Abstract:This study was conducted to evaluate the efficacy of the biocontrol yeast Cryptococcus laurentii and calcium chloride (CaCl2) in suppressing the blue and gray mold rots in pear fruit wounds and the possible mechanisms involved. The results from the presented investigation showed that combined treatment of pear fruit wounds with C. laurentii and CaCl2 was a much better approach for inhibition of Penicillium expansum and Botrytis cinerea infections than C. laurentii or CaCl2 alone. CaCl2 neither affected the growth of C. laurentii in vitro or in vivo, nor directly inhibited the mold rots in pear fruit. However, CaCl2 was shown to elicit the fruit resistance to mold rots when the time interval between CaCl2-treatment and pathogen-inoculation was increased up to 24h, being associated with an activation of the peroxidase activity of pear fruit. Therefore, it could be proposed that the mechanism by which CaCl2 reinforced the biocontrol efficacy of C. laurentii was mainly due to its ability to induce the fruit natural resistance.
Perinatal psychiatric disorders: a leading cause of maternal morbidity and mortality.

Abstract:The Confidential Enquiry into Maternal Deaths 1997 to 1999 finds that psychiatric disorder, and suicide in particular, is the leading cause of maternal death. Suicide accounted for 28% of maternal deaths. Women also died from other complications of psychiatric disorder and a significant minority from substance misuse. Some of the findings of the Confidential Enquiry confirm long established knowledge about postpartum psychiatric disorder. The findings highlight the severity and early onset of serious postpartum mental illness and of the risk of recurrence following childbirth faced by women with a previous history of serious mental illness either following childbirth or at other times. These findings led to the recommendation that all women should be asked early in their pregnancy about a previous history of serious psychiatric disorder and that management plans should be in place with regard to the high risk of recurrence following delivery. Other findings of the Enquiry were new and challenged some of the accepted wisdoms of obstetrics and psychiatry. It is likely that the suicide rate following delivery is not significantly different to other times in women's lives and for the first 42 days following delivery may be elevated. This calls into question the so-called 'protective effect of maternity'. The overwhelming majority of the suicides died violently, contrasting with the usual finding that women are more likely to die from an overdose of medication. Compared to other causes of maternal death, the suicides were older and socially advantaged. The Enquiry findings suggest that the risk profile for women at risk of suicide following delivery may be different to that in women at other times and in men. None of the women who died had been admitted at any time to a Mother and Baby Unit and their psychiatric care had been undertaken by General Adult Services. None of the women who died had had a previous episode correctly identified and none had had adequate plans for their proactive care. The conclusion is that there is a need for both Psychiatry and Obstetrics to acknowledge the substantial risk that women with a previous psychiatric history of serious mental illness face following delivery.
Synergistic Inhibitory Effect of 1-Methylcyclopropene (1-MCP) and Chlorine Dioxide (clo2) Treatment on Chlorophyll Degradation of Green Pepper Fruit During Storage

Abstract:Degreening indicates the ripening and senescence process of green pepper fruit, which mainly results from chlorophyll degradation. To date, however, the effect of 1-Methylcyclopropene (1-MCP) alone or in combination with chlorine dioxide (ClO2) on the chlorophyll degradation pathway of green pepper at the molecular level remains scarce. In this study, green peppers were treated with 1 mu L L-1 1-MCP, 30 mu L L-1 ClO2 alone and 1 mu L L-1 1-MCP plus 30 mu L L-1 ClO2, respectively and stored at 20 degrees C for 12 d. The results showed that 1-MCP + ClO2 combination was superior in inhibiting color changes, decreasing the respiration rate, and exhibiting chlorophyll content as compared with 1-MCP or ClO2 alone. Further study on the expression of genes related to chlorophyll degradation pathway revealed that pheophytinase (PPH), pheophorbide a oxygenase (PAO) and red chlorophyll catabolite reductase (RCCR) were suppressed by all treatments. The efficiency of the combined treatment (1-MCP + ClO2) was better than that of 1-MCP or ClO2 alone. While the expression of chlorophyllase (CLH) was not affected by ClO2, but was significant suppressed by 1-MCP and 1-MCP + ClO2. Therefore, our results indicate the different regulatory roles of 1-MCP and ClO2 on the chlorophyll degradation pathway and provide an efficient method to preserve green pepper.

