Abstract:BACKGROUND: Cryptococcus laurentii is a well-known postharvest yeast antagonist. N-6-benzyladenine (6-BA), a cytokinin plant hormone, has a role in retarding ripening and senescence of harvested produce. This study was conducted to evaluate the efficacy of C. laurentii and 6-BA in reducing the blue mould disease of apple fruit.RESULTS: The combination of C. laurentii with 6-BA (20 mu g mL(-1)) was more effective in suppressing the Penicillium expansum infection in apple fruit wounds than C. laurentii alone, although 6-BA (20 mu g mL(-1)) alone neither affected the growth of C. laurentii nor reduced the incidence of the blue mould disease in vivo. Moreover, treatment of apple fruit with C. laurentii and 6-BA (20 mu g mL(-1)) resulted in stimulation of superoxide dismutase activity but in inhibition of the increase in peroxidase activity.CONCLUSION: 6-BA (20 mu g mL(-1)) could enhance the efficacy of C. laurentii in reducing the postharvest blue mould disease of apple fruit, which offered great potential in minimizing the postharvest decay of apple fruit in an integrated pest management strategy. Isolation, Morphological Characterization, and Phylogenetic Identification of Botrytis cinerea Associated with Postharvest Gray Mold of Apple Fruits

Abstract:Apple (Malus domestica Borkh L., Family Rosaceae) is among the most popular fruits worldwide. Apple fruits are attacked by several fungal pathogens both pre- and post-harvest. The post-harvest fruit rot of apples is one of the most threatening dis-eases in apple cultivation. In the current study, we aimed to isolate, characterize, and identify the fungal phytopathogens associated with postharvest rot of apple fruits. Herein, we isolated eight genera from rotted Anna apple fruits during the 2018 season from orchards, wholesale, and retail markets as well as refrigerators. Then, the dif-ferent fungal pathogens were identified according to their cultural and microscopic characteristics as Penicillium expansum, Alternaria alternate, Botrytis cinerea, As-pergillus niger, Mucor sp., Rhizopus stolonifer, Stemphyllum sp., and Trichothecium roseum. In this study, investigations were made into the pathogenicity tests of the highest-frequency fungi. Pathogenicity tests on apples (Anna cv.) revealed that B. cinerea was the highest virulent fungus. Additionally, a simple linear regression (SLR) analysis was carried out to better understand the relationship between the di-ameter of the infected area caused by post-harvest pathogens and storage time. Therefore, B. cinerea was identified by molecular testing using internal transcribed spacer (ITS) sequencing and phylogenetic analyses. Postharvest gray mold, caused by Botrytis cinerea, is one of the most important diseases and seriously affects apple fruit quality, resulting in huge economic losses. Identification and evaluation of an endophytic antagonistic yeast for the control of gray mold (Botrytis cinerea) in apple and mechanisms of action

Abstract:Gray mold, caused by Botrytis cinerea, is a prevalent postharvest disease of apple that limits their shelf life, resulting in significant economic losses. The use of antagonistic microorganisms has been shown to be an effective approach for managing postharvest diseases of fruit. In the present study, an endophytic yeast strain PGY-2 was isolated from apples and evaluated for its biocontrol efficacy against gray mold and its mechanisms of action. Results indicated that strain PGY-2, identified as Bullera alba, reduced the occurrence of gray mold on apples and significantly inhibited lesion development in pathogen-inoculated wounds. Gray mold control increased with the use of increasing concentrations of PGY-2, with the best disease control observed at 108 cells/mL. Notably, Bullera alba PGY-2 did not inhibit the growth of Botrytis cinerea in vitro indicating that the yeast antagonist did not produce antimicrobial compounds. The rapid colonization and stable population of PGY-2 in apple wounds at 4 °C and 25 °C confirmed its ability to compete with pathogens for nutrients and space. PGY-2 also had a strong ability to form a biofilm and enhanced the activity of multiple defense-related enzymes (POD, PPO, APX, SOD, PAL) in host tissues. Our study is the first time to report the use of Bullera alba PGY-2 as a biocontrol agent for postharvest diseases of apple and provide evidence that Bullera alba PGY-2 represents an endophytic antagonistic yeast with promising biocontrol potential and alternative to the use of synthetic, chemical fungicides for the control of postharvest gray mold in apples.
Role of lipopeptides produced by Bacillus subtilis GA1 in the reduction of grey mould disease caused by Botrytis cinerea on apple

