Abstract:Enantiomers of various drug molecules have a specific effect on living organisms. Accordingly, developing a sample method for the efficient and rapid recognition of chiral drug enantiomers is of great industrial value and physiological significance. Here, inspired by the structure of ion channels in living organisms, we developed a chiral nanosensor based on an artificial tip-modified nanochannel system that allows efficient selective recognition of chiral drugs. In this system, l-alanine-pillar[5]arenes as selective receptors were introduced on the tip side of conical nanochannels to form an enantioselective "gate". The selective coefficient of our system toward R-propranolol is 4.96, which is higher than the traditional fully modified nanochannels in this work.

Abstract:Enantiomers, chiral isomers with opposite chirality, typically demonstrate differences in their pharmacological activity, metabolism, and toxicity. However, direct discrimination between enantiomers is challenging due to their similar physiochemical properties. Following the strategy of programmable nanoreactors for stochastic sensing (PNRSS), introduction of phenylboronic acid (PBA) to a Mycobacterium smegmatis porin A (MspA) assists in the identification of the enantiomers of norepinephrine and epinephrine. Using a machine learning algorithm, identification of the enantiomers has been achieved with an accuracy of 98.2%. The enantiomeric excess (ee) of a mixture of enantiomeric catecholamines was measured to determine the enantiomeric purity. This sensing strategy is a faster method for the determination of ee values than liquid chromatography-mass spectrometry and is useful as a quality control in the industrial production of enantiomeric drugs.

Abstract:Molecular chirality recognition plays a pivotal role in chiral generation and transfer in living systems and makes important contribution to the development of diverse applications spanning from chiral separation to soft nanorobots. To detect chirality recognition, most of the molecular sensors described to date are based on the design and preparation of the host-guest complexation with chromophore or fluorophore at the reporter unit. Nevertheless, the involved tedious procedures and complicated chemical syntheses hamper their practical applications. Here, we report the plasmonically chiroptical detection of molecular chirality recognition without the need for a chromophore or fluorophore unit. This facile methodology is based on plasmonic nanotransducers that can convert molecular chirality recognitions occurring at nanoscale interfaces into asymmetrically amplified plasmonic circular dichroism readouts, enabling enantiospecific recognition and quantitative determination of the enantiomeric excess of small amino acids. Importantly, such a plasmon-based chirality sensing shows 102-103 amplification in the plasmonic circular dichroism signals from the detections of racemate and near-racemate of molecular analysts, demonstrating an extraordinary sensitivity to the host-guest enantioselective interactions. Furthermore, with advantages of easy-processing, cost-effective, and specific to interfacial molecular chirality, our chiroptical sensing scheme could hold considerable promise toward applications of enantioselective high-throughput screening in biology, stereochemistry, and pharmaceutics.

Abstract:A novel electrochemical chiral sensor is proposed based on CNFs-SBE-β-CD composite.Chiral recognition of positively charged enantiomers containing primary and secondary amine groups is described.Mechanism and binding thermodynamics were explained in detail.Analysis of enantiomer in racemic mixture was achieved.A highly sensitive and selective electrochemical chiral sensor was developed based on the competitive supramolecular interaction of carbon nanofibers (CNFs) embedded sulfobutyl ether-β-cyclodextrin (SBE-β-CD) with optically active cationic drugs at glassy carbon electrode (GCE). The difference in intermolecular hydrogen bonding/stability constant/enantioselectivity coefficient and Gibbs free energy of anionic host SBE-β-CD with enantiomers of amlodipine (R-/S-AML), clenbuterol (R-/S-CBL) and metoprolol (R-/S-MET) as a guest paved the way for efficient discrimination. The proposed sensing platform (CNFs-SBE-β-CD/GCE) could recognize the aforementioned enantiomers based on the discernible difference of peak potential (S/R-AML (ΔEp = 135 mV), R/S-MET (ΔEp = 99 mV) and R/S-CBL (ΔEp = 111 mV). The binding mechanisms and thermodynamic study of the enantiospecific behavior have been investigated using the host-guest chemistry approach inside the nanocavity and results suggest that S-AML, R-MET, and R-CBL show stronger stability constants than their antipodes. Formation of the diastereomeric complex was taken as a measure of enantioselectivity and experimental results indicated that anionic SBE-β-CD is a better chiral ligand than neutral cyclodextrins. Abstract:Chirality is a fascinating and essential feature of life and highly associated with many significant pharmaceutical, chemical, and biological processes. The construction of chiral recognition platform is a hot research topic and challenging assignment. Herein, we report an electrochemical method by differential pulse voltammetry (DPV) for the enantioselective recognition of chiral drug propranolol (R/S-PPL) through a nanochannel platform based on the N-acetyl-L-cysteine functionalized pillar[5]arenes derivative NALC-P5 and the porous polycarbonate membrane. The chiral discrimination depends on the difference in the supramolecular host-guest interaction between the chiral NALC-P5 and the R/S-PPL. The transmission rate of the R/S-PPL can be regulated in the nanochannel and we can achieve the selective transport of the chiral drugs. This simple electrochemical technique has potential applications as a general platform for the recognition of chiral molecules.

