Abstract:Allelochemicals have been shown to inhibit cyanobacterial blooms for several years. In view of the disadvantages of "direct-added" mode, natural and pollution-free tea polyphenolic allelochemicals with good inhibitory effect on cyanobacteria were selected to prepare sustained-release particles by microcapsule technology. Results showed that the encapsulation efficiency of tea polyphenols sustained-release particles (TPSPs) was 50.6% and the particle size ranged from 700 to 970 nm, which reached the nanoscale under optimum preparation condition. Physical and chemical properties of TPSPs were characterized to prove that tea polyphenols were well encapsulated and the particles had good thermal stability. The optimal dosage of TPSPs was determined to be 0.3 g/L, at which the inhibition rate on Microcystis aeruginosa in logarithmic growth period could be maintained above 95%. Simultaneous decrease in algal density and chlorophyll-a content indicated that the photosynthesis of algal cells was affected leading to cell death. Significant changes of antioxidant enzyme activities suggested that Microcystis aeruginosa 's antioxidant systems had been disrupted. Furthermore, TPSPs increased the concentration of O 2− which led to lipid peroxidation of cell membrane and a subsequent increase in malondialdehyde (MDA) content. Meanwhile, the protein content, nucleic acid content, and electrical conductivity in culture medium rose significantly indicating the cell membrane was irreversibly damaged. This work can provide a basis for the utilization of environmentally friendly algal suppressants.

Abstract:For revealing the mechanism of allelopathic influence on phytoplankton by aquatic macrophytes, the growth and photosynthetic activities of cyanobacteria Microcystis aeruginosa and the chlorophyte Selenastrum capricornutum were investigated when they coexisted with submerged macrophyte Myriophyllum spicatum and were exposed to allelopathic polyphenols: pyrogallic acid (PA), gallic acid (GA), ellagic acid (EA) and (+)-catechin (CA). According to the results of coexistence assays, the non-photochemical quenching (NPQ) and effective quantum efficiency (YII) of M. aeruginosa were affected earlier and more rapidly than the cell density. However, the influence of M. spicatum on S. capricornutum was not found. When the Toxicity Index (TI) was applied to evaluate the combined effects of binary and multiple mixtures of polyphenols, it was found that the four tested polyphenols with the proportion identified in the M. spicatum-cultured solution were observed to present synergistic effect (0.36-0.49) according to the cell density, NPQ and YII of M. aeruginosa. With the combined effects of polyphenols on S. capricornutum, only additive action (0.52-1.62) was found. On the other hand, PA (2.97mgL(-1)), GA (2.65mgL(-1)) caused significant reductions of photosystem II (PSII) and whole electron transport chain activities of M. aeruginosa by 71.43 and 18.37%, 70.95 and 40.77% (P<0.05), respectively, after 24-h exposure, but no inhibition effect was found in S. capricornutum. The dark respiration and photosystem I (PSI) activities of M. aeruginosa were significantly increased by exposure to PA and GA (P<0.05). Nevertheless, EA and CA had no influence on the electron transport activities of the tested organisms. These results indicate that the reduction in photosynthetic activity of M. aeruginosa and the synergistic effect of allelochemicals may be two important causes for the inhibition of undesired phytoplankton by submersed macrophytes in natural aquatic ecosystems, and PSII in cyanobacteria is considered to be one of the target sites attacked by allelopathic polyphenols.
Continuous-release Beads of Natural Allelochemicals for the Long-Term Control of Cyanobacterial Growth: Preparation, Release Dynamics and Inhibitory Effects.

