Abstract:Heat stress is one of the primary environmental factors that harm both the productivity and health of buffaloes. The current study was conducted to estimate the threshold of temperature humidity index (THI)1 and genetic features for milk yield of first-lactation Mehsana buffaloes using an univariate repeatability test-day model. The data included 130,475 first lactation test-day milk yield (FLTDMY) records of 13,887 Mehsana buffaloes and the daily temperature and humidity. The statistical model included herd test day as fixed effects, days-in-milk (DIM) classes, age of the animal, as well as random factors such as the additive genetic effect (AGE) of animal in general conditions (intercept), AGE of the buffaloes subjected to heat stress (slope), permanent environmental effect of animal in general conditions (intercept), permanent environmental effect of animal under heat stress conditions (slope) and random residual effect. It was expected that the general effects and the heat-tolerance effects would be correlated, represented by the present investigation's repeatability models. The variance components of FLTDMY in the present study were computed using the REML method. The threshold for THI was 78. At the THI below the threshold, the heritability estimated for the FLTDMY trait was 0.29, and the additive genetic variance (AGV) for heat stress conditions was 0. At THI of 83, AGV for heat stress conditions was highest for FLTDMY. The genetic correlation of general AGE to heat-tolerant AGE was -0.40. The results indicated that a consistent selection for milk production, avoiding the thermal tolerance, may diminish the thermal tolerance capacity of Mehsana buffaloes.
Impact of heat stress on genetic evaluation of oocyte and embryo production in Gir dairy cattle

Abstract:Identifying and selecting genotypes tolerant to heat stress might improve reproductive traits in dairy cattle, including oocyte and embryo production. The temperature-humidity index (THI) was used, via random regression models, to investigate the impact of heat stress on genetic parameters and breeding values of oocyte and embryo production in Gir dairy cattle. We evaluated records of total oocytes (TO), viable oocytes (VO), cleaved embryos (CE), and viable embryos (VE) from dairy Gir donors. Twenty-four models were tested, considering age at ovum pick-up (AOPU) and THI means as a regressor in the genetic evaluation. We computed THI in eight periods, from 0 to 112 days before ovum pick-up, which were adjusted by different orders of Legendre polynomials (second, third, and fourth). The best-fit model according to Akaike's information criterion (AIC) and Model Posterior Probabilities (MPP) considered Legendre polynomials of third order and THI means of 112 days for TO, fourth order and 56 days for VO, second order and 28 days for CE, and second order and 42 days for VE, respectively. The heritability (h2) estimates across AOPU and THI scales ranged from 0.34 to 0.62 for TO, 0.31 to 0.58 for VO, 0.26 to 0.39 for CE, and 0.15 to 0.26 for VE, respectively. The fraction of the phenotypic variance explained by the permanent environment in different AOPU and THI scales ranged from 0.03 to 0.25 for TO, 0.05 to 0.26 for VO, 0.09 to 0.36 for CE, and 0.15 to 0.27 for VE, respectively. Spearman's rank correlation between the estimated breeding values in different AOPU and THI scale from the top 5% sires and females ranged from 0.18 to 0.90 for TO, 0.31 to 0.95 for VO, 0.14 to 0.85 for CE, and 0.47 to 0.94 for VE, respectively. The h2 estimates for all evaluated traits varied from moderate to high magnitude across AOPU and THI scales, indicating that genetic selection can result in rapid genetic progress for the evaluated traits. There was a reranking among the best animals in different AOPU and THI. It is possible to select dairy Gir cattle tolerant to heat stress to improve oocyte and embryo production.

