Abstract:When applied to a pure component suspension in an apolar solvent, a strong inhomogeneous electric field induces particle movement and the particles are collected at the surface of one of the two electrodes. This new phenomenon was used to separately isolate two organic crystalline compounds, phenazine and caffeine, from their suspension of 1,4-dioxane. First, the crystals of both compounds were collected at different electrodes under the influence of the electric field. Subsequent cooling crystallization allowed the immobilization and growth of the particles on the electrodes, which were separately collected after the experiment with purities higher than 91%. This method can be further developed into a technique for crystal separation and recovery in complex multicomponent suspensions of industrial processes. Crystallization is an effective and efficient separation technology that can, in a single process step, recover the desired compounds from solutions as high purity (>99%) crystalline solids [1,2,3,4] . However, such highly purified product is hard to obtain from a multi-component solution by direct crystallization, such as the product stream from a type-I Multicomponent Reaction (MCR), [5,6] or a racemic mixture of chiral pharmaceutical compounds, [7] since a mixed suspension is a likely result. Further purification of the solid phase usually requires additional steps (see Figure 1 (a) for instance), which will inevitably lead to the loss of valuable products. Alternatively, a single crystallization step coupled with simultaneous particle separation could diminish the product loss while effectively recovering desired solute from the mixed solution. Particle manipulation and subsequent separation can be achieved by the application of electromagnetic fields: a magnetic field could separate polymorphs, crystalline solids with different solid-state structures, to prepare seeds for large-scale crystallization, provided that a suitable paramagnetic medium can be found. [8] Alternatively, an electric field has been used in the successful capturing or manipulation of desired nanoparticles [9] and the assembly of colloid particles for the production of, for instance, optically tunable micropatterns, biosensors, and biofuel cells. [10-12] In all of these cases, the liquid phases did not contain any dissolved particle materials. On the other hand, the electric field has been used to localize protein crystallization and control its nucleation rate from a clear solution, in absence of any seed crystals. [13,14] However, up till now, the combination of particle manipulation by an electric field with the crystallization process for the in-situ product separation and recovery has, to our knowledge, never been attempted. In the present study, the experimental proof is given for an electric field enhanced crystallization (EEC) process by which the two solutes from a multi-component solution are separately recovered, as schematically demonstrated in Figure 1 (b). Subjected to an inhomogeneous electric field, particles in a suspension experience dielectrophoretic (DEP) and possibly electrophoretic (EP) forces: DEP originates from the difference in dielectric constants of the particles and the surrounding medium and drags the particles with larger dielectric constant towards the location of the electric field maxima, [15,16] while EP forces, which work only on charged particles, are directed towards the electrode with the counter charge. [17] To prevent electrochemical reactions, which would reduce the product yield, the current through a crystallizing suspension should be minimized. Meanwhile, the inhomogeneous field needs to be sufficiently strong and the dielectric constant of the solvent should be sufficiently small to ensure the effective manipulation of crystals by the DEP force. Therefore, the apolar and nonconductive solvent 1,4-dioxane was used in this study. We first applied an inhomogeneous electrostatic field to a 6 mL isonicotinamide (INA) suspension in 1,4-dioxane (ca. 18 mg solid/ml solvent), generated by a Direct Current (DC) potential difference between two parallel rod-shape electrodes roughly 6 mm apart, which were immersed into the suspension. Microfluidic platform for optimization of crystallization conditions

Abstract:We describe a universal, high-throughput droplet-based microfluidic platform for crystallization. It is suitable for a multitude of applications, due to its flexibility, ease of use, compatibility with all solvents and low cost. The platform offers four modular functions: droplet formation, on-line characterization, incubation and observation. We use it to generate droplet arrays with a concentration gradient in continuous long tubing, without using surfactant. We control droplet properties (size, frequency and spacing) in long tubing by using hydrodynamic empirical relations. We measure droplet chemical composition using both an off-line and a real-time on-line method. Applying this platform to a complicated chemical environment, membrane proteins, we successfully handle crystallization, suggesting that the platform is likely to perform well in other circumstances. We validate the platform for fine-gradient screening and optimization of crystallization conditions. Additional on-line detection methods may well be integrated into this platform in the future, for instance, an on-line diffraction technique. We believe this method could find applications in fields such as fluid interaction engineering, live cell study and enzyme kinetics.

