Abstract:Natural deep eutectic solvent (NADES) has received increasing interest as a green alternative to traditional organic solvents for efficient extraction of bioactive compounds from natural sources. In this study, phytochemicals in Carya cathayensis Sarg. peels extracted with Choline chloride-Malic acid (ChCl-MA) were identified using UPLC-Triple-TOF/MS. Effect of NADES on phenolic composition, antioxidant properties and inhibition of α-glucosidase and α-amylase were evaluated. Furthermore, extraction mechanism caused by different solvents were investigated by quantum chemical calculation combined with molecular dynamic simulation. A total of 29 phytochemicals were identified, and catechin, procyanidin B1, 2,3-dihydroxybenzoic acid, pinocembrin, procyanidin B3, myricetrin were the most abundant compounds. The extract using ChCl-MA exhibited the highest phenolic compounds content, antioxidant capacity, and α-glucosidase and α-amylase inhibition activities. Larger solvent accessible surface area, more hydrogen bonds between ChCl-MA and extract, longer lifetime of the hydrogen bonds, and lower intermolecular interaction energy account for higher extraction efficiency of ChCl-MA.

Natural deep eutectic solvent enhanced pulse-ultrasonication assisted extraction as a multi-stability protective and efficient green strategy to extract anthocyanin from blueberry pomace

Abstract:A deep understanding upon the relation between anthocyanin extraction performance and physicochemical properties of natural deep eutectic solvents (NADESs) was obtained, and a high-efficiency NADES (Choline chloride-Oxalic acid (ChOa)) was selected and coupled with pulse-ultrasonication for anthocyanin extraction from blueberry pomace. Response surface methodology was adopted to analyzed the significant factors' influence, interactive effects on extraction yield and optimize the extraction conditions. The theoretical optimal conditions with highest yield of total anthocyanins content (TAC) were 3.2 min of ultrasonic time, 325 W of ultrasonic power, 349.15 K of temperature, 60 mL/g of solvent to solid ratio, under which the experimental TAC value (24.27 +/- 0.05 mg C3GE/g DW) was highly correlated with theoretical result (24.28 mg C3GE/g DW). Multi-stability protective effect of ChOa on anthocyanin over acidified ethanol was revealed by stability assays in high temperature, varying pH and light exposure conditions. FT-IR and DSC analysis further characterized the influence of ChOa over acidic ethanol on typical anthocyanins functional groups and thermal behavior of anthocyanin extracts. UPLC-Triple-TOF/MS analysis identified 10 anthocyanins that mainly consisted of delphinidin, petunidin, and malvidin compounds. Overall, this study provided a green, high-efficiency and multistability protective strategy for the recovery of anthocyanins from plant source by-products.

Abstract:An excellent high-efficiency natural deep eutectic solvent (NADES, ChCl-MA) was screened out and integrated with pulse-ultrasonication technique for extracting phenolic compounds from Carya cathayensis Sarg. peels (CCSPs). Single factor experiment combined with response surface methodology (RSM) using Box-Behnken design (BBD) were employed to investigate significant factors and optimize their influence on extraction of phenolic compounds. Significant synergistic effect triggered by ChCl-MA based pulse-ultrasonication over other methods used alone were proved by comparative study concerning a variety of bioactive components and antioxidant activities. The second-order kinetic model was developed and validated (R2 > 0.99) to describe the extraction process and its mechanism; and second-order kinetic extraction rate constant (k), saturation concentration (Cs), and initial extraction rate (h) were calculated. FT-IR, DSC and SEM results further demonstrated synergistic effect and influence during extraction. Overall, this study provided a green and high-efficiency alternative for the recovery of various phenolics compounds from plant source by-products.
De-glycosylation and enhanced bioactivity of flavonoids from apple pomace during extraction with deep eutectic solvents