Abstract:The current investigation aimed to comprehend the inflammatory and related immune responses in intrauterine calves subjected to heat stress (HS) during late gestation. For this purpose, 48 Sahiwal cows in late gestation were chosen and categorized into four equal groups: naturally heat stressed (NHS), cooling-treated (CLT), spring, and winter, and likewise their neonate calves born in summer (IUHS - intrauterine heat stressed and IUCL – intrauterine cooled), spring, and winter seasons. Environmental parameters were recorded, and the temperature-humidity index (THI) was calculated daily throughout the study period. The average THI values ranged between 84.18 (summer-NHS), 73.88 (summer-CLT), 78.92 (spring), and 64.91 (winter). NHS and spring groups exhibited thermal stress based on THI (> 76.00). Various treatments significantly ( P < 0.01) impacted parameters like rectal temperature (RT), respiratory rate (RR), pulse rate (PR), and skin temperature (ST) in Sahiwal cows and their calves during the study, except for heart rate (HR). Blood samples collected during different seasons and from cows housed in a climatic chamber were used to extract plasma. Plasma cortisol, interleukin-6 (IL-6), tumour necrosis factor-α (TNF-α), and thiobarbituric acid reactive substances (TBARS) levels were notably higher ( P < 0.05) in the NHS compared to the CLT group. Conversely, total antioxidant capacity (TAC) and immunoglobulin G (IgG) levels were higher ( P < 0.05) in the CLT and winter groups. IUHS calves exhibited significantly ( P < 0.05) lower overall mean plasma TAC and IgG levels but higher inflammatory and oxidative biomarkers, such as IL-6, TNF-α, and TBARS. Additionally, significant impacts on body weight were observed for factors such as interval ( P < 0.01) and the interaction between treatment and interval ( P < 0.05), exhibiting consistently lower body weight in IUHS calves throughout the study period. These findings suggest that late gestation heat stress may lead to physiological alterations in future calves. Strategies aimed at mitigating heat stress during late gestation should be considered not only for the productivity and well-being of the pregnant dam but also for the development and future performance of the calf.

Heat Stress Impacts on Lactating Cows Grazing Australian Summer Pastures on an Automatic Robotic Dairy

Abstract:The objective of this study was to measure the impacts of summer heat events on physiological parameters (body temperature, respiratory rate and panting scores), grazing behaviour and production parameters of lactating Holstein Friesian cows managed on an Automated Robotic Dairy during Australian summer. The severity of heat stress was measured using Temperature-Humidity Index (THI) and impacts of different THIs—low (≤72), moderate (73–82) and high (≥83)—on physiological responses and production performance were measured. There was a highly significant (p ≤ 0.01) effect of THI on respiratory rate (66.7, 84.7 and 109.1/min), panting scores (1.4, 1.9 and 2.3) and average body temperature of cows (38.4, 39.4 and 41.5 °C), which increased as THI increased from low to moderate to high over the summer. Average milk production parameters were also significantly (p ≤ 0.01) affected by THI, such that daily milk production dropped by 14% from low to high THI, milk temperature and fat% increased by 3%, whilst protein% increased by 2%. The lactation stage of cow had no significant effect on physiological parameters but affected (p ≤ 0.05) average daily milk yield and milk solids. Highly significant (p ≤ 0.01) positive correlations were obtained between THI and milk temperature, fat% and protein% whilst the reverse was observed between THI and milk yield, feed intake and rumination time. Under moderate and high THI, most cows sought shade, spent more time around watering points and showed signs of distress (excessive salivation and open mouth panting). In view of the expected future increase in the frequency and severity of heat events, additional strategies including selection and breeding for thermotolerance and dietary interventions to improve resilience of cows need to be pursued.

Abstract:The performance of an adult dairy cow may be influenced by heat stress that occurs during her gestation. The present study investigated potential effects of temperature-humidity index (THI) experienced by a cow during pregnancy, on the gestated daughter's performance on her first lactation, for the French Holstein and Montbeliarde dairy cattle populations. Fourteen traits were analyzed, all measured on genotyped cows: 305-d milk, fat, and protein yields, 305-d somatic cell score, clinical mastitis (both occurrence and number of events), body conformation traits, and heifer and cow conception rate. To study the effect of heat stress, we considered the THI experienced by the gestating cow, averaged for each month of her pregnancy and then categorized in 7 classes (≤40; 40-45; 45-50; 50-55; 55-60; 60-65; and >65). These average THI classes were then fitted as categorical covariates in the regression models used for this study, which included other fixed effects, and the genomic estimated breeding value as a covariate, both specific to each trait, the latter previously obtained from the official French evaluations. The THI effect was therefore estimated as the deviation between the observed and the predicted performances. In general, the estimated heat stress effects were small, presenting limited practical impact on the studied traits, and particularly for fertility and udder health, the estimated heat stress effects were not statistically significant. For the production traits, i.e., milk, fat, and protein yields, the estimated effect associated to high THI experienced at the beginning of the gestation was negative, and lightly positive when associated to high THI experienced by the dam at the end of her pregnancy. Finally, our results suggest that under the current French climate conditions, heat stress experienced by cows during any stage of their pregnancy has limited impact on future performances of their gestated daughters, however we cannot exclude that significant in utero heat stress effect may be present in climate conditions warmer than the French.

