Abstract:OBJECTIVE To investigate the clinical significance of a rare chromosome abnormality der(Y)t(Y;1) in a patient with multiple myeloma (MM). METHODS The chromosome spread was prepared after 24 h culture of bone marrow. G-banding technique was used to analyze the karyotype. Fluorescence in situ hybridization (FISH) was performed to ascertain the origin of abnormal chromosome detected by conventional karyotypic analysis. Flow cytometry was used to detect the expression of the CD38/CD138/ZAP70. Immunoelectrophore was applied to identify the type of immunoglobulin. RESULTS A complex pattern of chromosome rearrangement was observed: 92,XXYY[3]/49,X,der(Y)t(Y;1)(q12;q21),t(11;14)(q13;q32),+18,+20,+21[47]/49,X,idem,del(13q22),ace[1]/98,XX,der(Y)t(Y;1) x 2,+18,+18,+20,+20,+21,+21[10]/46,XY[19]. The result was confirmed by metaphase-FISH. The type of immunoglobulin was IgD with the level of 6.24g/L. The CD38/CD138 was positive but ZAP70 was negative. CONCLUSION Structural abnormality of chromosome Y is rare in blood malignancy. Most of them were described in myelodysplastic syndrome or myeloproliferative disorders. It is the first report of der(Y)t(Y;1) abnormality in multiple myeloma.

Abstract:Drying treatments are an effective method of preserving the beneficial properties of postharvest mushrooms. The effects of natural-air drying (ND), hot-air drying (HD), vacuum-freeze drying (FD), heat pump drying (HPD) and microwave-vacuum drying (MVD) on the microstructure, flavor- and health-related compounds of F. velutipes root were investigated. The results showed that FD had the least impact on the microstructure of F. velutipes root and its original porous fiber structure appeared complete. It also possessed the highest content of volatile compounds. MVD gave the highest contents of umami amino acids, total phenolics and total flavonoids, and its extract exhibited high antioxidant activity. In addition, different drying treatments had significant effect on the chemical components of F. velutipes root, with FD and MVD may be the potent drying strategies for preservation of flavor and nutraceuticals respectively. Therefore, our results provided essential data support for F. velutipes root processing and functional product development.

Abstract:Plant genes usually have multiple potential poly(A) addition signals, and different sites are used for 3' processing of transcripts from a single gene. In contrast, we show here that four barley genes that also have multiple poly(A) addition signals conforming to the plant consensus use only one signal. In each of these genes, the region of cleavage for poly(A) addition is centered on a conserved pentanucleotide. This AGGCA is followed by a conserved sequence homologous to sequences involved in self-cleavage of plant viroid RNA precursors; immediately following, in turn, are four or five nucleotides complementary to the nucleotides immediately preceding AGGCA in each gene. The presence of these conserved sequences and their association with a single region for poly(A) addition in three different gene types (high-pI and low-pI alpha-amylase genes and a thiol protease) that otherwise are not homologous in their 3' untranslated/flanking sequences suggest that they might participate in some common regulatory mechanism shared by these genes.
The effect of microwave vacuum drying process on citrus: drying kinetics, physicochemical composition and antioxidant activity of dried citrus (Citrus reticulata Blanco) peel

Abstract:Hot air drying (HAD) is the most commonly used method for citrus peel drying. However, the long time and low efficiency of this process leads to the degradation of product quality and a reduction in antioxidant activity. Therefore, in this study, to improve the drying efficiency and increase the product quality of dried citrus peel, the effects of microwave vacuum drying (MVD) were compared with HAD and vacuum freeze drying (VFD) with respect to their drying kinetics, bioactive compound profile and antioxidant capacity. The results showed that the drying rate decreased with increasing sample load but increased with increasing microwave power. The Page model was the best model for fitting the drying curve of citrus peel during MVD, and the highest rehydration rate (84.5%) was obtained at a microwave vacuum power of 600 W. The microstructure of citrus peel dried by MVD was assessed by scanning electron microscopy, and a loose structure with highly porous material was observed, whereas citrus peel dried by HAD was compact. The total phenolic content (10.48 ± 0.21 mg GAE/g DW), total flavonoid content (6.54 ± 0.41 mg RE/g DW) and oxygen radical absorbance capacity (279.29 ± 11.37 µmol TE/g DW) values obtained for citrus peel dried via MVD were significantly higher than those obtained using HAD (p < 0.05). Several phenolic acid substances and flavonoid profiles were identified and quantified by HPLC-DAD, including coumaric acid, nobiletin, and tangeretin. The nobiletin content observed after MVD was significantly higher than that measured after HAD and VFD. The overall results of this study revealed that MVD at lower microwave powers appears to be a good alternative to the conventional HAD of citrus peel.