Abstract:Penicillium striatisporum strain P.st10 was showed to be highly antagonistic against mycelial growth of Phytophthora spp.and Sclerotinia sclerotiorum, while not antagonistic to other tested plant pathogens in plate tests.Investigation of antagonistic mechanism under light microscope showed that the dilution of filtrate from liquid fermentation of the fungus inhibited pathogen by leading to hyphal abnormality,inhibiting germination of spores,and suppressing formation and germination of sporangium of P.capsici.P.striatisporum could colonize in pepper rhizosphere.Organic fertilizer could improve the colonization ability.The rate of pepper blight disease reduced notably when soil was treated with the mycelium of the strain and its filtrate in pot test.
Effect of Ambient pH on Growth, Pathogenicity, and Patulin Production of Penicillium expansum

Abstract:Penicillium expansum is an important postharvest pathogen of pomaceous fruit and a causal agent of blue mold or soft rot. In this study, we investigated the effect of ambient pH on growth, ultrastructure alteration, and pathogenicity of P. expansum, as well as accumulation of patulin and expression of genes involved in patulin biosynthesis. Under different pH, the fungus was routinely cultured and collected for growth, pathogenicity, patulin production, and gene expression studies using transmission electron microscopy, apple inoculation, HPLC, and RT-qPCR methods. Different ambient pH had significant impact on expression of genes and growth factors involved in patulin biosynthesis. Under same range of pH, gene expression profile, growth factors, and patulin accumulation (in vivo and in vitro) all showed similar changing trends. A well-developed cell was observed in addition to upregulation of genes at pH between pH 5.0 and 7.0, while the opposite was observed when pH was too basic (8.5) or too acid (2.5). Additionally, ambient pH had direct or indirect influence on expression of PecreaA, PelaeA, and PepacC. These findings will help in understanding the effect of ambient pH on growth, pathogenicity, and patulin production and support the development of successful methods for combating P. expansum infection on apple fruits.

Comparison of the Antifungal Activity of Chlorine Dioxide, Peracetic Acid and Some Chemical Fungicides in Post-harvest Management of Penicillium digitatum and Botrytis cinerea Infecting Sweet Orange and Strawberry Fruits

Abstract:Penicillium digitatum and Botrytis cinerea are some of the most important post-harvest pathogenic fungi infecting vegetables and fruits. This study is aimed to assess the antifungal potency of chlorine dioxide (ClO 2 ) and peracetic acid (PAA) by examining the growth of P. digitatum and B. cinerea on inoculated potato-dextrose-agar medium (PDA), sweet orange and strawberry fruits under in vitro and storehouse conditions. Furthermore, the antifungal efficacy of ClO 2 and PPA was compared with four commercial fungicides namely Thiabendazole, Imazalil, Cyprodinil and Thiophanate-methyl (1000 mg/L). The results indicated that the antifungal activity of ClO 2 and PAA under both in vivo and in vitro conditions were similar to the chemical fungicides. At lower concentration, ClO 2 was more active than PAA. The radial growth of pathogenic fungi in PDA medium was completely inhibited by 100 ppm ClO 2 and 750–900 ppm PAA. Under the storehouse condition, application of 400 ppm ClO 2 and 1050 ppm PAA completely inhibited the test fungi on orange and strawberry fruits. No significant difference was observed in the total sugar content of the fruits following application of the test compounds at these concentrations. This study concludes that ClO 2 and PAA can be used as viable substitutes for chemical fungicides for the management of P. digitatum and B. cinerea in sweet orange and strawberry fruits.

Abstract:Patulin is a secondary metabolite mainly secreted by fungi and is the most common mycotoxin found in apples and apple-based products. For the past few years, numerous studies suggested the wide distribution and toxicity of patulin. In this study, we investigated the toxic effect of patulin on mouse oocytes and its possible mechanisms. The results showed that patulin treatment did not affect meiotic resumption, but inhibited oocyte maturation as indicated by failure of first polar body extrusion. Further mechanistic study showed that patulin treatment disturbed normal spindle assembly, chromosome alignment and morphology. We also found increased oxidative stress by testing the level of ROS and decreased mitochondrial membrane potential, indicating mitochondria dysfunction. In summary, our results suggest that patulin treatment causes oocyte meiotic arrest by disturbing normal spindle assembly and chromosome alignment, which may be caused by dysfunctions of mitochondria.

