Abstract:In this study, chitosan-gelatin conjugates were prepared by chemical crosslinking of tannic acid. The cryogel templates were developed through freeze-drying and immersed in camellia oil to construct cryogel-templated oleogels. Chemical crosslinking resulted in apparent colour changes and improved emulsion-related/rheological properties on conjugates. The cryogel templates with different formulas exhibited different microstructures with high porosities (over 96 %), and crosslinked samples might have higher hydrogen bonding strength. Tannic acid crosslinking also led to enhanced thermal stabilities and mechanical properties. Cryogel templates could reach a considerable oil absorption capacity of up to 29.26 g/g and prevent oil from leaking effectively. The obtained oleogels with high tannic acid content possessed outstanding antioxidant abilities. After 8 days of rapid oxidation at 40 °C, Oleogels with a high degree of crosslinking owned the lowest POV and TBARS values (39.74 nmol/kg, and 24.40 μg/g, respectively). This study indicates that the involvement of chemical crosslinking would favor the preparation and the application potential of cryogel-templated oleogels, and the tannic acid in the composite biopolymer systems could act as both the crosslinking agent and the antioxidant.

Modulating in vitro digestion of whey protein cold-set emulsion gels via gel properties modification with gallic acid and EGCG

Abstract:Gallic acid (GA) and epigallocatechin gallate (EGCG), cooperated at varied ratios (1:0, 3:1, 1:1, 1:3, and 0:1), were employed to modify gel properties of calcium induced-whey protein emulsion gel. The effects of GA/EGCG on emulsion morphology, as well as gel properties and in vitro digestive behavior of the emulsion gels were investigated. Compared with emulsions without phenolics, GA/EGCG induced slightly smaller particle size and stronger electrostatic repulsion between emulsion droplets. Moreover, GA/EGCG, notably at a ratio of 3:1, promoted electrostatic and hydrophobic interactions between protein molecules and the formation of a compact and filamentous gel microstructure, resulting in a remarkable increment in the gel strength (up to 106 %). Furthermore, in vitro oral digestion, dynamic gastric digestion (using an artificial gastric digestive system, AGDS), and intestinal digestion of the emulsion gels were simulated. Particle size and protein hydrolysis results revealed that GA/EGCG was prone to weaken the physical disintegration of gels, reduce protein hydrolysis, and enhance the stability of emulsified oil droplets during dynamic gastric digestion. As a consequence, delayed release of oil droplets was observed in the gels and more free fatty acids were released in the intestinal digestion, particularly in the gel with GA/EGCG (3:1). These findings would provide novel strategies for application of phenolic compounds in developing protein gel-based delivery systems.

Abstract:Biodegradable macroporous hydrogels were prepared using a biodegradable crosslinker based on chitosan derivatives. The chitosan crosslinker was synthesized through the amidating reaction of amino groups in the chitosan oligosaccharide (CSO) chains and carboxyl groups in acrylic acid (AA) under the catalysis of 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimidehydro-chloride (EDC) and N-hydroxysuccinimide (NHS). The amidating reaction is able to provide polymerizable carbon-carbon double bonds for CSO. The chemical structure of the resulting compound chitosan oligosaccharide-graft-acrylic acid (CSO-g-AA) was characterized by Fourier transform infrared spectroscopy (FTIR) and H-1 nuclear magnetic resonance (H-1 NMR) spectroscopy. The content of double bonds was measured by titration analysis. The measurement showed a grafting ratio of AA on CSO of about 3.5% (on average two acryoyl groups per CSO-g-AA chain) when the applied molar ratio of AA to CSO was 1.0. Polyacrylamide (PAM) cryogels were synthesized by the free radical polymerization of acrylamide using a modified freezing polymerization method crosslinked by CSO-g-AA. The obtained cryogels were macroporous hydrogels which could be degraded into solutions of linear polymer chains when treated with appropriate enzymes. Then we used snailase as a model enzyme to study the biodegradation process. The degradation was monitored by morphological studies using a confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM), mechanical strength, and swelling ratio. The duration of the degradation process was adjustable from one month to two months when using different concentrations of snailase solutions.

