Abstract:The aim of this study was to compare total phenolic content (TPC), radical-scavenging activity (RSA), total anthocyanin content (TAC), sugar and polyphenolic profiles of two apple cultivars (‘Discovery’ and ‘Red Aroma Orelind’) from organic and integrated production systems in climatic conditions of Western Norway. Sixteen sugars and four sugar alcohols and 19 polyphenols were found in the peel, but less polyphenols were detected in the pulp. The peel of both apples and in both production systems had significantly higher TPC and RSA than the pulp. The peel from integrated apples had higher TPC than the peel from organic apples, while organic apples had higher TAC than the integrated. Sucrose and glucose levels were higher in organic apples; fructose was cultivar dependent while minor sugars were higher in integrated fruits. The most abundant polyphenolic compound in the peel of the tested cultivars was quercetin 3-O-galactoside, while chlorogenic acid was most abundant in the pulp. Regarding polyphenols, phloretin, phloridzin, protocatechuic acid, baicalein and naringenin were higher in organic apple, while quercetin 3-O-galactoside, kaempferol 3-O-glucoside, chlorogenic acid and syringic acid was higher in integrated fruits. In conclusion, organic ‘Discovery’ and integrated ‘Red Aroma Orelind’ had higher bioavailability of health related compounds from the peel and the pulp.

Abstract:Apple (Malus pumila Mill.) is a popular fruit with high economic values and various biological activities that are beneficial to human health. In this study, 35 apple cultivars were collected and were evaluated for their basic quality indexes, phenolic compositions, antioxidant activity, anti-tumour, and anti-diabetic activities. The compositions of phenolics were detected by using high-performance liquid chromatography (HPLC) and high-resolution mass spectroscopy (HRMS) assays. The antioxidant activities of peel and pulp extracts from 35 apple cultivars were evaluated by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging assay and ferric reducing antioxidant power (FRAP) assay. Results showed that the contents of phenolic acids and proanthocyanidins showed significant correlations with the antioxidant activities. Phenolic-rich extracts significantly inhibited HepG2 cell proliferation, with the inhibition activity varied significantly between cultivars. ‘Gold Delicious’ pulp extract, ‘Xiboliyabaidian’ peel and pulp extracts showed protective effects on H2O2-induced injury of human umbilical vein endothelial cells (HUVEC). ‘Red Fuji’ peel extract, ‘Xiboliyabaidian’ peel and pulp extracts, as well as ‘Gold Delicious’ peel extract, significantly increased glucose consumption of HepG2 cells, in a dose-dependent manner. This research may provide theoretical guidance for further nutritional investigation of the apple resources.

Abstract:Apple is rich in secondary metabolites, which play a role in maintaining health and disease prevention. Polyphenols are the principal constituents of apples. In this study, 25 apple cultivars cultivated at the same field conditions were analyzed for their antioxidant potential and polyphenolic content. The total phenolic content of apples ranged from 7.22 to 143.57 mg GAE/100 g fw, the flavonoid content varied from 17.54 to 143.00 mg CE /100 g fw. The maximum catechin content 2310 ± 7.45 μg/g fw. The maximum epicatechin, quercetin and phloridzin content was observed in Malus Floribunda i.e., 1740 ± 5.03 μg/g fw , 5110 ± 4 93 μg/g fw, and 2170 ± 8.76 μg/g fw, respectively. Maximum content observed significant relations with catechin ( r = 0.2943) epicatechin ( r = 0.2597), quercetin ( r = 0.3995), apigenin ( r = 0.4054), phloridzin ( r = 0.3329). The most significant correlation was observed between DPPH and apigenin ( r = 0.4054). Principal component analysis and component score matrix show the distribution of polyphenolics between 25 apple cultivars of diverse origins. The HCA divided the cultivars into three main metabolically rich clusters. Our study elucidates the variation in antioxidant potential of different apple cultivars which could be useful for the development of new apple cultivars by conventional and modern breeders.

Abstract:Apples and apple-based products are particularly interesting due to being a good source of polyphenols in an everyday diet. Recently there has been increased interest in the preservation of traditional apple varieties due to studies that suggest that traditional apple varieties have a higher content of polyphenols and antioxidant activity compared to commercial varieties. This study shows that traditional apple varieties contain higher concentration of polyphenolic compounds than conventional ones, such as chlorogenic acid (1.29–456 mg/kg dw), catechin (0.70–312 mg/kg dw), epicatechin (1.75–244 mg/kg dw), procyanidin B1 + B2 (4.08–358 mg/kg dw) and quercetin-3-glucoside (0.96–231 mg/kg dw). This research underlies the great potential of traditional apple varieties as a source of natural antioxidants and polyphenolics.

