Abstract:Chinese kale is a widely cultivated plant in the genus Brassica in the family Brassicaceae. The origin of Brassica has been studied extensively, but the origin of Chinese kale remains unclear. In contrast to Brassica oleracea, which originated in the Mediterranean region, Chinese kale originated in southern China. The chloroplast genome is often used for phylogenetic analysis because of its high conservatism. Fifteen pairs of universal primers were used to amplify the chloroplast genomes of white-flower Chinese kale (Brassica oleracea var. alboglabra cv. Sijicutiao (SJCT)) and yellow-flower Chinese kale (Brassica oleracea var. alboglabra cv. Fuzhouhuanghua (FZHH)) via PCR. The lengths of the chloroplast genomes were 153,365 bp (SJCT) and 153,420 bp (FZHH) and both contained 87 protein-coding genes and eight rRNA genes. There were 36 tRNA genes in SJCT and 35 tRNA genes in FZHH. The chloroplast genomes of both Chinese kale varieties, along with eight other Brassicaceae, were analyzed. Simple sequence repeats, long repeats, and variable regions of DNA barcodes were identified. An analysis of inverted repeat boundaries, relative synonymous codon usage, and synteny revealed high similarity among the ten species, albeit the slight differences that were observed. The Ka/Ks ratios and phylogenetic analysis suggest that Chinese kale is a variant of B. oleracea. The phylogenetic tree shows that both Chinese kale varieties and B. oleracea var. oleracea were clustered in a single group. The results of this study suggest that white and yellow flower Chinese kale comprise a monophyletic group and that their differences in flower color arose late in the process of artificial cultivation. Our results also provide data that will aid future research on genetics, evolution, and germplasm resources of Brassicaceae.

Abstract:Brassinazole resistant 1 (BZR1), a brassinosteroid (BR) signaling component, plays a pivotal role in regulating numerous specific developmental processes. Our study demonstrated that exogenous treatment with 2,4-epibrassinolide (EBR) significantly enhanced the accumulation of carotenoids and chlorophylls in Chinese kale (Brassica oleracea var. alboglabra). The underlying mechanism was deciphered through yeast one-hybrid (Y1H) and dual-luciferase (LUC) assays, whereby BoaBZR1.1 directly interacts with the promoters of BoaCRTISO and BoaPSY2, activating their expression. This effect was further validated through overexpression of BoaBZR1.1 in Chinese kale calli and plants, both of which exhibited increased carotenoid accumulation. Additionally, qPCR analysis unveiled upregulation of carotenoid and chlorophyll biosynthetic genes in the T1 generation of BoaBZR1.1-overexpressing plants. These findings underscored the significance of BoaBZR1.1-mediated BR signaling in regulating carotenoid accumulation in Chinese kale and suggested the potential for enhancing the nutritional quality of Chinese kale through genetic engineering of BoaBZR1.1.
An Efficient Mesophyll Protoplast Isolation, Purification and PEG-mediated Transient Gene Expression for Subcellular Localization in Chinese Kale

Abstract:Transient gene expression serves as a valuable tool for gene functional study in plants. Here we reported protoplast isolation and purification from the plantlet young leaves of Chinese kale (Brassica oleracae var. alboglabra Bailey), and their transient gene expression with polyethylene glycol (PEG)-mediated transformation and sub cellular localization of phytoene desaturase (BaPDS1). The procedures of isolation andtransformation ofmesophyll protoplasts derived from Chinese kale were optimized, and the influencing factors were analyzed. The results showed that the optimal protocol of protoplast isolation and purification was initialized by digestion in enzyme solution (2.0% cellulase, 0.1% pectolase, and 0.6 M mannitol) for 9 h. After filtered through 400 mesh and centrifuged at 179 x g for purification, the total yield of protoplast reached as high as 6.04 x 107 protoplasts g(-1) fresh weight (FW) and the viability of the protoplasts was up to 95%. A maximum transformation efficiency of approximately 30% measured by using green fluorescent protein (GFP) as a detecting gene was obtained when PEG4000 was at a final concentration of 40% and transformation time was set to 15 min. In addition, the subcellular localization of BaPDS1 in Chinese kale was targeted to the chloroplast, confirming the efficiency and reliability of this transient transformation system. Taken together, an efficient protoplast isolation, purification and transformation system in Chinese kale was established in this study, laying a foundation for future research in molecular biology and gene function in Chinese kale and other Brassica vegetables.
A Novel Putative 2-Oxoglutarate-dependent Dioxygenase Gene (Boaaop-Like) Regulates Aliphatic Glucosinolate Biosynthesis in Chinese Kale