Abstract:Aim: Test of Bacillus subtilis strain GA1 for its potential to control grey mould disease of apple caused by Botrytis cinerea. Methods and results: GA1 was first tested for its ability to antagonize in vitro the growth of a wide variety of plant pathogenic fungi responsible for diseases of economical importance. The potential of strain GA1 to reduce post-harvest infection caused by B. cinerea was tested on apples by treating artificially wounded fruits with endospore suspensions. Strain GA1 was very effective at reducing disease incidence during the first 5 days following pathogen inoculation and a 80% protection level was maintained over the next 10 days. Treatment of fruits with an extract of GA1 culture supernatant also exerted a strong preventive effect on the development of grey mould. Further analysis of this extract revealed that strain GA1 produces a wide variety of antifungal lipopeptide isomers from the iturin, fengycin and surfactin families. A strong evidence for the involvement of such compounds in disease reduction arose from the recovery of fengycins from protected fruit sites colonized by bacterial cells. Conclusions: The results presented here demonstrate that, despite unfavourable pH, B. subtilis endospores inoculated on apple pulp can readily germinate allowing significant cell populations to establish and efficient in vivo synthesis of lipopeptides which could be related to grey mould reduction. Significance and impact of the study: This work enables for the first time to correlate the strong protective effect of a particular B. subtilis strain against grey mould with in situ production of fengycins in infected sites of apple fruits.

Abstract:The use of microbial biocontrol agents for control of postharvest disease has been the subject of intensive research over the past three decades resulting in commercialization of several biocontrol products. The objective of this research was to test endospore-forming bacteria collected from apple leaves for suppression of bitter rot and blue mold on apple. Bacteria were collected from abandoned, low-input, organic, and conventionally managed orchards in Pennsylvania and were screened for their ability to produce endospores, hydrolyze chitin, reduce pathogen growth in vitro, and suppress postharvest disease in vivo. Several isolates reduced bitter rot lesion size on ‘Rome Beauty’ from 40–89% compared to untreated controls. Bacillus megaterium isolates, A3-6 and Ae-1, resulted in the greatest suppression of bitter rot lesion size. One isolate, A3-2, suppressed blue mold lesion size. Scanning electron microscopy of inoculated apple wounds suggests parasitism as a mode of action explains the suppression of bitter rot lesion size by isolate A3-6. Of the top seventeen isolates exhibiting biocontrol potential, 70% were collected from abandoned or unmanaged locations. This research demonstrates abandoned apple orchards can be a source of new biocontrol agents for control of postharvest diseases of apple.

Abstract:Sustainable plant protection can be applied on apples against fungal pathogens such as Botrytis cinerea (which is responsible for gray mold)-a significant global postharvest disease. This pathogen can affect a wide range of hosts; and fruits may have variable susceptibilities to B. cinerea from different plant hosts. New possibilities to control gray mold in food production are under demand due to the emergence of resistance against antifungal agents in fungal pathogens. Cinnamon, pimento, and laurel extracts were previously assessed for antifungal activities under in vitro conditions and were found to have the potential to be effective against postharvest gray mold. Therefore, this study aimed to investigate the antifungal activity of cinnamon, pimento, and laurel extracts in vitro and against postharvest gray mold on apples to determine the susceptibility of apple fruits to B. cinerea from different plant hosts, and to analyze the chemical composition of the extracts. Apples (cv. "Connell Red") were treated with different concentrations of extracts and inoculated with B. cinerea isolates from apple and strawberry followed by evaluation of in vitro antifungal activity. The results reveal that most of the concentrations of the extracts that were investigated were not efficient enough when assessed in the postharvest assay, despite having demonstrated a high in vitro antifungal effect. Apples were less susceptible to B. cinerea isolated from strawberry. To conclude, cinnamon extract was found to be the most effective against apple gray mold; however, higher concentrations of the extracts are required for the efficient inhibition of B. cinerea in fruits during storage.

