Abstract:Penicillium expansum is an important postharvest pathogen of pomaceous fruit and a causal agent of blue mold or soft rot. In this study, we investigated the effect of ambient pH on growth, ultrastructure alteration, and pathogenicity of P. expansum, as well as accumulation of patulin and expression of genes involved in patulin biosynthesis. Under different pH, the fungus was routinely cultured and collected for growth, pathogenicity, patulin production, and gene expression studies using transmission electron microscopy, apple inoculation, HPLC, and RT-qPCR methods. Different ambient pH had significant impact on expression of genes and growth factors involved in patulin biosynthesis. Under same range of pH, gene expression profile, growth factors, and patulin accumulation (in vivo and in vitro) all showed similar changing trends. A well-developed cell was observed in addition to upregulation of genes at pH between pH 5.0 and 7.0, while the opposite was observed when pH was too basic (8.5) or too acid (2.5). Additionally, ambient pH had direct or indirect influence on expression of PecreaA, PelaeA, and PepacC. These findings will help in understanding the effect of ambient pH on growth, pathogenicity, and patulin production and support the development of successful methods for combating P. expansum infection on apple fruits.

Abstract:Penicillium marneffei is an important thermal dimorphic fungus that is endemic to Southeast Asia and China and causes penicilliosis, an AIDS-defining disease. Dimorphic switching is considered an important growth characteristic associated with its pathogenicity. In recent years, the molecular mechanisms underlying both dimorphic switching and monomorphic growth have been studied. However, little is known about the physical and chemical factors that impact either dimorphic switching or monomorphic growth of this organism. Further, the natural history of the disease is unclear. Our experiments focus upon the effects of temperature, pH and salinity on both growth phases of P. marneffei. We compared 11 isolates of P. marneffei and found that all could grow at a wide temperature range (8.0-39.8 degrees C), but growth was dramatically inhibited at 40 degrees C. The morphological switch from hyphae to yeast growth was initiated at 32 degrees C. However, the sensitivity to elevated temperatures during this transition varied among isolates. Both hyphae and yeast growth forms grew much better at acidic (pH 5, 6) and neutral pH than at alkaline conditions. While similar sensitivities were observed at high concentrations of NaCl and CaCl(2), in general, yeast cells displayed a greater sensitivity to both compounds. Our data demonstrate that isolate differences occur in growth patterns. Importantly, the growth requirements defined in our study may shed light on the environmental conditions that favor its survival, a subject that is not completely resolved in the current literature.
Recombinant Expression of Antimicrobial Peptides in Pichia Pastoris: A Strategy to Inhibit the Penicillium Expansum in Pears

Abstract:The alteration of safety-secured yeast is a crucial step before scale applications. Based on previous studies, Pichia pastoris showed great potential and value to improve its biocontrol ability. The original sequences of antimicrobial peptide Ac-AMP2 and MiAMP1 were optimized according to the preference of Pichia pastoris and ligated into pPICZ alpha A plasmid which is emerged as a high-performance vector for transformation and expression. The results of RT-qPCR and Western blotting could imply that pPICZ alpha A/Ac-AMP2 and pPICZ alpha A/MiAMP1 were successfully overexpressed in Pichia pastoris GS115. The peptide concentration of GS115/Ac-AMP2 reached a maximum value of 210 mg L-1 at 60 h while GS115/MiAMP1 was 220 mg L-1 at 96 h. The biocontrol experiment indicated that the recombinant strain GS115/Ac-AMP2 and GS115/MiAMP1 could highly suppress the pathogen Penicillium expansum in vivo, which was respectively 42 % (GS115/Ac-AMP2) and 29.2 % (GS115/MiAMP1) of incidence disease lower than the sterile distilled water treatment. In the case of the experimental results considered, the modified GS115/Ac-AMP2 and GS115/MiAMP1 might be promising biological agent in postharvest applications.
Effects of Environment Factors on Conidia Germination,appressoria Formation and Pathogenicity of the Persimmon Anthracnose Pathogens Colletotrichum Gloeosporioides

