Abstract:Flammulina velutipes residues, the by-products of Flammulina velutipes industry, cause environmental pollution. The aim of the current work was to evaluate the extraction efficiency of polysaccharides from Flammulina velutipes residues by an ultrasonic assisted snailase enzymatic extraction (UAEE) method, following the principles of green chemistry, mainly solvent and energy saving. The optimized parameters obtained by Response Surface Methodology (RSM) were determined as follows: ultrasonic power at 150 W, extraction time of 90 min, and extraction temperature of 55 °C. The experimental extraction yield (109.54 ± 5.44 mg/g) was closely matched the predicted yield (110.92 mg/g). Polysaccharides from Flammulina velutipes residues (UE-FVRP) had the molecular weight of 170 kDa and a composition comprising 8 monosaccharides, primarily xylose (Xyl, 24.71%), mannose (Man, 17.29%), arabinose (Ara, 11.48%), and galactose (Gal, 15.63%). FT-IR analysis suggested that UE-FVRP likely contained pyranose with α-glycoside bonds. In vitro antioxidant activity assays revealed that UE-FVRP displayed potent DPPH radical scavenging activity and reducing ability. Additionally, UE-FVRP demonstrated the ability to promote the growth of two Lactobacillus strains, indicating its potential as a novel polysaccharide with high in vitro prebiotic properties. Therefore, these findings proposed a novel, green, and efficient extraction method for extracting polysaccharides from Flammulina velutipes residues, highlighting the potential of utilizing Flammulina velutipes residues as a valuable ingredient in both food and feed applications, contributing to the sustainable reuse of agricultural by-products.

Study on Enzymatic Extraction of Isoflavone from Pueraria (Pueraria lobata) Root Residue

Abstract:Pueraria lobata(Willd.)Ohwi is a kind of raw material to produce starch in China.The root residue of pueraria from(pueraria-starch) manufactory still contains some isoflavone which is a kind of phytoestrogen with important physiological effects.By(pretreating) pueraria root residue with cellulase or xylanase before ethanol extraction,the total yield of isoflavone could be efficiently(increased.) The optimal cellulase dosage was 10 FPIU/g pueraria root residue and the suitable cellulase treatment time was 12 h.Total yield of isoflavone was 1.36 times as that by normal ethanol extraction.For pretreatment of pueraria root residue with xylanase,the(optimal) xylanase dosage was 300 IU/g residue for 6 h.Total yield of isoflavone was 1.52 times as that by regular ethanol extraction.It was proved that cellulase and xylanase could cooperate with each other to enhance extraction efficiency.When pueraria root residue was pretreated with both cellulase(7.5 FPIU/g residue) and xylanase(300 IU/g residue) for 6 h,total yield of isoflavone reached(138) mg/10 g pueraria root residue,which was 1.64 times as that by regular ethanol extraction.
Comprehensive understanding of impacts of steam explosion on facilitated extraction and transformation of flavonoids from Astragali Radix

Abstract:Recalcitrant structure of cell walls restricts the extraction of bioactive components from edible plants. In this study, the impacts of steam explosion (SE) on the release and transformation of flavonoids in Astragali Radix (AR) were evaluated. Results revealed that SE destroyed the compact structure of cell walls. Furthermore, the porous network was reformed due to the degradation of hemicelluloses and water-soluble components. The maximum extraction contents of ethanol-soluble and water-soluble flavonoids of 6.34 and 1.48 mg/g were obtained from the pretreated AR (1.5 MPa, 5 min), which were 5.22 and 2.88 times higher than those obtained from the untreated AR, respectively. SE not only released bound flavonoids from cell walls by cleaving glycoside or ester bonds, but also transformed some flavonoid glycosides into aglycones through deglycosylation. In conclusion, SE can reduce mass transfer hindrance and facilitate flavonoid transformation, thus providing a green and facile processing method for traditional edible plants.
The venous stasis syndrome after deep venous thrombosis or pulmonary embolism: a population-based study.

