Abstract:Flammulina velutipes residues (FVR) are the waste culture medium derived from the collection of Flammulina velutipes fruiting bodies, with an annual output that remains largely unexplored. The characteristics of digestion and fermentation of Flammulina velutipes residues polysaccharide (FVRP) are still relatively unknown. This study investigated the structure of the gut microbiota through 16 s rDNA gene sequencing and analyzed changes in short-chain fatty acid (SCFA) content via targeted metabolome analysis. The aim was to explore the prebiotic activity of FVRP based on a simulated digestion model combined with an in vitro anaerobic fermentation model. The results demonstrated that FVRP did not exhibit significant changes during in vitro digestion and fermentation but did enhance antioxidant activity. Furthermore, FVRP was found to rapidly reduce the pH value and increase SCFA production in the fermentation broth from lactic acid bacteria and human feces. Notably, FVRP altered the gut microbiota structure, significantly increasing the relative abundance of Firmicutes and Bacteroidota . Thus, FVRP could be considered a promising prebiotic food and feed additive that promotes the generation of short-chain fatty acids by modulating gut microbiota.

Abstract:Abstract Due to the problems of the hydrolysis of wasted activated sludge (WAS), fermentation stage takes a long time and volatile fatty acids (VFAs) output efficiency is low. This study investigated the effect of ultrasound combined with Fenton (US/Fenton) pretreatment on the sludge disintegration and anaerobic fermentation. The sludge was treated by three different pretreatments respectively: ultrasound alone, Fenton alone and US/Fenton. Results show that after three pretreatments, sludge floc structure was disintegrated and oxidized, the sludge particle morphology after US/Fenton pretreatment was changed the most obviously. After US/Fenton, specific surface area was the largest and the particle size reduction was remarkable from 50.24 μm to22.44 μm. The SCOD concentration after US/Fenton was 3461.72 mg COD L-1, which was raised 1.73 times, 2.68 times and13.29 times as against that after ultrasound, Fenton and the untreated. The degree of disintegration (DDCOD) of the US/Fenton pretreatment was 15.4% which was greater than ultrasound pretreatment of 9.97% and Fenton pretreatment of 3.18%. The VFAs concentration after US/Fenton pretreatment reached a maximum value of 4594 mg COD L-1 at the eighth day, it was raised 31.8% and 70.15% as separately compared with the Fenton and ultrasound pretreatments. The pretreatments changed the VFAs composition and proportion. The highest HAC concentration after US/Fenton was 2039 mg COD L-1, which was 1.72 and 1.78 times of that in Fenton and ultrasound, respectively. This study showed that the US/Fenton pretreatment had a certain synergistic promotion effect on the sludge anaerobic fermentation. The ultrasound and Fenton co-work, the ability of the VFAs production is further improved.

Abstract:The controllable ultrasonic modification was hindered due to the uncertainty of the relationship between ultrasonic parameters and polysaccharide quality. In this study, the ultrasonic degradation process was established with kinetics. The physicochemical properties and prebiotic activity of ultrasonic degraded Flammulina velutipes polysaccharides (U-FVPs) were investigated. The results showed that the ultrasonic degradation kinetic models were fitted to 1/Mt-1/M0 = kt. When the ultrasonic intensity increased from 531 to 3185 W/cm2, the degradation proceeded faster. The decrease of polysaccharide concentration contributed to the degradation of FVP, and the fastest degradation rate was at 60 °C. Ultrasound changed the solution conformation of FVP, and partially destroyed the stability of the triple helix structure of FVP. Additionally, the viscosity and gel strength of FVP decreased, but its thermal stability was improved by ultrasound. Higher ultrasonic intensity led to larger variations in physicochemical properties. Compared with FVP, U-FVPs could be more easily utilized by gut microbiota. U-FVPs displayed better prebiotic activity by promoting the growth of Bifidobacterium and Brautella and inhibiting the growth of harmful bacteria. Ultrasound could be effectively applied to the degradation of FVP to improve its physicochemical properties and bioactivities.