Abstract:Aureobasidin A (AbA), an antifungal cyclic depsipeptide antibiotic produced by Aureobasidium pullulans R 106, has previously been shown to be effective against a wide range of fungi and protozoa. Here we report the inhibitory effects of AbA on spore germination, germ tuber elongation and hyphal growth of five pathogenic fungi including Penicillium digitatum, P. italicum, P. expansum, Botrytis cinerea and Monilinia fructicola, which are major pathogens causing postharvest diseases of a variety of fruits. AbA inhibited five pathogenic fungi by reducing conidial germination rates, delaying conidial germination initiation, restricting elongation of germ tuber and mycelium, as well as inducing abnormal alternations of morphology of genii tubes and hyphae of these fungi. The sensitivity of these fungi to AbA was pathogen species-dependent. P digitatum was the most sensitive and M.fructicola the least. Importantly, AbA at 50 mu g/ml was effective in controlling the citrus green mold and in reducing the strawberry gray mold incidence and severity, caused by P. digitatum and B. cinerea, respectively, after artificial inoculation. AbA and/or its analogs, therefore, hold promise as relatively safe and promising fungicide candidates to control postharvest decays of fruits, because AbA targets the inositol phosphorylceramide (IPC) synthase, an enzyme essential for fungi but absent from mammals. Abstract:The efficacy of Rhodotorula mucilaginosa against postharvest gray mold, blue mold and natural decay development of apples and the possible mechanisms involved were investigated. The decay incidence and lesion diameter of gray mold and blue mold of apples treated by R. mucilaginosa were significantly reduced compared with the control fruits, and the higher concentration of R. mucilaginosa, the better the efficacy of the biocontrol. R. mucilaginosa also significantly reduced the natural decay development of apples following storage at 20°C for 35 days or at 4°C for 45 days followed by 20°C for 15 days. Germination and survival of spores of Penicillium expansum and Botrytis cinerea were markedly inhibited by R. mucilaginosa in an in vitro test. Rapid colonization of the yeast in apple wounds was observed whether stored at 20°C or 4°C. In apples, the activities of peroxidase (POD) and polyphenoloxidase (PPO) were significantly induced and lipid peroxidation (malondialdehyde (MDA) content) was highly inhibited by R. mucilaginosa treatment compared with those of the control fruits. All these results indicated that R. mucilaginosa has great potential for development of commercial formulations to control postharvest pathogens on fruits. Its modes of action were based on competition for space and nutrients with pathogens, inducement of activities of defense-related enzymes such as POD, PPO and inhibition of lipid peroxidation (MDA content) of apples, so as to enhance the resistance and delay the ripening and senescence of apples.

Abstract:The antagonistic activity of three isolates of Pseudomonas fluorescens, 1–112, 2–28 and 4–6, to control Botrytis cinerea (grey mold) under commercial cold storage conditions with two varieties of apple, ‘Ambrosia’ and ‘Spartan’, and possible mechanisms involved were investigated. All three isolates of P. fluorescens significantly reduced the size of the lesion and the incidence of grey mold on ‘Ambrosia’ apples after 15weeks in cold storage, in comparison to control fruits, and disease control was comparable to that of the commercial biofungicide Bio-Save® (Pseudomonas syringae). On ‘Spartan’ apples only isolates 1–112 and 4–6 significantly reduced the size of the lesion and the incidence of grey mold, after 15weeks in cold storage, and both isolates were comparable to the synthetic fungicide Scholar® (fludioxonil). Living cells of P. fluorescens or their metabolites markedly inhibited spore germination of B. cinerea in filter-sterilized apple juice (FSAJ). Cell-free supernatant and volatile organic compounds (VOCs) produced in vitro by P. fluorescens inhibited the mycelial growth of B. cinerea by 58.9 and 82.2%, respectively. Scanning electron microscopy (SEM) observations indicated that P. fluorescens colonized the wounds of apples and isolates 1–112 and 4–6 formed a biofilm at the wound site. Competition for nutrients and space, production of toxic metabolites and biofilm formation may play a role in the antagonism of P. fluorescens 1–112, 2–28 and 4–6 to B. cinerea.

Abstract:The biocontrol activity of Rhodotorula glutinis on gray mold decay and blue mold decay of apple caused by Botrytis cinerea and Penicillium expansum, respectively, was investigated, as well as its effects on postharvest quality of apple fruits. The results show there was a significant negative correlation between concentrations of the yeast cells and the disease incidence of the pathogens. The higher concentration of the R. glutinis, the better effect of the biocontrol capacity. At concentrations of R. glutinis 1×108CFUml−1, the amount of gray mold decay was completely inhibited after 5 days incubation at 20°C, after challenge with B. cinerea spores suspension of 1×105sporesml−1; While the blue mold decay was completely inhibited at concentrations of 5×108CFUml−1, at challenged with P. expansum spores suspension of 5×104sporesml−1. These results demonstrated that the efficacy of R. glutinis in controlling of gray mold decay of apples was better than the efficacy of controlling blue mold. R. glutinis within inoculated wounds on apples increased in numbers at 20°C from an initial level of 9.5×105CFU per wound to 2.24×107CFU at 20°C after 1 day. The highest population of the yeast was recovered 4 days after inoculation, the yeast population in wounds increased by 56.9 times. After that, the population of the yeast began to decline very slowly. R. glutinis significantly reduced the incidence of natural infections on intact fruit from 75% in the control fruit to 28.3% after 5 days at 20°C, and from 58.3 to 6.7% after 30 days at 4°C followed by 4 days at 20°C. R. glutinis treatment had no deleterious effect on quality parameters after 5 days at 20°C or after 30 days at 4°C followed by 4 days at 20°C.