Abstract:Deep eutectic solvents (DES) are attracting great attention as a green alternative to hazardous organic solvents for the extraction of valuable bioactive compounds from agricultural and processing waste. In this study, the simultaneous extraction and de-glycosylation of quercetin derivatives from apple pomace was achieved by using various DES made of choline chloride and different types of carboxylic acids. The content of quercetin derivatives was similar in DES and conventional methanol extracts, but their relative amounts were distinctively different due to de-glycosylation during DES extraction. Chromatographic and spectroscopic analyses allowed the determination of the structures of the glycosylated quercetins, and revealed that the proportion of the quercetin aglycon in DES extracts can be controlled by the DES composition, extraction temperature and duration of extraction. Of all quercetin glycoconjugates present, quercetin-3-O-alpha-arabinofuranoside was more prone to de-glycosylation during DES extraction than any other quercetin derivative. Furthermore, the antioxidant activity of the DES extracts was higher than that of methanol extracts, likely due to the increase of quercetin aglycon content during extraction. These data show the potential to extract and de-glycosylate quercetin derivatives in a single step using green DES to generate bioactive preparations with enhanced activity.

Abstract:Supercritical fluid extraction (SFE) was used to extract bioactive compounds from apple (Malus domestica) peel waste from three different Italian cultivars. The bioactive fractions were extracted applying a temperature of 60 °C and a pressure of 250 bar for 15 min with 20% ethanol as co-solvent, at a flow rate of 2 mL/min. The total polyphenol (TP), anthocyanin (TA), ascorbic acid (AA), and antioxidant activity contents (TACs) were measured, while chromatographic analyses were performed to highlight the differences between the extracts. The Stark cultivar had the highest levels of polyphenols, anthocyanins, and ascorbic acid, while the Royal Gala cultivar showed the highest total antioxidant activity. SFE extracts were then tested for their effect on the mitochondrial NADH–ubiquinone oxidoreductase (Complex I) activity on mitochondria isolated from human embryonic kidney cells (HEK239). The Stark extract showed the most positive response in terms of NADH oxidation. The results obtained in this work highlight the potential of apple peel waste as a source of functional phytocompounds and suggest that Stark cultivar extracts may be exploited for pharmacological applications. This study supports the circular bioeconomy by promoting the use of waste products as a valuable resource.

Abstract:UNLABELLED A method based on stir bar sorptive extraction combined with gas chromatography-mass spectrometry detection (SBSE-GC-MS) has been optimized with the aim of applying it to the analysis of apple pomace. The method allowed the identification of 124 volatile compounds after 3 h of extraction with a precision (RSDs) ranging between 2% (linalool) and 11% (ethyl hexanoate). Its use in analyzing varietal apple pomace revealed the interest of this substrate as regards its content in aromas. From a semi-quantitative point of view, the higher content in aldehydes and esters of the Blanquina variety is worth highlighting, as are the greater concentration of acids in the Clara variety and the higher content of terpenes and norisoprenoides in the Coloradona variety. In contrast, the Ernestina and Perico varieties presented the lowest levels of aromas. PRACTICAL APPLICATION The analysis of varietal apple pomaces showed the importance of this type of waste in the food industry, both for its content in aromas such as for its use as substrate in biotechnological processes.