Abstract:The thermal stress adversely affects crossbred dairy cattle production, especially in a hot, humid tropical state like Kerala. So, assessment of the intensity of stress and its relationship with the physiological alterations are crucial in adopting proper mitigation strategies to attain sustainable production. Hence this study was formulated to investigate the impact of heat stress on physio-biochemical parameters during early lactation of crossbred dairy cattle. The study comprised two sets of animals: six animals calved in month of December (control, season I) and six animals calved in the month of March (heat stressed,season II). Significant elevations (p < 0.05) in all the climatic and physiological parameters were noted in the second season. The heat stress did not result in significant differences (p > 0.05)in various blood and serum biochemical parameters such as potassium (K+), bicarbonate (HCO3−), calcium (Ca2+), phosphorus (P), Globulin, total erythrocyte count (TEC), Total leucocyte count (TLC), haemoglobin (Hb), the volume of packed red cells (VPRC), aspartate aminotransferase (AST), glucose, β hydroxyl butyrate (BHB), non-esterified fatty acid (NEFA) and triglyceride (TG).Significant differences (p < 0.05) were noted in sodium (Na+) on day zero postpartum and chloride (Cl−)on day 45 postpartum. Furthermore, there was a significant decrease (P < 0.05) in partial pressure of carbon dioxide (PCO2), gamma-glutamyl transferase (GGT), total protein, albumin, triiodothyronine (T3) and thyroxine (T4) during heat stressed period while cortisol and partial pressure of oxygen (PO2) levels increased. This study also revealed a highly significant reduction (p < 0.01) in milk yield, indicating the animals maintained their positive energy balance at the expense of milk production during the study period.

Abstract:Dairy cattle are highly susceptible to heat stress. Heat stress causes a decline in milk yield, reduced dry matter intake, reduced fertility rates, and alteration of physiological traits (e.g., respiration rate, rectal temperature, heart rates, pulse rates, panting score, sweating rates, and drooling score) and other biomarkers (oxidative heat stress biomarkers and stress response genes). Considering the significant effect of global warming on dairy cattle farming, coupled with the aim to reduce income losses of dairy cattle farmers and improve production under hot environment, there is a need to develop heat tolerant dairy cattle that can grow, reproduce and produce milk reasonably under the changing global climate and increasing temperature. The identification of heat tolerant dairy cattle is an alternative strategy for breeding thermotolerant dairy cattle for changing climatic conditions. This review synthesizes information pertaining to quantitative genetic models that have been applied to estimate genetic parameters for heat tolerance and relationship between measures of heat tolerance and production and reproductive performance traits in dairy cattle. Moreover, the review identified the genes that have been shown to influence heat tolerance in dairy cattle and evaluated the possibility of using them in genomic selection programmes. Combining genomics information with environmental, physiological, and production parameters information is a crucial strategy to understand the mechanisms of heat tolerance while breeding heat tolerant dairy cattle adapted to future climatic conditions. Thus, selection for thermotolerant dairy cattle is feasible.

Effect of c-Jun NH₂-terminal kinase-mediated p53 expression on neuron autophagy following traumatic brain injury in rats.

