Abstract:Morphology, the important feature of bloom-forming cyanobacteria, was studied for its impacts on the harmful cyanobacterial bloom (HCB) treatment by coagulative colloidal ozone microbubbles (CCOMBs). The globallyappeared HCB species - Microcystis aeruginosa (spherical cells, block mass colonies), Microcystis panniformis (spherical cells, flat penniform-like colonies) and Anabaena flos-aquae (filamentous morphology) were chosen as representative species. CCOMBs were generated by modifying the bubble surface and the gas core with coagulant and ozone, respectively. The removal of spherical cells and filaments was > 99.5% and <= 34.6%, individually, and the latter was ascribed to chain breakage. CCOMBs collected Microcystis panniformis via complexing with the fluorescent and non-fluorescent functional groups of cell colonies but captured Anabaena flos-aquae through the fluorescent ones. More Microcystis aeruginosa got membrane-damaged than Microcystis panniformis; nevertheless, the microcystin-LR (MC-LR) removal was guaranteed through efficiently oxidizing the released MC-LR. Although the outer peptidoglycan sheet of Anabaena flos-aquae was destroyed, the inner cyte membrane remained intact, preventing intracellular MC-LR from releasing. The HCBs dominated by single species with spherical cells were more readily treated than those with co-occurred species. The toxicological tests imply that, as a robust tool for HCB treatment, the CCOMB technology could be eco-environmentally friendly to the aquatic environment.
Double effects of mitigating cyanobacterial blooms using modified clay technology: regulation and optimization of the microbial community structure

Abstract:Harmful algal blooms (HABs) are global hazards under global climate change and eutrophication conditions. Modified clay (MC) method is widely used to control HABs in Asian and American coastal waters. However, little research has been conducted on the underlying mechanisms by which MC controls blooms in freshwater environments. Herein, experiments and bioinformatics analyses were conducted for MC-based control of freshwater blooms in a closed water body with an area of approximately 240 m 2 in the Fuchun River, China. Results revealed that the dominant bloom species were Microcystis , and an 87.68–97.01% removal efficiency of whole algal biomass was achieved after 3 h of MC treatment. The weaker zeta potentials of Microcystis species and hydrophilic groups such as O-H and P-O-P in the extracellular polymeric substances (EPS) surrounding Microcystis cells made them easier to be flocculated and removed by MC particles, and the relative abundance of Microcystis decreased to 29.12% and that of Cyanobium increased to 40.97%. Therefore, MC changes the cyanobacterial community structure, which is accompanied by the elimination of Microcystis sp. apical dominance and enhanced competition between Cyanobium and Microcystis in the phytoplankton community, increasing cyanobacterial community diversity. Under MC treatment, residual microorganisms, including cyanobacteria, had a high potential for DNA damage repair and were more likely to survive after being subjected to oxidative stress. In the meanwhile, the abundance of genes involved in genetic information processing, signal transduction, and photosynthesis was decreased indicating that the residual microbiome was week in proliferation and light energy harvesting. Therefore, accompanied with the destruction of Microcystis colonies, MC changes the function of cyanobacteria and phycosphere microbiome, further hindering bloom development. These findings illustrate that MC can regulate and optimize the microbial community structure through which MC controls cyanobacterial blooms in ecosystems. Graphical .

Effects of modified clay on the formation of Phaeocystis globosa colony revealed by physiological and transcriptomic analyses