Abstract:Background: Apple pomace has garnered significant attention within the life sciences domain due to its underutilized status as a waste material from apple processing. It represents a cost-effective and abundant source of triterpene acids due to its multifunctional clinical, nutritional, and pharmaceutical benefits. Purpose: The present study aimed to develop and validate a new, selective, effective, robust and reproducible laboratory methodology based on extraction, purification and analytical procedures to obtain and determining three major triterpene acids – Ursolic acid (UA), Oleanolic acid (OA) and Betulinic acid (BA) into the dry extracted product from apple pomace. Method: A new, cost-efficient, rapid, selective and high-yield two-stage ultrasound-assisted extraction procedure was developed and the effect of critical parameters: ultrasonic power, extraction time, solvent volume, temperature, and the amount of raw material on the extraction process were investigated. The dry column vacuum chromatography technique was used for purification to remove unwanted non-polar and polar impurities from the target bioactive compounds; A new, effective, specific, sensitive, and rapid HPLC analytical procedure was developed using analytical quality by design (AQbD) approach and validated according to ICH guidelines. Conclusion: The method has a good accuracy (the mean recovery >95 %) and linearity (R2>0.999). The limit of quantitation (LOQ) is 0.0001 mg/mL for UA, 0.00005 mg/mL for OA and 0.000025 mg/mL for BA. The validation results confirm that the method is specific, precise and robust. The purity of the extracted and purified target product from apple pomace is not less than 93 %. The developed laboratory methodology is capable of being considered for industrial purposes and through the appropriate technology transfer process can be successfully transferred to the industrial scale.

Abstract:The cider-making industry in Asturias generates between 9000 and 12,000 tons of apple pomace per year. This by-product, the remains of the apple pressing, and made up of peel, flesh, seeds and stems, is a valuable material, containing substantial amounts of antioxidant compounds associated with healthy properties. Polyphenols such as dihydrochalcones and quercetin glycosides, and triterpenic acids, among which ursolic acid is a major compound, are the main antioxidant families described in apple pomace. The simultaneous recovery of those families has been accomplished by low frequency ultrasound-assisted extraction. Working extraction conditions were optimised by response surface methodology (RSM): time, 5.1 min; extractant composition, 68% ethanol in water; solid/liquid ratio, 1/75 and ultrasonic wave amplitude, 90%. This procedure was further applied to analyse those components in the whole apple pomace (WAP), apple peel (AP) and apple flesh (AF). On average, dry WAP contained almost 1300 μg/g of flavonols, 1200 μg/g of dihydrochalcones and 4200 μg/g of ursolic acid. These figures increased in the apple peel to, respectively 2500, 1400 and 8500 μg/g dry matter. Two linear multivariate regression models allowed the antioxidant activity of apple by-products to be predicted on the basis of their bioactive composition. The results derived from this study confirm the potential of industrial cider apple pomace as a source of high-value bioactive compounds, and the feasibility of the ultrasound-assisted extraction technique to recover those components in a simple and efficient way.

Extraction, Characterization, and In Vitro Biological Activity of Polyphenols from Discarded Young Fig Fruits Based on Deep Eutectic Solvents

Abstract:(1) Background: Discarded young fig fruits (DYFFs) result in a waste of resources, such as sparse fruits and residual fruits, and there has been no research on the relationship between phenolic compounds and biological activity in DYFFs (2) Methods: Different deep eutectic solvents (DESs) and 80% ethanol were used to prepare DYFF extracts, and polyphenol extraction efficiency and bioactivities in the DYFFs extracts were compared. (3) Results: More than 1700 phytochemicals were identified in DYFFs, and thirteen of these typical phenolic compounds were analyzed quantitatively; chlorogenic acid, rutin, luteolin 8-C-glucoside, and epicatechin are the main polyphenols in DYFFs, especially chlorogenic acid with 2720-7980 mg/kg. Ferulic acid, caffeic acid, epicatechin, (+)-catechin, luteolin 8-C-glucoside, rutin, hesperetin, and chlorogenic acid showed different degrees of correlation with in vitro antioxidant activity. Moreover, the highest total phenol content found in the extracts of ChCl-Ethylene glycol (Choline chloride:Ethylene glycol = 1:2) was 8.88 mg GAE/g DW, and all quantitatively analyzed phenolic compounds had high levels in various DESs and 80% ethanol. The 80% ethanol and Choline chloride (ChCl) solvent system showed the greatest antioxidant properties, and the Choline chloride-Urea (Choline chloride: Urea = 1:2) extract of DYFFs exhibited the highest inhibitory activity. (4) Conclusions: DESs have demonstrated potential as promising green solvents, especially the ChCl solvent system, which facilitates the extraction of polyphenols.
Multifaceted analyses disclose the role of fruit size and skin-russeting in the accumulation pattern of phenolic compounds in apple