Abstract:[Objective] The aim was to study the inhibition mechanism of the chitosan-oligosaccharides prepared through enzymatic hydrolysis to Cladosporium cueumerinum. [Method] The inhibition rate of the ehitosan-oligosaceharides upon the hyphae and the spore germination of Cladosporium cueumerinum were determined. The morphology shape of hyphae and the cell membrane permeability of the pathogen were observed before and after the treatment [Result] When the concentration of the chitosan-oligosaccharides was 0.2 g/mL, the hyphae growth were inhibited. When the concentration was 0. 1g/mL, the spore germination were inhibited. According to the microscope observation, the chitosan-oligosaccharides with concentration of 0.8 mg/mL induced the morphological change, including excessive branching and hyphal segments in the new hyphal of Cladosporium cueumerimum. The formation of conidiospore was inhibited. The chitosan-oligosaccharides could induced the permeability of the cell membrane and the inclusion leakage of mycelium. [Conelusion] This study could provide references for the inhibition mechanism of Cladosporium cueumerinum.
Involvement of Organic Acid in the Control Mechanism of ε-Poly-L-lysine (ε-PL) on Blue Mold Caused by Penicillium expansum in Apple Fruits

Abstract:Blue mold is one of the most serious postharvest diseases in apples caused by Penicillium expansum. The purpose of this study is to determine the effect of ε-poly-L-lysine (ε-PL) on the pathogenicity of P. expansum and explore the potential mechanism from the perspective of organic acid. The study investigates the effect of ε-PL treatment on the growth and acid production of P. expansum in vitro and in vivo. When the concentration of ε-PL was 50 mg/L, the growth of P. expansum was inhibited and the decrease in pH value was delayed in the medium. For example, on the third day of culture, P. expansum reduced the pH of the medium from 6.1 to 4.15, and ε-PL inhibited the decrease in the pH value at most 34.4%. When the concentration reached 1000 or 2000 mg/L, the infection of P. expansum in fruits was effectively inhibited. During the growth and infection of P. expansum, gluconic acid is one of the main factors leading to the pH value falling in the local environment. After ε-PL treatment, the accumulation of gluconic acid decreased, the activity of glucose oxidase was suppressed, and then the decline in the local environmental pH slowed down. In addition, after ε-PL treatment, the activities of cell-wall-degrading enzymes, such as cellulase (CL) and polygalacturonase (PG), in the different areas of the P. expansum–apple interaction were also affected by pH change. The results show that ε-PL inhibited the pathogenicity of P. expansum by affecting the accumulation of gluconic acid and slowing the decline in pH in fruit tissues, so as to affect the pathogenicity of P. expansum. This is the first time that the mechanism of ε-PL interfering with the pathogenicity of P. expansum from the perspective of organic acids is clarified.

Abstract:Patulin, one of the most common mycotoxins produced primarily by the Penicillium, Aspergillus and Byssochlamys species, is often associated with fruits and fruit-based products. Biodegradation by microbes is an effective method to remove or detoxify mycotoxins. In this study, a bacterial strain with patulin degradation capability was selectively isolated using oxindole, an analogue to patulin, as the sole carbon source, and identified as Pseudomonas poae JSU-Y1 by phylogenetic analysis on the basis of 16S rRNA sequence. This isolated bacterium could inhibit the growth of Penicillium expansum both on plate medium and apple fruit with inhibition ratio of 30.3% and 44.9%, respectively. Up to 87.7% of the initial patulin (2.5 μg/mL) was removed after incubation with Pseudomonas poae JSU-Y1 in liquid medium at 30 °C for 72 h. When challenged with apple juice, 79% of patulin could be degraded by this isolated strain. Additionally, ascladiol was tentatively identified as the patulin degradation intermediate by LC-MS analysis. Taken together, the experiment results indicated that the isolated Pseudomonas poae JSU-Y1 would be a promising bacterial resource to control patulin contamination and toxigenic fungal growth in agricultural products.