Abstract:Glucosinolates are important secondary metabolites in Brassica vegetables, and the side-chain modifications of aliphatic glucosinolates are regulated by 2-oxoglutarate-dependent dioxygenases (AOPs). However, how the side-chain modifications of aliphatic glucosinolates are regulated in Chinese kale remains unclear. Here, we show that the novel gene BoaAOP-like affects the biosynthesis of aliphatic glucosinolates in Chinese kale. BoaAOP-like was localized in the cytoplasm of Chinese kale. The expression levels of BoaAOP-like increased with plant growth; among different organs, BoaAOP-like was the highest in the roots. BoaAOP-like was introduced into Chinese kale using Agrobacterium-mediated transient overexpression system, of which the expression levels were 2.20- to 49.56-fold higher in transiently overexpressed plants than in wild-type plants; the content of total aliphatic glucosinolates in transiently overexpressed plants was up to 70.26-fold higher than in wild-type plants. The content of each aliphatic glucosinolate was significantly accumulated in BoaAOP-like transiently overexpressed plants, with the gluconapin content increasing the greatest when compared to the wild-type plants. The expression levels of aliphatic glucosinolate-related genes, especially MYB28 and AOP2.1 were significantly up-regulated in transiently overexpressed plants. Overall, our results indicated that BoaAOP-like plays a critical role in regulating the biosynthesis of aliphatic glucosinolate in Chinese kale.

Abstract:ζ-Carotene desaturase（ZDS）is an important enzyme in carotenoid biosynthesis.Here,the Brassica oleracea var.alboglabra ZDS（Boa ZDS）gene was cloned from Chinese kale via reverse transcription-polymerase chain reaction（RT-PCR）and deposited in Gen Bank（accession number KY662297）.The Boa ZDS gene contains an open reading frame of 1 686 bp that encodes a 561-amino acid protein.Sequence analysis indicates that the ZDS protein is apparently conserved during plant evolution and is most closely related to B.oleracea var.capitata and B.rapa.The promoter sequence of the Boa ZDS gene was predicted to harbor several cis-acting elements that are related to light and phytohormone responses.Semiquantitative RT-PCR analysis showed that Boa ZDS expression varied among different developmental stages and organs.Relative ZDS expression remained stable during germination and seedling stages and rapidly increased at the mature leaf stage.The leaves showed the highest ZDS expression levels compared to the other organs.ZDS expression decreased in all flower tissues during blooming.The fused protein of Boa ZDS was obtained by prokaryotic expression.Heterologous expression of Boa ZDS in Escherichia coli confirmed that Boa ZDS encodes a functionalζ-carotene desaturase that increases β-carotene accumulation in E.coli cells harboring a β-carotene-producing plasmid.The findings of the present study provide a molecular basis for the elucidation of ZDS gene function in Chinese kale.
Overexpression of the Glucosyltransferase Gene BoaUGT74B1 Enhances the Accumulation of Indole Glucosinolates in Chinese Kale

Abstract:Glucosinolates are a diverse class of secondary metabolites that play a vital role in the plant defense response. Here, we investigated the function of the UDP-glucosyltransferase 74B1 (UGT74B1) gene in glucosinolate biosynthesis in Chinese kale. A 1401 bp coding sequence of BoaUGT74B1 was cloned from Chinese kale cultivar 'Sijicutiao'. Among different organs, BoaUGT74B1 has the highest expression level in young seeds. Among different flower tissues, BoaUGT74B1 has the highest expression level in the pistil; the gene was downregulated in flower tissues as flowers matured. Subcellular localization showed that BoaUGT74B1 is located in chloroplasts. A purified similar to 50 kD fusion protein was obtained from Escherichia coli after prokaryotic expression of BoaUGT74B1. BoaUGT74B1 introduced into the white-flower cultivar 'Sijicutiao' and yellow-flower cultivar 'Fuzhouhuanghua' of Chinese kale by Agrobacterium-mediated transformation was strongly in both cultivars. Expression levels of BoaUGT74B1 in the overexpressed plants 'Sijicutiao' and 'Fuzhouhuanghua' were 1.53- to 2.93-fold and 3.07- to 5.30-fold higher, respectively than that of the wild type; the total indole glucosinolate content in these two overexpressed plants was 1.30- to 7.11-fold and 2.13- to 4.44-fold higher, respectively, compared with that in wild-type plants. Each indole glucosinolate accumulated in significant amounts in all overexpressed plants; 4-methoxy glucobrassicin enhanced most in 'Sijicutiao' overexpressed plants, in concentrations up to 12.14-fold higher than the control; glucobrassicin enhanced most in 'Fuzhouhuanghua' overexpressed plants, reaching levels 22.76x that of its wild-type counterpart. Therefore, these findings indicate that BoaUGT74B1 plays an important role in the accumulation of indole glucosinolates in Chinese kale, and suggest the possible usefulness of this gene to improve the nutritional quality and resistance against biotic stress of Chinese kale.