Abstract:Fruits are nowadays considered important suppliers of anti-oxidant molecules. Apples are particularly rich in phenolic compounds, non-nutritional phytochemicals that play active roles in controlling severe chronic diseases. In this work, 19 phenolic compounds were investigated in both skin and pulp tissues of seven apple accessions across the Malus genus collected at two stages: during fruit development and at harvest. The primary difference in phenolic concentration between wild and domesticated accessions, especially in the pulp, could be explained by the larger growth rate of the domesticated varieties. The proposed dilution effect was also confirmed through the observation of the increased content of procyanidin B2+B4 and phloridzin in russet-skinned apples, known to have higher concentrations of these compounds. The metabolite screening was also accompanied by the expression analysis of 16 polyphenolic genes showing, for nine elements, a higher expression at harvest than during fruit development. Finally, a polyphenolic comparison with red-fleshed apples was also carried out, underlying a larger amount of procyanidins and quercetin-3rhamnoside in the white-fleshed accessions. The results presented and discussed in this work suggest that specific white-fleshed apples, especially with russeted-skin, may play an important role in ameliorating the nutraceutical potential of apple fruit.
Chemical Composition, Antioxidant Activity and Cytocompatibility of Polyphenolic Compounds Extracted from Food Industry Apple Waste: Potential in Biomedical Application

Abstract:Apple pomace (AP) from the food industry is a mixture of different fractions containing bioactive polyphenolic compounds. This study provides a systematic approach toward the recovery and evaluation of the physiochemical and biological properties of polyphenolic compounds from AP. We studied subcritical water extraction (SCW) and solvent extraction with ethanol from four different AP fractions of pulp, peel, seed, core, and stem (A), peel (B), seed and core (C), and pulp and peel (D). The subcritical water method at the optimum condition resulted in total polyphenolic compounds (TPC) of 39.08 ± 1.10 mg GAE per g of AP on a dry basis compared to the ethanol extraction with TPC content of 10.78 ± 0.94 mg GAE/g db. Phloridzin, chlorogenic acid, and quercetin were the main identified polyphenolics in the AP fractions using HPLC. DPPH radical scavenging activity of fraction B and subcritical water (SW) extracts showed comparable activity to ascorbic acid while all ethanolic extracts were cytocompatible toward human fibroblast (3T3-L1) and salivary gland acinar cells (NS-SV-AC). Our results indicated that AP is a rich source of polyphenolics with the potential for biomedical applications.
Extraction of Polyphenolic Antioxidants from Red Grape Pomace and Olive Leaves: Process Optimization Using a Tailor-Made Tertiary Deep Eutectic Solvent

Abstract:In the framework of introducing green strategies for food processing, the industrial orientation has shifted towards the replacement of conventional petroleum-based solvents with alternative eco-friendly ones. On this basis, the objective of this study was to synthesize a novel, tertiary, food-grade deep eutectic solvent, composed of glycerol, citric acid, and L-proline (GL-CA-Pro), and to test it as a solvent for the extraction of polyphenols from agri-food waste biomass. After an initial screening on various common residual materials (apple peels, lemon peels, orange peels, red grape pomace, olive leaves), evidence emerged that indicated GL-CA-Pro was more effective than other DESs commonly used for polyphenol extraction. Furthermore, extracts from red grape pomace (RGP) and olive leaves (OLL) were shown to contain higher level of total polyphenols and increased antioxidant activity. Process optimization for those two materials with the response surface methodology revealed that the major difference pertained to the extraction time. In addition, for both materials, GL-CA-Pro was shown to provide higher total polyphenol yields (53.25 and 42.48 mg gallic acid equivalents per g of dry mass, respectively) compared to water and 60% aqueous ethanol. However, the chromatographic analyses for OLL suggested aqueous ethanol was a more suitable solvent for some principal polyphenolic constituents. The RGP extract produced with GL-CA-Pro exhibited significantly stronger antioxidant effects compared to the aqueous and hydroethanolic extracts, but the outcome for the OLL extracts was diversified. It was concluded that GL-CA-Pro is a very efficient solvent for RGP polyphenols, but its efficiency regarding OLL was comparable to that of aqueous ethanol.