Abstract:The harmful algal bloom (HAB) species Phaeocystis globosa is commonly observed in global temperate and tropical oceans, and colonies of P. globosa exhibit a dominant morphotype during blooms. The use of polyaluminium chloride modified clay (PAC-MC) is an effective mitigation strategy for P. globosa blooms. Although previous studies have found that PAC-MC can stimulate P. globosa colony formation at low concentrations and inhibit it at higher concentrations, the underlying mechanisms of these effects are poorly understood. Here, we comprehensively compared the physiochemical indices and transcriptomic response of residual P. globosa cells after treatment with two concentrations of PAC-MC. The results showed that PAC-MC induced oxidative stress, photosynthetic inhibition, and DNA damage in residual cells. Moreover, it could activate antioxidant responses and enhance the repair of photosynthetic structure and DNA damage in cells. The biosynthesis of polysaccharides was enhanced and genes associated with cell motility were down-regulated after treatment with PAC-MC, resulting in the accumulation of colonial matrixes. After treatment with a low concentration of PAC-MC (0.1 g/L), the residual cells were slightly stressed, including physical damage, oxidative stress and other damage, and polysaccharide synthesis was enhanced to promote colony formation to alleviate environmental stress. Moreover, the damage to residual cells was slight; thus, normal cell function provided abundant energy and matter for colony formation. After treatment with a high concentration of PAC-MC (0.5 g/L), the residual cells suffered severe damage, which disrupted normal physiological processes and inhibited cell proliferation and colony formation. The present study elucidated the concentration-dependent mechanism of PAC-MC affecting the formation of P. globosa colonies and provided a reference for the application of PAC-MC to control P. globosa blooms.

Application of PAC-modified kaolin to mitigate Prorocentrum donghaiense: effects on cell removal and phosphorus cycling in a laboratory setting

Abstract:Among various strategies for mitigating harmful algal blooms (HABs), coagulation–flocculation of algal cells by modified clay (MC) has proven to be an effective method that is widely applied in East Asia. Inorganic nutrients produced by algal detritus decomposition after an HAB are quickly released into the overlying water, which may cause second eutrophication. For environmental safety reasons, it is of critical importance to consider the fate of flocculated organic matter and clarify its effects on nutrient cycling. The present study therefore compared cell density, in vivo chlorophyll-a fluorescence, pH, and nutrient concentrations (especially phosphorus) between control (A1) and MC-treated algal bloom waters (A2) in a laboratory setting. The results showed that 0.4 g L−1 of MC removed approximately 60 % of Prorocentrum donghaiense cells (original concentration = 4.63 ± 0.05 × 108 cells L−1) and delayed residual algal apoptosis. MC effectively scavenged 51.4 % of total phosphorus (TP) and 94.2 % of dissolved inorganic phosphorus (DIP) in seawater. Additionally, there was no excess DIP release from the MC-algae matrix into the water column, providing secondary eutrophication through algal degradation unlikely. This study newly demonstrated that MC treatment can significantly alter the velocities of phosphorus cycling (small scale) through two main pathways: (1) the direct coagulation–flocculation effect and (2) indirect blocking. This study provided fundamental insight into the quick downward sinking and slow decomposition of algal detritus in a laboratory setting and directly relates to natural processes occurring during HAB senescence/termination.

Insights into the differential removal of various red tide organisms using modified clay: Influence of biocellular properties and mechanical interactions

Abstract:In recent years, red tides have increased worldwide in frequency, intensity, involving a higher number of causative species during the events. As the most commonly used method for control of red tides, modified clay (MC) was found to have differential ability to remove various red tide species. However, the underlying mechanisms have not yet been completely elucidated. In this study, the use of MC to remove three typical disaster-causing species, Aureococcus anophagefferens, Prorocentrum donghaiense and Heterosigma akashiwo, was investigated, and differential removal of these species was probed with insights into their biocellular properties and mechanical interactions. The results showed that removal efficiencies of the three species by MC decreased in the order P. donghaiense > A. anophagefferens > H. akashiwo, while the sedimentation rates decreased in the order H. akashiwo > P. donghaiense > A. anophagefferens. Analyses of the cell surface properties and redundancy analysis (RDA) revealed that the highest surface zeta potential of -5.32±0.39 mV made P. donghaiense the most easily removed species; the smallest cell size of 3.30±0.03 μm facilitated the removal of A. anophagefferens; and the highest hydrophobicity with a H2O surface contact angle of 98.50±4.31° made the removal of H. akashiwo difficult. X-ray photoelectron spectroscopy (XPS) data indicated that the electronegativity of P. donghaiense was caused by carboxyl groups and phosphodiester groups, and the hydrophobicity of H. akashiwo was associated with a high C-(C, H) content on the cell surface. According to the extended Derjaguin, Landau, Verwey, and Overbeek (ex-DLVO) theory calculation, differences in the interaction energies between MC and the three red tide species effectively explained their different sedimentation rates. In addition, the degree of oxidative damage caused by MC to the three red tide species differed, which also affected the removal of red tide organisms.