Abstract:The effects of allelochemicals on cyanobacterial blooms have been observed for more than 20 years; however, the use of these compounds, usually involving a "direct-added" mode, has clear disadvantages, such as a short activity period or temporarily excessive localized concentration. Here, a simulated-allelopathy mode to facilitate the application of allelochemicals was proposed and tested on Microcystis aeruginosa. The continuous-release beads of 5,4'-dihydroxyflavone (DHF) were constitutive of a polymer matrix and showed a high drug-loading rate (47.18%) and encapsulation efficiency (67.65%) with a theoretical release time of approximately 120 d. Cyanobacterial growth tests showed that the DHF beads had long-term inhibition effects (>30 d), whereas those of "direct-added" DHF to cells lasted a maximum of 10 d. The beads also continuously affected the superoxide dismutase, catalase, and lipid peroxidation of M. aeruginosa. The inhibitory effects of DHF beads on cyanobacterial growth increased as initial cell densities of M. aeruginosa decreased, suggesting that the beads inhibit cyanobacterial activity more effectively in the early bloom phase. Consequently, the anti-cyanobacterial beads represent a novel application mode of allelochemicals with long-term inhibitory effects on cyanobacterial growth. Our study demonstrates that the successful application of allelochemicals offers great potential to control harmful cyanobacterial blooms, especially at the initial stage of development. Abstract:Eutrophication with a large number of Microcystis aeruginosa commonly occurs worldwide, thereby threatening the aquatic ecosystem and human health. In this study, four kinds of algicides were tested to explore their influence on cell density and chlorophyll-a of M. aeruginosa. Results showed that aluminum silicate agent, which inhibited more than 90% cell growth compared with the control group, demonstrated the strongest inhibition effect immediately on M. aeruginosa growth. Furthermore, the production and release of microcystin (MC)-LR were investigated. Aluminum silicate, CuSO4, and Emma-11 were more effective than pyrogallic acid in disrupting the cells of M. aeruginosa, thereby increasing the extracellular MC-LR concentration. Aluminum silicate caused the highest extracellular MC-LR concentration of more than 45 mg·L–1. Biotoxicity was also detected to evaluate the environmental risks of MC-LR release, which were related to the usages of different algicides. Extracellular MC-LR concentration mostly increased when the biotoxicity of algae solution increased. The experiments were also designed to reveal the effects of physical conditions in riverways, such as natural sunlight, aeration and benthal sludge, on MC-LR degradation. These findings indicated that UV rays in sunlight, which can achieve a MC-LR removal efficiency of more than 15%, played an important role in MC-LR degradation. Among all the physical pathways of MC-LR removal, benthal sludge adsorption presented the optimal efficiency at 20%.
The growth inhibitory effects and non-targeted metabolomic profiling of Microcystis aeruginosa treated by Scenedesmus sp

Abstract:The health of the aquatic ecosystem has recently been severely affected by cyanobacterial blooms brought on by eutrophication . Therefore, it is critical to develop efficient and secure methods to control dangerous cyanobacteria, such as Microcystis aeruginosa . In this research, we tested the inhibition of M. aeruginosa growth by a Scenedesmus sp. strain isolated from a culture pond. Scenedesmus sp. culture filtrate that had been lyophilized was added to M. aeruginosa , and cultivation for seven days, the cell density, chlorophyll a (Chl-a) concentration, maximum quantum yield of photosystem II (Fv/Fm), the activities of superoxide dismutase (SOD), catalase (CAT), and the concentration of malondialdehyde (MDA) and glutathione (GSH) were measured. Moreover, non-targeted metabolomics was carried out to provide light on the inhibitory mechanism in order to better understand the metabolic response. According to the results, M. aeruginosa is effectively inhibited by the lyophilized Scenedesmus sp. culture filtrate at a rate of 51.2%. Additionally, the lyophilized Scenedesmus sp. clearly inhibit the photosystem and damages the antioxidant defense system of M. aeruginosa cells, resulting in oxidative damage, which worsens membrane lipid peroxidation, according to changes in Chl-a, Fv/Fm, SOD, CAT enzyme activities and MDA, GSH. Metabolomics analysis revealed that the secondary metabolites of Scenedesmus sp. significantly interfere with the metabolism of M. aeruginosa involved in amino acid synthesis, membrane creation and oxidative stress, which is coherent with the morphology and physiology outcomes. These results demonstrate that the secondary metabolites of Scenedesmus sp. exert algal inhibition effect by breaked the membrane structure, destroyed the photosynthetic system of microalgae, inhibited amino acid synthesis, reduced antioxidant capacity, and eventually caused algal cell lysis and death. Our research provides a reliable basis for the biological control of cyanobacterial blooms on the one hand, and on other hand supply application of non-targeted metabolome on the study of microalgae allelochemicals.