Abstract:R2R3-MYB transcription factor MYB51 is known to control indole glucosinolate (indole GSL) biosynthesis in Arabidopsis. Here, two copies of BoaMYB51 have been isolated in Chinese kale (Brassica oleracea var. alboglabra Bailey), designated BoaMYB51.1 and BoaMYB51.2, which exhibit overlapping but distinct expression levels among different organs and respond to signaling molecules in a similar pattern. It has been demonstrated a structural and functional conservation between BoaMYB51s and AtMYB51 by phylogenetic analysis, complementation studies and transient expression assay. To further investigate the transcriptional mechanism, we identified the transcriptional activation domain (TAD) and putative interacting proteins of BoaMYB51s by means of yeast (Saccharomyces cerevisiae) two hybrid. Using tobacco (Nicotiana benthamiana) transient expression assay, we confirmed that the carboxy-end is required for transcriptional activation activity of BoaMYB51s. In addition, several BoaMYB51-interacting proteins have been identified by yeast two-hybrid screening. These results provide important insights into the molecular mechanisms by which MYB51 transcriptionally regulates indole GSL biosynthesis.
Fine-mapping of a Gene for the Lobed Leaf, BoLl , in Ornamental Kale ( Brassica Oleracea L. Var. Acephala )

Abstract:Leaf shape is an important agronomic trait of ornamental kale (Brassica oleracea L. var. acephala). In this study, we isolated a lobed-leaf DH line by microspore culture of ornamental kale. The genetic analysis indicated that the lobed-leaf trait was quantitatively inherited. By QTL-seq analysis, the candidate region for BoLl (lobed-leaf) was mapped on chromosome 9 (BRAD, Brassica oleracea, chromosome v1.0). The mapping region was confirmed and narrowed by simple sequence repeat (SSR) and insertion–deletion (Indel) markers in the F2 population; BoLl was located between the markers LYIn39 (0.17 cM) and LYIn40 (0.11 cM). According to the B. oleracea genome database (chromosome v1.0), the mapped interval (75.3 kb) contained eight genes, and seven of which were annotated in BRAD and one was not. According to the other B. oleracea genome information on EnsemblPlant, the mapped interval (79.6 kb) contained 11 genes. Two orthologous genes of AtLMI1/ATHB5 related to lobed leaves in Arabidopsis, Bol010029/Bo9g181710 (BRAD/EnsemblPlant) and Bol010030/Bo9g1181720, were identified as possible genes for BoLl, but the sequence analysis revealed no variation in their promoter and coding regions. The expression of Bol010029/Bo9g181710 and Bol010030/Bo9g1181720 was significantly higher in young lobed leaves than in young entire leaves. Among the other genes in the interval, only Bol010025 and Bol010031/Bo9g181730 showed co-dominance in the recombinant individuals, and their expression level was similar in lobed and entire leaves. Three hypotheses were proposed for the formation of lobed leaf. These results lay a foundation for uncovering the molecular mechanism of the lobed-leaf trait in ornamental kale.
Promoter variations in a homeobox gene, BrLMI1, contribute to leaf lobe formation in Brassica rapa ssp. chinensis Makino