Abstract:Jujubes have been favored by consumers because of their rich nutrition and wide use. Hot air drying has been commercially and typically used to prolong shelf life and acquire the dried produce. Jujube slices were dried with hot air combined with radio frequency (RF) at different drying stages, namely, early (0–2 h, E-HA + RF), middle (2–4 h, M-HA + RF), later (4–6 h, L-HA + RF), and whole (0–6 h, W-HA + RF) stages. This study aimed to investigate the effects of different RF application stages on the microstructure, moisture absorption rate, color, and ascorbic acid of jujube slices. Compared with the hot air drying (HA) group, the E-HA + RF group obtained the best results among the experimental groups because it reduced the cells with a roundness of less than 0.4 by 5%. Moreover, the M-HA + RF group showed better results than those of other groups, with an 18.6% and 48.8% reduction in cells for a cross-sectional area less than 200 μm2 and a perimeter less than 25 μm, respectively. The minimum total color difference (ΔE = 9.21 ± 0.31) and maximum retention of ascorbic acid (285.06 mg/100 g) were also observed in this group. Therefore, the method of hot air drying assisted by phased RF is viable in the drying industry to improve the quality of dried agricultural products and reduce energy consumption.

Abstract:Microwave vacuum drying (MVD) is a rapid drying method, which can achieve a good balance between drying rate and quality. In the present work, the effects of MVD processes on the quality, drying kinetics, and moisture diffusion of yam (Dioscorea opposita L.) were investigated. Results indicated that the loss of moisture in the MVD of yam slices mainly occurred in the stage with a constant and decreasing speed. When the moisture content of the dry base was decreased to about 1.3 g/g (dry basis, D.B.), it began to enter the deceleration phase. The effective moisture diffusivity (Deff) and mass transfer coefficient (km) increased following the power, loading amount at one time, and vacuum (pressure drop). The established equation of these parameters described well this variation law. Furthermore, a neural network model was established to predict the change in moisture content in the drying process, and the law of moisture diffusion was described. In terms of quality, the contribution ranked from high to low was loading, power, and pressure. Increasing the microwave power, loading, and maintaining a high vacuum degree could reduce energy consumption and ensure quality, thus improving the economic feasibility of microwave vacuum in the drying process.

Abstract:To examine the processing characteristics and high quality of an improved microwave vacuum drying system, litchi fruits were dried using intermittent microwave volumetric heating while microwave vacuum drying at 2 W/g was carried out for comparison; the intermittent microwave heating profiles were set as (1) 5 min drying-on, 5 min drying-off; (2) 5 min drying-on, 10 min drying-off; and (3) 5 min drying-on, 15 min drying-off. Energy consumption during drying was determined, and physicochemical properties such as moisture content, vitamin C, total phenolics, color, and sensory evaluation of dried products were assessed. In microwave vacuum drying, intermittent microwave volumetric heating was found to be energy-efficient (about 32 KJ/g to 45 KJ/g) and saved at least 31% of energy consumption compared with microwave vacuum drying as well as decreasing product browning. In addition, microwave volumetric heating had no substantial effects on sugar and protein contents, while antioxidants were affected significantly (p ≤ 0.05). Moreover, sensory evaluation showed that intermittent microwave-assisted vacuum drying (IMVD) increased the acceptance of the dried product compared with microwave vacuum drying (MVD).

Abstract:In this study, fresh wolfberry in the current season was used as the experimental material, and a radio frequency (RF)-hot air combined segmented drying process was adopted for berry preservation. Ultrasonic treatment, hot water blanching, sucrose infiltration, ultrasonic and blanching, ultrasonic and infiltration, and NaOH and NaCl solution impregnation were used for pre-treatment of combined drying. Taking natural drying and conventional Na 2 CO 3 dewaxing treatment as the control group, the effects of different pretreatment methods on drying characteristics, quality, and microstructure of wolfberry during combined drying were investigated. The drying time after ultrasonic and blanching pretreatment was the shortest (13.5 h), which effectively retained total flavonoids (1.79 mg/g) and enhanced their antioxidant activity ( I = 60.78%). NaCl impregnation treatment significantly increased the total phenol content (8.77 mg/g), whereas the retention rate of soluble sugar (0.73 g/g), ascorbic acid (3.96 mg/100 g), betaine (2.72%), and other nutrients increased significantly after ultrasonic and infiltration treatment, with improved color, rehydration rate and microstructure over other treatment methods. The Weibull distribution function can accurately describe the RF-hot air combined drying process of wolfberry after different pretreatment, and simulated results were consistent with the results of the combined drying characteristic curve of wolfberry. The aim of this study was to explore suitable pretreatment methods to improve the drying rate and ensure the quality of dried products of wolfberry.

