Abstract:The aim of this experiment was to investigate the effects of the partial substitution of fish meal with soybean products and chicken meal on the growth performance, antioxidant capacity and intestinal microbiota of Penaeus monodon. A total of 450 healthy, consistent shrimp were randomly divided into five groups, with three replicates per group and 30 shrimp per replicate. The proportion of fish meal substituted with soybean products and chicken meal in the five feed groups was 0% (FM), 40% (40SC), 60% (60SC), 80% (80SC) and 100% (100SC). The experiment lasted for 8 weeks. The results showed that, compared to the FM group, the 40SC and 60SC groups had a decrease in WG and SR, but there was no significant difference (p > 0.05). In contrast, compared to the FM group, the FCR in the 100SC group was significantly increased (p < 0.05), while there was no significant difference among the FM and 40SC, 60SC and 80SC groups (p > 0.05). Compared to the FM group, the ACP in the 80SC and 100SC groups significantly increased (p < 0.05), while the 40SC and 60SC groups had no significant difference (p > 0.05). The AKP in the 100SC group was significantly higher than that in the FM group (p < 0.05), while there were no significant differences among the other four groups (p > 0.05). There were no significant differences in T-AOC and T-SOD among all the treatment groups (p > 0.05). The next-generation sequencing of the intestinal microbiota showed that Proteobacteria was the most abundant phylum in the five groups, accounting for 37.67%, 66%, 40%, 40% and 43.33%, respectively. Compared to the FM group, the Fusobacteriota in the other four groups decreased significantly (p < 0.05). The functional prediction of FAPROTAX indicated that no functional components were observed which are harmful to the body. Considering the effects on growth performance, antioxidant capacity and intestinal microbiota, it is feasible to use soybean products and chicken meal to replace 60% of fish meal in the feed of P. monodon.

Abstract:Duplicate groups of rainbow trout (mean initial weight 16.7 ± 0.1 g) were fed six isonitrogenous (43.7 % crude protein) diets for 12 weeks. A fish-meal-based diet was designated the control. In the other five diets, 75 or 100 % of the fish meal was replaced with either poultry by-product meal (PBM), hydrolyzed feather meal (FEM), and spray-dried blood meal (BM) or with defatted soybean meal and corn gluten meal. Trout fed the diets in which 75 and 100 % of the fish meal was replaced with rendered animal protein showed comparable growth performance to trout fed the control diet except in terms of the protein efficiency ratio and feed conversion ratio. The feed intake of the trout fed a combination of fish meal and rendered animal protein (either with or without plant protein) was significantly higher than the feed intake of trout on the fish-meal-based diet. Apparent crude protein digestibility coefficients were significantly higher in trout fed the diets in which 75 % of the fish meal was replaced with plant protein sources than in trout fed the diets in which 75 % of the fish meal was replaced with rendered animal protein sources (P < 0.05). These results suggest that a combination of PBM, FEM, and BM is a good substitute for most of the fish meal in practical feed for rainbow trout.

Abstract:Reducing dependency on dietary fish meal (FM) and fish oil (FO) is extremely important for the sustainable development of the aquaculture industry. However, the metabolic consequences and mechanisms underlying the replacement of dietary FM and FO by terrestrial proteins (TPs) and lipids remain unclear. To reveal the effects of replacing dietary FM and/or FO on the metabolic changes, the integrated analysis of metabolomics and transcriptomics were employed to evaluate the changes in metabolites and genes of rainbow trout ( Oncorhynchus mykiss ) feeding different experimental diets. Four diets were formulated for the 84-day duration of the experiment: control group (FMFO), FM and vegetable oil (FMVO), terrestrial protein and FO (TPFO), and terrestrial protein and vegetable oil (TPVO). Integrated metabolomic and transcriptomic analyses revealed the significant difference in the metabolic pathways of O. mykiss among the three replacement schemes, i.e., single replacement of dietary FM by TP, single replacement of dietary FO by VO, and combined replacement of FM by TP and FO by VO. The combined replacement of FM and FO by TP and VO, respectively, disturbed immune function, energy metabolism, cellular protein biosynthesis capacity, and lipid metabolism of O. mykiss . The reduction of antioxidant capacity was only observed in individuals feeding diets with replacement of FM by TP. Furthermore, as soon as the dietary FM and/or FO were reduced, cellular protein biosynthesis ability was suppressed and accompanied by higher energy consumption in response to fluctuations of dietary quality, resulting in reduced growth performance. Interestingly, adenylosuccinate and adenosine monophosphate involved in purine metabolism were induced by both individual and combined replacement of FM and FO by TPs and lipids, respectively. It suggested that these two metabolites might be potential biomarkers for O. mykiss fed diets with reduction of FM and/or FO. This study constitutes a new understanding of the molecular and metabolic mechanisms of O. mykiss in response to the replacement of dietary FM and/or FO by TP and/or VO, respectively, and built a theoretical basis for further improvement of aquafeed formulation and sustainable development of aquaculture.