Abstract:Flammulina velutipes residues, the by-products of Flammulina velutipes industry, cause environmental pollution. The aim of the current work was to evaluate the extraction efficiency of polysaccharides from Flammulina velutipes residues by an ultrasonic assisted snailase enzymatic extraction (UAEE) method, following the principles of green chemistry, mainly solvent and energy saving. The optimized parameters obtained by Response Surface Methodology (RSM) were determined as follows: ultrasonic power at 150 W, extraction time of 90 min, and extraction temperature of 55 °C. The experimental extraction yield (109.54 ± 5.44 mg/g) was closely matched the predicted yield (110.92 mg/g). Polysaccharides from Flammulina velutipes residues (UE-FVRP) had the molecular weight of 170 kDa and a composition comprising 8 monosaccharides, primarily xylose (Xyl, 24.71%), mannose (Man, 17.29%), arabinose (Ara, 11.48%), and galactose (Gal, 15.63%). FT-IR analysis suggested that UE-FVRP likely contained pyranose with α-glycoside bonds. In vitro antioxidant activity assays revealed that UE-FVRP displayed potent DPPH radical scavenging activity and reducing ability. Additionally, UE-FVRP demonstrated the ability to promote the growth of two Lactobacillus strains, indicating its potential as a novel polysaccharide with high in vitro prebiotic properties. Therefore, these findings proposed a novel, green, and efficient extraction method for extracting polysaccharides from Flammulina velutipes residues, highlighting the potential of utilizing Flammulina velutipes residues as a valuable ingredient in both food and feed applications, contributing to the sustainable reuse of agricultural by-products.

Abstract:Different methods were used to degrade Tremella fuciformis polysaccharides (TFP) and prepare low molecular weight polysaccharides of Tremella fuciformis (TFLP) to improve their bioavailability. It was found that the TFLP prepared by ultrasonic-assisted H 2 O 2 -Vc method showed the highest level of antioxidant activity and stress resistance in C. elegans . The structural characteristics, in vivo antioxidant and stress resistance of TFLP-1 were evaluated after isolation and purification of TFLP, it was found that TFLP-1 was an acid polysaccharide with a molecular weight of 75770 Da, which mainly composed of mannose. Meanwhile, it could regulate the antioxidant activity and stress resistance in C. elegans by upregulating the transcription of fat-5 , fat-7 , acs-2 , glp-1 , hsf-1 , hsp-1 , mtl-1 , nhr-49 , skn-1 and sod-3 mRNA. The improvement effects were closely related to the significant regulation of galactose metabolism, alpha linolenic acid metabolism, and pantothenate and CoA biosynthesis metabolic pathways. These results provided insights into the high value application of Tremella fuciformis in the food industry and the development of antioxidant related functional foods.

Abstract:Sugarcane bagasse (SCB), a widely available lignocellulosic by-product, could be potentially used in macrofungal bioconversion for high-value bioactive compounds. For macrofungus with poor lignin degradation capacity, its recalcitrant lignin structure blocked the release of fermentable sugars for cell growth. Herein, the main objective was to construct an effective delignification pretreatment strategy for SCB biotransformation and synthesis of bioactive substances by Agaricus sinodeliciosus var. Chaidam. Different pretreatments including chemical (alkali and Fenton oxidation) and ultrasound methods were used to intensify the delignification process. Physico-chemical characteristics of native and pretreated SCB were examined via XRD, XPS, FT-IR and C-13 ssNMR to characterize crystallinity index, surface lignin and chemical structure. Lignocellulose-degrading enzymes were also used to assess the ability of the macrofungus to degrade SCB. By comparing morphological structure, chemical composition and fermentation characteristics, a suitable multiple sequential pretreatment method was finally identified. Our findings revealed that Fenton-ultrasound-alkali pre-treatment has an optimum delignification effect on SCB with never-uncovered excellent ability to detoxify harmful substances produced by Fenton oxidation. Under this pretreatment condition, the cellulose content was increased by 106.45%, whereas hemicelluloses and lignin were decreased by 48.34% and 74.15%, respectively. The macrofungal growth, intracellular polysaccharides, total phenols and terpenoids formation were 2.4, 1.46, 1.36 and 1.66 times higher than those of the untreated, respectively. Fermentation kinetic results showed outstanding fit and prediction of macrofungal growth and enzymatic activity secretion on above pretreated SCB. This study shed light on developing a novel pretreatment strategy for low-cost SCB lignocellulose transformation into high-value products.

Abstract:The high polysaccharide content of Lycii fructus agri-food waste should be reclaimed for value liberation from both environmental and economic perspectives. In this study, waste from L. fructus pigment products was valorized on a bench scale by upcycling into active polysaccharide-rich extracts. The methodological feasibility of polysaccharide recovery from L. fructus waste was evaluated using sequential extraction techniques. Three fractions LFP-30, LFP-100, and LFP-121, were obtained under stepwise increases in temperature and pressure. Highly heterogeneous xyloglucan (XG)-pectin macromolecules composed of linear homogalacturonan (HG) and alternating intra-RG-I-linkers, with topological neutral branches and XG participation, were predominant among the L. fructus polysaccharides (LFPs). Antioxidant activities in LFPs were unaffected by waste resources and severe extraction methodology conditions. Additionally, the direct investment potential of polysaccharide recovery was evaluated for full-scale implementation. This study demonstrated the necessity and feasibility of extracting bioactive polysaccharides with potential applications from L. fructus waste, and provided a sustainable strategy for transforming L. fructus waste disposal problems into value-added products using cost-effective methodologies.