Abstract:Antisolvent crystallization can be used as an alternative to cooling or evaporation for the separation and purification of solid product in the pharmaceutical industry. To improve the process understanding of antisolvent crystallization, the use of in-line tools is vital. In this study, the process analytical technology (PAT) tools including focused beam reflectance measurement (FBRM), particle video microscope (PVM), and near-infrared spectroscopy (NIRS) were utilized to monitor antisolvent crystallization of sodium scutellarein. FBRM was used to monitor chord count and chord length distribution of sodium scutellarein particles in the crystallizer, and PVM, as an in-line video camera, provided pictures imaging particle shape and dimension. In addition, a quantitative model of PLS was established by in-line NIRS to detect the concentration of sodium scutellarein in the solvent and good calibration statistics were obtained (r(2) = 0.976) with the residual predictive deviation value of 11.3. The discussion over sensitivities, strengths, and weaknesses of the PAT tools may be helpful in selection of suitable PAT techniques. These in-line techniques eliminate the need for sample preparation and offer a time-saving approach to understand and monitor antisolvent crystallization process.

Abstract:Continuous flow crystallization is an attractive mode of operation, due to its ability to generate consistent product quality while requiring a smaller footprint and lower production costs than its batch counterpart. We present a novel combination of a custom/in-house automated continuous crystallization platform integrated with self-optimization algorithms. We demonstrate the automated optimization of continuous crystallization of nirmatrelvir (PF-07321332), one of the active ingredients in PaxlovidTM, a potent, selective, and orally bioavailable inhibitor of SARS-CoV-2 Mpro. The continuous crystallization platform consists of three mixed suspension mixed product removal (MSMPR) crystallizers in series and includes an in-house designed automation user interface integrated with lab equipment. The platform also has an iterative design of experiments (DoE) based on mixed-integer nonlinear programming (MINLP) self-optimization algorithms. We implement automated controls for the lab equipment, including a flow sonication cell as the nucleation device, feed pumps, temperature controller units (TCUs), thermocouples, pressure sensors, stirrers, and coriolis mass flow meters. We enable integration of variety of in-situ process analytical technologies (PATs) via open platform communications unified architecture (OPC UA), including Mettler Toledo ParticleTrackTM with FBRM® (Focused Beam Reflectance Measurement) technology and BlazeTM Metrics imaging probe for the data visualization and real time process understanding.

Abstract:The ability to quickly generate and identify crystalline solids for organic compounds in a parallel fashion requires a rapid, adaptable crystallization screening strategy that delivers reliable, valuable, and consistent results. The key to the system is a standard platform small-scale (0.5–2 mg) crystallizer screening array that reproducibly crystallizes compounds and facilitates the presentation of crystallization samples to both an automated polarized light microscope and an instrument capable of PXRD analysis. Data science technologies were leveraged to streamline the workflow of data visualization and processing. The fully developed workflow successfully used both single-crystal and PXRD analyses to identify multiple polymorphs of a test compound in a single screening experiment on 200 mg of input material with commercially available crystallizers and instruments to perform a highly detailed crystallization screening study. The methods and techniques described herein are fully transferrable to those working in the synthetic organic chemistry field.

Abstract:With the growing problem of drug shortages worldwide, a solution could be a centralized compact pharmaceutical manufacturing platform that produces API and finished drug product. In this paper, the development of a combined crystallization, hybrid filtration-drying-dissolution apparatus to be used in such a compact platform is discussed. Experiments were conducted to evaluate each unit operation. Crystallization experiments using a conventional stirred tank and the newly designed scraped surface crystallizer demonstrated the advantages of the new design in terms of crystallization rates, yields and the ease of automation. Postcrystallization operations were operated stepwise using the custom hybrid device that delivered satisfactory results for each operation, e.g. after filtration, Fluoxetine HCl was dried in less than 20 min, with 99% yield after dissolution in the liquid excipient.