Abstract:Modified clay is considered to be the most direct and effective method for emergency mitigation of harmful algal blooms (HABs), and it has been widely applied. However, its application on Aphanizomenon flos-aquae blooms has not been reported. Additionally, the hydraulic characteristics of modified clay in settling algal cells and the mechanism of its influence by flow fields have not been well studied, which is detrimental to the effective application of modified clay in alleviating HABs. Therefore, this study investigates the feasibility of using modified clay to alleviate Aphanizomenon flos-aquae blooms through laboratory-scale experiments and field engineering applications, and analyzes the keinetic mechanism of algal cell settling by modified clay based on flow field characteristics. The results indicate that PAC-modified clay can effectively alleviate Aphanizomenon flos-aquae blooms, achieving a removal efficiency of over 90 %. Differences in mineral composition, particle size, mineral-to-PAC ratio, and hydraulic shear significantly affect the removal performance of algal cells. The flow field characteristics of the water not only affect the removal efficiency of algal cells, but also influence the settling speed of the cells. By artificially increasing hydraulic shear and enhancing the distribution of velocity and turbulence kinetic energy in the water, the interaction between algal cell-coagulant-clay particle is promoted, resulting in the formation of larger and denser flocs, yielding faster settling performance. In the practical engineering application at Changshuiyan Reservoir, the use of boat propellers to agitate the water also verified this reinforcing effect. Aphanizomenon flos-aquae blooms were effectively alleviated, and transparency increased from the initial 0.5 m to 5.5 m. This study, for the first time, combines flow field simulation of clay particle settling to analyze the kinetic mechanism of hydraulic shear in enhancing the efficiency of modified clay in alleviating HABs from a kinetic perspective.

Abstract:The application of modified clay (MC) to mitigate harmful algal blooms (HABs) is becoming a widespread practice in China because of its low cost, high efficiency and environmental friendliness. Due to its success in China, this technology has also gained international recognition in recent years. Nevertheless, the eco-environmental impacts of this technology still remain to be a concern to many customers and government agencies, which has motivated scientists in both China and many other countries to evaluate its potential effects on nontarget aquatic organisms, water quality and the benthic environment. This paper reviews the results from these studies both in the laboratory and in the field, which showed that MC does not harm nontarget aquatic organisms and has no negative impact on water quality or the benthic environment. Additionally, findings show that MC can alter nutrient cycling and reduce algal toxins in water bodies. Furthermore, researchers also found that MC affects cyst formation and germination in sedimentary environments. This review is expected to provide scientific guidance for mitigating HABs in China and worldwide using clay or MC.