Abstract:The free-radical degradation and antioxidant activity of polysaccharide from Tremella fuciformis was investigated. In present study, the combination of Fe(2+), ascorbic acid and H2O2 was used as degradation regents in order to obtain the lower molecular weight product. The result ascertained oxidative-reduce degradation did not change the main structure of polysaccharides in the test conditions. Five degraded polysaccharides were selected to evaluate their antioxidant activities in vitro. It was found that the degraded sample with lower molecular weight possessed the higher antioxidant activities. It was possible that free-radical degradation is an effective way for enhancing antioxidant activity to decrease molecular weight of polysaccharides.

Abstract:Fritillaria ussuriensis Maxim. (F.M.) has been widely used in both food and medication for more than 2000 years. In order to achieve its comprehensive utilization and investigate the structural characterization and biology activity, response surface methodology (RSM) was used to optimize the ultrasound-assisted extraction conditions of F.M. polysaccharides. The optimal extraction conditions were ultrasonic power of 174.2 W, ratio of liquid to material of 40.7 mL/g and ultrasonic time of 82.0 min. In addition, a neutral polysaccharide F-1 was obtained, and its structure characterization, antioxidant and immunological activity were evaluated. The structural properties of the polysaccharide were characterized by UV, IR, GC-MS, NMR and AFM. Monosaccharide composition of F-1 (MW 18.11 kDa) was rhamnose, arabinose, glucosamine hydrochloride, galactose, and glucose which under the ratio of 0.9: 3.8: 0.2: 2.9: 92.2. The fractions of F-1 were mainly linked by → 6)-α-D-Glcp-(1 → with branch chain α-D-Glcp-(1 → 4)-α-D-Glcp-(1 → and 4,6)-α-D-Glcp-(1 → residues. Moreover, F-1 has a significant scavenging activity, which can clear hydroxyl radicals, superoxide anion, DPPH and ABTS. In addition, the immunological activity showed that F-1 had an effect on macrophage phagocytic activity. And it can increase the release of inflammatory factors including TNF-α, IL-1β and IL-6. F-1 is a novel polysaccharide with significant activity in antioxidant and immunological activity, which has great potential for antioxidant and immunizer in food, pharmaceutical and cosmetic industries. The study can provide a methodological basis for polysaccharide research and theoretical basis for the industrialized production and practical application.

Abstract:Flammulina velutipes, a widely cultivated species of edible fungus, exhibits diverse functional activities attributed to its polysaccharides. In this study, we employed an in vitro model to investigate the impact of F. velutipes polysaccharides (FVP) fermentation on gut microbiota, with a particular focus on Bacteroides. FVP fermentation resulted in the proliferation of microbiota associated with short-chain fatty acid (SCFA) metabolism and suppression of Escherichia-Shigella. Bacteroides emerged as potential primary degraders of FVP, with species-level analysis identifying the preference of B. thetaiotaomicron and B. intestinalis in FVP degradation. Metabolomics analysis revealed significant increases in hypoxanthine and 7-methyladenine contents, with histidine metabolism emerging as the most enriched pathway. B. nordii and B. xylanisolvens exhibited the most influence on amino acid and SCFA metabolism. Understanding the mechanisms by which gut microbiota metabolize FVP can provide valuable insights into the potential of FVP to promote intestinal health and disease prevention.