Abstract:The ginger essential oil/β-cyclodextrin (GEO/β-CD) composite, ginger essential oil/β-cyclodextrin/chitosan (GEO/β-CD/CTS) particles and ginger essential oil/β-cyclodextrin/chitosan (GEO/β-CD/CTS) microsphere were prepared with the methods of inclusion, ionic gelation and spray drying. Their properties were studied by using scanning electron microscopy (SEM), differential scanning calorimetry (DSC), thermo-gravimetry analysis (TGA), Fourier transform infrared spectroscopy (FT-IR) and X-ray diffraction (XRD). The results showed that the particle size of GEO/β-CD composite was smaller than that of β-CD and GEO/β-CD/CTS particles were loose and porous, while the microsphere obtained by spray drying had certain cohesiveness and small particle size. Besides, results also indicated that β-CD/CTS could modify properties and improve the thermal stability of GEO, which would improve its application value in food and medical industries.

Abstract:The beta-lactoglobulin-chlorogenic acid (LG-CA) conjugate was explored to be formed through ultrasonication, redox-pair method and their combination, the ultrasonication used a probe ultrasonic machine with a 6 mm probe at 270 W, and the frequency was 20-25 kHz. The formation of the conjugate was confirmed by SDS-PAGE with a larger molecular weight. Besides, Fourier infrared spectroscopy (FTIR) and Circular dichroism (CD) indicated changes in the secondary structure of the LG-CA conjugate. The alpha-helix and beta-sheet contents of LG decreased and the unordered content increased significantly after the formation of covalent complexes. In addition, both the ultrasonic treatment and its combination with redox-pair method could significantly improve the antioxidant properties of LG. The former increased to 23.16 mu mol Trolox/g sample, the latter 82-106 mu mol Trolox/g sample. Therefore, ultrasonication could be used both individually and in combination with the redox-pair method to produce LG-CA conjugates with stronger antioxidant activities.

Abstract:This study was designed to investigate the formation of camellia oil body (OB) emulsion gels covalently cross-linked by oxidized polyphenols: catechin (OCT), caffeic acid (OCF), chlorogenic acid (OCA), and tannic acid (OTA). The structural characteristics, thermal stabilities, antioxidant activities, rheological properties, and lipid digestion kinetics of the cross-linked OB-polyphenol emulsion gels were studied. The results of free sulfhydryl and amino group contents, FT-IR, fluorescence spectroscopy, surface hydrophobicity and thermal stability analyses confirmed the formation of covalent interactions between polyphenols and OB emulsions. Based on the second-order structural kinetic model, OB emulsion gel cross-linked by OTA had stronger intermolecular interactions and more developed 3-D network structures than those of OCA, OCF and OCT. Furthermore, lipid digestion kinetics showed that the cross-linking of polyphenols with the OBs slowed down the disintegration of protein matrix under gastric conditions, resulting in delay the release of free fatty acid, which was confirmed by CLSM observations.
Effect of different gelators on the physicochemical properties and microstructure of coconut oleogels

