Abstract:We describe a new approach for the determination of volatile organic compounds (VOCs) at precise locations in the air using active sampling on sorbent tubes outfitted to an inexpensive multicopter and analysis by thermal desorption (TD) gas chromatography-mass spectrometry (GC-MS). The aerial sampling method permits for the simultaneous collection of multiple air samples on separate TD tubes concurrently, increasing sample throughput compared to single canister sampling. Furthermore, the method is relatively inexpensive when compared to similar approaches, with overall costs below about $2000 (U.S. dollars). To demonstrate applicability, we measured VOCs at several heights near anthropogenic sources in the mid-south USA, including a municipal landfill, petroleum refinery, and a coal-fired power plant (CFPP), and within the canopy of a loblolly pine (Pinus taeda) forest. Concentrations of benzene, toluene, and xylene (BTX) were higher (p < 0.05) downwind of the refinery and CFPP compared to upwind. We observed both a unique mixture of VOCs at each site and higher concentrations of abundant VOCs downwind compared to upwind of the point sources and within versus above the forest canopy. Overall, this feasibility study demonstrates that highly maneuverable multicopters can be used to probe VOC concentrations aloft and thus have great potential to be utilized in unique sampling situations and for vertical profiling.
Characterization of odorous industrial plumes by coupling fast and slow mass spectrometry techniques for volatile organic compounds

Abstract:This study aimed to develop a technique to chemically characterize odor issues in neighborhoods of designated industrial zones with pronounced emissions of volatile organic compounds (VOCs). Due to the elusive nature of odor plumes, speedy detection with sufficient sensitivity is required to capture the plumes. In this demonstration, proton-transfer-reaction mass spectrometry (PTR-MS) was used as the front-line detection tool in an industrial zone to guide sampling canisters for in-laboratory analysis of 106 VOCs by gas chromatography-mass spectrometry/flame ionization detector (GC-MS/FID). The fast but less accurate PTR-MS coupled with the slow but accurate GC-MS/FID method effectively eliminates the drawbacks of each instrument and fortifies the strength of both when combined. A 10-day PTR-MS field screening period was conducted to determine suitable trigger VOC species with exceedingly high mixing ratios that were likely the culprits of foul odors. Twenty canister samples were then collected, triggered by m/z 43, 61 (ethyl acetate, fragments, EA), m/z 73 (methyl ethyl ketone, MEK), or m/z 88 (morpholine) in all cases. Internal consistency was confirmed by the high correlation of critical species in the PTR-MS and trigger samples. Several long-lived halocarbons were exploited as the intrinsic internal reference for quality assurance. Oxygenated VOCs (OVOCs) accounted for 15%–75% of the total VOC mixing ratios in the triggered samples. However, EA and MEK, the most prominent OVOC species, did not appear to have common sources with morpholine, which presented with PTR-MS peaks incoherent with the other OVOCs. Nevertheless, these distinctive OVOC plumes were consistent with the multiple types of odor reported by the local residents. In contrast with the triggered sampling, random samples in the same industrial zone and roadside samples in a major metropolitan area were collected. The pronounced OVOC content in the triggered samples highlighted the advantage over random grab sampling to address odor issues.

Abstract:Assessing the odor risk caused by volatile organic compounds (VOCs) in water has been a big challenge for water quality evaluation due to the abundance of odorants in water and the inherent difficulty in obtaining the corresponding odor sensory attributes. Here, a novel odor risk assessment approach has been established, incorporating nontarget screening for odorous VOC identification and machine learning (ML) modeling for odor threshold prediction. Twenty-nine odorous VOCs were identified using two-dimensional gas chromatography-time of flight mass spectrometry from four surface water sampling sites. These identified odorants primarily fell into the categories of ketones and ethers, and originated mainly from biological production. To obtain the odor threshold of these odorants, we trained an ML model for odor threshold prediction, which displayed good performance with accuracy of 79%. Further, an odor threshold-based prioritization approach was developed to rank the identified odorants. 2-Methylisoborneol and nonanal were identified as the main odorants contributing to water odor issues at the four sampling sites. This study provides an accessible method for accurate and quick determination of key odorants in source water, aiding in odor control and improved water quality management. Environmental Implication Water odor episodes have been persistent and significant issues worldwide, posing severe challenges to water treatment plants. Unpleasant odors in aquatic environments are predominantly caused by the occurrence of a wide range of volatile organic chemicals (VOCs). Given the vast number of newly-detected VOCs, experimental identification of the key odorants becomes difficult, making water odor issues complex to control. Herein, we propose a novel approach integrating nontarget analysis with machine learning models to accurate and quick determine the key odorants in waterbodies. We use the approach to analyze four samples with odor issues in Changsha, and prioritized the potential odorants.