Abstract:In this work, 12 apple cultivars grown organically in three regions of Norway (Telemark, Ullensvang, Viken) were analyzed in terms of fruit quality, with the aim of equating different growing regions under specific climatic conditions. Apples were analyzed for concentration levels of minerals, sugars, sugar alcohols, organic acids, total phenolic content (TPC), radical scavenging activity (RSA), and phenolic profiles. Discovery "Rose" from Telemark stored the highest level of minerals (24,094.5 mg/kg dry weight). Glucose, fructose, sucrose, and sorbitol were the major carbohydrates, whereas the predominant organic acids were quinic acid and malic acid. Cultivar Discovery from Ullensvang had the highest TPC (9.22 g/kg) and RSA (229.32 mmol TE/kg). Of the polyphenols quantified, chlorogenic acid and kaempferol-3-O-glucoside were the most abounded, accounting for 85.50%. Principal component analysis (PCA) shows that the Ullensvang region is the richest source of most carbohydrates, organic acids (quinic, shikimic, and galacturonic), and most polyphenols, whereas the highest content of minerals and maleic acid characterized Viken. Regardless of location, the Discovery cultivar had, on average, the highest sugar and polyphenol contents. The results obtained suggest that organic apples from Norway are a rich source of beneficial compounds that can have a positive impact on human health. In addition, these results may be useful for consumers in identifying apple cultivars with desirable characteristics and for the fruit industry in tracing back the origin of apples. The findings could also be of great interest for locations with similar climate and soil conditions worldwide.

Abstract:Fruits are nowadays considered important suppliers of anti-oxidant molecules. Apples are particularly rich in phenolic compounds, non-nutritional phytochemicals that play active roles in controlling severe chronic diseases. In this work, 19 phenolic compounds were investigated in both skin and pulp tissues of seven apple accessions across the Malus genus collected at two stages: during fruit development and at harvest. The primary difference in phenolic concentration between wild and domesticated accessions, especially in the pulp, could be explained by the larger growth rate of the domesticated varieties. The proposed dilution effect was also confirmed through the observation of the increased content of procyanidin B2+B4 and phloridzin in russet-skinned apples, known to have higher concentrations of these compounds. The metabolite screening was also accompanied by the expression analysis of 16 polyphenolic genes showing, for nine elements, a higher expression at harvest than during fruit development. Finally, a polyphenolic comparison with red-fleshed apples was also carried out, underlying a larger amount of procyanidins and quercetin-3rhamnoside in the white-fleshed accessions. The results presented and discussed in this work suggest that specific white-fleshed apples, especially with russeted-skin, may play an important role in ameliorating the nutraceutical potential of apple fruit.
Electroactive Phenolic Contributors and Antioxidant Capacity of Flesh and Peel of 11 Apple Cultivars Measured by Cyclic Voltammetry and HPLC–DAD–MS/MS

Abstract:In this study, 11 apple cultivars were characterized by their total phenolic content (TPC) and total flavonoid content (TFC) and antioxidant, reducing, and chelating capacity by 2,2-diphenyl-1-picrylhydrazyl (DPPH) test, cyclic voltammetry (CV), and ferric reducing antioxidant power (FRAP) assays; and ferrous ion chelating capacity. The phenolic compounds in flesh and peel were determined by liquid chromatography coupled to mass spectrometry and diode array detector (HPLC–DAD–MS/MS) and their electroactivity by CV. The results showed higher TPC, TFC, and antioxidant capacity by DPPH test in the peels of all apple cultivars as compared to the respective flesh. The peel extracts also showed two-fold higher FRAP values as compared to the flesh extracts. The reducing capacity of the peel and flesh determined by CV measurements confirmed the results achieved by spectrophotometric methods of evaluating antioxidant capacity. There was no significant difference in chelating capacity in the peel and flesh. The HPLC–DAD–MS/MS analysis showed the presence of 11 phenolic compounds in the peel and flesh which varied in antioxidant, reducing, and chelating activity. The order of the phenolic compound content in flesh and peel in Quinte cultivar, which showed the highest antioxidant capacity, was as follows: epicatechin > chlorogenic acid > quercetin 3-arabinoside > quercetin 3-glucoside > cyanidin 3-galactoside > quercetin 3-rhamnoside > catechin > phloridzin > rutin > phloretin = quercetin. CV results were highly correlated with those obtained by spectrophotometry and HPLC–DAD–MS/MS, providing evidence to support the use of cyclic voltammetry as a rapid method to determine the phenolic profile and reducing the power of apple flesh and peel. The association between antioxidant assays and phenolic compound content showed that the highest contribution to the antioxidant capacity of apple peel and flesh was provided by catechin, epicatechin, and cyadinin-3-galactoside, while phloretin, phloridzin, and chlorogenic acid were the main contributors to chelating activity. Results from this study clearly indicate that removing the peel from apples may induce a significant loss of antioxidants.

