-type strains, suggesting bacteria responded to elevated temperatures, and may even play a role in C. proliferum thermal tolerance. To assess whether microbiome transplantation could enhance heat tolerance of the sensitive wild-type C. proliferum, we transplanted bacterial communities from heat-evolved to the wild-type strain and subjected it to acute heat stress. Microbiome transplantation resulted in the incorporation of only 30 low-abundance strains into the microbiome of wild-type cultures, while the relative abundance of 14 pre-existing strains doubled in inoculated versus uninoculated samples. Inoculation with either wild-type or heat-evolved bacterial communities boosted C. proliferum growth, although no difference in heat tolerance was observed between the two inoculation treatments. This study provides evidence that Symbiodiniaceae-associated bacterial communities respond to heat selection and may contribute to coral adaptation to climate change.

Abstract:BackgroundGlobal warming has triggered an increase in the prevalence and severity of coral disease, yet little is known about coral/pathogen interactions in the early stages of infection. The point of entry of the pathogen and the route that they take once inside the polyp is currently unknown, as is the coral’s capacity to respond to infection. To address these questions, we developed a novel method that combines stable isotope labelling and microfluidics with transmission electron microscopy (TEM) and nanoscale secondary ion mass spectrometry (NanoSIMS), to monitor the infection process between Pocillopora damicornis and Vibrio coralliilyticus under elevated temperature.ResultsThree coral fragments were inoculated with 15N-labeled V. coralliilyticus and then fixed at 2.5, 6 and 22 h post-inoculation (hpi) according to the virulence of the infection. Correlative TEM/NanoSIMS imaging was subsequently used to visualize the penetration and dispersal of V. coralliilyticus and their degradation or secretion products. Most of the V. coralliilyticus cells we observed were located in the oral epidermis of the fragment that experienced the most virulent infection (2.5 hpi). In some cases, these bacteria were enclosed within electron dense host-derived intracellular vesicles. 15N-enriched pathogen-derived breakdown products were visible in all tissue layers of the coral polyp (oral epidermis, oral gastrodermis, aboral gastrodermis), at all time points, although the relative 15N-enrichment depended on the time at which the corals were fixed. Tissues in the mesentery filaments had the highest density of 15N-enriched hotspots, suggesting these tissues act as a “collection and digestion” site for pathogenic bacteria. Closer examination of the sub-cellular structures associated with these 15N-hotspots revealed these to be host phagosomal and secretory cells/vesicles.ConclusionsThis study provides a novel method for tracking bacterial infection dynamics at the levels of the tissue and single cell and takes the first steps towards understanding the complexities of infection at the microscale, which is a crucial step towards understanding how corals will fare under global warming.

Abstract:Abstract As coral reefs struggle to survive under climate change, it is crucial to know whether they have the capacity to withstand changing conditions, particularly increasing seawater temperatures. Thermal tolerance requires the integrative response of the different components of the coral holobiont (coral host, algal photosymbiont, and associated microbiome). Here, using a controlled thermal stress experiment across three divergent Caribbean coral species, we attempt to dissect holobiont member metatranscriptome responses from coral taxa with different sensitivities to heat stress and use phylogenetic ANOVA to study the evolution of gene expression adaptation. We show that coral response to heat stress is a complex trait derived from multiple interactions among holobiont members. We identify host and photosymbiont genes that exhibit lineage-specific expression level adaptation and uncover potential roles for bacterial associates in supplementing the metabolic needs of the coral-photosymbiont duo during heat stress. Our results stress the importance of integrative and comparative approaches across a wide range of species to better understand coral survival under the predicted rise in sea surface temperatures.

Abstract:Endozoicomonas , a core bacterial group in corals, may also be a coral symbiont. Endozoicomonas communities often decrease rapidly in corals under heat stress. However, how the bacteria respond to changes in temperature and coral host during heat stress is unknown. Here, we employed the cultivable, dominant species E. montiporae as a working organism to explore how Endozoicomonas responds to heat stress. We designed two experiments to clarify the extent to which E. montiporae is influenced by temperature and coral host. We detected differentially expressed protein (DEP) profiles in this bacterium at 31 and 33°C compared to 25°C by tandem mass tags-based quantitative proteome analysis. Fifty DEPs, including many heat shock proteins, were detected when the temperature changed. The expression of antioxidant defense proteins and key pyruvate synthase proteins decreased, suggesting that E. montiporae were in a physiology of stress at 33°C. Furthermore, some proteins were differentially expressed because of the heat-stress-treated coral lysate specifically, suggesting that not only heat but also heat-induced host factors can affect the protein expression of the bacterium. This study provides an in-depth analysis of how the molecular mechanisms of Endozoicomonas are affected by heat stress and coral host.

Abstract:Coral reef health is tightly connected to the coral holobiont, which is the association between the coral animal and a diverse microbiome functioning as a unit. The coral holobiont depends on key services such as nitrogen and sulfur cycling mediated by the associated bacteria. However, these microbial services may be impaired in response to environmental changes, such as thermal stress. A perturbed microbiome may lead to coral bleaching and disease outbreaks, which have caused an unprecedented loss in coral cover worldwide, particularly correlated to a warming ocean. The response mechanisms of the coral holobiont under high temperatures are not completely understood, but the associated microbial community is a potential source of acquired heat-tolerance. Here we investigate the effects of increased temperature on the taxonomic and functional profiles of coral surface mucous layer (SML) microbiomes in relationship to coral-algal physiology. We used shotgun metagenomics in an experimental setting to understand the dynamics of microbial taxa and genes in the SML microbiome of the coral Pseudodiploria strigosa under heat treatment. The metagenomes of corals exposed to heat showed high similarity at the level of bacterial genera and functional genes related to nitrogen and sulfur metabolism and stress response. The coral SML microbiome responded to heat with an increase in the relative abundance of taxa with probiotic potential, and functional genes for nitrogen and sulfur acquisition. Coral-algal physiology significantly explained the variation in the microbiome at taxonomic and functional levels. These consistent and specific microbial taxa and gene functions that significantly increased in proportional abundance in corals exposed to heat are potentially beneficial to coral health and thermal resistance.