Automation plays a vital role in reducing development time in the pharmaceutical industry, both for drug substance (DS) and drug product (DP). Many aspects of crystallization development (solubility, kinetics, solid form characterization, concentration) have been positively impacted by automation; both in hardware and data processing, in the last two decades. However, wet milling, a critical unit operation used to manipulate particle properties in a crystallization process, has been non-conducive for automation. This study discusses efforts towards automation of wet milling, coupled with off-the-shelf automation tools for faster execution of physical property design workflows. The hardware and software configuration used, along with challenges and prospects to achieve milling automation are discussed. Lovastatin was used as a model compound to demonstrate the capabilities of the automated platform to achieve morphology and size manipulation. Five different batches of lovastatin were produced using different combinations of milling and thermal cycling. Milling followed by thermal cycling led to formation of rougher, fused particles with a lower aspect ratio compared to those produced by thermal cycling followed by milling. In-situ particle video microscopy was used to decipher that the initial increase in particle size during thermal cycling was driven by a fusion of primary particles followed by growth. Key material property descriptors such as bulk/tap density, flow function coefficient, surface area, and particle size by laser diffraction were collected using traditional means while particle size, aspect ratio and smoothness were also measured using image analysis. Lastly, principal component analysis (dimensionality reduction) was performed to determine the key property relationships impacting flowability. It was found that fines as measured by d10 (10 th percentile of the volumetric size distribution) and Hausner ratio had a high positive and negative correlation, respectively with flowability. The on demand milling platform combined with data rich trials using PAT is estimated to accelerate process development for particle engineering and reduce the drug substance delivery time to drug product team.

Abstract:Drug efficacy strongly relies on the solid state of the active pharmaceutical ingredient. Classical solid-state screening methods involve different solvent compositions and supersaturations. Moreover, the many repeat experiments needed to address the stochasticity of nucleation make this approach costly. This paper presents a newly developed modular microfluidic platform that provides a universal and flexible plug-and-play tool for crystallisation studies without use of surfactants. By dissolving a powder, our setup generates saturated solutions that can be used for solubility measurements or distributed in microdroplets. Here, we describe solubility measurements performed on different forms, stable and metastable, of pharmaceutical molecules (Irbesartan, Rimonabant and Aripiprazole) in organic and aqueous solvents. In addition, we provide nucleation statistics obtained for Sulfathiazole in water and in acetonitrile. Reporting polymorph screening on Sulfathiazole and statistics for nucleated forms, we find that the cooling rate influences both nucleation and polymorphism results, reflecting the competition between thermodynamics and kinetics. Three unknown forms were discovered, with XRD patterns and Raman spectra that do not match any referenced form. We also demonstrate the limitations of microfluidics for crystallisation by cooling: reducing the crystalliser volume considerably increases nucleation induction time.

Abstract:Protein crystallization has drawn great attention to replacing the traditional downstream processing for protein-based pharmaceuticals due to its advantages in stability, storage, and delivery. Limited understanding of the protein crystallization processes requires essential information based on real-time tracking during the crystallization process. A batch crystallizer of 100 mL fitted with a focused beam reflectance measurement (FBRM) probe and a thermocouple was designed for in situ monitoring of the protein crystallization process, with simutaneously record of off-line concentrations and crystal images. Three stages in the protein batch crystallization process were identified: long-period slow nucleation, rapid crystallization, and slow growth and breakage. The induction time was estimated by FBRM, i.e., increasing numbers of particles in the solution, which could be half of the time required for detecting the decrease of the concentration, by offline measurement. The induction time decreased with an increase in supersaturation within the same salt concentration. The interfacial energy for nucleation was analyzed based on each experimental group with equal salt concentration and different concentrations of lysozyme. The interfacial energy reduced with an increase in salt concentration in the solution. The yield of the experiments was significantly affected by the protein and salt concentrations and could achieve up to 99% yield with a 26.5 μm median crystal size upon stabilized concentration readings.

