Abstract:We utilized a microfluidic device with hydrodynamic flow focusing geometry to produce uniform agarose droplets in the range of 50 to 110 mum. The transport property of the thermally gelled particles was tailored by layer-by-layer (LBL) polyelectrolytes coating on the surface and was measured via the release rates of Rhodamine B. The mechanical strength of the capsules was further enhanced by a coating of silica nano-particles in addition to polyelectrolyte coatings. We demonstrated that yeast cells can be successfully encapsulated into agarose capsules.

Abstract:Hybrid CaCO3 particles were used as templates to fabricate two kinds of "micelles enhanced" polyelectrolyte capsules with three different polyel ectrolytes such as poly( allylamine hydrochloride), poly( sodium styrenesulfonate), and poly (di-allyldlmethylammonium chloride) via LbL technique. Both of (PAH/PSS)(6) and (PADA/PSS)(6) capsules had good spherical shape and good dispersion by CLSM. In contrast with conventional polyelectrolyte capsules, these novel micelles enhanced capsules had thick and compact capsule wall. Meanwhile, fine FESEM images showed that there was a significant difference in microstructure and topology between the two kinds of capsules made of different component polyelectrolytes.

Abstract:Reversible growth: Microcapsules were fabricated by layer-by-layer assembly of poly(diallyldimethyl ammonium chloride)/poly(styrene sulfonate) onto removable melamine formaldehyde particles. The capsules with swelling and shrinking features (see graphic) provide a novel route for microencapsulation. Loading of biomacromolecules such as dextran with ultrahigh molecular weight is achieved by spontaneously opening the capsule during core removal procedure, followed by closing the capsule in salt solution.

Abstract:“Micelle-enhanced” polyelectrolyte capsules were fabricated via a layer-by-layer technique, templated on hybrid calcium carbonate particles with built-in polymeric micelles based on polystyrene-b-poly(acrylic acid). Due to the presence of a large number of negatively charged micelles inside the polyelectrolyte capsule, which were liberated from templates, the capsule wall was reconstructed and had properties different to those of conventional polyelectrolyte capsules. This type of capsule could selectively entrap positively charged water-soluble substances. The encapsulation efficiency of positively charged substances was dependent on their molecular weight or size. For some positively charged compounds, such as rhodamine B and lysozyme, the concentration in the capsules was orders of magnitude higher than that in the incubation solution. In addition, in vitro release study suggested that the encapsulated compounds could be released through a sustained manner to a certain degree. All these results point to the fact that these capsules might be used as novel delivery systems for some water-soluble compounds.

Abstract:A gastro-retentive capsule has been prepared which is retained in the stomach for a period of 24h, providing a vehicle for the controlled delivery to the upper intestines. These "gastro cocoons" can resist passage through the sphincter of the stomach, and can retain a high drug payload (30%). They are made from oppositely charged polyelectrolytes and can swell to twice their initial volume. They are strong and also can resist 550 N of compressive force. They are based on filled pharmaceutical capsules which are visible to X-rays. Using ambroxol hydrochloride as a model drug linear, zero-order, release curves were obtained.

Abstract:Reprogramming of a differentiated cell into a cell capable of giving rise to many different cell types, a pluripotent cell, which in turn could repopulate or repair sick or damaged tissue, would present beneficial applications in regenerative medicine. Somatic cell nuclear transfer may offer this possibility, but technical hurdles and ethical frameworks currently prevent application of this technology in several countries. As a result, alternative strategies to reprogramming cell fate are being developed. This review briefly addresses somatic cell nuclear transfer and focuses on recent non-nuclear transfer-based approaches for reprogramming somatic cells and enhancing their differentiation potential. These include the fusion of somatic cells with embryonic stem cells, the treatment of somatic cells with extract of pluripotent cells and the retroviral transduction of somatic cells to overexpress pluripotency genes.