Abstract:The conidia germination and appressoria formation of Colletotrichum gloeosporioides, the pathogen of persimmon anthracnose, was tested on concave glass slides in different environment. As the concentration of glucose went up, rates of conidia germinating increased but the percentage of appressoria formation decreased, and the length of germ tubes augmented and the diameter of appressoria was steady; with time prolonging, the percentage of conidia germinating and appressoria formation all climbed. The conidia could germinate and form appressoria over wide pH range from 2. 0 to 9. 0; the optimal pH for conidia germination and appressoria formation was between pH 5. 0 and pH 6. 0. The pathogenicity test under different pH condition illuminated that the disease occurred on twigs at 23℃ within pH 4. 0 - 8. 0, also appeared at 17℃ and pH 6. 0 without spore masses on lesions of twigs surface, however, was absent at 17℃ and pH 5. 0 as well as at 15℃ and pH 5. 0 to pH 6. 0. The optimal temperature for the mycelium growth was about 25℃ , and higher temperature inhibited its growth. The scanning electron microscopy examination showed that the length of germ tube, growing longitudinally along the ridge and transversely cross the ridges and grooves, varied greatly, and the appressoria formation occurred at the bottom of groove or near.
Blue LED Light Treatment Inhibits Virulence and Patulin Biosynthesis in Penicillium Expansum

Abstract:Blue light-emitting diode (LED) light has been considered to influence the growth, quality, and preservation of certain fruit and vegetable. However, the blue LED light impact on apple postharvest blue mold remains elusive. In this study, the influence of blue LED light on Penicillium expansum virulence and patulin secretion was investigated in vitro and in vivo, as well as on the apple quality. The growth of P. expansum was inhibited by blue LED light via changes in mycelium morphology changes, cell membrane integrity, and potential mitochondrial membrane destruction, thereby causing reactive oxygen species accumulation, oxidative stress, and mitochondrial dysfunction. Furthermore, the expression level of LaeA relevant to the pathogenicity of P. expansum, was down-regulated. Specifically, the patulin production of P. expansum was inhibited via expression down-regulation of the patulin biosynthesis-related genes PatA, PatE, and PatN rather than direct degradation by the blue LED light treatment. Additionally, no significant effects on the fruit quality were observed when the light intensity was lower than 120 μmol m-2 s-1. In summary, blue LED light treatment has the potential for a low-cost, safe, and non-chemical approach to control apple postharvest decay.

Abstract:The effect of a strain of marine yeast Rhodosporidium paludigenum on postharvest blue mold and patulin accumulation in apples and pears stored at 23°C was evaluated. The occurrence and severity of apple and pear decay caused by Penicillium expansum were significantly inhibited by R. paludigenum. However, the application of the yeast at a high concentration (10(8) cells per ml) enhanced patulin accumulation after 7 days of storage; the amount of patulin increased 24.2 times and 12.6 times compared to the controls in infected apples and pears, respectively. However, R. paludigenum reduced the patulin concentration in the growth medium by both biological degradation and physical adsorption. Optimal in vitro patulin reduction was observed at 30°C and at pH 6.0. R. paludigenum incubated at 28°C was tolerant to patulin at concentrations up to 100 mg/liter. In conclusion, R. paludigenum was able to control postharvest decay in apples and pears and to remove patulin in vitro effectively. However, because the yeast induced patulin accumulation in fruit, the assessment of mycotoxin content after biological treatments in postharvest decay control is important. R. paludigenum may also be a promising source of gene(s) and enzyme(s) for patulin degradation and may be a tool to decrease patulin contamination in commercial fruit-derived products.
Inhibiting Penicillium Expansum Infection on Pear Fruit by Cryptococcus Laurentii and Cytokinin