Abstract:ment in the prediction of overall fruit mass accretion were obtained by GAGossym. Running GAGossym with the full parameter set resulted in an overall fitness improvement of ≈25% over all output types normally used to assess fit. Our discovery of the potential of GAs to derive improved parameter sets from high-level data will have profound implications on the speed at which we can develop locality-specific systems and on the accuracy of these systems to perform at that specific site. The GA was restricted to investigate parameter ranges that were within the realm of the field-observed variability. That is, parameters continued to maintain the original biological interpretations. We have demonstrated that we can develop a fine-tuned system that responds to local environments optimally given logical constraints on parameter ranges and functional forms. GOSSYM is currently used throughout U.S. cotton-growing regions. A self-correcting feature was developed that allowed the model to rederive its parameters when field data indicated that forecasts were beyond an arbitrarily set threshold of acceptability. The process of finding the optimal configuration of parameter sets has been completely automated. The process of model correction thus depends only on the model’s perceived goodness of fit. If it does not fit data well, it will invoke its associated GA and evolve a better set of parameters. This research implies that models may be applicable and have improved forecast efficacy over a wider range of conditions than is presently the case. This improvement in performance and applicability will affect natural resource management.

Abstract:Chiral drugs are very complex substances since individual enantiomers may differ in pharmacological and toxic effects, making it necessary to analyze enantiomers separately. In this study, we investigated the chiral differentiation of two ezetimibe enantiomers (i.e., SRS-EZM and RSR-EZM) and their mechanisms in complex with β-cyclodextrins (CDs) and metal ions as the auxiliary ligands. For this purpose, two complementary approaches have been employed: electrospray-tandem mass spectrometry (ESI-MS/MS) with collision induced dissociation (CID) and molecular modeling methods, including density functional theory (DFT) calculations and molecular dynamics (MD) simulations. The results showed a good agreement between experimental and theoretical data. It was demonstrated that SRS-EZM can be easily distinguished from RSR-EZM by applying CID in ESI-MS/MS. SRS-EZM is likely to form a more stable complex with β-CD and metal ions, and thus the [SRS-EZM]-Cu-[β-CD] cluster is more energetically difficult to separate from the SRS-EZM molecule compared with RSR-EZM. Such a difference may be attributed to the interactions between the drug molecule and the metal ion, as well as the cavity shape changes of the β-CDs upon complexation with molecular guests. Therefore, enantiomers in chiral drug can be recognized as ternary complexes of metal-analyte-β-CD by ESI-MS/MS with CID.

Abstract:Chiral recognition is of great significance in the field of life science and chemistry . The design and synthesis of chiral nanomaterials have become an important way to realize chiral recognition, while the application of non-chiral materials in this regard is still rare. Here, synthesis of achiral carbon dots (Asn-CDs) using racemate (DL-Asn, Asn) as nitrogen source is proposed, and chiral recognition of cysteine enantiomers (L-/ D -Cys) is achieved by employing metal Co 2+ as the inducible factor to co-structure a fluorescent probe (Asn-CDs+Co 2+ ). And then, based on the ability of Co 2+ to quench the fluorescence of Asn-CDs and L -cysteine ( L -Cys) could restore the fluorescence of the Asn-CDs+Co 2+ probe, so achieve an "on-off-on" detection mode to detect L -Cys. The Asn-CDs+Co 2+ probe can distinguish cysteine enantiomers not only by fluorescence spectra but also by circular dichroism spectra. The probe can be used for the quantitative detection of L -Cys by fluorescence method. Under the optimal conditions, the linear range of L -Cys detection is 1.824–625.0 μM, and the detection limit is 0.6081 μM. The method has the advantages of high sensitivity, good selectivity, a wide linear range, and fast response. This work may provide a new idea for chiral analysis based on achiral materials.

