Abstract:OBJECTIVE:To investigate the cytotoxicity of calcium hydroxide (CH) and composite resin on fibroblasts derived from human embryo body fibroblasts-H9 (EBf-H9), human dental pulp cells (hDPCs) and immortalized fibroblasts L929; and to evaluate the use of EBf-H9 as a cellular model for cytotoxicity screening of dental materials. METHODS:The EBf-H9 cells were derived from human embryonic stem cells (H9) via outgrowth of embryonic body (EB); hDPCs were isolated from healthy dental pulp, and identified by immunochemical staining. Cell Counting Kit-8 (CCK-8) assay was applied to analyze the cytotoxicity of CH and composite resin with serial concentrations on the 3 kinds of cells. RESULTS:Following 24 h and 48 h (or 72 h) post-treatment of CH and composite resin, the viability of L929 cells was significantly lower than that of EBf-H9 and hDPCs (P<0.05), and there was no significant difference between the last two groups (P>0.05). CONCLUSION:Immortalized fibroblasts L929 cells exhibited different response to CH and composite resin compared with EBf-H9 and hDPCs, and the last two cell types were similar to each other. This study indicated that fibroblasts derived from human embryonic stem cells were a potential cellular model instead of traditional immortalized murine cell line for cytotoxicity screening assay.
Mechanical and biological properties of silk fibroin/carbon nanotube nanocomposite films

Abstract:Multi-walled carbon nanotubes (MWCNTs) were homogeneously dispersed in silk fibroin (SF) solutions at different compositions, and a simple solvent-casting method was used to fabricate SF/MWCNT films. Structure, viscosity, and mechanical properties of the SF/MWCNT nanocomposites were characterized by FTIR (Fourier Transform Infrared Spectroscopy), viscometry, and tensile testing. Fibroblast cells were used to examine cell viability and attachment to nanocomposite films. Compared to a pure SF film, adding just 0.5 % (w/w) of MWCNT to the SF matrix could enhance the Young’s modulus and ultimate tensile strength by approximately 24 % and 39 %, respectively. In addition, with increasing MWCNTs content, the percentage of β -sheet structure increased significantly. When the content of MWCNTs increased from 0 to 1 % (w/w), the β -sheet content improved from 17.8 % to 53.6 %. Furthermore, adding up to 1 % (w/w) MWCNT increased the viscosity of the SF solution ten fold. From a biological point of view, no toxic effect was observed on fibroblast cells in the presence of MWCNTs after 3 and 7 days. This investigation suggests that SF/MWCNT nanocomposite films with improved mechanical properties and biocompatibility are well-suited as novel biomaterials.

Abstract:Silk fibroin (SF), which offers the benefits of biosafety, biocompatibility, and mechanical strength, has potential for use as a good biomedical material, especially in the tissue engineering field. This study investigated the use of SF biomaterials as a wound dressing compared to commercially available collagen materials. After human fibroblasts (WI-38) were cultured on both films and sponges, their cell motilities and gene expressions related to wound repair and tissue reconstruction were evaluated. Compared to the collagen film (Col film), the SF film induced higher cell motility; higher expressions of genes were observed on the SF film. Extracellular matrix production-related genes were up-regulated in WI-38 fibroblasts cultured on the SF sponges. These results suggest that SF-based biomaterials can accelerate wound healing and tissue reconstruction. They can be useful biomaterials for functional wound dressings.

As a versatile natural polymer, silk fibroin has been combined with various nanoparticles. The introduction of nanoparticles brings many optimized and new functionalities to silk fibroin, such as mechanical, biological, thermal, electrical, florescent and magnetical properties. This review focuses on the preparation methods and enhanced properties of silk fibroin-nanoparticle composites, especially their biomedical applications.