Abstract:More than ten thousand apple cultivars with high variability in the quality traits and year-round availability place apples at the forefront of consumed fruits. Yet consumers and producers alike are still looking for new apple cultivars with specific quality attributes such as plant resistance to biotic and abiotic stresses as well as a high health-promoting potential. The model plants were three cultivars: a new ‘Chopin’ and an old ‘Granny Smith’, scab resistant cultivars with green peel, and a red-skinned cultivar ‘Gala Schniga’. Apple peel and flesh were analyzed separately during two growing seasons: 2016 and 2017. The total ascorbate and phenolics as well as individual phenolic compounds, such as (+)-catechin, (−)-epicatechin, chlorogenic acid, phloridzin, and rutin concentrations, proved to be highly tissue-type and cultivar dependent. The apple of the ‘Chopin’ and ‘Granny Smith’ cultivars expressed much lower skin-to-flesh antioxidant potential differences as compared to ‘Gala Schniga’. The lowest differences between tissue types were observed in the case of chlorogenic acid and flavan-3-ols, followed by total phenolics and ascorbate concentrations. Except for phloridzin, ‘Gala Schniga’ exhibited the highest differences in global and individual phenolic compound concentrations as well as total antioxidant capacity between the apple peel and flesh. ‘Chopin’ was definitely distinguished by the highest concentration of ascorbate in both the peel and the flesh and expressed a higher concentration of flavanols, especially compared to ‘Granny Smith’. In contrast to ‘Gala Schniga’, ‘Chopin’ was richer in chlorogenic acid, (+)-catechin, and (−)-epicatechin in the flesh. The total antioxidant capacity of the green-peel apple cultivars was similar to that of the red-peel one. A narrower range of differences between the concentration of antioxidants in apple peel and flesh could mean better health-promoting properties and might be related to a greater resistance to environmental stress factors.

Abstract:The aim of this work was to investigate the influence of two locations and seven cultivars on the profiling of metabolites in organically grown plums (Prunus domestica L.) fruit in Norway. P, K, and Ca were most abundant in the studied fruits, while Ba and Sr formed a clear line between the locations. The most abundant sugars were glucose, fructose, sucrose, and sorbitol, which together accounted for up to 97.00%. Quinic acid and malic acid were the predominant organic acids, while chlorogenic acid, rutin, and kaempferol-3-O-glucoside were the most abundant polyphenols. Plums from Ullensvang were characterized by a higher content of minerals, sugars, organic acids, total polyphenol content (TPC), and radical scavenging activity (RSA), while plums from Telemark had a higher content of quantified polyphenols. The cultivar 'Mallard' had the highest mineral and radical scavenging activity, 'Opal' had the sweetest fruit, 'Jubileum' had the highest acidity, 'Excalibur' had the highest TPC content, and 'Valor' stored the highest content of quantified polyphenols, especially chlorogenic acid. These results provide comprehensive information on the chemical profiles of selected plum cultivars, suggesting that organic plums are a rich source of beneficial compounds that can have a positive impact on human health.

Abstract:Micro matrix solid phase dispersion (micro-MSPD) was optimized by response surface methodology for the extraction of polyphenols from the peel of twelve traditional and eight commercial apple varieties grown in Croatia. The optimized micro-MSPD procedure includes the use of 0.2 g of sample, 0.8 g of dispersant, a 57% solution of methanol in water as the solvent and 5 mL of extract volume. The total polyphenolic index (TPI) and antioxidant activity (AA) were measured by spectrophotometric assays. Eighteen polyphenolic compounds were identified in all investigated apples by HPLC-DAD and LC-(ESI)-MS. The peel of traditional apple varieties had higher contents of all investigated polyphenols. Calculated relative contribution of polyphenol groups indicated non-flavonoids (28.6%) and flavanols (46.2%) as the major contributors to the total polyphenolic content in traditional and commercial apple varieties, respectively. The most abundant polyphenol in traditional apple peel was chlorogenic acid, procyanidin B2 and epicatechin (1143 ± 755 μg/g dw, 954 ± 343 μg/g dw and 560 ± 362 μg/g dw, respectively). The peel of varieties ‘Apistar’, ‘Bobovac’ and ‘Božićnica’ could be highlighted as an important source of polyphenols.