Abstract:Abstract Leaves are the main consumed organ in leafy non-heading Chinese cabbage ( Brassica rapa L. ssp. chinensis Makino), and the shape of the leaves is an important economic trait. However, the molecular regulatory mechanism underlying lobed-leaf trait in non-heading Chinese cabbage remains unclear. Here, we identified a stable incompletely dominant major locus qLLA10 for lobed leaves formation in non-heading Chinese cabbage. Based on map-based cloning strategies, BrLMI1 , a LATE MERISTEM IDENTITY1 ( LMI1 )-like gene, was predicted as the candidate gene for qLLA10. Genotyping analysis showed that promoter variations of BrLMI1 in two parents are responsible for elevating the expression in the lobed-leaf parent and ultimately causing the alternation in leaf shape between the two parents, and the promoter activity of BrLMI1 was significantly affected by the promoter variations. BrLMI1 was exclusively localized in the nucleus and expressed mainly at the tips of each lobe. Leaf lobe development was perturbed in BrLMI1 -silenced plants produced by virus-induced gene silencing assays, and ectopic overexpression of BrLMI1 in Arabidopsis led to deeply lobed leaves never seen in the wild type, which indicates that BrLMI1 is required for leaf lobe formation in non-heading Chinese cabbage. These findings suggested that BrLMI1 is a positive regulatory factor of leaf lobe formation in non-heading Chinese cabbage and that cis-regulatory variations lead to the phenotype of lobed or entire leaf margins, thus providing the theoretical basis for unraveling the molecular mechanism underlying lobed leaves phenotype in Brassica crops.
Development and application of InDels and SNPs markers based on whole-genome resequencing in Chinese cabbage

Abstract:Abstract Chinese cabbage (Brassica rapa L. syn. B. campestris L., AA, 2n=20) is highly popular green leafy vegetable in the triangle of U, with China serving as its primary center of origin and diversification. Plant height, rosette leaf diameter, and disease resistance have always been important selection criteria for Chinese cabbage. In this study, we conducted re-sequencing on 55 Chinese cabbage accessions collected from China, Japan and Korea, followed population genetic evolutionary analysis and genome-wide association study (GWAS). Our findings unveiled a lack of significant population structure among these accessions, implying a possible evolutionary link or hybridization between Japanese and Korean Chinese cabbages with their Chinese counterparts. GWAS analysis results revealed that BrASD1, BrZHD13, BrGH3.3, and BrGH3.6 regarded as potential candidate genes highly associated with plant height in Chinese cabbage. Moreover, we also identified two single nucleotide polymorphisms (SNPs) loci associated with rosette leaf diameter and one SNP locus associated with downy mildew resistance, respectively. Furthermore, a novel tightly linked Insertion-deletion (InDel) marker on the end of A04 chromosome in B. rapa, SMB.4, has been developed. And SMB.4 can potentially serve as a useful molecular marker screening the genetic resources with downy mildew resistance and application for the downy mildew resistance breeding of Brassica crops.
Cloning and Characterization of the Microspore Development-Related Gene Bcmf2 in Chinese Cabbage Pak-Choi (brassica Campestris L. Ssp Chinensis Makino)