Abstract:The recognition and separation of chiral molecules with similar structure are of great industrial and biological importance. Development of highly efficient chiral recognition systems is crucial for the precise application of these chiral molecules. Herein, a homochiral zeolitic imidazolate frameworks (c-ZIF) functionalized nanochannel device that exhibits an ideal platform for electrochemical enantioselective recognition is reported. Its distinct chiral binding cavity enables more sensitive discrimination of tryptophan (Trp) enantiomer pairs than other smaller chiral amino acids owing to its size matching to the target molecule. It is found that introducing neighboring aldehyde groups into the chiral cavity will result in an inferior chiral Trp recognition due to the decreased adsorption-energy difference of D- and L-Trp on the chiral sites. This study may provide an alternative strategy for designing efficient chiral recognition devices by utilizing the homochiral reticular materials and tailoring their chiral environments.

Abstract:Chirality is a fundamental property of a molecule, and the significant progress in chirality detection and quantification of a molecule has inspired major advances in various fields ranging from chemistry, biology, to biotechnology and pharmacology. Chiral molecules have identical molecular formulas, atom-to-atom linkages, and bonding distances, and as such they are difficult to distinguish both sensitively and selectively. Today, most new drugs and those under development are chiral, which requires technological developments in the separation and detection of chiral molecules. Therefore, rapid and facile methods to detect and discriminate chiral compounds are necessary to accelerate advances in many research fields. The challenges in analysis stem from the obvious fact that chiral molecules have the same physical properties. Although significant progress on the detection of enantiomeric composition has been achieved in the past decade, in order to fully realize the capacity of chiral molecular interrogation, highly sensitive and selective, portable, and easy-to-use detection remains challenging because of the limitation of conventional techniques.Soft nanoarchitectonics is a new concept for the fabrication of functional soft material systems through harmonization of various actions including atomic/molecular-level manipulation, chemical reactions, self-assembly and self-organization, and their modulation by external fields/stimuli. Soft nanoarchitectonics has been widely used as a key enabling technology for integrating predefined molecular functionalities including electrochemical, optical, catalytic, or biological properties into biosensing devices, which provides exciting opportunities to design, assemble, and fabricate tailored nanosystems to enable new sensing strategies for chiral molecules.In this Account, we aim to concisely discuss how these molecule-inspired soft nanoarchitectonics work for enantioselective sensing. We will first outline the basic principle and mechanistic insights of the soft nanoarchitectonics approach for enantioselective sensing, and then we will describe the new breakthroughs and trends in the area that have been most recently reported by our groups and others. There will also be a discussion on the merits of soft nanoarchitectonics based sensing in comparison to conventional analytical methods. Finally, with this Account, we hope to spark new chiral molecule sensing strategies by fundamentally understanding chiral recognition and engineering soft nanoarchitectonics with programmable structures and predictable sensing properties.
Chiral Metafilms and Surface Enhanced Raman Scattering For Enantiomeric Discrimination of Helicoid Nanoparticles

Abstract:Chiral nanophotonic platforms provide a means of creating near fields with both enhanced asymmetric properties and intensities. They can be exploited for optical measurements that allow enantiomeric discrimination at detection levels greater than 6 orders of magnitude than is achieved with conventional chirally sensitive spectroscopic methods based on circularly polarized light. The optimal approach for exploiting nanophotonic platforms for chiral detection would be to use spectroscopic methods that provide a local probe of changes in the near field environment induced by the presence of chiral species. Here we show that surface enhanced Raman spectroscopy (SERS) is such a local probe of the near field environment. We have used it to achieve enantiomeric discrimination of chiral helicoid nanoparticles deposited on left and right-handed enantiomorphs of a chiral metafilm. Hotter electromagnetic hotspots are created for matched combinations of helicoid and metafilms (left-left and right-right), while mismatched combinations leads to significantly cooler electromagnetic hotspots. This large enantiomeric dependency on hotspot intensity is readily detected using SERS with the aid of an achiral Raman reporter molecule. In effect we have used SERS to distinguish between the different EM environments of the plasmonic diastereomers produced by mixing chiral nanoparticles and metafilms. The work demonstrates that by combining chiral nanophotonic platforms with established SERS strategies new avenues in ultrasensitive chiral detection can be opened.