Abstract:This study investigated the dietary effects of lipid and protein levels on growth performance, feed utilization, body composition, lipid metabolism, and antioxidant capacity of triploid rainbow trout, Oncorhynchus mykiss. A 3 × 2 two-factor design was conducted with three crude lipid levels of 4%, 9%, and 14% (L4, L9, and L14) and two crude protein levels of 44%, 49% (P44, P49). Therefore, a total of six diets were prepared as P44/L4, P44/L9, P44/L14, P49/L4, P49/L9, and P49/L14. Triploid rainbow trout (initial body weight 65.0 ± 0.1 g) were fed one of the six diets for 80 days. The results showed that weight gain (WG), protein retention (PR), and protein efficiency rate (PER) significantly increased with increasing the dietary lipid level at the same crude protein level, while feed conversion ratio (FCR) and hepatosomatic index significantly decreased ( P < 0.05 ). At the same lipid level, there was no difference in WG, FCR, PR, PER between 44% and 49% crude protein group ( P > 0.05 ). The P49/L14 group had the highest WG (374.6%) and lowest FCR (1.25), while P44/L14 group had the highest PER (1.80) and PR (25.06%) with similar WG and FCR to P49/L14 group. The crude lipid contents in whole fish were significantly higher in the L14 group than those in the L4 and L9 groups ( P < 0.05 ). Muscle n-3 PUFAs, n-6 PUFAs, and PUFAs levels were positively correlated with dietary lipid level, while n-6 PUFAs was negatively correlated with dietary protein level. Dietary protein, dietary lipid, and their interaction significantly affected hepatic malondialdehyde (MDA) content, aspartate aminotransferase, lipase (LPS), and fatty acid synthase (FAS) activities ( P < 0.05 ). In both P44 and P49 groups, LPS and FAS activities increased with increasing the dietary lipid level. MDA content significantly decreased in the P44 group and increased in the P49 group with increasing the dietary lipid level ( P < 0.05 ). As dietary protein level increased, serum total cholesterol level increased, while hepatic phosphoenolpyruvate carboxykinase activity decreased. With increasing the dietary lipid level, total superoxide dismutase, catalase, total nitric oxide synthase, and fructose-1,6-bisphosphatase activities showed an increasing trend, while the opposite was true for alanine aminotransferase activity. In conclusion, based on growth performance and feed utilization, dietary protein level of 44% and dietary lipid level of 14% (measured value, 43.71% and 13.62%) were suggested for young triploid rainbow trout.