Abstract:Background: The inherent properties of coconut oil (CO), including its elevated saturated fatty acid content and low melting point, make it suitable for application in plastic fat processing. The present study explores the physicochemical characteristics, micromorphology and oxidative stability of oleogels produced from CO using various gelators [ethylcellulose (EC), β-sitosterol/γ-oryzanol (PS) and glyceryl monostearate (MG)] to elucidate the formation mechanisms of coconut oleogels (EC-COO, PS-COO and MG-COO). Results: Three oleogel systems exhibited a solid-like behavior, with the formation of crystalline forms dominated by β and β'. Among them, PS-COO exhibited enhanced capability with respect to immobilizing liquid oils, resulting in solidification with high oil-binding capacity, moderate hardness and good elasticity. By contrast, MG-COO demonstrated inferior stability compared to PS-COO and EC-COO. Furthermore, MG-COO and PS-COO demonstrated antioxidant properties against CO oxidation, whereas EC-COO exhibited the opposite effect. PS-COO and EC-COO exhibited superior thermodynamic behavior compared to MG-COO. Conclusion: Three oleogels based on CO were successfully prepared. The mechanical strength, storage modulus and thermodynamic stability of the CO oleogel exhibited concentration dependence with increasing gelling agent addition. PS-COO demonstrated relatively robust oil-binding capacity and oxidative stability, particularly with a 15% PS addition. This information contributes to a deeper understanding of CO-based oleogels and offers theoretical insights for their application in food products. © 2024 Society of Chemical Industry.
Fabrication, Characterization, and in Vitro Digestion of Gelatin/gluten Oleogels from Thermally Crosslinked Electrospun Short Fiber Aerogel Templates

Abstract:In this work, the electrospun short fiber-based oleogels (ESF-O) were formed by thermal crosslinking. Gelatin and gluten nanofibers were obtained via electrospinning, then homogenized and transformed into short fiber dispersions. Through freeze-drying, electrospun short fiber-based aerogel (ESF-A) templates were obtained for oil adsorption. All ESF-A exhibited the micromorphology of loose fibrous pore structure and prominent changes of characteristic peaks in the thermal and infrared analyses. Moreover, the highly crosslinked templates owned excellent hydrophobicity and mechanical performances (elastic modulus: 0.25 kPa, yield strength: 14.56 kPa, compressive strength: 52.54 kPa, and the final compression recovery: 91.27%). Meanwhile, the oil adsorption/oil holding capacity could reach 76.56 g/g and 80.04%, respectively. Through thermal crosslinking, ESF-O presented good and controllable rheological/in vitro digestion properties, which were further confirmed by PCA analysis. According to different application conditions, ESF-O properties could be adjusted by different degrees of fiber addition or thermal crosslinking.

Abstract:Oleogelation based on emulsion using food-approved biopolymers is a feasible strategy to substitute solid fat with harmful trans and saturated fatty acids, providing food products with a better nutritional profile. However, oil with high content of unsaturated fatty acids is prone to oxidation, resulting in quality loss of food products. In the present study, proanthocyanidins from Chinese bayberry leaves (BLPs) were incorporated into emulsionbased oleogel to remedy the defect through the formation of colloidal complexes with gelatin. Oleogel stabilized with the colloidal complexes showed higher stability with lower oil loss and higher gel strength, compared to that with gelatin only. Then different polysaccharides were applied to further promote oleogel stability. However, polysaccharides abrogated the promotive effect of BLPs and resulted in comparable stability of oleogels with BLPs or not, which was confirmed with cryo-SEM observation. Then the application of oleogel with BLPs as margarine alternative was verified with batter properties, quality loss of cakes and oil stability in cakes. Batter properties of oleogel with BLPs were comparable to that without BLPs, and more close to margarine than oil. For quality loss, textural properties of oleogel cakes with BLPs or not resembled each other and were significantly better than oil cakes in hardness and chewiness and margarine cakes in cohesiveness. Particularly, BLPs were verified to retard oil oxidation in both oleogel and cakes prepared with oleogel. Overall, incorporation of BLPs into oleogel was feasible to improve its oil stability and keep its application in cake products.
Oleogels loaded with lycopene structured using Zein/EGCG/Ca2+ complexes: Preparation, characterization and potential application

Abstract:Oleogels have attracted considerable attention due to their excellent viscoelasticity and high content of polyunsaturated fatty acid. This study explored the potential of Zein/(-)-epigallocatechin-3-gallate/Ca2+ complexes oleogels loaded with lycopene as potential substitute for solid fats in biscuit formulations. Utilizing an emulsion-templated method, oleogels were prepared and characterized for visual appearance, droplet size, microstructure, and rheological properties. The incorporation of lycopene indicated a dose-dependent effect on these characteristics, achieving optimal properties at a concentration of 0.3 mg/mL. At this concentration, oleogels exhibited higher encapsulation efficiency (> 90 %), lower oil loss (< 2 %), and denser network structures. Rheological analysis highlighted the shear-thinning behavior, gel-like structure, and thixotropic recovery of oleogels. Substituting of margarine with lycopene-loaded oleogels in biscuits yielded products with regular appearance, uniform color, and potential health benefits, demonstrating the viability of these oleogels as a healthier alternative to traditional solid fats in baking.