Abstract:OBJECTIVES To estimate the incidence and determine predictors of venous stasis syndrome and venous ulcers after deep venous thrombosis and pulmonary embolism. PATIENTS AND METHODS This population-based retrospective cohort study reviewed medical records of 1527 patients with incident deep venous thrombosis or pulmonary embolism between 1966 and 1990. We recorded baseline characteristics, event type (deep venous thrombosis with or without pulmonary embolism or pulmonary embolism alone), leg side and site of deep venous thrombosis (proximal with or without distal deep venous thrombosis vs distal deep venous thrombosis alone), and venous stasis syndrome and venous ulcer. RESULTS Two hundred forty-five patients developed venous stasis syndrome. One-year, 5-year, 10-year, and 20-year cumulative incidence rates were 7.3%, 14.3%, 19.7%, and 26.8%, respectively. By 20 years the cumulative incidence of venous ulcers was 3.7%. Patients with deep venous thrombosis with or without pulmonary embolism were 2.4-fold (95% confidence interval, 1.7-fold-3.2-fold) more likely to develop venous stasis syndrome than patients with pulmonary embolism and no diagnosed deep venous thrombosis. In patients aged 40 years or younger with proximal compared with distal-only deep venous thrombosis, venous stasis syndrome was 3.0-fold more likely (95% confidence interval, 1.6-fold-4.7-fold). In patients with unilateral leg deep venous thrombosis, venous stasis syndrome usually developed in the concordant leg (P < .001). There was a 30% (95% confidence interval, 2%-62%) increased risk for venous ulcer per decade of age at the incident venous thromboembolism. CONCLUSIONS The cumulative incidence of venous stasis syndrome continues to increase for 20 years after venous thromboembolism. Pulmonary embolism alone is less likely to cause venous stasis syndrome.

Abstract:Ultrasonic-assisted extraction (UAE) and response surface methodology (RSM) were applied to the extraction of polysaccharides from the base of Flammulina velutipes stipe. Results showed that the optimum extraction parameters were ultrasonic power 680 W, time 19.8 min, the ratio of water to raw material 28 mL/g, and the yield of polysaccharides was 16.20%. After separation by DEAE cellulose-52 and Sephadex G-100 size-exclusion chromatography, three main fractions, FVSP -1, FVSP -2, and FVSP -3, were obtained. The relative molecular weights of which were 21 kDa, 224 kDa, 357 kDa, respectively, and all the three fractions were heteropolysaccharides with a glucan as backbone chain.

Abstract:Tremella fuciformis (TF) is a mushroom with rich nutritional and medicinal value. This study aimed to develop an efficient extraction technique for TF polysaccharide (TFP) to enhance its health benefits. TF was subjected to steam explosion (SE) pretreatment at 0.5, 1.0, and 1.5 MPa for 60 s, followed by polysaccharide extraction. The extraction yield of TFP increased from 15.42 % to 50.16 % at 1.0 MPa. SE disrupted the dense structure of TFP, significantly improving total sugar and uronic acid contents, monosaccharide molar percentages of mannose and glucose, specific surface area, and ζ potential by 0.16, 0.4, 0.01, 0.83, 0.19, and 0.26 times at 0.5 MPa (P < 0.05). With increasing SE pressure, the thermal stability of TFP was enhanced, while its elasticity, viscosity, molecular weight, and particle size were reduced. TFP at 0.5 MPa significantly extended the lifespan of Drosophila melanogaster, with Tmax reaching 74 d for females and 60 d for males at a dosage of 0.015625 %, indicating a 0.32-fold enhancement. TFP enhanced climbing ability and antioxidant stress resistance, increased antioxidant enzyme activities and total antioxidant capacity, and reduced malondialdehyde levels, indicating its anti-aging effects. These findings provide theoretical and technical support for the high-value development and utilization of TFP.

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.

Abstract:Morchella esculenta L. is a rare and the most valuable edible mushrooms. The polysaccharides of M. esculenta L. boast high nutrition and health values. This paper describes microwave extraction of the polysaccharides of M. esculenta L. by ultrasonic microwave. The optimal extraction conditions, optimized by single factor experiments and response surface method, were determined as follows: the ultrasonic power is 250 W, the microwave power is 220 W, and the extraction time is 7 min with a yield of polysaccharides at 4.425%. The crude polysaccharides of M. esculenta L. at 4 mg/mL exhibited hydroxyl radical, 2-Diphenyl-1-picrylhydrazyl radical and 2,2’-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging rates at 76.70%, 60.73%, and 80.87%, respectively. Its Fe2+ chelating rates increased from 45.63% to 82.50%, as the polysaccharide concentration increased from 0.5 to 4.0 mg/mL.