Abstract:The effectiveness of the cytokinin N6-benzyladenine (6-BA), alone or in combination with the biocontrol yeast Cryptococcus laurentii, in controlling blue mold on pear fruit was assessed. The application of 6-BA (500–2000μgmL−1) or C. laurentii was effective in reducing Penicillium expansum infection in pear fruit wounds, but its efficacy declined rapidly as the incubation time increased. Integrated application of C. laurentii and 6-BA at 500–2000μgmL−1, especially at the optimal concentration (1000μgmL−1), resulted in a more effective and stable inhibition of the mold rots than that of the 6-BA or C. laurentii alone. Treatments of pears with 6-BA at 1000μgmL−1, alone or with C. laurentii also led to an increase in catalase activity and an inhibition of the activities of both peroxidase and lipoxygenase as well as ethylene production. In addition, 6-BA from 20 to 2000μgmL−1 did not influence the population growth of C. laurentii in pear fruit wounds. These data suggested that a combination of 6-BA and C. laurentii could integrate the dual biological activities from 6-BA and C. laurentii and might be developed into a novel protection strategy for reduction of the blue mold rot of pear fruit.

Abstract:This study was conducted to evaluate the efficacy of the biocontrol yeast Cryptococcus laurentii and calcium chloride (CaCl2) in suppressing the blue and gray mold rots in pear fruit wounds and the possible mechanisms involved. The results from the presented investigation showed that combined treatment of pear fruit wounds with C. laurentii and CaCl2 was a much better approach for inhibition of Penicillium expansum and Botrytis cinerea infections than C. laurentii or CaCl2 alone. CaCl2 neither affected the growth of C. laurentii in vitro or in vivo, nor directly inhibited the mold rots in pear fruit. However, CaCl2 was shown to elicit the fruit resistance to mold rots when the time interval between CaCl2-treatment and pathogen-inoculation was increased up to 24h, being associated with an activation of the peroxidase activity of pear fruit. Therefore, it could be proposed that the mechanism by which CaCl2 reinforced the biocontrol efficacy of C. laurentii was mainly due to its ability to induce the fruit natural resistance.
Genetic identification, pathogenicity and patulin production of Penicillium species from apple blue mold in China

Abstract:Abstract Blue mold is a common postharvest disease that leads to serious rot and patulin contamination in fruits, which is a key factor endangering the storage and quality of apple products. However, the Penicillium species from apple blue mold in China and their pathogenicity and patulin (PAT) production ability have not been well studied. In this study, 62 strains of pathogenic Penicillium species were isolated from blue mold apples collected from different Chinese regions. The ITS, BenA and CaM genes were sequenced for molecular identification. Penicillium strains were identified as three species with 57 P. expansum, 3 P. crustosum, and 2 P. polonicum. Twelve variants of the ITS sequence, 61 variants of BenA and 67 variants of CaM genes were identified between Penicillium species. Fungal morphology was observed on PDA. After 10 d of cultivation, P. expansum cultures were detected with PAT concentrations ranging from 457.98 to 1333.85 mg/kg, and two P. polonicum strains were 6.02 and 6.30 mg/kg. Three P. crustosum strains could not produce PAT on PDA. In apple inoculation, P. expansum strains showed significantly faster infection rates than P. crustosum and P. polonicum strains. All P. expansum strains can produce PAT during infection, and the concentrations in the rot apple tissues were 2.31 to 88.67 mg/kg. The P. crustosum and P. polonicum strains could not produce PAT during infection. This study provided data and information regarding the morphology, virulence and PAT production of Penicillium pathogens that will improve the understanding of apple blue mold and postharvest disease control.

Abstract:Patulin, one of the most common mycotoxins produced primarily by the Penicillium, Aspergillus and Byssochlamys species, is often associated with fruits and fruit-based products. Biodegradation by microbes is an effective method to remove or detoxify mycotoxins. In this study, a bacterial strain with patulin degradation capability was selectively isolated using oxindole, an analogue to patulin, as the sole carbon source, and identified as Pseudomonas poae JSU-Y1 by phylogenetic analysis on the basis of 16S rRNA sequence. This isolated bacterium could inhibit the growth of Penicillium expansum both on plate medium and apple fruit with inhibition ratio of 30.3% and 44.9%, respectively. Up to 87.7% of the initial patulin (2.5 μg/mL) was removed after incubation with Pseudomonas poae JSU-Y1 in liquid medium at 30 °C for 72 h. When challenged with apple juice, 79% of patulin could be degraded by this isolated strain. Additionally, ascladiol was tentatively identified as the patulin degradation intermediate by LC-MS analysis. Taken together, the experiment results indicated that the isolated Pseudomonas poae JSU-Y1 would be a promising bacterial resource to control patulin contamination and toxigenic fungal growth in agricultural products.