Abstract:Modified clay compounds are used globally as a method of controlling harmful algal blooms, and their use is currently under consideration to control Karenia brevis blooms in Florida, USA. In 1400 L mesocosm tanks, chemical dynamics and lethal and sublethal impacts of MC II, a polyaluminum chloride (PAC)-modified kaolinite clay, were evaluated over 72 hours on a benthic community representative of Sarasota Bay, which included blue crab (Callinectes sapidus), sea urchin (Lytechinus variegatus), and hard clam (Mercenaria campechiensis) . In this experiment, MC II was dosed at 0.2 g L −1 to treat bloom-level densities of K. brevis at 1 × 10 6 cells L −1 . Cell removal in MC II-treated tanks was 57% after 8 hours and 95% after 48 hours. In the water column, brevetoxin analogs BTx-1 and BTx-2 were found to be significantly higher in untreated tanks at 24 and 48 hours, while in MC II-treated tanks, BTx-3 was found to be higher at 48 hours and BTx-B5 was found to be higher at 24 and 48 hours. In MC II floc, we found no significant differences in BTx-1 or BTx-2 between treatments for any time point, while BTx-3 was found to be significantly higher in the MC II-treated tanks at 48 and 72 hours, and BTx-B5 was higher in MC II-treated tanks at 24 and 72 hours. Among various chemical dynamics observed, it was notable that dissolved phosphorus was consistently significantly lower in MC II tanks after 2 hours, and that turbidity in MC II tanks returned to control levels 48 hours after treatment. Dissolved inorganic carbon and total seawater alkalinity were significantly reduced in MC II tanks, and partial pressure of CO 2 ( p CO 2 ) was significantly higher in the MC II-only treatment after 2 hours. In MC II floc, particulate phosphorus was found to be significantly higher in MC II tanks after 24 hours. In animals, lethal and sublethal responses to MC II-treated K. brevis did not differ from untreated K. brevis for either of our three species at any time point, suggesting MC II treatment at this dosage has negligible impacts to these species within 72 hours of exposure. These results appear promising in terms of the environmental safety of MC II as a potential bloom control option, and we recommend scaling up MC II experiments to field trials in order to gain deeper understanding of MC II performance and dynamics in natural waters.

Abstract:The frequent occurrence and long-term duration of Microcystis harmful algal blooms (HABs) are of great concern. Chemical flocculation is thought to be an effective way to deal with the HABs, while the application of the flocculants at a high dosage pose potential adverse impacts to the aquatic ecosystems. In this study, an alternative approach is proposed that involves the employment of polyaluminum chloride (PAC) combined with the Daphnia magna (D. magna) to achieve sustainable HABs removal efficiency with an acceptable ecological risk. It was found that under a dense Microcystis HABs (algal density of 1.5 × 107 cells/ml), a PAC dosage of 30 mg/l triggered >95% algae removal, but the released Al3+ caused 90% mortality of planktonic D. magna. Reducing the PAC dosage to 15 mg/l resulted in a slightly lower algal removal efficiency (>90%). In addition the reduced PAC dosage benefited the proliferation of the remaining unicellular algal cells, which tended to form a large colony during the 25-day experiment. Incubation of D. magna following flocculation with 15 mg/l PAC effectively grazed the remaining algal cells, meanwhile increasing the D. magna density by approximately 40-folds, and enlarging the body size by 1.37–1.50 times. This result implied that the released Al3+ was not detrimental to the D. magna. Flocculation with a reduced dosage is sufficient for colonial and large algal cells mitigation, which creates a window time for the biomanipulation of the residual tiny algae. Hence, the subsequent addition of D. magna triggered the sustainable removal of the HABs cells. The present study provides an environmentally friendly strategy for cleaning up the green tides without obvious detrimental effects on the aquatic ecosystem.

Abstract:Harmful algal blooms (HABs) due to eutrophication are becoming a serious ecological disaster worldwide, threatening human health and the optimal balance of aquatic ecosystems. The traditional approaches to eradicate HABs yield several drawbacks in practical application, while microbial algicidal technology is garnering mounting recognition due to its high efficiency, eco-friendliness, and low cost. In our previous study, we isolated a bacterium strain Raoultella sp. S1 from eutrophic water with high efficiency of algicidal properties. This study further investigated the flocculation and inactivation efficiency of S1 on Microcystis aeruginosa at different eutrophic stages by customizing the algal cell densities. The supernatant extract of S1 strain exhibited remarkable flocculation and inactivation effects against low (1 × 10 6 cell/mL)and medium (2.7 × 10 6 cell/mL)concentrations of algal cells, but unexceptional for higher densities. The results further revealed that algal cells at low and medium counts manifested a more apparent antioxidant defense response, while the photosynthetic efficiency and relative electron transport rate were considerably reduced within 24 h. TEM observations confirmed the disruption of thylakoid membranes and cell structure of algal cells by algicidal substances. Moreover, TMT proteomics revealed alterations in protein metabolic pathways of algal cells during the flocculation and lysis stages at the molecular biological level. This signified that the disruption of the photosynthetic system is the core algicidal mechanism of S1 supernatant. In contrast, the photosynthetic metabolic pathways in the HABs were significantly upregulated, increasing the energy supply for the NADPH dehydrogenation process and the upregulation of ATPases in oxidative phosphorylation. Insufficient energy provided by NADPH resulted in a dwindled electron transport rate, stagnation of carbon fixation in dark reactions, and blockage of light energy conversion into chemical energy. Nonetheless, carbohydrate metabolism (gluconeogenesis and glycolysis) proteins were down-regulated and hampered DNA replication and repair. This study aided in unveiling the bacterial management of eutrophication by Raoultella sp. S1 and further arrayed the proteomic mechanism of algal apoptosis.