Abstract:Numerous products and techniques are used to combat harmful cyanobacterial blooms in lakes. In this study, we tested nine products, the phosphate binders Phoslock® and Aqual-PTM, the coagulant chitosan, the phosphorus binder and coagulant aluminum salts (aluminum sulphate and sodium aluminate), the copper-based algicides SeClear, Captain® XTR and CuSO4·5H2O, the antibiotic Streptomycin and the oxidant hydrogen peroxide (H2O2) on their efficiency to manage the cyanobacterium Microcystis aeruginosa (M. aeruginosa). To this end, 7 days of laboratory experiments were conducted and effects were determined on chlorophyll-a, photosystem II efficiency (PSII), soluble reactive phosphorus (SRP) and intracellular and extracellular microcystin (MC) concentrations. The algicides, chitosan and H2O2 were the most powerful in reducing cyanobacteria biomass. Biomass reductions compared to the controls yielded: Chitosan (99.8%) > Hydrogen peroxide (99.6%) > Captain XTR (98.2%) > SeClear (98.1%) > CuSO4·5H2O (97.8%) > Streptomycin (86.6%) > Phoslock® (42.6%) > Aqual-PTM (28.4%) > alum (5.5%). Compounds that caused the largest reductions in biomass also strongly lowered photosystem II efficiency, while the other compounds (Phoslock®, Aqual-PTM, aluminum salts) had no effect on PSII, but strongly reduced SRP. Intracellular MC concentration followed the biomass patterns, extracellular MC was generally lower at higher doses of algicides, chitosan and H2O2 after one week. Recovery of PSII was observed in most algicides and chitosan, but not at the highest doses of SeClear and in all streptomycin treatments. Our results revealed that M. aeruginosa can be killed rapidly using several compounds, that in some treatments already signs of recovery occurred within one week. P fixatives are efficient in reducing SRP, and thus acting via resource suppression, which potentially may provide an addition to fast-acting algicides that kill most of the cells, but allow rapid regrowth as sufficient nutrients remain.

Abstract:Microcystis-dominated cyanobacterial blooms (MCBs) severely threaten ecological health by causing hypoxia and releasing microcystins (MCs). Luteolin has potential as low-cost eco-safe algaecide against Microcystis, but to enhance sustainability of its algicidal effect and elucidate underlying mechanisms at proteomic level are urgently desirable. This study optimally constructed continuous-release microsphere (CRM) of luteolin with strong solidity and durability even after long-term immersion. Applying luteolin CRM, this study developed a long-term algicidal option to strongly inhibit Microcystis growth and MC-release until 49 days, with inhibition ratios of growth and MC-release (both ≥ 98%) and inhibitory effect-lasting time (nearly 50 days) of CRM superior to most former reports, and long-term strong inhibitory effects of CRM on Microcystis growth and MC-release kept stable at various nitrogen levels. Also, luteolin CRM rendered extracellular MCs content decrease to nearby acceptable threshold for drinking water. These signified a promising prospect of luteolin CRM in sustained effective control against toxigenic MCBs in waters of different eutrophic states. Comparative proteomic analysis showed that luteolin CRM significantly up-regulated photosynthesis and protein homestasis, but down-regulated other processes including stress response, MC-synthesis/release, glycolysis, amino acid synthesis, fatty acid synthesis/β-oxidation, tricarboxylic acid cycle, transcription, translation, transport, cell shaping and cell division. These implied that continuous stress of luteolin released from CRM induced Microcystis proteome towards a shift of higher energy storage but lower energy release/consumption, which largely disturbed its physiological metabolic processes and thus negatively impact its growth. Proteomics results shed newly deep insights on algicidal mechanisms of flavonoid in the form of CRM.