Abstract:BACKGROUND Activation of c-Jun NH(2)-terminal kinase (JNK) has been implicated in neuron apoptosis as well as autophagy in response to various stressors after traumatic brain injury (TBI). However, the underlying molecular pathway remains unclear. Our study assessed whether JNK-mediated p53 phosphorylation might be an important mechanism for enhancing neuron autophagy in response to TBI. METHODS A total of 186 male Sprague-Dawley (SD) rats (300 - 350 g) were used in this study. By randomized block method rats were randomly divided into four groups: sham-operated (n = 46), TBI (n = 60), TBI + dimethyl sulfoxide (DMSO) (n = 40), and TBI + SP600125 (n = 40). JNK was treated with SP600125, a specific JNK inhibitor. JNK, p-P53, Beclin-1, damage-regulated autophagy modulator (DRAM) and p-bcl-2 were evaluated by Western blotting analysis. The cellular localization and expression of Beclin-1 and DRAM was observed by immunofluorescence and immunohistochemistry, and the expression of Beclin-1-Bcl-2/Bcl-xL complexes was evaluated by immunoprecipitation. Multiple-group comparisons were conducted using analysis of variance (ANOVA). P values of less than 0.05 were considered statistically significant. RESULTS It was observed that the expression of JNK, p-P53, Beclin-1, DRAM and p-bcl-2 was increasing after TBI, and the expression of Beclin-1 and DRAM was mainly located in the cytoplasm of neurons. But these were significantly inhibited in SP600125 group compared with sham group and TBI + SP600125 group (P < 0.05). The expression of Beclin-1-Bcl-2/Bcl-xL complexes was reduced after TBI. CONCLUSION JNK-mediated p53 phosphorylation might be an important mechanism for enhancing neuron autophagy in response to TBI.
Heat Stress: A Serious Disruptor of the Reproductive Physiology of Dairy Cows

Abstract:Global warming is a significant threat to the sustainability and profitability of the dairy sector, not only in tropical or subtropical regions but also in temperate zones where extreme summer temperatures have become a new and challenging reality. Prolonged exposure of dairy cows to high temperatures compromises animal welfare, increases morbidity, and suppresses fertility, resulting in devastating economic losses for farmers. To counteract the deleterious effects of heat stress, cattl e employ various adaptive thermoregulatory mechanisms including molecular, endocrine, physiological, and behavioral responses. These adaptations involve the immediate secretion of heat shock proteins and cortisol, followed by a complex network of disrupted secretion of metabolic and reproductive hormones such as prolactin, ghrelin, ovarian steroid, and pituitary gonadotrophins. While the strategic heat stress mitigation measures can restore milk production through modifications of the microclimate and nutritional interventions, the summer fertility records remain at low levels compared to those of the thermoneutral periods of the year. This is because sustainment of high fertility is a multifaceted process that requires appropriate energy balance, undisrupted mode of various hormones secretion to sustain the maturation and fertilizing competence of the oocyte, the normal development of the early embryo and unhampered maternal—embryo crosstalk. In this review, we summarize the major molecular and endocrine responses to elevated temperatures in dairy cows, as well as the impacts on maturing oocytes and early embryos, and discuss the consequences that heat stress brings about in dairy cattle fertility.

Abstract:Dairy farming is vulnerable to global warming and climate change. Improving and maintaining conception rates (CRs) have a paramount importance for the profitability of any dairy enterprise. There is an antagonistic relationship between fertility and milk yield, and intensive selection for milk yield has severely deteriorated reproductive efficiency. Irrespective of geography and husbandry, modern dairy cows experience heat stress (HS) effects leading to fertility declines, but it worsens in tropical climates. The threshold of HS experience among modern dairy cow has lowered, leading to decreased thermal comfort zone. Studies show that this threshold is lower for fertility than for lactation. HS abatement and robustness response to lactation yield lead to negative energy balance, and cow's reproductive requirements remain unfulfilled. The adverse effects of HS commence from developing oocyte throughout later stages and its fertilization competence; the oestrus cycle and oestrus behaviour; the embryo development and implantation; on uterine environment; and even extend towards foetal calf. Even cows can become acyclic under the influence of HS. These harmful effects of HS arise due to hyperthermia, oxidative stress and physiological modifications in the body of dairy cows. Proper assessment of HS and efficient cooling of dairy animals irrespective of their stage of life at farm is the immediate strategy to reduce fertility declines. Other long- and short-term mitigation strategies to reduce fertility declines during HS include feeding care, reducing disease and mastitis rates, using semen from cooled bulls, timed artificial inseminations (AI), allied hormonal interventions and use of embryo transfer technology. Ultimate long-term solution should be well-planned breeding for fertility improvement and HS tolerance.
5d CIDR-Heatsynch improves the circulatory estradiol levels, estrus expression and conception rate in anestrus buffalo (Bubalus bubalis)