Abstract:In the selection of apple ( Malus domestica Borkh) fruits, consumers predominantly use visual and organoleptic responses, such as aroma and texture, as quality/preference markers. Previous studies profiling the sensory properties and aroma profiles of apple varieties have provided a lexicon describing important attributes within the modalities of aroma, texture, and flavour. The range of volatile organic compounds (VOCs) produced by four apple cultivars was assessed by thermal desorption-gas chromatography-mass spectrometry (TD-GC-MS). To facilitate automated dynamic headspace extraction of the VOCs from bulk samples, a small multichamber thermal extraction system was used. Forty compounds were detected and putatively identified by mass spectral comparison with NIST libraries. Eight compounds were not previously identified in apples. Permutational multivariate analysis of variance (PerMANOVA) and canonical analysis of principal coordinates (CAP) done on the whole volatile organic compound profiles show separation of the four cultivars. The use of random forest classification enabled the determination of a minimum set of fifteen potential VOC markers that successfully differentiate the cultivars under study. Trends across samples, VOCs, and sensory evaluation are revealed. The association between 4-hexen-1-ol acetate with texture/consistency, and crispiness, and correlation of 2-methylbut-2-en-1-yl acetate with juiciness and acidity is shown for the first time. These sensory correlations indicate that the compounds determined by this experiment could be used as objective markers for the consumer appreciation of fresh apples, enabling the optimum conditions for processing and storage of individual cultivars to be identified without recourse to expensive sensory panels in every case. Such tests could also be used as part of routine quality control by the producer and retailer, reducing costs and eliminating waste due to batches of inferior product.

Abstract:In this research, we compared the phenotypical characters, total anthocyanins content, total phenols content, and antioxidant activity of red-fleshed apple cultivars ‘XJ4’, ‘QN-5’, ‘DH’ and ‘HX1’ at three fruit developmental stages. A further flavonoids metabolites study was conducted in ‘XJ4’ and ‘DH’. We found broader variation of total anthocyanins content in the peel of the four cultivars, which might result in larger differences of free radicals scavenging rate. The most significant difference in fruit phenotype, anthocyanins content, and DPPH scavenging rate was observed between ‘XJ4’ and ‘DH’ at mature stage. Therefore, the flavonoids metabolites of ‘XJ4’ and ‘DH’ at mature stage were compared to unveil the details of anthocyanins compounds. The unique compounds pelargonidin 3-O-β-d-glucoside and cyanidin-3-O-malonylhexoside were detected only in peel and flesh of ‘XJ4’ but not in ‘DH’, which might contribute to the purple peel and dark-red flesh color of ‘XJ4’. Significantly decreased upstream metabolites in the early biosynthetic genes regulated domain were found only in ‘XJ4’ peel but not in the flesh. This might explain why the anthocyanins content in ‘XJ4’ peel was decreased largely at the mature stage. Taken together, our findings will give some insight into the metabolites study in flavonoid biosynthetic pathway of red-fleshed apple.

Abstract:Apples (Malus domestica Borkh) are of particular interest for their high content of healthy phytochemicals. This study investigated the antioxidant activity and polyphenolic compounds of extracts from the fruits from Ozark Gold, Starkinson and Kosztela cultivars and additionally leaves from Ozark Gold cv. Phenolic compounds were identified and quantified by liquid chromatography- mass spectrometer (LC–MS) and high pressure liquid chromatography (HPLC). The samples were collected during fruit development at 60, 80, 130 and 145 days after full bloom. The concentration of apple phenolics was highest early in the season and decreased during fruit development. The leaf phenolics had a more steady level during all the period of collection than the fruits. Among the five groups of analyzed polyphenols, the procyanidins predominated in apple fruits and dihydrochalcones in leaves. The concentration of polyphenols decreased from 39.15, 5.97 and 33.39 g/kg dw (collected at 60 days after full bloom) to 14.22, 2.05 and 9.31 g/kg dw (collected at 145 days after full bloom) for apples Ozark Gold, Starkinson, Kosztela, respectively. The leaves characterized a much higher content of polyphenolic compounds and antioxidant capacity than unripe fruits. Antioxidant capacity measured by 2,2-azinobis-(3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-picrylhydrazyl (DPPH) methods was higher when the apple was unripe and the leaves were young. The results indicate that, unripe fruits and leaves are very good raw material for polyphenol nutraceutical production with high antioxidant potential.