Abstract:Complex inter-organismal communication among plants, insects, and microbes in natural and agricultural ecological systems is typically governed by emitted and perceived semiochemicals. To understand and ultimately utilize the role of volatile semiochemicals in these interactions, headspace volatiles are routinely collected and analyzed. Numerous collection systems are available (e.g., static or dynamic; adsorption or absorption) where the choice of technique should be dependent upon the plant, insect, or microbial ecological system studied, the information sought, and the limitations of each method. Within these constraints, it remains necessary that each method detects and provides the accurate in situ, or in vitro, volatile profile of the studied system. Herein, we analyzed and compared the pros and cons of three solventless, thermal desorption systems (SPME, Tenax/cold trap, SPDE) using a synthetic standard blend of compounds mimicking a simple natural blend (benzaldehyde, b-caryophyllene, (Z)-3-hexenol, 6-methyl-5-hepten-2-one, and limonene). Direct splitless injection and Super Q collections of the standard blend were used as controls. The results indicated that related qualitative, as well as quantitative differences, could be correlated with adsorbent sampling capacity and structural bias. The results for Tenax/cold trap and SPDE also were affected by sampled headspace volumes. All solventless techniques exhibited high analytical reproducibility, with SPME and SPDE providing ease of use, low cost, and minimal instrument modifications. The more complex Tenax/cold trap technique provided higher collection efficiency. Using these results, we provide guidance for technique selection for chemical communication applications

Abstract:Few analytical methods exist that combine chemical and sensory analysis of odorous compounds in whole air. Volatile organic compounds were collected by sampling air downwind from a small dairy through sorbent tubes of Tenax TA and Carboxen 569. Samples were analyzed by thermal desorption into a cryotrap and subsequent gas chromatographic separation, followed by simultaneous olfactometry and mass spectrometry. Because compounds are concentrated during sampling, sensory analysis encountered compounds at a concentration 40 times that in air, making this a useful method for identifying trace compounds participating in odor. Twenty odorous and nonodorous compounds were identified and quantified, including straight-chain and aromatic hydrocarbons, chlorinated compounds, alcohols, ketones, aldehydes, and organic acids, at air concentrations of 0.55-320.20 microg/m(3). Compound peaks were characterized by odors ranging from offensive to pleasant, demonstrating the integrative nature of olfaction. This method could be useful in studying many kinds of odors in air.

Abstract:ASTM international standard practice E679 prescribes the use of a 3-alternative forced-choice (3AFC) method with an ascending concentration series for measuring odour detection thresholds. It recommends obtaining an individual’s threshold by geometrically averaging the concentrations at which the judge’s detection response reverses from incorrect to correct. The legitimate reversal point is defined by a stopping rule. Previous researchers have identified some methodological flaws with this calculation approach, relating to the use of stopping rules and the method’s unconventional definition of thresholds. The current study aims to empirically investigate these issues in more depth.Thresholds for three odorants were obtained for 100 judges. 1,8-Cineole, isobutyraldehyde and β-damascenone were tested in accordance with the data collection procedure of ASTM E679. Initially, thresholds were estimated by the ASTM-based method with four different stopping rules. These estimates were subsequently compared against thresholds estimated by an alternative method; that of fitting psychometric functions. The theoretical grounds on which the latter method is based, and the clear connection between performance and stimulus concentration that it demonstrates, provide a thorough comparison of both conventionally-defined thresholds and method-independent thresholds against the ASTM-based method.Comparisons between the thresholds resulting from the various estimation methods demonstrated (1) the choice of stopping rule had a significant effect on threshold estimates (P<.05); (2) each stopping rule measured a specific level of performance, indicating a useful correspondence between the stopping rules and the psychometric function. These findings provide supporting evidence for the continued use of the ASTM-based method; albeit, if the stopping rule is standardized. One of the tested stopping rules (Stop 3) provided threshold estimates consistent with those estimated for d′=1.0, and is therefore recommended for future research.