Abstract:The present study aimed to investigate whether increasing estradiol (E 2 ) during preovulatory period would increase estrous expression, luteal profiles and conception rate in 5d CIDR based timed AI protocol. A total 156 anestrus buffalo allocated (78 per group) to either 5d CIDR-Cosynch (d-5: CIDR + GnRH; d0: PGF 2α +CIDR removal; 72 h post-CIDR removal: GnRH) or 5d CIDR-Heatsynch (d-5: CIDR + GnRH; d0: PGF 2α +CIDR removal; 24 h post-CIDR removal: estradiol benzoate) group. All the buffaloes inseminated at 72 and 84 h post-CIDR removal. A subset of buffalo ( n = 58) were subjected to examination of the follicle diameter and luteal profile during protocol, post-AI on days 5 and 12. The buffalo in 5d CIDR-Heatsynch had greater ( p < .05) E 2 concentrations, estrus induction and increasing trend ( p < .08) for conception rate (57.7% vs. 43.6%) than 5d CIDR-Cosynch. The percentage of pregnant buffalo that exhibited estrus signs was greater ( p < .01) in 5d CIDR-Heatsynch than 5d CIDR-Cosynch. Positive correlation ( p < .01) was observed between POF and E 2 concentrations; POF and CL diameter, CL diameter and P 4 concentrations. Estrus response and P 4 concentrations were indicators of probability of pregnancy. In conclusion, 5d CIDR-Heatsynch tended to improve conception rate. The estrus expression and P 4 concentrations (d5 and 12 post-first-AI) is the indicator of probability of pregnancy in buffalo.

Elucidating the effect of heat stress on milk production and composition in Jersey crossbred cows using test day records integrated with NASA POWER satellite data

Abstract:The present study was undertaken to determine the effect of heat stress on milk production (test day milk yield) and compositional traits (fat%, protein%, fat yield, protein yield) as well as to observe the pattern of response to increasing heat load on these traits in Jersey crossbred cows, maintained at ICAR-National Dairy Research Institute, Eastern Regional Station, Kalyani, West Bengal, India. The weather information, obtained from the NASA POWER database based on the location of the farm latitude and longitude, was used to calculate the Temperature Humidity Index (THI). To analyze the data, a linear model was fitted to the milk production and compositional records, which were adjusted for additive genetic effect of animal, permanent environmental effect of animals and known environmental sources of variations. Subsequently, a segmented linear regression model was fitted, and the least squares estimates of production and composition traits in different classes of THI were used as the dependent variable. Two THI break-points (BP) for milk yield and one THI BP for fat yield, protein %, and protein yield were found. The first and second BP for milk yield was at THI 59 and 77, respectively, with a significant decline in milk yield of -0.04 kg/unit of THI at second BP. The BP for fat and protein yield was at THI 76, with a decline rate of -1.18 and - 0.61 g/unit of THI increase, respectively. The findings revealed the significant adverse effects of THI on milk production and composition traits in Jersey crossbred cattle.
Impact of heat stress during close-up dry period on performance, fertility and immunometabolic blood indices of dairy cows: prospective cohort study

Abstract:This study aimed to investigate whether heat stress, as defined by the temperature-humidity index (THI) during the close-up dry period, had any impact on the productive performance, fertility, and immunometabolic blood indices of dairy cows in the subsequent lactation. Lactation performance was associated with increasing THI values on - 21, - 14, and - 7 d before calving resulting in decreased milk yield by about 2.30, 2.60, and 2.90 kg, respectively. The THI on the - 7 d before the calving was negatively associated with fertility parameters such as delayed first estrus postpartum, an elongated calving interval by approximately 32 d, a higher number of services per conception by 1.00, and an elongated artificial insemination service period, days open, and inter-calving period by about 20, 52, and 52 d, respectively. The study found that the immunometabolic blood indices were associated with increasing THI values during the close-up dry period. The study showed that exposing dairy cows to close-up dry period heat stress had negative consequences on performance, fertility, and immunometabolic blood indices in the subsequent lactation. Therefore, it is recommended that herd management and barn microclimate changes be implemented earlier, starting from the late dry period, to mitigate the negative impact of heat stress.