Abstract:In this work, an innovative and convenient approach was induced to construct aerogel-templated oleogels (ATO), which were simply made of camellia oil and gelatin. The freeze-dried gelatin aerogel templates were developed from uniformly dispersed electrospun short fibers (GF), which were further immersed in camellia oil within 5 min to form ATO facilely. Compared with traditional gelatin hydrogel-based (GH) aerogels, GF aerogel templates presented different micromorphology, thermal stabilities, and significant changes on infrared spectrum and secondary structure, where GF exhibited stronger internal interactions than GH. In compression tests, GH aer-ogels possessed strong resistance to stress while GF aerogels exhibited surprising resilient abilities. In addition, the oil adsorption capacity (OAC) and oil holding capacity (OHC) of GF aerogel templates considerably reached 125.31 g/g and 79.40%, respectively. Oleogels with different preparing methods and gelatin content presented outstanding rheological properties and in vitro digestibility. This work suggested that gelatin ATO could be fabricated from not only hydrogels but also electrospun short fibers, which owned a more desired physical structure and oil-related properties. Meanwhile, the properties of GF and GH systems could be significantly affected by the concentration of the biopolymer.
Fabrication of Oleogels Via a Facile Method by Oil Absorption in the Aerogel Templates of Protein-Polysaccharide Conjugates.

Abstract:In this study, a novel and facile method was developed to fabricate oleogels. The alginate/soy protein conjugates with excellent emulsifying activity and emulsion stability were prepared via Maillard reaction and freeze-dried to form the aerogel templates, which were then immersed in corn oil within 6 h to induce the oleogels. Compared with the alginate and soy protein solutions, the viscosity and elastic modulus G' of the conjugate solutions increased, indicating the formation of a new macromolecule and strengthened gel network from Maillard reaction. The conjugate aerogels presented the morphology of serious aggregation and conglutination but the higher elastic modulus and better thermal stability, due to the increasing covalent interactions. These aerogel templates showed a good oil absorption of up to 10.89 g/g aerogel and holding capacity of 40%. The resulting oleogels loaded with thymol showed excellent antimicrobial activities against Staphylococcus aureus and Escherichia coli. This work suggests that the fabrication of oleogels is not limited to the choice of existing oleogelators but from a wide variety of protein-polysaccharide conjugates to form the aerogel templates for oil absorption.
Synergetic effects of water-soluble polysaccharides for intensifying performances of oleogels fabricated by oil-absorbing cryogels

Abstract:Oleogels were prepared by the cryogel-templated method from porous cryogels, which were co-structured using hydroxypropyl-methylcellulose and structural enhancers (such as flaxseed gum, κ-carrageenan, carboxymethyl-cellulose, arabic gum, and guar gum). The hardness, network density, pore size, pore volume, and SEM micrographs of cryogels showed that κ-carrageenan and flaxseed gum could keep the integrity of aqueous foams during freeze-drying and endow cryogels with the high hardness and content to hold oils (>98%). Oil absorption curves indicated that flaxseed gum and guar gum-enhanced cryogels provided the fastest oil absorption rate due to bigger pores. The absorption model was fitted well with all experimental data. Physical and mechanical properties of cryogels were positively related to the rheological property and oil bonding capacity of oleogels. κ-carrageenan and flaxseed gum were more suitable as structural enhancers to improve hydroxypropyl-methylcellulose-based cryogels for preparing oleogels to replace plastic fats in foods.
Ovotransferrin Fibril—Gum Arabic Complexes as Stabilizers for Oleogel-in-Water Pickering Emulsions: Formation Mechanism, Physicochemical Properties, and Curcumin Delivery