Abstract:: For the sake of providing some important information relevant to the study of the molecular mechanism of genic male sterility in plants, gene differential expression in flower buds at different developmental stages, as well as in rosette leaves, florescence leaves, and scapes was analyzed using cDNA amplified fragment length polymorphism (cDNA-AFLP) in the genic male sterile A and fertile B line of Chinese cabbage pak-choi. Following amplification of 125 pairs of primer combinations, 11 differential fragments were obtained, of which eight were from the B line and the other three were from the A line. Of 11 differential fragments, four were verified by Northern hybridization that were expressed preferentially in fertile flower buds. Results of GenBank BLAST showed that one fragment was with unknown function, whereas the other fragments have strong nucleotide sequence similarities with the polygalacturonase (PG) gene, the pectinesterase (PE) gene, and the polygalacturonase inhibitory protein (PGIP4) gene. Only full-length cDNA from the differential fragment BcMF-A18T16-1 was amplified by rapid amplification of cDNA ends (RACE) and Northern analysis showed that this fragment was expressed only in medium and large-sized flower buds of the B line. The full-length cDNA, designated as BcMF2 (Brassica campestris Male Fertile 2), was 1 485 bp long and was composed of a 1 263-bp open reading frame, which had 83% nucleotide similarity to a PG gene from Arabidopsis encoding polygalacturonase. Analysis of the basic structure of the protein revealed that it had one polygalacturonase active site (RVTCGPGHGLSVGS) at 256th site of amino acids and was classified as being a member of family 28 of the glycosyl hydrolases. Abstract:KEY MESSAGE:QTL mapping plus bulked segregant analysis revealed a major QTL for shoot branching in non-heading Chinese cabbage. The candidate gene was then identified using sequence alignment and expression analysis. Shoot branching is a complex quantitative trait that contributes to plant architecture and ultimately yield. Although many studies have examined branching in grain crops, the genetic control of shoot branching in vegetable crops such as Brassica rapa L. ssp. chinensis remains poorly understood. In this study, we used bulked segregant analysis (BSA) of an F2 population to detect a major quantitative trait locus (QTL) for shoot branching, designated shoot branching 9 (qSB.A09) on the long arm of chromosome A09 in Brassica rapa L. ssp. chinensis. In addition, traditional QTL mapping of the F2 population revealed six QTLs in different regions. Of these, the mapping region on chromosome A09 was consistent with the results of BSA-seq analysis, as well as being stable over the 2-year study period, explaining 19.37% and 22.18% of the phenotypic variation across multiple genetic backgrounds. Using extreme recombinants, qSB.A09 was further delimited to a 127-kb genomic region harboring 28 annotated genes. We subsequently identified the GRAS transcription factor gene Bra007056 as a potential candidate gene; Bra007056 is an ortholog of MONOCULM 1 (MOC1), the key gene that controls tillering in rice. Quantitative RT-PCR further revealed that expression of Bra007056 was positively correlated with the shoot branching phenotype. Furthermore, an insertion/deletion marker specific to Bra007056 co-segregated with the shoot branching trait in the F2 populations. Overall, these results provide the basis for elucidating the molecular mechanism of shoot branching in Brassica rapa ssp. chinensis Makino.

Genotyping analysis showed that promoter variations of BrLMI1 in the two parents are responsible for elevating the expression in the lobed-leaf parent and ultimately causing the difference in leaf shape between the two parents, and the promoter activity of BrLMI1 was significantly affected by the promoter variations. BrLMI1 was exclusively localized in the nucleus and expressed mainly at the tip of each lobe. Leaf lobe development was perturbed in BrLMI1 -silenced plants produced by virus-induced gene silencing assays, and ectopic overexpression of BrLMI1 in Arabidopsis led to deeply lobed leaves never seen in the wild type, which indicates that BrLMI1 is required for leaf lobe formation in non-heading Chinese cabbage. These findings suggested that BrLMI1 is a positive regulatory factor of leaf lobe formation in non-heading Chinese cabbage and that cis-regulatory variations lead to the phenotype of lobed or entire leaf margins, thus providing a theoretical basis for unraveling the molecular mechanism underlying the lobed leaf phenotype in Brassica crops.

Abstract:Isolating and characterizing mutants with altered senescence phenotypes is one of the ways to understand the molecular basis of leaf aging. Using ethyl methane sulfonate mutagenesis, a new rice (Oryza sativa) mutant, brown midrib leaf (bml), was isolated from the indica cultivar ‘Zhenong34’. The bml mutants had brown midribs in their leaves and initiated senescence prematurely, at the onset of heading. The mutants had abnormal cells with degraded chloroplasts and contained less chlorophyll compared to the wild type (WT). The bml mutant showed excessive accumulation of reactive oxygen species (ROS), increased activities of superoxide dismutase, catalase, and malondialdehyde, upregulation of senescence-induced STAY-GREEN genes and senescence-related transcription factors, and down regulation of photosynthesis-related genes. The levels of abscisic acid (ABA) and jasmonic acid (JA) were increased in bml with the upregulation of some ABA and JA biosynthetic genes. In pathogen response, bml demonstrated higher resistance against Xanthomonas oryzae pv. oryzae and upregulation of four pathogenesis-related genes compared to the WT. A genetic study confirmed that the bml trait was caused by a single recessive nuclear gene (BML). A map-based cloning using insertion/deletion markers confirmed that BML was located in the 57.32kb interval between the L5IS7 and L5IS11 markers on the short arm of chromosome 5. A sequence analysis of the candidate region identified a 1 bp substitution (G to A) in the 5′-UTR (+98) of bml. BML is a candidate gene associated with leaf senescence, ROS regulation, and disease response, also involved in hormone signaling in rice. Therefore, this gene might be useful in marker-assisted backcrossing/gene editing to improve rice cultivars.