Abstract:The utilization of allelochemicals, which are regarded as eco-friendly algaecides for mitigating harmful cyanobacterial blooms (HCB), has attracted increasing attention, owing to their negligible toxicity, high selectivity and excellent degradability. In the present study, the inhibitory effects of malonic acid on Microcystis aeruginosa were investigated in terms of growth indicators, cell morphology, antioxidant enzymes activities and gene expression. The results indicated that the exposure to 0.5 mmol L −1 malonic acid for 7 d caused a high microalgae inhibition rate of 86.88 %. Furthermore, the analysis of scanning electron microscopy (SEM) and transmission electron microscope (TEM) showed that the presence of malonic acid resulted in the disruption of cell integrity and the changes in cell morphology. Specifically, malonic acid was observed to cleave microalgae cells, while concurrently oxidizing the microalgae organic matter. In addition, under the exposure of 0.5 mmol L −1 malonic acid, the levels of malondialdehyde (MDA) were 3.28 times as the control group on d-5, while the activities of superoxide dismutase (SOD) and catalase (CAT) were first increased by 5.96 and 11.73 fold compare to the control group on d-5, and then decreased, due to oxidative stress induced by malonic acid. Transcriptome-based gene expression analysis revealed that the presence of malonic acid significantly inhibited the transcription of genes ( rbcL , psbA1 ) associated with photosynthesis, and the expression of DNA repair gene ( recA ), stress response gene ( ftsH ) and microcystin synthesis genes ( mcyD , mcyH ) were up-regulated. These findings provide novel insights into the inhibitory effect of malonic acid on M. aeruginosa , suggesting that malonic acid has the potential to control M. aeruginosa blooms.

Sustained and Enhanced Inhibitory Effects of Allelochemicals on Microcystis Aeruginosa During Its Recruitment Stage

Abstract:Microcystis aeruginosa is the main toxic strain in cyanobacterial blooms, and the recruitment stage in its temperature-dependent seasonal succession is considered as the key to its subsequent growth. In this study, a protocol with specific temperature settings was developed as the simulated recruitment stage in order to investigate and confirm the superior inhibitory effects of allelochemicals on M. aeruginosa at that stage of recruitment. One of the most common allelochemicals, gallic acid (GA) (10 mg/L, 20 mg/L) was employed to treat M. aeruginosa under initially low temperature condition (15 °C), then intermediate (20 °C) and last normal (26 °C), which corresponds to the critical temperatures for cyanobacterial recruitment and growth. Growth, metabolism, photosynthetic activity, extracellular polysaccharides (EPS) and microcystins (MCs) release were analyzed and discussed in this study, and a more sustained and better inhibitory effect over a 20-day period was achieved. Notably, GA (10 mg/L) markedly delayed the recruitment of M. aeruginosa from low temperature, with an inhibition efficiency of 85.71 %, and suppressing Fv/Fm and photosynthetic pigments production. It is also observed that M. aeruginosa at recruitment stage exhibited higher sensitivity and poorer resistance to allelochemical treatment, with variable responses suggesting that optimal dosages may alter. The antioxidant enzyme activities remained high under prolonged stress, and the secretion of EPS was stimulated, indicating that cyanobacteria were more inclined to form colonies. While the laboratory-based inhibitory mechanism appeared to increase the release of microcystins in individual cells, the actual concentration of microcystins in natural aquatic environments requires further investigation.