Biocontrol Agents Increase the Specific Rate of Patulin Production by Penicillium expansum but Decrease the Disease and Total Patulin Contamination of Apples

Abstract:Synthetic fungicides are commonly employed for the control of postharvest diseases of fruits. However, due to health concerns about the use of these chemicals, alternative control methods including biocontrol based on antagonistic yeasts are gaining in popularity. In this study, we investigated the effects of two biocontrol yeasts, Rhodotorula mucilaginosa strain 3617 and Rhodotorula kratochvilovae strain LS11, on blue mold and patulin (PAT) contamination caused by Penicillium expansum strains PY and FS7 in artificially inoculated Fuji apples stored at 20°C for 9 days. To correlate the development of the P. expansum strains in yeast-treated and untreated apples with PAT production, we quantified their biomass in the infected fruits using a recently published quantitative real-time polymerase chain reaction method based on specific primers for patF, a gene from P. expansum that is involved in PAT biosynthesis. Both yeasts significantly reduced the disease incidence caused by the two strains of P. expansum up to 5-7 days of incubation, and lowered their biomass and the progression of symptoms up to 9 days. Interestingly, both yeasts strains increased the rate of PAT production (expressed as ng patulin/μg fungal DNA) by the two pathogenic strains. Nevertheless, both biocontrol agents reduced the total PAT contamination, especially in the case of P. expansum strain FS7, the higher PAT producer of the two tested P. expansum strains. Comparing between the yeast strains, R. kratochvilovae LS11 was more effective than R. mucilaginosa 3617 for the control of P. expansum.

Abstract:This study aimed to investigate the effect of chitin on the antagonistic activity of Rhodosporidium paludigenum against Penicillium expansum, the cause of blue mold in apple fruit, and the possible mechanisms involved. Our results showed that biocontrol efficacy and population growth of R. paludigenum were greatly enhanced when it was harvested from nutrient yeast dextrose with added chitin (NYCB)medium compared with that harvested from nutrient yeast dextrose (NYDB) medium. The ability of R. paludigenum produced in NYCB to induce resistance to blue mold in apple fruit was significantly enhanced. The enhanced disease control efficacy was correlated with higher levels of polyphenoloxidase (PPO) and superoxide dismutase (SOD) activities in apple fruit treated with R. paludigenum. Moreover, the SOD and catalase (CAT) activities of R. paludigenum were stimulated by cultivating in NYCB, while malondialdehyde ( MDA) accumulation in the yeast cells was suppressed. These results indicated that adding chitin to normal media might be an effective method to improve the antagonistic activity of R. paludigenum and the active oxygen metabolism of R. paludigenum might be closely related to the biocontrol activity of the yeast. Genetic identification, pathogenicity and patulin production of Penicillium species from apple blue mold in China

Abstract:Abstract Blue mold is a common postharvest disease that leads to serious rot and patulin contamination in fruits, which is a key factor endangering the storage and quality of apple products. However, the Penicillium species from apple blue mold in China and their pathogenicity and patulin (PAT) production ability have not been well studied. In this study, 62 strains of pathogenic Penicillium species were isolated from blue mold apples collected from different Chinese regions. The ITS, BenA and CaM genes were sequenced for molecular identification. Penicillium strains were identified as three species with 57 P. expansum, 3 P. crustosum, and 2 P. polonicum. Twelve variants of the ITS sequence, 61 variants of BenA and 67 variants of CaM genes were identified between Penicillium species. Fungal morphology was observed on PDA. After 10 d of cultivation, P. expansum cultures were detected with PAT concentrations ranging from 457.98 to 1333.85 mg/kg, and two P. polonicum strains were 6.02 and 6.30 mg/kg. Three P. crustosum strains could not produce PAT on PDA. In apple inoculation, P. expansum strains showed significantly faster infection rates than P. crustosum and P. polonicum strains. All P. expansum strains can produce PAT during infection, and the concentrations in the rot apple tissues were 2.31 to 88.67 mg/kg. The P. crustosum and P. polonicum strains could not produce PAT during infection. This study provided data and information regarding the morphology, virulence and PAT production of Penicillium pathogens that will improve the understanding of apple blue mold and postharvest disease control.

