Identification of a New Allele, BrPL, Regulating the Purple Leaf Trait in Chinese Cabbage (brassica Rapa L. Ssp. Pekinensis)

Abstract:Key message: Anthocyanin is an important secondary metabolite in plants. At present, there are few studies on the anthocyanin biosynthesis mechanism in Chinese cabbage. In this study, Chinese cabbage purple leaf line 19-(AV x V)-3 and green leaf line Y1211-1 were used to construct a six-generation population. Analysis of the F2 and BC1 phenotypes showed that the purple leaf trait was controlled by a single dominant gene. Purple leaf gene BrPL was finely mapped and cloned. After designing insertion-deletion (InDel) markers, BrPL was finely mapped between PX1 and PX2, and the candidate interval was 23.23 kb (23.195-23.218 Mb). Functional annotation showed that the BraA07g033110.3.5C gene was homologous to AT1G66390 in Arabidopsis thaliana, encoding an R2R3-MYB transcription factor that promoted anthocyanin synthesis. Compared with the Y1211-1 sequence, 19(AV x V)-3 had 58-bp and 3772-bp deletions in the promoter region and intron 1, respectively. We speculated that the deletion of the promoter region and intron 1 promotes anthocyanin biosynthesis. Combined transcriptome and metabolome analysis showed that cyanidin was the main component affecting the color of the purple leaves in Chinese cabbage. MYB2 promoted the high expression of CHS, CHI, F3H, F3 ' H, DFR, ANS, UFGTs, and GST in purple materials, while FLS expression was significantly upregulated in green materials, with the differential expression of these genes resulting in differences in their traits. The results of this study provide new insights into anthocyanin biosynthesis and purple germplasm production in Chinese cabbage.