Abstract:Background: Dental resin is frequently reinforced with synthetic dental fibers like glass fiber. Dental glass fiber can be challenging to find and costly in Indonesia. Bombyx mori silk fiber is a robust, convenient, and reasonably priced natural fiber substitute. Bombyx mori silk fiber is commonly used in medicinal applications as a post-surgical suture. Aim: The aim of this study was to assess the volumetric effect of Bombyx mori silk fiber on the water sorption of fiber-reinforced composite (FRC). Material and Methods: This study was an experimental laboratory. The materials used in this study were Bombyx mori silk fiber and flowable composite. Four groups of FRC samples were made using volumetric fibers at 0%, 5%, 10%, and 15% (n = 4). The test of water sorption was standardized (ISO 4049). The results were analyzed using one-way ANOVA (p<0.05). Results: The mean of water sorption of Bombyx mori silk fiber FRC for volumetric 0%, 5%, 10%, and 15% were: 0.475±0.030%; 0.681±0.033%; 0.889±0.008%; 1.318±0.039%, respectively. The present study showed that Bombyx mori silk fiber volumetric influenced the water sorption of FRC (p<0.05). Conclusion: The increased Bombyx mori silk fiber volumetric significantly increased the water sorption of FRC. The presence of a small gap caused by the weak interfacial bonds of the dental resin and fiber leads to water penetration.
The Effect of Assam Silk Fibroin on the Bacterial Cellulose Cytocompatibility as the Extracellular Matrix

Abstract:The integration of Assam silk fibroin improves the biocompatibility of the bacterial cellulose matrix and vice versa. The growing interest in natural polymers for biomedical and tissue engineering has fueled the search for materials with superior mechanical and biological properties. Bacterial cellulose (BC) and silk fibroin (SF) emerge as promising candidates meeting these criteria. This study focuses on enhancing BC through high‐pressure homogenization (HPH) and subsequent functionalization with Assam silk fibroin (ASF) using an ex‐situ approach. Analysis via attenuated total reflection–Fourier transform infrared (ATR–FTIR) spectroscopy confirms the successful integration of ASF into the HPH‐treated BC scaffold. The diffractogram of HPH‐BC/ASF indicates the prevalence of type I cellulose crystalline structures, with variations in lamellar and porous architecture based on component ratios. Mechanical testing, particularly the compressive test, reveals that the HPH‐BC/ASF formulation exhibits the highest compressive stress and modulus compared to other samples. Supplementary analyses, including swelling ratio and porosity measurements, support the superior compressive properties of HPH‐BC/ASF. Moreover, the cell viability of chondrocytes demonstrates compatibility with the BC‐based scaffold material. These findings underscore the potential applications of HPH‐BC and ASF in areas such as scaffolding for the development of extracellular matrix.

Abstract:3D printing technology has been a remarkably convincing strategy for fabricating scaffolds that can be used as a replacement for damaged tissue, which could help in regeneration and vascularization. In this study, polymer-ceramic composite 3D-printed scaffolds [Polycaprolactone(PCL), Hydroxyapatite(HAP) and Magnesium hydroxide(Mg(OH) 2 )] were fabricated, and infilled with silk fibroin(SF) to increase the cell adhesion in the scaffold. The cell adhesion was investigated by SEM analysis. It was observed that the presence of silk fibroin in the infilled region improves cell adhesion and cell viability; however, there was no significant difference in mechanical strength between SF-PCL Composite and PCL Composite, which were found to be 3.1 MPa and 2.92 MPa, respectively.

Abstract:Two kinds of gel materials were prepared from silk fibroin with polyepoxy compound at subfreezing temperature and higher temperature. In order to evaluate the feasibility of their application in biomaterials, we tested the cytotoxicity of silk fibroin gels by detecting the effect of the extracted liquid on the cell relative proliferation rate of L-929 mouse fibroblasts. The results indicated that both of the gel materials displayed high relative proliferation rate and grade 1 cytotoxicity, being in the allowed range of medical application. The cytotoxicity tests on polyepoxy compound and glutaraldehyde were conducted too, and the cytotoxicity of polyepoxy compound was obviously lower than that of glutaraldehyde. Polyepoxy compound can be used as a more safe cross-link reagent for silk fibroin modification.