Abstract:Protein crystallization as opposed to well-established chromatography processes has the benefits to reduce production costs while reaching a comparable high purity. However, monitoring crystallization processes remains a challenge as the produced crystals may interfere with analytical measurements. Especially for capturing proteins from complex feedstock containing various impurities, establishing reliable process analytical technology (PAT) to monitor protein crystallization processes can be complicated. In heterogeneous mixtures, important product characteristics can be found by multivariate analysis and chemometrics, thus contributing to the development of a thorough process understanding. In this project, an analytical set-up is established combining offline analytics, on-line ultraviolet visible light (UV/Vis) spectroscopy, and in-line Raman spectroscopy to monitor a stirred-batch crystallization process with multiple phases and species being present. As an example process, the enzyme Lactobacillus kefir alcohol dehydrogenase (L k ADH) was crystallized from clarified Escherichia coli ( E. coli ) lysate on a 300 mL scale in five distinct experiments, with the experimental conditions changing in terms of the initial lysate solution preparation method and precipitant concentration. Since UV/Vis spectroscopy is sensitive to particles, a cross-flow filtration (cross-flow filtration)-based bypass enabled the on-line analysis of the liquid phase providing information on the lysate composition regarding the nucleic acid to protein ratio. A principal component analysis (PCA) of in situ Raman spectra supported the identification of spectra and wavenumber ranges associated with productspecific information and revealed that the experiments followed a comparable, spectral trend when crystals were present. Based on preprocessed Raman spectra, a partial least squares (PLS) regression model was optimized to monitor the target molecule concentration in real-time. The off-line sample analysis provided information on the crystal number and crystal geometry by automated image analysis as well as the concentration of Lk ADH and host cell proteins (HCPs) In spite of a complex lysate suspension containing scattering crystals and various impurities, it was possible to monitor the target molecule concentration in a heterogeneous, multi-phase process using spectroscopic methods. With the presented analytical set-up of off-line, particle-sensitive on-line, and in-line analyzers, a crystallization capture process can be characterized better in terms of the geometry, yield, and purity of the crystals.

Abstract:Precompetitive collaborations on new enabling technologies for research and development are becoming popular among pharmaceutical companies. The Enabling Technologies Consortium (ETC), a precompetitive collaboration of leading innovative pharmaceutical companies, identifies and executes projects, often with third-party collaborators, to develop new tools and technologies of mutual interest. Here, we report the results of one of the first ETC projects: the development of a user-friendly population balance model (PBM)-based crystallization simulator software. This project required the development of PBM software with integrated experimental data handling, kinetic parameter regression, interactive process simulation, visualization, and optimization capabilities incorporated in a computationally efficient and robust software platform. Inputs from a team of experienced scientists at 10 ETC member companies helped define a set of software features that guided a team of crystallization modelers to develop software incorporating these features. Communication, continuous testing, and feedback between the ETC and the academic team facilitated the software development. The product of this project, a software tool called CrySiV, an acronym for Crystallization Simulation and Visualization, is reported herein. Currently, CrySiV can be used for cooling, antisolvent, and combined cooling and antisolvent crystallization processes, with primary and secondary nucleation, growth, dissolution, agglomeration, and breakage of crystals. This paper describes the features and the numerical methods of the software and presents two case studies demonstrating its use for parameter estimation. In the first case study, a simulated data set is used to demonstrate the capabilities of the software to find kinetic parameters and its goodness of fit to a known solution. In the second case study, the kinetics of an antisolvent crystallization of indomethacin from a ternary solvent system are estimated, providing a practical example of the tool.This article has not yet been cited by other publications.

From cloudy suspensions to clear data: Particle imaging enabled by automated dilution

Abstract:We present a novel dilution device designed for accurately and robustly diluting dense suspensions to enable real-time particle characterization using optical imaging systems. The device employs tangential flow filtration, allowing for continuous operation at high solid weight fractions typical in industrial crystallization processes. It demonstrates the ability to monitor suspensions continuously over 24 hours and to quickly adapt to changing particle densities, whilst continuously enabling accurate measurements. This paper presents also a study of the impact of particle density on measurement accuracy; extensive testing confirms the reliability of the dilution device for distributions of various particle sizes and shapes. The dilution device successfully tracks the evolution of the particle size and shape distribution (PSSD) during cooling crystallization. The innovation presented here, overcomes the limitations of existing methods hindered by high particle densities, providing a significant advancement in real-time monitoring and characterization capabilities under realistic conditions. This technology holds potential for broad applications, including the validation of new crystallization theories and, thus enhanced development of efficient crystallization processes.

