Abstract:Background and aim: Pregnancy and lactation have an impact on health status of animals and constitute burden on body metabolites and the oxidant-antioxidant equilibrium. This study is aimed at evaluating metabolic and oxidative stress patterns and parity impacts in both primiparous and multiparous dairy cows. Materials and methods: Twenty-seven primiparous and multiparous. Holstein cows were enrolled and categorized into four groups according to their physiologic status: Primiparous peripartum heifer (n=5), primiparous postpartum cow (n=9), multiparous peripartum cows (n=5), and multiparous postpartum cows (n=8). Blood sample was taken from each animal - peripartum groups at 3 weeks prepartum and postpartum groups at 3 weeks post-parturition - for complete blood picture, glucose, cholesterol, triglyceride, total protein, albumin, non-esterified fatty acids (NEFA), malondialdehyde (MDA), total antioxidant capacity, and haptoglobin estimation. Results: Postpartum primiparous cows showed significant decrease in glucose, total protein, and albumin while showing significant increase in MDA, NEFA, and globulin; on the other hand, multiparous postpartum cows showed significant decrease in glucose, total protein, and albumin, associated with significant increase in cholesterol and MDA when compared with prepartum PP and MP cows, respectively. Postpartum multiparous cows significantly showed reduction in NEFA when compared to primiparous postpartum cows. Hematologic profiles of postpartum primiparous and multiparous cows showed significant decrease in red blood cells and packed cell volume, significant increase in lymphocytes when compared with prepartum cows. Conclusion: Metabolic and oxidative abnormalities exist in both primiparous and multiparous cows during the transition phase, however postpartum primiparous cows show higher susceptibility to negative energy balance impacts. Oxidant/antioxidant imbalance occurred in both the primiparous and multiparous postpartum cows, highlighting the importance of oxidative stress profiles in the assessment of metabolic health status during transition.
Effect of Rumen-undegraded Fat on Milk Yield,Milk Composition and Peripheral Leukocyte Number in Post-parturition Dairy Cows

Abstract:Forty multiparous Holstein dairy cows with similar parity and milk yielding of the last year were allocated equally into a control and an experimental group according to matched-pair grouping design. Beginning at the day of parturition until 90 days after parturition, cows in the experimental group were fed basic diet supplemented with 400 g rumen-passed fat. It was found that rumen-passed fat did not alter dry matter intake (DIM)(P0.05). However, it increased milk yielding(P0.05). Milk fat percentage tended to be increased by supplemental fat, although the differences between the two groups were not pronounced(P0.05). There were no differences in milk protein percentage between the two groups(P0.05). Supplemental fat had minor effect on total number of peripheral leukocytes and the number of lymphocyte(P0.05); however, it tended to reduce somatic cell counts (SCC) in milk (P0.05). It was concluded that supplemental fat increased milk yielding, and improved immune function to a certain extent.
Impact of postpartum milking frequency on the immune system and the blood metabolite concentration of dairy cows