Abstract:The silk nanofibrils were isolated from wasted silk fibers to create biocompatible silk fibroin nanocomposites with high performance. Regenerated silk fibroin (SF) as a biomaterial has attracted increasing attention due to its outstanding biocompatibility and processibility. However, it faces great challenges in practical application because of the deterioration of the intrinsic mechanical properties of natural silk during the regeneration process. Natural silk nanofibrils (SNFs) has a typical hierarchical structure and maintains excellent mechanical properties of natural silk. Based on principle of homogeneous reinforced composites, herein, the SNFs were isolated as a reinforcing material and regenerated SF as matrix material to fabricate high‐performance SF nanocomposites. The SNFs with diameters ranging from 100 to 600 nm were prepared with yields of 87%. They not only maintained the crystallinity of natural silk, but also remained high stability in acid and alkaline solutions. Thanks to homogenous reinforcement of the SNFs, the tensile stress of the reinforced film was up to 2.2 MPa, the strain was 129%, and the water absorption was 50% in wet state. In vitro, the reinforced films supported adhesion and proliferation of human umbilical vein endothelial cells, which has potential in the application of biomedical dressings. Highlights Natural silk nanofibrils (SNFs) were exfoliated using a facile approach. The SNFs reinforced the silk nanocomposites significantly. The silk nanocomposites showed remarkable cytocompatibility. Homogeneous reinforcement is useful strategy to fabricate nanocomposites.

Abstract:Because of the crisis of the petrochemical era, environmentally friendly natural polymers and related material processes are receiving great attention. Cellulose and silk are typical fibrous materials that consist of polysaccharides and proteins, respectively, and have excellent mechanical properties and high physicochemical stabilities due to their unique self-assembly-based hierarchical structures. In this study, highly stable high-strength silk fiber (SF)–cellulose nanofibril (CNF) composites were prepared by the hierarchical fusion of micrometer-scale SFs and nanometer-scale CNFs. This manufacturing process is cost-effective because the raw materials (SFs and CNFs) are used as is. It is an eco-friendly process because it does not require the use of organic solvents or toxic reagents. In addition, it is an energy-efficient process because heat fusion (120°C) takes only 10 min. The results of the Direct Red 80 staining experiments confirm that up to 15 wt.% CNFs were added to the SF nonwoven. With the increase in the CNF amount, the nanometer-scale CNFs form a coating on the micro-scale SFs. At the same time, the CNFs form bonds with the SFs and increase the interfibrillar bonding strength of the CNF-coated SFs. Therefore, the mechanical properties of the SF/CNF composite and its stability in the water environment rapidly increase with increasing CNF concentration. In the case of SF/CNF15, the mechanical and impact strengths increase by 110 and 228%, respectively, compared with SF nonwoven without CNF. In addition, as CNFs are introduced, hydrophobicity of the surface and bulk of the SF/CNF composite can be imparted, thereby maintaining its structural stability in the water environment. This eco-friendly SF/CNF composite can be widely used as reinforcement preform of fiber-reinforced plastics as well as for other applications in the fibrous composite industry.

Abstract:Silk fibroin (SF), a natural biodegradable and biocompatible protein, has garnered significant attention in biomedical applications due to its impressive properties, including excellent biocompatibility, biodegradability, and mechanical resilience. Nevertheless, its broader usage faces obstacles by its insufficient mechanical strength and electrical conductivity. In order to address these constraints, recent studies have concentrated on combining SF with cutting-edge nanomaterials like MXene and carbon-based materials. This review comprehensively examines the applications and potential of silk fibroin-MXene/carbon-based nanocomposites in biomedical fields. The unique properties of SF, MXene, and carbon-based materials are explored, emphasizing how their combination enhances mechanical strength, conductivity, and biocompatibility. These composites show substantial enhancements in performance for several biomedical applications by utilising the excellent conductivity and mechanical capabilities of MXene and carbonaceous elements. The innovative potential of these nanocomposites is highlighted by critically discussing key applications such as tissue engineering, drug delivery, and biosensing. In addition, the work discusses the latest research progress, difficulties, and future prospects in the sector, providing valuable insights into possible breakthroughs and uses. This review seeks to comprehensively analyse the existing information on silk fibroin-MXene/carbon based nanocomposites in healthcare.
Electrospun Silk Fibroin Scaffolds for Tissue Regeneration: Chemical, Structural, and Toxicological Implications of the Formic Acid-Silk Fibroin Interaction