Abstract:The modified clay is the only worldwide-accepted practical method for mitigating algal bloom. Is it ecologically safe? To evidence it, a simulative bloom-occurring system of Karenia mikimotoi was set up, and the sentinel organisms of rotifer Brachionus plicatilis in sea surface and blue mussel Mytilus edulis on the benthos were respectively included. The organisms' physiological responses were determined as the indicators to reflect the ecological impacts when clay settled from surface to the bottom during the mimic bloom-mitigating process. Modified clay at a concentration of 0.1 g/L effectively removed the K. mikimotoi at an 81% removal rate, and its addition would not significantly strengthen the negative impacts on population dynamics and reproductive activities of B. plicatilis induced by sole K. mikimotoi within the first 2 h. Even an alleviation was observed at 2 d indicated by the increase of survival rate, egg and larva production after clay addition compared with those of 2 h. When the clay particles settled to benthos, the physical damage to the gills and digestive glands of M. edulis were found via the tissue and SEM observation, especially in higher treatment groups of 0.5 and 1.0 g/L, and filtering rate, digestive enzymes, condition index, water content and mortality were also influenced. However, little impact was found in group of 0.1 g/L. Risk assessment based on the adverse outcome pathway (AOP) model further revealed that the complete key event-key event relationship-adverse outcome pathway was only clearly observed in 0.5 g/L and 1 g/L groups but not in 0.1 g/L group, inferring the small ecological risk of 0.1 g/L. The integrated biomarker response (IBR) based on the mussel's physiological responses further backed up the AOP outcoming. The combined results from rotifer to bivalve emphasized on one conclusion that modified clay at 0.1 g/L was effective and ecologically safe in coastal bloom mitigation.

Abstract:Eutrophication with a large number of Microcystis aeruginosa commonly occurs worldwide, thereby threatening the aquatic ecosystem and human health. In this study, four kinds of algicides were tested to explore their influence on cell density and chlorophyll-a of M. aeruginosa. Results showed that aluminum silicate agent, which inhibited more than 90% cell growth compared with the control group, demonstrated the strongest inhibition effect immediately on M. aeruginosa growth. Furthermore, the production and release of microcystin (MC)-LR were investigated. Aluminum silicate, CuSO4, and Emma-11 were more effective than pyrogallic acid in disrupting the cells of M. aeruginosa, thereby increasing the extracellular MC-LR concentration. Aluminum silicate caused the highest extracellular MC-LR concentration of more than 45 mg·L–1. Biotoxicity was also detected to evaluate the environmental risks of MC-LR release, which were related to the usages of different algicides. Extracellular MC-LR concentration mostly increased when the biotoxicity of algae solution increased. The experiments were also designed to reveal the effects of physical conditions in riverways, such as natural sunlight, aeration and benthal sludge, on MC-LR degradation. These findings indicated that UV rays in sunlight, which can achieve a MC-LR removal efficiency of more than 15%, played an important role in MC-LR degradation. Among all the physical pathways of MC-LR removal, benthal sludge adsorption presented the optimal efficiency at 20%.