Abstract:The transition from pregnancy to lactation is marked by metabolic, hormonal, and immunological changes that have an impact on the incidence of infectious and metabolic diseases. The aim of this study was to evaluate the effect on immune function and blood metabolite concentration of limiting milk production in early lactation to reduce negative energy balance. Twenty-two multiparous Holstein cows were milked either once a day (1x) or twice a day (2x) for the first week postpartum. All cows were milked twice daily for the rest of lactation. Blood concentrations of nonesterified fatty acids (NEFA), beta-hydroxybutyric acid (BHBA), calcium, bilirubin, urea, phosphorus, glucose, leptin, stanniocalcin-1, and 17beta-estradiol were determined in samples collected from 5 wk before scheduled calving to 5 wk after calving. Polymorphonuclear leukocytes (PMNL) were isolated from blood to conduct assays for chemotaxis, phagocytosis, and respiratory burst. Peripheral blood mononuclear cells (PBMC) were isolated to evaluate lymphocyte proliferation and cytokine production (tumor necrosis factor-alpha, IL-4, and interferon-gamma). Cows milked 1x produced 31% less milk than cows milked 2x during the first week of lactation. Over the following 13 wk of lactation, the milk production of cows milked 1x during the first week was 8.1% lower than for cows milked 2x. However, because the percentages of fat and protein were greater in the milk from 1x cows, the yields of milk components and energy-corrected milk were similar. Calving induced an increase in the concentrations of NEFA, BHBA, urea, and bilirubin. The increases in levels of NEFA and BHBA were greater in cows milked 2x than in cows milked 1x. During the same period, the serum glucose concentration decreased but remained greater in cows milked 1x. Serum calcium on d 4 and serum phosphorus on d 4 and 5 were greater in cows milked 1x. The differences between the 2 groups persisted beyond treatment until postpartum d 24 for NEFA and glucose and until postpartum d 14 for BHBA. After calving, the concentrations of leptin and stanniocalcin-1 decreased. During the first week postpartum, the decrease of leptin was less marked in cows milked 1x. The immune functions of PBMC and PMNL isolated from experimental cows and incubated using a standard medium did not show clear-cut peripartum immunosuppression. These variables were not significantly affected by the treatments, with the exception of interferon-gamma secretion, which was greater on d 5 and 14 in cows milked 1x. In conclusion, limiting milk production in early lactation had positive effects on metabolite concentration, but larger studies are necessary to establish if this could reduce disease incidence.
Impact of parity on carcase and metabolic markers associated with oxidative stress during uterine involution in periparturient goat

Abstract:This study aimed to verify the impact of parity on tissue mobilisation, metabolic response, oxidative stress and reproductive tract traits during the peripartum period. Nineteen goats with singleton pregnancies were grouped into nine nulliparous (NU) and 10 multiparous (MU). The animals were followed from the fifth week before delivery to the fourth week after delivery. Does, kids and milk production were weighed; the loin muscle depth area, subcutaneous loin, kidney fat thicknesses, size of the uterus and haemodynamics of the uterine artery were measured by ultrasound. Placenta was weighed and the cotyledons were counted and measured. Plasma was assayed for total protein, glucose, cholesterol, triglycerides, creatinine, gamma-glutamyl transferase (GGT), glutamic-oxaloacetic acid transaminase (GOT) and glutamic-pyruvic acid transaminase (GPT), glutathione peroxidase and β-hydroxybutyrate. MU showed a greater live weight loss (+8%) and longer period of body mass loss (+12 days). Milk production was higher in MU and their kids were heavier at birth with greater weight gain. No differences in the effect of parity were found for uterine diameter and placental weight, while the NU goats had a larger cotyledonary surface. Additionally, MU goats had higher levels of glucose, GPT, GGT, cholesterol, total protein and glutathione peroxidase. In MU females, there was a greater muscle mass mobilisation throughout the peripartum period and a superior replacement of adipose tissue after delivery (+1.3 mm). Therefore, MU females appear to have better tissue mobilisation and productive performance dynamics, although parity does not affect uterine involution in goats. Parity influences metabolic response and oxidative stress during peripartum in goats. Multiparous goats show greater efficiency in the mobilisation of muscle and adipose tissue and better productive performance. Parity does not influence the process of uterine involution.