Abstract:Microcystis aeruginosa and Microcystis wesenbergii are two cyanobacteria commonly found in eutrophic shallow lakes. Previous studies reported that microcystin-producing M. aeruginosa could have an increased competitive potential on other algae and aquatic plants, and microcystin-LR (MC-LR) was regarded as an allelochemical. Based on this hypothesis, the allelopathic interaction between these two cyanobacteria was studied for the first time under laboratory conditions, and potential allelochemicals were screened. Cyanobacteria biomass and microcystin-LR (MC-LR) concentration were monitored under different culture conditions. The potential allelochemicals from M. aeruginosa were investigated by extract fractionation and GC(LC)/MS analysis. The growth of M. wesenbergii was inhibited by the addition of cell-free filtrates of M. aeruginosa whereas M. aeruginosa was promoted by the addition of cell-free filtrates of M. wesenbergii . The higher polarity the extract of M. aeruginosa is, the stronger the inhibition effect of the extract on M. wesenbergii will be. According to our results, M. aeruginosa has a significant allelopathic inhibition effect on M. wesenbergii . Allelopathic compounds from M. aeruginosa have synergistic effects on inhibition of M. wesenbergii . Besides microcystin, there may be other allelopathic compounds in M. aeruginosa .

Abstract:The outbreak of harmful algae blooms caused by water eutrophication seriously jeopardizes the aquatic ecological environment and human health. Therefore, algae control technology has attracted widespread attention between environmental scholars. Allelochemical sustained-release technology which releases the active ingredient to the target medium at a certain rate within the effective time, so that the system maintains a certain concentration, thus prolonging its influence on the target organism. Allelochemical sustained-release technology has become the focus of research due to the characteristics of high efficiency, safety, low-cost, environment friendly and no secondary pollution. This paper reviews the characteristics of allelochemical substances and the status quo of plant extraction, explains the detailed classification of allelochemical sustained-release microspheres (ASRMs) and the application of algae inhibition, summarizes the preparation method of ASRMs, elaborates on the mechanism of algae inhibition of sustained-release technology from the perspective of photosynthesis, cellular enzyme activity, algae cell structure, gene expression, and target site action. Focuses on the summary of the factors influencing the effect of algae inhibition of ASRMs, including particle size of sustained-release microspheres, selection of carrier materials, and the growth stage of algae. The future direction and prospect of algae inhibition by allelochemical sustained-release technology were prospected to provide the scientific basis for water ecological restoration.

Abstract:This work aims to identify adequate species, tissues, pretreatments and allelochemicals to inhibit algal growth. Physiological indexes were investigated to explore the mechanism underlying allelopathic effect. Results demonstrated that Microcystis aeruginosa growth could be suppressed by the four tested aquatic macrophytes, namely emergent Oenanthe javanica, Iris pseudacorus, Canna indica, and submerged Potamogeton crisp us. The leaves were responsible for releasing anti-algal inhibitors. Leaf extracts of I pseudacorus inhibited M aeruginosa by 76.60% after 11 d. Pretreatments influenced the release of inhibitors. The inhibition rate of I pseudacorus leaf extracts collected by water bath heating was 55.20% higher than that of fresh leaves. Furthermore, rare algal growth was observed after 5 d of exposure to I. pseudacorus and C. indica culture water. The allelochemicals in tissue extracts could inhibit the synthesis of chlorophyll a (Chl-a) and enhance the activity of superoxide dismutase (SOD), thus affecting algal growth. The highest removal rates of Chl-a was 78.83%. SOD activities were triggered 2 times by leaf extracts than the control. Besides, SOD activity may be impaired by the excessive algal damage. The results suggest that the major allelochemicals in I. pseudacorus culture water were identified to be aromatic compounds, including dimethyl phthalate, ethyl phthalate, phthalic acid, dibutyl phthalate, and 3-nitrophthalic acid.