Abstract:This investigation involved the extraction of a novel polysaccharide from the spore fermentation broth of Tremella fuciformis using a method that combined ultrasonic and enzymatic assistance with ethanol precipitation. It was then compared with enzymatic and ultrasonic extraction methods. The objective of this research is to offer a reference point for expanding the application of ultrasonic-assisted enzymatic extraction technology in T. fuciformis polysaccharides (TFPs). Based on single-factor experiments, Box-Behnken was used to optimize the extraction conditions of TFPs by ultrasonic-enzymatic-assisted ethanol precipitation extraction. The results revealed an optimal combination of enzymes, with a cellulase-to-papain ratio of 2:1, an enzyme addition of 4000U/100 mL, an enzymolysis temperature of 49 °C, ultrasonicpower at 3 W/mL and an ultrasonictime of 20 min. The extraction rate of TFPs and α- amylase inhibition rates were 23.94 % and 61.44 %, respectively. Comparing the physicochemical properties, structural characterization and in vitro activity of TFPs extracted through different methods, the results showed that ultrasonic treatment significantly influences the apparent morphology of polysaccharide and could enhance its in vitro biological activity. However, different extraction techniques exhibit insubstantial impact on the chemical composition, glycosidic bonds or glycosidic ring configurations within the polysaccharides. Among them, ultrasonic-enzymatic-assisted ethanol precipitation extraction of polysaccharide has the highest extraction rate and the lowest viscosity. It has significant effects on ABTS+ scavenging activity, α- amylase inhibition rate and glucose dialysis retardation index, polysaccharide treated with ultrasonic-enzymatic showed the best performance. These findings suggest that ultrasonic-enzymatic-assisted ethanol precipitation extraction can enhance the activities of TFPs, thereby providing a valuable insight for their future development and application.

Abstract:The most common type of fermentation that can change the physicochemical properties and biological activity of polysaccharide is caused by lactic acid bacteria. In this paper, the structures of Lactobacillus gasseri JM1 degraded and undegraded Hericium erinaceus polysaccharides (FHEP and HEP) were characterized, and the biological activities of the polysaccharides were determined. The result showed that polysaccharide molecular weight and monosaccharide composition changes due to fermentation degradation. FHEP had a molecular weight of 3.5 × 10 4 Da, which was lesser than the molecular weight of HEP (7.5 × 10 7 Da). HEP was composed of nine types of monosaccharides, while FHEP contained six types of monosaccharides. The results of methylation and NMR analysis results showed that after the structural link of HEP was degraded, it became FHEP, which has a branched structure. In vitro antioxidant activities and three enzyme-inhibitory activities found that FHEP had significantly higher activity than HEP ( P < 0.05), indicating that the degraded polysaccharides have better biological activity.
Structural characterization and human gut microbiota fermentation in vitro of a polysaccharide from Fucus vesiculosus

Abstract:In this study, an acidic polysaccharide (FVP-7 A) was isolated from Fucus vesiculosus by DEAE-Sepharose™ fast flow. The chemical composition, glycosidic bonds and in vitro fecal fermentation characteristics of FVP-7 A were studied. Results shown that FVP-7 A was a homogenous polysaccharide with average molecular weight of 30.94 kDa. Combined with FT-IR, monosaccharide composition, methylation and NMR analysis, the glycosidic bonds of FVP-7 A mainly composed of →4)-β-D-Manp-(1→, →3)-α-L-Fucp-(1→, α-D-Manp-(1→, →3)-β-D-Manp-(1 → and →4,6)-α-D-Manp-(1→. The zeta potential and atomic force microscopy images indicated that FVP-7 A could exist stably as a single chain-like structure in dilute solution. After gut fermentation, FVP-7 A was utilized and promoted multiple short-chain fatty acids production, especially acetic acid, butyric acid and valeric acid. For prebiotics, FVP-7 A significantly increased the relative abundance of short-chain fatty acids producing bacteria such as Bacteroides, Lachnospira, Faecalibacterium, Ruminococcus, Oscillospira and Dialister, and inhiited the growth of the harmful bacteria Shigella. These results indicated that FVP-7 A could be used as a potential dietary supplement to improve intestinal health.

Abstract:Flammulina velutipes polysaccharides (FVP) exhibit many biological activities, but the effects on gut microflora and metabolism were still unclear. Here, we explored the composition of FVP, their influence on human gut microflora composition and metabolites. FVP were used to vitro fermentation through human fecal inoculums. In addition, 16S rRNA sequencing were used to assess the effects of FVP on the gut microbiota. The metabolic profiles were investigated using untargeted metabolomics approaches in the LC-MS platform. The results showed that FVP was mainly consisted of glucose, mannose, xylose, fucose and galactose. FVP is shown to increase the relative abundances of Bifidobacteriaceae , as well as Bacteroidaceae and remarkably decrease the numbers of genera Lachnospiraceae coupled with Enterococcaceae . The differential metabolites were identified and mainly involved the metabolism of glycerophospholipid, linoleic acid and synthesis of unsaturated fatty acids. FVP may exhibit biological activity function by regulating gut microflora composition and metabolites.