Abstract:Simple Summary Serum biochemical parameters and hormones play a role in directly reflecting the physiological state and modulating the milk performance of dairy cows. However, the variability of the serum biochemical parameters and hormones in multiparous mid-lactation cows have not drawn much attention due to their supposedly stable states, especially for those cows under the same nutrition and management condition. The aim of this study was to evaluate the effects of days-in-milk (DIM, within the mid-lactation) and parity (ranging from 2-6) on serum biochemical parameters and hormone profiles based on a large cohort of dairy cows. The results showed that DIM and parity contribute to the variations in serum biochemical parameters and hormones related to protein status, energy supply, liver and kidney function, and oxidative stress of mid-lactation dairy cows, with the effect of DIM being dominant over parity. Our result suggested that the DIM periods and parity should be taken into consideration to optimize nutritional strategies in order to improve the milk performance traits more precisely.Abstract It is well known that serum biochemical parameters and hormones contribute greatly to the physiological and metabolic status of dairy cows. However, few studies have focused on the variation of these serum parameters in multiparous mid-lactation cows without the interference of diet and management. A total of 287 Holstein dairy cows fed the same diet and maintained under the same management regime were selected from a commercial dairy farm to evaluate the effects of days-in-milk (DIM) and parity on serum biochemical parameters and hormone profiles. Milk yield and milk protein content were affected by DIM and parity (p < 0.05). Milk protein yield showed a numerically decreasing trend with parity, and it was relatively constant in cows with parities between 2 and 4 but lower in cows with parity 6 (p = 0.020). Ten and five serum biochemical parameters related to protein status, energy metabolism, liver and kidney function, and oxidative stress were affected by DIM and parity, respectively (p < 0.05). Glucagon, insulin-like growth factor 1 concentration, and the revised quantitative insulin sensitivity check index were significantly different (p < 0.05) among cows with different DIM. Parity had no effect on hormone concentrations. An interaction between DIM and parity effect was only detected for glucagon concentration (p = 0.015), which showed a significantly increasing trend with DIM and overall decreasing trend with parity. In summary, DIM and parity played an important role in affecting the serum biochemical parameters and/or hormones of dairy cows, with serum parameters affected more by DIM than parity.

Abstract:The 16S rRNA gene sequences of 19 strains, 11 strains representing validated Aureobacterium or Microbacterium species and eight strains of non-valid species or isolates, were determined. These sequences were aligned with the sequences of other validated Aureobacterium and Microbacterium species and related actinobacteria. A comparative sequence analysis of 43 strains revealed that the species of the genera Aureobacterium and Microbacterium form a monophyletic association in which species of both genera are intermixed. The high similarity in phylogenetic properties found in the species within both genera and the close relationship in physiological and chemotaxonomic features other than the diamino acid in the cell wall, provided strong evidence that the genera Aureobacterium and Microbacterium should be unified. An emended genus Microbacterium is proposed for the two combined genera. The following validated Aureobacterium species were combined to the genus Microbacterium: Aureobacterium arabinogalactanolyticum to Microbacterium arabinogalactanolyticum, Aureobacterium barkeri to Microbacterium barkeri, Aureobacterium esteraromaticum to Microbacterium esteraromaticum, Aureobacterium flavescens to Microbacterium flavescens, Aureobacterium keratanolyticum to Microbacterium liquefaciens, Aureobacterium luteolum to Microbacterium luteolum, Aureobacterium saperdae to Microbacterium saperdae, Aureobacterium schleiferi to Microbacterium schleiferi, Aureobacterium terrae to Microbacterium terrae, Aureobacterium terregens to Microbacterium terregens, Aureobacterium testaceum to Microbacterium testaceum, and Aureobacterium trichothecenolyticum to Microbacterium trichothecenolyticum.
Effects of parity and early pregnancy on peripheral blood leukocytes in dairy cattle

Abstract:Subfertility remains a major problem in the dairy industry. Only 35-40% of high-yielding dairy cows and 55-65% of nonlactating heifers become pregnant after their first service. The immune system plays a critical role in the establishment of pregnancy. However, it can also create challenges for embryo survival and contribute to reduced fertility. We conducted 2 separate experiments to characterize changes in subsets of peripheral blood leukocytes (PBL) and their phenotype over the estrous cycle and early pregnancy in heifers and cows. We used flow cytometry and RT-qPCR to assess protein and mRNA expression of molecules important for immune function. We observed that monocytes and T cells were most affected by pregnancy status in heifers, whereas, CD8+ lymphocytes and natural killer (NK) cells were most affected during early pregnancy in cows. Changes in immune parameters measured appeared to be greater in heifers than cows including changes in expression of numerous immune function molecules. To test the hypothesis, we conducted a third experiment to simultaneously analyze the immunological responses to pregnancy between cows and heifers. We observed that cows had greater expression of proinflammatory cytokines and molecules associated with leukocyte migration and phagocytosis compared to heifers. Moreover, animals that failed to become pregnant showed altered expression of anti-inflammatory molecules. Overall, these findings support the hypothesis that early pregnancy signaling alters the proportions and functions of peripheral blood immune cells and differences between cows and heifers may yield insight into the reduced fertility of mature lactating dairy cows.