Abstract:Cyanobacterial bloom formation in freshwaters is a major socio-economic and health concern across the globe. Presently used chemical and physical control strategies are inefficient in complete removal of blooms and chemical application often lead to secondary pollution in water. Hence, the current study aimed to develop a bacteria-based formulation to control toxic bloom-forming Microcystis aeruginosa in freshwaters. Two bacterial strains, Exiguobacterium acetylicum and Pseudomonas previously isolated from Sri Lankan freshwaters and characterized for M. aeruginosa cell lysis and their microcystin (MC) toxin degradation were used in the study. Initially, suitability of bacterial strains to develop into solid formulations was evaluated through biofilm formation, antibiotic sensitivity and strain compatibility. Both strains formed biofilms on the surface of microtiter plates indicating their ability to attach and colonize on solid surfaces. The multiple antibiotic resistance indices for both strains were below the threshold risk level (0.2) against the seven tested classes of antibiotics. This result indicates that there is a low risk in introducing these bacterial strains to the natural environment. Further, plate assay showed that the two strains were compatible to stay together showing no antagonistic effect on the growth of each other. Having fulfilled all three criteria tested, the two strains were immobilized into beads (~5 mm) prepared from sodium alginate at 1:1 ratio of 1×108 cells/mL bacterial inoculum. Different weights (1.0, 2.0, 3.0 g) of bacteria-immobilized beads were enclosed in sachets made with Cambrella synthetic fabric. They were introduced to M. aeruginosa grown in BG-11 medium (OD=0.2, 730 nm) and kept at 26 °C. During incubation, growth stimulation of M. aeruginosa was visually observed in 1.0 and 2.0 g beads-containing cultures, whereas, gradual discoloration of colonies was observed with 3.0 g of beads. Microscopic observations also proved complete disintegration of M. aeruginosa colonies and lysis of cells in discolored cultures. After 15 days, M. aeruginosa cell lysis was estimated as a measure of chlorophyll degradation. The highest (8.4%) cell lysis was observed in cultures containing 3.0 g of beads. This indicates that bacteria cell lysis activity depends on the load of bacteria-immobilized beads. Degradation of MC toxin was estimated by enzyme-linked immunosorbent assay. The highest (57.6%) MC degradation was observed in cultures with 3.0 g of beads after 15 days of incubation. The efficiency of bacteria release from beads was tested by placing sachets in sterilized water. At 15 days, bacterial count in water was x107 CFU/mL, with all loadings of immobilized-bacteria indicating highly efficient bacteria release from the formulation. In conclusion, this study highlights that bacteria-based sodium alginate formulations can be made as a source of inoculum to control M. aeruginosa growth and MC degradation. Abstract:Cyanobacterial harmful algal blooms are a worldwide problem with substantial adverse effects on the aquatic environment as well as human health. Among the multiple physicochemical and biotic approaches, algicidal bacterium is one of the most promising and eco-friendly ways to control bloom expansion. In this study, Stenotrophomonas sp. KT48 isolated from the pond where cyanobacterial blooms occurred exhibited a strong inhibitory effect on Microcystis aeruginosa . However, the algicidal performance and mechanisms of Stenotrophomonas sp. remain under-documented. To explore the algicidal performance and physiological response againt M. aeruginosa , further works were implemented here. Our results indicated that the algicidal rate of strain KT48 cultured in 1/8 LB medium supplemented with 0.3% starch or glucose was about 30% higher than that in 1/8 LB medium. Strain KT48 culture, cell-free filtrate, and cells re-suspended were inoculated into the M. aeruginosa culture, and the Chl- a content was determined. Those results indicated that the algicidal activity of cells re-suspended was far higher than that of cell-free filtrate and culture. Thus, strain KT48 exhibited algicidal activity mainly through direct attacking M. aeruginosa rather than excretion of algicides. Furthermore, strain KT48 led to an increase in cellular reactive oxygen species (ROS) and caused lipid peroxidation as supported by the increase in malondialdehyde (MDA) levels. The ROS and MDA levels in algal cells treated with strain KT48 cells re-suspended were about 3.23-fold and 2.80-fold higher than those of untreated algal cells on day 11. And a further inhibition to the antioxidant system is suggested by a sharp decrease in the superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) activities. In addition, we also observed that the morphology of most algal cells changed from integrity to break. This study not only indicated strain KT48 with strong algicidal activity, but also explored the underlying algicidal mechanisms to provide a source of bacterial agent for the biocontrol of cyanobacterial blooms.