Abstract:Natural deep eutectic solvents (NADES) are promising green solvents for the extraction of compounds from natural products. This is the first study to use a sample preparation method based on NADES for the extraction of flavonoids (rutin, hesperidin, neohesperidin, naringenin, naringin, quercetin, hesperetin, and chrysin) from fruits, vegetables, and spices. In total, 17 types of NADES based on choline chloride, acetylcholine chloride, choline tartrate, betaine, and carnitine with different compositions were tailored to test their extraction efficiency. Operational conditions such as water content in NADES and liquid/solid ratios were also studied. A response surface methodology was used for multivariate optimization of some extraction parameters. Efficient recovery of extracted flavonoids (higher than 70%) was achieved using a 30% water solution of acetylcholine chloride/lactic acid ratio (2:1) as an extraction solvent. Other conditions for SLE-NADES were as follows: liquid/solid ratio 3:1, extraction temperature 60 °C, extraction time 30 min, and stirring speed 1400 rpm. Each extract was analyzed by UHPLC with UV and MS/MS detection. The developed method was applied for analysis of flavonoids in fruits (cranberry, fruits of Lycium barbarum L., grape, plum, and orange peel), vegetables (onion and broccoli), and spices (mustard, rosemary, and black pepper).

Abstract:Apple peel polyphenols were extracted by ultrasound-assisted extraction (UAE) and conditions for the extraction of polyphenols were optimized by the response surface methodology. The optimized conditions determined as 32 min of extraction time, 29 °C of extraction temperature and ethanol concentration of 56% with a yield of 35.08 ± 0.26 mg gallic acid equivalent/g dry weight. Six resins were used for the purification and D101 resin showed the highest ratio of desorption for polyphenols and further applied for dynamic adsorption/desorption test. Catechin, procyandin B, querecitin-3-galactoside and querecitin xyloside were identified in purified polyphenols by HPLC-MS/MS. Finally, the immunostimulatory activity in vitro of purified polyphenols on RAW264.7 cell lines was evaluated. The apple peel polyphenols exhibited a dose-dependent effect (p < 0.05) on the strongest propagation of RAW264.7 cells. The results demonstrate that apple peel is a valuable source of polyphenols and could be used as a natural immunostimulating agent for application in functional foods.

Approximately 65% of annual apple production is transformed into apple juice concentrate generating a large amount of waste material named apple pomace, which includes seeds, skin, and other components. Disposing of apple by-products directly into the environment constitutes a source of environmental pollution due to its high-water content and easily fermentable nature. Apple pomace is rich in polyphenols that can be utilized as active components in cosmetic, nutraceutical, or pharmaceutical products. The present study aims to describe and compare different physical methods for the extraction of polyphenols from apple pomace. Water was used as the extraction solvent in thermal-stirred extraction (TSE), ultrasound-assisted extraction (UAE), and microwave-assisted extraction (MAE). The best extraction conditions were identified in terms of solid to solvent ratio, temperature, power, and time through a kinetic study. The best extraction parameters were compared environmentally on a pilot scale through a life cycle assessment (LCA). All the results demonstrated the MAE is the best technique to extract polyphenol from apple pomace in terms yield and environmental impact proving that it is possible to transform waste into a sustainable source of bioactive ingredients.