Abstract:Characterization and quantification of livestock odorants is one of the most challenging analytical tasks because odor-causing gases are very reactive, polar and often present at very low concentrations in a complex matrix of less important or irrelevant gases. The objectives of this research is to develop a novel analytical method for characterization of the livestock odorants including their odor character, odor intensity, and hedonic tone and further quantitative analysis of the key odorants responsible for livestock odor emissions. Sorbent tubes packed with Tenax TA were employed for sampling. The automated one-step thermal desorption coupled with multidimensional gas chromatography-mass spectrometry-olfactometry system was developed for simultaneous chemical and odor analysis. Fifteen odorants identified from different livestock species operations are quantified. In addition, odor character, odor intensity and hedonic tone associated with each of the target compounds are also analyzed. The method developed in this research is being used on a multistate, multispecies project focused on quantifying odor and chemical analysis of odor.
PHYLOGENETIC ANALYSIS OF THE NEST ARCHITECTURE OF NEOTROPICAL OVENBIRDS (FURNARIIDAE)

Abstract:We reviewed the tremendous architectural diversity of ovenbird (Furnari- idae) nests based on literature, museum collections, and new field observations. With few exceptions, furnariids exhibited low intraspecific variation for the nest characters hypothe- sized, with the majority of variation being hierarchically distributed among taxa. We hy- pothesized nest homologies for 168 species in 41 genera (ca. 70% of all species and genera) and coded them as 24 derived characters. Forty-eight most-parsimonious trees (41 steps, CI = 0.98, RC = 0.97) resulted from a parsimony analysis of the equally weighted characters using PAUP, with the Dendrocolaptidae and Formicarioidea as successive outgroups. The strict-consensus topology based on these trees contained 15 clades representing both tra- ditional taxa and novel phylogenetic groupings. Comparisons with the outgroups demon- strate that cavity nesting is plesiomorphic to the furnariids. In the two lineages where the primitive cavity nest has been lost, novel nest structures have evolved to enclose the nest contents: the clay oven of Furnarius and the domed vegetative nest of the synallaxine clade. Although our phylogenetic hypothesis should be considered as a heuristic prediction to be tested subsequently by additional character evidence, this first cladistic analysis of the fur- nariids demonstrates the general utility of nest characters in reconstruction of avian rela- tionships, and it provides a test of monophyly for several furnariid taxa. Abstract:Volatile organic compounds (VOCs) are of interest in many different fields. Among them are food and fragrance analysis, environmental and atmospheric research, industrial applications, security or medical and life science. In the past, the characterization of these compounds was mostly performed via sample collection and off-site analysis with gas chromatography coupled to mass spectrometry (GC-MS) as the gold standard. While powerful, this method also has several drawbacks such as being slow, expensive, and demanding on the user. For decades, intense research has been dedicated to find methods for fast VOC analysis on-site with time and spatial resolution. We present the working principles of the most important, utilized, and researched technologies for this purpose and highlight important publications from the last five years. In this overview, non-selective gas sensors, electronic noses, spectroscopic methods, miniaturized gas chromatography, ion mobility spectrometry and direct injection mass spectrometry are covered. The advantages and limitations of the different methods are compared. Finally, we give our outlook into the future progression of this field of research.

Abstract:The lack of one-to-one olfactory thresholds (OTs) poses an obstacle to the comprehensive assessment of priority odorants emitted from swine slurry using mass spectrometric nontarget screening. This study screened out highly performing quantitative structure–activity relationship (QSAR) models of OT prediction to complement nontarget screening in olfactory perception evaluation. A total of 27 compounds emitted at different slurry removal frequencies were identified and quantified using gas chromatography–mass spectrometry (GC–MS), including thiirane, dimethyl trisulfide (DMTS), and dimethyl tetrasulfide (DMQS) without OT records. Ridge regression (RR, R 2 = 0.77, RMSE = 0.93, MAE = 0.73) and random forest regression (RFR, R 2 = 0.76, RMSE = 0.97, MAE = 0.69) rather than the commonly used principal component regression (PCR) and partial least squares regression (PLSR) were used to assign OTs and assess the contributions of emerging volatile sulfur compounds (VSCs) to the sum of odor activity value (SOAV). Priority odorants were p -cresol (25.0–58.9 %) > valeric acid (8.3–31.7 %) > isovaleric acid (6.7–19.0 %) > dimethyl disulfide (4.7–15.7 %) > methanethiol (0–13.6 %) > isobutyric acid (0–8.6 %), whereas the contributions of three emerging VSCs were below 10 %. Vital olfactory active structures were identified by QSAR models as having high molecular polarity, high hydrophilicity, high charge quantity, flexible structure, high reactivity, and a high number of sulfur atoms. This protocol can be further extended to evaluate odor pollution levels for distinct odor sources and guide the development of pertinent deodorization technologies.