Abstract:Pleurotus eryngii (PE) has been sought after for its various health benefits and high content of phenolic compounds. This study explored the feasibility of steam explosion (SE)-assisted extraction of polysaccharides with high antioxidant capacities from PE. An orthogonal experimental design (OED) was used to optimize the SE-assisted extraction of PE. The influence of the optimized SE-assisted extraction on the physicochemical properties of PE polysaccharides was determined by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), monosaccharide compositional analysis and antioxidant capacity assays. Under optimal SE conditions, SE-assisted extraction increased the polysaccharide yield by 138% compared to extraction without SE-assistance. In addition, SEM demonstrated that SE-assisted extraction markedly altered the spatial structure of Pleurotus eryngii polysaccharides (PEP), and monosaccharide compositional analysis revealed that this pretreatment significantly increased the proportions of some monosaccharides, such as glucose, rhamnose and arabinose, in the isolated PEP. FTIR spectra indicated no change in the major chemical functional groups of PEP. PEP extracted by SE-assisted extraction had significantly increased free radical scavenging and antioxidant capacities. In conclusion, SE-assisted extraction appears to be a novel polysaccharide extraction technology, which markedly increases extraction yields and efficiency and can increase the biological activity of polysaccharide extracts.

Abstract:A biological pretreatment of Eucommia ulmoides leaf biomass was carried out. Above all, the total flavonoids were isolated from Eucommia ulmoides leaves by the treatment of alkaline solution of sodium dodecyl benzene sulfonate combined with ultrasonic microwave. The extraction parameters were optimized by central composite design (CCD) software and were displayed as follows: surfactant concentration of 1.5%, microwave power of 700 W, extraction time of 30 min, and liquid–solid ratio of 50 mL/g. The actual yield of total flavonoids was 1.45%. The results of Sudan III color development showed that the cuticle of Eucommia ulmoides leaves was completely removed after dilute alkali pretreatment. Then, Eucommia ulmoides leaves were fermented by Trichoderma viride to remove the holocellulose and obtain gutta-percha. The content of cellulose and hemicellulose in Eucommia ulmoides leaves obviously decreased after Trichoderma viride fermentation. The optimum parameters were listed as follows: solid–liquid ratio of 0.06 g/mL, four extraction times, extraction time of 89.72 min, and extraction temperature of 85 °C. The actual yield of gutta-percha was 4.38%. The amount of extraction solvent per unit weight of gutta-percha from untreated Eucommia ulmoides leaves was 2.91 mL/mg, while that from leaves treated by Trichoderma viride was only 0.96 mL/mg. The amount of extraction solvent was grossly reduced, which is beneficial in terms of environmental protection. The characterizations of gutta-percha were verified by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), and proton nuclear magnetic resonance (1H NMR). This study laid a certain theoretical and experimental basis for the multi-stage extraction of Eucommia ulmoides leaves and the utilization of Eucommia ulmoides resources.

Abstract:Extraction conditions of mycelium polysaccharides from selenium-rich Ganoderma lucidum were studied by cellulase for increasing of yield. The factors influencing extraction of mycelium polysaccharides such as water,time,enzyme,pH and temperature were studied by orthogonal experiment design. The optimum conditions were obtained, water 50 mL per gram mycelia,time 80 min,enzyme 300 U/mL,pH 4.5 and temperature 55 ℃. The yield of mycelium polysaccharides was 33.6% under the optimum conditions. Mycelium polysaccharides were purified by Q Sepharose Fast Flow ion exehange chromatography and the recovery was up to 82.8%.

Abstract:One hundred six cases of phenylketonuria have been surveyed in 11 institutions in New England and Eastern Canada. Data not previously available (and of some degree of uncertainty as to reliability) are presented as to the ages at which patients with phenylketonuria achieve the ability to sit unsupported, to walk independently, and to say recognizable words. A small number of patients achieve these steps at reasonably near the normal age and the degree of variability is such that early developmental timetables should not be overemphasized in interpreting possible effects of any treatment of the disease. Both eczema and seizures are proportionately more frequent in severely retarded patients with phenylketonuria than in those with lesser degrees of intellectual defect. If present at the time an experimental treatment is instituted, they would provide more objective evidence of possible benefit. Electroencephalograms were abnormal in 79% of patients for whom tracings were available and would provide one means of evaluating plans of treatment even in the absence of seizures. While the majority of these patients have blue eyes and blond hair, there is enough evidence to suggest a tendency to darkening of the hair with age that this also should be used with caution in evaluation of treatment. Frequencies of various neurologic manifestations are presented, including unusual hand posturing, tremor, hyperreflexia, clonus and spasticity. These are all variable enough that a reasonably positive diagnosis [See FIG. 2. in Source Pdf.] by physical examination is not possible, and further, they are not well adapted to evaluation of treatment. The distribution of intelligence quotients in patients with phenylketonuria appears to follow a continuous curve and to include a small number of persons with borderline normal or better intelligence. In assessing the effect of any treatment, it is unwarranted to ascribe to that treatment the attainment of borderline normal intellectual levels that may be reached by small numbers of patients, especially if originally tested in infancy or early childhood. The effect of any therapy can be adequately evaluated only by comparing changes in a number of treated patients with phenylketonuria, with changes in comparable control patients, selected on the basis of intelligence, age, physical accomplishments, and status with respect to hair color, eczema, convulsive seizures and electroencephalographic findings.
Steam-explosion-modified Optimization of Soluble Dietary Fiber Extraction from Apple Pomace Using Response Surface Methodology