Abstract:Dietary supplementation of functional additives is an effective strategy to improve the nutritive value of low fish meal diets for fishes. The present study is aimed at investigating the dietary effects of a multienzyme complex, an organic acid-essential oil complex and prebiotic on growth, immune function, and intestinal health of rainbow trout (Oncorhynchus mykiss). A positive control diet (PC) and a negative control diet (NC) were formulated to contain 200 g/kg and 100 g/kg of fishmeal, respectively. Seven other diets were formulated by supplementing a multienzyme complex (MC, 0.125 g/kg), organic acid-essential oil complex (OEC, 0.5 g/kg), and prebiotic (P, 2 g/kg) alone or in combination to the NC diet (MC, OEC, P, MC+OEC, MC+P, OEC+P, MC+OEC+P). All diets were formulated to be isoproteic (434.3 g/kg-439.1 g/kg) and isolipidic (119.2 g/kg-125.9 g/kg) and fed to rainbow trout of 7.66 ± 0.05 g for 56 days. The weight gain, apparent digestibility coefficient of dry matter, protein efficiency ratio, and protein retention efficiency were significantly increased, and feed conversion ratio was significantly decreased by the three additives alone or in combination ( P < 0.05 ) compared to the NC. No significant differences in growth performance and feed utilization were observed among the treatment groups. Compared to the NC group, serum alanine transaminase, aspartate transferase activities, and malondialdehyde content reduced significantly, while serum superoxide dismutase increased in all except for the MC group, and alkaline phosphatase and lysozyme activity increased in all except for the MC and MC+P groups ( P < 0.05 ). The foregut protease activity of the MC+P and OEC+P groups and the amylase activity of the OEC and MC+OEC groups were significantly higher than those of the NC group ( P < 0.05 ). The villus height of P, MC+OEC, and OEC+P groups, the villus width of MC group, and the muscle thickness of MC, MC+OEC, and MC+OEC+P groups were also significantly higher than those of the NC group ( P < 0.05 ). Compared to the PC group, the richness and diversity of intestinal microorganisms in the NC group and all the supplemented groups were significantly reduced ( P < 0.05 ), but no differences among them ( P > 0.05 ). In conclusion, the individual or combined supplementation of multienzyme complex, organic acid-essential oil complex, and prebiotic in a low fish meal diet (100 g/kg) improved the growth performance, nutrient utilization, and immune function of rainbow trout, but no synergistic effects were observed in the combination of the three supplements.

Abstract:Duckweed (Lemna minor) meal was included in the formulation of three experimental feeds (L1, L2, L3) for rainbow trout at 10%, 20%, 28% of the protein source, respectively. Increasing the duckweed inclusion, the other protein sources were adjusted to get isonitrogenous (41%) and isolipidic (20%) diets, as the control diet (LC). 540 fish (mean body weight 124.5 ± 0.7 g) were randomly allocated in 12 tanks divided equally among the four different diets. After 90 days, fish were weighed and the most important productive performances, fillet quality and fatty acid profile were determined. The final body weight in L1 (340.53 g) and L2 (339.42 g) was not different from LC (348.80 g); L3 trout significantly (p < 0.05) exhibited the lowest one (302.16 g). Similar trends were found in final mean length, weight gain, specific growth rate, food conversion rate. Somatic indices were affected by duckweed inclusion. Diets had not significant effects on the proximate composition and fatty acids of the fillet in L1, L2, L3 respect to LC. Based on this study, duckweed meal derived from Lemna minor can be included in the feed for the rainbow trout without negative effects on the growth performances at 20% of the protein substitution.

Abstract:This 10-week feeding experiment examined the effects of supplementing vitamin C (VC) and E (VE) in oxidized oil diets on growth, fatty acid composition, blood physiological indicators, innate immunity, antioxidant capacity, and liver and intestine histology of rainbow trout ( Oncorhynchus mykiss ). Four diets were produced including a fresh fish oil containing diet (FFO diet), an oxidized fish oil containing diet (OFO diet), and OFO diet supplemented with 500 mg kg –1 VC and 400 mg kg –1 VE (OFO+C500+E400 diet) or 1,000 mg kg –1 VC and 800 mg kg –1 VE (OFO+C1000+E800 diet). Four hundred and twenty fish averaging 46.9 ± 0.32 g were stocked into 12 tanks and fed the diets twice a day to visual satiety. The results showed no significant effect of OFO or vitamins supplementation on growth, feed intake and feed utilization ( P > 0.05). The groups that received OFO and OFO+C1000+E800 diets had significantly ( P < 0.05) lower hepatosomatic indices than the other groups. Muscle fatty acid composition showed decreased proportion of 15:0, 16:0, 16:1n-7, 16:2n-4, 18:1n-5, 18:2n-6 (linoleic acid) and total saturated fatty acids in OFO group. Whereas higher percentages of 20:3n-6 and 20:3n-3 were found in OFO group. In addition, the highest ratio of docosahexaenoic acid (DHA, 22:6n-3) to eicosapentaenoic acid (EPA, 20:5n-3) was detected in OFO+C500+E400 group. The highest serum triglyceride concentration was recorded in the OFO group. Serum aspartate aminotransferase activity increased in OFO and OFO+C1000+E800 groups compared to OFO+C500+E400 group. Furthermore, significantly higher alkaline phosphatase activity in blood was found in OFO and OFO+C500+E400 groups. Significantly lower serum lysozyme, antiprotease, superoxide dismutase, and catalase activities were recorded in OFO group compared to FFO and OFO+C500+E400 groups, and an opposite trend was observed for malondialdehyde concentration. Muscle VC and VE concentrations, and liver and intestine histology remained unaffected. To conclude, feeding diet containing oxidized oil with peroxide value of 182 meq kg –1 with/without VC and VE supplementation did not influence growth and tissue VC and VE concentrations of rainbow trout. However, supplementing 500 mg kg –1 VC and 400 mg kg –1 VE reversed the adverse effects of OFO on non-specific immune response and antioxidant capacity.