Abstract:Traditionally, off-line measurements via image analysis or laser diffraction methods are used to analyze the crystal product. It is often beneficial to extract crystal samples intermittently during the crystallization process to better understand the complex dynamics in batch and continuous crystallization. However, frequent invasive product sampling can lead to undesired disturbances in the process dynamics. Various process analytical technologies have been developed for in situ monitoring of crystallization systems; however, obtaining quantitative crystal size distribution (CSD) information from these is challenging. While in the case of traditional concentration monitoring tools system-specific calibration is an accepted standard procedure, most image analysis-based techniques attempt to provide direct measurement of CSD information. To obtain a fast and accurate in situ image analysis measurement of particle size for a high aspect ratio crystallization system, a systematic off-line image analysis calibration methodology was performed to model off-line Malvern Morphologi image analysis results. An image analysis algorithm was calibrated to extract size distribution data from in situ images of varying sizes and solid concentrations. Using a genetic algorithm, the various methods and parameters in the image analysis algorithm were automatically optimized by minimizing the size distribution error between the model and the off-line image analysis measurement. Trends observed in the calibrated parameters were then fitted to continuous functions depending on solid density to be able to adapt to changes in the solid loading. The algorithms were then validated with a different particle data set with a known solid loading. Last, to demonstrate the proof of concept in sensor fusion and online application, the adaptive image analysis algorithm was coupled with a UV/vis sensor and tested on a dynamic data set to predict the size distribution with varying solid loadings throughout the crystallization process due to dissolution, nucleation, and growth.

Small-Scale Continuous Drug Product Manufacturing using Dropwise Additive Manufacturing and Three Phase Settling for Integration with Upstream Drug Substance Production

Abstract:The pharmaceutical industry has traditionally relied on mass manufacturing to make its products. This has created multiple problems in the drug supply network, including long production times, inflexible and sluggish manufacturing and lack of personalized dosing. The industry is gradually adapting to these challenges and is developing novel technologies to address them. Continuous manufacturing and 3D printing are two promising techniques that can revolutionize pharmaceutical manufacturing. However, most research studies into these methods tend to treat them separately. This study seeks to develop a new processing route to continuously integrate a 3D printing platform (Drop-on-Demand, DoD, printing) with crystallization that is generally the final step of the active ingredient manufacturing. Accomplishing this integration would enable harnessing the benefits of each method- personalized dosing of 3D printing and flexibility and speed of continuous manufacturing. A novel unit operation, three-phase settling (TPS), is developed to integrate DoD with the upstream crystallizer. To ensure on-spec production of each printed dosage, two process analytical technology tools are incorporated in the printer to monitor drug loading in manufactured drug products in real time. Experimental demonstration of this system is carried out via two case studies: the first study uses an active ingredient celecoxib to test the standalone operation of TPS; the second study demonstrates the operation of the integrated system (crystallizer - TPS - DoD) to continuously make drug products for the active ingredient- lomustine. A dissolution test is also performed on the manufactured and commercial lomustine drug products to compare their dissolution behavior.

Abstract:<p>Embedded droplet printing is a recent mode of generating and processing droplets within yield‐stress fluids. This technique has shown promise for performing sensitive processes like pharmaceutical crystallization, as well as chemical synthesis and biological experimentation due to the unique ability to process droplets that are quiescently suspended. Despite improving on conventional microfluidic technologies in numerous ways, current embedded droplet printing methods are limited to batch processes, severely hampering their overall utility. A new platform that enables continuous production of embedded droplets is presented and characterized. This platform expands the capabilities of embedded droplet printing and allows for its application to areas of continuous materials discovery, screening, and manufacturing. Here, the platform is used for the rapid production of pharmaceutical particles that are highly spherical and uniform, key targets for flowability, and ultimately manufacturability of pharmaceutical drug products. The presented platform achieves a maximum throughput of over 100 g per day, enabling characterization of the superior powder flow properties. The available operating space of this platform is demonstrated for an antisolvent crystallization process with an anti‐malarial drug. This understanding provides design guidelines for how similar platforms may be engineered for precise, rapid, customized, and distributed manufacturing of drug particles with superior flowability.</p>