Abstract:Apple pomace represents an underexploited source of bioactive compounds. This study examines the optimization of total phenolic content (TPC) and antioxidant extraction yield of apple pomace (variety: Belorusskoje malinovoje) using response surface methodology. The green extraction technique used was ultrasound-assisted extraction, and it was compared with conventional solvent extraction. The impact of extraction time and amplitude of ultrasound-assisted extraction on the yield of polyphenols and antioxidants has been evaluated. Total phenolic content was determined using an established TPC assay. The antioxidant activity of the apple pomace was determined using established assays 2,2-diphenyl-1-picrylhydrazyl (DPPH•) and 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS•+). Furthermore, the potential of apple pomace as a feed material was explored by assessing its nutritional composition, vitamins, minerals, fatty acids, and amino acid content. The extraction of antioxidants and phenolic compounds was efficiently optimized using RSM. The optimal conditions for TPC and DPPH• analyses were achieved with an extraction time of 17.5 min and an ultrasound-assisted extraction amplitude of 20%. Optimal conditions for ABTS•+ were 5 min extraction time and 20% amplitude. Conventional and ultrasound-assisted extraction methods yielded comparable results. Moreover, apple pomace exhibits potential as a feed ingredient despite its modest protein content. This study contributes to the utilization of apple pomace by providing additional information on its antioxidant content and nutritional composition, thus contributing to its sustainable utilization in various industries, especially the livestock feed sector.

Abstract:Herein, new function-oriented deep eutectic solvents (DESs) with on l-proline or betaine and DL-lactic acid were designed extraction of polyphenol compounds from grape pomace. The formations of new hydrogen between the two components of DESs were evidenced by FTIR and 1 H NMR spectra. Physicochemical characteristics of deep eutectic solvents under the effect of water on density, viscosity, electrical resistance have been revealed. The betaine-based DES could changes the electrical resistance depending on the concentration of water. The feasibility of using ultrasound-assisted extraction of phytochemicals combined with betaine-based and proline-based DES as a green chemistry technique to improve extraction of green (polyphenols) compounds from grape pomace was evaluated. Grape pomace extracts were analyzed by LC-DAD-MS to identify extracted compounds and analyze the impact of type DES on compound chemical profile. LC results indicated that ultrasound-assisted extraction of phytochemicals using betaine-based and proline-based deep eutectic solvent application allowed enhanced hydroxycinnamic acids extraction, while the use of proline-based DES resulted in a more diverse compound profile. The highest recovery efficiencies were recorded with DESs giving the highest total phenolic content (80–92 mg EGE//g d.w), total flavonoid content (47–51 QUE/g d.w), antiradical activity (80% at 100 mg/ml) and reducing power (310–468 mg ASE/g extract). Scanning electron microscope (SEM) results further verified DESs intensified the destruction of cell wall structure, and thereby allowed greater release of polyphenolic from grape pomace.

Abstract:Background: Pentacyclic triterpenoid acids are common in a number of food and spice plant species. Apples (Malus domestica) are the most common human food source for these potentially beneficial phytochemicals. Pre-20th century heritage apples have long been grown in mountainous western North Carolina and may be a wide-ranging source of these phytochemicals. Objectives: Existing extraction and analytical methods were improved and used to assess the content of three triterpenoid acids in heritage apple cultivars grown in western North Carolina, USA.Methods: Apples from a local farmers market were collected during the fall apple season. Apple peels were freeze dried, ground, and extracted with ethanol thrice. Extracts were analyzed by HPLC against external standards for betulinic, oleanolic, and ursolic acids.Results: The improved method was used to extract and to analyze the triterpenoid acid levels in 16 heritage apple cultivars grown in the Appalachian region of western North Carolina. Total triterpenoid acids ranged from 2 to 29 mg/g dry weight of peels. Content did not vary by apple color or time of harvest. Russeted varieties contained noticeably less triterpenoids.Conclusions: An improved and simplified method was used for the analysis of heritage apple varieties in western North Carolinas.