Abstract:When a linear or a four arm star-shaped polyglycidol is used as macroinitiator, densely grafted poly(glycidol-graft-epsilon-caprolactone) and poly(glycidol-graft-L-lactide) and loosely grafted poly[(glycidol-graft-epsilon-caprolactone)-co-glycidol] copolymers have been synthesized by chemical or, in the latter case, by enzymatic catalyzed ring-opening polymerization of epsilon-caprolactone and L-lactide. The well-defined copolymers possess similar molecular weights, but differ in their architecture, microstructure and chemical composition. The hydrolytic degradation behavior was studied in a phosphate buffer solution at pH 7.4 and 37 degrees C for up to 90 days. After different time periods, the mass loss was determined and the degraded copolymers were analyzed by means of NMR, size exclusion chromatography, and scanning electron microscopy. Compared to linear poly(epsilon-caprolactone), poly[(glycidol-graft-epsilon-caprolactone)-co-glycidol] shows a change of the degradation mechanism and a tremendous enhancement of polymer degradation. As this effect is attributed to the high concentration of hydroxy groups at the polyglycidol backbone, this work points out a new possibility to tailor the degradation profiles of polyesters by the introduction of functionality into the polymeric material.

Abstract:Core/shell microcapsules with low‐permeability membranes and controlled morphology are crucial for the delivery and controlled release of fragrance molecules, pharmaceuticals, inks, or vitamins. Design criteria for next generation microcapsules must include chemical and mechanical stability, and also provide enhanced substrate interactions to improve deposition onto relevant complex surfaces. Here, a coupled approach is presented to synthesize core/shell delivery systems by interfacial polymerization to enhance both the microcapsule–substrate interactions and the mechanical properties of the capsules to induce a burst‐type release. By combining membrane synthesis, nonlinear mechanics, interfacial rheology, analysis of mass transfer, and capsule morphology generated during interfacial polymerization, large permanent deformations into the capsule geometry are programmed, resulting in chemically stable, yet mechanically rupturing microcapsules with anisotropic geometry. To promote interactions and capsule adhesion onto complex substrates, the capsule contact area is controlled to form prominent “suction cup” shaped rims. These capsules have favorable, far‐reaching electrostatic interactions with oppositely charged substrates such as glass, hair, skin, or fabric. By modulating membrane mechanical properties and morphology during synthesis, formulation‐independent physical criteria are used to improve the overall performance of a functional delivery system while expanding knowledge of the key parameters influencing the interfacial polymerization process and membrane formation. Anisotropic core/shell microcapsules with “suction cup” morphologies provide enhanced affinity on complex substrates, such as the surface of human hair. The mechanics and permeability of the capsule membrane can be designed to synthesize chemically stable, yet mechanically rupturing targeted delivery systems for low‐molecular‐weight payloads.

Abstract:Stemming from early seminal notions of molecular recognition and encapsulation, three-dimensional, cavity-containing capsular compounds and assemblies have attracted intense interest due to the ability to modulate chemical and physical properties of species encapsulated within these confined spaces compared to bulk solution. With such a diverse range of covalent motifs and non-covalent (supramolecular) interactions available to assemble building blocks, an incredibly wide-range of capsular-type architectures have been developed. Furthermore, synthetic tunability of the internal environments gives chemists the opportunity to engineer systems for uses in sensing, sequestration, catalysis and transport of molecules, just to name a few. In this tutorial review, an overview is provided into the design principles, synthesis, characterisation, structural facets and properties of coordination cages, porous organic cages, supramolecular capsules and mechanically interlocked molecules. Using seminal and recent examples, the advantages and limitations of each system are explored, highlighting their application in various tasks and functions.