Abstract:The dissolution of Bombyx mori silk fibroin (SF) films in formic acid (FA) for the preparation of electrospinning dopes is widely exploited to produce electrospun SF scaffolds. The SILKBridge ® nerve conduit is an example of medical device having in its wall structure an electrospun component produced from an FA spinning dope. Though highly volatile, residual FA remains trapped into the bulk of the SF nanofibers. The purpose of this work is to investigate the type and strength of the interaction between FA and SF in electrospun mats, to quantify its amount and to evaluate its possible toxicological impact on human health. The presence of residual FA in SF mats was detected by FTIR and Raman spectroscopy (new carbonyl peak at about 1,725 cm −1 ) and by solid state NMR, which revealed a new carbonyl signal at about 164.3 ppm, attributed to FA by isotopic 13 C substitution. Changes occurred also in the spectral ranges of hydroxylated amino acids (Ser and Thr), demonstrating that FA interacted with SF by forming formyl esters. The total amount of FA was determined by HS-GC/MS analysis and accounted for 247 ± 20 μmol/g. The greatest part was present as formyl ester, a small part (about 3%) as free FA. Approximately 17% of the 1,500 μmol/g of hydroxy amino acids (Ser and Thr) theoretically available were involved in the formation of formyl esters. Treatment with alkali (Na 2 CO 3 ) succeeded to remove the greatest part of FA, but not all. Alkali-treated electrospun SF mats underwent morphological, physical, and mechanical changes. The average diameter of the fibers increased from about 440 nm to about 480 nm, the mat shrunk, became stiffer (the modulus increased from about 5.5 MPa to about 7 MPa), and lost elasticity (the strain decreased from about 1 mm/mm to about 0.8 mm/mm). Biocompatibility studies with human adult dermal fibroblasts did not show significant difference in cell proliferation (313 ± 18 and 309 ± 23 cells/mm 2 for untreated and alkali-treated SF mat, respectively) and metabolic activity. An in-depth evaluation of the possible toxicological impact of residual FA was made using the SILKBridge ® nerve conduit as case study, following the provisions of the ISO 10993-1 standard. The Potential Patient Daily Intake, calculated from the total amount of FA determined by HS-GC/MS, was 2.4 mg/day and the Tolerable Exposure level was set to 35.4 mg/day. This allowed to obtain a value of the Margin of Safety of 15, indicating that the amount of FA left on SF mats after electrospinning does not raise concerns for human health.