Abstract:To make full use of apple pomace, pretreatment with steam explosion (SE) was investigated to determine its effects on the yield and functionality of soluble dietary fiber (SDF). Response surface methodology (RSM) was used to optimize the effects of SE parameters. According to the quadratic polynomial model, the optimum SE parameters were as follows: steam pressure, 0.51 MPa; residence time, 168 s; and sieving mesh size, 60. Under the optimized conditions, the SDF yield from apple pomace after SE was 29.85%, which is 4.76 times the yield of SDF (6.27%) in untreated apple pomace. The SDF prepared with SE also exhibited higher functionality values. Scanning electron micrographs (SEM) indicated that the SDF surface from apple pomace was poriferous, rough and lax after SE treatment; therefore, it can be concluded that the SDF morphology was altered by the steam explosion.
Improved Method to Obtain Essential Oil, Asarinin and Sesamin from Asarum Heterotropoides Var. Mandshuricum Using Microwave-Assisted Steam Distillation Followed by Solvent Extraction and Antifungal Activity of Essential Oil Against Fusarium Spp

Abstract:An efficient microwave-assisted steam distillation followed by a solvent extraction method (MSDE) was established to isolate the essential oil, asarinin and sesamin from the roots and rhizomes of Asarum heterotropoides var. mandshuricum (AHVM). In the MSDE technology applied in this work, moistened roots and rhizomes of AHVM powders are applied to this system. After extracting the essential oil by steam distillation, the powder residue of the roots and rhizomes of AHVM is relatively dry. Hence, this method can save energy consumption, and the loss of asarinin and sesamin can be reduced during the subsequent extraction process and improve essential oil yield. The developed method extracted the essential oil to reach the maximum extraction amount (15.80 +/- 0.79 mL/ kg) after microwave irradiation for 30 min and maintained the yield balance. Through comparison, MSDE was chosen as the best approach, and under the optimum conditions (90 % aqueous ethanol, liquid-solid ratio of 19.0 mL/g and microwave irradiation power of 332 W) obtained by first order kinetic models and response surface analysis with Box-Behnken design, satisfactory yields of asarinin (1.81 +/- 0.72 mg/g) and sesamin (0.74 +/- 0.09 mg/g) were achieved. In accordance with the yields of asarinin and sesamin, our research optimized the process parameters. According to the comprehensive comparison of these parameters, the proposed process described in this study has greater advantages than traditional methods. Minimum inhibitory concentrations (MICS) of the essential oil, which obtain from the roots and rhizomes of AHVM of five kinds of Fusariums spp. (F. avenaceum, F. trichothecioides, F. sporotrioides, F. sambucinum and F. culmorum) were determination to be 1.25 mg/mL, 1.5 mg/mL, 1.5 mg/mL, > 2 mg/mL and > 2 mg/mL, respectively. The results of the antifungal test showed that the essential oil from the roots and rhizomes of AHVM showed remarkable antifungal activities, in which, F. avenaceum was the most sensitive to the essential oil, with the MIC50 values of 0.625 mg/mL.