Abstract:BACKGROUND:Members of the MtN3/saliva/SWEET gene family are present in various organisms and are highly conserved. Their precise biochemical functions remain unclear, especially in Chinese cabbage. Based on the whole genome sequence, this study aims to identify the MtN3/saliva/SWEETs family members in Chinese cabbage and to analyze their classification, gene structure, chromosome distribution, phylogenetic relationship, expression pattern, and biological functions.RESULTS:We identified 34 SWEET genes in Chinese cabbage and analyzed their localization on chromosomes and transmembrane regions of their corresponding proteins. Analysis of a phylogenetic tree indicated that there were at least 17 supposed ancestor genes before the separation in Brassica rapa and Arabidopsis. The expression patterns of these genes in different tissues and flower developmental stages of Chinese cabbage showed that they are mainly involved in reproductive development. The Ka/Ks ratio between paralogous SWEET gene pairs of B. rapa were far less than 1. In our previous study, At2g39060 homologous gene Bra000116 (BraSWEET9, also named BcNS, Brassica Nectary and Stamen) played an important role during flower development in Chinese cabbage. Instantaneous expression experiments in onion epidermal cells showed that the gene encoding this protein is localized to the plasma membrane. A basal nectary split is the phenotype of transgenic plants transformed with the antisense expression vector.CONCLUSION:This study is the first to perform a sequence analysis, structures analysis, physiological and biochemical characteristics analysis of the MtN3/saliva/SWEETs gene in Chinese cabbage and to verify the function of BcNS. A total of 34 SWEET genes were identified and they are distributed among ten chromosomes and one scaffold. The Ka/Ks ratio implies that the duplication genes suffered strong purifying selection for retention. These genes were differentially expressed in different floral organs. The phenotypes of the transgenic plants indicated that BcNs participates in the development of the floral nectary. This study provides a basis for further functional analysis of the MtN3/saliva/SWEETs gene family.
Molecular Characterization of MYB28 Involved in Aliphatic Glucosinolate Biosynthesis in Chinese Kale (Brassica oleracea var. alboglabra Bailey)

Abstract:Glucosinolates are Brassicaceae-specific secondary metabolites that act as crop protectants, flavor precursors, and cancer-prevention agents, which shows strong evidences of anticarcinogentic, antioxidant, and antimicrobial activities. MYB28, the R2R3-MYB28 transcription factor, directly activates genes involved in aliphatic glucosinolate biosynthesis. In this study, the MYB28 homology (BoaMYB28) was identified in Chinese kale (Brassica oleracea var. alboglabra Bailey). Analysis of the nucleotide sequence indicated that the cDNA of BoaMYB28 was 1257 bp with an ORF of 1020 bp. The deduced BoaMYB28 protein was a polypeptide of 339 amino acid with a putative molecular mass of 38 kDa and a pI of 6.87. Sequence homology and phylogenetic analysis showed that BoaMYB28 was most closely related to MYB28 homologs from the Brassicaceae family. The expression levels of BoaMYB28 varies across the tissues and developmental stages. BoaMYB28 transcript levels were higher in leaves and stems compared with those in cotyledons, flowers, and siliques. BoaMYB28 was expressed across all developmental leaf stages, with higher transcript accumulation in mature and inflorescence leaves. Over-expression and RNAi studies showed that BoaMYB28 retains the basic MYB28 gene function as a major transcriptional regulator of aliphatic glucosinolate pathway. The results indicated that over-expression and RNAi lines showed no visible difference on plant morphology. The contents of aliphatic glucosinolates and transcript levels of aliphatic glucosinolate biosynthesis genes increased in over-expression lines and decreased in RNAi lines. In over-expression lines, aliphatic glucosinolate contents were 1.5- to 3-fold higher than those in the wild-type, while expression levels of aliphatic glucosinolate biosynthesis genes were 1.5- to 4-fold higher than those in the wild-type. In contrast, the contents of aliphatic glucosinolates and transcript levels of aliphatic glucosinolate biosynthesis genes in RNAi lines were considerably lower than those in the wild-type. The results suggest that BoaMYB28 has the potential to alter the aliphatic glucosinolates contents in Chinese kale at the genetic level.