Abstract:Silk fibroin (SF) is a promising biomaterial due to its good biocompatibility, easy availability, and high mechanical properties. Compared with mulberry silk fibroin (MSF), nonmulberry silk fibroin (NSF) isolated from typical nonmulberry silkworm silk exhibits unique arginine-glycine-aspartic acid (RGD) sequences with favorable cell adhesion enhancing effect. This inherent property probably makes the NSF more suitable for cell culture and tissue regeneration-related applications. Accordingly, various types of NSF-based biomaterials, such as particles, films, fiber mats, and 3D scaffolds, are constructed and their application potential in different biomedical fields is extensively investigated. Based on these promising NSF biomaterials, this review firstly makes a systematical comparison between the molecular structure and properties of MSF and typical NSF and highlights the unique properties of NSF. In addition, we summarize the effective fabrication strategies from degummed nonmulberry silk fibers to regenerated NSF-based biomaterials with controllable formats and their recent application progresses in cell behavior regulation and tissue regeneration. Finally, current challenges and future perspectives for the fabrication and application of NSF-based biomaterials are discussed. Related research and perspectives may provide valuable references for designing and modifying effective NSF-based and other natural biomaterials. Statement of Significance There exist many reviews about mulberry silk fibroin (MSF) biomaterials and their biomedical applications, while that about nonmulberry silk fibroin (NSF) biomaterials is scarce. Compared with MSF, NSF exhibits unique arginine-glycine-aspartic acid sequences with promising cell adhesion enhancing effect, which makes NSF more suitable for cell culture and tissue regeneration related applications. Focusing on these advanced NSF biomaterials, this review has systematically compared the structure and properties of MSF and NSF, and emphasized the unique properties of NSF. Following that, the effective construction strategies for NSF based biomaterials are summarized, and their recent applications in cell behavior regulations and tissue regenerations are highlighted. Furthermore, current challenges and future perspectives for the fabrication and application of NSF based biomaterials were discussed.

Abstract:Silk, as a kind of natural fibrin, has been prepared into various biomaterials due to its excellent biocompatibility and mechanicalness. However, there are some controversies on the biocompatibility of silk fibroin (SF), especially when it coexists with sericin. In this study, two kinds of silk from Jiangsu and Zhejiang were degummed with two concentrations of Na2CO3 solution, respectively, to obtain four kinds of silk fibroin. The effects of different degumming treatments on silk fibroin properties were analyzed by means of color reaction, apparent viscosity measurement, and transmission electron microscope and isobaric tags for relative and absolute quantification analyses, and the effects of different silk fibroin membranes on the growth of Schwann cells were evaluated. The results showed that the natural silk from Zhejiang treated with 0.05% Na2CO3 solution had a fuller structure, higher apparent viscosity, and better protein composition. While SF obtained by degumming with 0.5% Na2CO3 solution was more beneficial to cell adhesion and proliferation due to the thorough removal of sericin. This study may provide important theoretical and experimental bases for the selection of biomaterials for fabricating artificial nerve grafts.

Abstract:Silk fibroin material has good mechanical properties and excellent biocompatibility as a natural biomaterial with broad application prospects. However, by applying regenerated silk fibroin in biomaterials with high mechanical strength requirements, such as bone materials, there are problems, such as insufficient mechanical properties and a significant decline in mechanical properties in the wet state. In this report, a silk fibroin composite that maintains high strength in the wet state was prepared by adding nano-SiO 2 as a nano-strengthening filler to the silk protein material and employing an epoxy-based silane coupling agent KH560 as an interfacial reinforcing agent. The results showed that the dry compressive strength of the composite material was substantially increased compared with that of the pure silk protein material; the wet compressive strength was significantly increased compared with that of the pure silk fibroin material, and the decrease of the mechanical properties in the wet state was low. The cytotoxicity test results of the composites showed that the materials were not cytotoxic. Rat bone marrow mesenchymal stem cells were cultured on the surface of the composites, and the results indicated that the composites could support the proliferation of bone marrow mesenchymal stem cells. The silk fibroin nanocomposites developed in this work can be applied as bone repair materials.