Abstract:This study evaluated the effect of a yeast antagonist Cryptococcus laurentii and a plant regulator indole-3-acetic acid (IAA) on inhibition of Botrytis cinerea infection in harvested apple fruit. The results showed that the combined treatment with C. laurentii and IAA at 20μg/ml was a more effective approach to reduce the gray mold rot in apple wounds than the C. laurentii alone. After 4 days of incubation, gray mold incidence in the combined treatment with C. laurentii and IAA was about 18%, which was a 50% reduction in incidence compared to the treatment with C. laurentii alone. Although IAA had no direct antifungal activity against B. cinerea infection when the time interval between IAA treatment and pathogen inoculation was within 2h, application of IAA strongly reduced gray mold infection when IAA was applied 24h prior to inoculation with B. cinerea in apple fruit wounds. Moreover, combination of IAA and C. laurentii stimulated the activities of superoxide dismutase, catalase and peroxidase with above 1.5-fold higher than that treatment with C. laurentii alone at 48h. Therefore, combination of C. laurentii with IAA, which integrated the dual biological activity from the antagonistic yeast and plant regulator, might be developed to be a useful approach to control gray mold in harvested apple fruit.
An evaluation of leucocyte adherence inhibition microassay using colorectal cancer cell lines.

Abstract:The leucocyte adherence inhibition (LAI) microassay using crude extracts from colorectal cancer cell lines (M-7609 and S-7512) was evaluated as a test of immunological antitumor reactivity in colorectal cancer patients. The optimal antigen concentration for differentiating cancer patients from control subjects was 0.05 mg/ml protein. The cut-off values of LAI index for determining reactive and nonreactive cases was -0.20. The numbers of colorectal cancer patients reactive against antigens M (derived from M-7609), S (derived from S-7512) and M + S were 9 of 19, 9 of 12 of 18, respectively, whereas against control antigens K (derived from K-7610, pulmonary cancer), SF (derived from cultured fibroblastic cells) and T (derived from fresh colorectal cancer tissue), they were only 1 of 18, 1 of 17, and 3 of 15, respectively. The mean LA indices of the Dukes A and B group, and of the Dukes C and recurrent cancer group were -0.28 and -0.24, but that of a tumor-free postoperative group, another malignant disease group and a group of healthy individuals were -0.04, -0.05, and -0.03, respectively. Judging from these results, LAI microassay using cultured human cancer cell lines as tumor antigen sources is suitable as a test for evaluating the antitumor immunmoreactivity of colorectal cancer patients.
Highlighting the complexity of pathogenesis: the host microbiota impacts disease development on apple fruit and is a cornerstone for its biocontrol

Abstract:Apple fruit is the most produced temperate fruit with a trade value estimated at 7.5 billion $ and is usually stored up to one year after harvest. Postharvest pathogens often compromise storage, responsible for up to 55% of fruit losses, depending on the country and fruit. They are also a source of mycotoxin contamination. A sustainable way to control that pathogen is using beneficial microorganisms called biocontrol agents (BCA). Despite promising efficacy in laboratory conditions, BCA's efficacy is variable and often reduced once applied at a large scale through either orchard or postharvest treatment. We hypothesized that the epiphytic microbiota plays a role in the variability of BCA efficiency (Pichia anomala, strain K) and postharvest disease development due to Botrytis cinerea on apples. A diverse set of 18 epiphytic microbial communities were harvested from apple carposphere and bio-banked. The analysis of their bacterial and fungal taxonomic composition and carbon metabolic footprint confirmed that contrasted microbiotas were harvested. Their impact on B. cinerea disease development was evaluated through a standardized in vivo bioassay. The reduction of B. cinerea rot development ranged from 20% to 80% when the microbiotas were applied alone. In addition, three microbiotas enhanced the biological control efficiency of strain K (up to +100%, whatever the tested microbiota concentrations) while others limited its action (down to -27%). A co-clustering analysis of biocontrol efficacy with carbon profiles or taxonomic composition was carried out. It identified promising molecules whose high metabolization was associated with high biocontrol by the microbiota and taxa with higher abundance in microbiota limiting B. cinerea rot development. Putative beneficial taxa were isolated from the most efficient microbiota. In vivo bioassays confirmed the efficacy of two molecules and two strains belonging to species never mentioned for their biocontrol properties against plant disease. This study, the first of its kind, demonstrates that natural epiphytic microbiota significantly influences postharvest disease development in apples and the variability in biocontrol efficacy. Our approach identified promising molecules and taxa that enhance biocontrol, offering new insights for sustainable postharvest pathogen management.