Use of Wet Milling Combined with Temperature Cycling to Minimize Crystal Agglomeration in a Sequential Antisolvent-Cooling Crystallization

Abstract:The objective of the research was to improve the process design of a combined antisolvent-cooling crystallization to reduce the degree of agglomeration of a real active pharmaceutical ingredient product, which was manufactured using a crystallization stage employing a methanol/water solvent system. Knowledge was gained from the use of process analytical technology (PAT) tools to monitor the process variables, allowing particle size, degree of agglomeration, solute concentration, and supersaturation to be tracked throughout the process. Based on knowledge of the solubility behavior and interpretation of the PAT histories, changes were made to the sequences of antisolvent addition and cooling within the crystallization process to reduce agglomeration in the final product. Different seed loadings and seeding addition points were also investigated to maintain operation within lower supersaturation regions of the phase diagram to limit agglomeration and avoid an undesired polymorphic transformation to an unstable form. The improved sequences of operations and seeding conditions did not provide sufficient improvement in the product quality and so were augmented by applying wet milling for further deagglomeration followed by temperature cycling to remove fine particles generated during milling. Open-loop heating and cooling cycles produced some limited improvements, whereas closed-loop direct nucleation control methods using FBRM as a feedback sensor for particle counts per second were much more successful at producing high-quality crystals of the desired polymorphic form. The work shows that understanding the trajectory of the process through the phase diagram to follow appropriate supersaturation profiles gives improved control of the various kinetic mechanisms and can be used to improve the quality of the final product.

Abstract:A miniaturized multi‐reactor system and knowledge‐based workflow were developed to identify optimal process conditions for controlling the crystal morphology of active pharmaceutical ingredients (APIs). This method, which features minimal material usage, inline imaging, and independent control of temperature T and supersaturation S, can be used to obtain more equant‐shaped crystals of diverse APIs. The morphology of crystalline active pharmaceutical ingredients (APIs) can significantly affect their product properties, so that its control during manufacturing is crucial. To address this, a newly augmented commercial milliliter‐scale facility and knowledge‐based workflow were developed with the aim of identifying optimal process conditions for producing more equant‐shaped crystals. The design includes minimal material usage, inline imaging, and independent temperature and supersaturation control through evaporative crystallization. The methodology enables the identification of process conditions for equant‐shaped crystals across diverse APIs. These findings contribute to advancing pharmaceutical research and development by providing a reliable approach to optimize crystal morphology.

Abstract:Crystallization of proteins is a critical step in structural biology, biopharmaceutical industry and materials science. Microfluidic technology has emerged as a promising tool for screening the crystallization conditions with advantages of increased throughput, reduced consumption of reagents and lower cost. However, current mirofluidic approches generally lack the module for high-speed data analysis, ignore the crystallization kinetics and suffer from inconsistency after scaling up due to the subnano-/nano-liter-scaled volume. To address the issues, we propose a deep learning-aided programmable microliter-droplet system, which allows the high-throughput screening of time-resolved protein crystallization in microliter scale. Based on the system, a series of temporal phase diagrams are acquired, which reveal the time-resolved crystallization of target proteins under different crystallization conditions. They provide precise guidance on the scale-up experiment (~93% in consistency), and help gain insight into the kinetic characteristics of protein crystallization.

Abstract:A workflow for the digital design of crystallization processes starting from the chemical structure of the active pharmaceutical ingredient (API) is a multi-step, multi-disciplinary process. A simple version would be to first predict the API crystal structure and from it the corresponding properties of solubility, morphology, and growth rates, assume that the nucleation would be controlled by seeding, and then use these parameters to design the crystallization process. This is usually an over-simplification as most APIs are polymorphic, and the most stable crystal of the API alone may not have the required properties for development into a drug product. This perspective, from the experience of a Lilly Digital Design project, considers the fundamental theoretical basis of crystal structure prediction (CSP), free energy, solubility, morphology and growth rate prediction, and the current state of nucleation simulation. This is illustrated by applying the modeling techniques to real examples, olanzapine and succinic acid. We demonstrate the promise of using ab initio computer modeling for solid form selection and process design in pharmaceutical development. We also identify open problems in the application of current computational modeling and achieving the accuracy required for immediate implementation that are currently limiting the applicability of the approach.