Abstract:This paper investigates the effect of different plasticizers (glycerol and dextrose), post-treatment methods, and metal oxide nanoparticles, i.e. zinc oxide and copper oxide on the structural properties of silk fibroin films and their effect on electrical properties, i.e. dielectric strength. Structural properties of the silk fibroin films have been evaluated using Fourier transform infrared spectroscopy and X-ray diffraction technique. It was found that plasticizers and post-treatment applied on the regenerated silk fibroin films have impacted the β-sheet conformation significantly. The dielectric strength of the silk fibroin films was measured using the parallel plate capacitor method and an interrelation with structural properties was observed. Nanoparticles have been introduced into the silk fibroin films as a progressive method to impart substantial effect on the dielectric strength of the silk fibroin films. Abstract:Synthetic dental fiber tape for fiber reinforcement (FRC) restoration is relatively costly and its availability is still limited in Indonesia, so natural dental fibers have been used as an alternative material. The aim of this study was to assess the effect of impregnation of various fiber tape toward the flexural strength and hardness of FRC. The materials used were natural Bombyx mori silk fibers (Indonesia), dental polyethylene tape (Construct Kerr, USA), dental Quartz tape (Quartz Splint UD, France), dental E-glass tape (Everstick TM, GC, Japan), silane coupling agent (Ultradent, Jordan), and composite resin (Denfil-Flow, USA). Five groups of samples consisted of FRC with various fiber tapes were prepared: unidirectional-silk, braided-silk, quartz, polyethylene, and E-glass. The five groups of FRC were tested to determine the flexural strength and hardness. The results were analyzed by one-way ANOVA, followed by LSD test. The results showed that the highest flexural strength was in the quartz group (496.84 ± 109.14 MPa), while the lowest was in the braided-silk group (139.39 ± 4.30 MPa). The highest hardness property was in the unidirectional-silk group (141.29 ± 25.17 VHN), while the lowest was in the braided-silk group (139.39 ± 4.30 VHN). The ANOVA showed that various fiber tapes significantly influenced the flexural strength and hardness of FRC (p < 0.05). The LSD showed that the unidirectional-silk, braided-silk, and polyethylene groups demonstrated no significant difference. The LSD for hardness showed that the unidirectional-silk group had a significant difference with the other groups (p < 0.05). It can be concluded that various fiber tapes influenced the flexural strength and hardness of FRC. Natural silk fibers showed comparable flexural strength and hardness with the other fiber tapes.

Abstract:Silk fibroin (SF) is a natural polymer with excellent biocompatibility and mechanical properties and moderate human body degradability, making SF an interesting candidate for regenerative medicine. Composite materials of SF and polyethylene glycidyl methacrylate (PEGDMA), a biocompatible polymer, attract attention as scaffold materials for regenerative medicine. To the authors' knowledge, SF–PEGDMA composite hydrogels have thus far not been manufactured using optical fabrication methods, and the change in their compressive properties during their degradation has not been studied. In addition, cellulose nanofiber (CNF), a plant-derived nanomaterial with excellent mechanical properties and biocompatibility, was added to the SF–PEGDMA hydrogels to enhance their mechanical properties. SF–PEGDMA composite hydrogels were three-dimensionally printed using digital light processing. The compressive strength of the obtained hydrogels stored in pure water or phosphate buffer solution temporarily increased and decreased after 4 days. However, after 7 days, the strength decreased to a level similar to that of the specimens which did not contain CNF. In the formability tests, the reproducibility of the model changed with the intensity of the light and the CNF concentration.

Abstract:Antibacterial materials are widely used to prevent hospital-acquired infections. In our previous report, metal (calcium, copper or zinc)-doped raw silk fabrics were shown to possess strong antibacterial activities against Escherichia coli. However, antibacterial materials may occasionally be harmful to the human body; thus, in this study, we investigated the cytotoxicities of extracts from metal-doped raw silk fabrics with respect to fibroblasts and osteoblasts indirectly. Calcium-doped raw silk fabric demonstrated cytocompatibility with fibroblasts. Contrarily, copper- and zinc-doped raw silk fabrics remarkably decreased the cell densities of fibroblasts, indicating their cytotoxic effects. This observation could be attributed to the high concentrations of the released copper or zinc ions. However, calcium-, copper- and zinc-doped raw silk fabrics did not demonstrate any cytotoxic effects on osteoblasts because a high concentration of the serum alleviated the effects of these metal ions released from the fabrics. Thus, calcium-doped raw silk fabric is a promising antibacterial material that does not induce strong cytotoxicity. This study will facilitate the design of materials that are both antibacterial and safe.