Abstract:Cyanobacterial bloom has been rising as a worldwide issue owing to its adverse effects to water quality and ecological health. To solve this problem, hydrogen peroxide (H2O2) has been considered as a potential algaecide because no by-products are generated after treatment and because it kills cyanobacteria selectively. In addition, cytochromes P450 (CYPs) was reported to be related with H2O2, but the roles of CYPs in the regulation of H2O2 in cyanobacteria have yet to be investigated. In this study, the CYPs suicide inhibitor 1-aminobenzotriazole (ABT) was added to the representative cyanobacteria Microcystis aeruginosa (M. aeruginosa) exposed to H2O2. The results showed that CYPs mediates the effects of H2O2 on M. aeruginosa. To be exact, the addition of ABT induced greater inhibitory effects on the growth and higher reactive oxygen species levels in M. aeruginosa comparing to those treated with H2O2 alone. At the same time, photosynthetic parameters significantly decreased, and the content of extracellular microcystins (MCs) increased but the total MCs decreased due to the combined effect of H2O2 and ABT. ABT also intensified the aggregation of Fe, which might explain the effects on photosynthesis and synthesis of MCs. Furthermore, the transcriptional levels of MCs-synthesis genes (mcyA and mcyD) decreased but MCs-release gene (mcyH) increased, and photosynthetic genes (psaB, psbD1 and rbcL) decreased, which confirmed the effects on the MC production/release and electron transport of photosynthesis, respectively. In summary, this study illuminated the mediation role of CYPs in the adverse effects on M. aeruginosa induced by H2O2, thus providing new theoretical basis for the explanation of H2O2 as potential algaecide.
Oxidative stress of Microcystis aeruginosa induced by algicidal bacterium Stenotrophomonas sp. KT48

Abstract:Cyanobacterial harmful algal blooms are a worldwide problem with substantial adverse effects on the aquatic environment as well as human health. Among the multiple physicochemical and biotic approaches, algicidal bacterium is one of the most promising and eco-friendly ways to control bloom expansion. In this study, Stenotrophomonas sp. KT48 isolated from the pond where cyanobacterial blooms occurred exhibited a strong inhibitory effect on Microcystis aeruginosa . However, the algicidal performance and mechanisms of Stenotrophomonas sp. remain under-documented. To explore the algicidal performance and physiological response againt M. aeruginosa , further works were implemented here. Our results indicated that the algicidal rate of strain KT48 cultured in 1/8 LB medium supplemented with 0.3% starch or glucose was about 30% higher than that in 1/8 LB medium. Strain KT48 culture, cell-free filtrate, and cells re-suspended were inoculated into the M. aeruginosa culture, and the Chl- a content was determined. Those results indicated that the algicidal activity of cells re-suspended was far higher than that of cell-free filtrate and culture. Thus, strain KT48 exhibited algicidal activity mainly through direct attacking M. aeruginosa rather than excretion of algicides. Furthermore, strain KT48 led to an increase in cellular reactive oxygen species (ROS) and caused lipid peroxidation as supported by the increase in malondialdehyde (MDA) levels. The ROS and MDA levels in algal cells treated with strain KT48 cells re-suspended were about 3.23-fold and 2.80-fold higher than those of untreated algal cells on day 11. And a further inhibition to the antioxidant system is suggested by a sharp decrease in the superoxide dismutase (SOD), catalase (CAT), and peroxidase (POD) activities. In addition, we also observed that the morphology of most algal cells changed from integrity to break. This study not only indicated strain KT48 with strong algicidal activity, but also explored the underlying algicidal mechanisms to provide a source of bacterial agent for the biocontrol of cyanobacterial blooms.