Abstract:Heat stress is a major threat to cattle reproduction today. It has been shown that the effect of high temperature not only has a negative effect on the hormonal balance, but also directly affects the quality of oocytes, disrupting the function of mitochondria, fragmenting their DNA and changing their maternal transcription. Studies suggest that the induction of HSP70 may reduce the apoptosis of granular layer cells caused by heat stress. It has been shown that the changes at the transcriptome level caused by heat stress are consistent with 46.4% of blastocyst development disorders. Cows from calves exposed to thermal stress in utero have a lower milk yield in their lifetime, exhibit immunological disorders, have a lower birth weight and display a shorter lifespan related to the expedited aging. In order to protect cow reproduction, the effects of heat stress at the intracellular and molecular levels should be tracked step by step, and the impacts of the dysregulation of thermal homeostasis (i.e., hyperthermy) should be taken into account.

Abstract:Early embryonic mortality is one of the major intriguing factors of reproductive failure that causes considerable challenge to the mammalian cell biologists. Heat stress is the major factor responsible for reduced fertility in farm animals. The present study aimed to investigate the influence of heat stress on prostaglandin production and the expression of key genes, including COX-2 , PGES , PGFS , ITGAV and LGALS15 , in buffalo endometrial epithelial cells.

Magnitude and persistence of higher estrus-associated temperatures in beef heifers and suckled cows

Abstract:Higher estrus-associated temperatures (HEAT) are a hallmark feature in sexually active females. The overarching aim of this study was to characterize the variability, magnitude, and persistence of HEAT in heifers and suckled beef cows as well as identify associated factors when occurring during thermoneutral conditions at the onset of the spring breeding season. In both heifers and cows, estrus was induced using a 7-d controlled internal drug release (CIDR)-PGF2α protocol. Vaginal temperature after prostaglandin F2α administration was recorded every 5 min using a Thermochron iButton affixed to a blank CIDR (containing no progesterone). Estrus was defined as when a heifer first stood to be mounted or when a cow had an Estrotect patch score of 3 or 4. Level of HEAT varied among individual animals. When comparing common HEAT variables using a mixed model with date nested within a year, maximum HEAT (39.9 ± 0.1 and 40.0 ± 0.1 °C) and duration (15.5 ± 0.8 and 15.4 ± 0.7) were similar in heifers and cows, respectively. However, the magnitude and persistence of HEAT differed. Total area under the HEAT curve was 117.1 ± 13.5 and 158.7 ± 12.3 for heifers vs cows, respectively (P = 0.0571). Further, 42.9% of heifers and 49% of cows had maximum HEAT ≥ 40 °C which persisted up to 6.5 and 10 h, respectively. When ambient conditions were predominantly thermoneutral, temperature humidity index had minimal impact on HEAT (mixed model, repeated measures over time). Toward identifying associated factors with different aspects of HEAT using best fit hierarchical linear regression models, baseline vaginal temperature and baseline duration were the most highly associated independent variables. Follicle size, estradiol and progesterone levels, and other available animal-related variables (e.g., age, weight, hair coat score) explained only a small amount of variation in HEAT. In summary, level of HEAT varies in estrus females even under thermoneutral conditions. Because HEAT can persist for an extended time, direct effects on fertility important components are unavoidable. Whether HEAT is a good or bad component of the periovulatory microenvironment is the basis of ongoing and future studies.

Abstract:Climate change is likely to affect both the productive and reproductive functions of Murrah buffaloes, the major milch breed of India. Hence, in the present study, weekly modules of Temperature Humidity Index (THI) based climate services were prepared and disseminated to the farmers of experimental villages of Haryana, a state of North-Western part of India through either WhatsApp, Text SMS, and Mobile Application. Difference-in-Difference (DiD), a quasi-experimental research design was used to study the impact of climate services on operational decision-making as well as the economic outcome of the farm. The findings of the study revealed that the climate services had a positive effect on the number of farmers adopting the practices like use of oil cakes, minerals, and feed additives in the animal diet. Treatment effect was found to be significant on quantity of oilcake (0.39, 0.45 and 0.51 kg/animal/day); concentrates during both summer (0.48, 0.56, 0.59 kg/animal/day) and in winter (0.35, 0.40 and 0.42 kg/animal/day); and mineral mixture (9.47, 12.34 and 13.08 gm/animal/day) in Text SMS, WhatsApp and MobileApp group, respectively. The treatment effect of climate services on economic outcome indicators such as summer milk yield was also found to be significant (0.38, 0.44, and 0.50 L/animal) from Text SMS, WhatsApp, and MobileApp, respectively. Overall, the benefit-to-cost ratio of climate services was found to be 1:1.8. Hence, the exclusive climate services developed for Murrah buffalo farmers were found to be an effective adaptive mechanism for helping vulnerable dairy farming to adapt to the climate of today and of the future.