The effects of caffeic acid and epicatechin treatment on gray mold resistance and antioxidant metabolism in apples

Abstract:Gray mold, caused by Botrytis cinerea, is a major disease affecting postharvest apples, where it significantly reduces their commercial value. In this study, we conducted in vitro tests at a laboratory scale that showed that two monomeric phenols, caffeic acid and epicatechin, can directly inhibit the growth of B. cinerea in a dose-dependent manner. In in vivo experiments, caffeic acid or epicatechin treatment significantly reduced the incidence of 'Fuji' "Fuji" apple gray mold and inhibited lesion development. Relative to the control group, caffeic acid-treated apples inoculated with B. cinerea showed greater accumulation of disease-related proteins, lower H2O2 synthesis, less residual malondialdehyde (MDA), and reduced damage to cell membrane lipids. However, caffeic acid had no significant effect on the accumulation of superoxide anions (O2.−), ascorbic acid (AsA), or glutathione (GSH). In contrast, epicatechin treatment not only induced the accumulation of reactive oxygen species and disease-related proteins in apples, but also activated AsA and GSH biosynthesis. Caffeic acid and epicatechin treatments both inhibited the invasion and expansion of pathogens but did so by regulating different disease-related proteins. Caffeic acid mainly promoted the accumulation of chitinase (CHI), while epicatechin mainly induced the synthesis of β-1,3-glucanase (GLU).

Abstract:Botryosphaeria dothidea causes apple ring rot, which is among the most prevalent postharvest diseases of apples and causes significant economic loss during storage. In this study, we investigated the biocontrol activity and possible mechanism of Bacillus velezensis strain P2-1 isolated from apple branches against B. dothidea in postharvest apple fruit. The results showed strain P2-1, one of the 80 different endophytic bacterial strains from apple branches, exhibited strong inhibitory effects against B. dothidea growth and resulted in hyphal deformity. B. velezensis P2-1 treatment significantly reduced the ring rot caused by B. dothidea. Additionally, the supernatant of strain P2-1 exhibited antifungal activity against B. dothidea. Re-isolation assay indicated the capability of strain P2-1 to colonize and survive in apple fruit. PCR and qRT-PCR assays revealed that strain P2-1 harbored the gene clusters required for biosynthesis of antifungal lipopeptides and polyketides. Strain P2-1 treatment significantly enhanced the expression levels of pathogenesis-related genes (MdPR1 and MdPR5) but did not significantly affect apple fruit qualities (measured in fruit firmness, titratable acid, ascorbic acid, and soluble sugar). Thus, our results suggest that B. velezensis strain P2-1 is a biocontrol agent against B. dothidea-induced apple postharvest decay. It acts partially by inhibiting mycelial growth of B. dothidea, secreting antifungal substances, and inducing apple defense responses.
Transcriptome and Biochemical Analysis Jointly Reveal the Effects of Bacillus cereus AR156 on Postharvest Strawberry Gray Mold and Fruit Quality