Abstract:Spatial and temporal controlled delivery of growth factors which contribute to efficient repair upon tissue injury or failure remains a major challenge in tissue engineering. In this paper, we evaluated polyelectrolyte multilayer microcapsules to control spatiotemporal release of transforming growth factor-β1 (TGF-β1). Polyelectrolyte microcapsules were bio-specifically engineered to enhance their ability to bind growth factors. Myofibroblast differentiation and contraction demonstrated that the bioactivity of TGF-β1 was not altered upon encapsulation into polyelectrolyte multilayer microcapsules and subsequent incorporation into gelatin based hydrogels. Finally, polyelectrolyte microcapsules were incorporated within a synthetic tissue engineering cryogel scaffold composed of gelatin without affecting the cryogel morphology, cell population capacity or mechanical properties of the scaffold.

Abstract:<p>A novel methodology, combining reactive gelation and droplet‐based microfluidics, is described for the synthesis of rigid, porous, and hollow polymeric nanoparticles (NPs). The precursors of such capsules are microfluidically generated latex droplets, with diameters tunable between 20 and 100 µm. The conversion of latex droplets to polymeric capsules involves two steps, namely self‐migration and gelation of the NPs toward the oil–water interface to form a solid‐like shell, and then postpolymerization to covalently fix the shell structure. The hollow structure of the capsules results from the interaction between negatively charged NPs inside the droplet and positive charges present on fluorosurfactant at the droplet interface. Significantly, the porosity and average pore size in the capsule shell can be controlled through variation of the initial NP concentration in the droplet. Based on the analysis of diffusion of fluorescent molecules of known size, it is shown that penetration of molecules into the internal volume of the capsules increases as the initial NP concentration in the droplet decreases, thus the porosity of the formed shell increases. This novel synthetic methodology defines a powerful tool in the generation of hollow capsules of controlled size and morphology, and has significant application in material sciences.</p>

Abstract:Hypothesis: Milli- and micro-capsules are developed to facilitate the controlled release of diverse active ingredients by passive diffusion or a triggered burst. As applications expand, capsules are required to be increasingly multi-functional, combining benefits like encapsulation, response, release, and even movement. Balancing the increasingly complex demands of capsules is a desire to minimize material usage, requiring efficient structural and chemical design. Designing multifunctional capsules with complex deformation should be possible even after minimizing the material usage through use of sparse fiber networks if the fibers are coated with responsive polymers. Experiments: Here capsules are created with a shell made from a mesh of nanoscale bacterial cellulose fibers that provide mechanical strength at very low mass levels, while a coating of thermoresponsive Poly(N-isopropylacrylamide), PNIPAM, on the fibers provides control of permeability, elastic response, and temperature response. These properties are varied by grafting different amounts of polymer using particular reaction conditions. Findings: The addition of PNIPAM to the cellulose mesh capsule enhances its mechanical properties, enabling it to undergo large deformations and recover once stress is removed. The increased elastic response of the capsule also provides reinforcement against drying-induced capillary stresses, limiting the degree of shrinkage during dehydration. Time-lapse microscopy demonstrates thermoreversible swelling of the capsules in response to temperature change. Cycles of swelling and shrinkage drive solvent convection to and from the capsule interior, allowing exchange of contents and mixing with the bulk fluid on a time scale of seconds. Because the cellulose capsules are produced via emulsion-templated fermentation, the polymer-modified biocapsule concept introduced here presents a pathway toward the sustainable and scalable manufacture of multifunctional responsive capsules.

Abstract:Proteins are desirable building blocks to create self-assembled, spatially defined structures and interfaces on length-scales that are inaccessible by traditional methods. Here, we describe a novel approach to create functionalized monolayers using the proteins BslA and SpyCatcher/SpyTag. BslA is a bacterial hydrophobin whose amphiphilic character underlies its ability to assemble into a monolayer at both air/water and oil/water interfaces. We demonstrate that Bsa1A having the SpyTag peptide fused at the N- or C-terminus does not affect the formation of such monolayers. We establish the creation of stable oil-in-water microcapsules using BslA, and also show the fabrication of capsules outwardly displaying the reactive SpyTag peptide by fusing it to the C-terminus of BslA. Such capsules can be covalently labeled by reacting the surface-displayed SpyTag with SpyCatcher fused to any desired protein. Injection-molded capsule bodies and caps based on polymer blends for controlled drug delivery