Abstract:Favorable uterine involution and ovarian activity are very important for the next reproductive cycle of postpartum cows. The objective of this study was to evaluate the effect of parity on uterine involution and resumption of ovarian activity in Chinese Holstein dairy cows after calving under similar postpartum nutritional conditions. Traits of the status of uterus and ovaries detected by ultrasonography, dry matter intake (DMI), milk yield, body condition score (BCS), and estradiol concentration in milk samples were analyzed for 46 Chinese Holstein dairy cows in various parities (primiparous=18; biparous=13; multiparous=15). The results showed that there was no significant difference for DMI, BCS, and milk yield among different parities; all cows were considered to be under similar nutritional conditions. Days of the previous gravid uterine horn involution were significantly greater in primiparous dairy cows than in biparous and multiparous dairy cows. Days from calving to ovulation (first and second) and the number of follicular waves to first ovulation were significantly greater in primiparous cows than in multiparous cows. In summary, there was a significant negative relationship between parity and postpartum uterine involution and resumption of ovarian activity in Chinese Holstein cows under similar body conditions.

Abstract:This study aimed to estimate the changes in the milk yield, milk fat, energy indicators [NEFA, BHBA, Dry MatterIntake (DMI) and Body Condition Score (BCS)] and concentration of innate immune molecules (Haptoglobin: Hp,Serum Amyloid A: SAA, TLR-4, TNF-α, IL-1β, IL-6, and IL-8), during the transition period in primiparous andmultiparous dual-purpose zebu (Deoni) cows. The blood sample was collected at weekly intervals during pre-partum (-21±2, -14±1, -7±1, d), date of calving (day 0), and postpartum period (3±1, 7±1, 14±1, 21±2 d) for estimationof the above plasma variables using commercially available bovine specific ELISA kits. DMI and BCS duringthe corresponding period were also recorded. Data were analyzed using a linear mixed model considering group,time and their interaction as fixed effects. Group, time and their interaction had significant effect on DMI whereprimiparous cows consumed higher DMI during early postpartum period as compared to multiparous cows. Groupalone had significant effect on milk yield, milk fat per cent and BHBA level while time alone influenced BCS.The interaction of group and time had significant effects on plasma TLR-4 and IL-8 concentration. Group alsohad significant effect on Hp and TNF-α levels. It was concluded that parity had significant effect on metabolicand immune indicators where higher DMI during transition period resulted in more milk yield in primiparous thanmultiparous indigenous (Deoni) cows.

Abstract:Dairy cows face metabolic challenges in the transition from late pregnancy to early lactation. The energy demands for the growing fetus and the onset of milk production are increasing but voluntary feed intake often decreases around parturition and cannot meet these demands. This energy balance, among others, can change the oxidative status. Oxidative stress occurs when antioxidant defense mechanisms are not sufficient to cope with the increasing generation of reactive oxygen species. Our objectives were to investigate (1) the effect of parity on the oxidative status of dairy cows (n = 247) in late pregnancy and early lactation; and (2) the effect of different inclusion rates of concentrate feeding (150 vs. 250 g/kg of energy-corrected milk) during early lactation on 2 farms including 87 cows in total. In addition, we aimed to compare the oxidative status across the 2 farms using equal portions of concentrate feeding. For these purposes, we measured concentrations of the derivatives of reactive oxygen metabolites (dROM) and the ferric reducing ability (FRAP) in serum on d -50, -14, +8, +28, and +100 relative to calving. Furthermore, we calculated the oxidative status index (OSi) as dROM/FRAP × 100. Data were analyzed using a linear mixed model. Cows in the first and second lactations had greater dROM, FRAP, and OSi than cows in their third and greater lactations. Hence, supporting the antioxidative side of the balance might be of particular importance in the first and second lactations. Feeding different amounts of concentrates did not affect dROM, FRAP, or OSi under our experimental conditions, suggesting that the relatively small differences in energy intake were not affecting the oxidative status. Comparing farms, cows from one farm were notable for having greater dROM and lower FRAP, resulting in a greater OSi compared with cows on the other farm. Milk yield showed a time by farm interaction with 7% less milk on d 100 on the farm with the greater OSi. Moreover, cows on that farm had 1.4-fold greater β-hydroxybutyrate concentrations. Our results emphasize the value of assessing oxidative status with regard to both the pro- and antioxidative sides, and support the association between oxidative and metabolic status. Further investigations are needed to determine the applicability of OSi as a prognostic tool during early lactation and to determine which factors have the greatest influence on oxidative status.