Abstract:This project aimed to explore the influence of the interaction between ovotransferrin fibrils (OTF) and gum arabic (GA) on the formation mechanism, physicochemical properties, and curcumin delivery of the oleogel-in-water Pickering emulsion. Cryo-scanning electron microscopy results showed that OTF—GA complexes effectively adsorbed on the oil–water interface, generating spatial hindrance to inhibit droplet coalescence. The texture analysis also proved that OTF—GA complexes endowed oleogel-in-water Pickering emulsion with preferable springiness (0.49 ± 0.03 mm), chewiness (0.43 ± 0.07 mJ), and adhesion (0.31 ± 0.01 mJ). By exploring the coalescence stability, droplet size, and rheological properties of OTF—GA complexes–stabilized oleogel-in-water Pickering emulsion (OGPE), the higher coagulation stability, larger average droplet size (46.22 ± 0.08 μm), and stronger gel strength were observed. The microrheological results also exhibited stronger attraction between the OGPE droplets, a more pronounced solid-like structure, and a slower speed of movement than OTF-stabilized oleogel-in-water Pickering emulsion (OPE). Meanwhile, OGPE significantly enhanced the extent of lipolysis, stability, and bioaccessibility of curcumin, suggesting that it possessed superior performance as a delivery system for bioactive substances. This project provided adequate theoretical references for protein–polysaccharide complexes–stabilized oleogel-in-water Pickering emulsion, and contributed to expanding the application of oleogel-in-water Pickering emulsion in the food industry.

Effects of Modified Oil Palm Kernel Expeller Fiber Enhanced via Enzymolysis Combined with Hydroxypropylation or Crosslinking on the Properties of Heat-Induced Egg White Protein Gel

Abstract:Due to its poor hydration properties, oil palm kernel expeller dietary fiber (OPKEDF) is rarely used in the food industry, especially in hydrogels, despite its advantages of high availability and low cost. To address this situation, the effects of enzymolysis combined with hydroxypropylation or crosslinking on the structure and hydration properties of OPKEDF were investigated, and the impact of these modified OPKEDFs on the properties of egg white protein gel (EWPG) was studied. Enzymolysis combined with hydroxypropylation or phosphate crosslinking improved the soluble fiber content (5.25–7.79 g/100 g), water-retention and expansion abilities of OPKEDF (p < 0.05). The addition of unmodified OPKEDF or modified OPKEDF increased the random coil content of EWPG and increased the density of its microstructure. Moreover, enzymolysis combined with hydroxypropylation or crosslinking enhanced the effect of OPKEDF on the properties of EWPG, including improvements in its water-retention ability, pH, hardness (from 97.96 to 195.00 g), chewiness (from 78.65 to 147.39 g), and gumminess (from 84.63 to 152.27) and a reduction in its transparency (p < 0.05). Additionally, OPKEDF and enzymolysis and hydroxypropylated OPKEDF increased the resilience (0.297 to 0.359), but OPKEDF treated via enzymolysis and crosslinking reduced it. Therefore, OPKEDF modified by means of enzymolysis in combination with hydroxypropylation or crosslinking improved the gel properties of EWPG. However, further work is required to determine the effects of these modifications on the nutritional profile, scalability, and economic feasibility of OPKEDF and egg white gel.