Abstract:Postharvest strawberry is susceptible to gray mold disease caused by Botrytis cinerea , which seriously damage the storage capacity of fruits. Biological control has been implicated as an effective and safe method to suppress plant disease. The aim of this study is to evaluate the postharvest disease control ability of Bacillus cereus AR156 and explore the response of strawberry fruit to this biocontrol microorganism. Bacillus cereus AR156 treatment significantly suppressed gray mold disease and postponed the strawberry senescence during storage. The bacterium pretreatment remarkably enhanced the reactive oxygen-scavenging and defense-related activities of enzymes. The promotion on the expression of the encoding-genes was confirmed by quantitative real-time PCR (qRT-PCR) that significantly increased the expression of the marker genes of salicylic acid (SA) signaling pathway, such as PR1 , PR2 , and PR5 , instead of that of the jasmonic acid (JA)/ethylene (ET) pathway, which was also shown. Moreover, through transcriptome profiling, about 6,781 differentially expressed genes (DEGS) in strawberry upon AR156 treatment were identified. The gene ontology (GO) classification and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment indicated that AR156 altered the transcription of numerous transcription factors and genes involved in the SA-related plant disease resistance, metabolism, and biosynthesis of benzoxazinoids and flavonoids. This study offered a non-antagonistic Bacillus as a method for postharvest strawberry storage and disease control, and further revealed that the biocontrol effects were arisen from the induction of host responses on the transcription level and subsequent resistance-related substance accumulation.

Inhibitory Abilities of Bacillus Isolates and Their Culture Filtrates Against the Gray Mold Caused by Botrytis Cinerea on Postharvest Fruit

Abstract:Botrytis cinerea, a major phytopathogenic fungus, has been reported to infect more than 200 crop species worldwide, and it causes massive losses in yield. The aim of this study was to evaluate the inhibitory abilities and effects of Bacillus amyloliquefaciens RS-25, Bacillus licheniformis MG-4, Bacillus subtilis Z-14, and Bacillus subtilis Pnf-4 and their culture filtrates and extracts against the gray mold caused by B. cinerea on postharvest tomato, strawberry, and grapefruit. The results revealed that the cells of Z-14, culture filtrate of RS25, and cells of Z-14 showed the strongest biocontrol activity against the gray mold on the strawberry, grape, and tomato fruit, respectively. All the strains produced volatile organic compounds (VOCs), and the VOCs of Pnf-4 displayed the highest inhibition values. Based on headspace solid-phase microextraction in combination with gas chromatography-mass spectrometry, esters accounted for the largest percentage of the VOCs produced by RS-25, MG-4, Z-14, and Pnf-4 (36.80%, 29.58%, 30.78%, and 36.26%, respectively). All the strains showed potent cellulase and protease activities, but no chitinase activity. RS-25, Z-14, and MG-4, but not Pnf-4, grew on chrome azurol S agar, and an orange halo was formed around the colonies. All the strains showed biofilm formation, fruit colonization, and lipopeptide production, which may be the main modes of action of the antagonists against B. cinerea on the fruit. This study provides the basis for developing natural biocontrol agents against the gray mold caused by B. cinerea on postharvest fruit.
Bacillus subtilis L1-21 as a biocontrol agent for postharvest gray mold of tomato caused by Botrytis cinerea

Abstract:Gray mold (Botrytis cinerea) is a serious disease causing considerable losses to harvested tomato during storage and transportation. Extensive and unjudicial use of fungicides have certain deleterious effects such as evolved resistance in pathogen and environmental pollution. The ice cold chain transportation is the most commonly used method, but the results are not satisfactory. Currently, the trend is shifting towards prevention and control measures which ensures environmental protection, therefore, the biological control of postharvest diseases has become a hot spot. In this study, Bacillus subtilis L1-21 was used to control gray mold on tomato fruits, by adopting plate confrontation and fruit acupuncture method. To evaluate the preservative effect of biocontrol agent L1-21 on tomato, the gfp-tagged L1-21 was used to study colonization potential in harvested tomatoes assay. Colonization of L1-21-gfp in postharvest tomatoes was stable at 103 cfu/g by soaking and fluorescence method. Whereas Inhibition rate of B. cinerea was 39.63±4.73% in plate confrontation. Culture suspension of L1-21 at a concentration of 108 cfu/ml displayed best control effect: 86.57%±2.55% against tomato gray mold. In addition, different components exhibited different results such as cell pellet+saline (72.94%±8.49%), supernatant (41.18%±16.22%), sterilized culture (28.56%±6.54%). Further, suspension of B. cinerea and Bacillus L1-21 injected in tested tomatoes led to maximum control effect (81.40%±2.74%) after 4 h. Injection of L1-21 culture into tomato followed by pathogen B. cinerea spore solution resulted in 100%±0% control effect after 48 h. There was no significant difference in fruit quality between treated and untreated tomatoes. Taken together, B. subtilis L1-21 may lay theoretical foundation to be used in prevention of gray mold and other postharvest diseases during storage time.