Abstract:A 90-day study was conducted to investigate the effects of substituting sunflower oil (SFO) for fish oil (FO) on various parameters in Labeo rohita (initial weight 18.21 ± 0.22 g). Five experimental diets with different levels of SFO (up to 7%) substitution for FO (0%, 25%, 50%, 75%, and 100%) were formulated, ensuring equal levels of nitrogen and lipids. The results indicated that even with 100% substitution of SFO with FO, there were no significant differences (P>0.05) were observed in growth performance. The survival rate (SR), hepato-somatic index (HSI), and viscero-somatic index (VSI) as well as whole-body composition were also nonsignificant by SFO substitution. However, the fatty acid profiles in both muscle and liver were influenced (P<0.05) by dietary substitution. Saturated fats (SFA) decreased, while monounsaturated fats (MUFA), and linoleic acid (LA) increased (P<0.05). On the other hand, the contribution of linolenic acid (ALA), docosahexaenoic acid (DHA), and eicosapentaenoic acid (EPA) decreased (P<0.05) as the amount of SFO in the diet increased. Hematology parameters, including red blood cells (RBCs), hemoglobin (Hb), and hematocrit (Hct), were not affected. Globulin (GLO) levels decreased significantly (P 0.05). Total protein (TP) increased (P<0.05) at 100% SFO inclusion in the diet, and albumin (ALB) levels increased (P 0.05), while low-density lipids (LDL) were significantly increased (P 0.05) changes. Superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT) activities in the liver and serum were not significantly (P>0.05) affected, while malondialdehyde (MDA) status was significantly (P<0.05) reduced. In conclusion, the fatty acid profile of the muscle and liver of fish was modified by the diets, and FO can be substituted with SFO up to 100% for L . rohita , which is beneficial for growth and immunity while marinating the lipid contents in fish. Our study revealed that fully replacing fish oil with SFO shows promise in fully replacing FO without compromising the growth and overall health status of the fish.

Abstract:Replacing dietary fish oil with DHA-rich microalgae Schizochytrium sp. and EPA-rich microalgae Nannochloropsis sp. for olive flounder (Paralichthys olivaceus) was examined. Three experimental isonitrogenous and isolipidic diets with lipid source provided by 50% fish oil (F50S50), 50% (M50F25S25) and 100% microalgae raw material (M100) respectively were compared with a soybean oil (S100) diet as control. Triplicate groups of olive flounder juveniles (16.5 +/- 0.91 g) were fed the experimental diets, and a group was fed the control diets for 8 weeks in a recirculation system. Results showed feed efficiency and growth performance were not significantly changed when fish oil (FO) was totally substituted by soybean oil (SO) or microalgae raw material (MRM). The whole-body composition, lipid content of liver and muscle, and lipid composition of plasma were not significantly influenced by the total substitution of FO by MRM. The polyunsaturated fatty acids (PUFA) content of muscle and liver declined in fish fed S100 diet, whereas it was not significantly reduced in fish fed M50F25S25 and M100 diets. The total substitution of FO by MRM not only maintained the levels of arachidonic acid, EPA or DHA but also increased n-3/n-6 ratio. In conclusion, MRM as the sole lipid source is sufficient to obtain good feed efficiency, growth performance and human health value in olive flounder juveniles.