Effect of postcalving serum nonesterified fatty acids concentration on the functionality of bovine immune cells

Abstract:The periparturient period is marked by metabolic, hormonal, and immunological changes, which have an effect on the incidence of infectious and metabolic diseases. In a previous study, a slower increase in milk production was induced by milking cows once daily during the first week of lactation, leading to an improvement in levels of several metabolites, including nonesterified fatty acids (NEFA) and β-hydroxybutyrate (BHBA). The aim was to determine the influence of serum collected on d 2, 5, and 61 postpartum from cows milked once or twice daily on immune cell functions and to determine which of the constituents were responsible for these effects. Peripheral blood mononuclear cells (PBMC) and polymorphonuclear leukocytes were collected from healthy midlactation cows and their immune functions (i.e., proliferation and interferon-γ production and chemotaxis, phagocytosis, and oxidative burst, respectively), were evaluated in presence of serum, NEFA, and BHBA. Proliferation of PBMC was greater with d-61 (65.1±1.6%) serum than with d-2 (37.3±2.4%) or d-5 (48.4±1.6%) serum and greater with d-2 and -5 serum from cows milked once (42.2±3.7 and 54.0±2.5) compared with cows milked twice daily (32.4±3.0 and 42.9±2.1). Proliferation was inversely correlated with the concentration of NEFA and BHBA in the serum (r=-0.86). Adding NEFA to d-61 serum to reach the level present in d-5 serum decreased proliferation to the level observed with d-5 serum. No effect of BHBA addition was observed. The release of interferon-γ by PBMC was lower in d-5 serum (766±63 pg/mL) than in d-61 serum (1,187±90 pg/mL) and by NEFA. Milking frequency did not affect chemotaxis, phagocytosis, or oxidative burst of polymorphonuclear leukocytes. Phagocytosis decreased over time in serum from d 2 to 61. Similarly, oxidative burst was greater with d-5 serum (12.7×10(8) ± 1.6×10(8) relative light units) than with d-61 serum (9.0×10(8) ± 1.6×10(8) relative light units). The NEFA had a negative effect on oxidative burst, but BHBA did not. In conclusion, several immune cell functions appear affected by the NEFA concentration. Therefore, strategies that prevent increases in blood NEFA during the transition period may limit postpartum immunosuppression.
[Medical disciplinary jurisprudence in The Netherlands; a 10-year review].

Abstract:OBJECTIVE To obtain information about the nature and the number of complaints, the complainants and the accused health professionals and about the sanctions imposed by the Dutch medical disciplinary courts. DESIGN Descriptive, retrospective study. SETTING Inspectorate for Health Care, Rijswijk, the Netherlands. METHODS All verdicts (n = 5333) given by the 5 medical disciplinary courts of first instance in the period 1983-1992 and the appeals arising from these verdicts were studied using the data of the Inspector in chief. These were related to the total number of practising clinicians obtained from the Inspectorate, the Royal Dutch Medical Association and Statistics Netherlands. The year of each verdict was noted, the number and nature of the verdicts, the categories of complainants and accused, the number of appeal cases and their consequences. RESULTS The number of complaints doubled in the period under study, rising from 379 to 720 per annum. Most of the complaints (92%) were against doctors, but the number of complaints were divided very unevenly over the clinical specialists. A large number of the complaints concerned 'lack of care or inadequate care' (27%), or 'incorrect treatment' (22%). Of all complaints 20% led to a sanction, mostly a warning (62%). Appeals were lodged against 29% of the verdicts. In 22 cases the licence to practice was withdrawn definitively. CONCLUSIONS Although the number of complaints increased over the study period, there was no reduction in the seriousness of the complaints. The effect of the disciplinary jurisdiction on the quality of care has not yet been adequately investigated.
Dairy cow parity affects relationships among nutritional parameters in the blood of dams, umbilical cords, and calves and placental development at calving