Abstract:The present study aimed to investigate the impact of Flammulina velutipes polysaccharide (FVSP) on the rheological properties and structural alterations of myofibrillar protein (MP) and oxidized MP (OMP), utilizing techniques such as rhehometer, fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). In the unoxidized system, the addition of 5.00% FVSP significantly improved (p < 0.05) the storage and loss moduli of the composite gel and promoted the α-helix to β-sheet transformation. These effects enhanced the protein's gel strength and water-holding capacity (WHC). In the oxidation system, 5.00% FVSP had significant effects (p < 0.05) on repair and improvement of the oxidized MP. These effects inhibited the cross-linking aggregation and degradation of the protein. In addition, the addition of FVSP significantly improved the gel properties of MPs after oxidation (p < 0.05), hindered fracture of the protein gel network structure. In summary, polysaccharides have a substantial effect on the functional characteristics of MP, and FVSP could potentially be applied in meat products.

Abstract:In an investigation of the mechanisms of the neuroprotective effects of theanine (γ-glutamylethylamide) in brain ischemia, inhibition by theanine of the binding of [3H](RS)-α-amino-3-hydroxy-5-methylisoxazole-4-propionic acid (AMPA), [3H]kainate, and [3H](E)-3-(2-phenyl-2-carboxyethenyl)-4,6-dichloro-1-H-indole-2-carboxylic acid (MDL 105,519) to glutamate receptors was studied in terms of its possible inhibiting effects on the three receptor subtypes (AMPA, kainate, and NMDA glycine), with rat cortical neurons. Theanine bound the three receptors, but its IC50 of theanine was 80- to 30,000-fold less than that of L-glutamic acid.
Effects of Flammulina velutipes polysaccharides on quality improvement of fermented milk and antihyperlipidemic on streptozotocin-induced mice

Abstract:Flammulina velutipes polysaccharide (FVP) has become a focus of attention due to its extensive physiological activity, and the addition of polysaccharides to probiotic fermented milk is a hot research topic in functional dairy products in recent years. In the present work, the physicochemical property and bioactivities of fermented milk contained with FVP isolated from fruiting body were evaluated. Our results indicated that fermented milk contained with FVP had better qualities and sensory scores. Moreover, in vivo data manifested that fermented milk contained with FVP could ameliorate hyperglycemia against diabetes by reducing blood glucose and insulin levels, and regulating dyslipidemia and the disorder of glycometabolism via the PI3K/Akt signaling pathway. Taken together, the study demonstrated that fermented milk contained with F. velutipes polysaccharides might be bioactive compounds for diabetes prevention, providing the basic theory for developing novel fermented drinks.

Abstract:In this study, two degraded polysaccharides were obtained by H2O2-Vc-ultrasonic and H2O2-Fe2+-ultrasonic treatment from Codium cylindricum. The basic structure of polysaccharides was characterized and the relationship between structure and antioxidant activity was studied. FTIR spectrum indicated that the degraded polysaccharides had similar functional groups (OH, CH, CO group) with ordinary polysaccharides. LC-MS analysis showed that the degraded polysaccharides were composed of the same monosaccharide units (mannose, galactose, arabinose, glucose, ribose) with Codium cylindricum polysaccharides, but the molar ratio was different. Meanwhile, the molecular weight and morphological feature of polysaccharides had been changed after degradation. Additionally, the antioxidant activity assay revealed that two degraded polysaccharides with lower molecular weight possessed better antioxidant property than ordinary polysaccharides. These results suggested that the basic structure of polysaccharides had not been damaged by two degradation methods, while the antioxidant activity was significantly enhanced.

Abstract:The degradation effect of ultrasound (US)/hydrogen peroxide (H2O2) on Tremella fuciformis polysaccharide (TFP) were studied. The main process parameters of degradation were evaluated and the structural changes and antioxidant activities of TFP before and after US/H2O2 were analyzed. The results showed that the degradation effect of US/H2O2 was significantly higher than that of US or H2O2 alone, and the degradation of TFP was dependent on the duration of its exposure to US, the ultrasonic amplitude, and the H2O2 and TFP concentrations. US/H2O2 reduced the molecular weight (from 8.14 × 105 Da to 1.27 × 104 Da) and particle size (from 710 nm to 182 nm) of the TFP within 50 min and narrowed its molecular weight and particle size distribution. High performance liquid chromatography, Fourier-transform infrared spectroscopy, Carbon-13 nuclear magnetic resonance, scanning electron microscopy, atomic force microscopy, and Congo red results indicated that the treatment could break down the polysaccharide chains, hinder the aggregation, and improve the conformation flexibility of the TFP molecules without changing the primary structure and monosaccharide composition of TFP. Additionally, the degraded TFPs with low molecular weight exhibited a higher antioxidant activity than the original TFP. These findings suggest that the US/H2O2 treatment is a simple and effective method to prepare a TFP of low molecular weight and high bioactivity.