Abstract:Aquafarms should reduce the use of fishmeal and fish oil in nutritional feed. One such accessible and relatively inexpensive food component that could successfully meet the challenge posed by aquaculture is algae. The objective of the present study was to evaluate the algae meal inclusion of Chlorella and Spirulina sp. in a diet for rainbow trout, evaluating its effects on fish growth, histological parameters and fillet quality. Experiments were carried out to replace 50% and 100% of fishmeal with Spirulina sp. and Chlorella vulgaris in feed for rainbow trout (Oncorhynchus mykiss W.) cultured in a recirculation system. At the end of the experimental period, the highest mean live weight was measured in rainbow trout fed a feed containing 50% algal meal in the feed. The absorptive vacuolization of cells was increased the most and lamina propria was average thickened when fed 50% algae diets. Fat droplets in the hepatocytes were larger in the 50% algae meal fed group, and their nuclei were replaced in the peripheral zone of the cells. Substitution of fishmeal with 50% algal meal in fish feed resulted in a 36.44% reduction in the lipid content of rainbow trout fillets compared to control fish.

The aim of this study was to determine the effect of diets including defatted black soldier fly larvae (Hermetia illucens), adult cricket (Gryllodes sigillatus) and superworm (Zophobas morio) on the growth performance, apparent nutrient digestibility, nutrient retention, and fatty acid profiles of juvenile rainbow trout (Oncorhynchus mykiss). Triplicate tanks of fish (100.5 +/- 0.6 g; mean +/- SD) were fed one of four diets, a control diet with 20% fishmeal, and three experimental diets containing either 15% defatted black soldier fly meal, 15% full-fat adult cricket meal or 15% full-fat superworm meal, where each insect meal partially replaced fishmeal and fish oil. After 84 days of feeding, no significant differences were observed between diets for growth performance indicators or body indices. Fish fed the control and superworm diets had a higher protein content and retention in whole-body carcass compared to the cricket and black soldier fly diet groups. No significant effects between diets were found on whole-body carcass regarding fatty acid classes SFA, MUFA, PUFA n-3 and PUFA n-6, although lauric acid and myristic acid were significantly higher in fish fed black soldier fly diet and linoleic acid was higher in fish fed superworm and cricket diets. Retention of fatty acids was higher for most classes in fish fed the black soldier fly diet, yet whole carcass lipid content did not differ significantly between diets. Additionally, apparent digestibility of phosphorous was significantly improved for all insect diets compared to the control. These results indicate that insect meals can partially replace fishmeal and fish oil in diets for rainbow trout without compromising growth performance and fatty acid composition, while improving phosphorous utilization.

Abstract:Aquaculture struggles with sustainability due to the use of fishmeal, and insects are one potential alternative. Insects have low long-chain omega−3 polyunsaturated fatty acid content, and insect-fed fish reflect this in their composition. In total, 500 rainbow trout (Oncorhynchus mykiss, ~46 g) were fed until the fish reached a commercial size (~415 g). Five diets were used: one control based on fishmeal as main source of protein; two with a 50% replacement of fishmeal with yellow mealworm meal (Tenebrio molitor), one with full-fat insect, and another partially defatted; two other diets similar to the one with the full-fat insect, but with the addition of an experimental algal oil rich in omega−3 in two different concentrations (one equivalent to control, the other one to the defatted insect diet). Growth was unaffected, as well as texture and organoleptic profile of the fillets. Lightness, brightness and colour of the fillets were slightly modified by the experimental diets. An increase in omega−3 levels over those of the full-fat insect diet is described. An omega−3 sparing effect was highlighted, causing lipid accumulation in fillets and liver, and a mild increase in oxidative damage. More research is encouraged to evaluate the fatty acid profile of the liver.