Cobalt-Doped Carbon Nitride for Efficient Removal of Microcystis aeruginosa via Peroxymonosulfate Activation

Abstract:Heterogeneous persulfate activation is an advanced technology for treating harmful algae in drinking water sources, while it remains a significant hurdle in the efficient management of cyanobacterial blooms. In this study, super-dispersed cobalt-doped carbon nitride (2CoCN) was prepared to activate peroxymonosulfate (PMS) for simultaneous Microcystis aeruginosa inhibition and microcystin (MC-LR) degradation. When the initial PMS and 2CoCN concentrations were 0.3 g/L and 0.4 g/L, respectively, the efficiency of algal cell removal reached 97% in 15 min, and the degradation of MC-LR reached 96%. Analyses by SEM, TEM, and EEM spectra revealed that the reaction led to changes in algal cell morphology, damage to the cell membrane and cell wall, and the diffusion of thylakoid membranes and liposomes. The activities of antioxidant enzymes (superoxide dismutase and catalase) and antioxidants (glutathione) in algal cells generally increased, and the content of malondialdehyde increased, indicating severe damage to the cell membrane. Radical capture experiments confirmed that singlet oxygen (1O2) was the key species destroying algal cells in the 2CoCN/PMS system. The 2CoCN/PMS system was effective in removing M. aeruginosa within a wide pH range (3–9), and 2CoCN had good reusability. Additionally, three degradation products of MC-LR were identified by LC–MS/MS analysis, and a possible mechanism for the inactivation of M. aeruginosa and the degradation of MC-LR was proposed. In conclusion, this study pioneered the 2CoCN/PMS system for inhibiting M. aeruginosa and degrading microcystin, aiming to advance water purification and algae removal technology.

Abstract:Using allelochemicals to suppress cyanobacteria growth is a prospective method for its high efficiency and ecological safety. However, the suppression efficiency is affected inevitably by the extracellular polymeric substances (EPS) produced by cyanobacteria, and the knowledge about the roles of EPS in resistance to allelochemical stress is scarce. For the study, two typical anti-cyanobacterial allelochemicals were adopted to investigate the role of EPS in resistance to allelochemical stress on Microcystis aeruginosa . Results show that EPS was crucial in alleviating the toxicity of allelochemicals to algae, especially in stabilizing the metabolism and photosynthetic activity of algal cells. The aggregation rate of algal cells increased with the increase of EPS secretion, which alleviated the stress of allelopathy. Tryptophan proteins and humic acids in EPS provided a binding site for allelochemicals, and the EPS-allelochemicals complex were formed by chemical bonding. This study improved our comprehension of the role of EPS in algal inhibition by allelochemicals.
Synergetic Suppression Effects Upon the Combination of UV-C Irradiation and Berberine on Microcystis Aeruginosa and Scenedesmus Obliquus in Reclaimed Water: Effectiveness and Mechanisms.

Abstract:The risk of harmful algal blooms (HABs) in the water recharged with reclaimed water is a bottleneck for water reuse. The suppression effects and mechanisms of the combination of UV-C and berberine on Microcystis aeruginosa and Scenedesmus obliquus in reclaimed water were investigated. Mono UV-C irradiation at 75 mJ cm(-2) could suppress the growth of M. aeruginosa for 7 d and that at 90 mJ cm(-2) could suppress the growth of S. obliquus for 5 d. UV-C irradiation combined with 02-2 mg L-1 berberine lengthened the inhibition period of M. aeruginosa to 10- > 22 d and that of S. obliquus to 7- > 22 d and induced more rapid lethal effects on the harmful microalgal cells, in significant synergetic patterns. The combination of UV-C and berberine suppressed total, intracellular and extracellular microcystin-LR (MC-LR) more effectively and decreased the MC-1.12 quota significantly, which further reduced the risks of microcystin production and release. Furthermore, synergetic mechanisms of the combined treatments were systematically investigated from the aspects of photosynthetic system (photosynthetic activity and pigments), metabolic activity (ATP and membrane potential), oxidation stress (reactive oxygen species (ROS) and glutathione (GSH)), and apoptosis-like cell death (phosphatidylserine (PS) ectropion, caspase-3 activity and terminal deoxynudeotidyl transferase dUTP nick end labeling (TUNEL) positive rate). The combination treatment provided a joint attack of UV-C and berberine on photosynthetic transport chain of photosynthetic system II (PS II), and a synergetic pathway to achieve more severe disruptions in energy metabolism as well as aggravated oxidative stress. The accumulated ROS enhanced increases in programmed cell death (PCD) indicators of both microalgal species, which contributed to the enhancement effects on growth suppression. The results showed that the combination treatment achieved lower dose requirements of both UV-C irradiation and berberine for inducing the same inhibition effects on microalgal cells, which was promising to be applied in the HABs control of reclaimed water. Controlling toxic Microcystis blooms: The power of a novel microalgal predator Poteriospumella lacustris in water safety improvement