Abstract:Thirteen apple cultivars were analyzed for their total phenolic content, total flavonoids, anthocyanins, ascorbic acid in methanolic extracts of both peel and cortex fractions. Three in vitro assays (FRAP, DPPH, and CUPRAC) were used to determine the antioxidant activity. Concentration of the phytochemicals studied varied greatly between the apple peel and the cortex region. Peels showed ~ 2.8 times higher total phenolic content and ~ 2.68% higher flavonoid content than the cortex. Principal component analysis could successfully explain 76.86% and 84.27% variability in the antioxidant determinants (antioxidants/assays) in the peel and cortex region of apple cultivars, respectively. Major contributor for antioxidant activity in both apple peel and cortex was total flavonoid content. Cultivars 'Well Spur' and 'Oregon Spur II' were found to be substantially rich in these two antioxidants. The antioxidant activity was best expressed by the in vitro FRAP assay in both the fractions. Non-hierarchical K-medoids clustering reflected the presence of an antioxidant/ assay protocol apart from the antioxidant/assay we considered in this study that needs further exploration to get full spectra of antioxidant profile across apple genotypes. Based on multivariate analysis and the concept of RACI, the FRAP antioxidant assay is recommended for determining antioxidant activity in apples.

Abstract:Significant variation in organic acid components was detected in mature fruits of 101 apple accessions using high-performance liquid chromatography. The Malus species predominantly accumulated malic acid and citric acid, whereas wild fruits exhibited significantly higher levels of organic acid content than that in cultivated fruits. Differential accumulation patterns during fruit developmental stages was detected between malic acid and citric acid, thus suggesting a complex genetic regulation mechanism of organic acid metabolism in apple fruit. A highly positive correlation was detected between fruit total organic acid content with malic acid and citric acid content, thus suggesting that malic acid and citric acid are the principal determinants of apple fruit acidity. In contrast to malic acid, citric acid was predominantly detected in partial wild apples, while extremely low to undetectable concentrations of citric acid were observed in cultivated apple fruits; this is likely due to the genetic effects of parental characters. Our results provide vital information that could be useful for future studies on genetic analysis and improvement of organic acid accumulation in apple fruits.

Abstract:Aronia (chokeberry, Aronia melanocarpa) is a valuable fruit that combines the health benefits of green tea and red wine and is gaining popularity worldwide. Aronia has a unique chemical composition with strong antioxidant properties, including anthocyanins and chlorogenic acids (CGAs). However, it remains unclear which specific compounds or groups are primarily responsible for the antioxidant properties of chokeberry. Therefore, an analysis of the antioxidant properties of aronia berries based on geographical region and their variability during ripening (from green to frostbitten fruit) was conducted. The fruits were collected from three locations for two years. The aim of our work was to identify the compounds responsible for the antioxidant properties of aronia berry extracts by using chemometric methods. The analyses of anthocyanins and CGAs were performed using HPLC-DAD, and the antioxidant capacity was assessed by FRAP and DPPH methods. The PCA analysis also considered variations in temperature and precipitation. The chemometric analysis revealed a strong correlation between radical-scavenging properties and the content levels of chlorogenic acids. The results obtained in this study show that unripe green chokeberry fruits exhibit the highest antioxidant properties, which can be attributed to the high content of CGAs at this stage.

Comparative Nutrient Element and Antioxidant Characterization of Berry Fruit Species and Cultivars Grown in Hungary

Abstract:Fruits of four berry species (strawberry, raspberry, red and black currants) were compared in their elemental composition (Al, B, Ba, Ca, Cu, Fe, K, Mg, Mn, Na, P, and Zn) and redox parameters involving total phenol content (TPC), ferric reducing ability (FRAP), 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, and total radical scavenger activity (TRSA). Berry cultivars contained significantly higher ( P ≤ 0.05) amounts of most detected elements as compared with apple with many maximum elemental values demonstrated by the black currant ‘Otelo’. Black currant also had the greatest antioxidant capacity as demonstrated through all conducted assays. The results obtained through FRAP, TPC, and TRSA assays were closely correlated, whereas TRSA and DPPH varied independently. Our study provides valuable information on the antioxidant capacity of several berry species grown in Hungary and highlights the crucial influence of cultivar on elemental content and antioxidant power of berry fruits. This study demonstrated the ability to select berry cultivars for specific nutritional purposes or assign parental lines in functional breeding programs.