Abstract:Heifer growth and milk production in lactating cows may diminish the nutrient supply to the fetus. This study aimed to analyze the characteristics of the nutrient supply to the fetus in primiparous and multiparous cows. We investigated maternal, umbilical cord, and calf blood glucose and amino acid levels, as well as placental development in 28 primiparous (PP) and 30 multiparous (MP) Holstein cows. Although the total cotyledonary weight and surface area showed no significant differences, the MP group exhibited larger individual cotyledons (P < 0.01) and fewer medium-sized cotyledons (P < 0.05). Within the PP group, total cotyledonary weight and surface area positively correlated with blood glucose (r = 0.71-0.77; P < 0.01) and total essential amino acid (r = 0.55; P < 0.05) concentrations in the umbilical veins. However, no significant correlation was observed in the MP group. Blood glucose and amino acid concentrations in the umbilical vein, umbilical artery, and calf were significantly lower in the MP group (P < 0.05), although no difference was observed in the dams between the groups. In conclusion, the nutrient status of primiparous cows can alter fetal nutrient supply. Moreover, multiparous cows have larger individual cotyledons as an adaptive response to increased milk production during pregnancy. However, this adaptive response in multiparous cows did not completely restore nutrient supply to the fetus to the same extent as that in primiparous cows. Therefore, the nutritional management of multiparous cows during pregnancy must be reconsidered.

Abstract:This study aimed to evaluate the effects of parity, body condition score (BCS) at calving, and milk yield on the metabolic profile of Gyr (Zebu) cows. Healthy cows in late pregnancy were grouped according to parity (primiparous, biparous, and multiparous); to BCS scale at calving (high—HBCS and normal—NBCS); and to milk yield (high—HP and moderate—MP production). BCS was assessed, and blood samples were collected on −21, −7, 0, 7, 21, and 42 days relative to parturition. The concentrations of non-esterified fatty acids (NEFA), beta-hydroxybutyrate (BHB), cholesterol, glucose, total protein (TP), albumin, total calcium (Ca), phosphorus (P), and magnesium (Mg); and activities of aspartate aminotransferase and gamma-glutamyltransferase were measured. Data were analyzed by two-way repeated measures ANOVA. The frequencies of high lipomobilization, subclinical ketosis, subclinical hypocalcemia (SCH), and the occurrence of diseases during early lactation were established. Regardless of grouping, NEFA, BHB, and cholesterol increased during early lactation; glucose showed higher values at calving; TP and albumin were higher at 21 and 42 DIM; and Ca, P, and Mg were lower at calving. Parity had little effect on the metabolic profile, HBCS did not differ from NBCS cows, and HP did not differ from MP cows in most metabolites. High lipomobilization in early lactation and SCH at calving were the most common imbalances but were not related to postpartum diseases. High-yielding Gyr cows have a balanced metabolic profile during the transition period, with few biologically relevant effects of parity, BCS at parturition, or milk yielded.

Abstract:Abstract Background: A standard chemotherapy for recurrent/metastatic salivary gland cancers has not been established. Combination chemotherapy of carboplatin and paclitaxel should be evaluated as a treatment option. Methods: This study retrospectively reviewed salivary gland cancer patients who received combination chemotherapy of carboplatin and paclitaxel. The differences in objective responses and in the prognoses according to the different pathological diagnoses were evaluated. Results: A total of 38 patients were enrolled in the study; of them, 18 had salivary duct carcinomas (SDCs), nine had adenoid cystic carcinomas (ACCs), and 11 had other pathological diagnoses. Objective responses were observed in 15 (39%) patients. The median progression-free survival (PFS) was 6.5 months, and the median overall survival (OS) was 26.5 months. ACC patients had relatively low response rates (9%), but there were no significant differences in PFS or OS compared to other sub-types. The treatment was well tolerated, with few adverse events. Conclusion: Salivary gland cancer patients showed a moderate clinical response to the combination chemotherapy of carboplatin and paclitaxel. The objective response rates differed according to the pathological diagnoses, but there were no significant differences in prognoses.