Abstract:Reproductive performance in Holstein dairy cattle managed during summer in southern Sonora is a challenge because of high ambient temperature and relative humidity. Both of these factors contribute to heat stress, which influences cow behavior. The physiological response of cows to heat stress is one component of a system-wide gene network. Within this environment, a superior cow's ability to get pregnant early during postpartum is favorable as to reduce the trait days open and to increase productive life. Recently, many reproductive specialists have recommended using pregnancy rate as a measure of reproductive success, after converting this trait into a quantitative value using a linear formula. In comparison to the traditional measure of days open, pregnancy rate calculation includes more easily cows that do not become pregnant; furthermore, the output variable indicates that larger values are more desirable, and therefore, more understandable by dairy producers. The objective herein was to predict pregnancy rate in lactating Holstein cows using molecular markers associated with fertility in Holstein cows under a heat-stressed environment. This study included 500 cows from three dairy herds located in the Yaqui Valley of Sonora. A blood sample was collected from every cow and spotted onto FTA cards. The DNA was extracted from each card and used to genotype 179 tag SNP within 43 genes in the prolactin and GH-IGF1 pathways. Five SNP within the genes IGFBP7, IGFBP2, PAPPA1, SSTR2, and STAT6 were associated with pregnancy rate using a mixed effects model. The genotype term was later included in this model to calculate allele substitution effects. Molecular breeding values of the individual cows were calculated by summing the additive genotype effect for each SNP that showed a significant independent association with pregnancy rate, and the average MBV was 0.46 ± 0.01%. Two statistical regression models were used to predict the variable pregnancy rate: a full model that included effects of days and number of lactations, contemporary group (e.g., farm management group), health status, and MBV and a reduced model that only included MBV. Coefficients of determination were 37.61% and 3.07% for full and reduced models, respectively (P < 0.01). These results indicate that five SNP explained only a small proportion of the additive genetic variance for pregnancy rate. Additional research is needed to understand if these results are due to low heritability/repeatability of a fertility and (or) if these results are also influenced by heat stress.

Abstract:This study aimed to access the effect of heat stress on milk yield, antioxidative levels, and serum metabolites in primiparous and multiparous Holstein dairy cows during the early lactation stage. A total of 200 cows were selected based on their month of calving (June, temperature humidity index (THI) = 66.72; July, THI = 70.30; August, THI = 69.32; September, THI = 67.20; October, THI = 59.45). Blood samples were collected on days 0, 21, 50, 80, and 100 after calving for serum oxidative status analysis and milk yield was recorded daily. The lower average daily milk yield was recorded among the cows that calved in June and July (P < 0.05), and the average daily milk yield of multiparous cows was higher than that of primiparous cows that calved in the same month (P < 0.05) from d1 to d100, suggesting that seasonal (June, July) heat stress negatively affected milk yield in both primiparous and multiparous cows at early lactation. The study also indicated that there was seasonal variation in most of the serum metabolites across the studied months. The study shows that heat stress (average THI = 70.30) was higher among the cows calving in June vis-à-vis those calving in October and differences were also observed among the primiparous cows and multiparous cows, respectively. These metabolites (e.g., glycine, serine, etc.) which showed significant variations were mainly involved in the pathways of aminoacyl-tRNA biosynthesis, glyoxylate and dicarboxylate metabolism, and the metabolism of glycine, serine and threonine. These data suggested that heat stress negatively affected the elevation of the serum oxidative and antioxidative index and thus badly influence milk yield. Metabolic biomarkers in serum associated with heat stress could be a reliable way to identify heat stress of primiparas and multiparas dairy cows.