Abstract:The present study aimed to investigate the effect of rotary microwave vacuum drying (RMVD), radio frequency vacuum drying (RFVD), vacuum far infrared drying (VFID), vacuum drying (VD), hot air drying (HD) and natural drying (ND) on the drying characteristics, active ingredients and microstructure of Codonopsis pilosulae slices. Compared with the fitting results of the four models, the Weibull model is the most suitable drying model for Codonopsis. The RFVD and HD color difference values were smaller compared to ND. The effective moisture diffusivity (Deff) under different drying methods was between 0.06 × 10−8 m2/s and 3.95 × 10−8 m2/s. RMVD-dried products had the shortest drying time and retained more active ingredients. The microstructure analysis revealed that the porous structure of RMVD is more favorable for water migration. RMVD is a promising dehydration method for obtaining high-value-added dried Codonopsis products.

Abstract:The objective of this study was to develop an efficient drying technology for poria cubes in order to improve product quality. Poria cubes were dried using different methods, including air impingement drying, infrared-assisted air impingement drying, vacuum drying, two-stage vacuum drying, and infrared-assisted air impingement drying. The results were compared with those from hot air drying. For the two-stage drying, the tested conditions were the first stage of vacuum drying with temperatures between 65-85 °C and a switching moisture ratio of 70-90%. The second stage infrared-assisted air impingement drying also had temperatures 65-85 °C. The drying kinetics (effective moisture diffusivity (Deff), Biot number (Bi), and mass transfer coefficient (k) were studied via the product qualities (broken ratio, firmness, microstructure, and water-soluble polysaccharide content) and specific energy consumption (SEC) of the drying processes. The results showed that two-stage drying led to the lowest drying time and energy consumption, and also obtained the best qualities. Box-Behnken experimental design with response surface methodology (RSM) was used to optimize the two-stage operating conditions as 82 °C under vacuum drying until a moisture content of 81% and a temperature of 69 °C with infrared-assisted air impingement drying was achieved. These findings suggested that two-stage vacuum and infrared-assisted air impingement drying is a promising method for producing high quality and energy efficient dried poria cubes.

Abstract:In this study, different drying methods namely, hot air drying (HAD: 60, 80 °C), vacuum freeze-drying (VFD: -40, − 80 °C) and vacuum-microwave drying (VMD: 160, 320 W), showed a significant effect (p < 0.05) on the drying kinetics and quality of WMR obtained from two largely cultivated watermelon cultivars. Wang and Singh and the Logarithmic model having higher R2 were selected as the best-fitted thin-layer drying model to explain the drying characteristics of WMR. The quality parameter studied includes chromatic parameters, bioactive and individual phenolic compounds of dried WMR. Chromatic parameters (L*, a*, b* and ΔE) were greatly influenced by the drying temperature during HAD process. VFD preserves the original nutritional composition of both cultivars and VMD results in increased phenolic and flavonoid compounds extraction as microwave power was increased from 160 to 320 W. The best quality dried product having a high bioactive composition, phenolic compounds and antioxidant capacity was observed in WMR of cultivar Black chairman dried after VFD (− 40 °C) and VMD (320 W) and the lowest was observed in HAD (80 °C). The VFD, with its higher retention of bioactive compounds and coloring parameters, could be used as a substitute for HAD, whereas the VMD, with its faster drying time and higher concentration of bioactive compounds, could be used as a substitute for HAD.Graphical abstract

Abstract:Fresh foods like vegetables, fruits, and aquatic products have high water activity and they are highly heat-sensitive and easily degradable. Dehydration is one of the most common methods used to improve food shelf-life. However, drying methods used for food dehydration must not only be efficient and economic but also yield high-quality products based on flavor, nutrients, color, rehydration, uniformity, appearance, and texture. This paper reviews some new drying technologies developed for dehydration of vegetables, fruits, and aquatic products. These include: infrared drying, microwave drying, radio frequency drying, electrohydrodynamic drying, etc., as well as hybrid drying methods combining two or more different drying techniques. A comprehensive review of recent developments in high-quality drying of vegetables, fruits and aquatic products is presented and recommendations are made for future research.

Radio frequency drying on functional diversity of tiger nut flour: Effects on physicochemical, structural, and rheological properties

Abstract:Tiger nut (TN) is a valuable nutrient and gluten-free tuber. To achieve high-quality TN flour as functional ingredients in food, it is essential to develop effective drying technologies for TN. Five drying methods including natural drying (Control), hot-air drying (HD), radio frequency single drying (RFSD), RF assisted hot-air drying (RFHD), and RF- vacuum drying (RFVD) were selected and compared to determine their effects on physiochemical, structural, and rheological properties of TN flour. Results showed that RF drying (RFD) significantly improved the hydration, oil-absorbing, and antioxidant activity capacity, especially for RFVD. RFHD exhibited greater color (BI = 13.80 ± 0.05 and C = 10.26 ± 0.05) and reducing sugar content (253.50 ± 2.27 mg d.b.) than RFSD and RFVD. The gelatinization temperature, enthalpy value, and particle size (57.30-269.33 μm) of TN flour were reduced. The structural property results indicated that RFD reduced the relative crystallinity and short-range ordering of the flour, altered protein secondary structure, and caused the damaged microstructure in comparison with Control and HD groups. All sample gels exhibited a weak strain overshoot behavior (type III) under large amplitude oscillations, and RFD resulted in a reduced viscoelastic behavior. RFD could be an effective method to produce functional TN flour.