Abstract:Abundant anthocyanins accumulate in many flowers, leaves, fruits, as well as stems. In this study, an F2 segregating population derived from a cross between Chinese kale with a purple stem and kale with a green stem was used to decipher the genetic basis for the stem color. Bulked segregant analysis in combination with RNA sequencing (BSR-seq) was used to genetically map the causal gene for the purple stem (Ps), and the allele frequency difference between the two pools showed a single peak on chromosome C09. Ps was fine-mapped to the 0.32 cM interval with 406 kb sequences. A gene-encoding dihydroflavonol 4-reductase (DFR), an enzyme in the anthocyanin biosynthesis pathway, was differentially expressed between the two pools, and was identified as the candidate gene for the purple stem in Chinese kale. Sequence analysis revealed that 1 bp was inserted in the coding sequence of exon 2 of the BoDRF gene in green kale, causing frameshift and loss of function. The expression level of the BoDFR gene was significantly lower than the Chinese kale cultivar with a purple stem. The genetic results of the Ps gene will be useful for future development of purple vegetables.

Abstract:To provide information relevant to understanding the molecular mechanism of genic male-sterility in plants, differential expression of genes in flower buds at different developmental stages, and in other organs, between a sterile A line and a fertile B line of Chinese cabbage-pak-choi (Brassica rapa ssp. chinensis; syn. B. campestris ssp. chinensis) was analysed by cDNA-AFLP. Four cDNA fragments, including 13cMF-A17T14, were obtained from flower buds of the fertile B line. In this paper, on the basis of BcMF-A17T14, a spliced full-length cDNA designated BcMF4 was generated by rapid amplification of cDNA ends (RACE) from flower buds of the fertile B line of Chinese cabbage-pak-choi. The complete nucleotide sequence of the cDNA of BcMF4 was then amplified only from flower buds of the fertile B line. BcMF4 was expressed specifically in tetrads and uni-nucleate pollen of the fertile B line.,ne 1,102 bp DNA sequence of BcMF4 contains a single 95 bp intron and two exons. Its largest open reading frame (999 nt) encodes a polypeptide of 332 amino acids. The putative BcMF4 protein contains a signal peptide and displays five possible N-glycosylation sites, which are typical structures found in polygalacturonase inhibiting proteins (PGIPs). It also has eight Leucine-Rich Repeat (LRR) regions, and has 69 - 90% identity to PGIPs from other plants. Functional interruption of its ortholog in Arabidopsis thaliana, using an RNAi vector, resulted in aborted pollen grains and decreased pollen germination, and the abnormal pollen phenotype was stably inherited. This study suggests that the product of BcMF4 plays an important role in the development of microspores or pollen.
Characterization of a Novel Dominant Mutation BrCRSL Conferring Curled Rod-Shaped Leaves in Chinese Cabbage

Abstract:Leaf morphogenesis requires precise genetic control and hormone regulation to develop into ideal plant archi-tectures. Leaf flattening is an adaptive trait to obtain efficient photosynthesis, for which the leaf adaxial-abaxial polar establishment is of great importance. Herein, a novel leaf morphological mutant, curled rod-shaped leaf (crsl), was isolated and characterized from Chinese cabbage (Brassica rapa L. ssp. pekinensis) EMS-induced population. The leaves in crsl were slender and rod-shaped, with the abaxial side folded upwards and the distal side curled downwards, the mesophyll cells were loosely arranged, and the epidermal morphology was opposite on the adaxial and abaxial surface. Genetic analysis revealed that the curled rod-shaped leaf mutant phenotype was controlled by a dominate nuclear gene, named BrCRSL. MutMap and Kompetitive Allele Specifc PCR (KASP) genotyping demonstrated that BraA02g016100.3C was the most likely candidate for BrCRSL. Compared with the wild-type, a non-synonymous SNP mutation (C to T) occurred in the 8th exon of BraA02g016100.3C in crsl. BraA02g016100.3C is orthologous of Arabidopsis thaliana MAB1 (AT5G50850), which encodes a putative mitochondrial PDH E1 & beta; subunit protein and contributes to PIN-dependent auxin transport in organ development. The amino acid substitution from proline (P) to leucine (L) is conserved among the ho-mologous proteins of BraA02g016100.3C. The crsl mutant exhibited abnormal auxin metabolism, primarily manifested by high level of L-tryptophan (TRP) accumulation that is the precursor of IAA biosynthesis. Furthermore, the expression of a wide range of leaf adaxial/abaxial polarity-related genes was affected in crsl. Our results contribute to a better understanding of the genetic control mechanism underlying leaf morphogenesis in Chinese cabbage and provide new insights into the auxin-related leaf axial growth.