Abstract:Microwave-assisted extraction (MAE) technique was employed for the extraction of himematsutake (Agaricus blazei Murrill) polysaccharide (PMAE) and optimized by Box-Behnken design. The optimum conditions were extraction time 29.37 min, microwave power 400 W, extraction temperature 74.64 degrees C and ratio of water to material 32.7:1 with an enhanced yield of 12.35%. Furthermore, PMAE had increased antioxidant activity in various oxidative systems in vitro when compared to polysaccharides extracted by conventional methods. The data obtained clearly showed that MAE is a fine way to extract the polysaccharides in A. blazei Murrill. Further, PMAE may be developed into a functional food as well as potential therapeutic agent.
Evaluation of the Extraction of Bioactive Compounds and the Saccharification of Cellulose as a Route for the Valorization of Spent Mushroom Substrate

Abstract:The extraction of bioactive compounds and cellulose saccharification are potential directions for the valorization of spent mushroom substrate (SMS). Therefore, investigating the suitability of different extraction methods for recovering bioactive compounds from SMS and how the extraction affects the enzymatic saccharification is of uppermost relevance. In this work, bioactive compounds were extracted from Pleurotus spp. SMS using four extraction methods. For Soxhlet extraction (SoE), a 40:60 ethanol/water mixture gave the highest extraction efficiency (EE) (69.9-71.1%) among the seven solvent systems assayed. Reflux extraction with 40:60 ethanol/water increased the extraction yield and EE compared to SoE. A shorter reflux time yielded a higher extraction of carbohydrates than SoE, while a longer time was more effective for extracting phenolics. The extracts from 240 min of reflux had comparable antioxidant activity (0.3-0.5 mM GAE) with that achieved for SoE. Ultrasound-assisted extraction (UAE) at 65 °C for 60 min allowed an EE (~82%) higher than that achieved by either reflux for up to 150 min or SoE. Subcritical water extraction (SWE) at 150 °C resulted in the best extraction parameters among all the tested methods. Vanillic acid and chlorogenic acid were the primary phenolic acids identified in the extracts. A good correlation between the concentration of caffeic acid and the antioxidant activity of the extracts was found. Saccharification tests revealed an enhancement of the enzymatic digestibility of SMS cellulose after the extraction of bioactive compounds. The findings of this initial study provide indications on new research directions for maximizing the recovery of bioactive compounds and fermentable sugars from SMS.

Abstract:A 43-year-old woman presented to our department with left breast lump, measuring 10 mm x 10mm x 6mm. On physical examination and mammography, the lump was suspected to be due to mastopathy, but malignancy could not be excluded based on ultrasonography. Excisional biopsy showed that it was an invasive lobular carcinoma. A modified radical mastectomy was performed after obtaining informed consent. She remained disease-free seven years after surgery.

Abstract:BACKGROUND/AIM: Flammulina velutipes (FV), also known as the golden needle mushroom, is an edible and medicinal fungus that contains bioactive substances regulating various physiological functions. While the fruiting bodies of FV are commonly consumed, their stipes are often discarded despite containing polysaccharides. In this study, the biological functions of FV stipes (FV-S) were investigated to reduce waste and pollution while increasing their value.MATERIALS AND METHODS: The antioxidant activity of FV was evaluated using three methods: the DPPH radical-scavenging capacity assay, ferrous ion chelating assay, and reducing power analysis. The anti-cancer potential was assessed through MTT viability and immunoblotting analyses.RESULTS: Results showed that FV-S had higher polysaccharide and total phenolic contents and greater antioxidant abilities, particularly in ethanolic extracts. FV-S also exhibited significant anticancer properties, specifically in hot water extracts with high polysaccharide contents, and suppressed prostate cancer cell viability by inhibiting androgen receptor and PCa-specific antigen mRNA expression while inducing caspase-3/7 activation.CONCLUSION: FV-S is rich in bioactive components, possesses higher antioxidant and anticancer abilities, and has potential as an anticancer agent, which could enhance the value of FV.

Abstract:This work developed a green, safe, efficient, and inexpensive extraction method for ergosterol from Lentinus edodes stem (LES) using natural deep eutectic solvent (NADES). The effects of different parameters such as the type, molar ratio, and water content of NADES and ultrasonic conditions on the extraction yield of ergosterol were investigated by single-factor experiments. The NADES was analyzed by Fourier Transform Infrared Spectroscopy (FT-IR), which proved that NADES was successfully synthesized. Compared with conventional organic solvents, the extraction efficiency of NADES was higher. The LES powder before and after extraction was characterized by a scanning electron microscope (SEM), demonstrating that NADES could release ergosterol better. The method had good linearity, precision and accuracy. In this study, NADES was recycled, and the extraction efficiency of ergosterol by NADES did not decrease significantly after seven cycles. In conclusion, the developed ultrasonic-assisted NADES can be a successful method for the extraction of ergosterol from LES.