Abstract:ABSTRACT: Acidified Oxine (AO) is a mixture of chlorite and chlorine dioxide. At 7.8 ppm available chlorine dioxide (ACD) for 10 min, it reduced 7.4 log CFU/mL of Erwinia carotovora, a potato pathogen, in phosphate buffer. Exposing potatoes to air purged through AO solutions proportionally reduced inoculated E. carotovora. A 40% reduction of spoilage was achieved by exposing to AO of 100 ppm ACD under an accelerated test. AO was demonstrated to be potentially effective in preventing potato spoilage without any significant risks of chemical residual or change of skin color.

Abstract:Fusarium graminearum is the main worldwide causal agent of Fusarium head blight grain disease. The production of mycotoxin caused by Fusarium head blight epidemics of wheat affects the flour and malt products, which results in heavy economic losses to farmers. In this paper, an inactivation procedure for F. graminearum F7875 spores was tested and found to follow a first-order kinetic model. We characterized the inactivation of F7875 by aqueous chlorine dioxide (ClO2) under different concentrations (5 -15 mg L-1). The results suggest that aqueous ClO2 significantly enhanced inactivation of F7875 at high concentration (15 mg L-1) compared to lower concentration of 5 and 10 mg L-1. The time required for a five-log reduction of F7875 spores was calculated based on the established models. Observed by scanning electron microscope, the ClO2 destroys the structure of the outer coats of F7875, the surface of F7875's hyphal appeared smooth prior to treatment, but after treatment with 30 mg L-1 aqueous ClO2, it appeared rugged and wrinkled. In conclusion, our study shows that treatment with aqueous ClO2 is an effective, fast, and safe method to control F. graminearum in grain production(.) (C) 2017 Published by Elsevier Ltd.
Transcriptomic Analysis Reveals the Inhibition Mechanism of Pulsed Light on Fungal Growth and Ochratoxin A Biosynthesis in Aspergillus Carbonarius.

Abstract:Pulsed light (PL) technology has a good effect on the control of fungi in postharvest fruit. In this present work, PL inhibited the growth of Aspergillus carbonarius in a dose-dependent manner, the mycelial growth decreased by 4.83 %, 13.91 % and 30.01 % at a fluence of 4.5 J.cm(-2) (PL5), 9 J.cm(-2) (PL10) and 13.5 J.cm(2) (PL15), respectively. When inoculated with PL15 treated A. carbonarius, the scab diameter of the pears, ergosterol and OTA content was reduced by 23.2 %, 27.9 % and 80.7 % after 7 days, respectively. The third-generation sequencing technique was applied to study the transcriptome response of A. carbonarius treated with PL. Compared with the blank control, a total number of 268 and 963 differentially expressed genes (DEGs) were discovered in the group of PL10 and PL15, respectively. To be specific, a large amount of DEGs involved in DNA metabolism were up-regulated, while most of DEGs related to cell integrity, energy and glucose metabolism, ochratoxin A (OTA) biosynthesis and transport were down-regulated. In addition, the stress response of A. carbonarius was imbalanced, including up-regulation of Catalase and PEX12 and down-regulation of taurine and subtaurine metabolism, alcohol dehydrogenase and glutathione metabolism. Meanwhile, the results of transmission electron microscopy, mycelium cellular leakage and DNA electrophoresis indicated that PL15 treatment caused mitochondrial swelling, the destroyed cell membrane permeability and imbalance of DNA metabolism. The expression of P450 and Hal involved in OTA biosynthesis pathway were down-regulated in PL treated samples detected by qRT-PCR. In conclusion, this study reveals the molecular mechanism of pulsed light on inhibiting the growth, development and toxin production of A. carbonarius.
Effect of Cinnamaldehyde and Citral Combination on Transcriptional Profile, Growth, Oxidative Damage and Patulin Biosynthesis of Penicillium expansum