Abstract:Microcystis aeruginosa blooms have become a global environmental concern. In particular, microcystins (MC) produced by algal cells pose a great threat to aquatic ecosystem security and human health. Therefore, it is necessary to develop environmentally friendly biological measures to reduce bloom hazards. In this study, a heterotrophic flagellate, NY1, that could quickly eliminate M. aeruginosa and efficiently remove MC was isolated and identified as Poteriospumella lacustris . When the initial cell density ratio of P . lacustris to M. aeruginosa was 1:200, P . lacustris rapidly ingested M. aeruginosa , and the clearance rates were more than 90 % within 24 h. P . lacustris effectively removed 83.06 % of MC (more than 200 μg/L) in 24 h and 93.88 % in 72 h. Although temperatures lower than 15 °C and acidic conditions (pH 6) slightly and temporarily inhibited the ingestion and MC removal activities of P . lacustris within 24 h of treatment, it rapidly recovered and ingested more than 90 % of M. aeruginosa and eliminated 97 % of MC within 72 h at various temperatures (15–35 °C) and different pH values (pH 6–10). The fluorescence emission-excitation matrix spectra analysis and acute toxicity test showed that P . lacustris treatment reduced dissolved organic matter (DOM) and eliminated the toxicity of M. aerugionsa culture to three representative species at different trophic levels ( Photobacterium phosphoreum , Daphnia magna , and Ctenopharyngodon idellus ), indicating the promising potential of P . lacustris in improving water safety. In addition to M. aeruginosa , P . lacustris ingested and digested Platymonas subcordiformis , Chlorella vulgaris, and Synechocystis sp. This study provides a natural and biological solution to global Microcystis blooms by using the grazing effect of P . lacustris to remove toxic cyanobacteria and eliminate MC and DOM, ultimately improving water quality and safety.

Abstract:Addressing notorious and worldwide Microcystis blooms, mechanical algae harvesting is an effective emergency technology for bloom mitigation and removal of nutrient loads in waterbodies. However, the absence of effective methods for removal of cyanobacterial toxins, e.g., microcystins (MCs), poses a challenge to recycle the harvested Microcystis biomass. In this study, we therefore introduced a novel approach, the "captured biomass-MlrA enzymatic MC degradation", by enriching microcystinase A (MlrA) via fermentation and spraying it onto salvaged Microcystis slurry to degrade all MCs. After storing the harvested Microcystis slurry, a rapid release of extracellular MCs occurred within the initial 8 h, reaching a peak concentration of 5.33 μg/mL at 48 h during the composting process. Upon spraying the recombinant MlrA crude extract (about 3.36 U) onto the Microcystis slurry in a ratio of 0.1% (v/v), over 95% of total MCs were degraded within a 24-h period. Importantly, we evaluated the reliability and safety of using MlrA extracts to degrade MCs. Results showed that organic matter/nutrient contents, e.g. soluble proteins, polysaccharides, phycocyanin and carotenoids, were not significantly altered. Furthermore, the addition of MlrA extracts did not significantly change the bacterial community composition and diversity in the Microcystis slurry, indicating that the MlrA extracts did not increase the risk of pathogenic bacteria. Our study provides an effective and promising method for the pre-treatment of harvested Microcystis biomass, highlighting an ecologically sustainable framework for addressing Microcystis blooms.