Biocontrol Agents Increase the Specific Rate of Patulin Production by Penicillium expansum but Decrease the Disease and Total Patulin Contamination of Apples

Abstract:Synthetic fungicides are commonly employed for the control of postharvest diseases of fruits. However, due to health concerns about the use of these chemicals, alternative control methods including biocontrol based on antagonistic yeasts are gaining in popularity. In this study, we investigated the effects of two biocontrol yeasts, Rhodotorula mucilaginosa strain 3617 and Rhodotorula kratochvilovae strain LS11, on blue mold and patulin (PAT) contamination caused by Penicillium expansum strains PY and FS7 in artificially inoculated Fuji apples stored at 20°C for 9 days. To correlate the development of the P. expansum strains in yeast-treated and untreated apples with PAT production, we quantified their biomass in the infected fruits using a recently published quantitative real-time polymerase chain reaction method based on specific primers for patF, a gene from P. expansum that is involved in PAT biosynthesis. Both yeasts significantly reduced the disease incidence caused by the two strains of P. expansum up to 5-7 days of incubation, and lowered their biomass and the progression of symptoms up to 9 days. Interestingly, both yeasts strains increased the rate of PAT production (expressed as ng patulin/μg fungal DNA) by the two pathogenic strains. Nevertheless, both biocontrol agents reduced the total PAT contamination, especially in the case of P. expansum strain FS7, the higher PAT producer of the two tested P. expansum strains. Comparing between the yeast strains, R. kratochvilovae LS11 was more effective than R. mucilaginosa 3617 for the control of P. expansum.
The Influence of Long-Term Storage on the Epiphytic Microbiome of Postharvest Apples and on Penicillium expansum Occurrence and Patulin Accumulation

Abstract:Patulin is a secondary metabolite primarily synthesized by the fungus Penicillium expansum, which is responsible for blue mold disease on apples. The latter are highly susceptible to fungal infection in the postharvest stages. Apples destined to produce compotes are processed throughout the year, which implies that long periods of storage are required under controlled atmospheres. P. expansum is capable of infecting apples throughout the whole process, and patulin can be detected in the end-product. In the present study, 455 apples (organically and conventionally grown), destined to produce compotes, of the variety "Golden Delicious" were sampled at multiple postharvest steps. The apple samples were analyzed for their patulin content and P. expansum was quantified using real-time PCR. The patulin results showed no significant differences between the two cultivation techniques; however, two critical control points were identified: the long-term storage and the deck storage of apples at ambient temperature before transport. Additionally, alterations in the epiphytic microbiota of both fungi and bacteria throughout various steps were investigated through the application of a metabarcoding approach. The alpha and beta diversity analysis highlighted the effect of long-term storage, causing an increase in the bacterial and fungal diversity on apples, and showed significant differences in the microbial communities during the different postharvest steps. The different network analyses demonstrated intra-species relationships. Multiple pairs of fungal and bacterial competitive relationships were observed. Positive interactions were also observed between P. expansum and multiple fungal and bacterial species. These network analyses provide a basis for further fungal and bacterial interaction analyses for fruit disease biocontrol.

Abstract:Meyerozyma guilliermondii is an antagonistic yeast that efficiently controls the blue mold decay of pears and potentially degrades patulin in vitro conditions. However, the possible patulin controlling efficacy of M. guilliermondii in pears remains unclear. In this study, the patulin controlling effect of M. guilliermondii in pears was investigated. The results showed that M. guilliermondii could effectively control patulin content production by Penicillium expansum in shuijing pear wounds from 3 d to 11 d. However, M. guilliermondii has a poor patulin control effect on fragrant pear and dangshan pear wounds. The patulin degradation ability of M. guilliermondii in shuijing pear wounds increased with the higher concentration of the yeast cells, and 1 × 108 cells/mL showed the best patulin control effect. In shuijing pears, M. guilliermondii could effectively control patulin at 20 °C and 4 °C in wounds as well as the whole fruits. Our results provide a safe and efficient strategy to detoxify patulin using M. guilliermondii, thereby limitting the risks of patulin in shuijing pears.