Abstract:Recently, biopolymer-based structured oil has emerged to substitute saturated and trans-fats. Herein, the porous biopolymeric networks with the cellular structure were developed with cross-linked dialdehyde cellulose (DAC) and gelatin to prepare oleogels. The determination of aldehyde content in the DAC molecular chain revealed a high degree of oxidation of cellulose. The FTIR data showed the covalent cross-linking bond between the DAC and gelatin. The covalent cross-linking between DAC and gelatin promoted the composite aerogel (DG) to form a porous three-dimensional network structure with enhanced thermal stability, mechanical properties, and water stability. The DAC/gelatin composite aerogel with 5 wt% DAC (DG-5) exhibited the highest porosity (91.27 %) and lowest pore diameter (10.52 μm) with greater capillary force, resulting in a high oil absorption capacity of 11.24 g/g and oil holding capacity of 55.57 %. The DG oleogels containing 6 % thymol in the oil phase showed good antibacterial activities against Escherichia coli and Staphylococcus aureus for 24 h. This work provides a facile and promising strategy for developing oleogels by DAC/gelatin cross-linked conjugates as aerogel templates for oil structuring.

Abstract:In this work, the alginate-gelatin conjugates were developed by Maillard reaction. The aerogel templates were obtained by freeze-drying the conjugates, and they were transformed into oleogels by being immersed in camellia oil. Degree of graft and browning index revealed the occurrence of Maillard reaction. Compared with pure alginate/gelatin samples, the rheology and mechanical properties of mixing samples were improved, indicating the interaction of alginate and gelatin. Aerogel templates formed by Maillard reaction presented the micromorphology of lamellar aggregation, obvious changes of peak intensity in infrared spectrum and better thermal stability. The materials reached a considerable oil adsorption capacity of 23.31 g/g and oil holding capacity of 84.75%. Through Maillard reaction, oleogels showed good in vitro digestion properties. This work suggested that oleogels could be constructed from alginate-gelatin conjugates that formed the aerogel templates for oil adsorption, and Maillard reaction significantly affected the physical structures and oil capacities of alginate-gelatin compound systems.

Abstract:In this work, cellulose nanofiber/epigallocatechin gallate (CNF/EGCG) complexes at different ratio were fabricated and the effects on characteristics and stability of emulsions under different oil phase fraction (φ) were investigated. The results of functional groups indicated the formation of CNF/EGCG complexes via hydrogen bond interaction, leading to enhanced antioxidant activity as the proportion of EGCG increased. The complexes exhibited more fragments but pure CNF were more homogenously dispersed in the aqueous phase. Enhancing the EGCG content reduced the droplet size of emulsions firstly before increased, while the apparent viscosity and viscoelastic properties displayed a contrary trend. Emulsion gels with smaller droplet size, higher apparent viscosity, excellent viscoelastic properties and physical stability could be formed at φ = 0.5 while emulsions at φ = 0.3 exhibited slight creaming. The emulsions at lower CNF/EGCG ratio presented better oxidative stability, which was ascribed to the higher antioxidant activity of the complexes and the thicker interfacial film. Results suggest that EGCG could be combined with CNF to improve the emulsion performance and be applied in extending the shelf-life of the emulsion-based products.
Composite-structure oleogels constructed by glycerol monolaurate and whey protein isolate: Preparation, characterization and in vitro digestion

Abstract:The Glycerol monolaurate (GML) oleogel was induced using Camellia oil by slowly raising the temp to the melting point (MP) of GML. Whey protein isolate (WPI) solution with different ratios was composited with GML oleogel by emulsion template methods, forming dense spines and honeycomb-like networks and impressed with an adjustable composite structure. Textural results showed that compared with single GML-based oleogels, the GML/WPI composite oleogels had the advantages of high hardness and molding, and structural stability. The composite oleogels had moderate thermal stability and maximal oil binding (96.36%). In particular, as up to 6 wt% GML/WPI, its modulus apparent viscosity was significantly increased in rheology and similar to commercial fats. Moreover, it achieved the highest release of FFA (64.07%) and the synergy provided a lipase substrate and reduced the body's burden. The resulting composite oleogel also showed intermolecular hydrogen bonding and van der Waals force interactions. These findings further enlarge the application in the plant and animal-based combined of fat substitutes, delivery of bioactive molecules, etc., with the desired physical and functional properties according to different proportions.