Neglected methane production and toxicity risk in low-frequency ultrasound for controlling harmful algal blooms

Abstract:Algal blooms are regarded as a significant source of CH4 emissions. Ultrasound has been gradually employed as a fast and efficient algae removal technology in recent years. However, the changes in water environment and potential ecological effects caused by ultrasonic algae removal are not fully clear. Here, a 40-day microcosm study was performed to simulate the collapse of Microcystis aeruginosa blooms after ultrasonic treatment. The results showed that low-frequency ultrasound at 29.4 kHz for 15 min removed 33.49% of M. aeruginosa and contributed to the destruction of cell structure, but it intensified the leakage of intracellular algal organic matter and microcystins. The accelerated collapse of M. aeruginosa blooms after ultrasonication promoted the rapid formation of anaerobic and reductive methanogenesis conditions, and elevated dissolved organic carbon content. Moreover, the release of labile organics, including tyrosine, tryptophan, protein-like compositions, and aromatic proteins, was facilitated by the collapse of M. aeruginosa blooms after ultrasonic treatment, and they supported the growth of anaerobic fermentation bacteria and hydrogenotrophic Methanobacteriales. This was also demonstrated by the increase in methyl-coenzyme M reductase (mcrA) genes in sonicated algae added treatments at the end of incubation. Finally, the CH4 production in sonicated algae added treatments was 1.43-fold higher than that in non-sonicated algae added treatments. These observations suggested that ultrasound for algal bloom control potentially increased the toxicity of treated water and its greenhouse gas emissions. This study can provide new insights and guidance to evaluate environmental effects of ultrasonic algae removal.
Controlling toxic Microcystis blooms: The power of a novel microalgal predator Poteriospumella lacustris in water safety improvement

Abstract:Microcystis aeruginosa blooms have become a global environmental concern. In particular, microcystins (MC) produced by algal cells pose a great threat to aquatic ecosystem security and human health. Therefore, it is necessary to develop environmentally friendly biological measures to reduce bloom hazards. In this study, a heterotrophic flagellate, NY1, that could quickly eliminate M. aeruginosa and efficiently remove MC was isolated and identified as Poteriospumella lacustris . When the initial cell density ratio of P . lacustris to M. aeruginosa was 1:200, P . lacustris rapidly ingested M. aeruginosa , and the clearance rates were more than 90 % within 24 h. P . lacustris effectively removed 83.06 % of MC (more than 200 μg/L) in 24 h and 93.88 % in 72 h. Although temperatures lower than 15 °C and acidic conditions (pH 6) slightly and temporarily inhibited the ingestion and MC removal activities of P . lacustris within 24 h of treatment, it rapidly recovered and ingested more than 90 % of M. aeruginosa and eliminated 97 % of MC within 72 h at various temperatures (15–35 °C) and different pH values (pH 6–10). The fluorescence emission-excitation matrix spectra analysis and acute toxicity test showed that P . lacustris treatment reduced dissolved organic matter (DOM) and eliminated the toxicity of M. aerugionsa culture to three representative species at different trophic levels ( Photobacterium phosphoreum , Daphnia magna , and Ctenopharyngodon idellus ), indicating the promising potential of P . lacustris in improving water safety. In addition to M. aeruginosa , P . lacustris ingested and digested Platymonas subcordiformis , Chlorella vulgaris, and Synechocystis sp. This study provides a natural and biological solution to global Microcystis blooms by using the grazing effect of P . lacustris to remove toxic cyanobacteria and eliminate MC and DOM, ultimately improving water quality and safety.

Abstract:Measures based on concurrent alterations of an environment’s physical, chemical, and biological factors are commonly adopted to control harmful algal blooms (HABs). It was postulated that the combinations and interactions of multiple measures could exert cumulative effects (as the overall effect may or may not be equal to the additive sum for each measure individually). However, few studies have further assessed whether the cumulative effect is synergistic, additive, or antagonistic. This study proposed a framework to distinguish and quantify the cumulative effects. We also designed an experiment to investigate the cumulative effect of the combined utilization of physical (flow velocity), chemical (copper), and biological (propionamide) measures on algae growth inhibition. The results show that the cumulative effect of physical and chemical measures on algae growth inhibition was antagonistic; the cumulative effect of physical and biological measures was antagonistic; the cumulative effect of chemical and biological measures was synergistic, and the cumulative effect of all the measures together tended to be antagonistic. These results showed that the synergistic interactions between chemical and biological measures produced antagonistic effects when physical measures were added. Through response surface methodology analysis, we also found that the physical factor was the most significant factor affecting the cumulative effect, followed by the chemical factor and then the biological factor. Our results provide a more detailed understanding of the interaction patterns among multiple measures that affect algal growth. Importantly, this understanding can be further integrated into future strategy development to fully exploit the potential of the cumulative effect at its maximum performance.