Abstract:Penicillium expansum is a major postharvest pathogen of apples, causing loss in fruits through tissue damage, as well as in apple products due to contamination with the mycotoxin patulin. During infections, patulin is a cultivar-dependent virulence factor that facilitates apple lesion development. Patulin also has characterized antimicrobial activity and is important for inhibiting other competitive phytopathogens, but the role of this inhibitory activity has not been investigated in the context of the apple microbiome. In our current study, we isolated 68 apple microbiota and characterized their susceptibility to P. expansum extracts. We found Gram-negative bacteria and Basidiomycete yeast to demonstrate largely patulin-specific growth inhibition compared to Gram-positive and Ascomycete isolates. From co-cultures, we identified a Hanseniaspora and Gluconobacter pairing that reduced P. expansum biomass and found that Hanseniaspora uvarum alone is sufficient to reduce apple disease progression in vivo. We investigated possible mechanisms of H. uvarum biocontrol activity and found modest inhibition on apple puree plates, as well as a trend toward lower patulin levels at the wound site. Active biocontrol activity required live yeast, which also were effective in controlling Botrytis cinerea apple infections. Lastly, we explored the breadth of H. uvarum biocontrol activity with over 30 H. uvarum isolates and found consistent inhibition of P. expansum apple disease.
Effect of Cinnamaldehyde and Citral Combination on Transcriptional Profile, Growth, Oxidative Damage and Patulin Biosynthesis of Penicillium expansum

Abstract:Penicillium expansum, as a main postharvest pathogen of fruits, can secrete patulin (PAT), causing fruit decay and health problems. In this study, the antifungal test, SEM (scanning electron microscope) observation, transcriptional profile, PAT biosynthesis, and physiological characters of P. expansum exposed to cinnamaldehyde and citral combination (Cin/Cit) were evaluated. Cin/Cit could inhibit the mycelial growth and spore germination of P. expansum in a dose-dependent manner. Besides, Cin/Cit caused spores and mycelia wrinkled and depressed by SEM observation. Gene expression profiles of P. expansum were conducted by RNA sequencing (RNA-seq) in the presence or absence of Cin/Cit treatment. A total of 1713 differentially expressed genes (DEGs) were obtained, including 793 down-regulated and 920 up-regulated genes. Most of the DEGs participated in the biosynthesis of secondary metabolites, amino acid metabolism, and oxidation-reduction process, etc. Cin/Cit induced the dysfunction of the mitochondrial membrane, causing the potential influence on energy metabolism and reactive oxidative species production. The changes of superoxide dismutase (SOD) and catalase (CAT) activities combing with the increase of hydrogen peroxide content indicated the oxidative stress on P. expansum induced by Cin/Cit, which corresponded well with the transcriptional results. Moreover, both the RNA-seq data and the qRT-PCR showed the remarkable down-regulation of genes included in the PAT biosynthetic pathway under the Cin/Cit treatment. These findings provided more useful information about the antifungal mechanism of Cin/Cit against P. expansum at molecular and gene levels and suggested that Cin/Cit is a potential candidate to control P. expansum.

Abstract:Penicillium digitatum is the most important pathogen causing green mold disease of postharvest citrus.Previous studies indicated that PdPG2 played an important role in pathogenicity,and disruption of PdPG2 resulted in attenuated virulence of P.digitatum.However,the expression profiles of PdPG2 were not well characterized.In this study,we investigated the expression profiles of PdPG2.The results indicated that PdPG2 was up-regulated under acidic conditions.The expression level of PdPG2 was approximately 10 fold at pH 3.0 and 0.36 fold at pH 8.0 compared with that in the control.Pectin could induce the expression of PdPG2 and the expression level was about 3.6 fold of that in the control.These results indicated that acidic condition and pectin could induce PdPG2 expression.The decreased pH of citrus rind during infection was suitable for PdPG2 expression,and pectin might be also helpful during infection.
Reactive Oxygen Species Metabolism Modulation on the Quality of Apple Fruits Inoculated with Penicillium expansum under Different Ambient pHs