Attomole enantiomeric discrimination of small molecules using an achiral SERS reporter and chiral plasmonics

Abstract:Biologically important molecules span a size range from very large biomacromolecules, such as proteins to small metabolite molecules. Consequently, spectroscopic techniques which can detect and characterize the structure of inherently chiral biomolecules over this range of scale at the femtomole level are necessary to develop novel biosensing and diagnostic technologies. Nanophotonic platforms uniquely enable chirally sensitive structural characterisation of biomacromolecules at this ultrasensitive level. However, they are less successful at achieving the same level of sensitivity for small chiral molecules, with less than nanomole typical. This poorer performance can be attributed to the optical response of the platform being sensitive to a much larger volume of the near field than is occupied by the small molecule. Here we show that by combining chiral plasmonic metasurfaces with Raman reporters, which can detect changes in electromagnetic environment at molecular dimensions, chiral discrimination can be achieved for attomole quantities of a small molecule, the amino acid cysteine. The signal-to-noise, and hence ultimate sensitivity, of the measurement can be further improved by combining the metasurfaces with gold achiral nanoparticles. This indirect enantiomeric detection is 9 orders of magnitude more sensitive than strategies relying on monitoring the Raman response of target chiral molecules directly. Given the generic nature of the phenomenon,this study provides a framework for developing novel technologies for detecting a broad spectrum of small biomolecules, which would be useful tools in the field of metabolomics.

Abstract:Chirality sensing is a very challenging task. Here, we report a method for ultrasensitive detection of chiral molecule L/D-carnitine based on changes in the recognition tunneling current across self-assembled core satellite gold nanoparticle (GNP) networks. The recognition tunneling technique has been demonstrated to work at the single molecule level where the binding between the reader molecules and the analytes in a nanojunction. This process was observed to generate a unique and sensitive change in tunneling current, which can be used to identify the analytes of interest. The molecular recognition mechanism between amino acid L-cysteine and L/D-carnitine has been studied with the aid of SERS. The different binding strength between homo- or heterochiral pairs can be effectively probed by the copper ion replacement fracture. The device resistance was measured before and after the sequential exposures to L/D-carnitine and copper ions. The normalized resistance change was found to be extremely sensitive to the chirality of carnitine molecule. The results suggested that a GNP networks device optimized for recognition tunneling was successfully built and that such a device can be used for ultrasensitive detection of chiral molecules.
A Disposable Dual-Signal Enantioselective Electrochemical Sensor Based on Stereogenic Porous Chiral Carbon Nanotubes Hydrogel

Abstract:As the highest form of molecular recognition, the chiral molecular recognition is the most difficult measurements. Herein, a disposable dual-signal enantioselective platform was fabricated based on stereoscopic porous chiral carbon nanotubes hydrogel modified screen printed electrode. This kind of chiral hydrogel was prepared by a simple heating method with l - cysteine and chiral single-walled carbon nanotubes of chirality (6,5 ), and its dispersion and morphology were characterized by several techniques including scanning electron microscopy, Fourier transform infrared spectroscopy, Raman spectroscopy, X-ray photoelectron spectroscopy and electrochemical impedance spectroscopy. The stereogenic chiral interface was successfully employed to discriminate mandelic acid enantiomers via both oxidation peak intensity and peak potential value of cyclic voltammetry. The chiral recognition mechanism was discussed specifically, which resulted from the formation of an efficient three-dimensional chiral nanospace. The inherent chirality of chiral carbon nanotubes hydrogel, together with their orderly spatial arrangement, can significantly improve the efficiency of chiral recognition compared with traditional electrochemical chiral sensors. As a novel chiral sensing interface, such chiral carbon nanotubes hydrogel was simple to prepare, fast to operate, with good sensitivity and excellent stability for the construction of efficient and practical electrochemical chiral sensors. Abstract:A range of electrochemical sensors capable of enantiomer discrimination have been developed, but the creation of their electrochemical interfaces remains complex. This often entails combining externally introduced chiral centers with conductive materials. This study marks a significant departure from this approach by exploiting the inherent chirality of single-walled carbon nanotubes (SWCNTs) for enantiomer discrimination. We have crafted a miniaturized, enantioselective electrochemical microsensor by seamlessly integrating high-purity SWCNTs, possessing inherent chirality, with an ultra-sensitive carbon fiber microelectrode. Using differential pulse voltammetry, we observed a notable difference in peak current, highlighting the sensor's precise ability to distinguish between enantiomers of 3,4-dihydroxyphenylalanine (DOPA). Furthermore, we investigated the intricacies of the interplay between enantioselective interfaces and the molecules under discrimination, offering deeper insights into the mechanisms governing this phenomenon. This study provides substantial momentum into the realm of enantioselective electrochemical sensing and sheds light on the unique attributes of inherently chiral carbon nanotubes in such applications. This paves the way for a new breed of chiral sensors that are simpler, more efficient, and cost-effective.