Abstract:Penicillium expansum, as a main postharvest pathogen of fruits, can secrete patulin (PAT), causing fruit decay and health problems. In this study, the antifungal test, SEM (scanning electron microscope) observation, transcriptional profile, PAT biosynthesis, and physiological characters of P. expansum exposed to cinnamaldehyde and citral combination (Cin/Cit) were evaluated. Cin/Cit could inhibit the mycelial growth and spore germination of P. expansum in a dose-dependent manner. Besides, Cin/Cit caused spores and mycelia wrinkled and depressed by SEM observation. Gene expression profiles of P. expansum were conducted by RNA sequencing (RNA-seq) in the presence or absence of Cin/Cit treatment. A total of 1713 differentially expressed genes (DEGs) were obtained, including 793 down-regulated and 920 up-regulated genes. Most of the DEGs participated in the biosynthesis of secondary metabolites, amino acid metabolism, and oxidation-reduction process, etc. Cin/Cit induced the dysfunction of the mitochondrial membrane, causing the potential influence on energy metabolism and reactive oxidative species production. The changes of superoxide dismutase (SOD) and catalase (CAT) activities combing with the increase of hydrogen peroxide content indicated the oxidative stress on P. expansum induced by Cin/Cit, which corresponded well with the transcriptional results. Moreover, both the RNA-seq data and the qRT-PCR showed the remarkable down-regulation of genes included in the PAT biosynthetic pathway under the Cin/Cit treatment. These findings provided more useful information about the antifungal mechanism of Cin/Cit against P. expansum at molecular and gene levels and suggested that Cin/Cit is a potential candidate to control P. expansum.

Inactivation of Inoculated Penicillium on Grape by Chlorine Dioxide Gas and Variation of Nutrients of Grape

Abstract:The bactericidal effect to the Penicillium on the grape by the different ClO2 gas concentrations at 30 min was studied.The experimental results showed that with the ClO2 gas concentration(5 mg/m3,10 mg/m3,15 mg/m3) increased the sterilization rate increased(respectively 2.62 log,3.05log and 3.21 log).After treatment by the ClO2 gas,the content of Vc,reducing sugar and titratable acid in the grape also been studied,and the results showed that When the concentration of ClO2 gas were 5 mg/m3 and 10 mg/m3,the content of Vc appeared the trend that first reduced and then upward along with the time prolong from 10 to 60 min.When the concentration of ClO2 gas was 15 mg/m3,the content of Vc and reducing sugar were both increased with the time(10~60 min) prolong.The titratable acid appeared an upward trend with the concentration of ClO2 and the time increased.
Inhibitory effects and mechanisms of cinnamaldehyde against Fusarium oxysporum, a serious pathogen in potatoes

Abstract:Background: Potatoes, a major economic crop, are significantly impacted by Fusarium dry rot, a prevalent postharvest disease. Despite the broad-spectrum antimicrobial properties of cinnamaldehyde, a naturally-derived plant substance, its efficacy against the causal pathogen of potato dry rot (Fusarium oxysporum) and the underlying mechanisms have not been extensively studied. Results: Our study demonstrates that cinnamaldehyde effectively inhibits the growth of Fusarium oxysporum, the pathogen responsible for potato dry rot, and increases its sensitivity to environmental stress factors such as extreme temperatures and high salt stress. Treatment with cinnamaldehyde results in altered fungal mycelium morphology, compromised cell wall stability, and disrupted cell membrane integrity, thereby reducing spore viability. Specifically, it interferes with the cell membrane and cell wall structures of the fungus, potentially disrupting fungal growth by modulating signaling pathways involved in cell wall maintenance, chitin metabolism, and GPI-anchored protein function. Notably, we show that cinnamaldehyde induces a form of regulated cell death in F. oxysporum, which is characterized not as typical apoptosis, as evidenced by Annexin V negative staining. However, the specific cell death type and underlying mechanism still needed to be further explored. Conclusion: Cinnamaldehyde, an environmentally friendly plant-based active compound, exhibits strong inhibitory effects on F. oxysporum, indicating its potential use in the prevention and control strategies for potato dry rot. This research contributes to the understanding of novel antifungal mechanisms and offers promising insights into eco-friendly alternatives for managing this economically significant postharvest disease. © 2024 Society of Chemical Industry.