Abstract:Bioaerosols (or biogenic aerosols) have largely been overlooked by molecular ecologists. However, this is rapidly changing as bioaerosols play key roles in public health, environmental chemistry and the dispersal ecology of microbes. Due to the low environmental concentrations of bioaerosols, collecting sufficient biomass for molecular methods is challenging. Currently, no standardized methods for bioaerosol collection for molecular ecology research exist. Each study requires a process of optimization, which greatly slows the advance of bioaerosol science. Here, we evaluated air filtration and liquid impingement for bioaerosol sampling across a range of environmental conditions. We also investigated the effect of sampling matrices, sample concentration strategies and sampling duration on DNA yield. Air filtration using polycarbonate filters gave the highest recovery, but due to the faster sampling rates possible with impingement, we recommend this method for fine -scale temporal/spatial ecological studies. To prevent bias for the recovery of Gram-positive bacteria, we found that the matrix for impingement should be phosphate-buffered saline. The optimal method for bioaerosol concentration from the liquid matrix was centrifugation. However, we also present a method using syringe filters for rapid in-field recovery of bioaerosols from impingement samples, without compromising microbial diversity for high -throughput sequencing approaches. Finally, we provide a resource that enables molecular ecologists to select the most appropriate sampling strategy for their specific research question.

Fifteen odorous VOCs identified from livestock operations were quantified. Method detection limits ranged from 30pg for indole to 3590pg for acetic acid per sample. In addition, odor character, odor intensity, and hedonic tone associated with each of the target odorants were also analyzed simultaneously. The mass of each VOC in the sample correlated well with the log stimulus intensity of odor. All of the coefficients of determination (R2) were greater than 0.74, and the top 10 R2s were greater than 0.90. Field air samples from swine and dairy operations confirmed that target compounds quantified represented typical odor-causing compounds emitted from livestock.
Real-Time Monitoring of Volatile Organic Compounds in Ambient Air Using Direct-Injection Mass Spectrometry

Abstract:Exposure to volatile organic compounds (VOCs) at low parts per billion (ppb) concentrations (by volume, ppbV) in ambient air is increasingly being recognized as a health risk. Furthermore, VOCs contribute to the formation of photochemical smog and particulate matter, which are hazardous and negatively impact the environment. Although most urban VOCs are anthropogenic in origin, some are produced by living organisms, especially trees. Gas chromatography (GC) is conventionally applied to VOC analysis of air, but it generally has a time resolution of tens of minutes and detection at low ppbV levels, frequently requiring sample preconcentration (such as using thermal desorption tubes (TDTs) or canister sampling with cryotrapping prior to analysis). Newer direct-injection mass spectrometry (DIMS) techniques can address the challenge of time resolution (seconds to tens of seconds) and eliminate the need for sampling via TDTs or canisters while still providing sufficient specificity. In this article, the results of a DIMS air monitoring campaign in Auckland, New Zealand, are described. Real-time selected ion flow tube mass spectrometry (SIFT–MS) measurements were obtained at three sites over a period of several weeks each: (i) adjacent to a freeway, (ii) in a residential area, and (iii) at a rural location. There were 13 compounds that were targeted in the study, covering various sources: fugitive emissions and incomplete combustion (benzene, toluene, and ethylbenzene, along with the xylene isomers, styrene, and ethanol), incomplete combustion (1,3-butadiene, formaldehyde, acetaldehyde, acrolein, benzaldehyde, and acetone), and biogenic origin (isoprene and monoterpenes (total of all isomers). At each site, SIFT–MS provided effective on-site analysis to sub-ppbV levels with better than 1-min time resolution.

Abstract:Anterior uveitis secondary to the herpes simplex virus (HSV) is a common finding in patients with herpetic corneal disease. Abstract:This review summarizes literature data from the past 5 years on new developments and/or applications of sample preparation methods for analysis of volatile organic compounds (VOC), mainly in air and water matrices. Next, focus is put on current tendencies in integrated sampling-extraction-sample introduction methods such as solid phase microextraction (SPME) and novel in-needle trapping devices. Particular attention is paid to emerging membrane extraction techniques such as membrane inlet mass spectrometry (MIMS) and membrane extraction with a sorbent interface (MESI). For VOC enrichment out of water, recent evolutions in direct aqueous injection (DAI) and liquid-liquid extraction (LLE) are highlighted, with main focus on miniaturized solvent extraction methods such as single drop microextraction (SDME) and liquid phase microextraction (LPME). Next, solvent-free sorptive enrichment receives major attention, with particular interest for innovative techniques such as stir bar sorptive extraction (SBSE) and solid phase dynamic extraction (SPDE). Finally, recent trends in membrane extraction are reviewed. Applications in both immersion and headspace mode are discussed.