Abstract:Apple blue mold is a significant postharvest disease caused by Penicillium expansum. pH modification in colonized tissues leads to the production of organic substances, the modulation of enzymes, and then increases fungal pathogenicity. This study evaluated Penicillium expansum-inoculated apple fruits’ quality responding to pH treatments ranging from 2.5 to 8.5 and analyzed the reactive oxygen species (ROS) metabolism modulation in inoculated apple fruits at the same pH. The results showed that the fruit quality of the firmness, total soluble solids, and titratable acid displayed a quick loss at pHs 5.0 and 7.0, compared with 2.5 and 8.5. Similarly, higher disease incidence was observed at pHs 5.0 and 7.0. Apple fruits infected with P. expansum at pHs 2.5 and 8.5 had less content of O2•−, H2O2, and malondialdehyde (MDA); lower enzymatic activity of NADPH oxidase (NOX); and greater cell membrane integrity than those at pHs 5.0 and 7.0. The analysis of the antioxidant enzymatic activities showed upregulation of superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) at pHs 2.5 and 8.5 compared with those at pHs 5.0 and 7.0. Similar trends were shown in ascorbic acid and glutathione. These results support the hypothesis that inoculated apple fruits at pHs 2.5 and 8.5 improve resistance to P. expansum by modulating ROS metabolism, compared with pHs 5.0 and 7.0.

Abstract:Patulin (PAT), mainly produced by Penicillium expansum, is a potential threat to health. In recent years, PAT removal using antagonistic yeasts has become a hot research topic. Meyerozyma guilliermondii, isolated by our group, produced antagonistic effects against the postharvest diseases of pears and could degrade PAT in vivo or in vitro. However, the molecular responses of M. guilliermondii over PAT exposure and its detoxification enzymes are not apparent. In this study, transcriptomics is used to unveil the molecular responses of M. guilliermondii on PAT exposure and the enzymes involved in PAT degradation. The functional enrichment of differentially expressed genes indicated that the molecular response mainly includes the up-regulated expression of genes related to resistance and drug-resistance, intracellular transport, growth and reproduction, transcription, DNA damage repair, antioxidant stress to avoid cell damage, and PAT detoxification genes such as short-chain dehydrogenase/reductases. This study elucidates the possible molecular responses and PAT detoxification mechanism of M. guilliermondii, which could be helpful to further accelerate the commercial application of antagonistic yeast toward mycotoxin decontamination.

Abstract:Patulin is a secondary metabolite produced primarily by the fungus Penicillium expansum, responsible for the blue mold disease on apples. It is found in apple products including apple cider when apple juice is added after fermentation. In the present study, two hundred and twenty-five cider-apples of the variety “Bedan”, cultivated in Brittany in France, were sampled from the orchard during harvesting until the storage step, right before processing. The patulin analysis on these samples reported a low contamination at the orchard and a significantly higher-level of contamination in the cider-apples starting from the transporting bin. The percentage of positive samples increased from 6% to 47% after 12 h in the harvesting bin before transporting and reached 95% after 24 h of transporting, decreasing then to 69% at the end of the storage. Penicillium expansum was quantified on the surface of apples using real-time PCR and was observed to be mostly consistent between the harvest and post-harvest steps. It was detected on average, on the surface of 85% of all sampled apples with a mean value around 2.35 × 106Penicillium expansum DNA/g of apple. Moreover, the changes in the fungal and bacterial epiphytic microbiota in the different steps were studied using a metabarcoding approach. The alpha and beta diversity analysis revealed the presence of unique and more diverse bacterial and fungal communities on the surface of apples picked from the orchard compared to the rest of the sampling steps. Potential indigenous biological control agents were identified on the surface of sampled apples. Future perspective includes developing actions of prevention and control of the contamination by Penicillium expansum during the harvest and along the various critical post-harvest stages before transformation in a sustainable development concern.