Abstract:A variety of polymer:polymer blends was used to prepare hot melt extrudates and empty capsules (bodies and caps) by injection-molding using a benchtop extruder (Babyplast). KollidonSR:inulin and Carbothane:inulin blends were investigated. The impact of the blend ratio on the water uptake and dry mass loss kinetics upon exposure to 0.1 MHCl, phosphate buffer pH6.8 and culture medium optionally inoculated with fecal samples from Inflammatory Bowel Disease (IBD) patients were studied. Hot melt extrudates were loaded with up to 60% theophylline, capsules were filled with drug powder. Increasing the inulin content led to increased water uptake and dry mass loss rates, resulting in accelerated drug release from the dosage forms, irrespective of the type of polymer blend. This can be attributed to the higher hydrophilicity/water-solubility of this polymer compared to KollidonSR and Carbothane. Interestingly, the presence of fecal samples in culture medium increased the water uptake and dry mass loss of hot melt extrudates to a certain extent, suggesting partial system degradation by bacterial enzymes. However, these phenomena did not translate into any noteworthy impact of the presence of colonic bacteria on theophylline release from the investigated extrudates or capsules. Hence, drug release can be expected to be independent of the location "small intestine vs. colon" from these dosage forms, which can be advantageous for long term release throughout the entire gastro intestinal tract.

Abstract:Various approaches have been described in the literature to demonstrate the possibility of designing biopolymer particles with well-defined characteristics, such as size, chemical composition or mechanical properties. From a biological point of view, the properties of particle have been related to their biodistribution and bioavailability. Among the reported core–shell nanoparticles, biopolymer-based capsules can be used as a versatile platform for drug delivery purposes. Among the known biopolymers, the present review focuses on polysaccharide-based capsules. We only report on biopolyelectrolyte capsules fabricated by combining porous particles as a template and using the layer-by-layer technique. The review focuses on the major steps of the capsule design, i.e., the fabrication and subsequent use of the sacrificial porous template, multilayer coating with polysaccharides, the removal of the porous template to obtain the capsules, capsule characterisation and the application of capsules in the biomedical field. In the last part, selected examples are presented to evidence the major benefits of using polysaccharide-based capsules for biological purposes.

Abstract:Microcapsules are a key class of microscale materials with applications in areas ranging from personal care to biomedicine, and with increasing potential to act as extracellular matrix (ECM) models of hollow organs or tissues. Such capsules are conventionally generated from non-ECM materials including synthetic polymers. Here, we fabricated robust microcapsules with controllable shell thickness from physically- and enzymatically-crosslinked gelatin and achieved a core-shell architecture by exploiting a liquid-liquid phase separated aqueous dispersed phase system in a one-step microfluidic process. Microfluidic mechanical testing revealed that the mechanical robustness of thicker-shell capsules could be controlled through modulation of the shell thickness. Furthermore, the microcapsules demonstrated environmentally-responsive deformation, including buckling by osmosis and external mechanical forces. A sequential release of cargo species was obtained through the degradation of the capsules. Stability measurements showed the capsules were stable at 37 °C for more than two weeks. Finally, all-aqueous liquid-liquid phase separated and multiphase liquid-liquid phase separated systems were generated with the gel-sol transition of microgel precursors. These smart capsules are promising models of hollow biostructures, microscale drug carriers, and building blocks or compartments for active soft materials and robots.

Abstract:Hollow permeable polysiloxane capsules with diameters between 180 and 210 nm were firstly fabricated by consecutively cocondensed methyltrimethoxysilane and dimethyldimethoxysilane monomers onto polydimethylsiloxane and subsequently removing the templated polydimethylsiloxane by exposure to solvents. The morphology of the polymer particles was demonstrated by transmission electron micrographs (TEM) and atomic force microscopy (AFM). One possible model was firstly proposed to gain the best illustration of the capsules. Indomethacin was originally encapsulated into the capsules using a solute-co-diffusion method.