Synergetic Suppression Effects Upon the Combination of UV-C Irradiation and Berberine on Microcystis Aeruginosa and Scenedesmus Obliquus in Reclaimed Water: Effectiveness and Mechanisms.

Abstract:The risk of harmful algal blooms (HABs) in the water recharged with reclaimed water is a bottleneck for water reuse. The suppression effects and mechanisms of the combination of UV-C and berberine on Microcystis aeruginosa and Scenedesmus obliquus in reclaimed water were investigated. Mono UV-C irradiation at 75 mJ cm(-2) could suppress the growth of M. aeruginosa for 7 d and that at 90 mJ cm(-2) could suppress the growth of S. obliquus for 5 d. UV-C irradiation combined with 02-2 mg L-1 berberine lengthened the inhibition period of M. aeruginosa to 10- > 22 d and that of S. obliquus to 7- > 22 d and induced more rapid lethal effects on the harmful microalgal cells, in significant synergetic patterns. The combination of UV-C and berberine suppressed total, intracellular and extracellular microcystin-LR (MC-LR) more effectively and decreased the MC-1.12 quota significantly, which further reduced the risks of microcystin production and release. Furthermore, synergetic mechanisms of the combined treatments were systematically investigated from the aspects of photosynthetic system (photosynthetic activity and pigments), metabolic activity (ATP and membrane potential), oxidation stress (reactive oxygen species (ROS) and glutathione (GSH)), and apoptosis-like cell death (phosphatidylserine (PS) ectropion, caspase-3 activity and terminal deoxynudeotidyl transferase dUTP nick end labeling (TUNEL) positive rate). The combination treatment provided a joint attack of UV-C and berberine on photosynthetic transport chain of photosynthetic system II (PS II), and a synergetic pathway to achieve more severe disruptions in energy metabolism as well as aggravated oxidative stress. The accumulated ROS enhanced increases in programmed cell death (PCD) indicators of both microalgal species, which contributed to the enhancement effects on growth suppression. The results showed that the combination treatment achieved lower dose requirements of both UV-C irradiation and berberine for inducing the same inhibition effects on microalgal cells, which was promising to be applied in the HABs control of reclaimed water. Abstract:Harmful algal blooms (HABs) impacted negatively the water ecosystem, and produced toxic microcystins that poses toxic effect on liver, nervous, and genital system. The introduction of useful and adaptive algae-degrading microbes or bio-augmentation can be regarded as an efficient way to inhibit the outbreak of HABs. The purpose of this study is to evaluate the application potential of algicidal bacteria named XMC, which is isolated from native snails. Response surface methodology (RSM) experiments showed that self-characteristic and various external conditions affected the actual algae inhibition ability of XMC. In particular, actual algicidal efficiency was strongly depend on the temperature and growth stage of XMC, and the maximum algicidal rate could reach 93.95% within 7 days. The degradation curve of Microcystis aeruginosa was compliant with the first-order kinetic model, which could be used to predict the degradation effect of Microcystis aeruginosa in engineering applications. The analysis results of algae dissolution products showed that algicidal bacteria XMC had both direct and indirect algicidal capacity. In addition, XMC had strong algicidal ability and greater environmental adaptability, and its algae dissolution products were environmentally friendly. All results indicated that XMC had the potential to be used in the bio-degradation of cyanobacteria bloom.