Abstract:A 12-week feeding trial was conducted to evaluate the effects of replacing soybean meal with different types of rapeseed meal (RSM; Chinese 95-type (oil press model) rapeseed meal [C95RM], Chinese 200-type rapeseed meal [C200RM], cold pressed rapeseed cake [CPRC], Indian rapeseed meal [IRM] and Canadian rapeseed meal [CRM]) on growth, antioxidant capacity, non-specific immunity and Aeromonas hydrophila infection tolerance in 990 fingering (average weight 12.77 ± 0.01g) rainbow trout (Oncorhynchus mykiss). A basal diet was prepared using fishmeal and soybean meal as the main protein sources, the other ten diets were formulated with five types of RSM at 20% (C95RM20, C200RM20, CPRC20, IRM20, CRM20) or 35% (C95RM35, C200RM35, CPRC35, IRM35, CRM35) inclusion levels to replace iso-nitrogenous soybean meal. Regardless of the RSM source, dietary inclusion of 20% RSM significantly reduced the weight gain rate (WGR) and digestive enzymes activities (except C200RM20) of fish, but increased the blood urea nitrogen (BUN) and hepatic malondialdehyde (MDA) content (except CRM20). Fish fed with CPRC20 and IRM20 exhibited relatively higher plasma cortisol and MDA content, but lower content/activities of triiodothyronine (T3), thyroxine (T4) and glutathione peroxidase (GPx) in plasma, lysozyme (LZM) and complement 3 (C3) in serum, catalase (CAT) in liver, and respiratory burst activity (RBA) of head kidney macrophages. The intestinal and hepatic tissues fed with 20% RSM were damaged to some extent, with the CPRC20 and IRM20 groups being the most severely affected. Regardless of the RSM source, dietary inclusion of 35% RSM significantly decreased WGR and digestive enzymes activities, but significantly increased plasma BUN and MDA content. The fish fed with CPRC35 and IRM35 exhibited relatively higher plasma cortisol, MDA, serum triglyceride, BUN content, but lower content/activities of T3, T4, C3 and LZM in serum, CAT, peroxidase and GPx in plasma, CAT in liver, RBA and phagocytic activity of head kidney macrophage. The hepatic and intestinal tissues damage was the worst in the IRM35 group among the 35% RSM inclusion groups. These results indicate that including ≥ 20% RSM in the diet, regardless of the source, reduced the growth, antioxidant capacity, immunity, and survival to Aeromonas hydrophila infection in rainbow trout.

Abstract:The effects of replacing fishmeal and fish oil with a plant-based diet were studied in juvenile (10g) and ongrowing (250-350g) rainbow trout from first-feeding. Feed-related differences in the intestinal and hepatic transcriptome were examined in juveniles after 7 months of feeding at 7°C. Based on microarray results obtained for juveniles, the expression of selected genes related to lipid, cholesterol and energy metabolisms, was assessed by RT-qPCR in ongrowing trout after 6 additional months of feeding at 17°C. Plasma glucose and cholesterol, lipid content and fatty acid profile of whole body were analyzed at both stages. After 7 months at 7°C, all juveniles reached the same body weight (10g), while at 13 months ongrowing fish fed the totally plant-based diet exhibited lower body weight (234 vs 330-337g). Body lipid content was higher in juveniles fed the totally plant-based diet (13.2 vs 9.4-9.9%), and plasma cholesterol was about 2-times lower in trout fed the plant-based diets at both stages. Fatty acid profile mirrored that of the respective diet, with low proportions of long-chain n-3 polyunsaturated fatty acids in fish fed plant-based diets. Genes involved in protein catabolism, carbohydrate metabolism and trafficking were down-regulated in the intestines of juveniles fed the plant-based diets. This was not true for ongrowing fish. Genes involved in lipid and cholesterol metabolisms were up-regulated in the livers of fish fed plant-based diets for both stages. In this study, feeding trout a totally plant-based diet from first-feeding affect a relatively low proportion of metabolism-related genes. In the longer term, when fish were reared at a higher temperature, only some of these changes were maintained (i.e. up-regulation of lipid/cholesterol metabolism). Although the plant-based diets tested in this study had no major deficiencies, small adjustments in the feed-formula are needed to further optimize growth performance while sparing marine resources.