Abstract:This study modified a passive sampling technique similar to the US EPA Method 325 A/B method but extended to include more toxic volatile organic compounds (VOCs) under varied climate conditions to enhance field applicability. A mixing chamber was built to determine uptake rates (Us) for the target compounds. It was found that the Us of 27 air toxics previously reported in the literature agreed reasonably well with our findings within 18%, thus proving the chamber's integrity. To broaden the compound coverage, both Carbopack X and Carboxen 569 were studied for a suite of toxic VOCs to meet stringent quality control (QC) criteria of correlation coefficients (R-square), method detection limits (MDL), back diffusion (BD), storage stability, as well as a wide range of climate conditions in temperature and humidity. After excluding the species that failed to pass any of the QC criteria, Carbopack X was found to fit 50 air toxics, whereas Carboxen 569 held 37. After excluding the overlapped species, 61 toxic VOCs can be determined with robust Us for a broad range of climate conditions when the two sorbents are used in pairs. A one-week field measurement was conducted to compare with the online thermal desorption gas chromatography-mass spectrometry (TD-GC-MS) with hourly data resolution. The field passive sampling showed comparable results to the means of the online hourly measurements, despite the high variability of selected target compounds, such as toluene from 0.3 ppbv as the 5th percentile to the maximum of about 80 ppbv. Passive sampling clearly demonstrated the ability to smooth out concentration variability and thus the time-averaging strength of toxic VOCs, revealing its ideal role as an exposure monitor over time. The passive sampling method can be more desired than active sampling or online methods when the aim is simply the knowledge of prolonged time-averaged concentrations.

Abstract:Disease detection and monitoring using Volatile Organic Compounds (VOCs) is becoming increasingly popular. For a variety of (gastrointestinal) diseases the microbiome should be considered. As its output is to large extent volatile, faecal volatilomics carries great potential. One technical limitation is that current faecal headspace analysis requires specialized instrumentation which is costly and typically does not work in harmony with Thermal Desorption units often utilized in e.g. exhaled breath studies. This lack of harmonization hinders uptake of such analyses by the Volatilomics community. Therefore, this study optimized and compared two recently harmonized faecal headspace sampling platforms: High-capacity Sorptive extraction (HiSorb) probes and the Microchamber thermal extractor (Microchamber). Statistical Design of Experiment (DOE) was applied to find optimal sampling conditions by maximizing reproducibility, the number of VOCs detected, and between subject variation. To foster general applicability those factors were defined using semi-targeted as well as untargeted metabolic profiles. HiSorb probes were found to result in a faster sampling procedure, higher number of detected VOCs, and higher stability. The headspace collection using the Microchamber resulted in a lower number of detected VOCs, longer sampling times and decreased stability despite a smaller number of interfering VOCs and no background signals. Based on the observed profiles, recommendations are provided on pre-processing and study design when using either one of both platforms. Both can be used to perform faecal headspace collection, but altogether HiSorb is recommended.

Abstract:Semi-volatile organic compounds (SVOCs) are important pollutants in indoor environments. Quantification of gaseous SVOC concentrations is essential to assess the pollution levels. Solid-phase microextraction (SPME) is considered to be an attractive sampling technique with merits, including simplicity of use, rapid sampling, and solvent free. However, the applications of SPME for sampling gaseous SVOCs are often limited by the fluctuating velocity of indoor air (leading to an unstable sampling rate) and the uncertainties associated with the traditional calibration of SPME. Therefore, we established an SPME-based active sampler to ensure the stable sampling of SVOCs in fluctuating air and developed a two-step calibration method based on the sampling principle of SPME. The presented method and a traditional method (sorbent tubes packed with Tenax TA) were simultaneously used to measure SVOC concentrations in an airstream generated in experiments. Three typical indoor SVOCs, diisobutyl phthalate (DiBP), tris (1-chloro-2-propyl) phosphate (TCPP), and benzyl butyl phthalate (BBzP) were chosen as the analytes. Mean concentrations measured by SPME agreed well with the sorbent tubes (relative deviations <12%), supporting the feasibility of the presented method. Further studies are expected to facilitate the application of the presented method (especially the problem associated with the sampling-tube loss of low volatile SVOCs).