Abstract:Radio frequency (RF) drying is an emerging technology for food and agricultural products, holding features of rapid, uniform, stable, and volumetric heating, high energy efficiency, and moisture leveling. However, the RF drying with a single stage commonly has drawbacks of unexpected product quality, non-uniform moisture distribution, and prolonged drying time. The multi-stage drying approach could overcome the shortcomings of one-stage strategy accordingly by applying different drying methods or operating parameter values in each phase separately. This review describes the principle of RF heating, presents the typical systems and superiorities of RF drying, and provides a comprehensive overview on recent development in applications of both the one-stage and the multi-stage RF drying, and analysis of drying characteristics and merits for different types of the two-stage strategy. This review finally proposes recommendations for future studies in improving and optimizing the existing RF drying protocols and scaling up them to industrial applications.

Abstract:The objective of this research was to examine the drying characteristics of pulsed vacuum drying (PVD) system as an alternative drying technology for a functional food—Poria cocos. Poria cubes in thin layer were dried at different temperatures (55, 65, and 75°C), vacuum duration (5, 15, and 25 min), and atmospheric pressure duration (4, 8, and 12 min) to evaluate the operating conditions on the drying kinetics and quality attributes of P. cocos. The quality attributes included volume shrinkage ratio, firmness, water-soluble polysaccharide content. Volume shrinkage ratio was determined by image processing technology and analyzed by Bigot’s curves. The material internal temperature was also recorded during the whole drying process. Considering the drying rate and quality attributes, drying temperature of 65°C, vacuum pressure duration of 15 min, and atmospheric pressure duration of 4 min were proposed as the most favorable drying conditions for drying Poria cubes. The current work indicates PVD is a promising technology for P. cocos drying as it not only enhances the drying process but also improves the quality attributes of it.

Abstract:In order to improve the shortcomings of uneven heating of traditional microwave drying and to maximally maintain food quality after harvest, a rotary microwave vacuum drying equipment was fabricated and used for drying experiments on Angelica sinensis to explore the effects of drying temperature, slice thickness, and vacuum degree on drying characteristics, physicochemical quality, and microstructure of dried Angelica sinensis products. The results showed that microwave vacuum drying can significantly shorten the drying time and improved the drying efficiency. Six different mathematical models were investigated and the Midilli model was the best-fitted model for all samples (R2 = 0.99903, Pearson's r = 0.99952), and drying methods had various effects on different indexes and were confirmed by Pearson's correlation analysis and principal component analysis. The optimal process parameters for microwave vacuum drying of Angelica sinensis were determined by entropy weight-coefficient of variation method as 45 °C, 4 mm, −0.70 kPa. Under this condition, well preserved of ferulic acid, senkyunolide I, senkyunolide H, ligustilide, total phenols and antioxidant activity, bright color (L* = 77.97 ± 1.89, ΔE = 6.77 ± 2.01), complete internal organizational structure and more regular cell arrangement were obtained in the samples. This study will provide a theoretical reference for the excavation of the potential value and the development of industrial processing of Angelica sinensis.

Abstract:A microwave–osmotic–vacuum combination drying technique was used to evaluate the effect of processing factors such as temperature and concentration with 15% inclusion of maltodetrin (dextrose equivalent, DE 10) during the microwave osmotic drying under continuous medium spray flow (MWODS) and power level during finish microwave vacuum drying (MVD). Moisture loss (ML), solids gain (SG), ML/SG ratio, weight reduction (WR), bulk density (BD), rehydration capacity (RC), drying times, and quality parameters, total color difference (Δ E ), texture (hardness and chewiness) were evaluated during the process to assess the product quality. Drying conditions were optimized to maximize ML, ML/SG, WR, BD, RC and minimize Δ E , hardness, chewiness, and drying times. Optimum levels were found at 58.5 °C temperature, 50.0% concentration, and 30 min contact time during MWODS and 20% initial power up to 50% moisture content and 10% final power level to the final moisture content of 20% (32 min). In addition to MVD, freeze drying, vacuum and air drying were also used for comparison for finish drying. Overall, the MWODS with maltodextrin moderated sucrose as solute and MVD for finished drying resulted in better product quality and remained closest to the freeze dried samples.