Enhanced Enantioselective Discrimination Regulated by Achiral Ligands in Chiral Metal-Organic Frameworks

Abstract:Enantioselective recognition is a fundamental property of chiral linkers in chiral metal-organic frameworks (CMOFs). However, clarifying the efficient enantioselective discrimination tailored by achiral linkers remains challenging to explain the chiral recognition mechanism and efficiency. Here, two CMOFs ([Zn2(l-Phe)2(bpa)2]n and [Zn2(l-Phe)2(bpe)2]n) with the completely different enantioselective recognition are synthesized from different nonchiral ligands and the same chiral ligands. The enantioselective recognition of CMOF is undoubtedly related to l-Phe, which differs in the hydrogen bonding to the Trp enantiomer. However, the electrochemical signals are weak and undifferentiated. [Zn2(l-Phe)2(bpe)2]n produces a flattened coplanar conformation with the -C═C- tether in the achiral ligand. The flattened achiral bpee ligand and its surrounding chiral phenylalanine molecules interact through multiple π-π stacking and hydrogen bonding, which together create a chiral sensor that facilitates the recognition of l-Trp. However, [Zn2(l-Phe)2(bpa)2]n produces a stepped conformation due to the -C-C- tether in the achiral ligand; despite the recognition effect of bpea, the recognition is unsatisfactory. Therefore, the chiral recognition of the two CMOFs stems from the synergistic effect between chiral and achiral ligands. This work shows that nonchiral ligands are also crucial in determining enantiomeric discrimination and opens up a new avenue for designing chiral materials.
Enantioselective Molecular Detection by Surface Enhanced Raman Scattering at Chiral Gold Helicoids on Grating Surfaces

Abstract:Distinct advantages of surface enhanced Raman scattering (SERS) in molecular detection can benefit the enantioselective discrimination of specific molecular configurations. However, many of the recent methods still lack versatility and require customized anchors to chemically interact with the studied analyte. In this work, we propose the utilization of helicoid-shaped chiral gold nanoparticles arranged in an ordered array on a gold grating surface for enantioselective SERS recognition. This arrangement ensured a homogeneous distribution of chiral plasmonic hot spots and facilitated the enhancement of the SERS response of targeted analytes through plasmon coupling between gold helicoid multimers (formed in the grating valleys) and adjacent regions of the gold grating. Naproxen enantiomers (R(+) and S(-)) were employed as model compounds, revealing a clear dependence of their SERS response on the chirality of the gold helicoids. Additionally, propranolol and penicillamine enantiomers were used to validate the universality of the proposed approach. Finally, numerical simulations were conducted to elucidate the roles of intensified local electric field and optical helicity density on the SERS signal intensity and on the chirality of the nanoparticles and enantiomers. Unlike previously reported methods, our approach relies on the excitation of a chiral plasmonic near-field and its interaction with the chiral environment of analyte molecules, obviating the need for the enantioselective entrapment of targeted molecules. Moreover, our method is not limited to specific analyte classes and can be applied to a broad range of chiral molecules.
Identification of Single Amino Acid Chiral and Positional Isomers Using an Electrostatically Asymmetric Nanopore.

Abstract:Chirality is essential in nearly all biological organizations and chemical reactions but is rarely considered due to technical limitations in identifying L/D isomerization. Using OmpF, a membrane channel from Escherichia coli with an electrostatically asymmetric constriction zone, allows discriminating chiral amino acids in a single peptide. The heterogeneous distribution of charged residues in OmpF causes a strong lateral electrostatic field at the constriction. This laterally asymmetric constriction zone forces the sidechains of the peptides to specific orientations within OmpF, causing distinct ionic current fluctuations. Using statistical analysis of the respective ionic current variations allows distinguishing the presence and position of a single amino acid with different chiralities. To explore potential applications, the disease-related peptide β-Amyloid and its d-Asp1 isoform and a mixture of the icatibant peptide drug (HOE 140) and its d-Ser7 mutant have been discriminated. Both chiral isomers were not applicable to be distinguished by mass spectroscopy approaches. These findings highlight a novel sensing mechanism for identifying single amino acids in single peptides and even for achieving single-molecule protein sequencing.
